Abstract Accurate and sensitive near‐infrared (NIR) luminescent lateral flow immunoassay (LFIA) has attracted considerable attention in the field of point‐of‐care testing (POCT). However, the detection accuracy and sensitivity are often compromised by the low fluorescence quantum efficiency ( 800 nm) completely eliminated background interference and allowed highly accurate and sensitive detection without any pre‐treatment steps. The limits of detection (LODs) for aflatoxin B1 (AFB1) in soy sauce, alpha hemolysin (Hla) of Staphylococcus aureus biomarker in joint fluid, and C‐reactive protein (CRP) in human haemolysed samples were 0.01 ng mL−1, 0.02 µg mL−1, and 0.156 mg L−1, respectively, commensurating with those of the corresponding gold standard assays and covering the detection range of interests. It is anticipated that the ultrabright NIR AIEgen nanoparticles will serve as a universally applicable signal probe for NIR‐LFIA diagnostics, promising to expand the range of applications for quantitative detection of complex samples.

Agropyron mongolicum Keng is a diploid perennial grass of triticeae in gramineae. It has strong drought resistance and developed roots that can effectively fix the soil and prevent soil erosion. GDSL lipase or esterases/lipase has a variety of functions, mainly focusing on plant abiotic stress response. In this study, a GDSL gene from A. mongolicum, designated as AmGDSL1, was successfully cloned and isolated. The subcellular localization of the AmGDSL1 gene (pCAMBIA1302-AmGDSL1-EGFP) results showed that the AmGDSL1 protein of A. mongolicum was only localized in the cytoplasm. When transferred into tobacco (Nicotiana benthamiana), the heterologous expression of AmGDSL1 led to enhanced drought tolerance. Under drought stress, AmGDSL1 overexpressing plants showed fewer wilting leaves, longer roots, and larger root surface area. These overexpression lines possessed higher superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and proline (PRO) activities. At the same time, the malondialdehyde (MDA) content was lower than that in wild-type (WT) tobacco. These findings shed light on the molecular mechanisms involved in the GDSL gene’s role in drought resistance, contributing to the discovery and utilization of drought-resistant genes in A. mongolicum for enhancing crop drought resistance.

Traditional drilling methods often face difficulty meeting the demand for efficient and accurate coring under complex geological conditions. Cordless coring is an advanced technology that uses hydraulic lifting to carry out coring, which can achieve automation and automated and intelligent drilling operations. In this research, a new type of hydraulic lifting cordless coring drilling tool is designed. Moreover, a numerical simulation model of the fluid flow in the annulus between the spearhead and spool of the cordless coring drilling tool was established. Orthogonal simulation tests are carried out, and according to the orthogonal test data, a numerical prediction model of the spool annulus fluid field based on the Backpropagation Neural Network (BP neural network) is established. The prediction of the flow rate of the drilling fluid and the spool back-pressure ratio was obtained when the structural parameters of the spearhead and the spool annulus were different. A multi-objective optimization of the annulus flow structure of the cordless core drilling tool has been carried out. The optimization objectives include deciding the back pressure ratio of the spool overcoming the spring and the flow rate of the drilling fluid. According to the established nonlinear optimization model and based on the improved Non-dominated Sorting Genetic Algorithm II (NSGA-II) multi-objective optimization algorithm, it is verified that the convergence speed and diversity of the improved algorithm are better than those before the improvement. The simulation and experimental validation are carried out. It is verified that the flow rate of drilling fluid increased by 33.56% after optimization, and the force ratio was lowered by 5.825%. Finally, based on the simulation and optimization results, the φ96 cordless core drilling tool was manufactured on a trial basis, and on-site concrete drilling, coring, and hydraulic lifting operations were conducted for smooth coring and lifting. This study could provide an important scientific basis and technical support for the application and development of hydraulic lifting cordless coring technology.

Abstract Nonconventional luminescent materials have been rising stars in organic luminophores due to their intrinsic characteristics, including water‐solubility, biocompatibility, and environmental friendliness and have shown potential applications in diverse fields. As an indispensable branch of nonconventional luminescent materials, polysiloxanes, which consist of electron‐rich auxochromic groups, have exhibited outstanding photophysical properties due to the unique silicon atoms. The flexible Si‐O bonds benefit the aggregation, and the empty 3d orbitals of Si atoms can generate coordination bonds including N → Si and O → Si, altering the electron delocalization of the material and improving the luminescent purity. Herein, we review the recent progress in luminescent polysiloxanes with different topologies and discuss the challenges and perspectives. With an emphasis on the driving force for the aggregation and the mechanism of tuned emissions, the role of Si atoms played in the nonconventional luminophores is highlighted. This review may provide new insights into the design of nonconventional luminescent materials and expand their further applications in sensing, biomedicine, lighting devices, etc.

Functional molecules have been attracting increasing attention in environmental and physiological studies. In particular, folic acid (FA) could be considered a key factor in estimating, adjusting, and making decisions in the treatment of neurodevelopmental disorders. It promotes the general significance and conceptual for considering FA molecular scientific research detections, which implies related advancement in both of biological structure and detection methods. Among these applications, the FA molecule acts as a coenzyme that incorporates carbon atoms and synthesizes purines and pyrimidines. Therefore, the calibration method has real applications and can be used as a sensing platform and for detection approaches, which conveys the internal relationship between the FA molecule and physiological characterization. This mini review briefly discusses multiple FA application fields and detection pathways and could supplement their utilization in anticipation of the onset of disease.

Due to the increase in nanoplastics (NPs) abundance in aquatic environments, their effects on phytoplankton have aroused large research attention. In this study, 100 nm sized polystyrene NPs were chosen to investigate their effecting performance and mechanisms on a typical dinoflagellates Alexandrium tamarense. The results indicated the population growth and photosynthetic efficiencies of A. tamarense were significantly inhibited by NPs exposure, as well as the increase in cellular total carotenoids and paralytic shellfish toxins (PSTs). Meanwhile, the cellar ROS levels increased, corresponding to the increased activities or contents of multiple antioxidant components, including SOD, CAT, GPX, GR, GSH and GSSG. The transcriptional results support the physiological–biochemical results and further revealed the down-regulation of genes encoding the light reaction centers (PSI and PSII) and up-regulation of genes encoding the antioxidant components. Up-regulation of genes encoding key enzymes of the Calvin cycle and glycolytic pathway together with the TCA cycle could accelerate organic carbon and ATP production for A. tamarense cells resistant to NPs stress. Finally, more Glu and acetyl-CoA produced by the enhanced GSH cycle and the glycolytic pathway, respectively, accompanied by the up-regulation of Glu and Arg biosynthesis genes supported the increase in the PST contents under NPs exposure. This study established a data set involving physiological–biochemical changes and gene information about marine dinoflagellates responding to NPs, providing a data basis for further evaluating the ecological risk of NPs in marine environments.

