Swimming pools are commonly treated with chlorine, which reacts with the natural organic matter and organic matter introduced by swimmers and form disinfection byproducts (DBPs) that are associated with respiratory-related issues, including asthma, in avid swimmers. We investigated a complementary disinfectant to chlorine, copper-silver ionization (CSI), with the aim of lowering the amount of chlorine used in pools and limiting health risks from DBPs. We sampled an indoor and outdoor pool treated with CSI-chlorine during the swimming season in 2017-2018 and measured 71 DBPs, speciated total organic halogen, in vitro mammalian cell cytotoxicity, and N-acetyl-l-cysteine (NAC) thiol reactivity as a cytotoxicity predictor. Controlled, simulated swimming pools were also investigated. Emerging DBP concentrations decreased by as much as 80% and cytotoxicity decreased as much as 70% in the indoor pool when a lower chlorine residual (1.0 mg/L) and CSI was used. Some DBPs were quantified for the first time in pools, including chloroacetaldehyde (up to 10.6 μg/L), the most cytotoxic haloacetaldehyde studied to date and a major driver of the measured cytotoxicity in this study. Three highly toxic iodinated haloacetic acids (iodoacetic acid, bromoiodoacetic acid, and chloroiodoacetic acid) were also quantified in pools for the first time. We also found that the NAC thiol reactivity was significantly correlated to cytotoxicity, which could be useful for predicting the cytotoxicity of swimming pool waters in future studies.

Recent studies used the sum of the measured concentrations of individual disinfection byproducts (DBPs) weighted by their Chinese hamster ovary (CHO) cell cytotoxicity LC50 values to estimate the DBP-associated cytotoxicity of disinfected waters. This approach assumed that cytotoxicity was additive rather than synergistic or antagonistic. In this study, we evaluated whether this assumption was valid for mixtures containing DBPs at the concentration ratios measured in authentic disinfected waters. We examined the CHO cell cytotoxicity of defined DBP mixtures based on the concentrations of 43 regulated and unregulated DBPs measured in eight drinking and potable reuse waters. The hypothesis for additivity was supported using three experimental approaches. First, we demonstrated that the calculated additive toxicity (CAT) and bioassay-based calculated additive toxicity (BCAT) of the DBP mixtures agree within 12% on a median basis. We also found an additive toxicity response (CAT ≈ BCAT) between the regulated and unregulated DBP classes. Finally, the empirical biological cytotoxicity of the DBP subset mixtures, independent of the calculated toxicity, was additive. These results support the validity of using the sum of cytotoxic potency-weighted DBP concentrations as an estimate of the CHO cell cytotoxicity associated with known DBPs in real disinfected waters.

This study investigated the speciation of halogen-specific total organic halogen and disinfection byproducts (DBPs) upon chlorination of natural organic matter (NOM) in the presence of iopamidol and bromide (Br−). Experiments were conducted with low bromide source waters with different NOM characteristics from Northeast Ohio, USA and varied spiked levels of bromide (2–30 μmol/L) and iopamidol (1–5 μmol/L). Iopamidol was found to be a direct precursor to trihalomethane (THM) and haloacetic acid formation, and in the presence of Br− favored brominated analogs. The concentration and speciation of DBPs formed were impacted by iopamidol and bromide concentrations, as well as the presence of NOM. As iopamidol increased the concentration of iodinated DBPs (iodo-DBPs) and THMs increased. However, as Br− concentrations increased, the concentrations of non-brominated iodo- and chloro-DBPs decreased while brominated-DBPs increased. Regardless of the concentration of either iopamidol or bromide, bromochloroiodomethane (CHBrClI) was the most predominant iodo-DBP formed except at the lowest bromide concentration studied. At relevant concentrations of iopamidol (1 μmol/L) and bromide (2 μmol/L), significant quantities of highly toxic iodinated and brominated DBPs were formed. However, the rapid oxidation and incorporation of bromide appear to inhibit iodo-DBP formation under conditions relevant to drinking water treatment.

Recycling wastewater from animal production for fertilizers using anaerobic digestion (AD) is a common method to recover the nutrients in the digestate. However, the digestate toxicity is not well understood because AD is mainly designed for chemical oxygen demand reduction. This study determined the toxicity during AD and the controlling factors with the goal to improve digestate safety during farmer handling to reuse the nutrients. Thermophilic and mesophilic AD of two swine wastewater sources were studied. Mammalian cell cytotoxicity revealed that the effluent after thermophilic digestion was at least 69% more toxic than the mesophilic effluent, owing to higher ammonia and total organic carbon in the former. Ammonia accounted for55% total cytotoxicity, and the organics of the thermophilic digestate were twice more toxic than those in the mesophilic digestate. Despite less toxicity contribution than the ammonia, the organics did demonstrate significant adverse effects on the thiol-mediated cellular protection mechanism. For swine wastewater nutrient recovery, converting ammonia to less toxic nitrogen forms could lower the toxic hazard of the AD digestate. With much less ammonia, the organics would be the remaining decisive factor for toxicity, which is favorably reduced using thermophilic AD over mesophilic. If the ammonia is not reduced, mesophilic AD would generate a less toxic digestate.

The reactions between disinfectants and organic matter or inorganic matter in source water generates disinfection by-products (DBPs) such as iodoacetic acid (IAA). DBPs are associated with health effects such as bladder cancer and adverse reproductive outcomes, but the effects of IAA on the ovary are not well known. This study determined whether IAA exposure affects ovarian follicle growth, steroidogenesis, and expression of apoptotic factors, cell cycle regulators, estrogen receptors, and steroidogenic factors in vitro. IAA exposure significantly decreased follicle growth, expression of cell cycle stimulators, and the proliferation marker Ki67. In contrast, IAA increased expression of the cell cycle inhibitor Cdkn1a. Moreover, IAA exposure increased expression of pro-apoptotic factors, whereas it decreased expression of anti-apoptotic factors. IAA exposure also altered expression of steroidogenic factors and estrogen receptors, disrupting steroidogenesis. These data demonstrate that IAA exposure inhibits follicle growth, decreases cell proliferation, and alters steroidogenesis in mouse ovarian follicles in vitro.

The increasing occurrence of harmful algal blooms in surface waters may increase the input of algal organic matter (AOM) to the dissolved organic matter pool. The formation of iodinated trihalomethanes (I-THMs) and noniodinated disinfection byproducts (DBPs) in synthetic waters containing AOM extracted from Microcystis aeruginosa was investigated in chloramination (preformed and in-situ formed chloramine, NH2Cl and Cl2-NH2Cl, respectively) and chlorination (Cl2) processes. AOM is much more favorable for iodine incorporation than natural organic matter (NOM). For example, the formation of I-THM from AOM is much higher than NOM isolate extracted from treated water (e.g., 3.5 times higher in the NH2Cl process), and thus higher iodine utilization and substitution factors from AOM were observed. Short contact time (2 min) chlorination in Cl2-NH2Cl process leading to the formation of halogenated intermediates favored I-THM formation, compared with NH2Cl process. However, further increasing chlorine contact time from 5 min to 24 h facilitated the conversion from iodide to iodate and thus I-THM formation decreased. Meanwhile, the formation of noniodinated THM4, haloacetonitriles (HANs), and haloacetaldehydes (HALs) increased. Factors including concentrations of AOM and bromide, pH, and chlorine/nitrogen ratios influenced the formation of I-THMs and noniodinated DBPs. To evaluate the benefit of mitigating I-THM formation over the risk of noniodinated DBP formation, measured DBPs were weighed against their mammalian cell toxicity indexes. Increasing the chlorine exposure increased the calculated cytotoxicity based on concentrations of measured I-THMs and noniodinated DBPs since unregulated HANs and HALs were the controlling agents.

Chloramines, in practice, are formed onsite by adding ammonia to chlorinated drinking water to achieve the required disinfection. While regulated disinfection byproducts (DBPs) are reduced during chloramine disinfection, other DBPs such as iodinated (iodo-) DBPs, that elicit greater toxicity are formed. The objective of this study was to investigate the impact of prechlorination time on the formation of both halogen-specific total organic halogen (TOX) and iodo/chlorinated (chloro-) DBPs during prechlorination/chloramination in source waters (SWs) containing iopamidol, an X-ray contrast medium. Barberton SW (BSW) and Cleveland SW (CSW) containing iopamidol were prechlorinated for 5–60 min and afterwards chloraminated for 72 hr with ammonium chloride. Chlorine contact time (CCT) did not significantly impact total organic iodine (TOI) concentrations after prechlorination or chloramination. Concentrations of total organic chlorine (TOCl) formed during prechlorination did not significantly change regardless of pH and prechlorination time, while TOCl appeared to decrease after 72 hr chloramination period. Dichloroiodomethane (CHCl2I) formation during prechlorination did not exhibit any significant trends as a function of pH or CCT, but after chloramination, significant increases were observed at pHs 6.5 and 7.5 with respect to CCT. Iodo-HAAs were not formed during prechlorination but were detected after chloramination. Significant quantities of chloroform (CHCl3) and trichloroacetic acid (TCAA) were formed during prechlorination but formation ceased upon ammonia addition. Therefore, prechlorination studies should measure TOX and DBP concentrations prior to ammonia addition to obtain data regarding the initial conditions.

