The A-kinase anchoring protein 150 (AKAP150) organizes kinases and phosphatases to regulate AMPA receptors (AMPARs) that are pivotal for synaptic plasticity. AKAP150 itself undergoes S-palmitoylation. However, the roles of AKAP150 and its palmitoylation in spinal nociceptive processing remain unknown. In this study, we found that intraplantar injection of complete Freund's adjuvant (CFA) significantly increased the synaptic expression of AKAP150 and caused a reorganization of AKAP150 signaling complex in spinal dorsal horn. Knockdown of AKAP150 or interruption of its interactions with kinases effectively suppressed the CFA-induced synaptic expression of GluA1 subunit of AMPARs. Our data also showed that an upregulation of AKAP150 palmitoylation was involved in the synaptic redistribution of AKAP150. Inhibition of AKAP150 palmitoylation by expression of palmitoylation-defective mutant AKAP150 (C36, 123S) effectively repressed the CFA-induced phosphorylation and synaptic expression of GluA1 subunit, meanwhile, attenuated the development of mechanical allodynia and thermal hyperalgesia. Furthermore, we found that an increased expression of palmitoyl acyltransferase ZDHHC2 contributed to the upregulation of AKAP150 palmitoylation and GluA1 accumulation in inflamed mouse. These data indicated that AKAP150 and its palmitoylation were involved in AMPA receptor-dependent modification of nociceptive transmission, and the manipulations of AKAP150 signaling complex and palmitoylation might serve as potential therapeutic strategies for persistent pain after inflammation.

Photochemistry to prepare platinum nanoparticles (Pt NPs) is an essential way to control Pt NPs catalyst size distribution. This article reports a series of morphologically controlled syntheses of Pt NPs loaded on modified carbon nanotube (Pt-CNTs). In the synthesis, Polyethylene glycol (PEG) participates in reactions both as a reducing agent and a stabilizer. Visible light irradiation was adopted as a kinetic controlling approach. Typical 4-nitrophenol (4-NP) reduction was adopted to probe the catalytic performances. Characterizations prove that visible light irradiation is an effective way to control the reaction process. In the optimized reaction conditions, i.e., when the ratio PEG:H2O is 1:9, and the pH is 10, the as-prepared Pt NPs are consequently in a very narrow sized distribution with an average diameter of 1.29 nm. The Pt-CNTs present a high reaction rate constant of 0.624 min-1 in the catalytic reduction of 4-NP. All the research results are beneficial for exploring more green chemistry and facile photochemical approaches in the controlled preparation of Pt nanocatalysts.

Colorectal cancer (CRC) is a common cancer with high metastatic property. Circular RNAs (circRNAs) have important involvement in cancer processes. This study focused on the regulation of circRNA RAD23 homologue B (circRAD23B) in CRC. The levels of circRAD23B, microRNA-1205 (miR-1205), and tripartite motif-44 (TRIM44) were examined by quantitative real-time polymerase chain reaction (qRT-PCR). Functional analyses were performed by Cell Counting Kit-8 (CCK-8) for cell proliferation, flow cytometry for cell cycle or cell apoptosis, and transwell assay for cell migration and invasion. Western blot was administrated for protein detection. The interaction of targets was analyzed by dual-luciferase reporter and RNA pull-down assays. The in vivo experiment was conducted via xenograft tumor in mice. We identified that circRAD23B was overexpressed in CRC tissues and cells. CRC cell proliferation, cell cycle progression, and cell metastasis were inhibited, while apoptosis was promoted by downregulating circRAD23B. Target analysis indicated that circRAD23B-targeted miR-1205 and TRIM44 were downstream genes of miR-1205. Moreover, the antitumor response of circRAD23B downregulation and miR-1205 overexpression was, respectively, achieved by increasing miR-1205 and decreasing TRIM44. CircRAD23B could regulate TRIM44 level by sponging miR-1205. In vivo, circRAD23B knockdown also reduced CRC tumorigenesis via the miR-1205/TRIM44 axis. These results suggested that the inhibition of circRAD23B retarded the progression of CRC via acting on the miR-1205/TRIM44 axis. CircRAD23B might be a novel target in CRC treatment.

A highly selective turn-on fluorescent and naked-eye colourimetric dual-channel probe for cyanide anions (CNˉ) has been designed and characterized. In the mixed solution (DMSO / H2O, 9:1, v / v), only CNˉ could cause an increase in the UV absorption intensity and the corresponding fluorescence intensity increased, and other anions had no significant effect on the probe. After treatment with cyanide in the probe solution, the solution showed a noticeable colour change, from light yellow to purple. Moreover, a fluorescence spectrophotometer can be used to observe that the fluorescence intensity of the solution is significantly enhanced. The response of the colourimetric and fluorescent dual-channel probe to CNˉ was attributed to nucleophilic addition, and the mechanism was determined by 1H NMR spectroscopy. In addition, this probe was used to detect CNˉ in actual water samples, including river water, drinking water, and tap water. Based on tetraphenylethylene as the skeleton, two novel salamo-type fluorescent probes (TPES1 = 2-hydroxy-1-naphthaldehyde O-(3-(((-2-hydroxy-5-(1,2,2-triphenylvinyl)benzylidene)amino)oxy)propyl) oxime and TPES2 = bis(2-hydroxy-1-naphthaldehyde O-(3-(((2-hydroxy-5-(1,2,2-triphenylvinyl)benzylidene)amino)oxy)propyl) dioxime) were designed and synthesized. Through the colorimetric and fluorescence dual-channel modes, cyanide ions could be specifically recognized by the two probe molecules. Interestingly, the probes TPES1 and TPES2 exhibited completely different spectral behaviors during the recognition process. Of the two, the TPES1 molecule is a ratio-type fluorescent probe, and double fluorescence emission peaks appeared during the titration process. The reason for this phenomenon is due to the rotation of the C–C single bond in tetraphenylethylene, causing the solution system to contain molecules in two states. On the other hand, the TPES2 molecule is a fluorescence-enhanced probe that has undergone two reaction processes of deprotonation and addition reaction successively, causing the fluorescence intensity to increase continuously. Theoretical calculations indicated that the configuration of probe TPES1 is reversed after interacting with cyanide ions (CN−). By calculating the minimum detection limits of TPES1 and TPES2 to CN−, the LOD values were 8.56 × 10−8 M and 5.37 × 10−8 M, respectively. In addition, probe-based test strips have been prepared for quick and simple detection of cyanide ions.