Carrier-based Unmanned Aerial Vehicle (CUAV) landing is an extremely critical link in the overall chain of CUAV operations on ships. Vision-based landing location methods have advantages such as low cost and high accuracy. However, when an aircraft carrier is at sea, it may encounter complex weather conditions such as haze, which could lead to vision-based landing failures. This paper proposes a runway line recognition and localization method based on haze removal enhancement to solve this problem. Firstly, a haze removal algorithm using a multi-mechanism, multi-architecture network model is introduced. Compared with traditional algorithms, the proposed model not only consumes less GPU memory but also achieves superior image restoration results. Based on this, We employed the random sample consensus method to reduce the error in runway line localization. Additionally, extensive experiments conducted in the Airsim simulation environment have shown that our pipeline effectively addresses the issue of decreased detection accuracy of runway line detection algorithms in haze maritime conditions, improving the runway line localization accuracy by approximately 85%.

Paclitaxel, a microtubule-stabilizing chemotherapy drug, can cause severe paclitaxel-induced peripheral neuropathic pain (PIPNP). The roles of transient receptor potential (TRP) ion channel vanilloid 1 (TRPV1, a nociceptor and heat sensor) and melastatin 8 (TRPM8, a cold sensor) in PIPNP remain controversial. In this study, Western blotting, immunofluorescence staining, and calcium imaging revealed that the expression and functional activity of TRPV1 were upregulated in rat dorsal root ganglion (DRG) neurons in PIPNP. Behavioral assessments using the von Frey and brush tests demonstrated that mechanical hyperalgesia in PIPNP was significantly inhibited by intraperitoneal or intrathecal administration of the TRPV1 antagonist capsazepine, indicating that TRPV1 played a key role in PIPNP. Conversely, the expression of TRPM8 protein decreased and its channel activity was reduced in DRG neurons. Furthermore, activation of TRPM8 via topical application of menthol or intrathecal injection of WS-12 attenuated the mechanical pain. Mechanistically, the TRPV1 activity triggered by capsaicin (a TRPV1 agonist) was reduced after menthol application in cultured DRG neurons, especially in the paclitaxel-treated group. These findings showed that upregulation of TRPV1 and inhibition of TRPM8 are involved in the generation of PIPNP, and they suggested that inhibition of TRPV1 function in DRG neurons via activation of TRPM8 might underlie the analgesic effects of menthol.

Abstract A series of new π‐conjugated macrocycles (AzoM‐n‐E, n = 1–3) incorporating azobenzene units have been synthesized by a facile strategy in one‐pot reaction. The resultant azobenzene‐embedded macrocycles feature intrinsic photoresponsive behaviors and intriguing supramolecular assembling properties. The smallest macrocycle AzoM‐1‐E with a rigid planar conjugated backbone structure is used to prepare the single crystal transistors, showing reversible optical tunability. The moderate size macrocycle AzoM‐2‐E assembles into a dimer in the form of interpenetration through π‐π stacking between azobenzene units. The largest macrocycle AzoM‐3‐E with enhanced flexibility can adaptively assemble with various types of electron‐deficient guests accompanied by distortion of azobenzene. Typically, AzoM‐3‐E assembles with the planar F4‐TCNQ to form a tetragonal geometry by C‐F···π and π‐π interactions, while the assembly with ellipsoidal C70 via π‐π interactions induces AzoM‐3‐E to form a boat‐shaped geometry. This work will shed new light on the development of functional conjugated macrocycles in organic electronics.

Members of the abscisic acid (ABA)-responsive element (ABRE) binding factor (ABF) and ABA-responsive element binding protein (AREB) families play essential roles in the regulation of ABA signaling pathway activity and shape the ability of plants to adapt to a range of stressful environmental conditions. To date, however, systematic genome-wide analyses focused on the ABF/AREB gene family in wheat are lacking. Here, we identified 35 ABF/AREB genes in the wheat genome, designated TaABF1–TaABF35 according to their chromosomal distribution. These genes were further classified, based on their phylogenetic relationships, into three groups (A–C), with the TaABF genes in a given group exhibiting similar motifs and similar numbers of introns/exons. Cis-element analyses of the promoter regions upstream of these TaABFs revealed large numbers of ABREs, with the other predominant elements that were identified differing across these three groups. Patterns of TaABF gene expansion were primarily characterized by allopolyploidization and fragment duplication, with purifying selection having played a significant role in the evolution of this gene family. Further expression profiling indicated that the majority of the TaABF genes from groups A and B were highly expressed in various tissues and upregulated following abiotic stress exposure such as drought, low temperature, low nitrogen, etc., while some of the TaABF genes in group C were specifically expressed in grain tissues. Regulatory network analyses revealed that four of the group A TaABFs (TaABF2, TaABF7, TaABF13, and TaABF19) were centrally located in protein–protein interaction networks, with 13 of these TaABF genes being regulated by 11 known miRNAs, which play important roles in abiotic stress resistance such as drought and salt stress. The two primary upstream transcription factor types found to regulate TaABF gene expression were BBR/BPC and ERF, which have previously been reported to be important in the context of plant abiotic stress responses. Together, these results offer insight into the role that the ABF/AREB genes play in the responses of wheat to abiotic stressors, providing a robust foundation for future functional studies of these genes.