Recent urban public water supply contamination events emphasize the importance of screening treated drinking water quality after distribution. In vitro bioassays, when run concurrently with analytical chemistry methods, are effective tools to evaluating the efficacy of water treatment processes and water quality. We tested 49 water samples representing the Chicago Department of Water Management service areas for estrogen, (anti)androgen, glucocorticoid receptor-activating contaminants and cytotoxicity. We present a tiered screening approach suitable to samples with anticipated low-level activity and initially tested all extracts for statistically identifiable endocrine activity; performing a secondary dilution-response analysis to determine sample EC50 and biological equivalency values (BioEq). Estrogenic activity was detected in untreated Lake Michigan intake water samples using mammalian (5/49; median: 0.21 ng E2Eq/L) and yeast cell (5/49; 1.78 ng E2Eq/L) bioassays. A highly sensitive (anti)androgenic activity bioassay was applied for the first time to water quality screening and androgenic activity was detected in untreated intake and treated pre-distribution samples (4/49; 0.93 ng DHTEq/L). No activity was identified above method detection limits in the yeast androgenic, mammalian anti-androgenic, and both glucocorticoid bioassays. Known estrogen receptor agonists were detected using HPLC/MS-MS (estrone: 0.72–1.4 ng/L; 17α-estradiol: 1.3–1.5 ng/L; 17β-estradiol: 1.4 ng/L; equol: 8.8 ng/L), however occurrence did not correlate with estrogenic bioassay results. Many studies have applied bioassays to water quality monitoring using only relatively small samples sets often collected from surface and/or wastewater effluent. However, to realistically adapt these tools to treated water quality monitoring, water quality managers must have the capacity to screen potentially hundreds of samples in short timeframes. Therefore, we provided a tiered screening model that increased sample screening speed, without sacrificing statistical stringency, and detected estrogenic and androgenic activity only in pre-distribution Chicago area samples.

Introduction of oil and gas extraction wastewaters (OGWs) to surface water leads to elevated halide levels from geogenic bromide and iodide, as well as enhanced formation of brominated and iodinated disinfection byproducts (DBPs) when treated. OGWs contain high levels of chemical additives used to optimize extraction activities, such as surfactants, which have the potential to serve as organic DBP precursors in OGW-impacted water sources. We report the first identification of olefin sulfonate surfactant-derived DBPs from laboratory-disinfected gas extraction wastewater. Over 300 sulfur-containing DBPs, with 43 unique molecular formulas, were found by high-resolution mass spectrometry, following bench-scale chlor(am)ination. DBPs consisted of mostly brominated species, including bromohydrin sulfonates, dihalo-bromosulfonates, and bromosultone sulfonates, with chlorinated/iodinated analogues formed to a lesser extent. Disinfection of a commercial C(12)-olefin sulfonate surfactant mixture revealed dodecene sulfonate as a likely precursor for most detected DBPs; disulfur-containing DBPs, like bromosultone sulfonate and bromohydrin disulfonate, originated from olefin disulfonate species, present as side-products of olefin sulfonate production. Disinfection of wastewaters increased mammalian cytotoxicity several orders of magnitude, with chloraminated water being more toxic. This finding is important to OGW-impacted source waters because drinking water plants with high-bromide source waters may switch to chloramination to meet DBP regulations.

The haloacetonitriles (HANs) is an emerging class of nitrogenous-disinfection byproducts (N-DBPs) present in disinfected drinking, recycled, processed wastewaters, and reuse waters. HANs were identified as primary forcing agents that accounted for DBP-associated toxicity. We evaluated the toxic characteristics of iodoacetonitrile (IAN), bromoacetonitrile (BAN), dibromoacetonitrile (DBAN), bromochloroacetonitrile (BCAN), tribromoacetonitrile (TBAN), chloroacetonitrile (CAN), dichloroacetonitrile (DCAN), trichloroacetonitrile (TCAN), bromodichloroacetonitrile (BDCAN), and chlorodibromoacetonitrile (CDBAN). This research generated the first quantitative, comparative analyses on the mammalian cell cytotoxicity, genotoxicity and thiol reactivity of these HANs. The descending rank order for HAN cytotoxicity was TBAN ≈ DBAN > BAN ≈ IAN > BCAN ≈ CDBAN > BDCAN > DCAN ≈ CAN ≈ TCAN. The rank order for genotoxicity was IAN ≈ TBAN ≈ DBAN > BAN > CDBAN ≈ BDCAN ≈ BCAN ≈ CAN ≈ TCAN ≈ DCAN. The rank order for thiol reactivity was TBAN > BDCAN ≈ CDBAN > DBAN > BCAN > BAN ≈ IAN > TCAN. These toxicity metrics were associated with membrane permeability and chemical reactivity. Based on their physiochemical parameters and toxicity metrics, we developed optimized, robust quantitative structure activity relationship (QSAR) models for cytotoxicity and for genotoxicity. These models can predict cytotoxicity and genotoxicity of novel HANs prior to analytical biological evaluation.

Seasonal algal blooms in surface waters can impact water quality through an input of algal organic matter (AOM) to the pool of dissolved organic matter as well as the release of cyanotoxins. The formation and speciation of disinfection byproducts (DBPs) during chlorination of algal-impacted waters, collected from growth of Microcystis aeruginosa were studied. Second-order rate constants for the reactions of microcystins (MCs) with chlorine and bromine were determined. Finally, the toxicity of chlorinated algal-impacted waters was evaluated by Chinese hamster ovary (CHO) cytotoxicity and genotoxicity assays. Under practical water treatment conditions, algal-impacted waters produced less regulated trihalomethanes (THMs) and haloacetic acids (HAAs), haloacetonitriles (HANs), and total organic halogen (TOX) than natural organic matter (NOM). For example, the weight ratios of DBP formation from AOM to NOM (median levels) were approximately 1:5, 1:3, 1:2 and 1:3 for THMs, HAAs, HANs, and TOX, respectively. Increasing initial bromide level significantly enhanced THM and HAN concentrations, and therefore unknown TOX decreased. The second-order rate constant for the reactions of MC-LR (the most common MC species) with chlorine was 60 M−1 s−1 at pH 7.5 and 21 °C, and the rate constants for MC congeners follow the order: MC-WR > MC-LW > MC-YR > MC-LY > MC-LR ≈ MC-RR. The reaction rate constant of bromine with MC-LR is two orders of magnitude higher than that of chlorine. Unchlorinated algal-impacted waters were toxic owning to the presence of MCs, and chlorination enhanced their cytotoxicity and genotoxicity due to the formation of toxic halogenated DBPs. However, the toxicity of treated waters depended on the evolution of cyanotoxins and formation of DBPs (particularly unknown or emerging DBPs).

In drinking water disinfection, switching from free chlorine to alternative chemical disinfectants such as monochloramine may result in the formation of different classes of toxic disinfection byproducts (DBPs). Haloacetonitriles (HANs) and haloacetamides (HAMs) are two currently unregulated nitrogen-containing DBP (N-DBP) groups commonly found in water disinfected with monochloramine that have been shown to be more cyto- and genotoxic than regulated DBPs. Following the Food Safety Modernization Act of 2011 in the U.S., guidelines for disinfection washes in food packaging facilities are under consideration to control pathogen risks. However, disinfectant exposures may need optimization because the high concentrations of chlorine disinfectant promote the formation of high levels of disinfection byproducts (DBPs). When chlorine doses up through the 200 mg/L as Cl2 range relevant to the current practice were applied to spinach and lettuce, significant DBP formation was observed, even within 5 min at 7 °C. Concentrations of volatile chlorinated DBPs in washwater were far higher than typically observed in disinfected drinking water (e.g., 350 μg/L 1,1-dichloropropanone). However, these DBPs partitioned to the aqueous phase and so represent a greater concern for the disposal or reuse of washwater than for consumer exposure via food. An in chemico high throughput assay based on N-acetylcysteine was developed and used in conjunction with previous and new mammalian cell cytotoxicity data. Our objective was to derive an empirical equation with confidence levels for mammalian cell cytotoxicity prediction. Modeling data included 16 unique sources of waters and wastewaters of distinct water qualities to encompass a wide range of real environmental samples. This approach provides a quick screen to identify those water and wastewaters that could be prioritized for in depth analytical biological analyses and toxicity. The resulting model can serve as a preliminary convenient tool to screen for potential mammalian cell cytotoxicity in organic extracts of a wide variety of water samples.

