The establishment of reliable age in the lake sediment profile mainly depends on the AMS 14C dating technique. However, the presence of the 14C lake reservoir effects (LREs) restricted for using radiocarbon dating in lake sediment, especially in dry and cold areas with a scarce plant cover in the Qinghai-Tibet Plateau. Hence, the discussion of influence factors of LREs is crucial. This paper selected 15 lakes (17 sediment and 3 plant samples) in the Qinghai-Tibet Plateau to examine the distribution characteristics of the modern LREs and their main influencing factors. In our study area, 14 lakes were all affected by the LREs. The minimum 14C year is 5900 a BP towards the deep water area, whereas the maximum 14C year is up to 7185 a BP in the margins of Lake Heihai. The maximum 14C year is up to 7750 a BP, and the minimum 14C year is present-day carbon in the 15 lakes. One further study indicated that the LRE differences in individual lake are mostly owing to the contribution of exogenous carbonate. The results displayed that the LREs tended to increase with the increase of the salinity, moreover, the LREs of saltwater lakes or salt lakes were significantly larger than freshwater lakes due to the possible supply of old total dissolved inorganic carbon with a long residence time in the lakes. Moreover, the contribution of calcite played a significant role on the LREs. Additionally, the LREs differences are affected by the source of organic matter. The lake with groundwater supply shows large LRE due to likely being influenced by crustal and ancient CO2 uprising.

Induced water hyacinth with purple roots (PRWH) exerts a significant inhibitory effect on the growth of blue-green algae. Interestingly, its chemical constituents differ from those of wild-type water hyacinth and have not yet been reported. This study aimed to explore the chemical constituents of PRWH and its bioactive components serving as allelopathic agents against blue-green algae. Phytochemical investigation of the bioactive ethyl acetate fraction of a crude methanol extract from PRWH led to the isolation of 56 compounds, including 11 new phenylphenalene derivatives. The structures of these compounds were elucidated by comprehensive analyses through NMR, HRMS, and X-ray techniques. Bioactivity evaluation against Microcystis aeruginosa indicated that compounds 7, 12, 15, 37, 39, 45, and 47 potently inhibited blue-green algae growth.

Background Spinal cord injury (SCI) is the most serious complication of spinal injury, often leading to severe dysfunction of the limbs below the injured segment. Conventional therapy approaches are becoming less and less effective, and gene therapy is a new research direction by now. Methods The Sprague-Dawley rats were haphazardly assigned to two groups, namely sham group and SCI model group, and lncRNA H19 and miR-370-3p levels were investigated using reverse transcription-quantitative polymerase chain reaction (RT-qPCR). Correlation between lncRNA H19 and miR-370-3p was ascertained by luciferase report assay and RT-qPCR. After transfection with si-H19, miR-370-3p inhibitor, negative controls (NC), or both, primary spinal neurons were subjected to the simulation of lipopolysaccharide (LPS) for inducing in vitro model of SCI. Cell viability, apoptotic rate, caspase-3 activity, Bax and Bcl-2 protein, ROS generation, TNF-α, IL-1β, and IL-6 protein, as well as IκBα and p65 phosphorylation ratio were evaluated adopting 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), apoptosis, caspase-3 activity, ROS generation, and western blot assays, thereby searching for the specific action mechanism on LPS-induced spinal never injury. Results SCI resulted in lncRNA H19 higher expression and miR-370-3p lower expression. LPS simulation raised a series of cellular biological changes, such as decreased viability, promoted apoptosis, generated ROS, and released inflammatory factors. lncRNA H19 inhibition reversed above LPS-induced changes. Besides, as the downstream target of lncRNA H19, miR-370-3p was oppositely regulated by lncRNA H19. The above biological changes induced by lncRNA H19 inhibition were reversed by miR-370-3p upregulation. Moreover, lncRNA H19 inhibition could block NF-κB pathway through miR-370-3p upregulation. Conclusion Inhibition of lncRNA H19/miR-370-3p mitigated spinal neuron apoptosis in an in vitro model of SCI. This provided the possibility for clinical use of gene therapy.

Donkey in China is well known for its draft purpose and transportation; however, donkey meat has attracted more and more consumers in recent years, yet it lacks sufficient information on its flavor components compared to other main meats. Therefore, in this study, volatile flavor compounds in neck meat of donkey, swine, bovine, and sheep were classified by electronic nose, then confirmed and quantified by gas chromatography-mass spectrometry. High-performance liquid chromatography (HPLC) and gas chromatography were used to quantify free fatty acid, amino acid, and flavor nucleotide. A total of 73 volatile compounds were identified, and aldehydes were identified as the characteristic flavor compounds in neck meat of donkey, bovine, swine and sheep in proportion of 76.39%, 46.62%, 31.64%, and 35.83%, respectively. Particularly, hexanal was the most abundant volatile flavor. Compared with other neck meat, much higher unsaturated free fatty acids were present in donkeys. Furthermore, neck meat of donkeys showed essential amino acid with highest content. Thus, special flavor and nutrition in donkey neck meat make it probably a candidate for consumers in other regions besides Asia.

The efficient separation of hexane isomers from the light naphtha fraction is a significant challenge in the petrochemical industry. 5A zeolite adsorbent is used commercially to sieve alkane isomers. In this study, 5A zeolites were synthesized using a low-cost natural clay mineral precursor, i.e. palygorskite (PAL), with the addition of crystallization directing agent (CDA). By varying the mass ratio of CDA/deionized water, 5A zeolites were obtained as CDA-5%, CDA-7.5%, and CDA-10%. All products were submicron particles with an average particle size of 400–800 nm. A sieving test of CDA-induced 5A zeolites was carried out on hexane adsorbates including n-hexane (nHEX), 2-methylpentane (2MP), and 3-methylpentane (3MP). According to vapor-phase batch adsorption experiments, a significant equilibrium amount (0.149 g/g) of nHEX and only 0.0321 g/g 2MP and 0.0416 g/g 3MP were adsorbed on the 5A zeolite product with CDA-5%. The dynamic adsorption performance of 5A zeolite (CDA-5%) was evaluated by breakthrough curves of binary mixtures of nHEX/2MP and nHEX/3MP. Palygorskite 5A (PAL 5A) zeolite achieved maximum dynamic adsorption capacities of nHEX (0.16 g/g in both cases) at 200°C and 1.2 MPa total pressure. This work provided an economic alternative for the synthesis of 5A zeolites using natural clay minerals instead of chemical raw materials.

