IL‐2 is a pleiotropic cytokine that regulates immune cell homeostasis. Its immunomodulatory function has been used clinically as an active immunotherapy agent for metastatic cancers. However, severe adverse effects, including the vascular leak syndrome and the preferential stimulation of anti‐immunogenic Treg rather than effector T cells, have been obstacles. We newly designed a mutein IL‐2, Mutakine‐6 (MK‐6), with reduced IL‐2Rα–binding capability. MK‐6 induced comparable cell growth potential toward IL‐2Rβγ–positive T cells but was far less efficient in in vitro Treg proliferation and STAT5 activation. Unlike IL‐2, in vivo administration of MK‐6 produced minimal adverse effects. Using CT26 and B16F10‐syngeneic tumor models, we found MK‐6 was highly efficacious on tumor regression. Serum albumin conjugation to MK‐6 prolonged in vivo half‐life and accumulated in CT26 tumors, showing enhanced antitumor effect. Tumor‐infiltrating leukocytes analysis revealed that albumin‐fused MK‐6 increased the ratio of effector CD8+ T cells to CD4+ Treg cells. These results demonstrated that MK‐6 is an efficient immunomodulator potentially used for improved immunotherapy with decreased adverse effects and attenuated Treg stimulation., Albumin‐conjugated IL‐2 mutein, MK‐6, exerts a potent antitumor effect in the CT26 tumor model.

In this study, a corrosion-stable silica membrane was developed to be used in H2 purification during the hydrogen iodide decomposition (2HI → H2 + I2), which is a new application of the silica membranes. From a practical perspective, the membrane separation length was enlarged up to 400 mm and one end of the membrane tubes was closed to avoid any thermal variation along the membrane length and sealing issues. The silica membranes consisted of a three-layer structure comprising a porous α-Al2O3 ceramic support, an intermediate layer, and a top silica layer. The intermediate layer was composed of γ-Al2O3 or silica, and the top silica layer that is H2 selective was prepared via counter-diffusion chemical vapor deposition of a hexyltrimethoxysilane. To the best of our knowledge, this is the first report of 400-mm-long closed-end silica membranes supported on Si-formed α-Al2O3 tubes produced via chemical vapor deposition method. A 400-mm-long closed-end membrane using a Si-formed α-Al2O3 tube exhibited a higher H2/SF6 selectivity of 1240 but lower H2 permeance of 1.4 × 10−7 mol Pa−1 m−2 s−1 with compared with the membrane using a γ-Al2O3-formed α-Al2O3 tube (907 and 5.6 × 10−7 mol Pa−1 m−2 s−1, respectively). The membrane using the Si-formed α-Al2O3 tube was more stable in corrosive HI gas than a membrane with a γ-Al2O3-formed α-Al2O3 tube after 300 h of stability tests. In conclusion, the developed silica membranes using the Si-formed α-Al2O3 tubes seem suitable for membrane reactors that produce H2 on large scale using HI decomposition in the thermochemical iodine–sulfur process.

OBJECTIVE Pheochromocytoma is a rare neuroendocrine tumour that secretes catecholamines and originates in the adrenal gland. Although surgical resection is the only curative therapy for pheochromocytoma, it is associated with a risk of haemodynamic instability (HDI), such as extremely high blood pressure and/or post tumour removal hypotension and shock. We investigated the risk factors for HDI during pheochromocytoma surgery. In the activation of cell-mediated adaptive immune responses that play major roles in the elimination of virus-infected or tumor cells, it is important that dendritic cells present antigen peptides on major histocompatibility complex (MHC) class I molecules and activate pathogen-specific cytotoxic T lymphocytes (CTL). As exogenous peptide antigens are generally presented on MHC class II but not class I, the development of a method for exogenous antigen delivery that facilitates MHC class I presentation is necessary for a potentially effective vaccine that is expected to provoke cell-mediated adaptive immune responses. Here, we developed extracellular vesicles that incorporate antigenic proteins by utilizing endosomal sorting complexes required for transport (ESCRT)-mediated vesicle formation pathway. Furthermore, we proved that these vesicles could deliver their contents to the cytoplasm of dendritic cells and activate antigen-specific CTLs. These technologies could be applied to the development of novel CTL-inducing peptide vaccines.

β-Catenin is an important component of the Wnt signalling pathway. As dysregulation or mutation of this pathway causes many diseases, including cancer, the β-Catenin level is carefully regulated by the destruction complex in the Wnt signalling pathway. However, the mechanisms underlying the regulation of β-Catenin ubiquitination and degradation remain unclear. Here, we find that WNK (With No Lysine [K]) kinase is a potential regulator of the Wnt signalling pathway. We show that WNK protects the interaction between β-Catenin and the Glucose-Induced degradation Deficient (GID) complex, which includes an E3 ubiquitin ligase targeting β-Catenin, and that WNK regulates the β-Catenin level. Furthermore, we show that WNK inhibitors induced β-Catenin degradation and that one of these inhibitors suppressed xenograft tumour development in mice. This study demonstrates the rapid fabrication and utility of 2-methacryloyloxyethyl phosphorylcholine (MPC) polymer film for cell patterning. The film was obtained on a cell culture surface by microcasting MPC polymer ethanol solution into a degassed polydimethylsiloxane mold with a desired pattern. After removal of the mold, 293AD cells were cultured on the surface of the polymer film with the patterned microstructures. Patterned cell adhesion restricted by the film was successfully maintained during at least a 168-h cultivation. The microcast MPC polymer film can be prepared rapidly and used for efficient long-term cell confinement.