Acute lung injury (ALI) is a common critical illness in clinical anesthesia and the intensive care unit that can cause acute hypoxic respiratory insufficiency. Despite various therapeutic regimes having been investigated, there is currently no effective pharmacotherapy available to treat ALI. Previous studies have reported that the NOD‑like receptor protein 3 (NLRP3) signaling pathway plays an important role in the inflammatory response and is involved in the pathogenesis of ALI. Moreover, dexmedetomidine (Dex), an α2‑adrenergic receptor activating agent, has been routinely used as an adjuvant therapy in treating inflammatory diseases, including ALI. However, the precise pathological mechanisms of Dex in ALI remain to be elucidated. Thus, the present study aimed to investigate the effects of the p38/heme oxygenase 1 (HO‑1) signaling pathways in the pathological mechanisms of Dex in ALI. Newborn male Sprague‑Dawley rats (n=48) were randomly divided into four groups (n=12 each), and an intravenous injection of lipopolysaccharide (LPS) was used to successfully induce the ALI model, with increased pulmonary damage, cell apoptosis, interleukin‑1β (IL‑1β) secretion and edema fluid in lungs. Moreover, the mRNA and protein expression levels of NLRP3 were significantly upregulated, while that of HO‑1 were downregulated by LPS treatment. Furthermore, the levels of phosphorylated p38 were also upregulated in ALI rats. It was demonstrated that Dex administration significantly alleviated LPS‑induced ALI, downregulated the secretion of IL‑1β, decreased the expression of NLRP3, inhibited the phospho‑activation of p38 and increased HO‑1 expression. In addition, pharmacological inhibition of p38 using the inhibitor SB20380 further enhanced the effect of Dex. Collectively, these preliminarily results identified the effects of Dex intervention on the pathogenesis of ALI via the regulation of p38/HO‑1 signaling pathways, which impacted the inflammatory effects, thus providing a theoretical basis and novel evidence for the development of new targets for clinical treatment of ALI.

A bifunctional secondary phosphine oxide (SPO) ligand-controlled method was developed for Ni-Al-catalyzed nonchelated dual C-H annulation of arylformamides with alkynes, providing a series of substituted amide-containing heterocycles in ≤97% yield. The SPO-bound bimetallic catalysis proved to be critical to the reaction efficiency.

Soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) have been implicated in the trafficking of postsynaptic glutamate receptors, including N-methyl-d-aspartate (NMDA)-subtype glutamate receptors (NMDARs) that are critical for nociceptive plasticity and behavioral sensitization. However, the components of SNAREs complex involved in spinal nociceptive processing remain largely unknown. Here we found that SNAP25, syntaxin4, VAMP2 and Munc18-1 were localized at postsynaptic sites and formed the complex in the superficial lamina of spinal cord dorsal horn of rats. The complex formation between these SNAREs components were accelerated after intraplantar injection of complete Freund's adjuvant (CFA), pharmacological removal of GABAergic inhibition or activation of NMDAR in intact rats. The increased SNAP25/syntaxin4/VAMP2/Munc18-1 interaction facilitated the surface delivery and synaptic accumulation of NMDAR during inflammatory pain. Disruption of the molecular interaction between SNAP25 with its SNARE partners by using a blocking peptide derived from the C-terminus of SNAP25 effectively repressed the surface and synaptic accumulation of GluN2B-containing NMDARs in CFA-injected rats. This peptide also alleviated inflammatory mechanical allodynia and thermal hypersensitivity. These data suggested that SNAREs complex assembly in spinal cord dorsal horn was involved in the inflammatory pain hypersensitivity through promoting NMDAR synaptic trafficking.

Layered molybdenum disulfide (MoS 2 ) is one of the representative two-dimensional (2D) semiconductors, offering a tantalizing prospect in the application of nanoelectronic devices and circuits. Here abnormal electrical and optoelectrical characteristics in transferred multilayer MoS 2 transistors are observed and discussed. Such phenomena can be explained by the MoS 2 intrinsic defects and the trapped charge impurities at the MoS 2 -dielectric and MoS 2 -MoS 2 interfaces, as well as its charge screening effect. Our findings highlight the significance of controlling the dielectric interface and the material quality of 2D semiconductors, which are critical for the reliability of future 2D nanoelectronic devices.

Three Ni(II) complexes {[NiL(μ-OAc)(CH3CH2OH)]2Ni} (1), {[NiL(μ-OAc)(CH3OH)]2Ni]·2CH2Cl2·2CH3OH (2) and {[NiL(μ-OAc)(CH3OH)]2Ni}·1.5CH3OH (3) based on a Salamo-type bis-oxime ligand (H2L = 2,2′-[ethylenedioxybis(nitrilo-methylidyne)]dinaphthol) have been synthesized and characterized by physicochemical and spectroscopic methods. The complexes 1, 2 and 3 are all trinuclear structures, and the center Ni(II) ions are six-coordinated and show the slightly distorted octahedral coordination. They all consist of three Ni(II) ions, two deprotonated L2- units, two μ-acetato ligands and two coordinated solvent molecules, as well as the crystallizing solvent molecules. Although the molecular structures of the Ni(II) complexes 1, 2 and 3 are similar to each other, obtained in different solvents, the supramolecular structures are entirely different. The complexes 1, 2 and 3 possess the self-assembled infinite 1D, 3D and 2D supramolecular structures via different intermolecular interactions (hydrogen bonds, C–H···π and π···π stacking interaction), respectively. But complex 2 formed a 3D structures by intramolecular hydrogen bonds. It is significant that the solvent molecules have obvious effects on the optical properties and molecular configuration. The Hirshfeld surface analysis showed that the short-range forces of the hydrogen bonds in complexes 1, 2 and 3 accounted for 67.8%, 44.6% and 52.7%, respectively. The molecular orbital energies for ligand H2L and complex 1 were obtained by DFT calculation, and the electron distribution, energy level and energy gap of HOMO and LUMO were given.

This paper presents the visible light-assisted synthesis of surfactant-free Pt nanoparticles (Pt-NPs) supported on multi-walled carbon nanotubes (MWCNTs) at ambient condition. Transmission electron microscopy (TEM) images indicate that Pt-NPs with high density and relative small in size (about 2 nm) were monodispersed on the surface of MWCNTs. The characterizations of Fourier transform infrared spectroscopy (FT-IR), X-ray photoelectron spectroscopy (XPS), X-ray diffractometer (XRD) and ultraviolet–visible spectroscopy (UV–Vis) prove that visible light irradiation significantly affect the species distribution of [PtCl4]2− precursor in the hydrolysis and then promote hydrolysate Pt oxides loading on MWCNTs. The loading process decides the morphology of Pt-NPs rather than the following precursor reduction, which does not change the morphology but only the valance state of platinum. The catalytic activity of Pt/MWCNTs was tested in subsequent methanol electrochemical reactions and 4-nitrophenol (4-NP) reduction.