Gastric cancer (GC) is one of the most common malignancies worldwide. The prognosis of GC is unsatisfied owning to widespread metastasis. P21-activated kinase 1 (PAK1), a member of serine/threonine kinases, is associated with the progression of multiple types of human cancers. Here, we demonstrated that CDK4/6 inhibitor reduced GC cell viability and decreased PAK1 expression. Consistently, PAK1 ablation increased GC cell sensitivity exposed to CDK4/6 inhibitor and promoted DNA damage. We also revealed PAK1 depletion notably affected PDK1-AKT pathway, and PDK1 overexpression totally abrogated the effect of PAK1 deletion on DNA damage in GC cells. Additionally, PDK1 overexpression also rescued the increased GC cell sensitivity towards CDK4/6 inhibitor and the cell cycle arrest caused by PAK1 depletion. Our findings, therefore, suggested that PAK1 silencing increased sensitivity to CDK4/6 inhibition in gastric cancer cells via PDK1-AKT pathway. We, therefore, thought PAK1 as a promising therapeutic target for the treatment of CDK4/6 inhibitor-resistant gastric cancer.

A series of carbazole substituted spiro[fluorene-9,9′-xanthene] (SFX) derivatives, namely SFX-2-Cz, SFX-2′-Cz, SFX-3′-Cz, SFX-2,7-DCz, SFX-2′,7′-DCz and SFX-3′,6′-DCz, were designed and prepared as hosts for phosphorescent organic light emitting diodes (PhOLEDs). All the target compounds exhibited excellent thermal stability, especially for the substitution onto the 3′,6′-position of SFX (SFX-3′,6′-DCz, up to 500 °C). The results of low temperature phosphorescence show that the triplet energy level (T1) will be reduced when the carbazole group is attached to the fluorene unit of SFX; however, the T1 is nearly intact when carbazoles were introduced into the xanthene moiety of SFX. In the following investigation of blue, green, and red PhOLEDs based on these SFX-carbazole hosts, all the host materials could realize effective energy transfer to dopant phosphors under electric field excitation. For the blue PhOLEDs, the SFX-3′,6′-DCz-based device reached the maximum current efficiency (CE) of 9.7 cd A−1, power efficiency (PE) of 6.4 lm W−1 and external quantum efficiency (EQE) of 5.1% due to high T1. However, the SFX-2′-Cz-based green PhOLED and the SFX-2,7-DCz-based red PhOLED show better performance of 28.2 cd A−1, 24.7 lm W−1, 8.5% and 17.4 cd A−1, 15.6 lm W−1, 10.1%, respectively, attributable to their well-matched frontier molecular orbital energy levels with the corresponding phosphorescent dopants. This work provides a systematic scheme for the rational design of efficient SFX-based hosts by carbazolyl functionalization.

Three polysaccharides (WZP1, WZP2, WZP3) and their Se-enriched products (SeWZP1, SeWZP2 and SeWZP3) were obtained from Pleurotus ostreatus using a simple, rapid method and HNO3-Na2SeO3 method, respectively. The molecular weight distribution profiles of all samples except SeWZP2 showed double peaks. The average molecular weights (Mw) of WZP1–3 were 48.6 kDa, 20.2 kDa and 11.8 kDa, respectively, and of SeWZP1–3 were 19.6 kDa, 37.7 kDa, 14.5 kDa, respectively. The complexity of monosaccharide composition of WZP1–3 was inversely proportional to the ethanol concentration used in the ethanol precipitation process. Additionally, the results of biological activity tests indicated that α-glucosidase inhibitory activity of WZP1–3 was related to the molecular weight and the monosaccharide composition complexity. The selenized modification can improve the α-glucosidase-inhibiting, hydroxyl radical-scavenging activity of P. ostreatus polysaccharides. Therefore, by improving their bioactivities by selenization, the polysaccharides of P. ostreatus could be utilized as a natural health food supplement.

Glaucoma is characterized by retinal ganglion cell (RGC) degeneration and is the second leading cause of blindness worldwide. However, current treatments such as eye drop or surgery have limitations and do not target the loss of RGC. Regenerative therapy using embryonic stem cells (ESCs) holds a promising option, but ethical concern hinders clinical applications on human subjects. In this study, we employed spermatogonial stem cells (SSCs) as an alternative source of ESCs for cell-based regenerative therapy in mouse glaucoma model. We generated functional RGCs from SSCs with a two-step protocol without applying viral transfection or chemical induction. SSCs were first dedifferentiated to embryonic stem-like cells (SSC-ESCs) that resemble ESCs in morphology, gene expression signatures, and stem cell properties. The SSC-ESCs then differentiated toward retinal lineages. We showed SSC-ESC-derived retinal cells expressed RGC-specific marker Brn3b and functioned as bona fide RGCs. To allow in vivo RGC tracing, Brn3b-EGFP reporter SSC-ESCs were generated and the derived RGCs were subsequently transplanted into the retina of glaucoma mouse models by intravitreal injection. We demonstrated that the transplanted RGCs could survive in host retina for at least 10 days after transplantation. SSC-ESC-derived RGCs can thus potentially be a novel alternative to replace the damaged RGCs in glaucomatous retina.