The development of systemic therapies, including vascular endothelial growth factor-tyrosine kinase inhibitors (VEGF-TKI) and mammalian target of rapamycin (mTOR) inhibitors, represents a major breakthrough in the treatment of patients with renal cell carcinoma (RCC). However, inherent resistance is observed in some patients and acquired resistance commonly develops in many patients within several months of the initiation of systemic therapies. Since these treatments rarely cure patients, their aim is to suppress tumor progression and prolong survival. Therefore, the establishment of dependable criteria that predict responses and resistance to systemic therapies is clinically important, and the underlying molecular mechanisms also need to be elucidated for the future development of more effective therapies. We herein review recent advances in research on the molecular mechanisms underlying resistance, with a focus on morphological characteristics, tumor angiogenesis, and the tumor immune microenvironment in RCC and their relationships with VEGF-TKI treatments. Recent therapies using immune checkpoint inhibitors (ICI) and newly developed VEGF-TKI also appear to be effective for advanced RCC, with stable and durable responses to ICI being observed in some RCC patients. These new drugs and their outcomes have been briefly described.

The thermochemical water-splitting iodine–sulfur (IS) process requires corrosion-resistant materials owing to usage of corrosive fluids, such as a mixture of HI–I2–H2O. Fluoropolymers, such as PTFE and PFA, are adaptable as lining materials for protecting plant components. However, there has been a concern: PTFE and PFA have the ability to permeate various permeants. From the viewpoint of corrosion of the base material, the permeation characteristics of HI and I2 should be evaluated to improve the integrity of the IS process. In this study, permeation tests on PTFE and PFA membranes were performed to measure the permeated fluxes of HI and I2, and the effects of the operating conditions on them were investigated. The introduction of a permeability parameter could be successful for normalizing the permeated fluxes for a specific membrane thickness and a vapor pressure. Then, the empirical formula of the permeability was given as an Arrhenius-type equation to use as a plant design. Finally, based on the results, the proper conditions for design of a lining material for the inhibition of HI and I2 permeation are summarized.

An electrochemical membrane Bunsen reaction using a cation exchange membrane (CEM) is a key to achieving iodine-sulfur (IS) thermochemical water splitting for the mass-production of hydrogen. In this study, we prepared a radiation-grafted CEM with a high ion exchange capacity (IEC) and a highly-porous Au-electroplated anode, and then used them for the membrane Bunsen reaction to reduce cell overvoltage. The high ionic content of our CEM led to low resistivity for proton transport, while the high porosity of the electrode led to a large effective surface area for anodic SO2 oxidation. The cell overvoltage for the membrane Bunsen reaction was significantly reduced to 0.21 V at 200 mA/cm2, one-third of that achieved using a commercial CEM and non-porous anode. From the analysis of the current-voltage characteristics, the grafted CEM was demonstrated to play a dominant role in the overvoltage reduction compared to the porous Au anode.

CONTEXT The oversecretion of plasma aldosterone by unilateral aldosterone-producing adenoma (APA) can be cured by adrenalectomy. However, the time needed for the endocrine environment to normalize remains unclear. OBJECTIVE To clarify adequate timing for a biochemical evaluation in unilateral APA patients after adrenalectomy. Cancer stem cells (CSCs) have tumour initiation, self-renewal, and long-term tumour repopulation properties, and it is postulated that differentiated somatic cells can be reprogrammed to CSCs by oncogenic signals. We previously showed that oncogenic HRASV12 conferred tumour initiation capacity in tumour suppressor p53-deficient (p53−/−) primary mouse embryonic fibroblasts (MEFs) through transcription factor NF-κB-mediated enhancement of glucose uptake; however, the underlying mechanisms of RAS oncogene-induced CSC reprogramming have not been elucidated. Here, we found that the expression of the reprogramming factor SOX2 was induced by HRASV12 in p53−/− MEFs. Moreover, gene knockout studies revealed that SOX2 is an essential factor for the generation of CSCs by HRASV12 in mouse and human fibroblasts. We demonstrated that HRASV12-induced cyclin-dependent kinase 1 (CDK1) activity and subsequent enhancement of protein O-GlcNAcylation were required for SOX2 induction and CSC generation in these fibroblasts and cancer cell lines containing RAS mutations. Moreover, the CDK inhibitor dinaciclib and O-GlcNAcylation inhibitor OSMI1 reduced the number of CSCs derived from these cells. Taken together, our results reveal a signalling pathway and mechanism for CSC generation by oncogenic RAS and suggest the possibility that this signalling pathway is a therapeutic target for CSCs.

Background Cabozantinib is an oral tyrosine kinase inhibitor in renal cell carcinoma (RCC), whose targets include oncogenic AXL and unique ligand GAS6. Critical gaps in basic knowledge need to be addressed to devise an exclusive biomarker and candidate when targeting the AXL/GAS6 axis. Methods To clarify the effects of the AXL/GAS6 axis on RCC, we herein performed a large-scale immunogenomic analysis and single-cell counts including various metastatic organs and histological subtypes of RCC. We further applied genome-wide mutation analyses and methylation arrays. Results Varying patterns of AXL and GAS6 expression were observed throughout primary RCC tumours and metastases. Scoring individual AXL/GAS6 levels in the tumour centre and invasive margin, namely, the AXL/GAS6 score, showed a good ability to predict the prognosis of clear cell RCC. Metastasis- and histological subtype-specific differences in the AXL/GAS6 score existed since lung metastasis and the papillary subtype were weakly related to the AXL/GAS6 axis. Cell-by-cell immunohistological assessments clarified an immunosuppressive environment in tumours with high AXL/GAS6 scores. Genomic alterations in the PI3K-mTOR pathway and DNA methylation profiling revealed distinct differences with the AXL/GAS6 score in ccRCC. Conclusion The AXL/GAS6 scoring system could predict the outcome of prognosis and work as a robust biomarker for the immunogenomic state in RCC.