Chimeric antigen receptor T (CAR-T) cell therapy is a novel cellular immunotherapy that is widely used to treat hematological malignancies, including acute leukemia, lymphoma, and multiple myeloma. Despite its remarkable clinical effects, this therapy has side effects that cannot be underestimated. Cytokine release syndrome (CRS) is one of the most clinically important and potentially life-threatening toxicities. This syndrome is a systemic immune storm that involves the mass cytokines releasing by activated immune cells. This phenomenon causes multisystem damages and sometimes even death. In this study, we reported the management of a patient with recurrent and refractory multiple myeloma and three patients with acute lymphocytic leukemia who suffered CRS during CAR-T treatment. The early application of tocilizumab, an anti-IL-6 receptor antibody, according to toxicity grading and clinical manifestation is recommended especially for patients who suffer continuous hyperpyrexia, hypotensive shock, acute respiratory failure, and whose CRS toxicities deteriorated rapidly. Moreover, low doses of dexamethasone (5-10 mg/day) were used for refractory CRS not responding to tocilizumab. The effective management of the toxicities associated with CRS will bring additional survival opportunities and improve the quality of life for patients with cancer.

Triptolide is a major active ingredient isolated from the traditional Chinese herb Tripterygium wilfordii Hook F. However, its use in clinical practice is limited due to its severe hepatotoxicity. Autophagy, a highly conserved intracellular process, is essential for maintaining cytoplasmic homeostasis. Considering that abnormalities in autophagy are closely associated with drug-mediated hepatotoxicity, we applied human normal liver HL7702 cells to elucidate the roles of autophagy in triptolide-induced hepatotoxicity. Our study revealed that triptolide was cytotoxic to HL7702 cells. It markedly increased autophagosome formation and expression of autophagy-related proteins, namely Beclin1 and microtubule-associated protein 1 light chain 3II, and induced oxidative stress. These proautophagic effects were counteracted by pretreatment with N-acetylcysteine, a reactive oxygen species scavenger. Moreover, the pharmacological suppression of autophagy further exacerbated triptolide-elicited decrease in cell viability, increase in lactate dehydrogenase leakage, and activation of apoptosis proteases (caspase 3 and caspase 9). Our findings suggest that triptolide-induced oxidative stress consequently enhances autophagic activity, and autophagy is a cytoprotective mechanism against triptolide-induced cytotoxicity in HL7702 cells.

Understanding the mechanisms of malignancy in acute myeloid leukemia (AML) cell is important for the targeted treatment and drug development. We found that visfatin, a 52-kDa adipokine, can positively regulate the proliferation of AML cells. Targeted inhibition of visfatin via its specific siRNAs or inhibitor can suppress the proliferation of AML cells. Further, knockdown of visfatin can increase the doxorubicin (Dox) and cisplatin (CDDP) sensitivity of AML cells. Among the tested six cytokines, si-visfatin can decrease the expression of interleukin-17 (IL-17). Over expression of IL-17 can reverse si-visfatin suppressed cell proliferation and increased Dox sensitivity. The upregulation of IL-17 was also involved in visfatin induced activation of PI3K/Akt signals in AML cells. The inhibitor of PI3K/Akt can synergistically suppress the proliferation of HL60 cells which were transfected with si-visfatin. Knockdown of visfatin can increase the expression of miR-135a, which can bind to the 3'UTR of IL-17 and decrease its expression. The inhibitor of miR-135a can attenuate si-visfatin suppressed expression of IL-17 and proliferation of AML cells. Collectively, our data suggested that visfatin can increase the malignancy of AML cells via regulation of miR-135a/IL-17/PI3K/Akt signals.

ObjectivesTo investigate the expression levels of aromatase cytochrome P450 enzyme (P450AROM) and related molecules—estrogen receptor-beta (ER-β), Ki-67, and p53—in prolactinoma tumor tissue from pre- and post-menopausal women, and to determine the associations of tumor invasiveness with expression levels of these genes.MethodsThis study recruited 90 patients with prolactinoma who underwent adenoidectomy between 2012 and 2017. Information was collected regarding clinical characteristics, hormones, laboratory tests, and magnetic resonance imaging-assessed tumor invasiveness. Expression levels of P450AROM, ER-β, Ki-67, and p53 were examined by immunohistochemistry in prolactinoma tissues.ResultsIncreased P450AROM expression was found in invasive prolactinoma tissues in post-menopausal women, compared with its expression in non-invasive prolactinoma tissues. ER-β level was significantly higher in patients resistant to treatment with bromocriptine, a dopamine agonist. However, there were no differences in rate of resistance to treatment (8.2% vs. 3.4%) or expression levels of P450AROM, Ki-67, p53, and ER-β between pre- and post-menopausal patients.ConclusionsOur results demonstrated that increased P450AROM expression in prolactinoma of post-menopausal women was positively associated with invasiveness. Moreover, ER-β level was higher in both pre- and post-menopausal patients who were resistant to dopamine agonist treatment.

ContextLH receptor (LHR) expression has been shown to be regulated posttranscriptionally by LHR mRNA binding protein (LRBP) in rodent and human ovaries. LRBP was characterized as mevalonate kinase. The gene that encodes mevalonate kinase is a member of a family of genes that encode enzymes involved in lipid synthesis and are regulated by the transcription factor sterol regulatory element binding proteins (SREBPs).ObjectiveThe current study examined the regulation of LHR mRNA expression in human granulosa-lutein cells in response to alterations in cholesterol metabolism.DesignUsing atorvastatin, an inhibitor of 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase to inhibit cholesterol biosynthesis, we examined its effect on LHR mRNA expression. The effect of atorvastatin on SREBP and mRNA expression as well as LHR mRNA binding protein expression was examined. Finally, the effect of atorvastatin on human chorionic gonadotropin (hCG)–stimulated progesterone production and the expression of key steroidogenic enzymes was also examined.ResultsStatin treatment reduced LHR mRNA expression by increasing the levels of SREBP1a and SREBP2, leading to an increase in LRBP. RNA gel shift assay showed that increased binding of LHR mRNA to LRBP occurred in response to atorvastatin, leading to LHR mRNA degradation. The granulosa-lutein cells pretreated with atorvastatin also showed decreased responsiveness to hCG by decreasing the mRNA and protein expression of steroidogenic enzymes. Atorvastatin also attenuated LH/hCG-induced progesterone production.ConclusionThese results imply that LHR mRNA expression by the human granulosa-lutein cells is regulated by cholesterol, through a mechanism involving SREBP and SREBP cleavage activating protein serving as the cholesterol sensor.

