Abstract: Systemic treatment of malignant pleural mesothelioma (MPM) is moderately active for the intrinsic pharmacological resistance of MPM cell and its ability to induce an immune suppressive environment. Here we showed that the expression of bromodomain (BRD) proteins BRD2, BRD4 and BRD9 was significantly higher in human primary MPM cells compared to normal mesothelial cells (HMC). Nanomolar concentrations of bromodomain inhibitors (BBIs) JQ1 or OTX015 impaired patient-derived MPM cell proliferation and induced cell-cycle arrest without affecting apoptosis. Importantly, BBIs primed MPM cells for immunogenic cell death, by increasing extracellular release of ATP and HMGB1, and by promoting membrane exposure of calreticulin and ERp57. Accordingly, BBIs activated dendritic cell (DC)-mediated phagocytosis and expansion of CD8+ T-lymphocyte clones endorsed with antitumor cytotoxic activity. BBIs reduced the expression of the immune checkpoint ligand PD-L1 in MPM cells; while both CD8+ and CD4+ T-lymphocytes co-cultured with JQ1-treated MPM cells decreased PD-1 expression, suggesting a disruption of the immune-suppressive PD-L1/PD-1 axis. Additionally, BBIs reduced the expansion of myeloid-derived suppressor cells (MDSC) induced by MPM cells. Finally, a preclinical model of MPM confirmed that the anti-tumor efficacy of JQ1 was largely due to its ability to restore an immune-active environment, by increasing intra-tumor DC and CD8+ T-lymphocytes, and decreasing MDSC. Thereby, we propose that, among novel drugs, BBIs should be investigated for MPM treatment for their combined activity on both tumor cells and surrounding immune-environment.

Abstract: The Protoplanetary Discussions conference --- held in Edinburgh, UK, from 7th --11th March 2016 --- included several open sessions led by participants. This paper reports on the discussions collectively concerned with the multiphysics modelling of protoplanetary discs, including the self-consistent calculation of gas and dust dynamics, radiative transfer and chemistry. After a short introduction to each of these disciplines in isolation, we identify a series of burning questions and grand challenges associated with their continuing development and integration. We then discuss potential pathways towards solving these challenges, grouped by strategical, technical and collaborative developments. This paper is not intended to be a review, but rather to motivate and direct future research and collaboration across typically distinct fields based on \textit{community driven input}, to encourage further progress in our understanding of circumstellar and protoplanetary discs., Accepted for publication in PASA. 25 pages

Abstract: In this paper we report on in-situ CCVD grown bilayer graphene transistors (BiLGFETs) in a Silicon-CMOS compatible fabrication process. By means of catalytic chemical vapor deposition (CCVD) the BiLGFETs are realized directly on oxidized silicon substrate without transfer. These BiLGFETs possess unipolar p-type device characteristics with a high on/off-current ratio between 1×10 5 and 1×10 7 at room temperature [1, 2]. At this stage, the maximal on-state current of a BiLGFET is clearly influenced by the contact resistance. In order to improve the performance of the produced BiLGFETs, an advanced fabrication step has been developed, by which means the contact resistance is lowered by a factor of 10.

Abstract: International audience; Co3O4/CoFe2O4 decorated on nickel foam (NF/Co3O4/CoFe2O4) was synthesized from a metal–organic framework by a solvothermal approach using nicotinic acid as an organic linker followed by annealing at 500 °C. The electrochemical activity of NF/Co3O4/CoFe2O4 for the oxygen evolution reaction (OER) was assessed in alkaline medium. Under basic conditions (pH > 10), the composite electrode revealed enhanced electrocatalytic OER activity requiring an overpotential of 215 mV versus RHE to reach 10 mA cm–2 with a Tafel slope of 90 mV dec–1. The enhanced OER activity was ascribed to the presence of Co3+ and Fe3+ in the octahedral sites of Co3O4 and CoFe2O4, respectively, and their synergic effect in Co3O4/CoFe2O4. This anode showed a stable current density of about 160 mA cm–2 for 20 h; the same Co3O4/CoFe2O4/NF anode was applied for several OER experiments without loss of activity.

Abstract: International audience; This study describes the development of a shaping process involving GDC10 (Ce0.9Gd0.1O1.95) as an electrolyte for Solid Oxide Cells operating at low temperature (500−600 °C). The goal is to build a single cell of porous/dense/porous GDC10 layers while adapting the porous layer thicknesses, which are infiltrated by catalysts to prepare efficient GDC-base composites. Two shaping processes were used to make the cell. The first process is die pressing of the electrolyte powder followed by the infiltration of a screen-printed porous backbone. The second shaping process consists of preparing a porous/dense/porous (30 μm/≈ 400 μm/30 μm) cell in a single step by combining tape-casting and screen-printing processes followed by the infiltration of the previous catalyst.

