Lithium-ion batteries (LIBs) have been used as energy storage devices for appliances of a wide range of sizes, for example, mobile phones, laptops, and electric vehicles. However, current commercial electrode materials in LIBs are facing severe challenges in energy density, safety, price, and recyclability, and this energy storage technology needs to be further developed for high power application in future. To address this challenge, nanostructured materials which possess large surface-to-volume ratios have been intensively studied as potential electrode materials to improve the battery performance. Among them, Tin (Sn) and nanostructured TiO2 have shown some advantages and disadvantages as electrode materials, for example Sn has high storage capacity but suffer from severe volume expansion during lithiation/delithiation. On the other hand, nanostructured TiO2 has small volume changes during Li ion insertion/extraction, but the poor ion mobility and electronic conductivity limited it electrochemical application. This review summarizes the recent development on Sn and TiO2 based nanostructure materials, particularly the synthesis method, their structure and battery performance as anodes for LIBs. Then, we discuss the strategies of developing new electrode materials and highlight a hybrid approach for designing and synthesizing high-performance electrode materials. The latest developments and related electrochemical performance of anode materials demonstrate that design hybrid nanostructure material is a low-cost and scalable approach to produce high-performance electrode materials for high-energy LIBs.

A method for regioselective ring openings of 3,4- and 2,3-epoxy alcohols with ambident nitrogen heterocycles is described. Using a diarylborinic acid catalyst, a single regioisomer is favored in couplings of nucleophile and electrophile partners that display low regioselectivity under conventional conditions. The method provides access to aromatic heterocycles bearing stereochemically defined, functionalized alkyl substituents, a product class similar in structure to medicinally relevant compounds such as the acyclic nucleoside analogues.

Anti-Wolbachia therapy has been identified as a viable treatment for combating filarial diseases. Phenotypic screening revealed a series of pyrazolopyrimidine hits with potent anti-Wolbachia activity. This paper focuses on the exploration of the SAR for this chemotype, with improvement of metabolic stability and solubility profiles using medicinal chemistry approaches. Organic synthesis has enabled functionalization of the pyrazolopyrimidine core at multiple positions, generating a library of compounds of which many analogues possess nanomolar activity against Wolbachia in vitro with improved DMPK parameters. A lead compound, 15f, was selected for in vivo pharmacokinetics (PK) profiling in mice. The combination of potent anti-Wolbachia activity in two in vitro assessments plus the exceptional oral PK profiles in mice puts this lead compound in a strong position for in vivo proof-of-concept pharmacodynamics studies and demonstrates the strong potential for further optimization and development of this series for treatment of filariasis in the future.

Gemcitabine and nab-paclitaxel (Abraxane®) is a standard of care chemotherapy combination used in the treatment of patients with advanced pancreatic cancer. While the combination has shown a survival benefit when compared to gemcitabine monotherapy, it is associated with significant off-target toxicity. Ultrasound targeted microbubble destruction (UTMD) has emerged as an effective strategy for the site-specific deposition of drug-payloads. However, loading a single microbubble formulation with two drug payloads can be challenging and often involves several manipulations post-microbubble preparation that can be cumbersome and generally results in low / inconsistent drug loadings. In this manuscript, we report the one-pot synthesis of a gemcitabine functionalised phospholipid and use it to successfully generate stable microbubble formulations loaded with gemcitabine (Lipid-Gem MB) or a combination of gemcitabine and paclitaxel (Lipid-Gem-PTX MB). Efficacy of the Lipid-Gem MB and Lipid-Gem-PTX MB formulations, following ultrasound (US) stimulation, was evaluated in a three-dimensional (3D) PANC-1 spheroid model of pancreatic cancer and a mouse model bearing ectopic BxPC-3 tumours. The results demonstrated a significant reduction in the cell viability in spheroids for both formulations reducing from 90 ± 10% to 62 ± 5% for Lipid-Gem MB and 84 ± 10% to 30 ± 6% Lipid-Gem-PTX MB following US irradiation. When compared with a clinically relevant dose of free gemcitabine and paclitaxel (i.e. non-particle bound) in a BxPC-3 murine pancreatic tumour model, both formulations also improved tumour growth delay with tumours 40 ± 20% and 40 ± 30% smaller than the respective free drug formulation when treated with Lipid-Gem MB and Lipid-Gem-PTX MB respectively, at the conclusion of the experiment. These results highlight the potential of UTMD mediated Gem / PTX as a treatment for pancreatic cancer and the facile preparation of Lipid-Gem-PTX MBs using a gemcitabine functionalised lipid should expedite clinical translation of this technology.

Halogen bonding represents a powerful tool in the field of noncovalent interactions. However, applications in enantioselective recognition and catalysis remain almost nonexistent, due in part to the distinct features of halogen bonds, including long covalent and noncovalent bond distances and high directionality. Herein, this work presents a novel chiral tetrakis‐iodo‐triazole structure as a neutral halogen bond donor for both chiral anion‐recognition and enantioinduction in ion‐pair organocatalysis. NMR‐titration studies revealed significant differences in anion affinity between the halogen bonding receptor and its hydrogen bonding parent. Selective recognition of chiral dicarboxylates and asymmetric induction in a benchmark organocatalytic reaction were demonstrated using the halogen bond donor. Inversions in the absolute sense of chiral recognition, enantioselectivity, and chiroptical properties relative to the related hydrogen donor were observed. Computational modeling suggested that these effects were the result of distinct anion‐binding modes for the halogen‐ versus hydrogen‐bond donors., Halogen bonding interactions at work: This work presents a novel chiral tetrakis‐iodo‐triazole as neutral halogen bond donor able to differentiate chiral dicarboxylates and induce chirality in an anion‐binding catalytic reaction. An inversion of chiral recognition, enantioselectivity and CD spectra compared to its parent hydrogen donor was observed and examined by DFT calculations, providing a hint on relevant halogen donor complex structures in solution.

