We discovered new B-site-ordered double perovskites Ln2LiFeO6 (Ln = La, Nd, Sm, and Eu) with most likely unusually high valence Fe5+, which was stabilized by strong oxidizing high-pressure synthesis. Despite large antiferromagnetic interactions between Fe5+ spins in these compounds, the magnetic ordering is strongly suppressed due to the geometrical frustration of Fe5+ located in a face-centered cubic lattice. In addition, canted magnetic structures are stabilized only in those with Ln = Sm and Eu, which is most likely due to significant Dzyaloshinskii Moriya interaction caused by large monoclinic structural distortion. These results provide a deep understanding of the structure-property relationships in geometrically frustrated B-site-ordered double perovskites.

Introduction Regorafenib is a multikinase inhibitor approved for the treatment of metastatic colorectal cancer (mCRC). Despite providing a statistically significant survival benefit, a substantial number of patients fail to respond to or continue with treatment, which has resulted in an unmet clinical need for a biomarker of regorafenib efficacy. Methods The JACCRO CC-12 study was a prospective, multicenter, single-arm phase II trial designed to evaluate the usefulness of [18F]fluorodeoxyglucose positron emission tomography (FDG-PET) as an imaging biomarker of regorafenib in patients with mCRC that progressed after standard chemotherapies. FDG-PET and contrast-enhanced computed tomography (CT) were performed before and after treatment with regorafenib 160 mg once daily 3 weeks on/1 week off. The primary end point was the change in the maximum standardized uptake value in the lesion with the highest uptake at pre-treatment FDG-PET. The secondary end points included overall survival (OS), progression-free survival (PFS), the objective response rate (ORR), safety, and the correlation between FDG-PET and CT. Results Twenty patients were enrolled from November 2014 to March 2016, 17 of whom were evaluated for metabolic and morphological changes. Metabolic response with FDG-PET was partial response (PR) in one case (5.9%), stable disease (SD) in four (23.5%), and progressive disease (PD) in 12 (70.6%). The metabolic response rate was 5.9%. On CT imaging, no complete response or PR was observed, and the ORR was 0%. Median PFS and OS were 1.7 and 9.8 months, respectively. The median PFS of patients who achieved PR or SD by FDG-PET was 3.7 months, whereas that of those assessed as PD was 1 month (p = 0.13). The median OS of patients who achieved PR or SD by FDG-PET was 13.0 months, whereas that of patients assessed as PD was 10.6 months (p = 0.43). Frequent adverse events were palmar–plantar erythrodysesthesia syndrome, hypertension, loss of appetite, and fatigue. Conclusions In this study, FDG-PET failed to demonstrate usefulness as an early imaging biomarker of regorafenib in patients with mCRC.

A method that separately quantifies the PSII with inactive oxygen-evolving complex (OEC) and active D1 retaining the primary quinone acceptor (QA)-reducing activity from the PSII with damaged D1 in the leaf was developed using PAM fluorometry. It is necessary to fully reduce QA to obtain Fm, the maximum fluorescence. However, QA in PSII with inactive OEC and active D1 would not be fully reduced by a saturating flash. We used the acceptor-side inhibitor DCMU to fully reduce QA. Leaves of cucumber (Cucumis sativus L.) were chilled at 4°C in dark or illuminated with UV-A to selectively inactivate OEC. After these treatments, Fv/Fm, the maximum quantum yield, in the leaves vacuum-infiltrated with DCMU were greater than those in water-infiltrated leaves. In contrast, when the leaves were illuminated by red light to photodamage D1, Fv/Fm did not differ between DCMU- and water-infiltrated leaves. These results indicate relevance of the present evaluation of the fraction of PSII with inactive OEC and active D1. Several examinations in the laboratory and glasshouse showed that PSII with inactive OEC and active D1 was only rarely observed. The present simple method would serve as a useful tool to clarify the details of the PSII photoinhibition.

Excessive accumulation of amyloid β-protein (Aβ) is one of the primary mechanisms that leads to neuronal death with phosphorylated tau in the pathogenesis of Alzheimer's disease (AD). Protofibrils, one of the high-molecular-weight Aβ oligomers (HMW-Aβo), are implicated to be important targets of disease modifying therapy of AD. We previously reported that phenolic compounds such as myricetin inhibit Aβ1-40, Aβ1-42, and α-synuclein aggregations, including their oligomerizations, which may exert protective effects against AD and Parkinson's disease. The purpose of this study was to clarify the detailed mechanism of the protective effect of myricetin against the neurotoxicity of HMW-Aβo in SH-SY5Y cells. To assess the effect of myricetin on HMW-Aβo-induced oxidative stress, we systematically examined the level of membrane oxidative damage by measuring cell membrane lipid peroxidation, membrane fluidity, and cell membrane potential, and the mitochondrial oxidative damage was evaluated by mitochondrial permeability transition (MPT), mitochondrial reactive oxygen species (ROS), and manganese-superoxide dismutase (Mn-SOD), and adenosine triphosphate (ATP) assay in SH-SY5Y cells. Myricetin has been found to increased cell viability by suppression of HMW-Aβo-induced membrane disruption in SH-SY5Y cells, as shown in reducing membrane phospholipid peroxidation and increasing membrane fluidity and membrane resistance. Myricetin has also been found to suppress HMW-Aβo-induced mitochondria dysfunction, as demonstrated in decreasing MPT, Mn-SOD, and ATP generation, raising mitochondrial membrane potential, and increasing mitochondrial-ROS generation. These results suggest that myricetin preventing HMW-Aβo-induced neurotoxicity through multiple antioxidant functions may be developed as a disease-modifying agent against AD.

A detailed chemical kinetic model of aluminum oxidation in gas phase has been constructed. Quantum chemical calculations are performed for obtaining molecular structures of intermediates on the reaction pathways. DFT level calculation is applied to search intermediates and transition states, and CBS-QB3 method is applied to calculate highly accurate potential energies. The rate coefficients of each reaction paths were also calculated based on VTST or RRKM theory for constructing the detailed chemical kinetic model. 76 elementary reactions were investigated for AlxOy intermediates, and chemical kinetics analysis was performed to find the major route of oxidation to Al8O12 as the final product of this model. The mainstream of oxidation and formation of large size AlxOy molecules were discussed on the model, and it was found that AlO, Al2O, and Al2O2 are important intermediates to form Al2O3 or Al2O4 as precursors for Al4O6.