Abstract: This study reports the only recent characterization of two contrasted wet deposition events collected during the PEACETIME (ProcEss studies at the Air–sEa Interface after dust deposition in the MEditerranean Sea) cruise in the open Mediterranean Sea (Med Sea) and their impact on trace metal (TM) marine stocks. Rain samples were analysed for Al, 12 TMs (Co, Cd, Cr, Cu, Fe, Mn, Mo, Ni, Pb, Ti, V and Zn) and nutrient (N, P, dissolved organic carbon) concentrations. The first rain sample collected in the Ionian Sea (Rain ION) was a typical regional background wet deposition event, whereas the second rain sample collected in the Algerian Basin (Rain FAST) was a Saharan dust wet deposition event. Even in the remote Med Sea, all background TM inputs presented an anthropogenic signature, except for Fe, Mn and Ti. The concentrations of TMs in the two rain samples were significantly lower compared to concentrations in rains collected at coastal sites reported in the literature, due to the decrease in anthropogenic emissions during the preceding decades. The atmospheric TM inputs were mainly dissolved forms, even in dusty Rain FAST. The TM stocks in the mixed layer (ML, 0–20 m) at the FAST station before and after the event showed that the atmospheric inputs were a significant supply of particulate TMs and dissolved Fe and Co for surface seawater. Even if the wet deposition delivers TMs mainly in soluble form, the post-deposition aerosol dissolution could to be a key additional pathway in the supply of dissolved TMs. At the scale of the western and central Mediterranean, the atmospheric inputs were of the same order of magnitude as ML stocks for dissolved Fe, Co and Zn, highlighting the role of the atmosphere in their biogeochemical cycles in the stratified Med Sea. In case of intense dust-rich wet deposition events, the role of atmospheric inputs as an external source was extended to dissolved Co, Fe, Mn, Pb and Zn. Our results suggest that the wet deposition constitutes only a source of some of dissolved TMs for Med Sea surface waters. Abstract: International audience; Oxygen-isotope measurements of fossil carbonates remain the most common method for paleoclimatic temperature reconstructions. A well-known limitation of this approach is the influence of the oxygen isotope composition of water in which mineralization occurs, which may vary significantly through space and time, and is often difficult to constrain precisely. Carbonate clumped-isotope thermometry is an alternative approach applicable to many carbonates. It is based on measurements of Δ47 (a tracer of small statistical anomalies in the abundance of rare, doubly-substituted carbonate isotopologues), and requires no independent information on the oxygen-isotope composition of parent waters. Here, we report new calibration observations of clumped isotopes in four species of calcitic marine bivalves (A. colbecki, N. cochlear, S. cucullata, M. gigas) from various ecosystems including coastal and deep-sea environments, with calcification temperatures ranging from −2 °C to 27 °C and very different amplitudes of seasonal temperature variability. At two localities with large seasonal temperature variability, calcification time intervals were constrained using a sclerochronological approach to test whether seasonal gradients of temperature can be accurately quantified based on Δ47 measurements.Our results indicate that the mature bivalves we analyzed have clumped-isotope compositions entirely consistent with earlier calibration studies processed in the I-CDES reference frame and based on biogenic/abiotic/synthetic materials. By contrast, juvenile M. gigas oysters yield substantially lower Δ47 values than expected based on their calcification environments, suggesting that their early growth phase is associated with yet poorly understood isotopic biases affecting both δ18Ο and Δ47 values. The link between seawater temperatures and bivalve Δ47 values is thus potentially applicable to seasonal reconstructions, but only if shell sections formed in cold seasons are precisely identified and precisely sampled, and taking into account that winter calcification is likely to be biased due to reduced growth rate. Moreover, the excellent agreement between our observations and the existing I-CDES calibrations further demonstrates the efficacy of the I-CDES standardization approach, and adds to the evidence that many different types of carbonates conform to statistically indistinguishable relationships between Δ47 and crystallization temperature.

Abstract: International audience; Hepatocellular carcinoma (HCC) development entails changes in liver metabolism. Current knowledge on metabolic perturbations in HCC is derived mostly from case-control designs, with sparse information from prospective cohorts. Our objective was to apply comprehensive metabolite profiling to detect metabolites whose serum concentrations are associated with HCC development, using biological samples from within the prospective European Prospective Investigation into Cancer and Nutrition (EPIC) cohort (>520 000 participants), where we identified 129 HCC cases matched 1:1 to controls. We conducted high-resolution untargeted liquid chromatography-mass spectrometry-based metabolomics on serum samples collected at recruitment prior to cancer diagnosis. Multivariable conditional logistic regression was applied controlling for dietary habits, alcohol consumption, smoking, body size, hepatitis infection and liver dysfunction. Corrections for multiple comparisons were applied. Of 9206 molecular features detected, 220 discriminated HCC cases from controls. Detailed feature annotation revealed 92 metabolites associated with HCC risk, of which 14 were unambiguously identified using pure reference standards. Positive HCC-risk associations were observed for N1-acetylspermidine, isatin, p-hydroxyphenyllactic acid, tyrosine, sphingosine, l,l-cyclo(leucylprolyl), glycochenodeoxycholic acid, glycocholic acid and 7-methylguanine. Inverse risk associations were observed for retinol, dehydroepiandrosterone sulfate, glycerophosphocholine, γ-carboxyethyl hydroxychroman and creatine. Discernible differences for these metabolites were observed between cases and controls up to 10 years prior to diagnosis. Our observations highlight the diversity of metabolic perturbations involved in HCC development and replicate previous observations (metabolism of bile acids, amino acids and phospholipids) made in Asian and Scandinavian populations. These findings emphasize the role of metabolic pathways associated with steroid metabolism and immunity and specific dietary and environmental exposures in HCC development.

Abstract: Background and aims Baclofen is a promising drug for treating patients with alcohol-related disorders. Nevertheless, the first randomized clinical trials (mainly with target doses) reported inconsistent efficacy, possibly because of the effective dose widely varying between patients. The Bacloville study aimed to test the efficacy of titrated baclofen for achieving low-risk alcohol consumption. Design Twelve-month multicenter pragmatic double-blind randomized clinical trial from June 2012 to June 2014. Setting Sixty-two French primary care centers. Participants Out-patients with high-risk alcohol consumption (> 40 g/day for women and > 60 g/day for men). Intervention and comparator Patients were randomly assigned (1 : 1 ratio) to receive titrated baclofen up to 300 mg/day or placebo for 12 months. Switching to open-label baclofen was allowed in cases of perceived inefficacy. Measurements The primary outcome defined success as no or low-risk alcohol consumption (≤ 20 g/day for women and ≤ 40 g/day for men) during the last month of the 1-year follow up, with patients who switched to open-label baclofen classified as failures. Findings A total of 320 patients were randomized, 162 to baclofen and 158 to placebo (consumption 129 g/day in both arms). Discontinuation rates were 30 and 34% in the baclofen and placebo arms, respectively, and return rates of the last-month diaries were 42 and 34%, respectively. Primary success rates were 57 and 36% in the baclofen and placebo arms, respectively [difference: 21 percentage points, 95% confidence interval (CI) = 8-34, P = 0.003]. When switchers were not classified as failures unless they failed, the success rates were 62 versus 55% (difference: 6 percentage points, 95% CI = -7 to 20). Over 12 months, daily consumption differed between both arms (11 g less in the baclofen arm), as did the number of abstinence days (3.3 days more in the baclofen arm). Adverse events were more frequent with baclofen than placebo and were mostly drowsiness, fatigue and insomnia. Serious adverse events occurred in 85 (seven deaths) and 36 (three deaths) patients with baclofen and placebo, respectively. Conclusions Baclofen was more effective than placebo in reducing alcohol consumption to low-risk levels. The number of adverse events and more serious adverse events was greater with baclofen than placebo.