Rationale: Vascular calcification in arterial intima is closely associated with atherosclerosis. Endothelial cells (ECs) sense blood flow and respond to the mechanical cues generated by different flow patterns. Laminar flow induces an anti-atherosclerotic EC phenotype whereas disturbed flow exerts an atheroprone effect. However, the contribution of blood flow to calcification in atherosclerotic arteries remains to be evaluated. Objective: We aim to investigate whether blood flow plays a determinant role in the distribution of vascular calcification and the underlying mechanisms involved. Methods and Results: Computed tomography angiography analysis of human coronary arteries (n=48) shows that calcification preferentially develops at flow perturbated sites. Similar phenomenon was observed in calcified human aortic valves and mouse arteries. Nonuniform shear stress produced in Y-shaped slide simulating live conditions in branched arteries promotes calcification in human umbilical vein ECs. The expression of KLF2 (Krüppel-like factor 2), a transcription factor inducible by laminar flow, is reduced in ECs of calcified human aortic valves and in endothelial calcification model, suggesting that KLF2 downregulation is likely involved in intimal calcification. Indeed, KLF2 silencing induces endothelial-to-mesenchymal transition and accelerates osteoinduction in both human aortic ECs and human umbilical vein ECs. EC-specific KLF2 knockdown promotes whereas AAV9 (adeno-associated virus serotype 9)-mediated EC-KLF2 overexpression ameliorates vascular calcification in ApoE −/− mice. Global mRNA profiling in human umbilical vein ECs reveals that KLF2 inhibits the BMP (bone morphogenetic proteins)/SMAD1/5 pathway which is critical in vascular calcification. Furthermore, KLF2 mediates laminar flow-induced inhibition of the BMP/SMAD1/5 pathway. By contrast, disturbed flow-induced activation of BMP/SMAD1/5 pathway is suppressed by KLF2 overexpression. Mechanistically, KLF2 binds to the promoters of BMP4 , BMPER , and SMAD1 to directly regulate their expression in ECs. Conclusions: Vascular calcification prefers to occur at branched or bifurcated areas of vasculature. Laminar flow inhibits vascular calcification through KLF2-mediated inhibition of endothelial BMP/SMAD1/5 signaling. Targeting KLF2 may represent a novel therapeutic approach against vascular calcification.

PDCD10, also known as CCM3, is a gene found to be associated with the human disease cerebral cavernous malformations (CCMs). PDCD10 forms a complex with GCKIII kinases including STK24, STK25, and MST4. Studies in C. elegans and Drosophila have shown a pivotal role of the PDCD10-GCKIII complex in maintaining epithelial integrity. Here, we found that mice deficient of Pdcd10 or Stk24/25 in the kidney tubules developed polyuria and displayed increased water consumption. Although the expression levels of aquaporin genes were not decreased, the levels of total and phosphorylated aquaporin 2 (Aqp2) protein in the apical membrane of tubular epithelial cells were decreased in Pdcd10- and Stk24/25-deficient mice. This loss of Aqp2 was associated with increased expression and membrane targeting of Ezrin and phosphorylated Ezrin, Radixin, Moesin (p-ERM) proteins and impaired intracellular vesicle trafficking. Treatment with Erlotinib, a tyrosine kinase inhibitor promoting exocytosis and inhibiting endocytosis, normalized the expression level and membrane abundance of Aqp2 protein, and partially rescued the water reabsorption defect observed in the Pdcd10-deficient mice. Our current study identified the PDCD10-STK-ERM signaling pathway as a potentially novel pathway required for water balance control by regulating vesicle trafficking and protein abundance of AQP2 in the kidneys.

A simple Schiff base fluorescent probe (SW) composed of oxime and salicylaldehyde units was designed and synthesized. The probe has high selectivity and sensitivity for Zn2+ ion in mixed solvent (DMSO/H2O, V/V = 9:1). The fluorescence has obvious redshift phenomenon and visible color change. The stoichiometric ratio of the probe to Zn2+ ion was confirmed to be 1:2 (mole) by 1H NMR, MS analysis and job curves. And the detection limit of fluorescence response of the SW to Zn2+ is down to 2.53 × 10–8 mol/L. At the same time, the probe SW can be applied to the test paper detection of Zn2+ ions under 365 nm UV light and the detection of Zn2+ ions in actual water samples.

A novel strategy for microRNAs (miRNAs) detection has been developed utilizing duplex-specific nuclease-assisted signal amplification (DSNSA) and guanine-rich DNA-enhanced fluorescence of DNA-templated silver nanoclusters (AgNCs). The combination between target miRNA, DSNSA, and AgNCs is achieved by the unique design of DNA sequences. Target miRNA opens the hairpin structure of the Hairpin DNA probe (HP) by hybridizing with the HP and initiates the duplex-specific nuclease-assisted signal amplification (DSNSA) reaction. The DSNSA reaction generates the release of the guanine-rich DNA sequence, which can turn on the fluorescence of the dark AgNCs by hybridizing with the DNA template of the dark AgNCs. The fluorescence intensity of AgNCs corresponds to the dosage of the target miRNA. This is measured at 630 nm by exciting at 560 nm. The constructed method exhibits a low detection limit (~8.3 fmol), a great dynamic range of more than three orders of magnitude, and excellent selectivity. Moreover, it has a good performance for miR-21 detection in complex biological samples. A novel strategy for microRNAs (miRNAs) detection has been developed utilizing duplex-specific nuclease-assisted signal amplification (DSNSA) and guanine-rich DNA-enhanced fluorescence of DNA-templated silver nanoclusters (AgNCs).