Site-selective oxidations of carbohydrates, employing acridinium photocatalysis and quinuclidine hydrogen atom transfer catalysis, are presented. Protocols have been developed for oxidations of all-equatorial carbohydrates as well as those containing cis-1,2-diols. Site-selectivity reflects the relative rates of hydrogen atom transfer from the carbohydrate C-H bonds, and can be enhanced using a phosphate hydrogen-bonding or boronic acid catalyst.

Methods for the O-arylation of hydroxyl and hemiacetal groups in carbohydrates via C(sp2)-O bond formation are discussed. Such methods provide an alternative disconnection to the traditional approach of nucleophilic substitution between a sugar-derived electrophile and a phenol or phenoxide nucleophile. They have led to new opportunities for stereoselectivity, site-selectivity and chemoselectivity in the preparation of O-aryl glycosides and carbohydrate-derived aryl ethers, compounds that are useful for a broad range of applications in medicinal chemistry, glycobiology and organic synthesis.

Introduction Sebaceous hyperplasia (SH) is a common skin presentation in adults. Due to their unwanted yellow papular appearance, patients may desire their removal. Although several treatment modalities have been reported, the full range and efficacy of options are unclear. Objective To determine the efficacy of laser modalities in the treatment of SH. The authors will also specifically assess the efficacy, recurrence rate and side effect profile of SH treatment with Er:YAG wavelength using a variable long pulsed (VLP) Er:YAG laser (SP Dynamis Fotona laser, Ljubljana, Slovenia) Methods a Materials: A comprehensive literature search was performed through PubMed, EMBASE, and Web of Science, using the search terms [(sebaceous hyperplasia)] and [(laser[s], Er:Yag, Er:Glass, Fraxel, CO2, PDL, Pulse dye laser, Diode, Xe-Cl, Excimer, Argon, KTP, Ruby, Alexandrite or Nd:YAG)]. The search yielded a total of 119 results and 8 were identified as relevant to this reviewResults: Pulse dye laser (PDL) provides a wide range of treatment results from complete reduction to flattening of the SH without significant adverse events; recurrence rates were unreported. Short PDL showed faster treatment response than long PDL. CO2 laser can produce considerable positive cosmetic outcomes with marked clinical improvement without any recurrence, but significant adverse effects have been reported. The 1450-nm diode laser has been described to produce good (75%) clinical improvement and lesion shrinkage ranging from 50% to greater than 75% without lasting adverse effects. In our clinic, Er:YAG has provided very significant cosmetic outcomes with a low recurrence rate and minimal adverse effects. Conclusions Laser modalities can provide satisfactory results for removing SH. It is crucial that the laser is being used by an expert who is familiar with the device as well as understand the laser tissue interaction to minimize patient adverse effects while providing the best cosmetic outcome. Background Current treatment options for onchocerciasis are sub-optimal, prompting research and development of a safe cure (macrofilaricide). Onchocerca ochengi, a parasite of cattle, is used as a close surrogate for the human parasite O. volvulus in a murine model for pre-clinical screening of macrofilaricides. Skin from naturally infected cattle have been used in previous studies as a reliable source of parasite material. However, there is limited knowledge on how source-related factors such as the microfilaridermia status of the cattle, the nodule load and nodular worm viability may affect survival of male O. ochengi worms implanted in the rodent hosts. Such relationships were investigated in this study. Methods Dermal tissue and nodules were obtained from Gudali cattle, dissected and cultured to obtain migrating microfilariae (mf) and male worms. Emerged male worms were implanted into SCID mice and Gerbils (Meriones unguiculatus) and recovery rates were determined upon 42 days post implantation. Finally, nodules were processed for histology and embryogram analyses to assess the nodular worm viability and fertility, respectively. Results Of the 69 cattle sampled, 24 (34.8%) were mf+ and 45 (65.2%) were mf–. The mean nodule loads were 180.5 ± 117.7 (mf+) and 110.6 ± 102.7 (mf-) (p = 0.0186). The mean male worm harvest from nodules were 76.8 ± 120.3 and 47.2 ± 33.4 (p = 0.2488) for mf+ and mf– cattle, respectively. The number of male worms per 100 nodules were 57/100 and 46/100 nodules for mf+ and mf– cows, respectively. Female worms from nodules of mf– cows had higher counts of both normal and abnormal embryos with higher proportions of dead nodular worms evinced by histology compared to those from mf+ cows. A total of 651 worms were implanted into mice and gerbils, out of which 129 (19.81%) were recovered. Logistic regression analysis indicated that the microfilaridermia status of the cattle (presence of mf) (OR = 4.3319; P = 0.001) is the single most important predictor of the success of male worm recovery after implantation into rodents. Conclusion Microfilaridermic cattle provide a promising source of adult O. ochengi. Male worms from this group of cattle have a better success rate of survival in a murine implant model. Tuber dormancy and sprouting are commercially important potato traits as long-term tuber storage is necessary to ensure year-round availability. Premature dormancy release and sprout growth in tubers during storage can result in a significant deterioration in product quality. In addition, the main chemical sprout suppressant chlorpropham has been withdrawn in Europe, necessitating alternative approaches for controlling sprouting. Breeding potato cultivars with longer dormancy and slower sprout growth is a desirable goal, although this must be tempered by the needs of the seed potato industry, where dormancy break and sprout vigour are required for rapid emergence. We have performed a detailed genetic analysis of tuber sprout growth using a diploid potato population derived from two highly heterozygous parents. A dual approach employing conventional QTL analysis allied to a combined bulk-segregant analysis (BSA) using a novel potato whole-exome capture (WEC) platform was evaluated. Tubers were assessed for sprout growth in storage at six time-points over two consecutive growing seasons. Genetic analysis revealed the presence of main QTL on five chromosomes, several of which were consistent across two growing seasons. In addition, phenotypic bulks displaying extreme sprout growth phenotypes were subjected to WEC sequencing for performing BSA. The combined BSA and WEC approach corroborated QTL locations and served to narrow the associated genomic regions, while also identifying new QTL for further investigation. Overall, our findings reveal a very complex genetic architecture for tuber sprouting and sprout growth, which has implications both for potato and other root, bulb and tuber crops where long-term storage is essential.