Iopamidol is a known direct precursor to iodinated and chlorinated DBP formation; however, the influence of iopamidol on both iodo/chloro-DBP formation has yet to be fully investigated. This study investigated the effect of iopamidol on the formation and speciation of halogen-specific total organic halogen (TOX), as well as iodo/chloro-DBPs, in the presence of 3 source waters (SWs) from Northeast Ohio and chlorinated oxidants. Chlorination and chloramination of SWs were carried out at pH 6.5-9.0 and, different iopamidol and dissolved organic carbon (DOC) concentrations. Total organic iodine (TOI) loss was approximately equal (22-35%) regardless of SW. Total organic chlorine (TOCl) increased in all SWs and was substantially higher in the higher SUVA254 SWs. Iopamidol was a direct precursor to chloroform (CHCl3), trichloroacetic acid (TCAA), and dichloroiodomethane (CHCl2I) formation. While CHCl3 and TCAA exhibited different formation trends with varying iopamidol concentrations, CHCl2I increased with increasing iopamidol and DOC concentrations. Low concentrations of iodo-acids were detected without discernible trends. Total trihalomethanes (THMs), total haloacetic acids (HAAs), TOCl, and unknown TOCl (UTOCl) were correlated with fluorescence regional volumes and SUVA254. The yields of all these species showed a strong positive correlation with fulvic, humic, and combined humic and fulvic regions, as well as SUVA254. Iopamidol was then compared to the 3 SWs with respect to DBP yield. Although the SUVA254 of iopamidol was relatively high, it did not produce high yields of THMs and HAAs compared to the 3 SWs. However, chlorination of iopamidol did result in high yields of TOCl and UTOCl.

Genotoxicity is considered a major concern for drinking water disinfection byproducts (DBPs). Of over 700 DBPs identified to date, only a small number has been assessed with limited information for DBP genotoxicity mechanism(s). In this study, we evaluated genotoxicity of 20 regulated and unregulated DBPs applying a quantitative toxicogenomics approach. We used GFP-fused yeast strains that examine protein expression profiling of 38 proteins indicative of all known DNA damage and repair pathways. The toxicogenomics assay detected genotoxicity potential of these DBPs that is consistent with conventional genotoxicity assays end points. Furthermore, the high-resolution, real-time pathway activation and protein expression profiling, in combination with clustering analysis, revealed molecular level details in the genotoxicity mechanisms among different DBPs and enabled classification of DBPs based on their distinct DNA damage effects and repair mechanisms. Oxidative DNA damage and base alkylation were confirmed to be the main molecular mechanisms of DBP genotoxicity. Initial exploration of QSAR modeling using moleular genotoxicity end points (PELI) suggested that genotoxicity of DBPs in this study was correlated with topological and quantum chemical descriptors. This study presents a toxicogenomics-based assay for fast and efficient mechanistic genotoxicity screening and assessment of a large number of DBPs. The results help to fill in the knowledge gap in the understanding of the molecular mechanisms of DBP genotoxicity.

We assessed mammalian cell cytotoxicity of ambient PM2.5 and investigated its association with the oxidative potential (OP) and chemical composition of the particles. Sixteen PM samples spanning in various seasons (fall, winter, spring and summer) were collected from an urban site in central Illinois. Cytotoxicity (LC50) in terms of the volume of air that kills 50% of the cells were calculated, which varied from 4.3 to 7.2 m3 of air. The OP was measured by two assays – the dithiothreitol (DTT) and the surrogate lung fluid (SLF) assay. In DTT assay, we measured two endpoints – hydroxyl radicals (•OH) generation and DTT consumption (the conventionally measured endpoint), while only •OH generation was measured in the SLF assay. Although, all three endpoints in the OP assays correlated significantly (P ≤ 0.05) with LC50, the correlation of reactive oxygen species (ROS) generation in DTT and SLF assays was much higher (r > 0.80 for •OH generation versus LC50) than the DTT consumption (r = 0.58). To further understand the components in PM that drive cytotoxicity and OP, concentration of water-soluble metals (Fe, Cu, Co, Cr, Mn, Ni, Pb, V, Hg, and Zn), organic carbon (OC), water soluble organic carbon (WSOC), and elemental carbon (EC) were measured. Among all the chemical components, Fe, Cu and WSOC correlated most (r > 0.70; P ≤ 0.01) with the cytotoxicity. DTT consumption correlated only with OC and WSOC (r > 0.80; P ≤ 0.01), while •OH generation in DTT and SLF assay correlated with both WSOC (r > 0.70; P ≤ 0.01) and metals (i.e. Fe and Cu; r > 0.75; P ≤ 0.01). Our results suggest a strong link between the PM2.5 OP and its cytotoxicity. Furthermore, the synergistic interactions among the organic compounds (i.e. WSOC) and metals (Fe and Cu) to enhance the ROS generation, which are more effectively captured in •OH generation endpoints in DTT and SLF assay than the DTT consumption, appear to be largely responsible for the observed mammalian cell cytotoxicity of PM2.5.

The disinfection of drinking water has been a major public health achievement. However, haloacetic acids (HAAs), generated as byproducts of water disinfection, are cytotoxic, genotoxic, mutagenic, carcinogenic, and teratogenic. Previous studies of monoHAA-induced genotoxicity and cell stress demonstrated that the toxicity was due to inhibition of glyceraldehyde-3-phosphate dehydrogenase (GAPDH), leading to disruption of cellular metabolism and energy homeostasis. DiHAAs and triHAAs are also produced during water disinfection, and whether they share mechanisms of action with monoHAAs is unknown. In this study, we evaluated the effects of mono-, di-, and tri-HAAs on cellular GAPDH enzyme kinetics, cellular ATP levels, and pyruvate dehydrogenase complex (PDC) activity. Here, treatments conducted in Chinese hamster ovary (CHO) cells revealed differences among mono-, di-, and triHAAs in their molecular targets. The monoHAAs, iodoacetic acid and bromoacetic acid, were the strongest inhibitors of GAPDH and greatly reduced cellular ATP levels. Chloroacetic acid, diHAAs, and triHAAs were weaker inhibitors of GAPDH and some increased the levels of cellular ATP. HAAs also affected PDC activity, with most HAAs activating PDC. The primary finding of this work is that mono- versus multi-HAAs address different molecular targets, and the results are generally consistent with a model in which monoHAAs activate the PDC through GAPDH inhibition-mediated disruption in cellular metabolites, including altering ATP-to-ADP and NADH-to-NAD ratios. The monoHAA-mediated reduction in cellular metabolites results in accelerated PDC activity by way of metabolite-ratio-dependent PDC regulation. DiHAAs and triHAAs are weaker inhibitors of GAPDH, but many also increase cellular ATP levels, and we suggest that they increase PDC activity by inhibiting pyruvate dehydrogenase kinase.

Hydraulic fracturing wastewaters discharged to surface water have led to elevated bromide and iodide levels, as well as enhanced formation of brominated trihalomethanes, haloacetic acids, haloacetonitriles, and iodo-trihalomethanes at downstream drinking water treatment plants, in chlorinated effluent from wastewater treatment plants, and in controlled laboratory studies. This enhanced formation of brominated and iodinated disinfection byproducts (DBPs) raises concerns regarding human health, because they are much more toxic than chlorinated DBPs. This study represents the first nontarget, comprehensive analysis of iodinated DBPs formed in chloraminated produced waters associated with hydraulic fracturing of shale and conventional gas formations. Haloacetamides (HAMs) are cytotoxic, genotoxic, and mutagenic byproducts of drinking water disinfection. They are soft electrophilic compounds that form covalent bonds with the free thiol/thiolate in cysteine residues through an SN2 reaction mechanism. Toxicity of the monohalogenated HAMs (iodoacetamide, IAM; bromoacetamide, BAM; or chloroacetamide, CAM) varied depending on the halogen substituent. The aim of this research was to investigate how the halogen atom affects the reactivity and toxicological properties of HAMs, measured as induction of oxidative/electrophilic stress response and genotoxicity. Additionally, we wanted to determine how well in silico estimates of electrophilic softness matched thiol/thiolate reactivity and in vitro toxicological endpoints. Each of the HAMs significantly induced nuclear Rad 51 accumulation and ARE signaling activity compared to a negative control. In each case the rank order of effect was IAM > BAM > CAM for Rad 51, and BAM ≈ IAM > CAM for ARE. In general, electrophilic softness and in chemico thiol/thiolate reactivity provided a qualitative indicator of toxicity, as the softer electrophiles IAM and BAM were more thiol/thiolate reactive and were more toxic.