In this study, the catalytic decomposition of hydrogen iodide was theoretically and experimentally investigated in a silica-based ceramic membrane reactor to assess the reactor's suitability for thermochemical hydrogen production. The silica membranes were fabricated by depositing a thin silica layer onto the surface of porous alumina ceramic support tubes via counter-diffusion chemical vapor deposition of hexyltrimethoxysilane. The performance of the silica-based ceramic membrane reactor was evaluated by exploring important operating parameters such as the flow rates of the hydrogen iodide feed and the nitrogen sweep gas. The influence of the flow rates on the hydrogen iodide decomposition conversion was investigated in the lower range of the investigated feed flow rates and in the higher range of the sweep-gas flow rates. The experimental data agreed with the simulation results reasonably well, and both highlighted the possibility of achieving a conversion greater than 0.70 at decomposition temperature of 400 °C. Therefore, the developed silica-based ceramic membrane reactor could enhance the total thermal efficiency of the thermochemical process.

The potential of the silica membrane reactors for use in the decomposition of hydrogen iodide (HI) was investigated by simulation with the aim of producing CO2-free hydrogen via the thermochemical water-splitting iodine-sulfur process. Simulation model validation was done using the data derived from an experimental membrane reactor. The simulated results showed good agreement with the experimental findings. The important process parameters determining the performance of the membrane reactor used for HI decomposition, namely, reaction temperature, total pressures on both the feed side and the permeate side, and HI feed flow rate were investigated theoritically by means of a simulation. It was found that the conversion of HI decomposition can be improved by up to four times (80%) or greater than the equilibrium conversion (20%) at 400 °C by employing a membrane reactor equipped with a tubular silica membrane. The features to design the membrane reactor module for HI decomposition of the thermochemical iodine-sulfur process were discussed under a wide range of operation conditions by evaluating the relationship between HI conversion and number of membrane tubes.

It is essential to understand the combined effects of elevated CO2 and temperature on phytoplankton-zooplankton link when attempting to predict climate change responses of freshwater ecosystems. Phytoplankton species differ in stoichiometric and fatty acids composition, and this may result in phytoplankton-mediated effect on zooplankton at elevated CO2 and temperature. Beyond the isolated analysis of CO2 or temperature effect, few studies have assessed zooplankton growth under the phytoplankton-mediated effects of elevated CO2 and temperature. In this study, three algal species (green alga, diatom, cyanobacteria) were fed on zooplankton Daphnia magna, under the conditions of CO2 concentrations of ambient (390 ppm) and elevated (1000 ppm) levels and temperatures at 20, 25 and 30 °C. Elevated CO2 increased the algal biomass, while it reduced the phosphorus (P) and ω3 polyunsaturated fatty acids (ω3 PUFAs) to carbon (C) ratios. Elevated temperature decreased the P/C ratios in all algal cultures and ω3 PUFAs/C ratios in the diatom and the cyanobacteria cultures. Phytoplankton-mediated effect of elevated CO2 reduced the growth of zooplankton fed on the green and the mixed three algae culture. The stimulation of zooplankton fed on the diatom and the cyanobacteria by elevated temperature can be offset by decreasing food P and ω3 PUFAs contents. The combined effects of elevated CO2 and temperature on the growth of daphnids were mainly mediated by ω3 PUFAs/C ratios in the phytoplankton. Rising temperature as a combined direct and indirectly phytoplankton-mediated effect on zooplankton may be able to ameliorate the negative effects of elevated CO2. The results indicated that the combined effects of increased CO2 and temperature increased the fatty acid content of the green alga but not the other algae. This study highlighted that climate change with simultaneously increasing temperature and CO2 may entangle the carbon transfer in freshwater planktonic communities.

Single cell analysis has gained attention as a means to investigate the heterogeneity of cells and amplify a cell with desired characteristics. However, obtaining a single cell from a large number of cells remains difficult because preparation of single-cell samples relies on conventional techniques such as pipetting that are labor intensive. In this study, we developed a system combining a 0.6-mm thin glass microfluidic device and machine vision approach to isolate single Euglena gracilis cells, as a model of microorganism with mobility, in a small/thin glass chamber. A single E. gracilis cell in a chamber was cultured for 4 days to monitor its multiplication. With this system, we successfully simplified preparation of single cells of interest and determined that it is possible to combine it with other analytical techniques to observe single cells continuously.

Inducing apoptosis is an effective treatment for cancer. Conventional cytotoxic anticancer agents induce apoptosis primarily through activation of tumor suppressor p53 by causing DNA damage and the resulting regulation of B-cell leukemia/lymphoma-2 (BCL-2) family proteins. Therefore, the effects of these agents are limited in cancers where p53 loss-of-function mutations are common, such as triple-negative breast cancer (TNBC). Here, we demonstrate that ultraviolet (UV) light-induced p53-independent transcriptional activation of NOXA, a proapoptotic factor in the BCL-2 family, results in apoptosis induction. This UV light-induced NOXA expression was triggered by extracellular signal-regulated kinase (ERK) activity. Moreover, we identified the specific UV light-inducible DNA element of the NOXA promoter and found that this sequence is responsible for transcription factor Krüppel-like factor 4 (KLF4)-mediated induction. In p53-mutated TNBC cells, inhibition of KLF4 by RNA interference reduced NOXA expression. Furthermore, treatment of TNBC cells with a KLF4-inducing small compound, APTO-253, resulted in the induction of NOXA expression and NOXA-mediated apoptosis. Therefore, our results help to clarify the molecular mechanism of DNA damage-induced apoptosis and provide support for a possible treatment method for p53-mutated cancers.