Mandrakes (Mandragora spp., Solanaceae) are known to contain tropane alkaloids and have been used since antiquity in traditional medicine. Tropane alkaloids such as scopolamine and hyoscyamine are used in modern medicine to treat pain, motion sickness, as eye pupil dilators and antidotes against organo-phosphate poisoning. Hyoscyamine is converted to 6β-hydroxyhyoscyamine (anisodamine) and scopolamine by hyoscyamine 6β-hydroxylase (H6H), a 2-oxoglutarate dependent dioxygenase. We describe here a marked chemo-diversity in the tropane alkaloid content in Mandragora spp. M. officinarum and M. turcomanica lack anisodamine and scopolamine but display up to 10 fold higher hyoscyamine levels as compared with M. autumnalis. Transcriptomic analyses revealed that H6H is highly conserved among scopolamine-producing Solanaceae. MoH6H present in M. officinarum differs in several amino acid residues including a homozygotic mutation in the substrate binding region of the protein and its prevalence among accessions was confirmed by Cleaved-Amplified-Polymorphic-Sequence analyses. Functional expression revealed that MaH6H, a gene isolated from M. autumnalis encodes an active H6H enzyme while the MoH6H sequence isolated from M. officinarum was functionally inactive. A single G to T mutation in nucleotide 663 of MoH6H is associated with the lack of anisodamine and scopolamine in M. officinalis.

Copper-mediated couplings of arylboroxines with glycosyl hemiacetals furnish O-aryl glycosides via Csp2-O bond formation. The method enables the anomeric O-arylation of protected pyranose and furanose derivatives, and is tolerant of functionalized arylboroxine partners. Whereas mixtures of anomers are formed from glucopyranose, galactopyranose and arabinofuranose hemiacetals, the α-anomer is generated selectively from mannopyranose and mannofuranose-derived substrates.

A protocol for selective monoesterification of glycerol and pentose alcohols with fatty acids is described, using phenylboronic acid as a phase-transfer reagent. Formation of arylboronic ester intermediates serves both to increase the solubility of the alcohol substrate in nonpolar organic solvent and to selectively protect diol groups, allowing for efficient and selective condensation with the aliphatic carboxylic acid. A phase-switching workup with basic aqueous sorbitol solution is used to cleave the boronic ester groups and to separate the boronic acid from the monoester product. The method provides an operationally simple means of access to a class of bio-derived products that have been broadly applied as food additives, components of cosmetic products and pharmaceutical formations, plasticizers, and non-ionic surfactants.

Diorganotin dihalides act as cocatalysts for site-selective and stereoselective couplings of diol-containing carbohydrates with electron-deficient alkenes in the presence of an Ir(III) photoredox catalyst and quinuclidine, a hydrogen atom transfer mediator. Quantum-chemical calculations support a proposed mechanism involving the formation of a cyclic stannylene acetal intermediate that shows enhanced reactivity toward hydrogen atom abstraction by the quinuclidinium radical cation. Addition of the carbon-centered radical to the alkene partner results in C-alkylation of the carbohydrate substrate.

The aim of the study was to investigate the impact of Crohn's disease (CD) on the performance of a lipid-based formulation of ciprofloxacin in a complex gastrointestinal simulator (TIM-1, TNO) and to compare the luminal environment in terms of bile salt and lipid composition in CD and healthy conditions. CD conditions were simulated in the TIM-1 system with a reduced concentration of porcine pancreatin and porcine bile. The bioaccessibility of ciprofloxacin was similar in simulated CD and healthy conditions considering its extent as well as its time course in the jejunum and ileum filtrate. Differences were observed in terms of the luminal concentration of triglycerides, monoglycerides, and fatty acids in the different TIM-1 compartments, indicating a reduction and delay in the lipolysis of formulation excipients in CD. The quantitative analysis of bile salts revealed higher concentrations for healthy conditions (standard TIM-1 fasted-state protocol) in the duodenum and jejunum TIM-1 compartments compared to published data in human intestinal fluids of healthy subjects. The reduced concentrations of bile salts in simulated CD conditions correspond to the levels observed in human intestinal fluids of healthy subjects in the fasted state.A lipidomics approach with ultra performance liquid chromatography (UPLC)/mass spectrometry (MS) has proven to be a time-efficient method to semiquantitatively analyze differences in fatty acid and bile salt levels between healthy and CD conditions. The dynamic luminal environment in CD and healthy conditions after administration of a lipid-based formulation can be simulated using the TIM-1 system. For ciprofloxacin, an altered luminal lipid composition had no impact on its performance indicating a low risk of altered performance in CD patients.

Carbon dust refers to carbon particles, originating from carbon inputs into the smelting process like anodes, that float on top of the bath, below anodes or suspended in the bath. The phenomenon has a deleterious effect on the specific energy consumption of cells and can lead to anode deformations, hot cells out of the process window and stoppage of cells. Trials were conducted in the TRIMET Hamburg Smelter focusing on the effect of dust at anode changes. The conditions were chosen to be best and worst practice as assessed by a visual carbon dust assessment in the tap hole. Contrasting with published literature, there was no relation in the experiments between spike formation and carbon dust in anodes after 8 h. Anodes set in cells with a high carbon level in the tap hole did not behave differently when compared to anodes with a low carbon dust content in the tap hole. Samples obtained from the frozen bath layer underneath the anodes showed carbon contents in the range of 0.0315–6.29%.