The presence of iodinated X-ray contrast media (ICM) in source waters is of high concern to public health because of their potential to generate highly toxic disinfection by-products (DBPs). The objective of this study was to determine the impact of ICM in source waters and the type of disinfectant on the overall toxicity of DBP mixtures and to determine which ICM and reaction conditions give rise to toxic by-products. Source waters collected from Akron, OH were treated with five different ICMs, including iopamidol, iopromide, iohexol, diatrizoate and iomeprol, with or without chlorine or chloramine disinfection. The reaction product mixtures were concentrated with XAD resins and the mammalian cell cytotoxicity and genotoxicity of the reaction mixture concentrates was measured. Water containing iopamidol generated an enhanced level of mammalian cell cytotoxicity and genotoxicity after disinfection. While chlorine disinfection with iopamidol resulted in the highest cytotoxicity overall, the relative iopamidol-mediated increase in toxicity was greater when chloramine was used as the disinfectant compared with chlorine. Four other ICMs (iopromide, iohexol, diatrizoate, and iomeprol) expressed some cytotoxicity over the control without any disinfection, and induced higher cytotoxicity when chlorinated. Only iohexol enhanced genotoxicity compared to the chlorinated source water.

Water reuse is receiving unprecedented attention as many areas around the globe attempt to better-manage their fresh water resources. Wastewaters in coastal regions may contain elevated levels of bromide (Br–) and iodide (I–) from seawater intrusion or high mineral content in the source waters. Disinfection of such wastewater is essential to prevent the spread of pathogens; however, little is known about the toxicity of the treated wastewater. In this study, we evaluated the genotoxicity to Chinese hamster ovary (CHO) cells induced by municipal secondary wastewater effluent amended with elevated Br– and I– after disinfection by chlorine, chloramines, or ozone. The formation of iodinated disinfection byproducts (I-DBPs) in drinking waters is of a concern due to their higher cyto- and genotoxicity than their chlorinated and brominated analogues. This study investigated the formation of I-DBPs under chloramination conditions using preformed chloramine and associated cyto- and geno-toxicities obtained with Chinese Hamster Ovary (CHO) cell assay. Cyto- and geno-toxicity of the samples were also calculated using DBP toxicity index values and correlated with total organic halide (TOX) formation. In low iodide (I-) (0.32 μM, 40 μg L−1) water, increasing dissolved organic carbon (DOC) concentration of selected waters from 0.1 to 0.25 mg L−1 increased the formation of iodinated trihalomethanes (I-THMs), while further increases from 0.25 to 4 mg L−1 produced an opposite trend. In high iodide water (3.2 μM, 400 μg L−1), increasing DOC from 0.5 to 4 mg L−1 gradually increased the I-THM formation, while a decrease was observed at 5.4 mg L−1 DOC. Iodoform was the most influenced species from the changes in DOC concentration. While increasing the initial iodide concentration from 0 to 5 μM increased the formation of iodoform, it did not make any considerable impact on the formation of other I-THMs. The measured cytotoxicity of samples was significantly correlated with increasing DOC concentration. Unknown TOCl and TOI showed a high correlation with measured cytotoxicity, while calculated total organic chlorine (TOCl) and total organic iodine (TOI) did not correlate. The comparison of measured and calculated cytotoxicity values showed that the calculated values do not always represent the overall cytotoxicity, since the formation of unknown DBPs are not taken into consideration during the toxicity calculations.

The process of natural transformation allows for the stable uptake, integration, and functional expression of extracellular DNA. This mechanism of horizontal gene transfer has been widely linked to the acquisition of antibiotic resistance and virulence factors. Here, we demonstrate that bromoacetic acid (BAA)-a regulated drinking water disinfection byproduct (DBP)-can stimulate natural transformation rates in the model organism Acinetobacter baylyi ADP1. We demonstrate that transformation stimulation in response to BAA is concentration-dependent and is linked to the ability of this compound to generate DNA damage via oxidative stress. In presence of BAA, transcription of recA was upregulated 20-40% compared to the nontreated controls, indicating that this component of the DNA damage response could be associated with the increase in transformation. Other genes associated with DNA translocation across the cytoplasmic membrane (i.e., pilX, comA) did not exhibit increased transcription in the presence of BAA, indicating that the enhancement of transformation is not associated with increased translocation rates of environmental DNA. Overall, these results lead us to speculate that elevated recA transcription levels could lead to increased integration rates of foreign DNA within the recipient cell during DNA repair. Lastly, we show that an artificial DBP cocktail simulating the environmental concentrations of five water DBP classes stimulates natural transformation by almost 2-fold. The results of this study suggest that mutagens like DBPs may play an important role in enhancing the fixation rates of extracellular DNA in the environmental metagenome.

Reusing wastewater in agriculture is becoming increasingly common, which necessitates disinfection to ensure reuse safety. However, disinfectants can react with wastewater constituents to form disinfection byproducts (DBPs), many of which are toxic and restrict the goal of safe reuse. Our objective was to benchmark the induction of mammalian cell cytotoxicity after ozonation against chlorination for three types of real wastewaters: municipal secondary effluent and two sources of minimally treated swine farm wastewaters. A new method to evaluate samples of suspected high cytotoxicity was devised. For the secondary effluent, ozonation reduced the cytotoxicity by as much as 10 times; chlorination lowered the cytotoxicity only when followed by dechlorination. The swine farm wastewaters were up to 2000 times more cytotoxic than the secondary effluent, and the highest reduction in cytotoxicity was 17 times as achieved by ozonation. These results indicate that secondary effluent is preferred over swine wastewaters for agricultural reuse regardless of the tested disinfectants. Ozonation consistently reduced the cytotoxicity of both the full strength and the organic extracts of all tested wastewaters more than chlorination. The only significant correlation was observed in the secondary wastewater between total haloacetonitriles and cytotoxicity. While the association of reduced toxicity with the modification or reduction of specific compound(s) is unclear, regulated DBPs may not be the primary forcing agents.

Water disinfection greatly reduced the incidence of waterborne diseases, but the reaction between disinfectants and natural organic matter in water leads to the formation of drinking water disinfection by-products (DBPs). DBPs have been shown to be toxic, but their effects on the ovary are not well defined. This study tested the hypothesis that monohalogenated DBPs (chloroacetic acid, CAA; bromoacetic acid, BAA; iodoacetic acid, IAA) inhibit antral follicle growth and steroidogenesis in mouse ovarian follicles. Antral follicles were isolated and cultured with either vehicle or DBPs (0.25–1.00 mM of CAA; 2–15 µM of BAA or IAA) for 48 and 96 h. Follicle growth was measured every 24 h and the media were analyzed for estradiol levels at 96 h. Exposure to DBPs significantly inhibited antral follicle growth and reduced estradiol levels compared to controls. These data demonstrate that DBP exposure caused ovarian toxicity in vitro.

When the X-ray contrast pharmaceutical iopamidol was chlorinated in natural source water, a toxic mixture of many disinfection byproducts (DBPs) was formed. We isolated and identified five high-molecular weight iopamidol chlorination DBPs and analyzed their cytotoxicity and genotoxicity using mammalian cells. Of the five DBPs, four exhibited low levels of cytotoxicity, while none of the iopamidol DBPs was genotoxic. These data support the hypothesis that these high-molecular weight DBPs were not the forcing agents that induced the cytotoxicity and genotoxicity observed after iopamidol was disinfected in natural source waters.