Tube-like outgrowths from root epidermal cells, known as root hairs, enhance water and nutrient absorption, facilitate microbial interactions, and contribute to plant anchorage by expanding the root surface area. Genetically regulated and strongly influenced by environmental conditions, longer root hairs generally enhance water and nutrient absorption, correlating with increased stress resistance. Wheat, a globally predominant crop pivotal for human nutrition, necessitates the identification of long root hair genotypes and their regulatory genes to enhance nutrient capture and yield potential. This study focused on 261 wheat samples of diverse genotypes during germination, revealing noticeable disparities in the length of the root hair among the genotypes. Notably, two long root hair genotypes (W106 and W136) and two short root hair genotypes (W90 and W100) were identified. Transcriptome sequencing resulted in the development of 12 root cDNA libraries, unveiling 1180 shared differentially expressed genes (DEGs). Further analyses, including GO function annotation, KEGG enrichment, MapMan metabolic pathway analysis, and protein–protein interaction (PPI) network prediction, underscored the upregulation of root hair length regulatory genes in the long root hair genotypes. These included genes are associated with GA and BA hormone signaling pathways, FRS/FRF and bHLH transcription factors, phenylpropanoid, lignin, lignan secondary metabolic pathways, the peroxidase gene for maintaining ROS steady state, and the ankyrin gene with diverse biological functions. This study contributes valuable insights into modulating the length of wheat root hair and identifies candidate genes for the genetic improvement of wheat root traits.

Tau is a microtubule-associated protein essential for microtubule assembly and stability in neurons. The abnormal intracellular accumulation of tau aggregates is a major characteristic of brains from patients with Alzheimer’s disease (AD) and other tauopathies. In AD, the presence of neurofibrillary tangles (NFTs), which is composed of hyperphosphorylated tau protein, is positively correlated with the severity of the cognitive decline. Evidence suggests that the accumulation and aggregation of tau cause synaptic dysfunction and neuronal degeneration. Thus, the prevention of abnormal tau phosphorylation and elimination of tau aggregates have been proposed as therapeutic strategies for AD. However, currently tau-targeting therapies for AD and other tauopathies are limited. A number of dietary bioactive compounds have been found to modulate the posttranslational modifications of tau, including phosphorylation, small ubiquitin-like modifier (SUMO) mediated modification (SUMOylation) and acetylation, as well as inhibit tau aggregation and/or promote tau degradation. The advantages of using these dietary components over synthetic substances in AD prevention and intervention are their safety and accessibility. This review summarizes the mechanisms leading to tau pathology in AD and highlights the effects of bioactive compounds on the hyperphosphorylation, aggregation and clearance of tau protein. The potential of using these bioactive compounds for AD prevention and intervention is also discussed.

Super-hydrophilic and oleophobic functional materials can prevent pollution or adsorption by repelling oil, and have good circulation. However, traditional strategies for preparing these functional materials either use expensive fabrication machines or contain possibly toxic organic polymers, which may prohibit the practical application. The research of multifunctional ZnO microstructures or nanoarrays thin films with super-hydrophilic, antifouling, and antibacterial properties has not been reported yet. Moreover, the exploration of underwater oleophobic and self-cleaning antifouling properties in ZnO micro/nanostructures is still in its infancy. Here, we prepared ZnO microstructured films on fluorine-doped tin oxide substrates (F-ZMF) for the development of advanced self-cleaning type super-hydrophilic and oleophobic materials. With the increase of the accelerators, the average size of the F-ZMF microstructures decreased. The F-ZMF shows excellent self-cleaning performance and hydrophilic (water contact angle ≤ 10°) and oleophobic characteristics in the underwater antifouling experiment. Under a dark condition, F-ZMF-4 showed good antibacterial effects against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) with inhibition rates of 99.1% and 99.9%, respectively. This study broadens the application scope of ZnO-based material and provides a novel prospect for the development of self-cleaning super-hydrophilic and oleophobic materials.

To improve acquisition efficiency and achieve super high-resolution reconstruction, a computational integral imaging reconstruction (CIIR) method based on the generative adversarial network (GAN) network is proposed. Firstly, a sparse camera array is used to generate an elemental image array of the 3D object. Then, the elemental image array is mapped to a low-resolution sparse view image. Finally, a lite GAN super-resolution network is presented to up-sample the low-resolution 3D images to high-resolution 3D images with realistic image quality. By removing batch normalization (BN) layers, reducing basic blocks, and adding intra-block operations, better image details and faster generation of super high-resolution images can be achieved. Experimental results demonstrate that the proposed method can effectively enhance the image quality, with the structural similarity (SSIM) reaching over 0.90, and can also reduce the training time by about 20%.

In recent years, the phenomenon of acute poisoning and organ damage caused by organophosphorus pesticides (OPs) has been a frequent occurrence. Chlorpyrifos (CPF) is one of the most widely used organophosphorus pesticides. The main active components of ginseng stems and leaves are total ginseng stem-and-leaf saponins (GSLSs), which have various biological effects, including anti-inflammatory, antioxidant and anti-tumor activities. We speculate that these could have great potential in the treatment of severe diseases and the relief of organophosphorus-pesticide-induced side effects; however, their mechanism of action is still unknown. At present, our work aims to evaluate the effects of GSLSs on the antioxidation of CPF in vivo and in vitro and their potential pharmacological mechanisms. Mice treated with CPF (5 mg/kg) showed severe intestinal mucosal injury, an elevated diamine oxidase (DAO) index, the decreased expression of occlusive protein-1 (ZO-1) and occlusive protein, an impaired intestinal mucosal oxidation system and intestinal villi relaxation. In addition, chlorpyrifos exposure significantly increased the contents of the inflammatory factor TNF-α and the oxidative-stress-related indicators superoxide dismutase (SOD), catalase (CAT), glutathione SH (GSH), glutathione peroxidase (GSH-PX), reactive oxygen species (ROS) and total antioxidant capacity (T-AOC); elevated the level of lipid peroxide malondialdehyde (MDA); reversed the expression of Bax and caspase; and activated NF-κB-related proteins. Interestingly, GSLS supplementation at doses of 100 and 200 mg/kg significantly reversed these changes after treatment. Similar results were observed in cultured RAW264.7 cells. Using flow cytometry, Hoechst staining showed that GSLSs (30 μg/mL, 60 μg/mL) could improve the cell injury and apoptosis caused by CPF and reduce the accumulation of ROS in cells. In conclusion, GSLSs play a protective role against CPF-induced enterotoxicity by inhibiting NF-κB-mediated apoptosis and alleviating oxidative stress and inflammation.