Tumour-promoting inflammation is a hallmark of cancer, and chronic inflammatory disease increases the risk of cancer. In this context, MYD88, a downstream signalling molecule of Toll-like receptors that initiates inflammatory signalling cascades, has a critical role in tumour development in mice and its gene mutation was found in human cancers. In inflammation-induced colon cancer, tumour suppressor p53 mutations have also been detected with high frequency as early events. However, the molecular mechanism of MYD88-induced cancer development is poorly understood. Here, we demonstrated that MYD88 induced the protein accumulation of the transcription factor HIF-1α through NF-κB in p53-deficient cells. HIF-1α accumulation was not caused by enhanced protein stability but by NF-κB-mediated transcriptional activation, the enhanced translation of HIF-1α and JNK activation. In contrast, MYD88-induced mRNA expressions of HIF-1α and HIF-1-target genes were attenuated in the presence of p53. Furthermore, constitutively active forms of MYD88 induced tumour-initiating cell (TIC) generation in p53-deficient cells, as determined by tumour xenografts in nude mice. TIC generating activity was diminished by the suppression of NF-κB or HIF-1α. These results indicate that MYD88 signals induce the generation of TICs through the NF-κB-HIF-1α activation cascade in p53-deficient cells and suggest this molecular mechanism underlies inflammation-induced cancer development.

Patterned cell culturing is one of the most useful techniques for understanding the interaction between geometric conditions surrounding cells and their behaviors. The authors previously proposed a simple method for cell patterning with an agarose gel microstructure fabricated by microcasting with a degassed polydimethylsiloxane (PDMS) mold. Although the vacuum pressure produced from the degassed PDMS can drive a highly viscous agarose solution, the influence of solution viscosity on the casting process is unknown. This study investigated the influences of micro-channel dimensions or solution viscosity on the flow of the solution in a micro-channel of a PDMS mold by both experiments and numerical simulation. It was found experimentally that the degassed PDMS mold was able to drive a solution with a viscosity under 575 mPa·s. A simulation model was developed which can well estimate the flow rate in various dimensions of micro-channels. Cross-linked albumin has low viscosity (1 mPa·s) in aqueous solution and can undergo a one-way dehydration process from solution to solid that produces cellular repellency after dehydration. A microstructure of cross-linked albumin was fabricated on a cell culture dish by the microcasting method. After cells were seeded and cultivated on the cell culture dish with the microstructure for 7 days, the cellular pattern of mouse skeletal myoblast cell line C2C12 was observed. The microcasting with cross-linked albumin solution enables preparation of patterned cell culture systems more quickly in comparison with the previous agarose gel casting, which requires a gelation process before the dehydration process.

2-Methacryloyloxyethyl phosphorylcholine (MPC) polymers including hydrophobic units such as poly(MPC-co-butyl methacrylate) (PMB) and poly(MPC-co-dodecyl methacrylate) (PMD) are used as coating agents for medical devices because of their antifouling effects. In this study, the whole hydration process of MPC polymer-coated surfaces was investigated using air-injection-mediated liquid exclusion (AILE) methods in which the liquid exclusion diameter during air injection was correlated to the water-repelling property. The prejetted and standard AILE methods showed the initial change from a dry to a wet state and the swelling behaviors of the MPC polymers, respectively. The liquid exclusion diameter of the MPC polymer-coated surfaces increased with an increase in the immersion time in various aqueous solutions such as deionized water, phosphate-buffered saline (PBS), and cell culture media. Moreover, the liquid exclusion diameter of the PMD-coated surface was larger than that of the PMB-coated one. Ellipsometry directly indicated the polymer layers swollen in water. Scanning probe microscopy (SPM) revealed that nanosized protuberances were formed in water, especially at the PMD-coated surface. The different swelling behaviors of these MPC polymer-coated surfaces affected the liquid exclusion diameters. Thus, the AILE methods are a powerful tool to elucidate the hydration process in various liquid media.

The extracellular matrix (ECM) surrounding cancer cells becomes stiffer during tumor progression, which influences cancer cell behaviors such as invasion and proliferation through modulation of gene expression as well as remodeling of the actin cytoskeleton. In this study, we show that MMP24 encoding matrix metalloproteinase (MMP)-24 is a novel target gene of Yes-associated protein (YAP), a transcription coactivator known as a mechanotransducer. We first examined the effect of substrate stiffness on MMP24 expression in MCF-7 human breast cancer cells and showed that the expression of MMP24 was significantly higher in cells grown on stiff substrates than that on soft substrates. The MMP24 expression was significantly reduced by knockdown of YAP. In contrast, the expression of constitutively active YAP increased MMP24 promoter activity. In addition, binding of YAP to the MMP24 promoter was confirmed by the chromatin immunoprecipitation (ChIP) assay. These results show that ECM stiffening promotes YAP activation, thereby inducing MMP24 expression. Based on the Human Protein Atlas database, breast cancer patients with lower MMP24 expression exhibit the worse survival rates overall. Thus, MMP24 may negatively regulate the aggressiveness of cancer cells under the stiff ECM environment during tumor progression.