Saccharomyces cerevisiae is a facultative anaerobic organism that grows well under both aerobic and hypoxic conditions in media containing abundant fermentable nutrients such as glucose. In order to deeply understand the physiological dependence of S. cerevisiae on aeration, we checked endoplasmic reticulum (ER)-stress status by monitoring the splicing of HAC1 mRNA, which is promoted by the ER stress-sensor protein, Ire1. HAC1-mRNA splicing that was caused by conventional ER-stressing agents, including low concentrations of dithiothreitol (DTT), was more potent in hypoxic cultures than in aerated cultures. Moreover, growth retardation was observed by adding low-dose DTT into hypoxic cultures of ire1∆ cells. Unexpectedly, aeration mitigated ER stress and DTT-induced impairment of ER oxidative protein folding even when mitochondrial respiration was halted by the ro mutation. An ER-located protein Ero1 is known to directly consume molecular oxygen to initiate the ER protein oxidation cascade, which promotes oxidative protein folding of ER client proteins. Our further study using ero1-mutant strains suggested that, in addition to mitochondrial respiration, this Ero1-medaited reaction contributes to mitigation of ER stress by molecular oxygen. Taken together, here we demonstrate a scenario in which aeration acts beneficially on S. cerevisiae cells even under fermentative conditions.

Light energy causes damage to Photosystem I (PSI) and Photosystem II (PSII). The majority of the previous photoinhibition studies have been conducted with PSII, which shows much larger photoinhibition than PSI; therefore, relatively little is known about the mechanism of PSI photoinhibition so far. A previous report showed that the photoinhibition action spectrum measured with PSI activity of isolated thylakoid is similar to the absorption spectrum of chlorophyll. However, it is known that the extent of PSI photoinhibition is much smaller in vivo compared to in vitro. It is also possible that the different extent of PSII photoinhibition, caused by different spectral light qualities, can affect the photoinhibition of PSI in vivo because PSI receives electrons from PSII. In the present research, to study the effect of light quality and the effect of the extent of PSII photoinhibition on the PSI photoinhibition in vivo, intact leaves were photoinhibited under four different light qualities. The rate coefficient of PSI photoinhibition was significantly higher in blue and red light compared to white light. The rate of PSI photoinhibition at the same photon-exposure was the largest in blue and red light and followed by white and green light. These results support the notion that light absorption by chlorophyll is responsible for the PSI photoinhibition, even in intact leaves. The variation among light colors in the relationships between the extent of photoinhibition of PSII and that of PSI indicate that PSI and PSII are independently photoinhibited with different mechanisms in the early stage of in vivo photoinhibition.

The trajectory of glomerular filtration rate (GFR) in relation to glomerular hyperfiltration (GHF) has been unknown. It was evaluated retrospectively in 23,982 GHF-free health examinees who were followed for 2−10 yr (mean: 5.1 yr). GFR was estimated by the serum creatinine concentration, and GHF was defined as age- and sex-specific estimated GFR (eGFR) ≥ 95% of the Japanese general population. The temporal profile of eGFR was plotted in a GHF-centered way, which was fitted to a random coefficient linear mixed model. Of the 23,982 subjects, 797 and 23,185 subjects developed or did not develop GHF, respectively, so that they were termed as the GHF(+)and GHF(−)groups. At baseline, median eGFR was significantly elevated in the GHF(+)group compared with in the GHF(−)group: 94.1 versus 77.3 mL/min/1.73 m2( P < 0.001). Elevation of basal eGFR lasted for a mean (SD) of 3.3 (1.9) yr in the GHF(+)group; mean eGFR then rose to the GHF range, which was 108.5 mL/min/1.73 m2. The eGFR decline after the peak was steeper in the GHF(+)group than in the GHF(−)group: −0.984 versus −0.497 mL/min/1.73 m2/yr ( P < 0.001). Baseline eGFR, but no other variable, well predicted incident GHF, with an area under the receiver operating characteristic curve of 0.87 (95% confidence interval: 0.86–0.88). In conclusion, GHF occurs as a chronic, multiphasic phenomenon: initially with a sustained GFR elevation for years, followed by a GFR surge to the GHF range, which was accompanied by accelerated GFR declining.

Protamine causes cardiac depression, which may be mediated by tumor necrosis factor alpha (TNF-α). Ulinastatin, a human urinary protease inhibitor, inhibits TNF-α. Here, we aimed to investigate whether ulinastatin prevented protamine-induced myocardial depression by inhibiting TNF-α. Rat hearts were perfused using a Langendorff system, and three protocols were followed. Protocol 1: The hearts were divided into saline, ulinastatin-low, and ulinastatin-high groups. Protamine was administered to each group, and myocardial contractility was the primary outcome. Protocol 2: The hearts were allotted to saline or ulinastatin group. Protamine was administered to each group. TNF-α expression in the coronary effluent and myocardial tissue was measured. Protocol 3: The hearts were allotted to saline and ulinastatin groups. Recombinant rat-TNF-α was administered to each group. Protamine alone reduced the maximum left ventricular pressure derivative (LV dP/dt max) by 45 ± 4%. In contrast, the reduction in LV dP/dt max was 4 ± 3% in the ulinastatin-high group. Compared with that in the saline group, the increase in TNF-α in the coronary effluent was attenuated in the ulinastatin group. Recombinant TNF-α alone reduced LV dP/dt max (- 21 ± 14%). In contrast, when TNF-α was added in the presence of ulinastatin, the decrease in LV dP/dt max was prevented significantly (- 3 ± 8%). We showed, for the first time, that ulinastatin protected against protamine-induced myocardial damage, both by inhibiting TNF-α synthesis and by directly preventing the cardiodepressant action of TNF-α.