Background: To explore the intervention mechanism of Baixiangdan Capsule(BXD) and Shuyu Capsule(SY) in the treatment of anger-out and anger-in. A kind of GABABR1(GB1) mediated GABA in the dorsal raphe nucleus(DRN) regulating serotonin(5-HT) levels in the Prefrontal Cortex(PFC), Hippocampus and Hypothalamus in Anger-out and Anger-in male rats. To further explore the difference of Baixiangdan Capsule(BXD) and Shuyu Capsule(SY) in the treatment of anger-out and anger-in.Methods: The anger rat model was established by social isolation combined with resident-intruder paradigm, and behavioral evaluation was used to screen and distinguish anger-out and anger-in model rats. BXD and SY were intervention drugs of anger–out and anger-in rats respectively. On this basis, ELISA was used to detect GABA content in DRN and 5-HT contents in PFC, hippocampus and hypothalamus after different time course (0,1,3,5,7 days) treated with BXD and SY. The co expression of 5-HT and GB1 in DRN was detected by immunofluorescence double labeling technique. Finally, brain stereotactic localization was performed after baclofen, the GB1 specific agonist, and CGP35348, the GB1 specific inhibitor, were injected into the DRN, the 5-HT contents in PFC, hippocampus and hypothalamus was detected by ELISA.Results: After drug treatment, ABT scores and OFT total distance in BXD group were significantly decreased to the level of control group (PConclusions: BXD and SY can effectively improve the abnormal behavior changes of anger-out and anger-in rats, and the optimal duration of action is 7 days. The improvement way is to correct the following abnormal changes: The significantly increased GABA in DRN combined with a significantly increased GB1 on 5-HT neurons in DRN, which further mediated the synaptic inhibition effect, thereby reducing 5-HT level of 5-HT neurons in DRN, resulting in a significant decrease of 5-HT levels in PFC, hypothalamus and hippocampus. Therefore, GB1 mediated GABA in DRN can regulate 5-HT levels in PFC, hypothalamus and hippocampus, which may be one of the ways that BXD and SY treat anger-out and anger-in.

The identity of the mechanism that controls aggressive behavior in rodents is unclear. Serotonin (5-HT) and GABA are associated with aggressive behavior in rodents. However, the regulatory relationship between these chemicals in the different brain regions of rats has not been fully defined. This study aimed to clarify the role of GABABR1 in DRN-mediated GABA to regulate 5-HT expression in multiple brain regions in male rats with high and low aggressive behavior. Rat models of highly and less aggressive behavior were established through social isolation plus resident intruder. On this basis, GABA content in the DRN and 5-HT contents in the PFC, hypothalamus, hippocampus and DRN were detected using ELISA. Co-expression of 5-HT and GB1 in the DRN was detected by immunofluorescence and immunoelectron microscopy at the tissue and subcellular levels, respectively. GB1-specific agonist baclofen and GB1-specific inhibitor CGP35348 were injected into the DRN by stereotaxic injection. Changes in 5-HT levels in the PFC, hypothalamus and hippocampus were detected afterward. After modeling, rats with highly aggressive behavior exhibited higher aggressive behavior scores, shorter latencies of aggression, and higher total distances in the open field test than rats with less aggressive behavior. The contents of 5-HT in the PFC, hypothalamus and hippocampus of rats with high and low aggressive behavior (no difference between the two groups) were significantly decreased, but the change in GABA content in the DRN was the opposite. GB1 granules could be found on synaptic membranes containing 5-HT granules, which indicated that 5-HT neurons in the DRN co-expressed with GB1, which also occurred in double immunofluorescence results. At the same time, we found that the expression of GB1 in the DRN of rats with high and low aggressive behavior was significantly increased, and the expression of GB1 in the DRN of rats with low aggressive behavior was significantly higher than that in rats with high aggressive behavior. Nevertheless, the expression of 5-HT in DRN was opposite in these two groups. After microinjection of baclofen into the DRN, the 5-HT contents in the PFC, hypothalamus and hippocampus of rats in each group decreased significantly. In contrast, the 5-HT contents in the PFC, hypothalamus and hippocampus of rats in each group increased significantly after injection with CGP35348. The significant increase in GABA in the DRN combined with the significant increase in GB1 in the DRN further mediated the synaptic inhibition effect, which reduced the 5-HT level of 5-HT neurons in the DRN, resulting in a significant decrease in 5-HT levels in the PFC, hypothalamus and hippocampus. Therefore, GB1-mediated GABA regulation of 5-HT levels in the PFC, hypothalamus and hippocampus is one of the mechanisms of highly and less aggressive behavior originating in the DRN. The increased GB1 level in the DRN of LA-behavior rats exhibited a greater degree of change than in the HA-group rats, which indicated that differently decreased 5-HT levels in the DRN may be the internal mechanisms of high and low aggression behaviors.

In this paper, the effect of binder decay rather than a change in the pigments on the blurring of ancient wall paintings was researched. The simulated wall paintings were prepared by brushing an aqueous solution containing gelatine and ochre grains on the surface of cylindrical compressed soil samples. Then, the dried samples were calcined at 650 °C for 2 h to obtain the simulated wall paintings with the degraded binder gelatine. Next, the calcined samples were brushed with a certain amount of acetone solution containing an ionic liquid ([BMIm]PF6) to obtain the corresponding repaired samples. Based on the results from various characterization methods (UV–vis, FTIR, XRD, XPS, SEM, TG), the following conclusions were drawn. The degradation of the binder caused by calcination increases the surface roughness of the painting layer, resulting in enhanced scattering. In this case, because scattering decrease the light absorption by the pigments, even if unchanged pigment exists in the painting layer, its colour can become blurred. The filling of the ionic liquid into the pores caused by gelatine decay in the painting layer can decrease the scattering, and the blurred colour can be restored to some extent. As typical examples, this principle was successfully applied to restore the blurred colour of an ancient Chinese wall painting (Tang Dynasty) and a pottery (Eastern Han Dynasty).

Thermally activated delayed fluorescent (TADF) materials, as the third generation of organic electroluminescent materials, have many advantages over other organic light-emitting diodes (OLEDs) materials, such as 100% internal quantum efficiency, no doping of heavy metals, and avoiding the shortages of ordinary fluorescent materials and phosphorescent materials. So it is considered to be the most competitive organic light-emitting materials, and has great application prospects in the field of OLEDs. So far, small-molecule TADF materials have achieved high quantum yield and full-color range of red, green, and blue. However, TADF polymers suitable for low-cost and easily scalable solution processing are less developed, which are confined by the preparation methods and polymers designing, and there are still challenges of increasing quantum efficiency and strengthening device performance. This review mainly summarizes different synthesis strategies of TADF polymers and the latest development in the field. Special attention is focused on illustrating the designing and structure-property relationship of TADF polymers, and finally, an outlook is given for the design and application prospect of TADF polymers in the future.