A flexible polymer chain in the presence of inert macromolecular crowders will experience a loss of configurational entropy due to the crowder excluded volume. This entropy reduction will be most pronounced in good solvent conditions where the chain assumes an expanded coil conformation. For polymers that undergo a folding transition from a coil to a compact ordered state, as is the case for many globular proteins, macromolecular crowding is expected to stabilize the folded state and thereby shift the transition location. Here, we study such entropic stabilization effects for a tangent square-well sphere chain (monomer diameter σ) in the presence of hard-sphere (HS) crowders (diameter D ≥ σ). We use the Wang–Landau simulation algorithm to construct the density of states for this chain in a crowded environment and are thus able to directly compute the reduction in configurational entropy due to crowding. We study both a chain that undergoes all-or-none folding directly from the coil state and a chain that folds via a collapsed-globule intermediate state. In each case, we find an increase in entropic stabilization for the compact states with an increase in crowder density and, for fixed crowder density, with a decrease in crowder size (concentrated, small crowders have the largest effect). The crowder significantly reduces the average size for the unfolded states while having a minimal effect on the size of the folded states. In the athermal limit, our results directly provide the confinement free energy due to crowding for a HS chain in a HS solvent.

Summary Inflammatory response heterogeneity has impeded high-resolution dissection of diverse immune cell populations during activation. We characterize mouse cutaneous immune cells by single-cell RNA sequencing, after inducing inflammation using imiquimod and oxazolone dermatitis models. We identify 13 CD45+ subpopulations, which broadly represent most functionally characterized immune cell types. Oxazolone pervasively upregulates Jak2/Stat3 expression across T cells and antigen-presenting cells (APCs). Oxazolone also induces Il4/Il13 expression in newly infiltrating basophils, and Il4ra and Ccl24, most prominently in APCs. In contrast, imiquimod broadly upregulates Il17/Il22 and Ccl4/Ccl5. A comparative analysis of single-cell inflammatory transcriptional responses reveals that APC response to oxazolone is tightly restricted by cell identity, whereas imiquimod enforces shared programs on multiple APC populations in parallel. Synthetic microfibers have been reported in most aquatic environments and represent a large proportion of environmental microplastics. However, they remain largely under-represented in microplastic ecotoxicity studies. The present study aims to investigate particle interaction with, and retention time in, aquatic organisms comparing microfibers, and microbeads. We used brine shrimp (Artemia sp.) and fish (Gasterosteus aculeatus) as invertebrate and vertebrate models, respectively. Organisms were exposed to a mixture of microbeads (polyethylene, 27–32 μm) and microfibers (dope dyed polyester; 500 μm-long) for 2 h, at high concentrations (100,000 part./L) in order to maximize organism-particles interaction. Artemia were exposed in the presence or absence of food. Fish were exposed either via the trophic route or directly via water, and water exposures were performed either in freshwater or seawater. In the absence of food, Artemia ingested high numbers of microbeads, retained in their digestive tract for up to 96 h. Microfiber ingestion was very limited, and its egestion was fast. In the presence of food, no microfiber was ingested, microbead ingestion was limited, and egestion was fast (48 h). Limited particle ingestion was observed in fish exposed via water, and particle retention time in gut did not exceed 48 h, both for direct and trophic exposure. However, water exposures resulted in a higher number of particles present in gills, and average retention time was higher in gills, compared to gut. This suggests that gills are organs susceptible to microplastic exposure and should be taken into account in fish exposure and effect studies. Our results show that particle ingestion and retention by organisms differ between microbeads and microfibers, suggesting particle selection based on size, shape, and/or color and species-specific selective feeding. We also showed that the presence of food results in limited particle ingestion and retention in Artemia and that microbeads are more likely to be transferred to organisms from upper trophic levels than microfibers. Finally, fish exposure to particles was not significantly different between freshwater and seawater conditions.

Potato, S. tuberosum, is one of the most important global crops, but has high levels of waste due to tuber greening under light, which is associated with the accumulation of neurotoxic glycoalkaloids. However, unlike the situation in de-etiolating seedlings, the mechanisms underlying tuber greening are not well understood. Here, we have investigated the effect of monochromatic blue, red, and far-red light on the regulation of chlorophyll and glycoalkaloid accumulation in potato tubers. Blue and red wavelengths were effective for induction and accumulation of chlorophyll, carotenoids and the two major potato glycoalkaloids, α-solanine and α-chaconine, whereas none of these accumulated in darkness or under far-red light. Key genes in chlorophyll biosynthesis (HEMA1, encoding the rate-limiting enzyme glutamyl-tRNA reductase, GSA, CHLH and GUN4) and six genes (HMG1, SQS, CAS1, SSR2, SGT1 and SGT2) required for glycoalkaloid synthesis were also induced under white, blue, and red light but not in darkness or under far-red light. These data suggest a role for both cryptochrome and phytochrome photoreceptors in chlorophyll and glycoalkaloid accumulation. The contribution of phytochrome was further supported by the observation that far-red light could inhibit white light-induced chlorophyll and glycoalkaloid accumulation and associated gene expression. Transcriptomic analysis of tubers exposed to white, blue, and red light showed that light induction of photosynthesis and tetrapyrrole-related genes grouped into three distinct groups with one group showing a generally progressive induction by light at both 6 h and 24 h, a second group showing induction at 6 h in all light treatments, but induction only by red and white light at 24 h and a third showing just a very moderate light induction at 6 h which was reduced to the dark control level at 24 h. All glycoalkaloid synthesis genes showed a group one profile consistent with what was seen for the most light regulated chlorophyll synthesis genes. Our data provide a molecular framework for developing new approaches to reducing waste due to potato greening.