Li-rich layered oxides (LrLOs) attracted much attention due to their high specific capacity. However, LrLOs have disadvantages such as fast voltage decay and poor cycle stability. In this work, we propose a dual-doping strategy based on Ce and F ions to respond to the defects of LrLOs. This work shows that Ce and F have a strong synergistic effect in LrLOs cathode materials. Dual-doping makes the structure of the cathode materials more stable, which is mainly manifested by inhibiting the extraction of lattice oxygen, reducing the migration of cations during the cycle, and reducing the corrosion of the electrolyte to the cathode materials. Thereby, the work improves the cycle performance of the cathode materials. The capacity retention rate is 84.3% for 200 cycles at 1[Formula: see text]C (versus 59.6%). The median discharge voltage is 3.2155[Formula: see text]V after 200 cycles at 1[Formula: see text]C (versus 2.9485[Formula: see text]V). The voltage decay is 0.4706[Formula: see text]V for 200 cycles at 1[Formula: see text]C (versus 0.6923[Formula: see text]V). Interestingly, our research found that Ni[Formula: see text] of the Li layer also plays an important role in the process of improving cycle performance. This work provides new ideas for solving the cycle stability of LrLOs cathode materials and suppressing voltage decay.

Resveratrol has been reported as an ideal medicine in the treatment of colorectal cancer. Meanwhile, cadmium could affect the occurrence and development of tumors in various ways. Epithelial–mesenchymal transition is a major progress regulated with colorectal cancer (CRC). We aimed to determine the effect and mechanism of resveratrol on the Cd-promoted EMT in CRC cells. First, we investigated the migration and invasion of CRC cells with or without the treatment of different concentrations of Cd in vitro by the transwell assay. Second, Western blot and RT-qPCR assay were used to detect the expressions of EMT-related markers (ZEB1, vimentin, E-cadherin, and N-cadherin) in Cd-exposed CRC cells. Subsequently, after treating with different concentrations of resveratrol, the migration and invasion of Cd-exposed CRC cells were detected again, as well as the expressions of EMT-related markers. Moreover, m6A-related RNAs in Cd-exposed CRC cells after treating with resveratrol were immunoprecipitated and validated by Me-RIP and RT-qPCR. These indicated that Cd promoted the migration and invasion of CRC cells. In addition, Cd up-regulated the expressions of N-cadherin, vimentin, and ZEB1, while it down-regulated that of E-cadherin in CRC cells. Resveratrol could reverse the Cd-promoted migration, invasion, and EMT procession by regulating the expression of ZEB1.

The application of indocyanine green (ICG) imaging in laparoscopic radical gastrectomy is in the preliminary stages of clinical practice, and its safety and efficacy remain controversial.To investigate the safety and efficacy of ICG near-infrared tracer-guided imaging during laparoscopic D2 lymphadenectomy in patients with gastric cancer.Patients with potentially resectable gastric adenocarcinoma (clinical tumor stage cT1-cT4a, N0/+, M0) were enrolled in a prospective randomized clinical trial at a tertiary referral teaching hospital between November 2018 and July 2019. Patients were randomly assigned to the ICG group or the non-ICG group. The number of retrieved lymph nodes, rate of lymph node noncompliance, and postoperative recovery data were compared between the groups in a modified intention-to-treat analysis. Statistical analysis was performed from August to September 2019.The ICG group underwent laparoscopic gastrectomy using near-infrared imaging after receiving an endoscopic peritumoral injection of ICG to the submucosa 1 day before surgery.Total number of retrieved lymph nodes.Of 266 participants randomized, 133 underwent ICG tracer-guided laparoscopic gastrectomy, and 133 underwent conventional laparoscopic gastrectomy. After postsurgical exclusions, 258 patients were included in the modified intention-to-treat analysis, which comprised 129 patients (86 men and 43 women; mean [SD] age, 57.8 [10.7] years) in the ICG group and 129 patients (87 men and 42 women; mean [SD] age, 60.1 [9.1] years) in the non-ICG group. The mean number of lymph nodes retrieved in the ICG group was significantly more than the mean number retrieved in the non-ICG group (mean [SD], 50.5 [15.9] lymph nodes vs 42.0 [10.3] lymph nodes, respectively; P .001). Significantly more perigastric and extraperigastric lymph nodes were retrieved in the ICG group than in the non-ICG group. In addition, the mean total number of lymph nodes retrieved in the ICG group within the scope of D2 lymphadenectomy was also significantly greater than the mean number retrieved in the non-ICG group (mean [SD], 49.6 [15.0] lymph nodes vs 41.7 [10.2] lymph nodes, respectively; P .001). The lymph node noncompliance rate of the ICG group (41 of 129 patients [31.8%]) was lower than that of the non-ICG group (74 of 129 patients [57.4%]; P .001). The postoperative recovery process was comparable, and no significant difference was found between the ICG and non-ICG groups in the incidence (20 of 129 patients [15.5%] vs 21 of 129 [16.3%], respectively; P = .86) or severity of complications within 30 days after surgery.Indocyanine green can noticeably improve the number of lymph node dissections and reduce lymph node noncompliance without increased complications in patients undergoing D2 lymphadenectomy. Indocyanine green fluorescence imaging can be performed for routine lymphatic mapping during laparoscopic gastrectomy, especially total gastrectomy.ClinicalTrials.gov Identifier: NCT03050879.

To detect the changes in the liver function in both male and female OVE26 mice from young to adults for better understanding of type 1 diabetes‐induced hepatic changes, OVE26 mice and wild‐type FVB mice were raised in the same environment without any intervention, and then killed at 4, 12, 24 and 36 weeks for examining liver's general properties, including pathogenic and molecular changes. The influence of diabetes on the bodyweight of male and female mice was different. Both male and female OVE26 mice did not obtain serious liver injury or non‐alcoholic fatty liver disease, manifested by mild elevation of plasma alanine transaminase, and less liver lipid content along with significantly suppressed lipid synthesis. Uncontrolled diabetes also did not cause hepatic glycogen accumulation in OVE26 mice after 4 weeks. Oxidative stress test showed no change in lipid peroxidation, but increased protein oxidation. Changed endoplasmic reticulum stress and apoptosis along with increased antioxidant capacity was observed in OVE26 mice. In conclusion, uncontrolled type 1 diabetes did not cause hepatic lipid deposition most likely because of reduced lipids synthesis in response to insulin deficiency. Enhanced antioxidant capacity might not only prevent the occurrence of severe acute liver injury but also the self‐renewal, leading to liver dysfunction.