HER3, a member of the receptor tyrosine kinase super family, is overexpressed in a number of cancers, and is associated with malignant phenotypes. Control of the protein stability of the membrane, as well as nuclear receptors, has been known to be an important process affecting tumor cells; however, their relationships have yet to be elucidated. In this study, we demonstrate that estradiol promotes rapid degradation of HER3 via the proteasome pathway in ER-positive breast cancer, MCF-7. ER prevented HER3 degradation, and knockdown of ER expression by si-RNA promoted rapid degradation of HER3. Breakdown of HER3 and ER were regulated by a ubiquitin ligase Nedd4-1 in the presence of estradiol stimulation. We speculate that estradiol quickly degrades ER, making HER3 accessible by Nedd4-1, and leads to the rapid degradation of HER3. In addition, knockdown of ubiquitin ligase Nedd4-1 enhances estradiol induced cell proliferation. These results indicate that HER3 and Nedd4-1 in ER-positive breast cancers might be an important therapeutic target., Highlights • First time we found that estradiol promotes rapid degradation of HER3 in ER-positive breast cancer MCF-7 cells. • Knockdown of ubiquitin ligase Nedd4-1 decreased the degradation efficiency of HER3 in the presence of estradiol. • Knockdown of Nedd4-1 enhanced estradiol induced tumor cell proliferation.

In the realm of stationary energy storage, a plurality of candidate chemistries continues to vie for acceptance, among them the Na–NiCl2 displacement battery, which has eluded widespread adoption owing to the fragility of the β″-Al2O3 membrane. Here we report a porous electronically conductive membrane, which achieves chemical selectivity by preferred faradaic reaction instead of by regulated ionic conduction. Fitted with a porous membrane of TiN, a displacement cell comprising a liquid Pb positive electrode, a liquid Li–Pb negative electrode and a molten-salt electrolyte of PbCl2 dissolved in LiCl–KCl eutectic was cycled at a current density of 150 mA cm−2 at a temperature of 410 °C and exhibited a coulombic efficiency of 92% and a round-trip energy efficiency of 71%. As an indication of industrial scalability, we show comparable performance in a cell fitted with a faradaic membrane fashioned out of porous metal. Molten-salt batteries such as Na–NiCl2 are promising candidates for grid storage, but suffer from fragility of ion-selective ceramic membranes. Here the authors report the operation of a Li–Pb||PbCl2 battery fitted with a robust TiN mesh membrane that functions by protective faradaic reaction.

Hereditary leiomyomatosis and renal cell carcinoma (HL (RCC)) entails cutaneous and uterine leiomyomatosis with aggressive type 2 papillary RCC-like histology. HLRCC is caused by pathogenic variants in the FH gene, which encodes fumarate hydratase (FH). Here, we describe an episode of young-onset RCC caused by a genomic FH deletion that was diagnosed via clinical sequencing. A 35-year-old woman was diagnosed with RCC and multiple metastases: histopathological analyses supported a diagnosis of FH-deficient RCC. Although the patient had neither skin tumors nor a family history of HLRCC, an aggressive clinical course at her age and pathological diagnosis of FH-deficient RCC suggested a germline FH variant. After counseling, the patient provided written informed consent for germline genetic testing. She was simultaneously subjected to paired tumor profiling tests targeting the exome to identify a therapeutic target. Although conventional germline sequencing did not detect FH variants, exome sequencing revealed a heterozygous germline FH deletion. As such, paired tumor profiling, not conventional sequencing, was required to identify this genetic deletion. RCC caused by a germline FH deletion has hitherto not been described in Japan, and the FH deletion detected in this patient was presumed to be of maternal European origin. Although the genotype-phenotype correlation in HLRCC-related tumors is unclear, the patient’s family was advised to undergo genetic counseling to consider additional RCC screening.

Hydrogen production from nuclear energy has attracted considerable interest as a clean energy solution to address the challenges of climate change and environmental sustainability. With respect to the large-scale and economical production of hydrogen using nuclear energy, the thermochemical water-splitting iodine-sulfur (IS) process is a promising method. The IS process uses sulfur and iodine compounds to decompose water into its elemental constituents, hydrogen and oxygen, by using three coupled chemical reactions: the Bunsen reaction; sulfuric acid decomposition; and hydrogen iodide (HI) decomposition. The decomposition of HI is the efficiency-determining step of the process. In this work, a membrane reactor with a silica membrane closed on one end was designed, and its potential for hydrogen production from HI decomposition was explored. In the reactor-module design, only one end of the membrane tube was fixed, while the closed-end of the tube was freely suspended to avoid thermal expansion effects. The closed-end silica membranes were prepared for the first time by a counter-diffusion chemical vapor deposition of hexyltrimethoxysilane. In application, HI conversion of greater than 0.60 was achieved at a decomposition temperature of 400 °C, which is three times greater than the equilibrium conversion (0.20). Thus, the membrane reactor with closed-end silica membrane was shown to produce a successful equilibrium shift in the production of hydrogen via HI decomposition in the thermochemical IS process.