Abstract: We present a theory-experiment investigation of the helically chiral compounds Ru(acac)(3) and Os(acac)(3) as candidates for next-generation experiments for detection of molecular parity violation (PV) in vibrational spectra. We used relativistic density functional theory calculations to identify optimal vibrational modes with expected PV effects exceeding by up to 2 orders of magnitude the projected instrumental sensitivity of the ultrahigh resolution experiment under construction at the Laboratoire de Physique des Lasers in Paris. Abstract: PTPN2 is an important protein tyrosine phosphatase (PTP) that plays a key role in cell signaling. Deletions or inactivating mutations of PTPN2 has been described in different pathologies and underline its critical role in hematopoiesis, auto-immunity and inflammation. Surprisingly, despite the major pathophysiological implications of PTPN2, the structural analysis of this PTP and notably of its pathogenic mutants remain poorly documented. Contrary to other human PTP enzymes, to date, only one structure of PTPN2 (wild-type form) has been reported. Here, we report the first crystal structure of a pathogenic mutant of PTPN2 (Cys216Gly) that causes an autoimmune enteropathy. We show in particular that this mutant adopts a classical PTP fold. More importantly, albeit inactive, the mutant retains its ability to bind substrates and to adopt the characteristic catalytically competent closed form of PTP enzymes. This novel PTPN2 structure may serve as a new tool to better understand PTP structures and the structural impacts of pathogenic mutations. Abstract: International audience; Conducting polymers have been used as support for catalysts towards distinct electrocatalytic reactions. The overall activity of the modified interfaces critically depends on both the processes on the catalyst surface and of the charge compensation within the polymer matrix. Therefore, understand the coupling between surface and volumetric processes in such systems is of utmost importance. In this contribution we report on the experimental investigation of the interplay between surface and volumetric processes during the electrooxidation of formic acid (and also formaldehyde) on platinum modified polyaniline electrodes. The study was performed through in situ nanogravimetry, with the Electrochemical Quartz Crystal Nanobalance (EQCN). We have explored the dynamics of charge and mass changes along several experimental conditions, including the self-organized potential oscillations. The systematic study included the analysis of several configurations: gold electrodes covered with a PANI film and dispersed platinum (Au/PANI/Pt); platinum electrodes covered with PANI (Pt/PANI); and bare platinum electrodes. The methodology introduced includes the detailed analysis of the charge compensation process and of the time-derivative of the mass variations. Typical results for the electro-oxidation of formic acid on PANI supported platinum indicate that the electrocatalysis itself is reflected in the current profile, whereas the charge compensation rules the gravimetric response. This general observation also holds for the oscillatory electro-oxidation of formic acid. The physicochemical description of the support/catalyst systems described here opens interesting perspectives for the investigation of parent systems.

Abstract: International audience; Surface modification of electrodes with glycans was investigated as a strategy for modulating the development of electrocatalytic biofilms for microbial fuel cell applications. Covalent attachment of phenyl-mannoside and phenyl-lactoside adlayers on graphite rod electrodes was achieved via electrochemically assisted grafting of aryldiazonium cations from solution. To test the effects of the specific bio-functionalities, modified and unmodified graphite rods were used as anodes in two-chamber microbial fuel cell devices. Devices were set up with wastewater as inoculum and acetate as nutrient and their performance, in terms of output potential (open circuit and 1 kΩ load) and peak power output, was monitored over two months. The presence of glycans was found to lead to significant differences in startup times and peak power outputs. Lactosides were found to inhibit the development of biofilms when compared to bare graphite. Mannosides were found, instead, to promote exoelectrogenic biofilm adhesion and anode colonization, a finding that is supported by quartz crystal microbalance experiments in inoculum media. These differences were observed despite both adlayers possessing thickness in the nm range and similar hydrophilic character. This suggests that specific glycan-mediated bioaffinity interactions can be leveraged to direct the development of biotic electrocatalysts in bioelectrochemical systems and microbial fuel cell devices.

Abstract: International audience; Nitrogen-co-doped MoS2@rGO composites (named hereafter N-MoS2@N-RGO) are prepared by the hydrothermal method using dicyandiamide or hydrazine monohydrate as the nitrogen source. N-MoS2@rGO is obtained by the hydrothermal reaction of thioacetamide, ammonium molybdate and GO precursors in the presence of dicyandiamide, while N-MoS2@N-RGO is synthesized using the same precursors in the presence of hydrazine monohydrate as the nitrogen source. On the one hand, N-MoS2@RGO displays excellent performance and good capacitance value of 335 F/g acquired at 0.5A/g and improved stability (∼94% of the original capacitance is preserved after 5000 cycles). On the other hand, compared with N-MoS2@RGO, N-MoS2@N-RGO possesses a slightly higher capacitance of 340 F/g recorded at 0.5A/g and better cycling stability (∼98% of its original value after 5000 cycles). Taken together, the results of this investigation clearly indicate that the fabricated composites have application prospect in energy storage devices.

Abstract: International audience; Speciation of Cd in soil solutions strongly determines the fate of this toxic metal in the environment. Generally, in soil solutions, Cd predominantly binds to the dissolved organic matter (DOM). The determination of the quantity and reactivity of DOM that actually complexes Cd in soil solutions is challenging for operational purposes. Therefore, this study tested whether Cd2+ concentration in soil solutions could effectively be predicted by considering complexation with a single mean organic ligand assumed to be a fraction of DOM of unspecified nature or assumed to be purely fulvic acids (FA) with reactivity as described in WHAM VII. The reactivity of the unspecified ligand and the concentration of FA were modelled and fitted to experimental data from 76 agricultural soils with low Cd contents. The optimal reactivity and FA concentration that minimized the relative error (RE) of predictions of the concentration of Cd2+ in soil solutions were either considered constant across soils or modelled from soil properties by multiple linear regressions (MLR) or random forests (RF), giving 6 models, the predictive value of which was assessed by 10-folds cross-validation. When the reactivity of the mean ligand and the optimal FA concentration were considered constant across soils, the models were biased and 66.9% of predictions had relative errors below a factor of 2. By contrast, if the reactivity of the mean ligand or the optimal FA concentration were allowed to vary with soil characteristics, these performances increased to 95.5%, soil pH being the main predictor and RF being slightly more efficient than MLR. With more than 95% of the relative errors of prediction below a factor of 2, the models developed in this work could be valuable for assessing Cd speciation in the solution of soils having a low Cd content.

