This paper describes a spontaneous outbreak of toxic myopathy in finishing pigs due to the ingestion of feed with salinomycin associated with tiamulin and addresses the control methods performed during the outbreak to minimize economic losses resulting from the intoxication. A lot of 940 pigs fed feed containing 30 ppm salinomycin received 100 ppm tiamulin (via water) to control recurrent respiratory diseases on the farm. After ingesting tiamulin, some animals manifested clinical signs of motor incoordination, stiff gait, reluctance to move, muscle weakness and tremors, dyspnea, depression, and decubitus, remaining in “sitting-dog position” or with the abduction of the pelvic limbs, and rested on tip-toes when in a standing position. Two animals were euthanized for macro-and microscopic evaluation. The other sick animals received supportive anti-inflammatory treatment. The most relevant macroscopic finding observed during necropsy was the slight pallor of the pelvic limb muscles. The main histopathological findings consisted of multifocal areas of hyaline degeneration and marked necrosis of skeletal myofibers, with macrophage infiltrate associated with cell regeneration and skeletal fiber phagocytosis. These lesions were more intense in the longissimus dorsi, diaphragm, and masseter muscles. The definitive diagnosis was based on epidemiological aspects and clinical lesional conditions compatible with toxic myopathy secondary to ionophore intoxication.

Electrospinning serves as a versatile means of understanding the effects of strong cylindrical confinement on encapsulated liquid crystals (LCs) and is a promising technique for developing functional fabrics and surfaces. In this work, we use electrospinning to create core-sheath nanofibers composed of a polyvinylpyrrolidone (PVP) sheath doped with a photochromic, azobenzene-based surfactant, and a low-molecular weight nematic liquid crystal core. The incorporation of an azobenzene surfactant into the polymer sheath allows for photochemical control over the nematic to isotropic transition temperature of the LC core resulting in ultra violet (UV) irradiation-induced depression of the phase transition temperature. Owing to the photochromic nature of the surfactant, the photoinduced decrease in the transition temperature is reversed with visible light irradiation with no notable fatigue resistance. At high surfactant loading rates, the temperature of the photoinduced phase transition is reduced to below room temperature allowing for the birefringence of the nanofibers “on” and “off” with irradiation and requires no external heating. Based on these oberservations, we propose photoisomerization of the azobenzene-based surfactant at the PVP/LC interface leads to surface induced disorder of the LC core changing the orientation from planar axial to random with UV irradiation and reversing that change with visible irradiation. This is further substantiated by examinig the LC alignment within nanofibers by imaging the fibers with the addition of a quarter-wave retardation plate. This method adds to the growing set of tools available for the control of the phase behavior of encapsulated liquid crystals and expands the functionality of electrospun materials.

Biological sand filtration systems are widely used for groundwater Mn removal. Bioaugmentation of sand filters through inoculation with Mn(II)-oxidizing bacteria (MOB) have contributed to the optimization of this biofiltration. However, a challenging aspect of this bioaugmentation process on a large scale is keeping fresh MOB cultures viable and transporting large culture volumes to groundwater treatment plants. In this work, powdered MOB inoculums were prepared by vacuum lyophilization. Bacterial lyophilization was performed in different growth conditions and the best performance was observed in biofilms covered with biogenic Mn oxides. On the other hand, a culture medium to produce the inoculum was developed using crude glycerol waste. Inoculums grown and lyophilized in this glycerol medium were able to be immobilized onto sand filters and to enhance the performance of groundwater Mn removal, reaching the optimal removal efficiency faster than fresh MOB cultures. These results generate new tools to simultaneously, re-use crude glycerol waste and improve large scale bioaugmentation approaches for groundwater Mn removal.

The Met80Ala and Met80Ala/Tyr67Ala variants of S. cerevisiae iso-1 cytochrome c (ycc) and their adducts with cardiolipin immobilized onto a gold electrode coated with a hydrophobic self-assembled monolayer (SAM) of decane-1-thiol were studied through cyclic voltammetry and surface-enhanced resonance Raman spectroscopy (SERRS). The electroactive species - containing a six-coordinate His/His axially ligated heme and a five-coordinate His/- heme stable in the oxidized and reduced state, respectively - and the pseudoperoxidase activity match those found previously for the wt species and are only slightly affected by CL binding. Most importantly, the reduced His/- ligated form of these variants is able to catalytically reduce the nitrite ion, while electrode-immobilized wt ycc and other His/Met heme ligated variants under a variety of conditions are not. Besides the pseudoperoxidase and nitrite reductase functions, which are the most physiologically relevant abilities of these constructs, also axial heme ligation and the equilibria between conformers are strongly affected by the nature - hydrophobic vs. electrostatic - of the non-covalent interactions determining protein immobilization. Also affected are the catalytic activity changes induced by a given mutation as well as those due to partial unfolding due to CL binding. It follows that under the same solution conditions the structural and functional properties of immobilized ycc are surface-specific and therefore cannot be transferred from an immobilized system to another involving different interfacial protein-SAM interactions.

Background: The work presented here is focused on the development of a comprehensive theoretical model for the description of drug release from a double - layer bioresorbable suture thread and the therapeutic efficacy of the active compounds delivered in the surrounding tissue. Methods: In particular, the system under investigation is composed of a core of slow-degrading polylactic- acid-co-ε-caprolactone (PLCL), where an antibiotic compound (Vancomycin) is loaded, surrounded by a shell of a fast-degrading polylactic-co-glycolic acid (PLGA) which contains an anesthetic drug (Lidocaine hydrochloride) for the post-surgical pain relief. Results: This system is of potential interest for the combined effects provided by the different active molecules, but the different release and polymer degradation dynamics, as well as their mutual influence, do not allow an intuitive a priori evaluation of device behavior, which can be rationalized through mathematical modeling. The model takes into account the main involved phenomena (polymer degradation and diffusion of the drugs within the device and the tissue, where they are metabolized) and their synergic effects on the overall system behavior. Conclusion: Model results are discussed in order to quantify the impact of the main design parameters on device performances, thanks to the use of phase diagrams (which show drug effect in time and space) whose insights are summarized in order to determine a design space according to the specific needs.