Pigs are susceptible to cold stress due to the absence of brown fat caused by the partial deletion of uncoupling protein 1 during their evolution. Some local pig breeds in China exhibit potential cold adaptability, but research has primarily focused on fat and intestinal tissues. Skeletal muscle plays a key role in adaptive thermogenesis in mammals, yet the molecular mechanism of cold adaptation in porcine skeletal muscle remains poorly understood. This study investigated the cold adaptability of two pig breeds, Mashen pigs (MS) and Large White pigs (LW), in a four-day cold (4 °C) or normal temperature (25 °C) environment. We recorded phenotypic changes and collected blood and longissimus dorsi muscle for transcriptome sequencing. Finally, the PRSS8 gene was randomly selected for functional exploration in porcine skeletal muscle satellite cells. A decrease in body temperature and body weight in both LW and MS pigs under cold stress, accompanied by increased shivering frequency and respiratory frequency, were observed. However, the MS pigs demonstrated stable physiological homeostasis, indicating a certain level of cold adaptability. The LW pigs primarily responded to cold stress by regulating their heat production and glycolipid energy metabolism. The MS pigs exhibited a distinct response to cold stress, involving the regulation of heat production, energy metabolism pathways, and robust mitochondrial activity, as well as a stronger immune response. Furthermore, the functional exploration of PRSS8 in porcine skeletal muscle satellite cells revealed that it affected cellular energy metabolism and thermogenesis by regulating ERK phosphorylation. These findings shed light on the diverse transcriptional responses of skeletal muscle in LW and MS pigs under cold stress, offering valuable insights into the molecular mechanisms underlying cold adaptation in pigs.

The fundamental aspects of site preference of alloying elements on sublattice of the strengthen γ′ phase with L12 structure have not been well understood, which hinders the optimized design of advanced Ni-based high-temperature alloys. In this contribution, the temperature- and composition-dependent site occupying preferences of the binary, ternary, and quaternary of Ni3Al-based γ′ phase alloyed with Mi where Mi represents the additional transitional metals Co, Cr, Cu, Fe, Mn, Mo, Re, Ta, Ti, V, or W atoms (arranged in alphabetical order) chosen frequently, were studied using a two-sublattice thermodynamic model (Ni, Al, Mi)1a(Ni, Al, Mi)3c. The site occupying fractions (SOFs) were calculated based on a thermodynamic database established in this work, where the thermodynamic data of the end-members involved were obtained using first-principles calculations and phonon spectrum calculations. The calculated SOFs results show that there is an obvious site preference for stoichiometry binary Ni3Al, and its site configuration changes from (Al)1a(Ni)3c at room temperature to (Al0.9984Ni0.0015)1a (Al0Ni0.9994)3c at 1273 K. For the γ′ phase with the composition 78Ni-26Al-4Mi (atom ratio and xNi/xAl = 3:1), Mo atoms always preferred to occupy the 1a sublattice (Al site), Co, Mn, and Ti atoms always prefer the 3c sublattice (Ni site) in the whole temperature range, while the site preference of Cr, Cu, Fe, Re, Ta, V, or W atom is affected by temperature. For example, when the heat treatment temperature is lower than 700 K, Cr, Cu, Fe, Ta, V, and W atoms occupy the 1a and 3c sublattice randomly, and Re atoms prefer to 3c sublattice, while when the heat treatment temperature is higher than 1273 K, Cr, Cu, and W atoms prefer 3c sublattice, Fe and Ta atoms prefer to 1a sublattice, while all Re atoms occupy the 3c sublattice exclusively, and all V atoms occupy the 1a sublattice exclusively, respectively. Likewise, the site preference of the quaternary system with selective compositions 78Ni-26Al-2 M1-2 M2 was also predicted. Based on calculated SOFs results, the mechanical and thermodynamic properties were studied at the ground state. It has been revealed that Cr, Re, and V doping can improve the microhardness of Ni3Al alloys; in particular, the effect of Cr is extraordinary; and all elements, except Mn, Mo, and Ti, would enhance the bulk modulus of Ni3Al-based γ′ phase, in which Mn have the greatest influence on reducing the bulk and shear modulus, respectively. Furthermore, all the B/G ratios of the computed Ni3Al-based γ′ phase are >1.75, showing inherent ductility. Only Cr doping significantly enhances the Debye temperature of the Ni3Al-based γ′ phase.

Summary: Syntheses using classic coupling reactions can be tedious, leading to a dearth of ambipolar semiconducting polymers. Here, we detail the rapid synthesis of balanced ambipolar polymers by using a carbon-hydrogen (C–H) activation strategy. Using this protocol, two C–H reactive hydrogenated monomers and six ambipolar polymers can be rapidly synthesized within 2 h via C–H activation and the optoelectronic properties of the resultant polymers can be characterized. We also describe the fabrication process and performance evaluation of the ambipolar polymers-based organic transistors.For complete details on the use and execution of this protocol, please refer to Shen et al. To improve the gel properties of duck egg white gel and increase the industrial value of duck egg white, the mechanisms of ultrasound and synergetic phosphorylation/ultrasound treatments were examined in this study. It was found that as the ultrasound power increased, the surface hydrophobicity, hardness, and cohesiveness of the gel system increased, and the ζ-potential and water mobility decreased. Of the two treatments, phosphorylation/ultrasound had the strongest impact on the conformation and crystallinity of the gel system and promoted the formation of high molecular polymers. Both gel systems displayed enhanced compactness, stability, and gel strength because of the enhanced protein–protein interactions via hydrogen bonds and protein aggregation, and increased the content of intramolecular β-sheets following ultrasound treatment, and synergetic phosphorylation/ultrasound further improved the stability, water binding and gel properties. This experiment showed that ultrasound and, particularly, phosphorylation/ultrasound are effective methods to improve the gel properties of duck egg white. This study enhanced our understanding of the interactions of sodium pyrophosphate and egg white under ultrasound treatment, and promote the potential application of sodium pyrophosphate and ultrasound treatment of novel food products.