Abstract: Advances in materials science have accelerated the development of diagnostic tools with the last decade witnessing the development of enzyme-free sensors, owing to the improved stability, low cost and simple fabrication of component materials. However, the specificity of non-enzymatic sensors for certain analytes still represents a challenging task, for example the determination of cholesterol level in blood is vital due to its medical relevance. In this work, a reagent displacement assay for cholesterol sensing in serum samples was developed. It is based on coating of a glassy carbon electrode with a polymer of intrinsic microporosity (PIM) that forms a host-guest complex with methylene blue (MB). In the presence of cholesterol, the MB electroactive probe was displaced due to the stronger association of cholesterol guest to the PIM host. The decrease in the oxidative current was proportional to the cholesterol concentration achieving a detection limit of approximately 0.1 nM. Moreover, to further assist the experimental studies, comprehensive theoretical calculations are also performed by using density functional theory (DFT) calculations. Abstract: The molecular mechanisms and forces involved in the translocation of bacterial toxins into host cells are still a matter of intense research. The adenylate cyclase (CyaA) toxin from Bordetella pertussis displays a unique intoxication pathway in which its catalytic domain is directly translocated across target cell membranes. The CyaA translocation region contains a segment, P454 (residues 454–484), which exhibits membrane‐active properties related to antimicrobial peptides. Herein, the results show that this peptide is able to translocate across membranes and to interact with calmodulin (CaM). Structural and biophysical analyses reveal the key residues of P454 involved in membrane destabilization and calmodulin binding. Mutational analysis demonstrates that these residues play a crucial role in CyaA translocation into target cells. In addition, calmidazolium, a calmodulin inhibitor, efficiently blocks CyaA internalization. It is proposed that after CyaA binding to target cells, the P454 segment destabilizes the plasma membrane, translocates across the lipid bilayer and binds calmodulin. Trapping of CyaA by the CaM:P454 interaction in the cytosol may assist the entry of the N‐terminal catalytic domain by converting the stochastic motion of the polypeptide chain through the membrane into an efficient vectorial chain translocation into host cells., The mechanism of cell invasion by bacterial toxins is still poorly understood. The adenylate cyclase (CyaA) toxin from Bordetella pertussis, the causative agent of whooping cough, directly translocates its catalytic domain across plasma membranes. The membrane‐permeabilizing segment (yellow) of CyaA translocates across the plasma membrane and binds calmodulin (red), which assists the entry of the catalytic domain (blue) into host cells while the hydrophobic and acylation domains (green) interact with the membrane and the C‐terminal Repeat‐in‐Toxin domain (grey) remains in the extra‐cellular milieu.

Abstract: In order to decrease the number of lenses and the weight of thermal imaging devices, specific optical design are required by using gradient refractive index (GRIN) elements transparent in the infrared waveband. While widely used for making visible GRIN lenses with silicate glasses, the ion exchange process is very limited when applied to chalcogenide glasses due to their low Tg and relatively weak mechanical properties. In this paper, we develop chalco-halide glasses based on alkali halide (NaI) addition in a highly covalent GeSe2–Ga2Se3 matrix, efficient for tailoring a significant and permanent change of refractive by ion exchange process between K+ and Na+. Optical and structural properties of the glass samples were measured showing a diffusion length reaching more than 2 mm and a Gaussian gradient of refractive index Δn of 4.5.10–2. The obtained GRIN lenses maintain an excellent transmission in the second (3–5 µm) and third (8–12 µm) atmospheric windows.

Abstract: Aimed at eliminating or at least significantly reducing the use of solvents, sodium hypochlorite pentahydrate crystals (NaOCl·5H2O) in the presence of a catalytic amount of a nitrosyl radical (TEMPO or AZADO) have been successfully used to induce mechanochemical oxidative processes on several structurally different primary and secondary alcohols. The proposed redox process is safe, inexpensive and performing effectively, especially on the macroscale. Herein, an Ertalyte® jar has been successfully used, for the first time, in a mechanochemical process.

Abstract: International audience; Among the positive electrode materials for Na-ion batteries, Na3V2(PO4)2F3 is considered as one of the most promising and generates high interest. Here, we study the influence of the sol–gel synthesis parameters on the structure and on the electrochemical signature of the partially substituted Na3V2−zAlz(PO4)2(F,O)3 materials. We demonstrate that the acidity of the starting solution influences the vanadium oxidation state of the final product. For the first time we report on the possibility of controlling the double Al/V and O/F substitution that leads to the preparation of the Na3V2−zAlz(PO4)2F1+zO2−z solid solution.

Abstract: International audience; INTRODUCTION: Adrenomedullin receptors are highly expressed in human alveolar capillaries and provide a molecular target for imaging the integrity of pulmonary microcirculation. In this work, we aimed to develop a NOTA-derivatized adrenomedullin analog (DFH17), radiolabeled with [18F]AlF, for PET imaging of pulmonary microcirculation.METHODS: Highly concentrated [18F](AlF)2+ (15 μL) was produced from purified fluorine-18 in NaCl 0.9%. Various complexation experiments were carried out at Al-to-NOTA molar ratios ranging from 1:1 to 1:40 to assess optimal radiolabeling conditions before using the peptide. DFH17 peptide (2 mM, pH 4) was radiolabeled with [18F](AlF)2+ for 15 min at 100 °C in a total volume of 60 μL. As part of the radiolabeling process, parameters such as fluorine-18 activity (~37 and 1480 MBq), concentration of AlCl3 (0.75, 2, 3, 6 or 10 mM) and the effects of hydrophilic organic solvent (aqueous vs ethanol 50%) were studied. The final formulation was tested for purity, identity and stability in saline. Initial in vivo evaluation of [18F]AlF-DFH17 was performed in normal rats by PET/CT.RESULTS: The scaled-up production of [18F]AlF-DFH17 was performed in high radiochemical and chemical purities in an overall radiochemical yield of 22-38% (at end-of-synthesis) within 60 min. The final formulation was stable in saline at different radioactive concentrations for 8 h. PET evaluation in rats revealed high lung-to-background ratios and no defluorination in vivo up to 1 h post-injection.CONCLUSION: The novel radioconjugate [18F]AlF-DFH17 appears to be a promising PET ligand for pulmonary microcirculation imaging.