The Buddhist sacred site of Taya Caves is a gem hidden underground in Yokohama, Japan. The caves were excavated and sculpted into bare rock by Shingon Buddhist monks from the Kamakura until the Edo period (thirteenth–nineteenth century), and dedicated to ascetic training, rituals, and pilgrimage. They are a maze of halls and galleries decorated with hundreds of rock-cut reliefs, picturing deities and masters of Buddhism, temples and shrines, real and fantastic animals, vegetal motifs, mandalas, zodiac signs, family crests, etc. The history and rock art of Taya Caves and the urge to preserve their cultural value led to this first-ever scientific investigation, dealing with the stone properties, deterioration, and environmental setting. Textural, mineralogical, geochemical, and petrophysical investigations were combined with a microclimate monitoring and chemical analyses of groundwater and rainwater. The caves are excavated into a clay-rich fossiliferous siltstone, extremely soft and porous and highly susceptible to water-driven weathering. Water represents a constant in Taya Caves, either flowing, dripping, and stagnant; or rising from the subsoil; or related to the extremely high relative humidity. Crusts and efflorescences represent important indicators of mineral dissolution and mobilization. The crusts are made of gypsum, crystallized from the dissolution of calcareous bioclasts and oxidation of pyrite, with minor calcite. The efflorescences are composed of chlorides, phosphates, sulfates, and carbonates, possibly deriving from agrochemicals and the surface vegetation cover. The salt weathering is strictly related to the microenvironmental variables and physico-chemical properties of the phases and waters involved. Rock-water interaction is particularly damaging even considering just the physical mechanisms. The stone is strongly sensitive to water absorption, hygroscopic adsorption, and slaking: the stresses generated by in-pore water and air movement and the swelling clay minerals may lead to rapid disintegration, especially during cyclic processes. This research is expected to raise concerns about the safeguard of Taya Caves and support future monitoring and conservation plans, and to foster a wider promotion and valorization of this heritage site.

The methylenation reagent 1-methylbenzimidazol-2-yl methyl sulfone 2 reacts with various aldehydes and ketones in the presence of t-BuOK (room temperature, 1 h) in dimethylformamide to give the corresponding terminal alkenes generally in high yields. For sensitive substrates, the reaction is better carried out at low temperature using sodium hexamethyldisilazide in 1,2-dimethoxyethane. The byproduct is easily removed from the products, and the reaction conditions are mild and practical. Reagent 2 can be easily prepared from commercially available 2-mercaptobenzimidazole 5 in 95% yield without any expensive reagents.

Mexoryl SX (terephthalylidene-3,3′-dicamphor-10,10′-disulfonic acid, Ecamsule) is a water-soluble UV-A absorber. The near-IR phosphorescence spectrum of singlet oxygen generated by photosensitization with Mexoryl SX was not observed in air-saturated water. On the other hand, the time-resolved near-IR phosphorescence spectrum was observed in oxygen-saturated phosphate buffer (pH 7.4). The quantum yield of the singlet oxygen generation (ΦΔ) was determined to be 0.0021 ± 0.0005. The ability of Mexoryl SX as a photosensitizer is quite low. The question arises as to the quite low ΦΔ value. No phosphorescence was detectable from Mexoryl SX in ethanol at 77 K. We elucidated the nature of the lowest excited triplet (T1) state of Mexoryl SX using a time-resolved EPR technique, because this technique is powerful for the study of short-lived and nonphosphorescent T1 molecules. The strong time-resolved EPR signals were observed. This fact shows that a considerable proportion of the lowest excited singlet (S1) state molecules undergoes intersystem crossing (ISC) to the T1 state and the deactivation process of the T1 state is mainly radiationless. The observed zero-filed splitting parameters, T1 lifetime, and S1 → T1 ISC anisotropy suggest that the T1 state can be regarded as a 3nπ*–3ππ* mixing state in character and the two unpaired electrons in the T1 state do not localize on (4-methylbenzylidene)camphor, a closely related component. Although the shorter T1 lifetime (47 ns) prevents T1 state quenching by ground-state oxygen, the 3nπ* character may contribute something to the low ΦΔ value.

Using the density functional theory, we investigated the geometric, electronic structure, phase stability and electrochemical properties of a potential P2 layer orthorhombic cathode material NaxMnO2 (0 ≤ x ≤ 1) applied for sodium-ion batteries. Herein, we shed the light on the undeniable effect of the polaron formation and polaron migration on the diffusion of Na+ ions in the orthorhombic P2 layered oxides. Both GGA+U and HSE06 methods agree that, when a Na+ ion is removed from the fully charged state of NaMnO2, the accompanying polaron preferably forms at one of the third nearest Mn (3NN) octahedra to the Na vacancy, implying the oxidization of the Mn3+ ion at one of these 3NN sites to Mn4+. The positive polaron migrates simultaneously with the Na vacancy and would hinder the diffusion of Na ions. Two kinds of elementary diffusion processes, named parallel and crossing, have been explored which required almost same activation energy of about 423 meV (518 meV) by GGA+U (HSE06). In the fully discharged state, GGA+U and HSE06 methods indicate that the negative polaron forms at one of the second nearest Mn neighbours (2NN). The activation energy of 273 meV (327 meV) is needed for diffusion in a structure with a low Na concentration, which is much lower than that required for diffusion in the Na-rich regime. Consequently, Na+ ions can diffuse easier at lower Na concentrations. With the overall activation energy of 423 meV (518 meV), this material exhibits a faster ion diffusion in comparison with the prevailing lithium-based materials such as olivine phosphate.

Acrocarpospora is a rare, recently established actinomycete genus of the family Streptosporangiaceae. In the present study, we sequenced whole genomes of the type strains of Acrocarpospora corrugate, Acrocarpospora macrocephala, and Acrocarpospora pleiomorpha to assess their potency as secondary metabolite producers; we then surveyed their nonribosomal peptide synthetase (NRPS) and polyketide synthase (PKS) gene clusters. The genome sizes of A. corrugate NBRC 13972T, A. macrocephala NBRC 16266T, and A. pleiomorpha NBRC 16267T were 9.3 Mb, 12.1 Mb, and 11.8 Mb, respectively. Each genome contained 12-17 modular NRPS and PKS gene clusters. Among the 23 kinds of NRPS and PKS gene clusters identified from the three strains, eight clusters were conserved in all the strains, six were shared between A. macrocephala and A. pleiomorpha, and the remaining nine were strain-specific. We predicted the chemical structures of the products synthesized by these gene clusters based on bioinformatic analyses. Since the chemical structures are diverse, Acrocarpospora strains are considered an attractive source of diverse nonribosomal peptide and polyketide compounds.