Circulating nucleic acids, such as short interfering RNA (siRNA), regulate many biological processes; however, the mechanism by which these molecules enter the cell is poorly understood. The role of extracellular‐matrix‐derived polymers in binding siRNAs and trafficking them across the plasma membrane is reported. Thermal melting, dynamic light scattering, scanning electron microscopy, and computational analysis indicate that hyaluronic acid can stabilize siRNA via hydrogen bonding and Van der Waals interactions. This stabilization facilitated HA size‐ and concentration‐dependent gene silencing in a CD44‐positive human osteosarcoma cell line (MG‐63) and in human mesenchymal stromal cells (hMSCs). This native HA‐based siRNA transfection represents the first report on an anionic, non‐viral delivery method that resulted in approximately 60 % gene knockdown in both cell types tested, which correlated with a reduction in translation levels. acceptedVersion

Onychomycosis is a common nail infection mainly caused by species belonging to the F. oxysporum, F. solani, and F. fujikuroi species complexes. The aim of this study was to evaluate the in vitro susceptibility of six representative strains of clinically relevant Fusarium spp. toward a set of natural-occurring hydroxycinnamic acids and their derivatives with the purpose to develop naturally occurring products in order to cope with emerging resistance phenomena. By introducing a prenylated chain at one of the hydroxy groups of trans-cinnamic acids 1&ndash, 3, ten prenylated derivatives (coded 4&ndash, 13) were preliminarily investigated in solid Fusarium minimal medium (FMM). Minimal inhibitory concentration (MIC) and lethal dose 50 (LD50) values were then determined in liquid FMM for the most active selected antifungal p-coumaric acid 3,3&prime, dimethyl allyl ester 13, in comparison with the conventional fungicides terbinafine (TRB) and amphotericin B (AmB), through the quantification of the fungal growth. Significant growth inhibition was observed for prenylated derivatives 4&ndash, 13, evidencing ester 13 as the most active. This compound presented MIC and LD50 values (62&ndash, 250 &micro, M and 7.8&ndash, 125 &micro, M, respectively) comparable to those determined for TRB and AmB in the majority of the tested pathogenic strains. The position and size of the prenylated chain and the presence of a free phenol OH group appear crucial for the antifungal activity. This work represents the first report on the activity of prenylated cinnamic esters and ethers against clinical Fusarium spp. and opens new avenues in the development of alternative antifungal compounds based on a drug repositioning strategy.

Background Hypoalbuminemia is frequently observed in patients with SARS‐CoV‐2 infection although its underlying mechanism and relationship with the clinical outcome still need to be clarified. Methods We retrospectively evaluated in patients with COVID‐19 hospitalised at the Fatebenefratelli‐Sacco Hospital in Milan, the prevalence of hypoalbuminemia, its association with the severity of COVID‐19, with the levels of C‐reactive protein, d‐dimer and interleukin‐6 and with clinical outcome over a follow‐up period of 30 days. Aims KIAA1549-BRAF fusions occur in certain brain tumours and provide druggable targets due to a constitutive activation of the MAP-kinase pathway. We introduce workflows for calling the KIAA1549-BRAF fusion from DNA methylation-array-derived copy number as well as DNA panel sequencing data. Methods Copy number profiles were analysed by automated screening and visual verification of a tandem duplication on chromosome 7q34, indicative of the KIAA1549-BRAF fusion. Pilocytic astrocytomas of the ICGC cohort with known fusion status were used for validation. KIAA1549-BRAF fusions were called from DNA panel sequencing data using the fusion callers Manta, Arriba with modified filtering criteria and deFuse. We screened DNA methylation and panel sequencing data of 7790 specimens from brain tumour and sarcoma entities. Results We identified the fusion in 337 brain tumours with both DNA methylation and panel sequencing data. Among these, we detected the fusion from copy number data in 84% and from DNA panel sequencing data in more than 90% using Arriba with modified filters. While in 74% the KIAA1549-BRAF fusion was detected from both methylation-array-derived copy number and panel sequencing data, in 9% it was detected from copy number data only and in 16% from panel data only. The fusion was almost exclusively found in pilocytic astrocytomas, diffuse leptomeningeal glioneuronal tumours and high-grade astrocytomas with piloid features. Conclusions The KIAA1549-BRAF fusion can be reliably detected from either DNA methylation array or DNA panel data. The use of both methods is recommended for the most sensitive detection of this diagnostically and therapeutically important marker.

In this paper we report on the use of an Ullmann-like aryl halide homocoupling reaction to obtain long Graphyne Molecular Wires (GY MWs) organized in dense, ordered arrays. Instead of using highly reactive terminal alkynes, we resort to a precursor wherein the acetylenic functional group is internal, namely protected by two phenyl rings, each bearing a Br atom in the para position to allow for linear homocoupling. In addition, two further factors concur with the production of dense and highly ordered arrays of very long GY MWs, namely the geometric compatibility between the substrate and both the organometallic intermediates and the final polymeric products of the synthesis, coupled with the presence of surface-adsorbed bromine atoms separating the MWs, which minimize inter-wire cross-linking secondary reactions.