N-methyl-d-aspartate (NMDA) glutamate receptors (NMDARs) containing GluN2B subunits are prevalent early after birth in most brain regions in rodents. Upon synapse maturation, GluN2B is progressively removed from synapses, which affects NMDAR function and synaptic plasticity. Aberrant recruitment of GluN2B into mature synapses has been implicated in several neuropathologies that afflict adults. We found that the E3 ubiquitin ligase Cbl-b was enriched in the spinal cord dorsal horn neurons of mice and rats and suppressed GluN2B abundance during development and inflammatory pain. Cbl-b abundance increased from postnatal day 1 (P1) to P14, a critical time period for synapse maturation. Through its N-terminal tyrosine kinase binding domain, Cbl-b interacted with GluN2B. Ubiquitination of GluN2B by Cbl-b decreased the synaptic transmission mediated by GluN2B-containing NMDARs. Knocking down Cbl-b in vivo during P1 to P14 led to sustained retention of GluN2B at dorsal horn synapses, suggesting that Cbl-b limits the synaptic abundance of GluN2B in adult mice. However, peripheral inflammation induced by intraplantar injection of complete Freund's adjuvant resulted in the dephosphorylation of Cbl-b at Tyr363, which impaired its binding to and ubiquitylation of GluN2B, enabling the reappearance of GluN2B-containing NMDARs at synapses. Expression of a phosphomimic Cbl-b mutant in the dorsal horn suppressed both GluN2B-mediated synaptic currents and manifestations of pain induced by inflammation. The findings indicate a ubiquitin-mediated developmental switch in NMDAR subunit composition that is dysregulated by inflammation, which can enhance nociception.

Four new hetero-pentanuclear 3d–4f complexes [Zn4(L)2La(NO3)2(OEt)(H2O)] (1), [Zn4(L)2Ce(NO3)2(OMe)(MeOH)] (2), [Zn4(L)2Pr(NO3)2(OEt)(EtOH)] (3) and [Zn4(L)2Nd(NO3)2(OMe)(MeOH)] (4) were synthesized by the reactions of a newly synthesized octadentate bis(salamo)-based tetraoxime ligand (H4L) with Zn(OAc)2·2H2O and Ln(NO3)3·6H2O (Ln = La, Ce, Pr and Nd), respectively, and characterized via elemental analyses, FT-IR, UV-Vis spectroscopy and single crystal X-ray crystallography. The X-ray crystallographic investigation revealed that all ZnII ions were located in N2O3 coordination spheres, and possessed a trigonal bipyramid coordination environment. The LnIII ion lay in an O8 coordination sphere, and adopted a distorted square antiprismatic coordination environment. Furthermore, supramolecular interactions and fluorescence properties were investigated.

MLAA-34 is a novel leukemia-associated gene closely related to the carcinogenesis of acute monocytic leukemia (AML). MLAA-34 over expression has been observed to inhibit apoptosis in vitro. JAK2/STAT3 pathway plays an important role in cell proliferation, differentiation and inhibition of apoptosis in number of cancers. However, the relationship and interaction between MLAA-34 and JAK2/STAT3 has never been investigated in AML. This study investigates and reports a novel relationship between MLAA-34 and JAK2/STAT3 pathway in AML both in vitro and in vivo. We constructed MLAA-34 knockdown vector and transfected U937 cells to observe its apoptotic activities in relation to JAK2/STAT3 signaling pathway in vitro and then in vivo in mouse model. Levels of expression of MLAA-34 and JAK2/STAT3 and its downstream targets were also measured in AML patients and a few volunteers. We found that MLAA-34 knockdown increased U937 apoptosis in vitro and inhibited tumor growth in vivo. Components of the canonical JAK2/STAT3 pathway or its downstream targets, including c-myc, bcl-2, Bax, and caspase-3, were shown to be involved in the carcinogenesis of AML. We also found that the JAK2/STAT3 pathway positively regulated MLAA-34 expression. We additionally identified a STAT3 binding site in the MLAA-34 promoter where STAT3 binds directly and activates MLAA-34 expression. In addition, MLAA-34 was found to form a complex with JAK2 and was enhanced by JAK2 activation. Correlation of MLAA-34 and JAK2/STAT3 was further confirmed in AML patients. In conclusion, MLAA-34 is a novel regulator for JAK2/STAT3 signaling, and in turn, is regulated by this interaction in a positive feedback loop. Thus we report a novel model of interaction mechanism between MLAA-34 and JAK2/STAT3 which can be utilized as a potential target for a novel therapeutic approach in AML.

Background Cultured human cells are pivotal models to study human gene functions, but introducing complete loss of function in diploid or aneuploid cells has been a challenge. The recently developed CRISPR/Cas9-mediated homology-independent knock-in approach permits targeted insertion of large DNA at high efficiency, providing a tool for insertional disruption of a selected gene. Pioneer studies have showed promising results, but the current methodology is still suboptimal and functional outcomes have not been well examined. Taking advantage of the promoterless fluorescence reporter systems established in our previous study, here, we further investigated potentials of this new insertional gene disruption approach and examined its functional outcomes. Results Exemplified by using hyperploid LO2 cells, we demonstrated that simultaneous knock-in of dual fluorescence reporters through CRISPR/Cas9-induced homology-independent DNA repair permitted one-step generation of cells carrying complete disruption of target genes at multiple alleles. Through knocking-in at coding exons, we generated stable single-cell clones carrying complete disruption of ULK1 gene at all four alleles, lacking intact FAT10 in all three alleles, or devoid of intact CtIP at both alleles. We have confirmed the depletion of ULK1 and FAT10 transcripts as well as corresponding proteins in the obtained cell clones. Moreover, consistent with previous reports, we observed impaired mitophagy in ULK1−/− cells and attenuated cytokine-induced cell death in FAT10−/− clones. However, our analysis showed that single-cell clones carrying complete disruption of CtIP gene at both alleles preserved in-frame aberrant CtIP transcripts and produced proteins. Strikingly, the CtIP-disrupted clones raised through another two distinct targeting strategies also produced varied but in-frame aberrant CtIP transcripts. Sequencing analysis suggested that diverse DNA processing and alternative RNA splicing were involved in generating these in-frame aberrant CtIP transcripts, and some infrequent events were biasedly enriched among the CtIP-disrupted cell clones. Conclusion Multiallelic gene disruption could be readily introduced through CRISPR/Cas9-induced homology-independent knock-in of dual fluorescence reporters followed by direct tracing and cell isolation. Robust cellular mechanisms exist to spare essential genes from loss-of-function modifications, by generating partially functional transcripts through diverse DNA and RNA processing mechanisms. Two newly di- and tri-nuclear Cu(II) and Ni(II) complexes constructed from a half-salamo chelating ligand (HL: 2-[O-(1-ethyloxyamide)]oxime-6-methoxyphenol), [Cu2(L)2(μ-OAc)2] (1) and [{Ni3(μ-OAc)2(L)2(OAc)2]·CHCl3 (2), and one tetranuclear Co(II) complex [Co4(L′)4(MeOH)4] (3) based on an unexpected ligand H2L′ derived from the cleavage of HL have been synthesized and characterized by elemental analyses, IR, UV–vis spectra, and single crystal X-ray diffraction analyses. The single crystals of complexes 1, 2 and 3 in the process of synthesis are obtained through different routes. The complex 1 includes two Cu(II) atoms, two completely deprotonated (L)− units and two μ-acetate ions, which displays a propeller-type motif. The complex 2 contains three Ni(II) atoms, two completely deprotonated (L)− units, two μ-acetate ions, two chelating acetate ions and one crystallizing chloroform molecule, the crystal structure of complex 2 reveals a butterfly-type motif. The complex 3 is made up of four Co(II) atoms, four completely deprotonated (L′)2− units (HL′: 2-[O-(1-hydroxyethyl)]oxime-6-methoxyphenol) and four coordinated methanol molecules, which shows a windmill-type motif. Besides, magnetic properties and Hirshfeld surface analyses were discussed.