Daidzein, an aglycone-type isoflavone, is useful in the prevention of atherosclerotic cardiovascular diseases. However, the solubility of daidzein remains relatively low even with pharmaceutical interventions (e.g., γ-cyclodextrin inclusion complex). In the present study, daidzein-cyclodextrin-metal organic framework solid dispersion complexes were prepared by the solvent evaporation method. The physicochemical properties of the complex and its effect on the solubility of daidzein were evaluated. The enhancement effect of a cyclodextrin-metal organic framework on the antioxidant properties of daidzein was verified using a diphenyl-picrylhydrazyl radical scavenging test. Powder X-ray diffraction results showed that the characteristic diffraction peaks of daidzein and cyclodextrin-metal organic framework disappeared and new peaks (2θ = 7.1°, 16.5°) were observed. FT-IR measurements showed that the peak derived from the carbonyl group of daidzein shifted to the lower wavenumber. NOESY 1H-1H NMR showed cross peaks at the proton on the resorcinol side of daidzein and the proton (H-5, H-6) in a cyclodextrin-metal organic framework. Dissolution rate of daidzein at 5 min in distilled water was 0.06% for daidzein alone while the daidzein inclusion complex was about 100%. When fasted state simulated intestinal fluid was used, the dissolution rate of the daidzein complex was about 71% compared with that of daidzein alone (~ 3.0%) at 5 min. The daidzein inclusion complex improved the antioxidant capacity to ~ 1.3 times (17.8 µg/mL) compared to the IC50 of daidzein alone (22.9 µg/mL). Preparations of cyclodextrin-metal organic framework inclusion complexes will be a platform in developing pharmaceutical formulations to enhance the bioavailability and activity of drugs.

The mechanism of antioxidant defense system is still controversial. As islet ß-cell is weak in oxidative condition, that causes diabetes mellitus, therefore, antioxidant defense system of human pancreatic islet derived 1.1B4 cell was analyzed. Cells were exposed to H2O2 and comprehensive gene expression was analyzed by Agilent human microarray. HMOX1 and NR4A3, member of orphan receptor, were up-regulated. Therefore, NR4A3 was knocked down with siRNA, then analyzed gene expression by microarray, and found that the knocked down cells were weak in oxidative stress. HMOX1 expression was strongly inhibited by siRNA of NR4A3, and NR4A3 responsible sequence of aaggtca was found near the HMOX1 gene, suggesting NR4A3 is oxidative stress responsible transcription factor through HMOX1 expression. The expression of CCNE1 and CDK2 was also inhibited by knocked down of NR4A3, it is suggested NR4A3 is also important transcription factor for cell growth regulation.Graphical abstractHydrogen peroxide induces NR4A3 and binds to aaggtca sequence of HMOX1, and increased transcription of HMOX1. Resulting heme oxygenase produces biliverdin, antioxidants, from heme. NR4A3 also bind to aaggtca sequence of CDK2 and CCNE1, resulting CDK2 and Cyclin E. CDK2 bind to cyclin E and cell goes from G1 to S phase.

Several efforts have been made to use virtual reality (VR) and augmented reality (AR) for medical and dental education and surgical support. The current methods still require users to wear devices such as a head-mounted display (HMD) and smart glasses, which pose challenges in hygiene management and long-term use. Additionally, it is necessary to measure the user’s inter-pupillary distance and to reflect it in the device settings each time to accurately display 3D images. This setting is difficult for daily use. We developed and implemented a novel anatomy education method using a spatial reality display capable of stereoscopic viewing with the naked eye without an HMD or smart glasses. In this study, we developed two new applications: (1) a head and neck anatomy education application, which can display 3D-CG models of the skeleton and blood vessels of the head and neck region using 3D human body data available free of charge from public research institutes, and (2) a DICOM image autostereoscopic viewer, which can automatically convert 2D CT/MRI/CBCT image data into 3D-CG models. In total, 104 students at the School of Dentistry experienced and evaluated the system, and the results suggest its usefulness. A stereoscopic display without a head-mounted display is highly useful and promising for anatomy education.

The anti-inflammatory effects of linagliptin in LPS-induced inflammation in the TLR4-independent pathway have not been examined before. We examined the anti-inflammatory effects of linagliptin in the TLR4- and the LBP-independent pathway.Methods: U937 cells were cultured in the medium supplemented with 10% fetal bovine serum (FBS) and treated with 100 nM phorbol myristate acetate for 48 h. Cells were then left untreated or were treated with 10 μg/mL anti-TLR4 antibodies alone or in combination with linagliptin for 1 h in media supplemented with or without 10% FBS. The cells were divided into 5 groups: a) control cells (untreated) b) cells treated with LPS c) cells treated with 10 μg/mL anti-TLR4 antibodies d) cells treated with LPS and 10 μg/mL anti-TLR4 antibodies and e) cells treated with LPS, 10 μg/mL anti-TLR4 antibodies, and linagliptin. The LPS concentrations used were 50 pg/mL or 100 pg/mL for cells treated in the presence of 10% FBS and 100 pg/mL or 1 μg/mL for cells treated in the absence of FBS. Linagliptin concentrations of 1 nM, 10 nM, and 100 nM were used for treatment. The supernatants were analyzed for interleukin (IL)-6 production after 24 h of various treatments.Results: LPS increased IL-6 production compared to the untreated control cells, and anti-TLR4 antibody suppressed LPS-induced increased IL-6 levels. Linagliptin suppressed LPS-induced IL-6 production in a concentration-dependent manner in the presence of FBS. However, only 100 nM linagliptin could suppress LPS-induced IL-6 production in the absence of FBS.Conclusion: Concentration-dependent and -independent inflammatory suppression was observed following linagliptin treatment after LPS induction in an experimental model of TLR4 inhibition by anti-TLR4 antibodies. The physical fractionation of cytoplasmic versus nuclear components of cells is a key step for studying the subcellular localization of molecules. The application of an electric field is an emerging method for subcellular fractionation of proteins and nucleic acids from single cells. However, the multibiophysical process that involves electrical lysis of cytoplasmic membranes, electrophoresis, and diffusion of charged molecules remains unclear. Here we study RNA dynamics in single cells during the electrophoretic extraction via a microfluidic system that enables stringent fractionation of the subcellular components leveraging a focused electric field. We identified two distinct kinetics in the extraction of RNA molecules, which were respectively associated with soluble RNA and mitochondrial RNA. We show that the extraction kinetics of soluble RNA is dominated by electrophoresis over diffusion and has a time constant of 0.15 s. Interestingly, the extraction of mitochondrial RNA showed unexpected heterogeneity in the extraction with slower kinetics (3.8 s), while reproducibly resulting in the extraction of 98.9% ± 2% after 40 s. Together, we uncover that the microfluidic system uniquely offers length bias-free fractionation of RNA molecules for quantitative analysis of correlations among subcellular compartments by exploiting the homogeneous electrophoretic properties of RNA.