Abstract: International audience; Microfluidization was used to develop methyl cellulose (MC)/cellulose nanocrystal CNC based nanocomposite films containing a plant essential oil (EO) blend (oregano: thyme) emulsion. A three factorial experimental design was used to systematically optimize the microfluidization pressure based on size and antifungal activity of the prepared emulsion. Results showed that microfluidization of the film forming dispersions provide a novel approach for the development of high strength bionanocomposite films. Incorporation of 7.5% CNC into MC containing 0.50–0.75% EO and application of a pressure of 15 k psi (103 MPa) created a nanoemulsion with particle size ≤ 100 nm which exhibited significant antifungal activity in vitro against Aspergillus niger, A. flavus, A. parasiticus and P. chrysogenum. In situ tests with MC/CNC based bioactive films containing EO emulsion produced a 2 log reduction in fungal growth in infected rice during 8 weeks of storage at 28 °C. Methyl cellulose nanocomposite films containing EOs nanoemulsion showed a slow release (35%) of volatile component over 12 weeks of storage period. The addition of CNC as reinforcing filler improved the tensile strength of the MC based films by 30% and decreased water barrier and release properties by 9 and 25% respectively. In addition, combined treatment of bioactive films with an irradiation treatment at 750 Gy showed more pronounced antifungal and mechanical properties than treatment with the bioactive film or irradiation alone. These results show the potential for EO-loaded methyl cellulose-based films to prolong shelf life of food products.

Abstract: International audience; Metal fluorides with 3D open structures, pyrochlore (pyr) or hexagonal tungsten bronze (HTB), are promising materials as positive electrodes for rechargeable batteries or catalysts. Herein, we have developed a two-step synthesis procedure to obtain new anhydrous mixed-metal-cation fluorides crystallizing in the pyrochlore structure. The first step consists of preparing mixed-metal ammonium fluorides (NH4)M2+Fe3+F6 (M = Mn, Fe, Co Ni) using different synthesis strategies. For M = Mn, three allotropic varieties of (NH4)Mn2+Fe3+F6 are obtained; two phases adopt the expected pyrochlore network with either the cubic Fm3̅m or the orthorhombic Pnma space group, and the third phase exhibits a 3D network with narrow pseudotriangular cavities. 57Fe Mössbauer spectrometry indicates that the crystal structures are governed by the Fe3+/Mn2+ cationic order or disorder. The second step is a topotactic oxidation of pyr-(NH4)M2+Fe3+F6 under a molecular F2 flow. To better understand the reaction mechanism, the topotactic oxidation was followed by thermogravimetry, XRD, FTIR, and Mössbauer spectrometry. The successful synthesis of the first anhydrous pyr-M3+0.5Fe0.5F3 provides a new route to prepare anhydrous mixed-metal fluorides pyrochlore with empty cavities of the open framework.

Abstract: Polyesters, as they exist in planta, are promising materials with which to begin the development of “green” replacements. Cutin and suberin, polyesters found ubiquitously in plants, are prime candidates. Samples enriched for plant polyesters, and in which their native backbones were largely preserved, were studied to identify “natural” structural features; features that influence critical physical properties. Quantitative nuclear magnetic resonance (NMR), differential scanning calorimetry (DSC), and X-ray scattering methods were used to quantify structure–property relationships in these polymeric materials. The degree of esterification, namely, the presence of acylglycerol linkages in suberin and of secondary esters in cutin, and the existence of mid-chain epoxide groups defining the packing of the aliphatic chains were observed. This packing determines polymer crystallinity, the resulting crystal structure, and the melting temperature. To evaluate the strength of this rule, tomato cutin from the same genotype, studying wild-type plants and two well-characterized mutants, was analyzed. The results show that cutin’s material properties are influenced by the amount of unbound aliphatic hydroxyl groups and by the length of the aliphatic chain. Collectively, the acquired data can be used as a tool to guide the selection of plant polyesters with precise structural features, and hence physicochemical properties., A detailed insight into plant polyesters identifies correlations of structural features with physical properties that are translated into five essential rules for the development of novel green materials.

Abstract: International audience; The boreal biome plays an important role in the global carbon cycle. However, current estimates of its sink-source strength and responses to changes in climate are primarily derived from models and thus remain uncertain. A major challenge is the validation of these models at a regional scale since empirical flux estimates are typically confined to ecosystem or continental scales. The Integrated Carbon Observation System (ICOS)-Svartberget atmospheric station (SVB) provides observations including tall tower eddy covariance (EC) and atmospheric concentration measurements that can contribute to such validation in Northern Sweden. Thus, the overall aim of this study was to quantify the carbon balance in Northern Sweden region by integrating land-atmosphere fluxes and atmospheric carbon dioxide (CO2) concentrations. There were three specific objectives. First, to compare flux estimates from four models (VPRM, LPJ-GUESS, ORCHIDEE, and SiBCASA) to tall tower EC measurements at SVB during the years 2016–2018. Second to assess the fluxes' impact on atmospheric CO2 concentrations using a regional transport model. Third, to assess the impact of the drought in 2018. The comparison of estimated concentrations with ICOS observations helped the evaluation of the models' regional scale performance. Both the simulations and observations indicate there were similar reductions in the net CO2 uptake during drought. All the models (except for SiBCASA) and observations indicated the region was a net carbon sink during the 3-year study period. Our study highlights a need to improve vegetation models through comparisons with empirical data and demonstrate the ICOS network's potential utility for constraining CO2 fluxes in the region.

Abstract: International audience; The bioadsorbent C1, which is a chitosan derivative prepared in a one-step synthesis, was successfully used to adsorb Cr(VI) and Cu(II) simultaneously. Here, for the first time the simultaneous adsorption of a cation and an anion was modeled using the Corsel model for kinetics and the Real Adsorbed Solution Theory model for equilibrium data. Batch studies of the adsorption of Cu(II) and Cr(VI) in single and binary aqueous solutions were performed as a function of initial solute concentration, contact time, and solution pH. The maximum adsorption capacities of C1 in single and binary aqueous solutions were 1.84 and 1.13 mmol g−1 for Cu(II) and 3.86 and 0.98 mmol g−1 for Cr(VI), respectively. The reuse of C1 was investigated, with Cu(II) ions being almost completely desorbed and fully re-adsorbed. For Cr(VI), the desorption was incomplete resulting in a lower re-adsorption. Energy-dispersive X-ray spectroscopy was used for mapping the distributions of Cr(VI) and Cu(II) adsorbed on the C1 surface in single and binary adsorption systems. Isothermal titration calorimetry experiments were performed for Cr(VI) and Cu(II) adsorption in single solutions. The thermodynamic parameters of adsorption showed that the adsorption of both metal ions was enthalpically driven, but entropically unfavorable.