Endothelial-mesenchymal transition (EndMT) drives endothelium to contribute to atherosclerotic calcification. In a previous study, we showed that glycogen synthase kinase-3β (GSK3β) inhibition induced β-catenin and reduced mothers against DPP homolog 1 (SMAD1) in order to redirect osteoblast-like cells towards endothelial lineage, thereby reducing vascular calcification in Matrix Gla Protein (Mgp) deficiency and diabetic Ins2Akita/wt mice. Here, we report that GSK3β inhibition or endothelial-specific deletion of GSK3β reduces atherosclerotic calcification. We also find that alterations in β-catenin and SMAD1 induced by GSK3β inhibition in the aortas of Apoe−/− mice are similar to Mgp−/− mice. Together, our results suggest that GSK3β inhibition reduces vascular calcification in atherosclerotic lesions through a similar mechanism to that in Mgp−/− mice.

bHLH transcription factors are involved in multiple aspects of plant biology, such as the response to abiotic stress. Erigeron breviscapus is a composite plant, and its rich flavonoids have strong preventive and therapeutic effects on cardio cerebral vascular disease. EbbHLH80, a gene from E. breviscapus that positively regulates flavonoid synthesis, was previously characterized. However, it is unclear whether EbbHLH80 increases flavonoid accumulation, which affects salt tolerance. The function of EbbHLH80 in transgenic tobacco seeds was identified by phylogenetic analysis and metabolome-transcriptome analysis. We investigated the role of EbbHLH80 in salt stress response. Our results showed that the expression of EbbHLH80 increased following salt treatment. Integrating the metabolome and transcriptome analysis of EbbHLH80-OE and Yunyan 87 (WT) seeds, we identified several genes and metabolites related to flavonoid biosynthesis and salt stress. Moreover, EbbHLH80-OE plants displayed higher salt tolerance than wild-type plants during seed germination and seedling growth. After salt treatment, transgenic tobacco had significantly lower levels of reactive oxygen species (ROS) than WT, with enhanced levels of antioxidant enzyme expression. Altogether, our results demonstrated that EbbHLH80 might be a positive regulator, promoting salt tolerance by modulating ROS scavenging and increasing stress-responsive genes.

The radial gate corbel beam is a kind of gate support structure more often used in large and medium-sized reservoirs, but the current corresponding structural design code does not give a review calculation method for it. Because there are obvious differences between a corbel and a corbel beam in structural form and force, if the corbel beam is just simplified in the calculation as a corbel structure, it can easily lead to misjudgment of insufficient bearing capacity. This misjudgment has a significant impact on later safety assessment, danger removal, and reinforcement. Currently, there are limited studies available on the internal stress distribution of the corbel beam. In this study, taking the danger removal and reinforcement of the radial gate beam of a medium reservoir in Beijing as an example, the concrete quality of the dam was tested by the core drilling method, and two safety review methods of the corbel beam for different types of reservoirs were proposed in combination with the Code for Design of Hydraulic Concrete Structures (SL191-2008). Then, three different types of calculation model were established by the method of theoretical mechanics calculation and finite element simulation. Combined with the safety test data, the stress state of the combined stress structure of the corbel beam and gate pier under the real state was analyzed and the safety evaluation was carried out. The calculation results of these two corbel beam safety review methods were respectively reduced by 32% and 47% compared with the current calculation method. Engineering practice has proved the rationality of the two safety evaluation methods proposed in this paper, which can provide a certain reference for similar engineering reinforcement.

Artificial photosynthesis is promising to convert solar energy and CO2 into valuable chemicals, and to alleviate the problems of the greenhouse effect and the climate change crisis. Here, we fabricated a novel photocatalyst by directly growing Bi2MoO6 nanosheets on three-dimensional (3D) N,O-doped carbon (NO-C). Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) show that the designed photocatalyst ensured the close contact between Bi2MoO6 and NO-C, and reduced the stacking of the NO-C layers to provide abundant channels for the diffusion of CO2, while NO-C can allow for fast electron transfer. The charge transfer in this composite was determined to follow a step-scheme mechanism, which not only facilitates the separation of charge carriers but also retains a strong redox capability. Benefiting from this unique 3D structure and the synergistic effect, BMO/NO-C showed excellent performance in photocatalytic CO2 reductions. The yields of the best BMO/NO-C catalysts for CH4 and CO were 9.14 and 14.49 μmol g−1 h−1, respectively. This work provides new insights into constructing step-scheme photocatalytic systems with the 3D nanostructures.

Early and sensitive detection of δ-aminolevulinic acid (δ-ALA) and porphobilinogen (PBG) is the cornerstone of diagnosis and effective treatment for acute porphyria. However, at present, the quantifying strategies demand multiple solvent extraction steps or chromatographic approaches to separate δ-ALA and PBG prior to quantification. These methods are both time-consuming and laborious. Otherwise, in conventional spectrofluorimetry, the overlapping spectra of the two analytes cause false diagnosis. To overcome this challenge, we present a two-step approach based on derivative matrix-isopotential synchronous fluorescence spectrometry (DMISFS) and the Hantzsch reaction, realizing the simple and simultaneous detection of δ-ALA and PBG in urine samples. The first step is chemical derivatization of the analytes by Hantzsch reaction. The second step is the determination of the target analytes by combining MISFS and the first derivative technique. The proposed approach accomplishes following advantages: 1) The MISFS technique improves the spectral resolution and resolves severe spectral overlap of the analytes, alleviating tedious and complicated pre-separation processes; 2) First derivative technique removes the background interference of δ-ALA on PBG and vice versa, ensuring high sensitivity; 3) Both the analytes can be determined simultaneously via single scanning, enabling rapid detection. The obtained detection limits for δ-ALA and PBG were 0.04 μmol L−1 and 0.3 μmol L−1, respectively. Within-run precisions (intra and inter-day CVs) for both the analytes were

Amomum tsaoko (AT) is a dietary botanical with laxative properties; however, the active ingredients and mechanisms are still unclear. The active fraction of AT aqueous extract (ATAE) for promoting defecation in slow transit constipation mice is the ethanol-soluble part (ATES). The total flavonoids of ATES (ATTF) were the main active component. ATTF significantly increased the abundance of Lactobacillus and Bacillus and reduced the dominant commensals, such as Lachnospiraceae, thereby changing the gut microbiota structure and composition. Meanwhile, ATTF changed the gut metabolites mainly enriched in pathways such as the serotonergic synapse. In addition, ATTF increased the serum serotonin (5-HT) content and mRNA expression of 5-hydroxytryptamine receptor 2A (5-HT2A), Phospholipase A2 (PLA2), and Cyclooxygenase-2 (COX2), which are involved in the serotonergic synaptic pathway. ATTF increased Transient receptor potential A1 (TRPA1), which promotes the release of 5-HT, and Myosin light chain 3(MLC3), which promotes smooth muscle motility. Notably, we established a network between gut microbiota, gut metabolites, and host parameters. The dominant gut microbiota Lactobacillus and Bacillus, prostaglandin J2 (PGJ2) and laxative phenotypes showed the most significant associations. The above results suggest that ATTF may relieve constipation by regulating the gut microbiota and serotonergic synaptic pathway and has great potential for laxative drug development in the future.