Encapsulation of the nematic liquid crystal N-(4-methoxybenzylidene)-4-butylaniline (MBBA) into the core of poly(vinylpyrrolidone) (PVP) microfibers has been accomplished via coaxial electrospinning for the first time. Data from optical microscopy, two-dimensional Raman mapping, differential scanning calorimetry, and dielectric spectroscopy have been employed to gain detailed insights into the confinement effects on MBBA in a semiflexible polymer sheath. The electrospun fiber diameters could be tuned easily by modifying the flow rate of MBBA, and both the MBBA core and PVP sheath diameters were determined via Raman line cuts. The PVP sheath induced alignment of MBBA, where its long axis is parallel to the fiber axis. Further, the alignment of MBBA led to iridescence in the fibers, which was found to be temperature-sensitive and reversible. Synthetic scaffolds that possess an intrinsic capability to protect and sequester sensitive growth factors is a primary requisite for developing successful tissue engineering strategies. Growth factors such as recombinant human bone morphogenetic protein-2 (rhBMP-2) is highly susceptible to premature degradation and to provide a meaningful clinical outcome require high doses that can cause serious side effects. We discovered a unique strategy to stabilize and sequester rhBMP-2 by enhancing its molecular interactions with hyaluronic acid (HA), an extracellular matrix (ECM) component. We found that by tuning the initial protonation state of carboxylic acid residues of HA in a covalently crosslinked hydrogel modulate BMP-2 release at physiological pH by minimizing the electrostatic repulsion and maximizing the Van der Waals interactions. At neutral pH, BMP-2 release is primarily governed by Fickian diffusion, whereas at acidic pH both diffusion and electrostatic interactions between HA and BMP-2 become important as confirmed by molecular dynamics simulations. Our results were also validated in an in vivo rat ectopic model with rhBMP-2 loaded hydrogels, which demonstrated superior bone formation with acidic hydrogel as compared to the neutral counterpart. We believe this study provides new insight on growth factor stabilization and highlights the therapeutic potential of engineered matrices for rhBMP-2 delivery and may help to curtail the adverse side effects associated with the high dose of the growth factor. Cellulose nanopaper (CNP) features appealing properties, including transparency, flatness, a low thermal expansion coefficient and thermal stability, often outperforming conventional paper. However, free-standing crystalline cellulose films usually swell in water or upon moisture sorption, compromising part of their outstanding properties. This remains a major problem whenever working in a water environment is required. Freestanding cellulose nanopaper is prepared by solution casting water suspensions of cellulose nanocrystals with an average width of 10 nm and an average aspect ratio of 28, isolated from Avicel by acid hydrolysis and extensively characterized by AFM and FE-SEM measurements and GPC detection of their degree of polymerization. We demonstrate by elemental analyses, FT-IR, Raman spectroscopy, XRD measurements and water contact angle detection that wet treatment with lauroyl chloride results in surface hydrophobization of nanopaper. The hydrophobized nanopaper, C12-CNP, shows a more compact surface morphology than the starting CNP, due to the effect of chemical functionalization, and presents enhanced resistance to water, as assessed by electrochemical permeation experiments. The new hydrophobized nanopaper is a promising substrate for thin film devices designed to work in a humid environment.

Aberrant regulation of BCL-2 family members enables evasion of apoptosis and tumor resistance to chemotherapy. BCL-2 and functionally redundant counterpart, MCL-1, are frequently over-expressed in high-risk diffuse large B-cell lymphoma (DLBCL). While clinical inhibition of BCL-2 has been achieved with the BH3 mimetic venetoclax, anti-tumor efficacy is limited by compensatory induction of MCL-1. Voruciclib, an orally bioavailable clinical stage CDK-selective inhibitor, potently blocks CDK9, the transcriptional regulator of MCL-1. Here, we demonstrate that voruciclib represses MCL-1 protein expression in preclinical models of DLBCL. When combined with venetoclax in vivo, voruciclib leads to model-dependent tumor cell apoptosis and tumor growth inhibition. Strongest responses were observed in two models representing high-risk activated B-cell (ABC) DLBCL, while no response was observed in a third ABC model, and intermediate responses were observed in two models of germinal center B-cell like (GCB) DLBCL. Given the range of responses, we show that CIVO, a multiplexed tumor micro-dosing technology, represents a viable functional precision medicine approach for differentiating responders from non-responders to BCL-2/MCL-1 targeted therapy. These findings suggest that the combination of voruciclib and venetoclax holds promise as a novel, exclusively oral combination therapy for a subset of high-risk DLBCL patients.

The dynamics of polypropylene glycol, both neat and attached to silica nanoparticles, were investigated using elastic neutron backscattering and dielectric spectroscopy. The mean square displacement measured by the former is suppressed by the particles at temperatures corresponding to a dielectric secondary relaxation (that involves only a portion of the repeat unit) and the segmental relaxation (glass transition). Despite the suppression of the displacements, the motions are faster in the nanocomposite, primarily due to poorer packing (lower density) at the particle interface. At very low temperatures we discovered a new dynamic process in the polymer. Reflecting its very local nature, this process is unaffected by attachment of the chains to the silica.

Polylactic acid (PLA)-based polymers are ubiquitous in the biomedical field thanks to their combination of attractive peculiarities: biocompatibility (degradation products do not elicit critical responses and are easily metabolized by the body), hydrolytic degradation in situ, tailorable properties, and well-established processing technologies. This led to the development of several applications, such as bone fixation screws, bioresorbable suture threads, and stent coating, just to name a few. Nanomedicine could not be unconcerned by PLA-based materials as well, where their use for the synthesis of nanocarriers for the targeted delivery of hydrophobic drugs emerged as a new promising application. The purpose of the here presented review is two-fold: on one side, it aims at providing a broad overview of PLA-based materials and their properties, which allow them gaining a leading role in the biomedical field; on the other side, it offers a specific focus on their recent use in nanomedicine, highlighting opportunities and perspectives.