Abstract: International audience; Epidemiologic studies examining the association between specific fatty acids and colorectal cancer (CRC) risk are inconclusive. We investigated the association between dietary estimates and plasma levels of individual and total saturated (SFA), monounsaturated (MUFA), industrial-processed trans (iTFA), and ruminant-sourced trans (rTFA) fatty acids, and CRC risk in the European Prospective Investigation into Cancer and Nutrition (EPIC). Baseline fatty acid intakes were estimated in 450 112 participants (6162 developed CRC, median follow-up = 15 years). In a nested case-control study, plasma phospholipid fatty acids were determined by gas chromatography in 433 colon cancer cases and 433 matched controls. Multivariable-adjusted hazard ratios (HRs) and odds ratios (ORs) with 95% confidence intervals (CIs) were computed using Cox and conditional logistic regression, respectively. Dietary total SFA (highest vs lowest quintile, HRQ5vsQ1 = 0.80; 95%CI:0.69-0.92), myristic acid (HRQ5vsQ1 = 0.83, 95%CI:0.74-0.93) and palmitic acid (HRQ5vsQ1 = 0.81, 95%CI:0.70-0.93) were inversely associated with CRC risk. Plasma myristic acid was also inversely associated with colon cancer risk (highest vs lowest quartile, ORQ4vsQ1 = 0.51; 95%CI:0.32-0.83), whereas a borderline positive association was found for plasma stearic acid (ORQ4vsQ1 = 1.63; 95%CI:1.00-2.64). Dietary total MUFA was inversely associated with colon cancer (per 1-SD increment, HR1-SD = 0.92, 95%CI: 0.85-0.98), but not rectal cancer (HR1-SD = 1.04, 95%CI:0.95-1.15, Pheterogeneity = 0.027). Dietary iTFA, and particularly elaidic acid, was positively associated with rectal cancer (HR1-SD = 1.07, 95%CI:1.02-1.13). Our results suggest that total and individual saturated fatty acids and fatty acids of industrial origin may be relevant to the aetiology of CRC. Both dietary and plasma myristic acid levels were inversely associated with colon cancer risk, which warrants further investigation.

Abstract: International audience; Nanostructuration is one of the most promising strategies to develop performant electrode materials for energy storage devices, such as hybrid supercapacitors. In this work, we studied the influence of precipitation medium and the use of a series of 1-alkyl-3-methylimidazolium bromide ionic liquids for the nanostructuration of β(III) cobalt oxyhydroxides. Then, the effect of the nanostructuration and the impact of the different ionic liquids used during synthesis were investigated in terms of energy storage performances. First, we demonstrated that forward precipitation, in a cobalt-rich medium, leads to smaller particles with higher specific surface areas (SSA) and an enhanced mesoporosity. Introduction of ionic liquids (ILs) in the precipitation medium further strongly increased the specific surface area and the mesoporosity to achieve well-nanostructured materials with a very high SSA of 265 m2/g and porosity of 0.43 cm3/g. Additionally, we showed that ILs used as surfactant and template also functionalize the nanomaterial surface, leading to a beneficial synergy between the highly ionic conductive IL and the cobalt oxyhydroxide, which lowers the resistance charge transfer and improves the specific capacity. The nature of the ionic liquid had an important influence on the final electrochemical properties and the best performances were reached with the ionic liquid containing the longest alkyl chain.

Abstract: Quantification of CO2 fluxes at the Earth’s surface is required to evaluate the causes and drivers of observed increases in atmospheric CO2 concentrations. Atmospheric inversion models disaggregate observed variations in atmospheric CO2 concentration to variability in CO2 emissions and sinks. They require prior constraints fossil CO2 emissions. Here we describe GCP-GridFED (version 2019.1), a gridded fossil emissions dataset that is consistent with the national CO2 emissions reported by the Global Carbon Project (GCP). GCP-GridFEDv2019.1 provides monthly fossil CO2 emissions estimates for the period 1959–2018 at a spatial resolution of 0.1°. Estimates are provided separately for oil, coal and natural gas, for mixed international bunker fuels, and for the calcination of limestone during cement production. GCP-GridFED also includes gridded estimates of O2 uptake based on oxidative ratios for oil, coal and natural gas. Abstract: International audience; Cost-effective and efficient sorbents with emulsion-breaking capability, especially surfactant-stabilized emulsions, are required to remediate oil spills from water. Herein, in situ growth of magnetic Mn0.01Co0.90Fe3O4 (MCFO) nanoparticles (NPs) on reduced graphene oxide (RGO) was performed to prepare MCFO/RGO nanocomposite via a simple solvothermal method. The nanocomposite was coated onto a polyurethane (PU) sponge to fabricate a novel magnetic, superhydrophobic/superoleophilic and flame-retardant material. The prepared magnetic MCFO/RGO/PU was superhydrophobic with a contact angle of 165 ± 1°. Compared with neat PU, the MCFO/RGO/PU sponge could be driven by a magnet for oil collection in water. More importantly, the MCFO/RGO/PU sponge could effectively collect oil droplets from surfactant-free and stabilized toluene/water emulsions. In addition, the fabricated sponge exhibited excellent performance for oils and organic solvents sorption, achieving up to 131 times of its own weight and retaining its ability even after 10 sorption cycles. Interestingly, the thermal stability and fire retardancy of the prepared PU sponge were improved with respect to neat PU.

Abstract: Regorafenib significantly prolonged progression-free survival (PFS) in pretreated patients with advanced non-adipocytic sarcoma (HR = 0.46, p &lt, 0.001) in a placebo-controlled, randomized, phase-II trial (NCT01900743). Thus, here, we assessed the prevalence of 57 biomarkers and their prognostic and predictive values for PFS and overall survival (OS). We analyzed 134/182 patients included in this trial, treated with regorafenib (n = 71, 53%) or placebo (n = 63, 47%). Mutational analyses were performed via full coding sequence analysis for 10 genes, and mutation hotspot panel for 50 genes (four genes in common). H19 was studied with RNA in-situ hybridization. The prognostic and predictive biomarkers&rsquo, values were studied only for biomarkers found positive/mutated in at least 10 patients. Overall, 25 out of 57 studied biomarkers, including five out of seven genes involved in angiogenesis, were found mutated/positive in at least one patient, of which 23 biomarkers had low prevalence (fewer than eight out of 134 patients), contrasting with H19 (n = 24, 18%), and TP53 (n = 35, 26%). However, in multivariable models of PFS and OS, including treatment effects and interactions, no significant prognostic or predictive values of the tested biomarkers were observed. Though several promising biomarkers were found to be positive/mutated, none of them were identified as viable predictive and prognostic biomarkers.