Transient receptor potential vanilloid 4 channels (TRPV4) are pivotal regulators of vascular homeostasis. Altered TRPV4 signaling has recently been implicated in various cardiovascular diseases, including hypertension and atherosclerosis. These versatile nonselective cation channels increase endothelial Ca2+ influx in response to various stimuli including shear stress and G protein-coupled receptor (GPCR) activation. Recent findings suggest TRPV4 channels produce localized Ca2+ transients at the endothelial cell plasma membrane that may allow targeted effector recruitment and promote large-scale Ca2+ events via release from internal stores (endoplasmic reticulum). However, the specific impact of TRPV4 channels on Ca2+ signaling in the intact arterial intima remains unknown. In the current study, we employ an endothelium-specific TRPV4 knockout mouse model (ecTRPV4&minus, /&minus, ) to identify and characterize TRPV4-dependent endothelial Ca2+ dynamics. We find that carotid arteries from both ecTRPV4&minus, and WT mice exhibit a range of basal and acetylcholine (ACh)-induced Ca2+ dynamics, similar in net frequency. Analysis of discrete Ca2+ event parameters (amplitude, duration, and spread) and event composite values reveals that while ecTRPV4&minus, artery endothelium predominantly produces large Ca2+ events comparable to and in excess of those produced by WT endothelium, they are deficient in a particular population of small events, under both basal and ACh-stimulated conditions. These findings support the concept that TRPV4 channels are responsible for generating a distinct population of focal Ca2+ transients in the intact arterial endothelium, likely underlying their essential role in vascular homeostasis.

Potato production is often constrained by abiotic stresses such as drought and high temperatures which are often present in combination. In the present work, we aimed to identify key mechanisms and processes underlying single and combined abiotic stress tolerance by comparative analysis of tolerant and susceptible cultivars. Physiological data indicated that the cultivars Desiree and Unica were stress tolerant while Agria and Russett Burbank were stress susceptible. Abiotic stress caused a greater reduction of photosynthetic carbon assimilation in the susceptible cultivars which was associated with a lower leaf transpiration rate. Oxidative stress, as estimated by the accumulation of malondialdehyde was not induced by stress treatments in any of the genotypes with the exception of drought stress in Russett Burbank. Stress treatment resulted in increases in ascorbate peroxidase activity in all cultivars except Agria which increased catalase activity in response to stress. Transcript profiling highlighted a decrease in the abundance of transcripts encoding proteins associated with PSII light harvesting complex in stress tolerant cultivars. Furthermore, stress tolerant cultivars accumulated fewer transcripts encoding a type-1 metacaspase implicated in programmed cell death. Stress tolerant cultivars exhibited stronger expression of genes associated with plant growth and development, hormone metabolism and primary and secondary metabolism than stress susceptible cultivars. Metabolite profiling revealed accumulation of proline in all genotypes following drought stress that was partially suppressed in combined heat and drought. On the contrary, the sugar alcohols inositol and mannitol were strongly accumulated under heat and combined heat and drought stress while galactinol was most strongly accumulated under drought. Combined heat and drought also resulted in the accumulation of Valine, isoleucine, and lysine in all genotypes. These data indicate that single and multiple abiotic stress tolerance in potato is associated with a maintenance of CO2 assimilation and protection of PSII by a reduction of light harvesting capacity. The data further suggests that stress tolerant cultivars suppress cell death and maintain growth and development via fine tuning of hormone signaling, and primary and secondary metabolism. This study highlights potential targets for the development of stress tolerant potato cultivars.

Anode cover material (ACM) partly fuses at the bottom where it interacts with anode and bath in the aluminium reduction cell. This consolidated material is defined as the top crust in the cell, and plays a crucial role in maintaining heat balance, protecting the carbon anode from air-burning and absorbing the fluorides evaporation in the cells. Previous studies have been conducted to illustrate the crust formation and properties. However, there is rarely a systematic structural analysis of crust in the vertical direction due to the complexity and inhomogeneity of industrial crust. In the present study, the phase composition of crust was systematically analysed in the vertical direction, and five zones were identified based on the phase composition. Furthermore, two types of particles are identified in top zone (1) namely: smelter grade alumina (SGA) and crushed bath particles (CBPs). A systematic analysis on CBPs have shown four types of structural features.

The Hevea brasiliensis rubber tree is the largest commercial producer of natural rubber. The ability to synthesize a naturally occurring polymer is one of the main attractions of studying latex rubber biosynthesis. Natural rubber consists of high molecular weight cis-polyisoprene, a branch product of the isoprenoid network, which is synthesized by sequential condensations of isopentenyl diphosphate (IPP). To date, genomic technologies have enabled the generation of Hevea transcriptomes, draft genomes and rubber particle proteomes for a number of key tree clones (or cultivars). These have contributed significantly to the molecular genetics of genes encoding the mevalonate (MVA) and methylerythritol 4-phosphate (MEP) pathways of IPP synthesis, including rubber transferase candidates that participate in the final steps of cis-polyisoprene formation. Rapidly developing sequencing technologies will continue to provide opportunities to yield new insights into rubber biosynthesis. This chapter discusses the prospects of furthering genomics analysis of rubber biosynthesis in the following areas: regulation of pathways supplying and utilising IPP for rubber and non-rubber isoprenoids, characterization of rubber particle proteins in relation to molecular weight distribution and lipid droplet membrane proteins, and the potential benefits of haplotype analysis of rubber biosynthesis pathway genes for genomics-driven breeding.

Methods for site-selective transformations of hydroxyl groups in carbohydrate derivatives are reviewed. The construction of oligosaccharides of defined connectivity hinges on such transformations, which are also needed for the preparation of modified or non-natural sugar derivatives, the installation of naturally occurring postglycosylation modifications, the selective labeling or conjugation of carbohydrate derivatives, and the preparation of therapeutic agents or research tools for glycobiology. The review begins with a discussion of intrinsic factors and processes that can influence selectivity in reactions of unprotected or partially protected carbohydrate derivatives, followed by a description of transformations that engage two OH groups in cyclic adducts (acetals, ketals, boronic esters, and related species). An overview of the various classes of site-selective transformations of OH groups in sugars is then provided: the reactions discussed include esterification, thiocarbonylation, alkylation, glycosylation, arylation, silylation, phosphorylation, sulfonylation, sulfation, and oxidation. Emphasis is placed on recently developed methods that employ reagent or catalyst control to achieve otherwise challenging transformations or site-selectivities.