Desmoplastic/nodular medulloblastomas (DNMB) and medulloblastomas with extensive nodularity (MBEN) were outlined in the current WHO classification of tumors of the nervous system as two distinct histological MB variants. However, they are often considered as cognate SHH MB entities, and it is a reason why some clinical MB trials do not separate the patients with DNMB or MBEN histology. In the current study, we performed an integrated DNA/RNA-based molecular analysis of 83 DNMB and 36 MBEN to assess the etiopathogenetic relationship between these SHH MB variants. Methylation profiling revealed "infant" and "children" SHH MB clusters but neither DNMB nor MBEN composed separate epigenetic cohorts, and their profiles were intermixed within the "infant" cluster. In contrast, RNA-based transcriptional profiling disclosed that expression signatures of all MBEN were clustered separately from most of DNMB and a set of differentially expressed genes was identified. MBEN transcriptomes were enriched with genes associated with synaptic transmission, neuronal differentiation and metabolism, whereas DNMB profiling signatures included sets of genes involved in phototransduction and NOTCH signaling pathways. Thus, DNMB and MBEN are distinct tumor entities within the SHH MB family whose biology is determined by different transcriptional programs. Therefore, we recommend a transcriptome analysis as an optimal molecular tool to discriminate between DNMB and MBEN, which may be of benefit for patients' risk stratification in clinical trials. Molecular events identified in DNMB by RNA sequencing could be considered in the future as potent molecular targets for novel therapeutic interventions in treatment-resistant cases.

International audience; Conformationally rigid multipodal molecules should control the orientation and packing density of functional head groups upon self-assembly on solid supports. Common tripods frequently fail in this regard because of inhomogeneous bonding configuration and stochastic orientation. These issues are circumvented by a suitable tetrapodal diazatriptycene moiety, bearing four thiol-anchoring groups, as demonstrated in the present study. Such molecules form well-defined self-assembled monolayers (SAMs) on Au(111) substrates, whereby the tetrapodal scaffold enforces a nearly upright orientation of the terminal head group with respect to the substrate, with at least three of the four anchoring groups providing thiolate-like covalent attachment to the surface. Functionalization by condensation chemistry allows a large variety of functional head groups to be introduced to the tetrapod, paving the path toward advanced surface engineering and sensor fabrication.

Different approaches to modeling the gas phase polymerization of ethylene that have been considered in the literature are reviewed. It is shown that while simple, well-mixed models can give an adequate representation of the average performance of a given polymerization reactor, they do not allow one to analyze certain modeling problems such as the existence of temperature gradients or particle segregation, nor can they be used to treat operational modes such as condensed cooling where there can be up to three different phases in different parts of the reactor. For this, more complex models are required. These can take the form of compartmentalized models or even models based on computation fluid dynamics. However, long computational times can be required to fully exploit these models. The local segmental and global dynamics of a series of polypropylene glycol / silica nanocomposites were studied using rheometry and mechanical and dielectric spectroscopies. The particles cause substantial changes in the rheology, including higher viscosities that become non-Newtonian and the appearance of stress overshoots in the transient shear viscosity. However, no change was observed in the mean relaxation times for either the segmental or normal mode dynamics measured dielectrically. This absence of an effect of the particles is due to masking of the interfacial response by polymer chains remote from the particles. When the unattached polymer was extracted to isolate the interfacial material, very large reductions in the relaxation times were measured. This speeding up of the dynamics is due in part to the reduced density at the interface, presumably a consequence of poorer packing of tethered chains. In addition, binding of the ether oxygens of the polypropylene glycol chains truncates the normal mode, which shifts the corresponding relaxation peak to higher frequencies., 6 figures

Hydrogen peroxide (H2O2) and voltage-dependent K+ (KV) channels play key roles in regulating coronary blood flow in response to metabolic, ischemic, and paracrine stimuli. The KV channels responsible have not been identified, but KV7 channels are possible candidates. Existing data regarding KV7 channel function in the coronary circulation (limited to ex vivo assessments) are mixed. Thus we examined the hypothesis that KV7 channels are present in cells of the coronary vascular wall and regulate vasodilation in swine. We performed a variety of molecular, biochemical, and functional (in vivo and ex vivo) studies. Coronary arteries expressed KCNQ genes (quantitative PCR) and KV7.4 protein (Western blot). Immunostaining demonstrated KV7.4 expression in conduit and resistance vessels, perhaps most prominently in the endothelial and adventitial layers. Flupirtine, a KV7 opener, relaxed coronary artery rings, and this was attenuated by linopirdine, a KV7 blocker. Endothelial denudation inhibited the flupirtine-induced and linopirdine-sensitive relaxation of coronary artery rings. Moreover, linopirdine diminished bradykinin-induced endothelial-dependent relaxation of coronary artery rings. There was no effect of intracoronary flupirtine or linopirdine on coronary blood flow at the resting heart rate in vivo. Linopirdine had no effect on coronary vasodilation in vivo elicited by ischemia, H2O2, or tachycardia. However, bradykinin increased coronary blood flow in vivo, and this was attenuated by linopirdine. These data indicate that KV7 channels are expressed in some coronary cell type(s) and influence endothelial function. Other physiological functions of coronary vascular KV7 channels remain unclear, but they do appear to contribute to endothelium-dependent responses to paracrine stimuli.