Peroxisomes play a central role in lipid metabolism. We previously demonstrated that Pex11a deficiency impairs peroxisome abundance and fatty acid β‐oxidation and results in hepatic triglyceride accumulation. The role of Pex11a in dyslipidaemia and obesity is investigated here with Pex11a knockout mice (Pex11a−/−). Metabolic phenotypes including tissue weight, glucose tolerance, insulin sensitivity, cholesterol levels, fatty acid profile, oxygen consumption, physical activity were assessed in wild‐type (WT) and Pex11a−/− fed with a high‐fat diet. Molecular changes and peroxisome abundance in adipose tissue were evaluated through qRT‐PCR, Western blotting, and Immunofluorescence. Pex11a−/− showed increased fat mass, decreased skeletal muscle, higher cholesterol levels, and more severely impaired glucose and insulin tolerance. Pex11a−/− consumed less oxygen, indicating a decrease in fatty acid oxidation, which is consistent with the accumulation of very long‐ and long‐chain fatty acids. Adipose palmitic acid (C16:0) levels were elevated in Pex11a−/−, which may be because of dramatically increased fatty acid synthase mRNA and protein levels. Furthermore, Pex11a deficiency increased ventricle size and macrophage infiltration, which are related to the reduced physical activity. These data demonstrate that Pex11a deficiency impairs physical activity and energy expenditure, decreases fatty acid β‐oxidation, increases de novo lipogenesis and results in dyslipidaemia and obesity.

Herein we describe the design and synthesis of a new series of heteroarylpyrimidine/heteroaryltriazine derivatives on the basis of quinazoline-2,4(1H,3H)-diones as CB2 R-selective ligands using a bioisosterism strategy. An acetamide group was explored to displace the enamine linker of the lead compound for the purpose of stereoisomerism elimination and hydrophilicity increase. As a result, some of the synthesized compounds showed high bioactivity and selectivity for CB2 R in calcium mobilization assays, and four displayed CB2 R agonist activity, with EC50 values below 30 nm. The compound exhibiting the highest agonist activity toward CB2 R (EC50 =7.53±3.15 nm) had a selectivity over CB1 R of more than 1328-fold. Moreover, structure-activity relationship (SAR) studies indicated that the substituents on the nucleus play key roles in the functionality of a ligand, with one such example demonstrating CB2 R antagonist activity. Additionally, molecular docking simulations were conducted with the aim of better understanding of these new derivatives in relation to the structural requirements for agonists/antagonists binding to CB2 R.

Background Oncostatin M (OSM) and leukemia inhibitory factor (LIF) are two important pro-inflammatory cytokines of the interleukin-6 (IL-6) family. The two cytokines mediated signaling was recently found to be closely associated with cancer and chronic inflammation, which represent promising therapeutic targets for the treatment of many solid tumors and inflammatory disease. As the most closely related members, cross-reactivity of them may result in undesired activation of off-target cells, leading to toxicity or lack of efficacy of the therapeutic effects. However, the mechanism of the cross-reactivity of OSM and LIF is not well understood. Methods In this work, protein-protein docking, molecular dynamics (MD) simulations with explicit solvent and post endpoints binding free energy (BFE) analysis were carried out to further understand the structural and energetic principles of interactions between the two cytokines and the shared receptor LIFR. Results For the first time, the simulation given a computational model of OSM-LIFR interaction, and provided significant insights into the mechanism of OSM and LIF cross-react with LIFR. The identified common features shared by OSM and LIF bind to LIFR involving 10 “conserved” residues (90% similarity) distributed at the binding site III comprised of AB loop, BC loop and D helix. In addition, 11 shared residues were identified in LIFR contribute 77.85% and 84.63% energies for OSM and LIF binding, which play a critical role in the formation of the two cytokine-receptor complexes. Moreover, the “nonconserved” residues at the same position of cytokines such as Asp41 in OSM and Pro51 in LIF as well as the three residues (Glu338, Asn201 and Glu260) in LIFR were also discovered. Conclusions These important information may facilitate the rational design of novel chemical or biological agents with less toxicity and improved efficacy.

Objective The IL‐33/ST2 axis has been extensively investigated in type 2 eosinophilic inflammation. Here, we aimed to investigate the role of the IL‐33/ST2 axis in neutrophil‐dominant allergic airway inflammation. Methods House‐dust mite (HDM) extract and lipopolysaccharide (LPS) were administered to establish a murine model of neutrophil‐dominant allergic airway inflammation. The formation of neutrophilic extracellular traps (NETs) in the lung tissues was demonstrated by immunofluorescence imaging. Mature IL‐33 in bronchoalveolar lavage fluid (BALF) was detected by Western blotting. The neutrophilic chemokine KC produced by bone marrow‐derived macrophages (BMDMs) or primary alveolar epithelial cells was measured with a commercial ELISA kit. Results In the present study, we observed neutrophilic inflammation and tight junction damage in the lungs of mice sensitised with HDM and LPS. Furthermore, sensitisation with HDM and LPS resulted in the formation of NETs, accompanied by increased levels of mature IL‐33 in the BALF. Moreover, LPS damaged the epithelial tight junction protein occludin directly or indirectly by inducing NET formation. Surprisingly, IL‐33 deficiency augmented neutrophilia and epithelial barrier injury in the lungs of mice after sensitisation with HDM and LPS. Similarly, the absence of ST2 exacerbated the neutrophilic inflammatory response, decreased the expression of occludin and exacerbated the severity of neutrophil‐dominant allergic airway inflammation in an HDM/LPS‐induced mouse model. Mechanistically, BMDMs and alveolar epithelial cells from IL‐33‐ or ST2‐deficient mice tended to produce higher levels of the neutrophilic chemokine KC. Conclusions These results demonstrated that the IL‐33/ST2 axis may play a protective role in neutrophil‐dominant allergic airway inflammation., Most research on the IL‐33/ST2 axis has been conducted in type 2 immunity‐mediated eosinophilic asthma, but less research has focused on neutrophilic asthma. In this study, we found that IL‐33/ST2 axis deficiency aggravated neutrophil‐dominant allergic airway inflammation induced by house‐dust mite and lipopolysaccharide coexposure, which may account for the more proinflammatory status of IL‐33/ST2 axis‐deficient macrophages and lung epithelial cells.