An anaerobic membrane bioreactor was configured for methanogenic degradation of cellulose-containing sewage. The degradation performance and microbial changes were evaluated under five hydraulic retention times (HRTs). The results indicated the methane production was largely enhanced with 92.6% efficiency of chemical oxygen demand (COD) converting to methane and 80% proportion of methane in produced biogas, meanwhile the biomass yield presented the fewest at the shortest HRT 8 h. Enhanced methane production with decreased biomass yield was attributed to an association between fungi and methanogens. Microbial analysis showed fungi Basidiomycota and methanogen Methanoregula apparently established the association, especially Basidiomycota reaching 93% relative abundance at HRT 8 h. Specific methanogenic activity (SMA) and biochemical methane potential (BMP) tests suggested the association was derived from H 2 production by fungi and H 2 consumption by methanogens, during the process of cellulose degradation. The methanogenic degradation of cellulose-containing sewage was markedly promoted via the fungi-methanogens syntrophic association.

High-throughput microscopy of bacterial cells elucidated fundamental cellular processes including cellular heterogeneity and cell division homeostasis. Polydimethylsiloxane (PDMS)-based microfluidic devices provide advantages including precise positioning of cells and throughput, however device fabrication is time-consuming and requires specialised skills. Agarose pads are a popular alternative, however cells often clump together, which hinders single cell quantitation. Here, we imprint agarose pads with micro-patterned ‘capsules’, to trap individual cells and ‘lines’, to direct cellular growth outwards in a straight line. We implement this micro-patterning into multi-pad devices called CapsuleHotel and LineHotel for high-throughput imaging. CapsuleHotel provides ~65,000 capsule structures per mm2 that isolate individual Escherichia coli cells. In contrast, LineHotel provides ~300 line structures per mm that direct growth of micro-colonies. With CapsuleHotel, a quantitative single cell dataset of ~10,000 cells across 24 samples can be acquired and analysed in under 1 hour. LineHotel allows tracking growth of > 10 micro-colonies across 24 samples simultaneously for up to 4 generations. These easy-to-use devices can be provided in kit format, and will accelerate discoveries in diverse fields ranging from microbiology to systems and synthetic biology.

The IS process is the most deeply investigated thermochemical water-splitting hydrogen production cycle. It is in a process engineering stage in JAEA to use industrial materials for components. Important engineering tasks are verification of integrity of the total process and stability of hydrogen production in harsh environment. A test facility using corrosion-resistant materials was constructed. The hydrogen production ability was 100 L/h. Operation tests of each section were conducted to confirm basic functions of reactors and separators, etc. Then, a trial operation for integration of the sections was successfully conducted to produce hydrogen of about 10 L/h for 8 hours.

The decomposition of hydrogen iodide in the thermochemical water splitting iodine–sulfur process at an intermediate temperature (400 °C) using a catalytic membrane reactor was reported here, for the first time. The performance of a catalytic membrane reactor based on a hexyltrimethoxysilane-derived silica membranes (H2 permeance of 9.4 × 10−7 mol Pa−1 m−2 s−1 and H2/N2 selectivity of over 80.0.) was evaluated at 400 °C by varying the HI flow rates of 2.6, 4.7, 6.9, 8.4, and 9.7 mL min−1. The silica membranes were prepared by counter-diffusion chemical vapor deposition method on γ-alumina-coated α-alumina tubes. Hydrogen was successfully extracted from the membrane reactor using the silica membrane at 400 °C. A significant increase in HI conversion was achieved. The conversion achieved at an HI flow rate of 2.6 mL min−1 was approximately 0.60, which was greater than the equilibrium conversion in HI decomposition (0.22).

A scaffold-hopping strategy towards a new pyrazolo[1,5- a ]pyridine based core using molecular hybridization of two structurally distinct EP 1 antagonists, followed by structure-activity relationship-guided optimization, resulted in the identification of potent EP 1 antagonists exemplified by 4c , 4f , and 4j , which were shown to reduce pathological intravesical pressure in rats when administered at 1 mg/kg iv.

Current R&D on the thermochemical water splitting iodine-sulfur (IS) process in Japan Atomic Energy Agency (JAEA) is summarized. Reactors were fabricated with industrial materials and verified by test operations: a Bunsen reactor, a H2SO4 decomposer, and a HI decomposer. Component materials of the reactors were stable in the operation environment. Small amount of H2SO4 in the anolyte solution in an electro-electrodialysis (EED) cell had no negative impact on cell performance parameters. Relationship between cell solution composition and temperature and cell parameters was formulated by experimental data. Demonstration of the test facility with process design of 100 L/h hydrogen production is performed to verify integrity of process components and stability of hydrogen production. Tests of sections were first conducted individually to show material processing rates were controllable. Based on the result, an 8-h continuous operation of the total IS process was performed in February 2016 with H2 production rate of 10 L/h. Demonstrations are planned for longer operation period and higher H2 production rate after improvement of components to prevent troubles.

Novel pyrazolo[1,5- a ]pyridine derivatives were designed, synthesized and evaluated as orally active EP 1 antagonists for the treatment of overactive bladder. Matched molecular pair analysis (MMPA) allowed the design of a new series of pyrazolo[1,5- a ]pyridine derivatives 4 – 6 . Structure-activity relationships (SAR) studies of 4 – 6 were performed, leading to identification of the nanomolar-level EP 1 antagonist 4c , which exhibited good pharmacological effect through intraduodenal (id) administration in a 17-phenyltrinor prostaglandin E2-induced bladder contraction model in rats.