We report the lithium ionic conductivities of closo‑type complex hydrides synthesized from various molar ratios of lithium borohydride (LiBH4) and decaborane (B10H14) as starting materials. The prepared closo‑type complex hydrides comprised [B12H12]2−, [B11H11]2−, and [B10H10]2− complex anions. In addition, increasing the LiBH4 content in the starting materials increased the amounts of [B11H11]2− and [B10H10]2−, leading to an improved ion conductivity of the prepared sample. The present study offers useful insights into strategies for controlling the complex anion composition in emerging solid electrolytes of closo-type complex hydrides at the molecular level, and improving their ionic conductivities.

Purpose Dipeptidyl peptidase-4 inhibitors, including linagliptin, prevent inflammation. However, the in vitro effects of linagliptin are unclear. Moreover, although linagliptin inhibits lipopolysaccharide (LPS)-induced inflammation, the anti-inflammatory effects of linagliptin in this context are not concentration-dependent. In the absence of LPS-binding protein (LBP), the pro-inflammatory effects of LPS involve pathways other than the Toll-like receptor (TLR) 4 pathway. Here, we aimed to determine the anti-inflammatory mechanisms of linagliptin in an experimental model in which LBP was added to the medium. Methods Human U937 monocytes were cultured at 1 × 106 cells/mL in Roswell Park Memorial Institute medium and differentiated into macrophages using phorbol myristate acetate. All processes were carried out in medium containing 10% fetal bovine serum (FBS). After 48 hrs of culture, we replaced the medium and pretreated the cells with 100, 250, 500, or 2500 nM linagliptin for 1 hr. We exchanged the medium again, and the cells were treated with 1 ng/mL LPS with or without 100, 250, 500, or 2500 nM linagliptin. Interleukin (IL)-6 and LBP in the supernatant, nuclear factor (NF)-κB/p65 in the nucleus, and reactive oxygen species (ROS) in the cells, as important markers of the mechanism of inflammation induction by LPS, were measured using enzyme-linked immunosorbent assay kits. Results Linagliptin significantly prevented LPS-stimulated IL-6 production and intranuclear NF-κB/p65 levels in a concentration-dependent manner. LPS-induced intracellular ROS levels were significantly decreased by linagliptin at all concentrations. LBP levels were markedly higher in FBS-containing medium than in medium without FBS. However, LBP levels did not change following administration of linagliptin and/or LPS. Conclusion Concentration-dependent and -independent inflammatory suppression was observed following linagliptin treatment in the context of LPS-induced pro-inflammatory responses. Thus, our findings suggested that linagliptin induced two different mechanisms to repress inflammation, i.e., TLR4-dependent and -independent mechanisms.

Redox affected pumice tuff was used to investigate the changes of pore properties due to oxidation and its possible consequences to adsorption of nuclides on it. Porosity and pore size distribution were analyzed by mercury injection technique and scanning electron microscopy. Batch experiment was carried out with 1 cm3 block shaped intact pumice tuff samples to determine adsorption coefficient, Kd at several pH, ionic strength and initial nuclide concentrations of stable cesium (133Cs). Pore area, surface area and specific surface area were determined from pore size distribution data before and after adsorption experiment and later compared between fresh and oxidized samples. The results showed that reduction of less than 0.33 μm size micro pores due to oxidation is mainly responsible for reducing surface areas, eventually reducing the sorption quantity. Influence of pore properties became negligible in redox affected pumice tuff indicating inverse relation of oxidation phenomena with adsorption coefficients values. Immunomodulatory function of all-trans retinoic acid (ATRA) has been gathering much attention for the therapy of autoimmune diseases. ATRA is a chemically unstable molecule which requires proper formulation for targeted delivery. Here we examined nanostructured lipid carrier (NLC) for the formulation of ATRA. NLC is a representative nanoparticle formulation especially suited for oral delivery. We established the preparation procedures of ATRA-containing NLC (NLC-RA) which minimizes the degradation of ATRA during the preparation process. NLC-RA thus obtained was taken up by macrophages and induced anti-inflammatory response via suppressing NF-κB signaling as well as via enhancing the production of anti-inflammatory cytokines. Moreover, NLC-RA enhanced differentiation of naïve T cells to regulatory T cells in the co-culture system with dendritic cells. These results suggest that NLC-RA is a promising alternative therapy for the autoimmune diseases especially intestinal bowel disease.

Salt weathering can cause substantial deterioration of natural rocks, building stones, masonry materials, monuments, and engineering structures. Nearly two centuries of salt weathering studies, both theoretically and empirically, have manifested its power as well as its complexity. This paper attempts to unite the kinds of literature assess the various theories in the light of the combined information. The theoretical approaches concerning the most cited mechanisms of salt weathering such as crystallization, hydration and thermal expansion of crystalline salts are thoroughly reviewed. It is understood that there is no universally acceptable hard and sound theoretical information on this topic yet. More precise theories should be developed to elucidate the complications of the mechanisms of salt weathering as well as to interpret the results of empirical studies.