Endothelial–mesenchymal transition (EndMT) drives the endothelium to contribute to vascular calcification in diabetes mellitus. In our previous study, we showed that glycogen synthase kinase-3β (GSK3β) inhibition induces β-catenin and reduces mothers against DPP homolog 1 (SMAD1) to direct osteoblast-like cells toward endothelial lineage, thereby reducing vascular calcification in Matrix Gla Protein (Mgp) deficiency. Here, we report that GSK3β inhibition reduces vascular calcification in diabetic Ins2Akita/wt mice. Cell lineage tracing reveals that GSK3β inhibition redirects endothelial cell (EC)-derived osteoblast-like cells back to endothelial lineage in the diabetic endothelium of Ins2Akita/wt mice. We also find that the alterations in β-catenin and SMAD1 by GSK3β inhibition in the aortic endothelium of diabetic Ins2Akita/wt mice are similar to Mgp−/− mice. Together, our results suggest that GSK3β inhibition reduces vascular calcification in diabetic arteries through a similar mechanism to that in Mgp−/− mice.

Abstract Protein derived from purple wheat bran was hydrolyzed sequentially using alcalase proteases for the production of antioxidant peptides. Purple wheat bran protein (PWBP) hydrolysates were fractionated using size-exclusion (G-25) and ion-exchange chromatography methods to identify the structure of antioxidant peptides. The free radical scavenging activity of peptides purified from PWBP hydrolysates was evaluated using superoxide anion radical-scavenging activity and determination assays of Trolox equivalent antioxidant capacity (TEAC). Results demonstrated that purple wheat bran peptide F4-4 exhibited the highest antioxidant activity among other hydrolysates. F4-4 was further identified as Cys-Gly-Phe-Pro-Gly-His-Cys, Gln-Ala-Cys, Arg-Asn-Phe, Ser-Ser-Cys, and Trp-Phe by high performance liquid chromatography (HPLC) spectrometer coupled with Orbitrap Elite™ mass spectrometer (LC–MS/MS). Antioxidant peptides 2 and 4 showed improved stability when the temperature was lower than 80 °C. These peptides also demonstrated good digestive stability in vitro system by simulating gastrointestinal digestion.

In this paper, a series of quasi-static cyclic shear tests were performed on artificial rock joint specimens with natural morphologies at different normal stress levels. After shearing, combined with 3D scanning technology and image processing, the 3D morphology parameter, A0θmax∗/(C+1), of 36 analyzed directions was measured to investigate the damage and degradation laws of the joint surfaces. The polar curves of the roughness parameter shrank from a circular to an elliptical shape, with the shear direction as the minor axis. The roughness degradation rates and the shear cycles were mutually interrelated with the normal stress levels, which had a tendency to be fast initially and then to slow down under higher normal stress. Influenced by the cyclic shear direction, the 3D roughness of the joint surfaces degenerated anisotropically and sustained the most serious damage along the shear direction. Two damage and degradation fitting formulas were proposed, which could aid the assessment of residual roughness under different stress histories and the cumulative wear degree.

In order to improve the characteristics of egg white protein gel and the stability of egg white protein gel products. In this work, the changes of the texture, physicochemical properties, secondary structure and microstructure of ultrasound modified egg white protein gel (UEP) and tea polyphenols (TP) assisted ultrasound modification of egg white protein gel (TUEP) with different ultrasonic power (0–360 W) were studied. With the increase of ultrasonic power, soluble protein, surface hydrophobicity and disulfide bonds of TUEP and UEP showed an increasing trend. In particular, with the increase of ultrasonic power, the content of intramolecular β-sheets and α-helices of TUEP showed an increasing trend, and significantly improved the relaxation time and microstructure, thus enhancing TUEP gel stability. In addition, the hardness and water holding capacity (WHC) of TUEP and UEP can be increased by ultrasonic treatment, and when the ultrasonic power reached 120 W, the hardness and WHC reached the maximum. The hardness and WHC of TUEP were significantly better than that of UEP. SDS-PAGE results showed that the peptide chain of protein without being broken under ultrasonic treatment. Ultrasonic treatment can improve the gel strength of egg white protein by promoting the cross-linking of proteins to form a dense gel structure, and egg white protein gel form more disulfide bonds and a more stable gel conformation under TP assisted ultrasound. In conclusion, ultrasound and TP assisted ultrasound modification of egg white protein gel is a reliable technique, which can improve the value of egg white protein in food processing.

The heavy use of antibiotics in medicine, stock farming and agriculture production has led to their gradual accumulation in environmental media, which poses a serious threat to ecological environment and human safety. As an efficient and promising catalyst for the degradation of antibiotics, nanoscale zero valent iron (nZVI) has attracted increasing attention in recent years. In this study, sepiolite nanofiber supported zero valent iron (nZVI/SEP) composite was prepared via a facile and environmentally friendly method. The nZVI particles (with size of 20–60 nm) were dispersed evenly on the surface of sepiolite nanofibers, and the catalytic performance for the removal of tetracycline hydrochloride (TC-HCl) in aqueous system was investigated. The effect of nZVI loading amount, catalyst dosage, H2O2 concentration and pH on the removal efficiency of TC-HCl were studied. It was revealed that the sepiolite supporter effectively inhibited the agglomeration of nZVI particles and increased the contact area between contaminant and the active sites, resulting in the higher catalytic performance than pure nZVI material. The TC-HCl removal efficiency of nZVI/SEP composite was up to 92.67% when TC-HCl concentration of 20 mg/L, catalyst dosage of 1.0 g/L, H2O2 concentration of 1.0 mM, pH value of 7. Therefore, the nZVI/SEP composites possess high catalytic activity for TC-HCl removal and have great application prospects in antibiotic wastewater treatment.