Since the first regulation of disinfection by-products (DBPs) in the 1970s, >700 DBPs have been identified, and many of these are much more toxic than those regulated. Moreover, drinking water today is not the same as it was in the past, with increasing use of alternative disinfectants like chloramine, ozone, chlorine dioxide, and UV (which can form other types of DBPs), as well as new impacts on our source waters from climate change, population increases, wastewater intrusion, and energy exploration. The question today is whether we are regulating the right DBPs to protect human health, and if not, what should be done. New approaches may involve (1) the use of in vitro data and a Precautionary Principle approach, (2) using surrogate metrics of finished waters, such as total organic bromine/iodine, total nitrosamines, or total organic nitrogen rather than creating longer lists of regulated DBPs, and (3) using toxicity assays for whole drinking water extracts to pinpoint potential problems, then using chemical analyses to identify the toxic agents, and finally, (4) invoking different treatment strategies to reduce the toxicity. While the early regulations likely significantly improved the safety of drinking water, DBP exposure is a constant in modern life, and there is more that we can do.

An integrated manure treatment system, including a mixed algal-bacterial bioreactor (MABB) and hydrothermal processing of biomass solids, was found to remove 76.4–97.0% of the total estrogenic hormones (estrone (E1), 17β-estradiol (E2), and estriol (E3)) from the liquid portion of animal manure (LPAM). The mixed biomass was converted into either biocrude oil with a yield up to 40% via hydrothermal liquefaction (HTL) or syngas with a yield up to 54% yield via catalytic hydrothermal gasification (CHG). Adding granular activated carbon (GAC) in the MABB enhanced the removal of estrogenic hormones (+ 7.2%), cytotoxicity (+ 58%), and heavy metals (+ 10.5%). After the integrated system with the MABB, HTL, and CHG processes, the overall percent removal of heavy metals from the LPAM ranged from 27.1 to 40.3%. The concentrations of potentially toxic heavy metals (lead (Pb), copper (Cu), zinc (Zn), cadmium (Cd), nickel (Ni), chromium (Cr)) in the aqueous phase after HTL and CHG tests ranged from 0.01 to 25.3 mg/L.

Toxicity arising from toxic disinfection byproducts is an unintended result of disinfection during water reclamation. To ensure safe water reclamation treatment, it is important to develop a disinfection strategy with minimal formation of overall toxicity in the reclaimed water. The cumulative disinfectant concentration over time (Ct) is a useful concept for pathogen control during reuse water disinfection. We evaluated the toxicity impact of Ct values and different methods to achieve identical Ct values by ozonation or chlorination of wastewaters from four agricultural sources on mammalian cells. N-acetylcysteine (NAC) reactivity of the wastewater organic extracts was determined to reveal their impact on the thiol-specific biological detoxification mechanism. The results demonstrated that for two sources and for both ozonation and chlorination, higher Ct values enhanced cytotoxicity. The ozonated waters were at least 10% less toxic and as much as 22.4 times less toxic than either the non-disinfected controls or the chlorinated waters. Chlorination consistently induced higher cytotoxicity than ozonation by between 2.2 and 22.4 fold, respectively, and induced similar or higher cytotoxicity than the non-disinfected controls, by at most 4.4 fold. Given the same Ct values, the combination of high disinfectant concentration and short contact time produced finished wastewaters with higher toxicity, than the combination of low disinfectant concentration and long contact time. NAC thiol reactivity was positively and significantly correlated with mammalian cell cytotoxicity, and agreed with 80% of the cytotoxicity rank order. This suggests that the induction of cytotoxicity involved reactions with agents that acted as thiol pool quenchers. The overall results indicate that the cytotoxicity of wastewaters may increase when higher Ct values are applied to inactivate recalcitrant pathogens. To counteract the potential increase in cytotoxicity at high Ct values, for both ozonation and chlorination, lower disinfectant dose and longer contact time may be adopted.

The process of disinfecting drinking water inadvertently leads to the formation of numerous disinfection byproducts (DBPs). Some of these are mutagenic, genotoxic, teratogenic, and cytotoxic, as well as potentially carcinogenic both in vivo and in vitro. We investigated the in vitro biological activity of five DBPs: three monohaloacetic acids (monoHAAs) [chloroacetic acid (CAA), bromoacetic acid (BAA), and iodoacetic acid (IAA)] and two novel halobenzoquinones (HBQs) [2,6-dichloro-p-benzoquinone (DCBQ) and 2,6-dibromo-p-benzoquinone]. We focused particularly on cytotoxicity and induction of two adaptive stress response pathways: the oxidative stress responsive Nrf2/ARE and DNA-damage responsive p53 pathways. All five DBPs were cytotoxic to the Caco-2 cell line after a 4 h exposure, and all DBPs induced both of the adaptive stress response pathways, Nrf2/ARE and p53, in the micromolar range, as measured by two β-lactamase-based reporter gene assays. The decreasing order of potency for all three endpoints for the five DBPs was IAA ∼ BAA > DCBQ ∼ DBBQ > CAA. Induction of oxidative stress was previously proposed to be the molecular initiating event (MIE) for both classes of DBPs. However, comparing the levels of activation of the two pathways uncovered that the Nrf2/ARE pathway was the more sensitive endpoint for HAAs, whereas the p53 pathway was more sensitive in the case of HBQs. Therefore, the DNA damage-responsive p53 pathway may be an important piece of information to fill in a gap in the adverse outcome pathway framework for the assessment of HBQs. Finally, we cautiously compared the potential risk of the two novel HBQs using a benchmarking approach to that of the well-studied CAA, which suggested that their relative risk may be lower than that of BAA and IAA.

The introduction of drinking water disinfection greatly reduced waterborne diseases. However, the reaction between disinfectants and natural organic matter in the source water leads to an unintended consequence, the formation of drinking water disinfection byproducts (DBPs). The haloace-taldehydes (HALs) are the third largest group by weight of identified DBPs in drinking water. The primary objective of this study was to analyze the occurrence and comparative toxicity of the emerging HAL DBPs. A new HAL DBP, iodoacetaldehyde (IAL) was identified. This study provided the first systematic, quantitative comparison of HAL toxicity in Chinese hamster ovary cells. The rank order of HAL cytotoxicity is tribromoacetaldehyde (TBAL) ≈ chloroacetaldehyde (CAL) > dibromoacetaldehyde (DBAL) ≈ bromochloroacetaldehyde (BCAL) ≈ dibromochloroacetaldehyde (DBCAL) > IAL > bromoacetaldehyde (BAL) ≈ bromodichloroacetaldehyde (BDCAL) > dichloroacetaldehyde (DCAL) > trichloroacetaldehyde (TCAL). The HALs were highly cytotoxic compared to other DBP chemical classes. The rank order of HAL genotoxicity is DBAL > CAL ≈ DBCAL > TBAL ≈ BAL > BDCAL > BCAL ≈ DCAL > IAL. TCAL was not genotoxic. Because of their toxicity and abundance, further research is needed to investigate their mode of action to protect the public health and the environment.

We investigated chronic mammalian cell cytotoxicity of wastewaters from four sources and their optical spectroscopic properties with or without chlorination or ozonation. Samples from effluents of activated sludge, nitrification tower, facultative waste stabilization pond, and maturation waste stabilization pond were either chlorinated or ozonated. The wastewater samples were analyzed for fluorescence excitation emission matrix, specific fluorescence index (SFI), and specific UV absorbance at 254 nm (SUVA). Before and after disinfection the wastewater samples were quantitatively analyzed for in vitro mammalian cell cytotoxicity. We found that the organic extracts from the ozonated samples induced lower cytotoxicity responses than those from the chlorinated or the nondisinfected samples. To develop correlations between SFI, SUVA, and cytotoxicity, we analyzed 21 independent samples. Significant linear correlations found among these samples suggest that under the tested conditions, cytotoxicity was preferentially influenced by the fluorescence and SUVA of their composite organic agents. These two spectroscopic parameters may be used as indicators for the potential cytotoxicity of nondisinfected, ozonated, or chlorinated municipal wastewaters.

International audience; The reclamation and disinfection of waters impacted by human activities (e.g., wastewater effluent discharges) are of growing interest for various applications but has been associated with the formation of toxic nitrogenous disinfection byproducts (N-DBPs). Monochloramine used as an alternative disinfectant to chlorine can be an additional source of nitrogen in the formation of N-DBPs. Individual toxicity assays have been performed on many DBPs, but few studies have been conducted with complex mixtures such as wastewater effluents. In this work, we compared the cytotoxicity and genotoxicity of wastewater effluent organic matter (EfOM) before and after chloramination. The toxicity of chloraminated EfOM was significantly higher than the toxicity of raw EfOM, and the more hydrophobic fraction (HPO) isolated on XAD-8 resin was more toxic than the fraction isolated on XAD-4 resin. More DBPs were also isolated on the XAD-8 resin. N-DBPs (i.e., haloacetonitriles or haloacetamides) were responsible for the majority of the cytotoxicity estimated from DBP concentrations measured in the XAD-8 and XAD-4 fractions (99.4% and 78.5%, respectively). Measured DBPs accounted for minor proportions of total brominated and chlorinated products, which means that many unknown halogenated compounds were formed and can be responsible for a significant part of the toxicity. Other non-halogenated byproducts (e.g., nitrosamines) may contribute to the toxicity of chloraminated effluents as well.