We have previously shown that Rab34 is an important regulator of ciliogenesis and that its unique long N-terminal region (amino acids 1-49) is essential for ciliogenesis in certain cultured mammalian cells. In the present study, we performed an in-depth deletion analysis of the N-terminal region of Rab34 together with Ala-based site-directed mutagenesis to identify the essential amino acids that are required for serum-starvation-induced ciliogenesis in hTERT-RPE1 cells. The results showed that a Rab34 mutant lacking an N-terminal 18 amino acids and a Rab34 mutant carrying an LPQ-to-AAA mutation (amino acids 16-18) failed to rescue a Rab34-KO phenotype (i.e., defect in ciliogenesis). Background In continuous renal replacement therapy (CRRT), administration of anticoagulants is necessary for achieving a certain level of filter lifetime. Generally, anticoagulant doses are controlled to keep activated partial thromboplastin time and other indicators within a certain target range, regardless of the membrane materials used for the filter. However, in actual clinical practice, the filter lifetime varies significantly depending on the membrane material used. The objective of this study was to demonstrate that the minimum anticoagulant dose necessary for prolonging the filter lifetime while reducing the risk of hemorrhagic complications varies depending on the type of membrane. Methods In three beagles, hemodiafiltration was performed with hemofilters using polysulfone (PS), polymethylmethacrylate (PMMA), and AN69ST membranes separately. The minimum dose of nafamostat mesylate (NM) that would allow for 6 h of hemodiafiltration (required dose) was investigated for each membrane material. Results The NM doses required for 6 h of hemodiafiltration were 2 mg/kg/h for the PS membrane, 6 mg/kg/h for the PMMA membrane, and 6 mg/kg/h for the AN69ST membrane. Conclusion For hemodiafiltration performed in beagles, the required NM dose varied for each filter membrane material. Using the optimal anticoagulant dose for each membrane material would allow for safer CRRT performance.

Salt weathering is one of the most damaging processes affecting stone conservation and represents an extensively debated topic in heritage science. The main subject of previous research is the built heritage, whereas other types of assets are often neglected, such as underground cultural heritage. This paper introduces an investigation of stone decay in the subterranean environment focused on salt weathering, its patterns, compositional features, and variability in time and space, aiming at finding its driving forces in the rock properties and environmental setting. Field explorations and the mineralogical analysis of salt efflorescences and crusts were integrated by the petrographic, geochemical, and petrophysical characterization of the rock substrates, a microclimate monitoring, and the chemical analysis of groundwater and rainwater. The subject of study is the underground archaeological-historical site of Yoshimi Hyaku Ana in Japan, a complex of Kofun tombs of the 6th–7th century and galleries of the WWII era. The site is affected by intense salt weathering, involving efflorescences and crusts composed of mixed soluble sulfates, mostly hydrated: gypsum, alunogen, alum-(Na), halotrichite, epsomite, polyhalite, tamarugite, thenardite, and mirabilite. They derive principally from the dissolution of rock-forming minerals and components (pyrite, glass, feldspars, etc.) from the Miocene volcanic tuffs into which the site is excavated. The tuffs show a certain lithological diversity (e.g., glass amount and chemico-mineralogical composition) that controls the space variability of salt composition. Another major influencing factor is the underground microclimate, which also affects the time variability and seasonality of salt weathering. The innermost underground areas have an extremely high relative humidity (~100%) and are essentially salt-free, whereas, nearby the site entrances, the wider fluctuations of air temperature and humidity create conditions for salt crystallization in the dry winter season and deliquescence in summer. Depending on the solubility of each salt phase, cycles of crystallization/dissolution and hydration/dehydration can occur both seasonally and in the short-term, causing severe stresses to the stone and damage.

BACKGROUND: The efficacy and safety of benralizumab, an interleukin-5 receptor alpha-directed cytolytic monoclonal antibody, for the prevention of exacerbations in patients with moderate to very severe chronic obstructive pulmonary disease (COPD) are not known.METHODS: In the GALATHEA and TERRANOVA trials, we enrolled patients with COPD (at a ratio of approximately 2:1 on the basis of eosinophil count [≥220 per cubic millimeter vs. RESULTS: In GALATHEA, the estimates of the annualized exacerbation rate were 1.19 per year (95% confidence interval [CI], 1.04 to 1.36) in the 30-mg benralizumab group, 1.03 per year (95% CI, 0.90 to 1.19) in the 100-mg benralizumab group, and 1.24 per year (95% CI, 1.08 to 1.42) in the placebo group; the rate ratio as compared with placebo was 0.96 for 30 mg of benralizumab (P = 0.65) and 0.83 for 100 mg of benralizumab (P = 0.05). In TERRANOVA, the estimates of the annualized exacerbation rate for 10 mg, 30 mg, and 100 mg of benralizumab and for placebo were 0.99 per year (95% CI, 0.87 to 1.13), 1.21 per year (95% CI, 1.08 to 1.37), 1.09 per year (95% CI, 0.96 to 1.23), and 1.17 per year (95% CI, 1.04 to 1.32), respectively; the corresponding rate ratios were 0.85 (P = 0.06), 1.04 (P = 0.66), and 0.93 (P = 0.40). At 56 weeks, none of the annualized COPD exacerbation rate ratios for any dose of benralizumab as compared with placebo reached significance in either trial. Types and frequencies of adverse events were similar with benralizumab and placebo.CONCLUSIONS: Add-on benralizumab was not associated with a lower annualized rate of COPD exacerbations than placebo among patients with moderate to very severe COPD, a history of frequent moderate or severe exacerbations, and blood eosinophil counts of 220 per cubic millimeter or greater (Funded by AstraZeneca [GALATHEA and TERRANOVA] and Kyowa Hakko Kirin [GALATHEA]; GALATHEA and TERRANOVA ClinicalTrials.gov numbers, NCT02138916 and NCT02155660.).

Primary cilia form by two pathways: an extracellular pathway in which the cilium grows out from the cell surface and an intracellular pathway in which the nascent cilium forms inside the cell. Here we identify the GTPase Rab34 as a selective mediator of intracellular ciliogenesis. We find that Rab34 is required for formation of the ciliary vesicle at the mother centriole and that Rab34 marks the ciliary sheath, a unique sub-domain of assembling intracellular cilia. Rab34 activity is modulated by divergent residues within its GTPase domain, and ciliogenesis requires GTP binding and turnover by Rab34. Because Rab34 is found on assembly intermediates that are unique to intracellular ciliogenesis, we tested its role in the extracellular pathway used by MDCK cells. Consistent with Rab34 acting specifically in the intracellular pathway, MDCK cells ciliate independently of Rab34 and paralog Rab36. Together, these findings reveal a new context-specific molecular requirement for ciliary membrane biogenesis.