A two-photon fluorescent covalent organic framework nanopore is designed for the first time for H2S interference-resistant bioimaging., The complex environment of living organisms significantly challenges the selectivity of classic small-molecule fluorescent probes for bioimaging. Due to their predesigned topological structure and engineered internal pore surface, covalent organic frameworks (COFs) have the ability to filter out coexisting interference components and help to achieve accurate biosensing. Herein, we propose an effective interference-resistant strategy by creating a COF-based hybrid probe that combines the respective advantages of COFs and small-molecule probes. As a proof of concept, a two-photon fluorescent COF nanoprobe, namely TpASH-NPHS, is developed for targeting hydrogen sulfide (H2S) as a model analyte. TpASH-NPHS exhibits limited cytotoxicity, excellent photostability and long-term bioimaging capability. More importantly, compared with the small-molecule probe, TpASH-NPHS achieves accurate detection without the interference from intracellular enzymes. This allows us to monitor the levels of endogenous H2S in a mouse model of cirrhosis.

Ni−Al bimetallic catalysis proves to be an efficient catalytic strategy for unreactive bond transformations. Recently, chiral bifunctional ligands, especially amphoteric secondary phosphine oxide (SPO) ligand, are used for a more powerful synergistic effect in the bimetal-catalyzed reactions, providing not only milder reaction conditions and higher reactivity but also excellent reaction selectivity. Herein, we give a brief review on the development of Ni−Al bimetallic catalytic system and highlight recent advances in enantioselective Ni−Al bimetallic catalysis for unreactive bond transformation.

A redox-neutral Ni-catalyzed intramolecular hydroarylation of alkene with simple arene has been developed, in which DMF played a proton shuttle role in facilitating the intramolecular coupling, avoiding the use of additional reductants and oxidants. A series of oxindoles with a quaternary center were obtained in up to 99% yield.

Three novel N4-cavity Cu(II), Ni(II) and Zn(II) mononuclear complexes, [Cu(L)(NO3)2][Cu(L)(NO3)]NO3 (1) [Ni(L)(NO3)2] (2) and [Zn(L)(NO3)2] (3) were successfully synthesized via the reactions of a pyridine-containing N4-cavity bisoxime ligand L (L = 2,2'-[Ethylenedioxobis(azetidinyl)]pyridine) with Cu(II), Ni(II) and Zn(II) nitrates, respectively. Complexes 1-3 were characterized via elemental analysis, IR, UV-Vis spectra and X-ray crystallography. All three complexes are monoclinic systems, containing one M(II) atom and one (L) ligand moiety. Complexes 2 and 3 form centrosymmetric molecules. In addition, supramolecular interactions, Hirshfeld surface analysis were used to analyze the crystal properties that have been studied.

A new salamo-type fluorescence probe H2L was synthesized by a series of reactions and corroborated by single-crystal X-ray crystallography and 1H NMR spectra. Fluorescence of Co2+ selectively response showed “off” when cations were sequentially detected. The LOD of H2L to Co2+ was calculated to be 3.73 × 10-7 M in DMF/water (9:1, v/v) solution system. Meanwhile, the single crystal of the Co2+ complex with H2L was obtained by solvent evaporation. The structure of the complex [Co3(L)2(MeOH)2(μ-OAc)2]·2MeOH was validated by single-crystal X-ray diffraction, IR, and elemental analyses, and the mechanism of recognition of Co2+ by H2L was got. Density functional theory (DFT) and Hirshfeld surface analyses were investigated to further understand the experimental results observed.

Binuclear CoIII complex [Co2(L)3(DMF)3] containing a Schiff ligand H2L was synthesized and analyzed by elemental analysis, single crystal X-ray crystallography, FT-IR, UV–Vis absorption spectroscopy, Hirshfeld surface analysis and DFT calculation. The CoIII complex was a 3:2 ((L)2− : CoIII) binuclear structure. The spatial configuration of central CoIII ions was six-coordinated slightly twisted octahedron. The 3D supramolecular network structure was linked by intermolecular hydrogen bonds, C H···π and π···π stacking interactions. The Schiff ligand H2L exhibits weak luminescence, but this weak fluorescence is turned on after coordination with the ZnII ion. Furthermore, H2L could as an “OFF-ON” chemosensor for fluorescent recognition of ZnII.

The K+-Cl- co-transporter 2 (KCC2) is a neuron-specific Cl- extruder in the dorsal horn of spinal cord. The low intracellular Cl- concentration established by KCC2 is critical for GABAergic and glycinergic systems to generate synaptic inhibition. Peripheral nerve lesions have been shown to cause KCC2 dysfunction in adult spinal cord through brain-derived neurotrophic factor (BDNF) signaling, which switches the hyperpolarizing inhibitory transmission to be depolarizing and excitatory. However, the mechanisms by which BDNF impairs KCC2 function remain to be elucidated. Here we found that BDNF treatment enhanced KCC2 ubiquitination in the dorsal horn of adult mice, a post-translational modification that leads to KCC2 degradation. Our data showed that spinal BDNF application promoted KCC2 interaction with Casitas B-lineage lymphoma b (Cbl-b), one of the E3 ubiquitin ligases that are involved in the spinal processing of nociceptive information. Knockdown of Cbl-b expression decreased KCC2 ubiquitination level and attenuated the pain hypersensitivity induced by BDNF. Spared nerve injury significantly increased KCC2 ubiquitination, which could be reversed by inhibition of TrkB receptor. Our data implicated that KCC2 was one of the important pain-related substrates of Cbl-b and that ubiquitin modification contributed to BDNF-induced KCC2 hypofunction in the spinal cord.