A high-performance, H2-permselective silica membrane derived from hexyltrimethoxysilane (HTMOS) was developed for application in the thermochemical water-splitting iodine–sulfur process. Silica membranes, referred to here as HTMOS membranes, were prepared via counter-diffusion chemical vapor deposition on γ-alumina-coated α-alumina support tubes with outer diameters of 10 mm. Special attention was devoted to obtain high H2/HI selectivity, high H2 permeance, and good stability in the presence of corrosive HI gas. The effects of the deposition conditions, temperature, and period were investigated. The HTMOS membrane prepared at 450 °C for 5 min exhibited high H2/HI selectivity (>175) with H2 permeance on the order of 10−7 mol Pa−1 m−2 s−1. On the basis of stability experiments, it was found that the HTMOS membrane was stable upon HI exposure at a temperature of 400 °C for 11 h.

Techniques for partitioning cell adhesion are useful tools in biological and medical experiments. However, conventional cell patterning methods require special apparatus, special materials or high-level skills. Therefore, we have developed a new cell patterning methodology which can be easily carried out in biological laboratories. Non-cell adhesive material including hydrogel or gas patterns to restrict cell adhesion on a culture dish or glass substrates can be constructed by exploiting a polydimethylsiloxane (PDMS) mold with microchannels. The PDMS molds suck non-adhesive materials into microchannels from the inlet of the microchannels and the materials are immobilized onto the substrates with a desired pattern. High resolution under a few micrometers and long-term stability can be realized. This method has been used for analysis of stem cells, muscle cells, neuron development and other cells in collaboration with many biological researchers. Several examples to use this technique are introduced in this review.

Tumor suppressor p53 plays an integral role in DNA-damage induced apoptosis, a biological process that protects against tumor progression. Cell shape dramatically changes when cells undergo apoptosis, which is associated with actomyosin contraction, however, it remains entirely elusive how p53 regulates actomyosin contraction in response to DNA-damaging agents. To identify a novel p53 regulating gene encoding the modulator of myosin, we conducted DNA microarray analysis. We found that, in response to DNA-damaging agent doxorubicin, expression of myotonic dystrophy protein kinase (DMPK), which is known to upregulate actomyosin contraction, was increased in a p53-dependent manner. The promoter region of DMPK gene contained potential p53-binding sequences and its promoter activity was increased by overexpression of the p53 family protein p73, but, unexpectedly, not of p53. Furthermore, we found that doxorubicin treatment induced p73 expression, which was significantly attenuated by downregulation of p53. These data suggest that p53 induces expression of DMPK through upregulating p73 expression. Overexpression of DMPK promotes contraction of the actomyosin cortex, which leads to formation of membrane blebs, loss of cell adhesion, and concomitant caspase activation. Taken together, our results suggest the existence of p53-p73-DMPK axis which mediates DNA-damage induced actomyosin contraction at the cortex and concomitant cell death.

Transcription factor GLI1 is an effecter of Hedgehog (HH) signalling and activated in a broad spectrum of cancers. However, the role of the HH-GLI1 pathway in cancer and the activation mechanism of GLI1 in HH signalling after dissociation from its inhibitor, SUFU, are not fully understood. Here, we found that GLI1 associated with the methylosome protein 50 (MEP50)/protein arginine methyltransferase 5 (PRMT5) complex and was methylated. Association of MEP50/PRMT5 with GLI1 was enhanced and expression of MEP50 and PRMT5 was activated by HH signals, suggesting their role in positive feedback regulation. Methylated GLI1 lost its ability to bind ubiquitin ligase ITCH/NUMB, resulting in nuclear accumulation and activation of GLI1. Moreover, protein expression of GLI1 was enhanced by MEP50/PRMT5 and expression of MEP50, PRMT5, and GLI1 target genes was upregulated in HH-expressing cancers. These results suggest that MEP50/PRMT5 is important for HH signal-induced GLI1 activation, especially in cancers., Yoshinori Abe et al. find that the Hedgehog (HH) signalling effector GLI1 is methylated by and associates with the MEP50/PRMT5 methyltransferase complex upon HH activation. They find that this complex plays a key role in positive feedback regulation of HH signalling in cancer.

Cation-exchange membranes (CEMs) were developed using a radiation-grafted polymerization method to improve HI-mediated electro-electrodialysis (EED) in thermochemical water-splitting iodine-sulfur processes. The performance of the HI-mediated reaction was improved using the developed CEMs by introducing divinylbenzene (DVB) as the crosslinker. The effects of the crosslinked CEMs on performance indices such as proton conductivity, transport number, and water permeation factor were investigated. Introducing the crosslinks improved the selectivity of H+ and water transport. The permeation mechanism of the proposed EED model was investigated to determine the role of the crosslinkers in this operation. The results indicated that even though the crosslinkers had little effect on the absorption of the HIx solution and the H+ diffusion coefficient, a notable decrease was observed in the I− diffusion coefficient. The crosslinkers inhibited swelling caused by the absorption of the HIx solution that led to volume changes known to suppress H2O permeation.

Apprehension over exhaustion of fossil fuels and global warming, due to increasing amounts of CO2, has generated a lot of attention for the subject of renewable energy. Renewable energy has an intermittency problem and its output fluctuates depending on natural conditions. Biohydrogen is one of the promising renewable energy sources. Hydrogen produced by photosynthetic bacteria depends on the intensity of light irradiation and also fluctuates with the daily variation of sunlight. The co-culture system of dark-fermentative and photosynthetic bacteria is one solution for reducing the dependency of hydrogen production on light intensity. Because these two strains of bacteria have different processes of hydrogen production, it is possible to combine different outputs so far as the co-culture system works well. This study performed hydrogen production by the co-culture system composed of agar gels embedded with both dark-fermentative bacteria, Clostridium butyricum MIYAIRI, and photosynthetic bacteria, Rhodobacter sphaeroides RV, under a fluctuating light-irradiation. The time-course of hydrogen production was determined for the different conditions of co-culture in the mixing ratios of the two bacterial strains and light-irradiation patterns. As a result, the co-culture system succeeded in producing hydrogen exceeding that in the case of a single culture system and improved its stability against light fluctuation. Hydrogen production by the co-culture system would be applicable to the reduction of intermittency in renewable energies.