Notwithstanding previous studies have proved the anti-apoptotic effect of Bcl-2 associated athanogene3 (BAG3) in myocardium, the structural domains PXXP and BAG responsible for its protection are not reformed. Since BAG3 in cardiomyocytes is a new target for inhibiting apoptosis induced by hypoxia/reoxygenation (H/R) stress, we demonstrated that over-expression of BAG3 reduced the injury induced by H/R in either neonatal or adult rat cardiomyocytes (NRCMs and ARCMs, respectively) and PXXP and BAG domains play an important role in cellular protection in H/R stress. Apoptosis in cardiomyocytes induced by hypoxia-reperfusion was evaluated with propidium iodide (PI) staining, cleaved caspase-3, and terminal deoxynucleotidyl transferase dUTP nick end labelling (TUNEL) staining in cultured NRCMS. Either increasing expression of BAG3 or its mutants was performed to manipulate the level of BAG3. Co-immunoprecipitation (Co-IP) was used to demonstrate the complex that BAG3 is binding to HSC70 and JNK. PXXP and BAG domains of BAG3 played an essential role in BAG3 attenuating cardiomyocytes apoptosis induced by H/R through the JNK signalling pathway. The cellular protection of BAG3 with its structural domain PXXP or BAG is associated with the binding with HSC70 and JNK. These results showed that the protective effect of BAG3 on apoptosis induced by H/R stress is closely related to its structural domains PXXP and BAG. The mechanism may provide a new therapeutic strategy for the patients suffering from ischemic cardiomyopathy and may be a critical role of its PXXP and BAG3 domains.

Moringa oleifera Lam. (M. oleifera) is valuable plant distributed in many tropical and subtropical countries. It has a number of medicinal uses and is highly nutritious. M. oleifera has been shown to inhibit tumor cell growth, but this effect has not been demonstrated on prostate cancer cells. In this study, we evaluated the inhibitory effect of M. oleifera alkaloids (MOA) on proliferation and migration of PC3 human prostate cancer cells in vitro and in vivo. Furthermore, we elucidated the mechanism of these effects. The results showed that MOA inhibited proliferation of PC3 cells and induced apoptosis and cell cycle arrest. Furthermore, MOA suppressed PC3 cell migration and inhibited the expression of matrix metalloproteinases (MMP)-9. In addition, MOA significantly downregulated the expression of cyclooxygenase 2 (COX-2), β-catenin, phosphorylated glycogen synthase 3β, and vascular endothelial growth factor, and suppressed production of prostaglandin E2 (PGE2). Furthermore, FH535 (β-catenin inhibitor) and MOA reversed PGE2-induced PC3 cell proliferation and migration, and the effects of MOA and FH535 were not additive. In vivo experiments showed that MOA (150 mg/kg) significantly inhibited growth of xenograft tumors in mice, and significantly reduced the protein expression levels of COX-2 and β-catenin in tumor tissues. These results indicate that MOA inhibits the proliferation and migration, and induces apoptosis and cell cycle arrest of PC3 cells. Additionally, MOA inhibits the proliferation and migration of PC3 cells through suppression of the COX-2 mediated Wnt/β-catenin signaling pathway.

BACKGROUND: The diagnosis of severe asthma (SA) is difficult due to a necessary long-term treatment history currently, while there are few studies on biomarkers in the diagnosis of SA. Long non-coding RNA (lncRNA) growth arrest specific-5 (GAS5) has the potential of playing this role because its binding with glucocorticoid receptor (GR). The purpose of this article is to explore the possibility of lncRNA GAS5 acting as a biomarker for early diagnosis of severe asthma (SA). METHODS: Peripheral blood was obtained from healthy volunteers, patients with non-severe asthma (nSA) and SA, and peripheral blood mononuclear cells (PBMCs) were separated. Twenty-four female BALB/c mice (aged 6 weeks) were randomly and averagely divided into 3 groups, i.e., control group, asthma group and dexamethasone group. The mice were sensitized and challenged with ovalbumin (OVA) and lipopolysaccharide (LPS) to establish a murine model of steroid-insensitive asthma. Human bronchial epithelial cells (HBECs) were cultured, transfected with miR-9 mimics, JNK1 inhibitor and treated with interleukin (IL)-2 + IL-4 and dexamethasone. Western blot was used to detect glucocorticoid receptor phosphorylation at serine 226 (GR(ser226)), and quantitative real-time PCR was used to detect GAS5 level. Staphylococcus aureus is one of the most common pathogens in bacterial biofilm infections. Antibiotic treatment for infection caused by S. aureus biofilms is challenging, and few effective strategies have been developed to combat these infections. The aim of this study was to investigate the effect and possible mechanisms of artesunate on the biofilm formation of S. aureus. Bacterial growth curves were determined by a microtiter plate. Biofilm formation was determined by the crystal violet staining method and confocal laser scanning microscopy. Bacterial adhesion was assayed by the colony-counting method. The expression of virulence and adhesion genes was determined by real-time PCR. The hemolytic activity and expression of ɑ-hemolysin were analyzed using rabbit erythrocytes and Western blotting. The results showed that artesunate could significantly inhibit the biofilm formation of S. aureus in a dose-dependent manner. Artesunate could also inhibit bacterial adhesion and the expression of hla, RNAIII and agrA as well as ɑ-hemolysin production. The effect of artesunate on adhesion genes (clfA, clfB, fnbA, fnbB) had strain specificity, but it did not affect the expression of ica genes. The results indicated that artesunate might inhibit ɑ-hemolysin synthesis by the agr system, which inhibits biofilm formation.