In the last decades bioresorbable and biodegradable polymers have gained a very good reputation both in research and in industry thanks to their unique characteristics. They are, indeed, able to ensure high performances and biocompatibility, at the same time avoiding post-healing surgical interventions for devices removal. In the medical device industrial use of such biopolymers, it is widely known that product formulation and manufacturing need to follow specific procedures in order to ensure both proper mechanical properties and desired degradation profile. Moreover, also the sterilization method is crucial and its impact on physical properties is generally underestimated. In this work we focused our attention on the effect of different terminal sterilization methods on two commercially available poly(L-lactide-co-ε-caprolactone) with equivalent chemical composition (70% PLA and 30% PCL) and relatively similar initial molecular weight, but different chains arrangement and crystallinity. Results obtained show that crystallinity plays a key role, helping in preserving the narrow distribution of chains and, as a consequence, defined physical properties. These statements can be used as guidelines for a better choice of the most adequate biodegradable polymers in the production of resorbable medical devices.

Nonlinear dielectric measurements were carried out on two strongly polar liquids, 4-vinyl-1,3-dioxolan-2-one (VPC) and 4-ethyl-1,3-dioxolan-2-one (EPC), having chemical structures differing from propylene carbonate (PC) only by the presence of a pendant group. Despite their polarity, the compounds are all non-associated, "simple" liquids. From the linear component of the dielectric response, the α relaxation peak breadth was found to be invariant at a fixed value of the relaxation time, τα. From spectra from the nonlinear component, the number of dynamically correlated molecules was determined; it was also constant at fixed τα. Thus, two manifestations of dynamic heterogeneity depend only on the time constant for structural reorientation. More broadly, the cooperativity of molecular motions for non-associated glass-forming materials is connected to (i.e., reciprocally governs) the time scale. The equation of state for the two liquids was also obtained from density measurements made over a broad range of pressures and temperatures. Using these data, it was determined that the relaxation times of both liquids conform to density scaling. The effect of density, relative to thermal effects, on the α relaxation increases going from PC < VPC < EPC.

In this work we focused our attention on the effect of different terminal sterilization methods on two commercially available poly(l-lactide-co-ε-caprolactone) with equivalent chemical composition (70% PLA and 30% PCL) and relatively similar initial molecular weights, but different chain arrangements and crystallinity. Results obtained show that crystallinity plays a key role in helping preserve the narrow distribution of chains and, as a consequence, defined physical properties. The increase of heavy metal concentrations in atmospheric aerosols, due to both natural and anthropogenic emissions, keeps high the interest around the resulting damages to health and to climate change. In this paper, we investigate the feasibility of using the atmospheric aerosol as a source of secondary raw materials, by estimating its contents in economically significant elements. We characterised the geochemical composition of the atmospheric aerosol of urban (Firenze), sub-urban (Arezzo), and rural (Piancastagnaio, Siena) areas in Tuscany, analysing common major, minor, and trace constituents of silicate rocks using inductively-coupled plasma atomic emission and mass spectrometry (ICP-AES and ICP-MS). We also determined the radiogenic isotope composition of the aerosols (Sr, Nd, and Pb) through thermal ionization mass spectrometry (TIMS). Although the results indicate that elemental quantities are not at exploitable grade, we think that this challenge has more advantages than simply the recovery of inorganic pollutants dispersed into the atmosphere: with appropriate technologies, the designed industrial plant could also serve to drastically reduce the content of Volatile Organic Compounds occurring in atmosphere, especially in urban areas. Considering this perspective, benefits undoubtedly overwhelm the high-costs of the business, providing the most precious commodity for life: clean air for the next generations to be living in our Planet.

Four thiolated oligoarylene molecules (i) 4-methoxy-terphenyl-4 ''-methanethiol (MTM), (ii) 4-methoxy-terphenyl-3 '',5 ''-dimethanethiol (MTD), (iii) 4-nitro-terphenyl-4 ''-methanethiol (NTM), and (iv) 4-nitro-terphenyl-3 '',5 ''-dimethanethiol (NTD) were synthesized and self-assembled as monolayers (SAMs) on polycrystalline Au electrodes of organic field-effect transistors (OFETs). SAMs were characterized by contact angle and AC/DC electrochemical measurements, whereas atomic force microscopy was used for imaging the pentacene films grown on the coated electrodes. The electrical properties of functionalized OFETs, the electrochemical SAMs features and the morphology of pentacene films were correlated to the molecular organization of the thiolated oligoarylenes on Au, as calculated by means of the density functional theory. This multi-methodological approach allows us to associate the systematic replacement of the SAM anchoring head group (viz. methanethiol and dimethanethiol) and/or terminal tail group (viz. nitro-, -NO2, and methoxy, -OCH3) with the change of the electrical features. The dimethanethiol head group endows SAMs with higher resistive features along with higher surface tensions compared with methanethiol. Furthermore, the different number of thiolated heads affects the kinetics of Au passivation as well as the pentacene morphology. On the other hand, the nitro group confers further distinctive properties, such as the positive shift of both threshold and critical voltages of OFETs with respect to the methoxy one. The latter experimental evidence arise from its electron-withdrawing capability, which has been verified by both DFT calculations and DC electrochemical measurements

A multilayer self-assembled monolayer (MuL-SAM) composed by i) cysteamine, ii) 4-formilphenyl boronic acid and iii) dopamine has been used to functionalize polycrystalline Au. Its electrochemical characterization shows how this surface engineering makes Au reactive toward dopamine. Kelvin probe measurements have shown how MuL-SAM tunes the surface potential of the bare Au. Such measurements are correlated with the electrical performances of organic field-effect transistors (OFETs), whose source/drain electrodes have been functionalized by MuL-SAM. Such devices, operated as charge-injection organic gauges (CIOGs), have shown sensitivity to the concentration of dopamine in the range of [1, 50] nM.