Rational design of sulfur hosts for lithium-sulfur (Li-S) batteries is essential to address the shuttle effect and accelerate reaction kinetics. Herein, the composites of bimetallic sulfide CuCo2S4 loaded on carbon nanotubes (CNTs) are prepared by hydrothermal method. By regulating the loading of CuCo2S4 nanoparticles, it is found that when Cu2+ and CNT are prepared in a 10:1 ratio, the CuCo2S4 nanoparticles loaded on the CNT are relatively uniformly distributed, avoiding the occurrence of agglomeration, which improves the electrical conductivity and number of active sites. Through a series of electrochemical performance tests, the S/CuCo2S4-1/CNT presents a discharge specific capacity of 1021 mAh g−1 at 0.2 C after 100 cycles, showing good cycling stability. Even at 1 C, the S/CuCo2S4-1/CNT cathode delivers a discharge capacity of 627 mAh g−1 after 500 cycles. This study offers a promising strategy for the design of bimetallic sulfide-based sulfur hosts in Li-S batteries.

Brassinosteroid (BR) signaling is very important in plant developmental processes. Its various components interact to form a signaling cascade. These components are widely studied in Arabidopsis; however, very little information is available on tomatoes. Brassinosteroid Insensitive 2 (BIN2), the downstream suppressor of BR signaling, plays a critical role in BR signal pathway, while FRIGIDA as a key suppressor of Flowering Locus C with overexpression could cause early flowering; however, how the BR signaling regulates FRIGIDA homologous protein to adjust flowering time is still unknown. This study identified 12 FRIGIDA-LIKE proteins with a conserved FRIGIDA domain in tomatoes. Yeast two-hybrid and BiFC confirmed that SlBIN2 interacts with 4 SlFRLs, which are sub-cellularly localized in the nucleus. Tissue-specific expression of SlFRLs was observed highly in young roots and flowers. Biological results revealed that SlFRLs interact with SlBIN2 to regulate early flowering. Further, the mRNA level of SlBIN2 also increased in SlFRL-overexpressed lines. The relative expression of SlCPD increased upon SlFRL silencing, while SlDWF and SlBIN2 were decreased, both of which are important for BR signaling. Our research firstly provides molecular evidence that BRs regulate tomato flowering through the interaction between SlFRLs and SlBIN2. This study will promote the understanding of the specific pathway essential for floral regulation.

Silicon-based photovoltaic technology is helpful in reducing the cost of power generation; however, it suffers from economic losses and environmental pollution caused by silicon cutting waste. Herein, a hydrothermal method accompanied by heat treatment is proposed to take full advantage of the photovoltaic silicon cutting waste and biomass excrementum bombycis to fabricate flake-like porous Si@C (FP-Si@C) composite anodes for lithium-ion batteries (LIBs). The resulting FP-Si@C composite with a meso-macroporous structure can buffer the severe volume changes and facilitate electrolyte penetration. Meanwhile, the slightly graphitic carbon with high electrical conductivity and mechanical strength tightly surrounds the Si nanoflakes, which not only contributes to the ion/electron transport but also maintains the electrode structural integrity during the repeated lithiation/delithiation process. Accordingly, the synergistic effect of the unique structure of FP-Si@C composite contributes to a high discharge specific capacity of 1322 mAh g−1 at 0.1 A g−1, superior cycle stability with a capacity retention of 70.8% after 100 cycles, and excellent rate performance with a reversible capacity of 406 mAh g−1 at 1.0 A g−1. This work provides an easy and cost-effective approach to achieving the high-value application of photovoltaic silicon cutting waste, as well as obtaining high-performance Si-based anodes for LIBs.

Carbon dots (CDs) have caught enormous attention owing to their distinctive properties, such as their high water solubility, tunable optical properties, and easy surface modification, which can be generally used for the detection of heavy metals and organic pollutants. Herein, nitrogen and fluorine co-doped carbon dots (NFCDs) were designed via a rapid, low-cost, and one-step microwave-assisted technique using DL-malic acid and levofloxacin. The NFCDs emitted intense green fluorescence under UV lighting, and the optical emission peak at 490 nm was observed upon a 280 nm excitation, with a high quantum yield of 21.03%. Interestingly, the spectral measurements illustrated excitation-independent and concentration-independent single-color fluorescence owing to the presence of nitrogen and fluorine elements in the surface functional groups. Additionally, the NFCDs were applied for the selective detection of Fe3+ and ascorbic acid based on the “turn-off” mode. The detection limits were determined as 1.03 and 4.22 µM, respectively. The quenching mechanisms were explored using the static quenching mechanism and the inner filter effect. Therefore, a NFCDs fluorescent probe with single color emission was successfully developed for the convenient and rapid detection of Fe3+ and ascorbic acid in environments.

Plant laccase genes belong to a multigene family, play key roles in lignin polymerization, and participate in the resistance of plants to biotic and abiotic stresses. Switchgrass is an important resource for forage and bioenergy production, yet information about the switchgrass laccase gene family is scarce. Using bioinformatic approaches, a genome-wide analysis of the laccase multigene family in switchgrass was carried out in this study. In total, 49 laccase genes (PvLac1 to PvLac49) were identified; these can be divided into five subclades, and 20 of them were identified as targets of miR397. The tandem and segmental duplication of laccase genes on Chr05 and Chr08 contributed to the expansion of the laccase family. The laccase proteins shared conserved signature sequences but displayed relatively low sequence similarity, indicating the potential functional diversity of switchgrass laccases. Switchgrass laccases exhibited distinct tissue/organ expression patterns, revealing that some laccases might be involved in the lignification process during stem development. All five of the laccase isoforms selected from different subclades responded to heavy metal. The immediate response of lignin-related laccases, as well as the delayed response of low-abundance laccases, to heavy-metal treatment shed light on the multiple roles of laccase isoforms in response to heavy-metal stress.