Heterotrinuclear 3d-4f complexes with a naphthalenediol-based acyclic bis(salamo)-type ligand have been synthesized and structurally characterized. Spectral titrations clearly show that the heterotrinuclear complexes [Zn2(L)La(OAc)3] (1), [Zn2(L)Ce(OAc)3] (2) and [Zn2(L)Dy(OAc)3(CH3OH)]·CH2Cl2 (3) are acquired by the substitution reaction of the obtained homotrinuclear Zn(II) complex with 1 equiv. of Ln(NO3)3 (Ln3+=La3+, Ce3+ and Dy3+). Two Zn(II) ions are penta- and hexa-coordinated with geometries of distorted tetragonal pyramid and octahedron. La(III) ion is deca-coordinated, adopting a distorted bicapped square antiprism geometry. Ce(III) ion is nona-coordinated with geometry of distorted capped square antiprism as well as Dy(III) ion. The different coordination modes of acetate ions in complexes 1, 2 and 3 lead to different coordination numbers of the lanthanide(III) ions. Furthermore, the structures and fluorescence properties have been discussed.

An ionic liquid-based microwave-assisted extraction (ILMAE) method was developed to extract 6-, 8-, 10-gingerols and 6-, 8-, and 10-shogaols from ginger (Zingiber officinale Roscoe). A series of water-miscible ionic liquids (ILs) with different kinds of anions and cations were investigated. Results indicated that among the investigated ILs, the carbon chains of alkyl cations remarkably affected the extraction efficiency of the six compounds and 1-decyl-3-methylimidazolium bromide ([C10MIM]Br) was the optimal IL. The selected conditions for ILMAE were as follows: [C10MIM]Br concentration, 0.80 M; extraction temperature, 75 °C; irradiation time, 30 min; and irradiation power, 400 W, in the case of solid–liquid ratio of 0.1:10 g/mL. This study showed that compared with the total extraction yield of 0.595 ± 0.031% by methanol marinated extraction and 0.673 ± 0.043% by methanol-based microwave-assisted extraction, ILMAE attained higher yield of 0.716 ± 0.051%; and ILMAE obtained a shorter time of 30 min for extraction of gingerols and shogaols from ginger. This study provides a basis for separating and purifying bioactive components from ginger. Moreover, the use of small amounts of [C10MIM]Br as extraction solution in the method was considered environmentally friendly.

Three trinuclear complexes constructed from a naphthalenediol-based bis(Salamo)-type tetraoxime ligand (H4L1), [Cu3(L1)(OAc)2]·CH3OH (1), [Zn3(L1)(OAc)2(CH3OH)2]·4CHCl3 (2), and [Cu2(L1)Na(NO3)(CH3OH)] (3), and one dinuclear complex, [{Cu(L2)}2] (4), based on an unexpected ligand H2L2 derived from the cleavage of H4L1 have been synthesized and characterized by X-ray diffraction analyses. Different anionic sources, transition metal salts, and rationally controlled reaction conditions play important roles in the construction of the resulting complexes with variable coordination geometries (coordination number ranges from 4 to 7), and four types of supramolecular network were controlled by noncovalent interactions, such as hydrogen bond, π···π stacking, C–H···π, Cu···π, and halogen-related interactions. The fluorescence properties of H4L1 and its complexes were investigated, and 2 exhibits unique bright cyan–yellow visible fluorescent emissions in different solvents.

An unexpected polyhydroxyl-bridged tetranuclear ZnII complex and a benzoquinone compound derived from metal-ion promoted reactivity of Schiff base ligands were synthesized and characterized. The reaction of zinc(II) acetate dihydrate with oxime-type Schiff base ligand HL1 [HL1 = 1-(3-((3,5-dibromosalicylaldehyde)amino)phenyl)ethan-1-one O-benzyl oxime] in methanol, acetone, and acetonitrile resulted in the chemoselective cleavage of the C=N bond of the Schiff base HL1, and then the further addition of acetone to two salicylaldehyde molecules derived from cleavage of the C=N bond in situ α,α double aldol reaction promoted by ZnII ions. The newly formed ligands H4L2 coordinate to four ZnII ions forming a defect-dicubane core structure [ZnII4(H2L2)2(μ3-OCH3)2(μ-OCH3)2(CH3OH)2] (1) bridged exclusively by oxygen-based ligands. The similar ligand HL3 [HL3 = 1-(3-((3,5-dichlorosalicylaldehyde)amino)phenyl)ethan-1-one O-benzyl oxime)] was employed to react with CdII acetate dihydrate under the same reaction conditions. No aldol addition occurred but a unexpected benzoquinone compound 2,5-bis(((3-(1-((benzyloxy)imino)ethyl)phenyl)imino)methyl)-1,4-benzoquinone (2) formed. The results provided interesting insights into one-pot routes involving in situ reactions act as a strategy for obtaining a variety of polymeric/polynuclear complexes which are inconvenient to obtain from directly presynthesizing the ligands.

Twofold C-H annulation of readily available formamides and alkynes without built-in chelating groups was achieved. Ni-Al bimetallic catalysis enabled by a bulky BINOL-derived chiral secondary phosphine oxide (SPO) ligand proved to be critical for high reactivity and high selectivity. This reaction uses readily available formamides as starting materials and provides a concise synthetic pathway to a broad range of chiral ferrocenes in 40-98 % yield and 93-99 % ee.

The hydrolysis of K2[PtCl4] under visible light, the equilibrium, morphology, and size of Pt NPs combined with CDs, the valence states of Pt element and the bonding of Pt NPs with CDs, the influence of different kinds of cyclodextrin (α, β, γ-CDs), the acidity of the solution, and the visible light color on the catalytic ability of Pt NPs were all researched by typical approaches. All results prove visible light irradiation is effectively playing a key role in the reaction process to form Pt NPs in a very narrow size distribution. pH = 9, β-CDs, and purple visible light are the optimized reaction conditions. The Pt-CDs present a high rate constant of 0.88 min−1 in the catalytic reduction of 4-NP. This research is beneficial for expanding photochemical approaches from ultraviolet light to visible light, which is more of green chemistry, low cost, and facile.