Disinfectants inactivate pathogens in source water; however, they also react with organic matter and bromide/iodide to form disinfection byproducts (DBPs). Although only a few DBP classes have been systematically analyzed for toxicity, iodinated and brominated DBPs tend to be the most toxic. The objectives of this research were (1) to determine if monochloramine (NH2Cl) disinfection generated drinking water with less toxicity than water disinfected with free chlorine (HOCl) and (2) to determine the impact of added bromide and iodide in conjunction with HOCl or NH2Cl disinfection on mammalian cell cytotoxicity and genomic DNA damage induction. Water disinfected with chlorine was less cytotoxic but more genotoxic than water disinfected with chloramine. For both disinfectants, the addition of Br(-) and I(-) increased cytotoxicity and genotoxicity with a greater response observed with NH2Cl disinfection. Both cytotoxicity and genotoxicity were highly correlated with TOBr and TOI. However, toxicity was weakly and inversely correlated with TOCl. Thus, the forcing agents for cytotoxicity and genotoxicity were the generation of brominated and iodinated DBPs rather than the formation of chlorinated DBPs. Disinfection practices need careful consideration especially when using source waters containing elevated bromide and iodide.

Amine-based CO2 capture is a prime contender for the first full-scale implementation of CO2 capture at fossil fuel-fired power plants postcombustion. However, the formation of potentially carcinogenic N-nitrosamines and N-nitramines from reactions of flue gas NOx with the amines presents a potential risk for contaminating airsheds and drinking water supplies. Setting regulatory emission limits is hampered by the dearth of toxicity information for the N-nitramines. This study employed quantitative in vitro bioassays for mutagenicity in Salmonella typhimurium, and chronic cytotoxicity and acute genotoxicity in Chinese hamster ovary (CHO) cells to compare the toxicity of analogous N-nitrosamines and N-nitramines relevant to CO2 capture. Although the rank order was similar for genotoxicity in CHO cells and mutagenicity in S. typhimurium, the Salmonella assay was far more sensitive. In general, mutagenicity was higher with S9 hepatic microsomal activation. The rank order of mutagenicity was N-nitrosodimethylamine (NDMA)N-nitrosomorpholineN-nitrodimethylamine1,4-dinitrosopiperazineN-nitromorpholine1,4-dinitropiperazineN-nitromonoethanolamineN-nitrosodiethanolamineN-nitrodiethanolamine. 1-Nitrosopiperazine and 1-nitropiperazine were not mutagenic. Overall, N-nitrosamines were ∼15-fold more mutagenic than their N-nitramine analogues.

Combined chlorine is increasingly being used as an alternative disinfectant to free chlorine to maintain a residual in drinking water distribution systems mainly because it would reduce the formation of regulated disinfection byproducts (DBPs) trihalomethanes and haloacetic acids. However, the use of combined chlorine could promote the formation of currently unregulated nitrogenous DBPs (N-DBPs) such as haloacetonitriles and haloacetamides that are found to be more cyto- and genotoxic than regulated DBPs. Monochloramine quickly reacts with chloroacetaldehyde, a DBP formed during primary disinfection with free chlorine, forming and reaching pseudoequilibrium (equilibrium constant K1 = 1.87 × 10(3) M(-1)) with the carbinolamine 2-chloro-1-(chloroamino)ethanol. 2-Chloro-1-(chloroamino)ethanol undergoes slow dehydration to form the imine 1-chloro-2-(chloroimino)ethane that decomposes at a faster rate to chloroacetonitrile. 2-Chloro-1-(chloroamino)ethanol is also oxidized by monochloramine to produce the previously unreported DBP N,2-dichloroacetamide. The carbinolamine dehydration step was found to be acid/base catalyzed (k2(0) = 3.30 × 10(-6) s(-1), k2(H) = 2.43 M(-1) s(-1), k2(OH) = 3.90 M(-1) s(-1)). In contrast, N,2-dichloroacetamide formation was observed to be only base catalyzed (k3(OH) = 3.03 × 10(4) M(-2) s(-1)). N,2-dichloroacetamide cytotoxicity (LC50 = 2.56 × 10(-4) M) was found to be slightly lower compared to that reported for chloroacetamide but higher than those of di- and trichloroacetamide.

Monohaloacetic acids (monoHAAs) are a major class of drinking water disinfection by-products (DBPs) and are cytotoxic, genotoxic, mutagenic, and teratogenic. We propose a model of toxic action based on monoHAA-mediated inhibition of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) as a target cytosolic enzyme. This model predicts that GAPDH inhibition by the monoHAAs will lead to a severe reduction of cellular ATP levels and repress the generation of pyruvate. A loss of pyruvate will lead to mitochondrial stress and genomic DNA damage. We found a concentration-dependent reduction of ATP in Chinese hamster ovary cells after monoHAA treatment. ATP reduction per pmol monoHAA followed the pattern of iodoacetic acid (IAA) > bromoacetic acid (BAA) >> chloroacetic acid (CAA), which is the pattern of potency observed with many toxicological endpoints. Exogenous supplementation with pyruvate enhanced ATP levels and attenuated monoHAA-induced genomic DNA damage as measured with single cell gel electrophoresis. These data were highly correlated with the SN 2 alkylating potentials of the monoHAAs and with the induction of toxicity. The results from this study strongly support the hypothesis that GAPDH inhibition and the possible subsequent generation of reactive oxygen species is linked with the cytotoxicity, genotoxicity, teratogenicity, and neurotoxicity of these DBPs.

Hydrothermal liquefaction (HTL) is an attractive method for converting wet biomass into petroleum-like biocrude oil that can be refined to make petroleum products. This approach is advantageous for conversion of low-lipid algae, which are promising feedstocks for sustainable large-scale biofuel production. As with natural petroleum formation, the water in contact with the produced oil contains toxic compounds. The objectives of this research were to: (1) identify nitrogenous organic compounds (NOCs) in wastewater from HTL conversion of Spirulina; (2) characterize mammalian cell cytotoxicity of specific NOCs, NOC mixture, and the complete HTL wastewater (HTL-WW) matrix; and (3) investigate mitigation measures to reduce toxicity in HTL-WW. Liquid-liquid extraction and nitrogen-phosphorus detection was used in conjunction with gas chromatography-mass spectrometry (GC-MS), which detected hundreds of NOCs in HTL-WW. Reference materials for nine of the most prevalent NOCs were used to identify and quantify their concentrations in HTL-WW. Mammalian cell cytotoxicity of the nine NOCs was quantified using a Chinese hamster ovary (CHO) cell assay, and the descending rank order for cytotoxicity was 3-dimethylamino-phenol2,2,6,6-tetramethyl-4-piperidone2,6-dimethyl-3-pyridinol2-picolinepyridine1-methyl-2-pyrrolidinoneσ-valerolactam2-pyrrolidinoneε-caprolactam. The organic mixture extracted from HTL-WW expressed potent CHO cell cytotoxic activity, with a LC(50) at 7.5% of HTL-WW. Although the toxicity of HTL-WW was substantial, 30% of the toxicity was removed biologically by recycling HTL-WW back into algal cultivation. The remaining toxicity of HTL-WW was mostly eliminated by subsequent treatment with granular activated carbon.

Disinfection by-products occur widely as the unintended effect of water disinfection and are associated with toxicity and adverse human health effects. Yet the molecular mechanisms of their toxicity are not well understood. To investigate the molecular basis of hyperploidy induction by monohaloacetonitriles, the interaction of monohaloacetonitriles with topoisomerase II in Chinese hamster ovary cells was examined. We showed a concentration-dependent inhibition of DNA decatenation activity of topoisomerase under acellular conditions while in vitro monohaloacetonitrile treatment expressed mixed results. The working hypothesis, that topoisomerase II is a molecular target of monohaloacetonitriles, was only partially supported. Nevertheless, this research serves as a starting point toward molecular mechanisms of toxic action of monohaloacetonitriles.