Renewable energy is regarded as a clean energy source but has some problems, one of which is intermittency. To reduce this, the time-delay of hydrogen production by photosynthetic bacteria can be effective. In this study, we qualitatively evaluated the time-delay of hydrogen production by photosynthetic bacteria under various irradiation conditions, and we also quantitatively evaluated it by fitting the experimental data and the hydrogen production model with a genetic algorithm. As a result of model fitting, we found that the relationship between the lengths of the optimized time-delay of hydrogen production by photosynthetic bacteria and the amount of light irradiation is linear. And we also found that the time-delay of hydrogen production by photosynthetic bacteria had an upper limit under low light intensity. We have suggested the existence of an energy store mechanism in photosynthetic bacteria.

A technique for partitioning cell adhesion with long-term stability is an important tool in biology and medical experiments. This study has developed a simple vapor-based immobilization method using a compound bearing a fluoro-functional group on a cell culture surface with micro/nanoscale patterns. The vapor of a silane coupling agent containing the fluoro-functional group was introduced into the micro-/nanochannels of polydimethylsiloxane elastomer placed on the target surface in a vacuum desiccator. The agent was immobilized with a pattern shown only on the areas of the channels on the surface. Although it is difficult to introduce the agent solution into a sub-micrometer-width channel via a conventional liquid-based approach, the vapor can be introduced into nanochannels with an exceptionally narrow width of 800 nm and an exceptionally low height of 700 nm. The fluoro-functional group area was clearly found to repel the adhesion of fluorescent-dye-conjugated fibronectin molecules. Four different types of cells, mouse skeletal myoblast cell line C2C12 cells, human cervix adenocarcinoma cell line HeLa cells, bovine artery endothelial cells HH, and mouse mammary gland epithelial cell line NMuMG cells, were prepared and each type was seeded onto the immobilized culture surfaces of glass and polystyrene dishes. The cells were adhered on the non-fluoro-functional group immobilized area and the cell pattern was maintained during atleast a 39 days of cultivation. This method is expected to be useful for simple and stable cell patterning in biological and medical fields.

Hydrogel patterning methods are widely used for cell patterning because they offer better long-term stability than protein patterning methods such as micro-contact printing, but conventional hydrogel patterning methods require special apparatuses such as a laser or an electron beam lithography system or they have complicated chemical operations which prevent their practical use in biological laboratories. A simple method was developed to cast a hydrogel solution without external power sources using a polydimethylsiloxane (PDMS) mold with micro-channels. This study employed “the accumulation of vacuum pressure” in a degassed lump of PDMS as a driving force for introducing agarose solution into the micro-channels. Sufficient vacuum pressure could be accumulated within 1 h in the PDMS elastomer that was acting as a vacuum tank, and 2 w/v% agarose solution could be aspirated into the micro-channels with widths from 100 to 2000 μm and a height of 19 μm, fully filling them. After the gelation and dehydration of agarose solution in the micro-channels, the patterns of agarose gel on the channels were successfully cast with a 90%-width accuracy. By using the repellency of agarose gel toward cell adhesion, patterned cultures of myoblasts and cortical neurons were successfully prepared. This technique is expected to be useful in repellency-guided cell patterning for various types of cells, with applications to cell–cell interactions and axon guidance.

We demonstrated that our previously developed gas-phase fluoroalkylsilane patterning method was applicable to polydimethylsiloxane (PDMS) and we compared the stability of patterned proteins and cultured cells between PDMS and glass surfaces. The shapes of the protein patterns were stable on both glass and PDMS surfaces for more than 1 week. The cell patterns were stable on glass surfaces for 1 week, while those on PDMS collapsed within a few days. These results indicated that our method was applicable to PDMS, although, compared with glass, PDMS has an unsolved issue for its application to long-term patterning of cells.

Three anthocyanins, four flavonols, three aromatic acids and five gallotannins were isolated from Sapria himalayana f. albovinosa in Myanmar. They were identified as cyanidin 3-O-glucoside (1), cyanidin 3-O-xyloside (2) and peonidin 3-O-glucoside (3) (anthocyanins), quercetin 3-O-glucoside (4), quercetin 7-O-glucoside (5), quercetin 3-O-glucuronide (6) and isorhamnetin 3-O-glucoside (7) (flavonols), ellagic acid (8), gallic acid (9) and ethyl gallate (10) (aromatic acids), and 1,2,4,6-tetragalloylglucose (11), 1,4,6-trigalloylglucose (12), 1,2,6-trigalloylglucose (13), 1,2,4-trigalloylglucose (14) and 1,6-digalloylglucose (15) (gallotannins) by UV, LC-MS, acid hydrolysis, NMR and/or HPLC comparisons with authentic samples. The chemical composition of S. myanmarensis was qualitatively the same with that of S. himalayana f. albovinosa. Phenolic compounds of the Rafflesiaceae species including Sapria, Rafflesia and Rhizanthes were isolated and identified in this survey for the first time.
