In this study, the solubilisation of waste coarse wool as a precursory step for the large scale valorisation of keratin was investigated using a green deep eutectic solvent (DES) based on L-cysteine and lactic acid. The investigation was undertaken via the response surface methodology and based on the Box-Behnken design for four process variables of temperature (70-110 °C), dissolution time (2-10 h), the mass of L-cysteine (0.5-2.5 g) in 20 mL of lactic acid, and wool load in the DES (0.2-0.6 g). The effect of variations in temperature was established to be the most significant process variable influencing keratin yield from waste coarse wool in the current work. An optimum keratin yield (93.77 wt.%) was obtained at the temperature of 105 °C, 8 h dissolution time, with 1.6 g L-cysteine in 20 mL of lactic acid using 0.5 g of wool. This study suggests L-cysteine and lactic acid as a green solvent with the potential to scale up keratin recovery from waste wool without significant destruction in the structure of the recovered keratin.HighlightsKeratin recovery from wool using deep eutectic solvent was assessedThe basis for the use of the new deep eutectic solvent was discussedThe effects of the process variables on keratin yield were exploredKeratin recovered was optimised and characterised.

Background HIV-2 resistance to integrase strand-transfer inhibitors (INSTIs) is characterized by two main pathways: (i) mutations at codons 143, 148 and155; and (ii) amino acid insertion after integrase codon 231 (231ins). Objectives To complete INSTI resistance data on HIV-2 by determining the viral replicative capacity and INSTI phenotypic susceptibility of integrase mutants obtained through site-directed mutagenesis. Methods Site-directed mutants (SDMs) were constructed and viral stocks produced. Viral replicative capacity was assessed by measuring HIV-2 viral load at days 3, 7 and 14. In vitro phenotypic susceptibility was measured using the ANRS PBMC assay. Results Viruses bearing 231ins did not present impaired replicative capacity, except the 231ins GIRGK mutant. A 231ins GK SDM was resistant to raltegravir and cabotegravir, but remained susceptible to dolutegravir and bictegravir. SDMs harbouring a 5 amino acid insertion (GYKGK or SREGK) were both resistant to all INSTIs. The SDM with T97A+N155H, with or without E92Q, was resistant to all INSTIs, except bictegravir. Conclusions These first data on the newly described resistance pathway 231ins, using site-directed mutagenesis, showed no measurable impact on viral fitness and confirmed the decreased susceptibility to a first-generation INSTI (raltegravir) and cabotegravir. Resistance to second-generation INSTIs (dolutegravir and bictegravir) occurred for mutants with a 5 amino acid 231ins.

Adipocytes increase energy expenditure in response to prolonged sympathetic activation via persistent expression of uncoupling protein 1 (UCP1)1,2. Here we report that the regulation of glycogen metabolism by catecholamines is critical for UCP1 expression. Chronic β-adrenergic activation leads to increased glycogen accumulation in adipocytes expressing UCP1. Adipocyte-specific deletion of a scaffolding protein, protein targeting to glycogen (PTG), reduces glycogen levels in beige adipocytes, attenuating UCP1 expression and responsiveness to cold or β-adrenergic receptor-stimulated weight loss in obese mice. Unexpectedly, we observed that glycogen synthesis and degradation are increased in response to catecholamines, and that glycogen turnover is required to produce reactive oxygen species leading to the activation of p38 MAPK, which drives UCP1 expression. Thus, glycogen has a key regulatory role in adipocytes, linking glucose metabolism to thermogenesis. Increased glycogen metabolism in adipocytes leads to expression of uncoupling protein 1, thereby linking glucose metabolism to thermogenesis.