In this work, SnO2 nanoflowers were prepared by a simple one-step hydrothermal process. The morphology and structure of SnO2 nanoflowers were characterized by SEM, TEM, Raman spectroscopy, and XRD, which demonstrated the good crystallinity of the SnO2 tetrahedron structure of the as-synthesized materials. In addition, the sensing properties of SnO2 nanoflowers were studied in detail. It was found that the SnO2 nanoflower-based gas sensor exhibits excellent gas response (9.2 to 120 ppm), fast response and recovery (2/15 s to 6 ppm), good linearity of correlation between response (S) vs. concentration (C) (lgS = 0.505 lgC − 0.147, R2 = 0.9863), superb repeatability, and selectivity at 300 °C. The outstanding performance can also be attributed to the high specific surface area ratio and size of SnO2 nanoflowers close to the thickness of the electron depletion layer that can provide abundant active sites, promote the rate of interaction, and make it easier for gas molecules to diffuse into the interior of the material. Therefore, SnO2 nanoflowers can be an ideal sensing material for real-time monitoring of low-concentration HCHO.

Witches’-broom (WB, excessive initiation, and outgrowth of axillary buds) is one of the remarkable symptoms in plants caused by phytoplasmas, minute wall-less intracellular bacteria. In healthy plants, axillary bud initiation and outgrowth are regulated by an intricate interplay of nutrients (such as sugars), hormones, and environmental factors. However, how these factors are involved in the induction of WB by phytoplasma is poorly understood. We postulated that the WB symptom is a manifestation of the pathologically induced redistribution of sugar and phytohormones. Employing potato purple top phytoplasma and its alternative host tomato (Solanum lycopersicum), sugar metabolism and transportation, and the spatiotemporal distribution of phytohormones were investigated. A transmission electron microscopy (TEM) analysis revealed that starch breakdown was inhibited, resulting in the degradation of damaged chloroplasts, and in turn, premature leaf senescence. In the infected source leaves, two marker genes encoding asparagine synthetase (Sl-ASN) and trehalose-6-phosphate synthase (Sl-TPS) that induce early leaf senescence were significantly up-regulated. However, the key gibberellin biosynthesis gene that encodes ent-kaurene synthase (Sl-KS) was suppressed. The assessment of sugar content in various infected tissues (mature leaves, stems, roots, and leaf axils) indicated that sucrose transportation through phloem was impeded, leading to sucrose reallocation into the leaf axils. Excessive callose deposition and the resulting reduction in sieve pore size revealed by aniline blue staining and TEM provided additional evidence to support impaired sugar transport. In addition, a spatiotemporal distribution study of cytokinin and auxin using reporter lines detected a cytokinin signal in leaf axils where the axillary buds initiated. However, the auxin responsive signal was rarely present in such leaf axils, but at the tips of the newly elongated buds. These results suggested that redistributed sucrose as well as cytokinin in leaf axils triggered the axillary bud initiation, and auxin played a role in the bud elongation. The expression profiles of genes encoding squamosa promoter-binding proteins (Sl-SBP1), and BRANCHED1 (Sl-BRC1a and Sl-BRC1b) that control axillary bud release, as determined by quantitative reverse transcription (qRT)-PCR, indicated their roles in WB induction. However, their interactions with sugars and cytokinins require further study. Our findings provide a comprehensive insight into the mechanisms by which phytoplasmas induce WB along with leaf chlorosis, little leaf, and stunted growth.

Responsive slippery lubricant-infused surfaces (SLIS) have attracted substantial attention because of the high demand of fundamental research and practical applications, such as controllable liquid-repellency, intelligent, and easy-to-implement wettability switching. In this review, advanced development of responsive slippery surfaces is briefly summarized upon various external stimuli, including stress, electrical field, magnetic field, and temperature. In addition, remaining challenge and prospect are also discussed.

Fluorine is widely dispersed in nature and has multiple physiological functions. Although it is usually regarded as an essential trace element for humans, this view is not held universally. Moreover, chronic fluorosis, mainly characterized by skeletal fluorosis, can be induced by long-term excessive fluoride consumption. High concentrations of fluoride in the environment and drinking water are major causes, and patients with skeletal fluorosis mainly present with symptoms of osteosclerosis, osteochondrosis, osteoporosis, and degenerative changes in joint cartilage. Etiologies for skeletal fluorosis have been established, but the specific pathogenesis is inconclusive. Currently, active osteogenesis and accelerated bone turnover are considered critical processes in the progression of skeletal fluorosis. In recent years, researchers have conducted extensive studies in fields of signaling pathways (Wnt/β-catenin, Notch, PI3K/Akt/mTOR, Hedgehog, parathyroid hormone, and insulin signaling pathways), stress pathways (oxidative stress and endoplasmic reticulum stress pathways), epigenetics (DNA methylation and non-coding RNAs), and their inter-regulation involved in the pathogenesis of skeletal fluorosis. In this review, we summarised and analyzed relevant findings to provide a basis for comprehensive understandings of the pathogenesis of skeletal fluorosis and hopefully propose more effective prevention and therapeutic strategies.

Hepatitis B virus (HBV) infection is closely related to hepatocellular carcinoma (HCC) development. To investigate the mechanism of HBV causing HCC, we previously analyzed the transcription of the HBV-transgenic cell line HepG2-4D14 and parental HepG2 cells and identified a subset of long noncoding RNAs (lncRNAs) differentially expressed between them. In this study, we focus on lncRNA LINC01010, as it is significantly downregulated in HepG2-4D14 cells and in liver tissues of HCC patients, and positively correlated with survival. We found that HBV-encoded HBx can reduce the transcription of LINC01010. Functional analysis showed that the overexpression of LINC01010 inhibits proliferation, migration and invasion of HepG2 cells while the knockdown of LINC01010 promotes these processes. By taking the approach of RNA immunoprecipitation (RIP) and mass spectrometry, we identified that LINC01010 can interact with vimentin. Further studies demonstrated that LINC01010 negatively affects the vimentin network extension and causes more rapid subunit exchange and lower stability of vimentin filaments. In addition, LINC01010 can reduce the amount of insoluble vimentin within cells, which suggests that LINC01010 interfers with vimentin polymerization. These data indicate that LINC01010 can inhibit the assembly of vimentin filament. Thus, we revealed that HBV HBx-downregulated LINC01010, which suppresses cell proliferation and migration by negatively regulating the formation of vimentin filament. Taken together, LINC01010 is a potential tumor suppressor that may restrain HBV-related HCC development.