Among the manifold advantages of the Comet assay is the ability to evaluate very limited amounts of test agents. In many cases compounds for a research study are not commercially available and the chemicals must be individually synthesised at great expense and with large amounts of time involved. In this chapter we shall describe some of our work with a series of high-priority nitrogen-containing drinking-water disinfection byproducts (DBPs) that were synthesised as high-purity chemical analytical standards. The use of 96-well microplates allowed us to analyse the genotoxicity of these DBPs in mammalian cells.

Disinfection of drinking water protects public health against waterborne pathogens. However, during disinfection, toxic disinfection byproducts (DBPs) are formed. Exposure to DBPs was associated with increased risk of bladder cancer in humans. DBPs are generated at concentrations below their carcinogenic potencies; it is unclear how exposure leads to adverse health outcomes. We used computational estimates of the energy of the lowest unoccupied molecular orbital (ELUMO) to predict thiol reactivity and additive toxicity among soft electrophile DBPs. Bromoacetic acid (BAA) was identified as non-thiol-reactive, which was supported by in chemico and in vitro data. Bromoacetonitrile (BAN) and bromoacetamide (BAM) were thiol-reactive. Genotoxicity induced by these compounds was reduced by increasing the thiol pool with N-acetyl l-cysteine (NAC), while NAC had little effect on BAA. BAN and BAM shared depletion of glutathione (GSH) or cellular thiols as a molecular initiating event (MIE), whereas BAA induces toxicity through another pathway. Binary mixtures of BAM and BAN expressed a potentiating effect in genotoxicity. We found that soft electrophile DBPs could be an important predictor of common mechanism groups that demonstrated additive toxicity. In silico estimates of ELUMO could be used to identify the most relevant DBPs that are the forcing factors of the toxicity of finished drinking waters.

Alternative technologies to disinfect drinking water such as ultraviolet (UV) disinfection are becoming more widespread. The benefits of UV disinfection include reduced risk of microbial pathogens such as Cryptosporidium and reduced production of regulated drinking water disinfection byproducts (DBPs). The objective of this research was to determine if mammalian cell cytotoxicity and genotoxicity varied in response to different chlorination protocols with and without polychromatic medium pressure UV (MPUV) and monochromatic low pressure UV (LPUV) disinfection technologies. The specific aims were to analyze the mammalian cell cytotoxicity and genotoxicity of concentrated organic fractions from source water before and after chlorination and to determine the cytotoxicity and genotoxicity of the concentrated organic fractions from water samples treated with UV alone or UV before or after chlorination. Exposure of granular activated carbon-filtered Ohio River water to UV alone resulted in the lowest levels of mammalian cell cytotoxicity and genotoxicity. With combinations of UV and chlorine, the lowest levels of cytotoxicity and genotoxicity were observed with MPUV radiation. The best combined UV plus chlorine methodology that generated the lowest cytotoxicity and genotoxicity employed chlorination first followed by MPUV radiation. These data may prove important in the development of multibarrier methods of pathogen inactivation of drinking water, while limiting unintended toxic consequences.

Iodinated X-ray contrast media (ICM) were investigated as a source of iodine in the formation of iodo-trihalomethane (iodo-THM) and iodo-acid disinfection byproducts (DBPs), both of which are highly genotoxic and/or cytotoxic in mammalian cells. ICM are widely used at medical centers to enable imaging of soft tissues (e.g., organs, veins, blood vessels) and are designed to be inert substances, with 95% eliminated in urine and feces unmetabolized within 24 h. ICM are not well removed in wastewater treatment plants, such that they have been found at elevated concentrations in rivers and streams (up to 100 μg/L). Naturally occurring iodide in source waters is believed to be a primary source of iodine in the formation of iodo-DBPs, but a previous 23-city iodo-DBP occurrence study also revealed appreciable levels of iodo-DBPs in some drinking waters that had very low or no detectable iodide in their source waters. When 10 of the original 23 cities' source waters were resampled, four ICM were found--iopamidol, iopromide, iohexol, and diatrizoate--with iopamidol most frequently detected, in 6 of the 10 plants sampled, with concentrations up to 2700 ng/L. Subsequent controlled laboratory reactions of iopamidol with aqueous chlorine and monochloramine in the absence of natural organic matter (NOM) produced only trace levels of iodo-DBPs; however, when reacted in real source waters (containing NOM), chlorine and monochloramine produced significant levels of iodo-THMs and iodo-acids, up to 212 nM for dichloroiodomethane and 3.0 nM for iodoacetic acid, respectively, for chlorination. The pH behavior was different for chlorine and monochloramine, such that iodo-DBP concentrations maximized at higher pH (8.5) for chlorine, but at lower pH (6.5) for monochloramine. Extracts from chloraminated source waters with and without iopamidol, as well as from chlorinated source waters with iopamidol, were the most cytotoxic samples in mammalian cells. Source waters with iopamidol but no disinfectant added were the least cytotoxic. While extracts from chlorinated and chloraminated source waters were genotoxic, the addition of iopamidol enhanced their genotoxicity. Therefore, while ICM are not toxic in themselves, their presence in source waters may be a source of concern because of the formation of highly toxic iodo-DBPs in chlorinated and chloraminated drinking water.

Due to their efficacy in deactivating a range of microbial pathogens, particularly amoebic cysts, iodine-based disinfectants have been a popular option for point-of-use (POU) drinking water disinfection by campers, the military, and rural consumers in developing countries. Recently, concerns regarding the formation of cytotoxic and genotoxic iodinated disinfection byproducts (I-DBPs) have arisen during chloramine disinfection of iodide-containing waters in the developed world; similar concerns should pertain to iodine-based POU disinfection. Because there are alternative POU disinfection techniques, including chlorine-based disinfectants, this paper compared disinfection byproduct formation from a range of iodine-based disinfectants at their recommended dosages to chlorination and chloramination under overdosing conditions. Just as chloroform was the predominant trihalomethane (THM) forme during chlorination or chloramination, iodoform was the predominant THM formed during iodination. Conditions fostering THM formation were similar between these treatments, except that THM formation during chlorination increased with pH, while it was slightly elevated at circumneutral pH during iodination. Iodoform formation during treatment with iodine tincture was higher than during treatment with iodine tablets. On a molar basis, iodoform formation during treatment with iodine tincture was 20-60% of the formation of chloroform during chlorination, and total organic iodine (TOI) formation was twice that of total organic chlorine (TOCl), despite the 6-fold higher oxidant dose during chlorination. Based upon previous measurements of chronic mammalian cell cytotoxicity for the individual THMs, consumers of two waters treated with iodine tincture would receive the same THM-associated cytotoxic exposure in 4-19 days as a consumer of the same waters treated with a 6-fold higher dose of chlorine over 1 year. Iodoacetic acid, diiodoacetic acid, and other iodo-acids were also formed with iodine tincture treatment, but at levels11% of iodoform. However, testing of a Lifestraw Personal POU device, which combines an iodinated anion exchange resin with activated carbon post-treatment, indicated minimal formation of I-DBPs and no iodine residual. Although N-nitrosamines have been associated with oxidant contact with anion exchange resins, N-nitrosamine formation rapidly declined to low levels (4 ng/L) using the Lifestraw device after the first few flushes of water.

The monohaloacetic acids (monoHAAs), iodoacetic, bromoacetic and chloroacetic acids are toxic disinfection byproducts. In vitro toxicological end points were integrated with DNA damage and repair pathway-focused toxicogenomic analyses to evaluate monoHAA-induced alterations of gene expression in normal nontransformed human cells. When compared to concurrent control transcriptome profiles, metabolic pathways involved in the cellular responses to toxic agents were identified and provided insight into the biological mechanisms of toxicity. Using the Database for Annotation, Visualization and Integrated Discovery to analyze the gene array data, the majority of the altered transcriptome profiles were associated with genes responding to DNA damage or those regulating cell cycle or apoptosis. The major pathways involved with altered gene expression were ATM, MAPK, p53, BRCA1, BRCA2, and ATR. These latter pathways highlight the involvement of DNA repair, especially the repair of double strand DNA breaks. All of the resolved pathways are involved in human cell stress response to DNA damage and regulate different stages in cell cycle progression or apoptosis.