The purpose of this study was to prepare solid dispersions of triamterene (TRT) with ascorbic acid (AA) or ascorbic acid 2 glucoside (AA2G) and to evaluate their physical properties. Solid dispersions were prepared by dissolving each sample in an organic solvent and evaporation (EVP). Powder X-ray diffraction (PXRD) revealed a halo pattern for EVP1 (AA/TRT = 1/1) and EVP2 (AA2G/TRT = 1/1). In differential scanning calorimetry (DSC), endothermic peaks due to the melting of TRT and AA disappeared for EVP1 (AA/TRT = 1/1), and the melting peaks of TRT and AA2G disappeared for EVP2 (AA2G/TRT = 1/1). Fourier transform infrared (FT-IR) spectroscopy revealed broadened peaks for EVP1 (AA/TRT = 1/1) and EVP2 (AA2G/TRT = 1/1) due to the hydroxyl groups (-OH) of AA and the amino groups (-NH2) of TRT and also revealed a peak shift due to the pteridine skeleton (C = N) of TRT. In near-infrared absorption (NIR) spectroscopy, peaks due to the hydroxyl groups (-OH) of AA and AA2G were found for EVP1 (AA/TRT = 1/1) and EVP2 (AA2G/TRT = 1/1), respectively. A peak due to the amino groups (-NH2) was evident. This suggested the formation of an evaporation, in which TRT interacted with AA or AA2G. In the dissolution test, the dissolved fraction of TRT alone after 3 min was 30%, whereas the fractions were enhanced to approximately 90% for EVP1 (AA/TRT = 1/1) and EVP2 (AA2G/TRT= 1/1). Results confirmed that dissolution properties were improved as a result of complex formation. The above findings indicated improvement the dissolution properties of TRT.

One of the most popular and intensively extracted building stones in Japan is a Miocene dacite-rhyolite ignimbrite known as Oya-ishi, quarried nearby the city of Utsunomiya (Tochigi). Traces of its historical usage in the last 1,500 years survive in vernacular architecture, construction, rock-cut and relief sculpture, but large-scale exploitation commenced only in the Edo period (i.e., from the 17th century), an epoch of economic growth and flourishing arts and culture. Among the over 200 underground quarries in the region, few are still active, the others abandoned or converted into geoheritage and tourist attractions (e.g., History Museum, Heiwa Kannon monument, Keikan Park). Salt weathering is one of the decay aspects of Oya stone jeopardizing the preservation of those sites of historical and geological interest and, indirectly, visitor safety. The efflorescences on the tuff quarry walls turn out to be composed of sulfates, namely gypsum, mirabilite, and thenardite, their crystallization being controlled by the relevant microenvironmental conditions. In the extremely humid underground spaces, the phases having a very high deliquescence relative humidity are stable: gypsum is essentially ubiquitous, even in the deepest quarry levels, the most environmentally isolated; mirabilite needs a slightly dryer environment, so is observable in the middle levels or semi-underground quarries; thenardite requires further dryer conditions, and is mainly detected in the open air. The mechanisms of formation of these efflorescences are still under investigation: the classic minero-petrographic and geochemical characterization of the rock and its weathering phases is being supported by a microclimatic monitoring in different sites and seasons, and the chemical analysis of rainwater and groundwater. The research direction is aimed at the identification of the environmental and lithological constraints on the salt weathering of Oya tuff, that is: the spatial and temporal variability of relative humidity, and its influence on the cycles of salt crystallization/dissolution and the resulting mechanical stresses; the chemical driving forces, related to the rock mineralogy (zeolites, feldspar alteration, etc.) and water quality.

We obtain a quantum dimer model (QDM) containing a Rokhsar-Kivelson (RK) point expressed by spin-1/2 Heisenberg antiferromagnets on a diamond-like decorated square lattice. This lattice has macroscopically degenerated nonmagnetic ground states, which are equivalent to the Hilbert space of a square-lattice QDM. Then, a square-lattice QDM containing the RK point as a second-order effective Hamiltonian is obtained by introducing further neighbor couplings as perturbation. Objectives: The deficit of Glyoxalase I (Glo1) and the subsequent increase in methylglyoxal (MG) has been reported to be one the five mechanisms by which hyperglycemia causes diabetic late complications. Aldo-keto reductases (AKR) have been shown to metabolize MG; however, the relative contribution of this superfamily to the detoxification of MG in vivo, particularly within the diabetic state, remains unknown. Methods: CRISPR/Cas9-mediated genome editing was used to generate a Glo1 knock-out (Glo1−/−) mouse line. Streptozotocin was then applied to investigate metabolic changes under hyperglycemic conditions. Results: Glo1−/− mice were viable and showed no elevated MG or MG-H1 levels under hyperglycemic conditions. It was subsequently found that the enzymatic efficiency of various oxidoreductases in the liver and kidney towards MG were increased in the Glo1−/− mice. The functional relevance of this was supported by the altered distribution of alternative detoxification products. Furthermore, it was shown that MG-dependent AKR activity is a potentially clinical relevant pathway in human patients suffering from diabetes. Conclusions: These data suggest that in the absence of GLO1, AKR can effectively compensate to prevent the accumulation of MG. The combination of metabolic, enzymatic, and genetic factors, therefore, may provide a better means of identifying patients who are at risk for the development of late complications caused by elevated levels of MG. Novel transgenic Eucalyptus camaldulensis trees expressing the bacterial choline oxidase A (codA) gene by the Cauliflower mosaic virus (CaMV) 35S promoter and the Arabidopsis thaliana heat shock protein (HSP) terminator was developed. To evaluate the codA transcription level and the metabolic products and abiotic stress tolerance of the transgenic trees, a six-month semi-confined screen house cultivation trial was conducted under a moderate-stringency salt-stress condition. The transcription level of the CaMV 35S promoter driven-codA was more than fourfold higher, and the content of glycine betaine, the metabolic product of codA, was twofold higher, with the HSP terminator than with the nopaline synthase (NOS) terminator. Moreover, the screen house cultivation revealed that the growth of transgenic trees under the salt stress condition was alleviated in correlation with the glycine betaine concentration. These results suggest that the enhancement of codA transcription by the HSP terminator increased the abiotic stress tolerance of Eucalyptus plantation trees.