In this work we quantified the role of electrostatic interactions in the self-assembly of a model amphiphilic peptide (RADA 16-I) into fibrillar structures by a combination of size exclusion chromatography and molecular simulations. For the peptide under investigation, it is found that a net charge of +0.75 represents the ideal condition to promote the formation of regular amyloid fibrils. Lower net charges favor the formation of amorphous precipitates, while larger net charges destabilize the fibrillar aggregates and promote a reversible dissociation of monomers from the ends of the fibrils. By quantifying the dependence of the equilibrium constant of this reversible reaction on the pH value and the peptide net charge, we show that electrostatic interactions contribute largely to the free energy of fibril formation. The addition of both salt and a charged destabilizer (guanidinium hydrochloride) at moderate concentration (0.3-1 M) shifts the monomer-fibril equilibrium toward the fibrillar state. Whereas the first effect can be explained by charge screening of electrostatic repulsion only, the promotion of fibril formation in the presence of guanidinium hydrochloride is also attributed to modifications of the peptide conformation. The results of this work indicate that the global peptide net charge is a key property that correlates well with the fibril stability, although the peptide conformation and the surface charge distribution also contribute to the aggregation propensity.

The thermal, rheological, and mechanical and dielectric relaxation properties of exfoliated dispersions of montmorillonite clay in a molecular liquid, polychlorobiphenyl (PCB), were studied. The viscosity enhancement at low concentrations of clay (≤5%) exceeded by a factor of 50 the increase obtainable with conventional fillers. However, the effect of the nanoclay on the local dynamics, including the glass transition temperature, was quite small. All materials herein conformed to density-scaling of the reorientation relaxation time of the PCB for a common value of the scaling exponent. A new relaxation process was observed in the mixtures, associated with PCB molecules in proximity to the clay surface. This process has an anomalously high dielectric strength, suggesting a means to exploit nanoparticles to achieve large electrical energy absorption. This lower frequency dispersion has a weaker dependence on pressure and density, consistent with dynamics constrained by interactions with the particle surface.

Electrolyte-gated organic field-effect transistors (EGOFETs), based on ultrathin pentacene films on quartz, were operated with electrolyte solutions whose pH was systematically changed. Transistor parameters exhibit nonmonotonic variation versus pH, which cannot be accounted for by capacitive coupling through the Debye–Helmholtz layer. The data were fitted with an analytical model of the accumulated charge in the EGOFET, where Langmuir adsorption was introduced to describe the pH-dependent charge buildup at the quartz surface. The model provides an excellent fit to the threshold voltage and transfer characteristics as a function of the pH, which demonstrates that quartz acts as a second gate controlled by pH and is mostly effective from neutral to alkaline pH. The effective capacitance of the device is always greater than the capacitance of the electrolyte, thus highlighting the role of the substrate as an important active element for amplification of the transistor response.

A series of hydrophilic alkanethiols, HS(CH2)(n)OH with n = 3, 4, 6, 8, 9 and 11, have been self-assembled on gold electrodes of pentacene-based thin-film transistors. The multiparametric and ultra-sensitive response of these devices allows us to characterize both charge-injection and electrical stability moving from vacuum to air. The decay exponent beta of charge injection is found to be 0.7-1.1 angstrom(-1) in agreement with earlier measurements of charge tunneling done by electrochemistry and large-area molecular junctions. We find that the intermediate chain lengths (n = 4 and 6) yield an optimum response as they represent the best compromise in terms of decrease of the contact-resistance and bias stress.

An experimental study was carried out of various factors affecting the ballistic penetration resistance of elastomer/steel bilayers. For blunt penetrators, the contribution of the coating to performance is optimized using the hardest substrates, front surface placement of the elastomer, and (when normalizing by added weight) thin, ca. 2–3 mm, coatings. These results, none of which are predicted by existing models, evince the marked coupling of coating and substrate in the impact response of the bilayer. We also show that nanoparticle fillers have a modest effect on ballistic performance of polyurea coatings, changing the penetration velocity by a few percent or less. This contrasts with the linear dynamic mechanical behavior, which shows much more significant increases in energy absorption due to nano-reinforcement.

The relaxational dynamics of poly(propyleneglycol) (PPG) and poly(propyleneglycol)-diglycidylether (PPGDE) compounds, the latter with different average number of repeating units, was studied by broadband dielectric spectroscopy (10 mHz to 10 GHz) from above to below the glass transition temperature in the range 115–350 K. The dielectric response of all systems showed a high temperature single relaxation while below a crossover temperature TB it split in a main (α-) and a secondary (β-) relaxation. The effect on the main and secondary relaxation of different groups terminating the polymer chain was analyzed: in particular, a decrease of the secondary relaxation time was observed for PPGDE with respect to PPG with the same molecular weight. Moreover the change of the Vogel temperature T0 paralleled what previously found for similar systems. The relationship between main and secondary relaxation and the microscopic nature of this latter were analyzed in the framework of the coupling model.

Triple negative breast cancer (TNBC) is a very aggressive subgroup of breast carcinoma, still lacking specific markers for an effective targeted therapy and with a poorer prognosis compared to other breast cancer subtypes. In this study we investigated the possibility that TNBC cells contribute to the establishment of tumor vascular network by the process known as vasculogenic mimicry, through endothelial cell differentiation. Vascular-like functional properties of breast cancer cell lines were investigated in vitro by tube formation assay and in vivo by confocal microscopy, immunofluorescence or immunohistochemistry on frozen tumor sections. TNBCs express endothelial markers and acquire the ability to form vascular-like channels in vitro and in vivo, both in xenograft models and in human specimens, generating blood lacunae surrounded by tumor cells. Notably this feature is significantly associated with reduced disease free survival. The impairment of the main pathways involved in vessel formation, by treatment with inhibitors (i.e. Sunitinib and Bevacizumab) or by siRNA-mediating silencing, allowed the identification of PDGFRβ and FGFR2 as relevant players in this phenomenon. Inhibition of these tyrosine kinase receptors negatively affects vascular lacunae formation and significantly inhibits TNBC growth in vivo. In summary, we demonstrated that TNBCs have the ability to form vascular-like channels in vitro and to generate blood lacunae lined by tumor cells in vivo. Moreover, this feature is associated with poor outcome, probably contributing to the aggressiveness of this breast cancer subgroup. Finally, PDGFRβ and FGFR2-mediated pathways, identified as relevant in mediating this characteristic, potentially represent valid targets for a specific therapy of this breast cancer subgroup.