International audience; For the first time, the possibility of obtaining porous germanium from both single-crystal and polycrystalline starting materials using environmentally friendly (in contrast to the commonly used HF and HCl) imidazolium ionic liquids (IL) was shown. It was demonstrated that pore formation depended significantly on the viscosity of the IL, on the etching current density and etching time, and on defects and imperfections of the substrate surface. By varying these parameters, the rates of two competing processes - formation and growth of pores and electropolishing - could be adjusted, consequently allowing germanium with a various structured surface, including a porous surface. In particular, using a more viscous IL (for example, [BMIM][PF6] versus [BMIM][BF4]) made it possible to reduce the rate of dissolution of the germanium dioxide surface layer; as a result, the formation and growth of pores became a dominant process. At the same time, the dissolution rate of the already formed porous layer (electropolishing) increased with the etching current density and the number of defects and imperfections of the germanium surface.

The objective of this research is to know the levels ofureu and creatinine serum for pre hemodialysis and post hemodialysis at RSU RoyalPrima Medan. The methods that are being used for this research is descriptive with54 patients as samples. The results of this research is loss of urea and creatinineserum level, with the average of urea level for pre hemodialysis is 128.11 mg/dL, andthe average of urea level for post hemodialysis is 43.26 mg/dL with 1,8% patient withlow urea level, 50% with normal urea level, and 48.2% with high urea level. Theaverage of creatine level for pre hemodialysis is 11.56 mg/dL, and the average ofcreatine levels is 4.3 mg/dL, with all of the patient still have a high creatinine levels.The conclusion is there is drop for both urea and creatinen serum level, but the urealevel for half of the patient did not go down to normal, and for the creatinine levelthere is none of the patient creatinine level that go down to normal.

Acetylene and ethylene are the smallest molecules that contain an unsaturated carbon–carbon bond and can be efficiently utilized in a large variety of cycloaddition reactions. In this account, pectin components were obtained from akebia trifoliate koiaz peels with the acid extraction technology. The chemical constituent of the akebia trifoliate koiaz peels extract (ATKPE) was analyzed and its anti-corrosion performance for mild steel in HCl medium was investigated by electrochemical methods, surface analysis and theoretical calculations. Experimental results show that ATKPE exhibits excellent corrosion inhibition characteristic, and its inhibition efficiency can be as high as 90% when the concentration of ATKPE is 800 mg/L. Electrochemical measurements show that ATKPE is a mixed-type corrosion inhibitor consisting with Langmuir adsorption model. In addition, SEM, AFM and contact angle observations suggests that the ATKPE may attach firmly to the metal surface by forming a barrier film. XPS and ATR-FTIR results further verify the bonding interaction between the functional groups and steel substrate. Meanwhile, quantum chemical calculations and molecular dynamics (MD) simulations were performed to reveal the inhibition mechanism at molecular/atomic level. In brief, our findings demonstrate a novel environmentally benign, efficient corrosion inhibitor for steel protection in acidic media.

Prolonged endoplasmic reticulum (ER) stress can mediate inflammatory myopathies and insulin signaling pathways. The double-stranded RNA (dsRNA)-activated protein kinase R (PKR) has been implicated in skeletal muscle dysfunction. However, pathological roles of PKR in ER stress in muscle are not fully understood. The current study aimed to investigate the effect of imoxin (IMX), a selective PKR inhibitor, on tunicamycin (TN)-induced promotion of ER stress and suppression of insulin signaling in C2C12 myotubes. Cells were pretreated with 5 µM IMX for 1 h and exposed to 0.5 µg/mL TN for 23 h. A subset of cells was stimulated with 100 nM insulin for the last 15 min. mRNA expression and protein levels involved in ER stress were measured by RT-PCR and Western blotting, respectively. TN significantly augmented PKR phosphorylation by 231%, which was prevented by IMX. In addition, IMX reduced mRNA and protein levels of ER stress-related markers, including CCAAT-enhancer-binding protein homologous protein (CHOP, mRNA: 95% decrease; protein: 98% decrease), activating transcription factor 4 (ATF4, mRNA: 69% decrease; protein: 99% decrease), cleavage of ATF6, and spliced X-box-binding protein 1 (XBP-1s, mRNA: 88% decrease; protein: 79% decrease), which were induced by TN. Furthermore, IMX ameliorated TN-induced suppression of phospho-insulin receptor β (317% increase) and Akt phosphorylation (by 36% at Ser473 and 30% at Thr308) in myotubes, while augmenting insulin-stimulated AS160 phosphorylation and glucose uptake (by ∼30%). These findings suggest that IMX may protect against TN-induced skeletal muscle ER stress and insulin resistance, which are potentially mediated by PKR.