To gain insight into the genetic regulation of lipid metabolism in tomato, we conducted metabolic trait loci (mQTL) analysis following the lipidomic profiling of fruit pericarp and leaf tissue of the Solanum pennellii introgression lines (IL). To enhance mapping resolution for selected fruit-specific mQTL, we profiled the lipids in a subset of independently derived S. pennellii backcross inbred lines, as well as in a near-isogenic sub-IL population. We identified a putative lecithin:cholesterol acyltransferase that controls the levels of several lipids, and two members of the class III lipase family, LIP1 and LIP2, that were associated with decreased levels of diacylglycerols (DAGs) and triacylglycerols (TAGs). Lipases of this class cleave fatty acids from the glycerol backbone of acylglycerols. The released fatty acids serve as precursors of flavor volatiles. We show that LIP1 expression correlates with fatty acid-derived volatile levels. We further confirm the function of LIP1 in TAG and DAG breakdown and volatile synthesis using transgenic plants. Taken together, our study extensively characterized the genetic architecture of lipophilic compounds in tomato and demonstrated at molecular level that release of free fatty acids from the glycerol backbone can have a major impact on downstream volatile synthesis.

Diarylborinic acids (Ar2BOH) catalyze the C3-selective ring opening of 3,4-epoxy alcohols with aniline, dialkylamine and arenethiol nucleophiles. The regiochemical outcome is consistent with a catalytic tethering mechanism in which the borinic acid interacts with both the electrophile and the nucleophile. The rate acceleration resulting from this induced intramolecularity effect is sufficient to overcome steric biases that would otherwise favor C4-selective opening of the substituted epoxy alcohols.

A new mode of catalysis of the semipinacol rearrangement of 2,3-epoxy alcohols is described. In combination with a halide salt additive, diarylborinic acids promote a Type II rearrangement that occurs with net retention of configuration. This unusual stereochemical outcome is consistent with a mechanism involving regioselective ring opening of the epoxy alcohol by halide, followed by rearrangement of the resulting halohydrin-derived borinic ester. The protocol is applicable to a range of substrates, enabling ring contractions and expansions as well as stereospecific syntheses of acyclic β-hydroxycarbonyl compounds.

A powerful acquired thermotolerance response in potato was demonstrated and characterised in detail, showing the time course required for tolerance, the reversibility of the process and requirement for light. Potato is particularly vulnerable to increased temperature, considered to be the most important uncontrollable factor affecting growth and yield of this globally significant crop. Here, we describe an acquired thermotolerance response in potato, whereby treatment at a mildly elevated temperature primes the plant for more severe heat stress. We define the time course for acquiring thermotolerance and demonstrate that light is essential for the process. In all four commercial tetraploid cultivars that were tested, acquisition of thermotolerance by priming was required for tolerance at elevated temperature. Accessions from several wild-type species and diploid genotypes did not require priming for heat tolerance under the test conditions employed, suggesting that useful variation for this trait exists. Physiological, transcriptomic and metabolomic approaches were employed to elucidate potential mechanisms that underpin the acquisition of heat tolerance. This analysis indicated a role for cell wall modification, auxin and ethylene signalling, and chromatin remodelling in acclimatory priming resulting in reduced metabolic perturbation and delayed stress responses in acclimated plants following transfer to 40 °C.

Uterine contractions prior to 37 weeks gestation can result in preterm labor with significant risk to the infant. Current tocolytic therapies aimed at suppressing premature uterine contractions are largely ineffective and cause serious side effects. Calcium (Ca2+) dependent contractions of uterine smooth muscle are physiologically limited by the opening of membrane potassium (K+) channels. Exploiting such inherent negative feedback mechanisms may offer new strategies to delay labor and reduce risk. Positive modulation of small conductance Ca2+-activated K+ (KCa2.3) channels with cyclohexyl-[2-(3,5-dimethyl-pyrazol-1-yl)-6-methyl-pyrimidin-4-yl]-amine (CyPPA), effectively decreases uterine contractions. This study investigates whether the receptor agonist oxytocin might solicit KCa2.3 channel feedback that facilitates CyPPA suppression of uterine contractions. Using isometric force myography, we found that spontaneous phasic contractions of myometrial tissue from nonpregnant mice were suppressed by CyPPA and, in the presence of CyPPA, oxytocin failed to augment contractions. In tissues exposed to oxytocin, depletion of internal Ca2+ stores with cyclopiazonic acid (CPA) impaired CyPPA relaxation, whereas blockade of nonselective cation channels (NSCC) using gadolinium (Gd3+) had no significant effect. Immunofluorescence revealed close proximity of KCa2.3 channels and ER inositol trisphosphate receptors (IP3Rs) within myometrial smooth muscle cells. The findings suggest internal Ca2+ stores play a role in KCa2.3-dependent feedback control of uterine contraction and offer new insights for tocolytic therapies.

Senescent sweetening results in the accumulation of reducing sugars in potato tubers following extended periods of storage at moderate temperatures used to avoid the separate condition of cold-induced sweetening. It represents a significant problem for the potato processing industry due to the development of dark fry colour and the accumulation of acrylamide in processed products. Previous studies have implicated oxidative stress in the accumulation of reducing sugars in potato tubers over long term storage. However, in the present analysis we found no evidence for a correlation between oxidative stress as estimated from quantification of hydrogen peroxide, lipid peroxidation or activity of redox enzymes and the accumulation of reducing sugars. On the contrary, transcriptional profiling indicated changes in carbohydrate metabolism were associated with the onset of senescent sweetening and qRT-PCR indicated that reduced abundance of transcripts encoding a plastidial glucose-6-phosphate/phosphate translocator was widely observed during sweetening onset in multiple genotypes. Our data suggest that reduction in the capacity of plastids to import glucose-6-phosphate reduces the capacity for starch resynthesis in the stored tuber thereby shifting the metabolic balance towards starch turnover resulting in reducing sugar accumulation.