Background Arginine:glycine amidinotransferase deficiency (AGAT-d) is a very rare inborn error of creatine synthesis mainly characterized by absence of brain Creatine (Cr) peak, intellectual disability, severe language impairment and behavioural disorder and susceptible to supplementary Cr treatment per os. Serial examinations by magnetic resonance spectroscopy are required to evaluate Cr recovery in brain during treatment of high doses of Cr per os, which have been proved beneficial and effective in treating main clinical symptoms. A long term study with detailed reports on clinical, neurochemical and neuropsychological outcomes of the first Italian patients affected by AGAT-d here reported can represent a landmark in management of this disorder thus enhancing medical knowledge and clinical practice. Results We have evaluated the long term effects of Cr supplementation management in four Italian patients affected by AGAT-d, correlating specific treatments with serial clinical, biochemical and magnetic resonance spectroscopy examinations as well as the neuropsychological outcome by standardized developmental scales. Consecutive MRS examinations have confirmed that Cr depletion in AGAT-d patients is reversible under Cr supplementation. Cr treatment is considered safe and well tolerated but side effects, including weight gain and kidney stones, have been reported. Conclusions Early treatment prevents adverse developmental outcome, while patients diagnosed and treated at an older age showed partial but significant cognitive recovery with clear improvements in adaptive functioning.

To analyze the Johari–Goldstein (JG) secondary process, dielectric relaxation measurements were carried out on poly(methyl methacrylate) (PMMA) and poly(thiomethyl methacrylate) (PTMA). The latter has a sulfur atom replacing the oxygen in the ester of PMMA, making the pendant group much less polar. This weakens the intensity of the secondary relaxation, enabling facile resolution of the JG and primary structural relaxation peaks. We find that the JG relaxation time of PTMA has a substantially larger activation energy (factor of 2) in the liquid compared to the glassy state. Although deconvolution of the peaks in PMMA is more ambiguous due to the large dielectric strength of its JG relaxation, applying the same analysis leads to a very similar result. In light of previous results, we conclude very generally from their temperature-dependence that JG secondary motions sense the glass transition, consistent with their role as precursor to structural relaxation.

Emerging evidence suggests that Ret oncoproteins expressed in medullary thyroid cancer (MTC) might evade the pro-apoptotic function of the dependence receptor proto-Ret by directly impacting the apoptosis machinery. Identification of the molecular determinants of the interplay between Ret signaling and apoptosis might provide a relevant contribution to the optimization of Ret-targeted therapies. Here, we describe the cross-talk between Ret-M918T oncogenic mutant responsible for type 2B multiple endocrine syndrome (MEN2B), and components of death receptor-mediated extrinsic apoptosis pathway. In the human MEN2B-type MTC cell line MZ-CRC-1 expressing Ret-M918T, Ret was found associated with Fap-1, known as inhibitor of the CD95 death receptor trafficking to the cell membrane, and with procaspase-8, the initiator pro-form caspase in the extrinsic apoptosis pathway. Cell treatment with the anti-tumor Ret kinase inhibitor RPI-1 inhibited tyrosine phosphorylation of procaspase-8, likely inducing its local activation, followed by downregulation of both Ret and Fap-1, and translocation of CD95 into lipid rafts. According to the resulting increase of CD95 cell surface expression, the CD95 agonist antibody CH11 enhanced RPI-1-induced cell growth inhibition and apoptosis. RET RNA interference downregulated Fap-1 protein in MZ-CRC-1 cells, whereas exogenous RET-M918T upregulated Fap-1 in HEK293 cells. Overall, these data indicate that the Ret oncoprotein exerts opposing controls on Fap-1 and CD95, increasing Fap-1 expression and decreasing CD95 cell surface expression. The functional interplay of the Ret mutant with the extrinsic apoptosis pathway provides a mechanism possibly contributing to MTC malignant phenotype and a rational basis for novel therapeutic strategies combining Ret inhibitors and CD95 agonists.

Matrix metalloproteinases (MMPs) are important factors in gliomas since these enzymes facilitate invasion into the surrounding brain and participate in neovascularization. In particular, the gelatinases (MMP-2 and MMP-9), and more recently MMP-25, have been shown to be highly expressed in gliomas and have been associated with disease progression. Thus, inhibition of these MMPs may represent a promising non-cytotoxic approach to glioma treatment. We report herein the synthesis and biological evaluation of a series of 4-butylphenyl(ethynylthiophene)sulfonamido-based hydroxamates. Among the new compounds tested, a promising derivative, 5a, was identified, which exhibits nanomolar inhibition of MMP-2, MMP-9, and MMP-25, but weak inhibitory activity towards other members of the MMP family. This compound also exhibited anti-invasive activity of U87MG glioblastoma cells at nanomolar concentrations, without affecting cell viability.

Structural characterization of poly-l-lactic acid (P(L)LA) and poly(glycolic acid) (PGA) oligomers containing three units was carried out with an atomistic approach. Oligomer structures were first optimized through quantum chemical calculations, using density functional theory (DFT); rotational barriers concerning dihedral angles along the chain were then investigated. Diffusion coefficients of l-lactic acid and glycolic acid in pure water were estimated through molecular dynamic (MD) simulations. Monomer structures were obtained with quantum chemical computation in implicit water using DFT method; atomic charges were fitted with Restrained Electrostatic Potentials (RESP) formalism, starting from electrostatic potentials calculated with quantum chemistry. MD simulations were carried out in explicit water, in order to take into account solvent presence.