To investigate associations between central visual function and inner retinal structure in primary open-angle glaucoma (POAG). This study enrolled 78 POAG patients and 58 healthy controls. POAG was classified into early glaucoma and moderate to advanced glaucoma. The following tests were performed on all participants: isolated-check visual evoked potential (icVEP) testing, 24-2 standard automated perimetry (SAP), and Cirrus optical coherence tomography (OCT) examinations. Signal-to-noise ratio (SNR) measures obtained from icVEP responses to isolated checks presented at four depths of modulation (DOMs; 8%, 14%, 22%, and 32%) were explored. Mean macular sensitivity (mMS) was assessed by calculating the mean sensitivities of central 12 SAP points. Ganglion cell layer+ inner plexiform layer thickness (GCL+IPLT) and peripapillary retinal nerve fiber layer thickness (pRNFLT) were measured by OCT scanning. Therapeutics targeting cytokines such as the oncostatin M (OSM)-mediated inflammation represent a potential strategy for the treatment of inflammatory bowel disease (IBD). Despite the investigation of the specific role of the interactions between OSM and the receptor (OSMR) in IBD pathogenesis, the 3D structure of the OSM–OSMR complex remains elusive. In this work, the interaction mode between OSM and OSMR at atomic level was predicted by computational simulation approach. The interaction domain of the OSMR was built with the homology modeling method. The near-native structure of the OSM–OSMR complex was obtained by docking, and long-time scale molecular dynamics (MD) simulation in an explicit solvent was further performed to sample the conformations when OSM binds to the OSMR. After getting the equilibrated states of the simulation system, per-residue energy contribution was calculated to characterize the important residues for the OSM–OSMR complex formation. Based on these important residues, eight residues (OSM: Arg100, Leu103, Phe160, and Gln161; OSMR: Tyr214, Ser223, Asp262, and Trp267) were identified as the “hot spots” through computational alanine mutagenesis analysis and verified by additional MD simulation of R100A (one of the identified “hotspots”) mutant. Moreover, six cavities were detected at the OSM–OSMR interface through the FTMap analysis, and they were suggested as important binding sites. The predicted 3D structure of the OSM–OSMR complex and the identified “hot spots” constituting the core of the binding interface provide helpful information in understanding the OSM–OSMR interactions, and the detected sites serve as promising targets in designing small molecules to block the interactions.

Biogenic volatile organic compounds (BVOC) play an important role in global environmental chemistry and climate. In the present work, biogenic emissions from China in 2017 were estimated based on the Model of Emissions of Gases and Aerosols from Nature (MEGAN). The effects of BVOC emissions on ozone and secondary organic aerosol (SOA) formation were investigated using the WRF-CMAQ modeling system. Three parallel scenarios were developed to assess the impact of BVOC emissions on China's ozone and SOA formation in July 2017. Biogenic emissions were estimated at 23.54 Tg/yr, with a peak in the summer and decreasing from southern to northern China. The high BVOC emissions across eastern and southwestern China increased the surface ozone levels, particularly in the BTH (Beijing-Tianjin-Hebei), SCB (Sichuan Basin), YRD (Yangtze River Delta) and central PRD (Pearl River Delta) regions, with increases of up to 47 μg m−3 due to the sensitivity of VOC-limited urban areas. In summer, most SOA concentrations formed over China are from biogenic sources (national average of 70%). And SOA concentrations in YRD and SCB regions are generally higher than other regions. Excluding anthropogenic emissions while keeping biogenic emissions unchanged results that SOA concentrations reduce by 60% over China, which indicates that anthropogenic emissions can interact with biogenic emissions then facilitate biogenic SOA formation. It is suggested that controlling anthropogenic emissions would result in reduction of both anthropogenic and biogenic SOA.

A facile protocol for the synthesis of acridone derivatives has been developed through palladium-catalyzed carbonylation/C H activation sequence from 2-bromo-diarylamines in moderate to excellent yields. The reactions occurred smoothly and allowed both electron-rich and electron-deficient substrates converted to the corresponding acridones.

The survival rate of childhood acute lymphoblastic leukemia (ALL) has increased while that of Philadelphia-positive (Ph+) ALL remains low. CD19 is a B-cell specific molecule related to the survival and proliferation of normal B cells. However, there is little information available on the effects of CD19 on the biological behavior of Ph+ ALL cells. In this study, we explored a lentiviral vector-mediated short hairpin RNA (shRNA) expression vector to stably reduce CD19 expression in Ph+ ALL cell line SUP-B15 cells and investigated the effects of CD19 downregulation on cell proliferation, apoptosis, drug sensitivity, cell adhesion, cell migration and cell invasion in vitro. CD19 mRNA and protein expression levels were inhibited significantly by CD19 shRNA. Down-regulation of CD19 could inhibit cell proliferation, adhesion, migration and invasion, and increase cell apoptosis and the efficacy of chemotherapeutic agents and imatinib in SUP-B15 cells. Moreover, we found that down-regulation of CD19 expression inhibits cell proliferation and induces apoptosis in SUP-B15 cells in a p53-dependent manner. Taken together, our results suggest that lentiviral vector-mediated RNA interference of CD19 gene may be a promising strategy in the treatment of Ph+ ALL.

In this paper, an electrochemical nonenzymatic sensor using three dimensional Fe 3 O 4 accumulated sphere-like nanocomposites as the carriers was designed. The decorated materials with homogeneous stacked sphere-like morphologies were synthesized through a facile one-step solvothermal reaction by selecting Fe(CO) 5 as the iron sources and absolute ethanol as the solvent. We used cyclic voltammetric (CVs) and current-time (I-t) curves to detect the current response towards H 2 O 2 . Achieved consequences demonstrated that the as-prepared nonenzymatic H 2 O 2 sensor revealed the low detection potential (−0.1 V) as well as high sensitivity (205 μA mM −1 cm −2 ), wide linear range (5.0 μM-4.995 mM) and low detection limit (2 μM, S/N = 3). In addition, the novel nonenzymatic sensor was successfully applied to the detection of H 2 O 2 in pasteurized milk samples.

A green protocol with catalyst-free photo-induced cyclization for the synthesis of a spiro-fused organic semiconductor molecule based on spiro[fluorene-9,9′-xanthene] (SFX) was developed. The photophysical and electrochemical properties of the spiro-fused compound (DPCzSFX) have been characterized. The results showed that the molecular stability and the property of the charge injecting/transporting were improved due to the cyclization. And the broadening of the emission spectrum in film makes the sipro-fused compound possible for the application as a host in the white phosphorescent organic light-emitting diodes. However, the same reaction for the spirobifluorene-based dipenylamine derivative did not occur under the same condition, which also highlights the importance of the oxygen heteroatom in the ring to the reaction of cyclization.