In the aluminum electrolysis process, new industrial aluminum/electricity power markets demand a new cell technology to extend the cell heat balance and amperage operating window of smelters by shifting the steady states. The current work investigates the responses of lithium-modified bath system when the input/output balance is shifted in a laboratory analogue to the industrial heat balance shift. Li2CO3 is added to the cryolite-AlF3-CaF2-Al2O3 system as a bath modifier. A freeze deposit is formed on a ‘cold finger’ dipped into the bath and investigated by X-ray diffraction analysis and electron probe X-ray microanalysis. The macro- and micro-structure of the freeze lining varies with the bath superheat (bath temperature minus bath liquidus temperature) and an open crystalline layer with entrapped liquid dominates the freeze thickness. Compared with the cryolite-AlF3-CaF2-Al2O3 bath system, the lithium-modified bath freeze is more sensitive to the heat balance shift. This freeze investigation provides primary information to understand the variation of the side ledge in an industrial cell when the lithium-modified bath system is used.

Site-selective functionalization of hydroxy groups in sugar derivatives is a major challenge in carbohydrate synthesis. Methods for achieving this goal will provide efficient access to new sugar-derived chemical building blocks and will facilitate the preparation or late-stage modification of complex oligosaccharides for applications in glycobiology research and drug discovery. Here, we describe site-selective, copper-promoted couplings of boronic acids with carbohydrate derivatives. These reactions generate sugar-derived aryl ethers, a structural class that is challenging to generate by other means and has not previously been accessed in a site-selective fashion. Experimental evidence and computational modeling suggest that the formation of a sugar-derived boronic ester intermediate is crucial to the selectivity of these processes, accelerating the arylation of an adjacent hydroxy group. The results demonstrate how the interactions of sugars with boron compounds can be combined with transition metal catalysis to achieve new chemical reactivity.

In an aluminum electrolysis cell, the side ledge forms on side walls to protect it from the corrosive cryolitic bath. In this study, a series of laboratory analogue experiments have been carried out to investigate the microstructure and composition of side ledge (freeze linings) at different heat balance steady states. Three distinct layers are found in the freeze linings formed in the designed Cryolite-CaF2-AlF3-Al2O3 electrolyte system: a closed (columnar) crystalline layer, an open crystalline layer, and a sealing layer. This layered structure changes when the heat balance is shifted between different steady states, by melting or freezing the open crystalline layer. Phase chemistry of the freeze lining is studied in this paper to understand the side ledge formation process upon heat balance shifts. Electron probe X-ray microanalysis (EPMA) is used to characterize the microstructure and compositions of distinct phases existing in the freeze linings, which are identified as cryolite, chiolite, Ca-cryolite, and alumina. A freeze formation mechanism is further developed based on these microstructural/compositional investigations and also thermodynamic calculations through the software—FactSage. It is found that entrapped liquid channels exist in the open crystalline layer, assisting with the mass transfer between solidified crystals and bulk molten bath.

A flexible homopolymer chain with sufficiently short-range interactions undergoes a discontinuous transition from an expanded coil to a compact crystallite analogous to the all-or-none folding transition exhibited by fast-folding proteins. In this work we investigate the effects of both tethering and confinement on this type of folding transition. In particular, we study a flexible square-well N-mer chain (monomer diameter σ) located between two hard walls forming a slitlike pore (width W) with the chain end tethered to one wall. We carry out Monte Carlo simulations with Wang–Landau sampling to construct the single-chain density of states and use both microcanonical and canonical analyses to characterize phase transitions. We find that in a very wide pore (W > Nσ) the all-or-none folding transition is only slightly affected by the tethering. Intensifying legislation and increased research on the toxicological and persistent nature of per- and polyfluoroalkyl substances (PFASs) have recently influenced the direction of liquid repellent chemistry use; environmental, social, and sustainability responsibilities are at the crux. Without PFAS chemistry, it is challenging to meet current textile industry liquid repellency requirements, which is a highly desirable property, particularly in outdoor apparel where the technology helps to provide the wearer with essential protection from adverse environmental conditions. Herein, complexities between required functionality, legislation and sustainability within outdoor apparel are discussed, and fundamental technical performance of commercially available long-chain (C8) PFASs, shorter-chain (C6) PFASs, and non-fluorinated repellent chemistries finishes are evaluated comparatively. Non-fluorinated finishes provided no oil repellency, and were clearly inferior in this property to PFAS-finished fabrics that demonstrated good oil-resistance. However, water repellency ratings were similar across the range of all finished fabrics tested, all demonstrating a high level of resistance to wetting, and several non-fluorinated repellent fabrics provide similar water repellency to long-chain (C8) PFAS or shorter-chain (C6) PFAS finished fabrics. The primary repellency function required in outdoor apparel is water repellency, and we would propose that the use of PFAS chemistry for such garments is over-engineering, providing oil repellency that is in excess of user requirements. Accordingly, significant environmental and toxicological benefits could be achieved by switching outdoor apparel to non-fluorinated finishes without a significant reduction in garment water-repellency performance. These conclusions are being supported by further research into the effect of laundering, abrasion and ageing of these fabrics.

Resolving the source of environmental contamination is the critical first step in remediation and exposure prevention. Australia's oldest silver-zinc-lead mine at Broken Hill (>130 years old) has generated a legacy of contamination and is associated with persistent elevated childhood blood lead (Pb) levels. However, the source of environmental Pb remains in dispute: current mine emissions; remobilized mine-legacy lead in soils and dusts; and natural lead from geological weathering of the gossan ore body. Multiple lines of evidence used to resolve this conundrum at Broken Hill include spatial and temporal variations in dust Pb concentrations and bioaccessibility, Pb isotopic compositions, particle morphology and mineralogy. Total dust Pb loading (mean 255 μg/m2/day) and its bioaccessibility (mean 75% of total Pb) is greatest adjacent to the active mining operations. Unweathered galena (PbS) found in contemporary dust deposits contrast markedly to Pb-bearing particles from mine-tailings and weathered gossan samples. Contemporary dust particles were more angular, had higher sulfur content and had little or no iron and manganese. Dust adjacent to the mine has Pb isotopic compositions (208Pb/207Pb: 2.3197; 206Pb/207Pb: 1.0406) that are a close match (99%) to the ore body with values slightly lower (94%) at the edge of the city. The weight of evidence supports the conclusion that contemporary dust Pb contamination in Broken Hill is sourced primarily from current mining activities and not from weathering or legacy sources.