Bromination of 3,4-dihydro-2H-thiopyran derivatives leads to bromo-substituted 3,4-dihydro-2H-thiopyrans or 4H-thiopyrans, depending on the reaction conditions and the structure of the starting heterocycles.

The analysis of products synthesized by Cu-catalyzed click reactions can be complicated due to the presence of metal impurities in isolated substances, which may "selectively" distort some signals in NMR spectra. Such a pronounced impurity effect was found in both 1H and 13C NMR spectra for a number of 1,4-substituted 1,2,3-triazoles. Recording of the full undistorted spectra is possible with additional product treatment, with more thorough purification, or by recording the spectra at low temperatures. The reasons for the distortion and disappearance of signals have been thoroughly studied; it was shown that impurities of paramagnetic metal ions in small amounts lead to this effect. Here, we want to deliver a warning message to the community: when all NMR signals in a spectrum are distorted, this situation is easy to detect. However, if only a few signals are "selectively" removed by impurities and the rest of the spectrum appears normal, this situation is much harder to notice. Therefore, incorrect conclusions about chemical structure may be obtained. Here, we demonstrated the example of Cu2+ ions, but one may anticipate a similar effect for other paramagnetic metal contaminants if the organic molecule has a functional group capable of coordination (heteroatom or a multiple bond).

Acetylene surrogates are efficient tools in modern organic chemistry with largely unexplored potential in the construction of heterocyclic cores. Two novel synthetic paths to 3,6-disubstituted pyridazines were proposed using readily available acetylene surrogates through flexible C2 unit installation procedures in a common reaction space mode (one-pot) and distributed reaction space mode (two-chamber): (1) an interaction of 1,2,4,5-tetrazine and its acceptor-functionalized derivatives with a CaC2 -H2 O mixture performed in a two-chamber reactor led to the corresponding pyridazines in quantitative yields; (2) [4+2] cycloaddition of 1,2,4,5-tetrazines to benzyl vinyl ether can be considered a universal synthetic path to a wide range of pyridazines. Replacing water with D2 O and vinyl ether with its trideuterated analog in the developed procedures, a range of 4,5-dideuteropyridazines of 95-99% deuteration degree was synthesized for the first time. Quantum chemical modeling allowed to quantify the substituent effect in both synthetic pathways.

The reaction space of the furanics-to-aromatics (F2A) conversion process for 5-hydroxymethylfurfural (HMF)-based platform chemicals has been explored both experimentally and by quantum chemistry methods. For the first time, a structure-activity relationship was established in furan-yne cycloaddition for a number of different HMF derivatives. Correlations between the activation energy of the cycloaddition stage and the structure of the substrates were established by molecular modeling methods. Analysis of the concerted and stepwise mechanisms of cycloaddition in the singlet and triplet electronic states of the molecular system was carried out. A series of biobased 7-oxanorbornadienes was obtained in the reaction with dimethyl acetylenedicarboxylate. Various methods of aromatization of the obtained [4+2] adducts have been examined. Rearrangement catalyzed by a Lewis acid leads to the formation of a phenol derivative, whereas reduction by diiron nonacarbonyl leads to the formation of functionalized benzene. Systematic study of the cycloaddition process has revealed a simple way to analyze and predict the relative reactivity of furanic substrates.