Iron based catalysts supported on aluminum pillared clays (Fe/Al-PILC) was prepared by impregnation method and the selective catalytic reduction of NO with propene by the Fe/Al-PILC catalysts was investigated in a fixed bed reactor. The physical and chemical properties of the catalysts were characterized by N2 adsorption/desorption, XRD, H2-TPR, UV-Vis, Py-IR, etc. Results showed that 9% Fe/Al-PILC reduced 98% of NO at 400-550°C. SO2 and water vapor slightly influenced the catalytic activity of the catalysts. XRD and N2 adsorption/desorption characterization results revealed that the iron oxides in the catalyst were highly dispersed on the surface of the carrier and the catalyst has a large specific surface area and pore volume. H2-TPR results indicated that the activity of the catalysts was mainly determined by the reduction performance of Fe2O3 phase. UV-Vis results showed that the activity of the catalysts was positively correlated with the iron oxide oligomer FexOy. Py-IR results indicated that both Lewis acid and Bronsted acid were formed on the catalyst surface and the Lewis acid sites were the main catalytic activity center of NO and C3H6 reaction.

Kidney biopsy is considered the golden criterion for diagnosing the etiology of kidney disease but accompanied by non-negligible complications. We explored the possibility of using urinary microRNA (miRNA) as a non-invasive biomarker for hypertensive kidney injury. We assessed differential miRNA expressions in the kidneys and urine of hypertensive mice with kidney injury induced by deoxycorticosterone acetate (DOCA)-salt compared to the controls. DOCA-salt treatment significantly increased renal tubular lesions from day 2 and mRNA expression of fibrosis-related genes from day 4 compared to the controls, respectively. Urinary albumin and N-acetyl-beta-D-glucosaminidase was significantly increased on day 8 compared to the controls. Array results showed that 20 out of 585 miRNAs were highly expressed in the kidneys and significantly increased on day 8 compared to the controls, including miR-21, miR-146b, miR-155 and miR-132, which were confirmed by real-time polymerase chain reaction and were significantly higher from day 4. The miR-21/creatinine in the urine from day 4 was significantly higher than that of the controls and was detected earlier than urinary albumin. In conclusion, we have identified urinary miR-21 that correlates with histopathological lesions and functional markers of kidney damage to facilitate a potential noninvasive detection for hypertensive kidney injury.

Materials with Dirac point are so amazing since the charge carriers are massless and have an effective speed of light. However, among the predicted two-dimensional silicon allotropes with Dirac point, no one has been directly proved by experiment. This fact motivates us to search for other two-dimensional silicon allotropes. As a result, another stable single atomic layer thin silicon allotrope is found with the help of CALYPSO code in this work. This silicene allotrope is composed of eight-membered rings linked by Si–Si bonds with buckling formation. The electronic calculation reveals that it behaves as a nodal line semimetal with the linear energy dispersion relation near the Fermi surface. Notably, the ab initio molecular dynamics simulations display that the original atomic configuration can be remained even at an extremely high temperature of 1000 K . Additionally, hydrogenation could induce a semimetal-semiconductor transition in this silicene allotrope. We hope this work can expand the family of single atomic layer thin silicon allotropes with special applications.

Sol-gel and impregnation methods were used to prepare the Fe-Ag/Al2O3 catalysts supported on the monolithic cordierite with different Fe/Ag loading ratios. The catalytic performance to reduce NO with C3H6 was evaluated in a one-dimensional electrically heated temperature programmed ceramic tubular reactor in simulated flue gas atmosphere at 200–700°C. The results show that the NO reduction efficiency on 7.2Fe/1.9Ag/20Al2O3/CM with C3H6 is more than 90% and reaches about 100% at the temperatures of 500°C and 550°C respectively. Iron can effectively improve the ability of Ag/20Al2O3/CM catalysts to resist SO2 and H2O in flue gas. When SO2 and H2O are 0.02% and 8% in the flue gas, the NO reduction efficiency is almost not influenced on 7.2Fe/1.9Ag/20Al2O3/CM at 500°C. The 90% NO reduction efficiency is maintained during 6 h without decrease. However, the catalytic activity of 2Ag/20Al2O3/CM without iron modification is strongly influenced by SO2 and H2O in the flue gas. The NO reduction efficiency on Ag/20Al2O3/CM decreases rapidly from about 70% to 46% and 25% respectively, when the SO2 and H2O are 0.02% and 8% in the flue gas. The results of XRD and SEM of the catalyst show that AgFeO2 and Fe3+ are formed in the 7.2Fe/1.9Ag/20Al2O3/CM catalyst after the modification by iron, and the surface of the catalyst become loose and porous, forming Fe3O4-based needle-like and flaky crystals. H2-TPR results show that 7.2Fe/1.9Ag/20Al2O3/CM has better reduction properties than Ag/20Al2O3/CM in the wider temperature range. Pyridine adsorption Infrared Spectroscopy (Py-FTIR) experimental results show that Fe increases the Lewis acid sites in the catalyst surface.