Diethylhexyl 2,6-naphthalate (DEHN, tradename Corapan TQ) is used as a photostabilizer for a widely-used UV-A absorber, butylmethoxydibenzoylmethane (BMDBM). The photophysical properties of DEHN have been studied through measurements of the transient absorption, fluorescence, phosphorescence and EPR spectra in ethanol. The energy level of the lowest excited singlet state of DEHN is higher than that of BMDBM (enol form). The singlet-singlet energy transfer from BMDBM to DEHN is energetically unfavourable. The energy level of the lowest excited triplet (T1) state of DEHN is lower than that of BMDBM. It is probable that the mechanism of photostabilization of BMDBM by DEHN is the triplet-triplet energy transfer from BMDBM to DEHN. The observed phosphorescence spectrum, T1 lifetime and zero-field splitting parameters indicate that the T1 state of DEHN can be regarded mainly as a locally excited 3ππ* state within naphthalene. The sum of the quantum yields of fluorescence of DEHN (0.59 ± 0.09) and singlet oxygen generation by DEHN (0.44 ± 0.04) is almost unity in ethanol. About 60% of the UV energy absorbed by DEHN is released as fluorescence, while about 40% is used to produce the excited triplet state. The photoexcited states of 2-naphthoic acid have been studied for comparison.

Prognosis of patients with localized nasal extranodal natural killer/T-cell lymphoma, nasal type (ENKL) has been improved by non-anthracycline-containing treatments such as concurrent chemoradiotherapy (CCRT). However, some patients experience early disease progression. To clarify the clinical features and outcomes of these patients, data from 165 patients with localized nasal ENKL who were diagnosed between 2000 and 2013 at 31 institutes in Japan and who received radiotherapy with dexamethasone, etoposide, ifosfamide, and carboplatin (RT-DeVIC) were retrospectively analyzed. Progression of disease within 2 years after diagnosis (POD24) was used as the definition of early progression. An independent dataset of 60 patients with localized nasal ENKL who received CCRT at Samsung Medical Center was used in the validation analysis. POD24 was documented in 23% of patients who received RT-DeVIC and in 25% of patients in the validation cohort. Overall survival (OS) from risk-defining events of the POD24 group was inferior to that of the reference group in both cohorts (P < .00001). In the RT-DeVIC cohort, pretreatment elevated levels of serum soluble interleukin-2 receptor (sIL-2R), lactate dehydrogenase, C-reactive protein, and detectable Epstein-Barr virus DNA in peripheral blood were associated with POD24. In the validation cohort, no pretreatment clinical factor associated with POD24 was identified. Our study indicates that POD24 is a strong indicator of survival in localized ENKL, despite the different CCRT regimens adopted. In the treatment of localized nasal ENKL, POD24 is useful for identifying patients who have unmet medical needs.

We have fabricated bismuth (Bi) ultrathin films on a charge-density-wave (CDW) compound 1T-TaS2 and elucidated electronic states by angle-resolved photoemission spectroscopy and first-principles band-structure calculations. We found that the Bi film on 1T-TaS2 undergoes a structural transition from (111) to (110) upon reducing the film thickness, accompanied by a drastic change in the energy band structure. We also revealed that while two-bilayer-thick Bi(110) film on Si(111) is characterized by a dispersive band touching the Fermi level (EF), the energy band of the same film on 1T-TaS2 exhibits holelike dispersion with a finite energy gap at EF. We discuss the origin of such intriguing differences in terms of the CDW proximity effect.

The fluid volume balance between intracellular water (ICW) and extracellular water (ECW) gradually changes with age and various medical conditions. Comprehension of these physiological changes would aid in clinical decision-making related to body fluid assessments. A total of 1,992 individuals (753 men and 1,239 women) aged ≥15 yr included in this study had their body composition measurements performed at training gyms in 2014. We developed a regression formula to assess the association of age with the ratio of ECW to ICW in these subjects. The mean ages of male and female subjects were 51.2 ± 15.2 and 57.4 ± 15.2 yr, and their mean body mass indexes were 23.4 ± 3.3 and 21.1 ± 2.8 kg/m2, respectively. The total fluid volumes of male and female subjects were 39.6 ± 4.9 and 27.7 ± 3.0 liters, whereas the percent body fat mass per kilogram of body weight were 19 and 26%, respectively. The ECW-to-ICW ratio increased with age because of the steeper decrease in the ICW content than in the ECW content, especially after the age of 70 yr. The regression formulas used for calculating the age-adjusted ECW/ICW ratio were as follows: 0.5857 + 7.4334 × 10−6 × (age)2 in men and 0.6062 + 5.5775 × 10−6 × (age)2 in women. In conclusion, the fluid imbalance between ICW and ECW contents is driven by decreased cell volume associated with aging and muscle attenuation. Therefore, our proposed formula may serve as a useful assessment tool for the calculation of body fluid composition.

closo-type complex hydrides contain large cage-type complex polyanions in their crystal structures and thus can exhibit superior ion-conducting properties (e.g., Li and Na). However, the unique structures of complex polyanions have made it challenging to modify crystal structures, making systematic control of ion conductivity difficult. Here, we report an atom deficiency approach to enhance lithium-ion conductivity of complex hydrides. We find that lithium and hydrogen could be simultaneously extracted from Li2B12H12 by applying a small external energy, enabling the formation of atom deficiencies. These atom deficiencies lead to an increase in carrier concentration, improving lithium-ion conductivity by 3 orders of magnitude compared to that of a pristine material. Okayama University-type retinal prosthesis (OURePTM) is a photoelectric dye-coupled polyethylene film which generates electric potential in response to light and stimulates nearby neurons. This study aims to test surgical feasibility for subretinal film implantation and to examine functional durability of films in subretinal space. Dye-coupled films were implanted subretinally by vitrectomy in the right eye of normal white rabbits: 8 rabbits for 1 month and 8 rabbits for 6 months. The implanted films were removed by vitrectomy in 4 of these 8 rabbits in 1-month or 6-month implantation group. The films were also implanted in 4 rhodopsin-transgenic retinal dystrophic rabbits. Visual evoked potential was measured before film implantation as well as 1 or 6 months after film implantation, or 1 month after film removal. The films were successfully implanted in subretinal space of retinal detachment induced by subretinal fluid injection with a 38G polyimide tip. The retina was reattached by fluid-air exchange in vitreous cavity, retinal laser coagulation, and silicone oil injection. The ratios of P2 amplitudes of visual evoked potential in the implanted right eye over control left eye did not show significant changes between pre-implantation and post-implantation or post-removal (paired t-test). In Kelvin probe measurements, 4 pieces each of removed films which were implanted for 1 or 6 months showed proportional increase of surface electric potential in response to increasing light intensity. The film implantation was safe and implanted films were capable of responding to light.