In this study hydrogels synthesized from agarose and carbomer 974P macromers were selected for their potential application in spinal cord injury (SCI) repair strategies following their ability to carry cells and drugs. Indeed, in drug delivery applications, one of the most important issues to be addressed concerns hydrogel ability to provide a finely controlled delivery of loaded drugs, together with a coherent degradation kinetic. Nevertheless, solute effects on drug delivery system are often neglected in the large body of literature, focusing only on the characterization of unloaded matrices. For this reason, in this work, hydrogels were loaded with a chromophoric salt able to mimic, in terms of steric hindrance, many steroids commonly used in SCI repair, and its effects were investigated both from a structural and a rheological point of view, considering the pH-sensitivity of the material. Additionally, degradation chemistry was assessed by means of infrared bond response (FT-IR) and mass loss.

Agar-Carbomer (branched poly (acrylic acid)) hydrogel, an injectable bio-resorbable scaffold with a controlled nanostructure specifically designed for neural cell housing, was developed together with a new protocol for building three dimensional biohybrid cell/hydrogel systems. In order to overcome classic structural analysis inconveniences due to the high water amount, which affects instruments results and reliability, agar-Carbomer hydrogels were synthesized by microwave-assisted block copolymerization together with La 3+ salts. Propylene glycol, glycerol and buffered saline solution were used as cross-linking agents and solvent, respectively. Biomaterial properties were not affected by the presence of lanthanum, and were checked via swelling and rheological analysis. Moreover, the presence of La 3+ within the polymeric network was characterized by thermogravimetric analysis, environmental scanning electron microscopy and Fourier transformed infrared spectroscopy. The results showed that the rare earth presented uniform distribution in the hydrogel network due to the formation of chemical bonds after polymerization without being modified its luminescence emission spectrum that allowed hydrogel detection. These results made the obtained host-guest system a useful tool for analytical research studies concerning regenerative medical applications that could also be potentially taken up with in vivo experiments.

Replacement of the axial Met80 heme ligand in electrode-immobilized cytochrome c with a noncoordinating Ala residue and alteration of the hydrogen bonding network in the region nearby following substitution of Tyr67 were investigated as effectors of the thermodynamics and kinetics of the protein-electrode electron transfer (ET) and the heme-mediated electrocatalytic reduction of H(2)O(2). To this end, the voltammetry of the Met80Ala, Met80Ala/Tyr67His, and Met80Ala/Tyr67Ala variants of yeast iso-1-cytochrome c chemisorbed on carboxyalkanethiol self-assembled monolayers was measured at varying temperature and hydrogen peroxide concentration. The thermodynamic study shows that insertion of His and Ala residues in place of Tyr67 results mainly in differences in protein-solvent interactions at the heme crevice with no relevant effects on the E degrees' values at pH 7, which for single and double variants range from approximately -0.200 to -0.220 V (vs SHE). On the contrary, both double variants show much lower ET rates compared to Met80Ala, most likely as a consequence of a change in the ET pathways. In the present nondenaturing immobilizing conditions, and with hydrogen peroxide concentrations in the micromolar range, the variants catalyze H(2)O(2) reduction at the electrode, whereas wild-type cytochrome c does not. H(2)O(2) electrocatalysis occurs with an efficient mechanism likely involving a fast catalase-like process followed by electrocatalytic reduction of the resulting dioxygen at the electrode. Comparison of Met80Ala/Tyr67His with Met80Ala/Tyr67Ala shows that the presence of a general acid-base residue for H(2)O(2) recognition and binding through H-bonding in the distal heme site is a key requisite for the reductive turnover of this substrate.

The retinoid 4-oxo-N-(4-hydroxyphenyl)retinamide (4-oxo-4-HPR), a metabolite of fenretinide (4-HPR) present in plasma of 4-HPR–treated patients, is very effective in inducing growth inhibition and apoptosis in several cancer cell lines. 4-Oxo-4-HPR and 4-HPR have different mechanisms of action because 4-oxo-4-HPR, unlike 4-HPR, causes marked cell accumulation in G2-M phase. Here, we investigated the molecular events involving 4-oxo-4-HPR–induced cell cycle perturbation in ovarian (A2780 and IGROV-1) and breast (T47D, estrogen receptor+ and BT-20, estrogen receptor-) cancer cells. 4-Oxo-4-HPR induced a delay of mitosis (with mitotic index increasing 5- to 6-fold in all cell lines) without progression beyond the anaphase, as shown by cyclin B1 expression. 4-Oxo-4-HPR induced multipolar spindle formation and phosphorylation of BUBR1, resulting in activation of the spindle checkpoint. Multipolar spindles were not due to impairment of pole-focusing process, loss of centrosome integrity, or modulation of the expression levels of molecules associated with spindle aberrations (Kif 1C, Kif 2A, Eg5, Tara, tankyrase-1, centractin, and TOGp). We show here that 4-oxo-4-HPR targets microtubules because, in treated cells, it interfered with the reassembly of cold-depolymerized spindle microtubules and decreased the polymerized tubulin fraction. In cell-free assays, 4-oxo-4-HPR inhibited tubulin polymerization (50% inhibition of microtubule assembly at 5.9 μmol/L), suggesting a direct molecular interaction with tubulin. In conclusion, by showing that 4-oxo-4-HPR causes mitotic arrest through antimicrotubule activities, we delineate a new molecular mechanism for a retinoid. [Mol Cancer Ther 2009;8(12):3360–8]