The responses of a cryolite-based bath to shifts in heat input/output balance have been investigated in previous work. The effect of such heat balance shifts on bath superheat and on the frozen ledge, protecting the cell walls, is of most interest and has been investigated primarily in an experimental analogue to a smelting cell. In this paper, the microstructure and morphology of the freeze itself, as a function of the applied heat balance shift is investigated. Different layers were detected in the freeze, showing that both the freeze morphology and the phases present vary depending on bath conditions. The phase composition change was also analysed and compared with the bulk bath in different thermal states, created when the heat balance was shifted. Based on the investigation, a freeze growth mechanism is proposed.

A structurally novel iodoperfluoroarene-substituted polystyrene has been synthesized, and its noncovalent halogen bonding interactions with polymeric Lewis bases have been studied. RAFT polymerization was used to generate halogen bond donating polystyrene derivatives with low dispersities. Coassembly of the halogen bond donor polymer with an amine-functionalized methacrylate block copolymer was achieved using a solvent switching protocol and studied by electron microscopy and dynamic light scattering analysis. The structures obtained include spheres, wormlike structures, and vesicles as well as “inverted” morphologies such as hexagonally packed hoops and bicontinuous structures, which have been challenging to access by conventional assembly methods. The ability to access such inverted structures suggests that complexes generated by noncovalent interactions between complementary macromolecules can display high effective packing parameters, giving rise to behavior that has generally required unconventional ...

When reaction progress kinetic analysis is implemented on data obtained using a spectroscopic or chromatographic technique such as ATR–FTIR, NMR, absorbance spectroscopy, HPLC or GC, an initial step involves fitting a mathematical function to the experimental concentration versus time data. This function is then differentiated to give graphs of rate versus substrate concentration (graphical rate equations). It is important that the function be able to accurately describe the experimental data without introducing bias or artifacts. Here, we put forward a nonlinear ordinary differential equation that can be fitted to experimental concentration versus time data for applications in reaction progress kinetic analysis. We show that it can be applied to data from catalytic transformations showing diverse types of kinetic behavior.

This study examines current soil contamination in an Australian industrial city, Newcastle. Public (roadside verges and parks) and private (homes) surface soils (n = 170) contained metal(loid)s elevated above their respective Australian Health Investigation Levels (HIL). Lead (Pb), the most common contaminant in the city, exceeds the HIL for residential soils (HIL-A, 300 mg/kg) in 88% of private soils (median: 1140 mg/kg). In-vitro Pb bio-accessibility analysis of selected soils (n = 11) using simulated gastric fluid showed a high affinity for Pb solubilisation (maximum Pb concentration: 5190 mg/kg, equating to 45% Pb bio-accessibility). Highly soluble Pb-laden Fe- and Mn-oxides likely contribute to the bio-accessibility of the Pb. Public and private space surface soils contain substantially less radiogenic Pb (range: 208 Pb/ 207 Pb: 2.345–2.411, 206 Pb/ 207 Pb: 1.068–1.312) than local background soil ( 208 Pb/ 207 Pb: 2.489, 206 Pb/ 207 Pb: 1.198), indicating anthropogenic contamination from the less radiogenic Broken Hill type Pb ores ( 208 Pb/ 207 Pb: 2.319, 206 Pb/ 207 Pb: 1.044). Source apportionment using Pb isotopic ratio quantification and soil mineralogy indicate the city's historic copper and steel industries contributed the majority of the soil contaminants through atmospheric deposition and use of slag waste as fill material. High-temperature silicates and oxides combined with rounded particles in the soil are characteristic of smelter dust emissions. Additionally, a preliminary investigation of polycyclic aromatic hydrocarbons in soils, sometimes associated with ferrous metal smelting, coal processing or burning of fossil fuels, shows that these too pose a health exposure risk (calculated in comparison to benzo( a )pyrene: n = 12, max: 13.5 mg/kg, HIL: 3 mg/kg).

The synthesis of stereodefined β-aminophosphines having both carbon- and phosphorus-based chirality centers is described. The method involves resolution of a mixture of β-aminophosphine oxide diastereomers accessed by ring-opening of an amino alcohol-derived cyclic sulfamidate. A stereospecific, borane-promoted reduction of β-aminophosphine oxides, which occurs under mild conditions and with inversion of configuration at phosphorus, is a key step in this process. The products obtained are new building blocks for the synthesis of P-chiral ligands and organocatalysts. In a proof-of-concept application in organocatalysis, the diastereomeric P-chiral β-aminophosphine-based bifunctional thioureas displayed significantly different activities in the Morita-Baylis-Hillman reaction of methyl acrylate with benzaldehyde derivatives.

A series of N4-benzylamine-N2-isopropyl-quinazoline-2,4-diamine derivatives has been synthesized and tested for antibacterial activity against five bacterial strains. Twelve different substituents on the N4-benzylamine group have been investigated along with replacement of the quinazoline core (with either a benzothiophene or regioisomeric pyridopyrimidine ring systems). In order to develop structure activity relationships, all derivatives were tested for their antibacterial activities against Escherichia coli and Staphylococcus aureus via Kirby–Bauer assays and minimum inhibitory concentration assays. Eight of the most potent compounds against S. aureus and E. coli were also screened against one strain of methicillin-resistant S. aureus (MRSA), Staphylococcus epidermidis and Salmonella typhimurium to further examine their antibacterial activities. Lead compound A5 showed good activities with MICs of 3.9 μg mL−1 against E. coli, S. aureus and S. epidermidis and 7.8 μg mL−1 against MRSA. Selected front runners were also screened for their DMPK properties in vitro to assess their potential for further development.