Abstract: Molecular dynamics (MD) simulations have been recorded on the complex between the edema factor (EF) of Bacilllus anthracis and calmodulin (CaM), starting from a structure with the orthosteric inhibitor adefovir bound in the EF catalytic site. The starting structure has been destabilized by alternately suppressing different co-factors, such as adefovir ligand or ions, revealing several long-distance correlations between the conformation of CaM, the geometry of the CaM/EF interface, the enzymatic site and the overall organization of the complex. An allosteric communication between CaM/EF interface and the EF catalytic site, highlighted by these correlations, was confirmed by several bioinformatics approaches from the literature. A network of hydrogen bonds and stacking interactions extending from the helix V of of CaM, and the residues of the switches A, B and C, and connecting to catalytic site residues, is a plausible candidate for the mediation of allosteric communication. The greatest variability in volume between the different MD conditions was also found for cavities present at the EF/CaM interface and in the EF catalytic site. The similarity between the predictions from literature and the volume variability might introduce the volume variability as new descriptor of allostery.

Abstract: International audience; Ethylene Glycol oxidation reaction on nickel and ruthenium modified palladium nanocatalysts was investigated with electrochemical, spectroelectrochemical, and chromatographic methods. These carbon-supported materials prepared by a revisited polyol approach, exhibited high activity towards the ethylene glycol electrooxidation in alkaline medium. Electrolysis coupled with High Performance Liquid Chromatography/Mass Spectrometry (HPLC-MS) and in situ Fourier Transform Infrared Spectroscopy (FTIRS) measurements allowed to determine the different compounds electrogenerated in the oxidative conversion of this two-carbon molecule. High value-added products such as oxalate, glyoxylate, and glycolate were identified in all the electrolytic solutions, while glyoxylate was selectively formed at the Ru45@Pd55/C electrode surface. In situ FTIRS results also showed a decrease of the pH value in the thin layer near the electrode as a consequence of OH- consumption during the spectroelectrochemical experiments.

Abstract: International audience; Processes using water at sub- and supercritical conditions (T > 300 °C) are used in industrial wastewater treatment, chemical recycling of wastes, biomass conversion and also in material synthesis, because of their rapidity and high efficiency. However, the salts injected or generated during the processes can lead to complex multiphasic water-salts systems with a behavior far to be understood in all the cases. Drastic changes of physics and chemistry of the systems are expected from water intrinsic properties; however, the formation of new phases, sometimes unexpected, near the critical point of water (374 °C; 22.1 MPa) also has a direct effect on the properties of the system. It is thus important to be conscious of the existence of these new phases to have the best control as possible on the physico-chemistry occurring inside the reactor. The aim of this review paper is to give an overview of these poorly-known new phases and to show that they are more encountered in SCW than it is thought.

Abstract: International audience; Nano-oxides and hydroxides generate great interest as promising positive electrode materials for the development of high energy density supercapacitors. However, their usually limited ionic and electronic conductivities significantly decrease their energy storage performances when increasing the electrode's mass loading. Here, we report on a sonochemical approach to functionalize the surface of Co(OH)2 nanomaterials by EmimBF4 ionic liquid that greatly improves the stability and the electrochemical performances of high mass loading electrodes (13 mg cm−2). This surface functionalization boosts the transport properties and strongly enhances the capacity as well as the capacity retention at higher current densities compared to basic Co(OH)2 (e.g. 113.5 C g−1 vs 59.2 C g−1 at 1 A g−1). Additionally, the protective layer formed by the ionic liquid stabilizes the electrode material upon cycling in KOH aqueous electrolyte and protects the material from oxidation upon open-air storage.

Abstract: International audience; The ability to design complex copper (Cu) parts into the most efficient thermal structures is an old dream, but difficult to realize with conventional manufacturing techniques. The recent development of laser 3D printing techniques makes it possible to fully explore intricate designs and maximize the thermal performance of Cu-based thermal management components but present significant challenges due to its high optical reflectivity. In this study, we demonstrated the laser 3D printing of pure Cu with a moderate laser power (400 W). Dense Cu parts (95 %) with smooth surface finishing (Ra ∼18 μm) were obtained at a scan speed of 400 mm/s, a hatch distance of 0.12 mm, and a layer thickness of 0.03 mm. The hardness, electrical, and thermal conductivity of the printed Cu parts are 108 MPa, 5.71 × 107 S/m, and 368 W/m·K, respectively which are close to those of bulk Cu. Additionally, complex heat sink structures were printed with large surface areas (600 mm2/g), and their cooling performances were compared to a commercial heat sink with a smaller surface area (286 mm2/g) on an electronic chip. The complex heat sinks printed cools the electronic chip 45 % more efficiently than the commercial one. The introduction of selective laser melting to additively manufacturing Cu heat sinks offers the promise to enhance the performance beyond the scope of exciting thermal management components.

Abstract: The development of advanced heat transfer fluids (HTF) with enhanced heat transfer properties has been identified as a key target to increase the efficiency of industrial processes. In this work, heat transfer performance improvements of a novel nanofluid, consisting of metallic nanoparticles dispersed in a commercial thermal oil, were investigated. Nanofluids combining tin nanoparticles (1 mass %) with Therminol 66 (TH66) were synthesised using the two step-method and experimentally analysed. The effectiveness of biosurfactant addition and nanoparticle polyethylene terephthalate (PET) nanocoating for high temperature nanofluid stabilisation were independently investigated. The PET nanoscale coatings were grown by molecular layer deposition, which has been used for the first time in this field. The thermal conductivity, dynamic viscosity and specific heat capacity of the stable, oil-based nanofluids were characterised at high temperatures, and the results were compared and in good agreement with models found in the relevant literature. Finally, the heat transfer performance of the nanofluids with respect to their base fluids was evaluated, employing empirical values for the thermophysical properties of the involved materials. In this way, increments of the heat transfer coefficients up to 9.3% at 140 °C, relevant to industrial applications were obtained.

Abstract: Six niobium rich glasses presenting different sodium contents were synthetized. On the basis of complementary infrared and Raman analysis, the influence of the sodium content and its role on the structure of the glasses prior to and following poling was examined. Correlative second harmonic generation (SHG)/Raman microscopy on the poled glasses cross section has shown a co-localization of the SHG signal and the structural variations. It also evidenced similar structural rearrangements whether sodium is removed through poling (230–300 °C) or through the alkali content defined by the starting glass compositions (melted at 1300–1500 °C). The effect of the sodium content on the optical properties, prior to and after thermal poling, is also discussed. It was found that refractive index variations induced by poling (ranging between 10−3 and 3 × 10−2) are mainly the result of a density decrease in the poled region rather than compositional and structural changes. The electro-optic origin of the poling-induced second order nonlinear response is confirmed by the Maker Fringe SHG analysis, and the evolution of χ(2) (2–2.5 pm/V) with the sodium content is discussed based on a selection of parameters influencing either the third order susceptibility [χ(3)] or the internal electric field strength of the poled layer.