Characterizing the transformations of polypeptides is important across a broad range of scientific disciplines. As polypeptides are an important constituent of dissolved organic matter within seawater and freshwater, it is important to understand their (bio)geochemical fate. Oxidants, formed in blood as part of the immunological response or applied to waters for disinfection, react with polypeptides to form transformation products that may exert toxicity. An analytical method was developed to characterize and quantify modifications to the side chains of amino acid residues within polypeptides. Enzymatic digestion of polypeptides using Pronase E, a protease cocktail, proved preferable to common strong acid digestion techniques, because the circumneutral pH conditions employed during enzymatic digestion prevent artifacts arising from extreme pH conditions. Lysine nitrile, one of the predicted transformation products of lysine residues within polypeptides, was destroyed during strong acid digestion but not enzymatic digestion. Due to the potential variability in enzymatic digestion efficiencies, the liberation of a mass-labeled leucine monomer from an octapeptide spiked standard was employed as a measure of complete digestion efficiency for each sample and enabled quantification of modified amino acid residues within polypeptides. A multivariate statistical analysis was conducted to evaluate the influence on digestion efficiency of Pronase E loadings, salinity, natural organic matter concentration, and pH across the range of conditions relevant to blood, seawater, and concentrated freshwaters and disinfected drinking/recreational waters. At Pronase E loadings of 10 mg, the analysis indicated that digestion efficiencies ranged from 25 to 55% over the range of conditions expected for typical drinking waters concentrated from 1 L to 10 mL. The analytical method was applied to triplicate 1 L samples of a chlorinated tap water and a chlorinated indoor pool water. For the tap water, the digestion efficiency was 47.2% (+/-11.1% relative standard deviation), and the lysine nitrile concentration was 104.6 ng/L (+/-6.8 ng/L standard deviation). For the pool water, the digestion efficiency was 23% (+/-15% relative standard deviation); although the occurrence of lysine nitrile was verified, matrix effects must be overcome for quantification.

An occurrence study was conducted to measure five iodo-acids (iodoacetic acid, bromoiodoacetic acid, (Z)-3-bromo-3-iodo-propenoic acid, (E)-3-bromo-3-iodo-propenoic acid, and (E)-2-iodo-3-methylbutenedioic acid) and two iodo-trihalomethanes (iodo-THMs), (dichloroiodomethane and bromochloroiodomethane) in chloraminated and chlorinated drinking waters from 23 cities in the United States and Canada. Since iodoacetic acid was previouslyfound to be genotoxic in mammalian cells, the iodo-acids and iodo-THMs were analyzed for toxicity. A gas chromatography (GC)/negative chemical ionization-mass spectrometry (MS) method was developed to measure the iodo-acids; iodo-THMs were measured using GC/high resolution electron ionization-MS with isotope dilution. The iodo-acids and iodo-THMs were found in waters from most plants, at maximum levels of 1.7 microg/L (iodoacetic acid), 1.4 microg/L (bromoiodoacetic acid), 0.50 microg/L ((Z)-3-bromo-3-iodopropenoic acid), 0.28 microg/L ((E)-3-bromo-3-iodopropenoic acid), 0.58 microg/L ((E)-2-iodo-3-methylbutenedioic acid), 10.2 microg/L (bromochloroiodomethane), and 7.9 microg/L (dichloroiodomethane). Iodo-acids and iodo-THMs were highest at plants with short free chlorine contact times (1 min), and were lowest at a chlorine-only plant or at plants with long free chlorine contact times (45 min). Iodide levels in source waters ranged from 0.4 to 104.2 microg/L (when detected), but there was not a consistent correlation between bromide and iodide. The rank order for mammalian cell chronic cytotoxicity of the compounds measured in this study, plus other iodinated compounds, was iodoacetic acid(E)-3-bromo-2-iodopropenoic acidiodoform(E)-3-bromo-3-iodo-propenoic acid(Z)-3-bromo-3-iodo-propenoic aciddiiodoacetic acidbromoiodoacetic acid(E)-2-iodo-3-methylbutenedioic acidbromodiiodomethanedibromoiodomethanebromochloroiodomethane approximately chlorodiiodomethanedichloroiodomethane. With the exception of iodoform, the iodo-THMs were much less cytotoxic than the iodo-acids. Of the 13 compounds analyzed, 7 were genotoxic; their rank order was iodoacetic aciddiiodoacetic acidchlorodiiodomethanebromoiodoacetic acidE-2-iodo-3-methylbutenedioic acid(E)-3-bromo-3-iodo-propenoic acid(E)-3-bromo-2-iodopropenoic acid. In general, compounds that contain an iodo-group have enhanced mammalian cell cytotoxicity and genotoxicity as compared to their brominated and chlorinated analogues.

The haloacetamides, a class of emerging nitrogenous drinking water disinfection byproduct (DBPs), were analyzed for their chronic cytotoxicity and for the induction of genomic DNA damage in Chinese hamster ovary cells. The rank order for cytotoxicity of 13 haloacetamides was DIAcAmIAcAmBAcAmTBAcAmBIAcAmDBCAcAmCIAcAmBDCAcAmDBAcAmBCAcAmCAcAmDCAcAmTCAcAm. The rank order of their genotoxicity was TBAcAmDIAcAm approximately equal to IAcAmBAcAmDBCAcAmBIAcAmBDCAcAmCIAcAmBCAcAmDBAcAmCAcAmTCAcAm. DCAcAm was not genotoxic. Cytotoxicity and genotoxicity were primarily determined by the leaving tendency of the halogens and followed the order IBrCl. With the exception of brominated trihaloacetamides, most of the toxicity rank order was consistent with structure-activity relationship expectations. For di- and trihaloacetamides, the presence of at least one good leaving halogen group (I or Br but not Cl) appears to be critical for significant toxic activity. Log P was not a factor for monohaloacetamides but may play a role in the genotoxicity of trihaloacetamides and possible activation of dihaloacetamides by intracellular GSH and -SH compounds.

Disinfection by-products (DBPs) are formed when disinfectants (chlorine, ozone, chlorine dioxide, or chloramines) react with naturally occurring organic matter, anthropogenic contaminants, bromide, and iodide during the production of drinking water. Here we review 30 years of research on the occurrence, genotoxicity, and carcinogenicity of 85 DBPs, 11 of which are currently regulated by the U.S., and 74 of which are considered emerging DBPs due to their moderate occurrence levels and/or toxicological properties. These 74 include halonitromethanes, iodo-acids and other unregulated halo-acids, iodo-trihalomethanes (THMs), and other unregulated halomethanes, halofuranones (MX [3-chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone] and brominated MX DBPs), haloamides, haloacetonitriles, tribromopyrrole, aldehydes, and N-nitrosodimethylamine (NDMA) and other nitrosamines. Alternative disinfection practices result in drinking water from which extracted organic material is less mutagenic than extracts of chlorinated water. However, the levels of many emerging DBPs are increased by alternative disinfectants (primarily ozone or chloramines) compared to chlorination, and many emerging DBPs are more genotoxic than some of the regulated DBPs. Our analysis identified three categories of DBPs of particular interest. Category 1 contains eight DBPs with some or all of the toxicologic characteristics of human carcinogens: four regulated (bromodichloromethane, dichloroacetic acid, dibromoacetic acid, and bromate) and four unregulated DBPs (formaldehyde, acetaldehyde, MX, and NDMA). Categories 2 and 3 contain 43 emerging DBPs that are present at moderate levels (sub- to low-mug/L): category 2 contains 29 of these that are genotoxic (including chloral hydrate and chloroacetaldehyde, which are also a rodent carcinogens); category 3 contains the remaining 14 for which little or no toxicological data are available. In general, the brominated DBPs are both more genotoxic and carcinogenic than are chlorinated compounds, and iodinated DBPs were the most genotoxic of all but have not been tested for carcinogenicity. There were toxicological data gaps for even some of the 11 regulated DBPs, as well as for most of the 74 emerging DBPs. A systematic assessment of DBPs for genotoxicity has been performed for approximately 60 DBPs for DNA damage in mammalian cells and 16 for mutagenicity in Salmonella. A recent epidemiologic study found that much of the risk for bladder cancer associated with drinking water was associated with three factors: THM levels, showering/bathing/swimming (i.e., dermal/inhalation exposure), and genotype (having the GSTT1-1 gene). This finding, along with mechanistic studies, highlights the emerging importance of dermal/inhalation exposure to the THMs, or possibly other DBPs, and the role of genotype for risk for drinking-water-associated bladder cancer. More than 50% of the total organic halogen (TOX) formed by chlorination and more than 50% of the assimilable organic carbon (AOC) formed by ozonation has not been identified chemically. The potential interactions among the 600 identified DBPs in the complex mixture of drinking water to which we are exposed by various routes is not reflected in any of the toxicology studies of individual DBPs. The categories of DBPs described here, the identified data gaps, and the emerging role of dermal/inhalation exposure provide guidance for drinking water and public health research.