Hebei is one of the most air-polluted provinces located in China. In this study, NOx, volatile organic compounds (VOCs), and ozone (O3) observational data obtained at 7 monitoring sites in Hebei Province in the warm season (May to September) in 2019 were analyzed to investigate the spatiotemporal distribution, composition characteristics, and chemical reactivity of VOCs. Results indicated that VOCs concentrations were the highest in Shijiazhuang and Langfang (47.8 and 39.2 ppb, respectively) and the lowest in Zhangjiakou and Chengde (16.6 and 12.0 ppb, respectively). In the warm season in 2019, the monthly average concentrations of VOCs increased initially and decreased subsequently, and the highest concentration found in July and August. The diurnal variation patterns of VOCs exhibited a double-peak curve, with the first peak occurring between 07:00–09:00, influenced by the production and life activities of residents. The second peak occurred between 23:00–0:00 due to the poor atmospheric diffusion conditions at nighttime and the possible transportation from residual layer. Alkanes, haloalkanes, and alkenes were responsible for the largest components to VOCs in Hebei Province, with the average proportions at the 7 monitoring sites of 43.25%, 30.39% and 15.00%, respectively. The ratios of toluene to benzene (T/B) of the monitored sites indicated that the VOCs concentrations at the Shijiazhuang and Xingtai sites were mainly influenced by emissions from coal combustion. In comparison, the VOCs concentrations at the Baoding and Langfang sites were mainly influenced by motor vehicle emissions, while the sites at Zhangjiakou and Chengde were mainly influenced by solvent-based coating and industrial process emissions. Alkenes had the highest chemical reactivity of the VOCs in Hebei Province, ethylene, propene, toluene, and 1-butene were also the high chemical reactivity species. Lastly, tailored schemes for the reduction of O3 precursor were proposed based on the NOx, VOCs, and O3 data obtained from the various monitoring sites.

Among the reported two-dimensional silicon materials, only hexagonal and kagome-like silicene have been reported experimentally. Thus, designing such materials with another strategy is desirable. Based on the experimentally synthesized pentagonal silicon cluster recently and via a swarm structure search method, herein we report a two-dimensional silicene allotrope by combining pentagonal silicon clusters. Further calculations show that this silicene allotrope exhibits great anisotropic conductance, it behaves as a metal with linear energy dispersions along the b -direction and Dirac point located at about 0.10 eV below the Fermi energy level, while it exhibits semiconducting state along the a -direction. This strong anisotropic conductance is driven by the different coordination numbers of silicon atoms. Through hydrogenating the three-fold coordinated silicon atoms, the silicene allotrope would turn into a semiconductor with a direct band gap of 1.60 eV (2.05 eV under HSE level). Particularly, the hydrogenated model possesses high carrier mobility of and at room temperature for electrons and holes, respectively. This finding promotes potential applications of silicon nanomaterials in in-plane anisotropic nanoelectronic, high-speed electronic, and photovoltaic devices.

Dbl-family guanine nucleotide exchange factors (GEFs) can activate RhoGTPases by facilitating the exchange of GDP for GTP, the aberrant expression of which has been implicated in tumorigenicity and metastasis of human cancers. ARHGEF39, as a member of Dbl-family GEFs, was reported to be a potential oncogene in human hepatocellular carcinoma previously. However, the role of ARHGEF39 in gastric cancer (GC) remains unclear so far. In the current study, we demonstrated that ARHGEF39 expression was significantly upregulated in GC tissues compared with paired adjacent normal tissues by quantitative real-time PCR analysis. Functional analyses revealed that ARHGEF39 overexpression could promote proliferation, colony formation, and migration of GC cells in vitro, whereas ARHGEF39 knockdown markedly suppressed these phenotypes. Moreover, ARHGEF39 enhanced tumorigenicity and lung metastasis potential of GC cells in nude mice model. Mechanistically, we found that overexpressed ARHGEF39 significantly increased the phosphorylation level of Akt (p-Akt), and its effect on cell proliferation was attenuated by PI3K inhibitor LY294002. Thus, our findings suggest that ARHGEF39 may contribute to cell proliferation and migration in GC via a possible mechanism involving Akt signaling.

Herein, we described the design and synthesis of a series of pyridazine-3-carboxamides to be CB2-selective agonists via a combination of scaffold hopping and bioisosterism strategies. The compounds were subjected to assessment of their potential activities through calcium mobilization assays. Among the tested derivatives, more than half of these compounds exhibited moderate to potent CB2 agonist activity. Six compounds showed EC50 values below 35 nM, and several derivatives also exhibited significantly enhanced potency and high selectivity at the CB2 receptor over the CB1 receptor. Specifically, compound 26 showed the highest CB2 agonist activity (EC50 = 3.665 ± 0.553 nM) and remarkable selectivity (Selectivity Index > 2729) against CB1. In addition, logPs of some representative compounds were measured to display significantly decreased values in comparison with GW842166X. Furthermore, docking simulations were conducted to explain the interaction mode of this series.

In this paper, 15 batches of volatile oil from Schizonepeta tenuifolia herbs (STVO) originating from different areas of China were used to identify and quantify eight main components by GC-MS and to evaluate the antioxidant activity with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity by the HPLC method. The results indicated that with respect to the antioxidant activity and the contents of the main compounds, there were notable differences among the STVOs. IC50 values of the STVOs ranged from 0.016 to 1.518 mg/mL. To determine the correlation of the chemical components and the antioxidant activities, multiple linear regression analysis (MLRA) and partial least square analysis (PLSA) were performed. As a result, (+)-pulegone and (-)-menthone showed much closer relationships than the other components in each analysis. Hence, it was concluded that (+)-pulegone and (-)-menthone were the characteristic components that may exhibit potent antioxidant activities in STVO.

Starting from a prototypical structure 1, we describe our efforts to design and obtain novel quinazoline/pyrimidine-2,4(1H,3H)-diones with high CB2 agonist potency and selectivity as well as improved physicochemical characteristics, mainly hydrophilicity. The most potent and selective CB2 agonists, 8 and 36, in this series were also endowed with lower logP values than that of GW842166X and lead compound 1. These derivatives appear to be promising lead compounds for the development of future CB2 agonists.

Designing selective inhibitor of protein kinase B (PKB/Akt) is an area of intense research to develop potential anticancer drugs. As a general point of strategy, the peptide substrate-binding site only responds to a highly specific sequence of amino acids. Targeting the substrate-mimetic inhibitors to the peptide substrate-binding site has the potential for better selectivity. It is therefore of great interest to understand the peptide substrate binding mode of PKB, as well as its specificity and affinity for different substrate-mimetic inhibitors. In the present study, we used molecular dynamic simulations to better understand the interactions of the PKB substrate-binding site with the substrate-mimetic inhibitors. Our computational models successfully mirrored PKB’s selectivity for the substrate-mimetic inhibitors.