Abstract: International audience; Exposure of mononuclear phagocytes to β-glucan, a naturally occurring polysaccharide, contributes to the induction of innate immune memory, which is associated with long-term epigenetic, metabolic, and functional reprogramming. Although previous studies have shown that innate immune memory induced by β-glucan confers protection against secondary infections, its impact on autoinflammatory diseases, associated with inflammasome activation and IL-1β secretion, remains poorly understood. In particular, whether β-glucan-induced long-term reprogramming affects inflammasome activation in human macrophages in the context of these diseases has not been explored. We found that NLRP3 inflammasome-mediated caspase-1 activation and subsequent IL-1β production were reduced in β-glucan-reprogrammed macrophages. β-Glucan acted upstream of the NLRP3 inflammasome by preventing potassium (K+) efflux, mitochondrial ROS (mtROS) generation, and, ultimately, apoptosis-associated speck-like protein containing a CARD (ASC) oligomerization and speck formation. Importantly, β-glucan-induced memory in macrophages resulted in a remarkable attenuation of IL-1β secretion and caspase-1 activation in patients with an NLRP3-associated autoinflammatory disease, cryopyrin-associated periodic syndromes (CAPS). Our findings demonstrate that β-glucan-induced innate immune memory represses IL-1β-mediated inflammation and support its potential clinical use in NLRP3-driven diseases.

Abstract: International audience; WO3−x (oxygen deficient tungsten oxide) thin films and WO3−x/Ta2O5 (tantalum oxide) composite films exhibit a huge photochromic effect with 60% near-infrared transmission modulation, in comparison to results in the literature. The colouring efficiency of such films results from both the stoichiometry of the particles (oxygen/tungsten deficient ratio) and the quality of the films (films are thin, homogeneous, and constituted of nanoparticles of about 5 nm in diameter and so with low scattering). Additionally, the high colouring efficiency obtained with a low fluence ultraviolet (UV) lamp is followed by a nearly complete bleaching phenomenon after a few hours in the dark. Beyond the attractiveness of our films toward multiple applications, and especially as films able to regulate the solar flow through smart windows, the optical properties and the colouring and bleaching kinetics were deeply investigated. For both the WO3−x thin films and the WO3−x/Ta2O5 composite films, colouring under irradiation and bleaching in the dark are phenomena that both proceed with a complex kinetics combining ultra-fast processes (from charge transfer, with a characteristic time of about one minute) and slower processes (linked to ionic diffusion, with a characteristic time of about one hour or even longer).

Abstract: Acyl coenzyme A binding protein (ACBP), also known as diazepam binding inhibitor (DBI) is a multifunctional protein with an intracellular action (as ACBP), as well as with an extracellular role (as DBI). The plasma levels of soluble ACBP/DBI are elevated in human obesity and reduced in anorexia nervosa. Accumulating evidence indicates that genetic or antibody-mediated neutralization of ACBP/DBI has anorexigenic effects, thus inhibiting food intake and inducing lipo-catabolic reactions in mice. A number of anorexiants have been withdrawn from clinical development because of their side effects including an increase in depression and suicide. For this reason, we investigated the psychiatric impact of ACBP/DBI in mouse models and patient cohorts. Intravenously (i.v.) injected ACBP/DBI protein conserved its orexigenic function when the protein was mutated to abolish acyl coenzyme A binding, but lost its appetite-stimulatory effect in mice bearing a mutation in the γ2 subunit of the γ-aminobutyric acid (GABA) A receptor (GABAAR). ACBP/DBI neutralization by intraperitoneal (i.p.) injection of a specific mAb blunted excessive food intake in starved and leptin-deficient mice, but not in ghrelin-treated animals. Neither i.v. nor i.p. injected anti-ACBP/DBI antibody affected the behavior of mice in the dark–light box and open-field test. In contrast, ACBP/DBI increased immobility in the forced swim test, while anti-ACBP/DBI antibody counteracted this sign of depression. In patients diagnosed with therapy-resistant bipolar disorder or schizophrenia, ACBP/DBI similarly correlated with body mass index (BMI), not with the psychiatric diagnosis. Patients with high levels of ACBP/DBI were at risk of dyslipidemia and this effect was independent from BMI, as indicated by multivariate analysis. In summary, it appears that ACBP/DBI neutralization has no negative impact on mood and that human depression is not associated with alterations in ACBP/DBI concentrations.

Abstract: Based on the evolution of the positions and intensities of the diffraction peaks, high energy X-ray diffraction (HEXRD) is recognized as the ultimate method to follow quantitatively in situ phase transformations in steels. However, the possible asymmetricity of diffraction peaks is seldom considered, and is known to bear information. A procedure for quantifying their skewness is proposed. In the case of a third generation high strength steel obtained by quench and partitioning (Q&P), the skewness is shown to be due to carbon heterogeneities at austenite/martensite interfaces developed at nanoscale, in agreement with prior post mortem atom probe tomography (APT) investigations.

Abstract: Gut microbiota composition influences the clinical benefit of immune checkpoints in patients with advanced cancer but mechanisms underlying this relationship remain unclear. Molecular mechanism whereby gut microbiota influences immune responses is mainly assigned to gut microbial metabolites. Short-chain fatty acids (SCFA) are produced in large amounts in the colon through bacterial fermentation of dietary fiber. We evaluate in mice and in patients treated with anti-CTLA-4 blocking mAbs whether SCFA levels is related to clinical outcome. High blood butyrate and propionate levels are associated with resistance to CTLA-4 blockade and higher proportion of Treg cells. In mice, butyrate restrains anti-CTLA-4-induced up-regulation of CD80/CD86 on dendritic cells and ICOS on T cells, accumulation of tumor-specific T cells and memory T cells. In patients, high blood butyrate levels moderate ipilimumab-induced accumulation of memory and ICOS + CD4 + T cells and IL-2 impregnation. Altogether, these results suggest that SCFA limits anti-CTLA-4 activity., The gut microbiota has been reported to regulate the efficacy of cancer therapy. Here, the authors show that short-chain fatty acids, which are generated through bacterial fermentation, increases immune tolerance leading to resistance to anti-CTLA-4 immunotherapy in mice and patients with metastatic melanoma.