Coronavirus (CoV) nonstructural proteins (nsps) assemble to form the replication-transcription complex (RTC) responsible for viral RNA synthesis. nsp7 and nsp8 are important cofactors of the RTC, as they interact and regulate the activity of RNA-dependent RNA polymerase and other nsps. To date, no structure of the full-length SARS-CoV-2 nsp7:nsp8 complex has been published. The current understanding of this complex is based on structures from truncated constructs, with missing electron densities, or from related CoV species where SARS-CoV-2 nsp7 and nsp8 share upward of 90% sequence identity. Despite available structures solved using crystallography and cryo-EM representing detailed static snapshots of the nsp7:nsp8 complex, it is evident that the complex has a high degree of structural plasticity. However, relatively little is known about the conformational dynamics of the individual proteins and how they complex to interact with other nsps. Here, the solution-based structural proteomic techniques, hydrogen-deuterium exchange mass spectrometry (HDX-MS) and cross-linking mass spectrometry (XL-MS), illuminate the dynamics of SARS-CoV-2 full-length nsp7 and nsp8 proteins and the nsp7:nsp8 protein complex. Results presented from the two techniques are complementary and validate the interaction surfaces identified from the published three-dimensional heterotetrameric crystal structure of the SARS-CoV-2 truncated nsp7:nsp8 complex. Furthermore, mapping of XL-MS data onto higher-order complexes suggests that SARS-CoV-2 nsp7 and nsp8 do not assemble into a hexadecameric structure as implied by the SARS-CoV full-length nsp7:nsp8 crystal structure. Instead, our results suggest that the nsp7:nsp8 heterotetramer can dissociate into a stable dimeric unit that might bind to nsp12 in the RTC without significantly altering nsp7-nsp8 interactions.

Xanthan is an important polysaccharide widely used in many industrial fields. It is produced by the bacteria Xanthomonascampestris. Chemical modification of xanthan can open up new horizons for its use. In this work, xanthan butyl ester was obtained for the first time by the interaction of xanthan and bromobutane using sodium hydroxide as a catalyst. The composition and structure of the obtained new xanthan derivative was studied by elemental analysis, IR spectroscopy, X-ray phase analysis, scanning electron microscopy and thermal analysis. The introduction of a butyl group into the xanthan molecule was proved by elemental analysis and IR spectroscopy by the appearance of corresponding bands. It was shown by X-ray phase analysis that xanthan butyl ether has a more X-ray amorphous structure in comparison with the original xanthan. It was shown by scanning electron microscopy that xanthan butyl ether powder consists of particles of a larger size and a layered structure in comparison with the original xanthan. It has been shown by thermal analysis that xanthan butyl ether is less thermostable than the starting xanthan

The organotypic human air‐liquid‐interface (ALI) airway tissue model has been used as an in vitro cell culture system for evaluating the toxicity of inhaled substances. ALI airway cultures are highly differentiated, which has made it challenging to evaluate genetic toxicology endpoints. In the current study, we assayed DNA damage with the high‐throughput CometChip assay and quantified mutagenesis with Duplex Sequencing, an error‐corrected next‐generation sequencing method capable of detecting a single mutation per 107 base pairs. Fully differentiated human ALI airway cultures were treated from the basolateral side with 6.25 to 100 μg/mL ethyl methanesulfonate (EMS) over a period of 28 days. CometChip assays were conducted after 3 and 28 days of treatment, and Duplex Sequencing after 28 days of treatment. Treating the airway cultures with EMS resulted in time‐ and concentration‐dependent increases in DNA damage and a concentration‐dependent increase in mutant frequency. The mutations observed in the EMS‐treated cultures were predominantly C → T transitions and exhibited a unique trinucleotide signature relative to the negative control. Measurement of physiological endpoints indicated that the EMS treatments had no effect on anti‐p63‐positive basal cell frequency, but produced concentration‐responsive increases in cytotoxicity and perturbations in cell morphology, along with concentration‐responsive decreases in culture viability, goblet cell and anti‐Ki67‐positive proliferating cell frequency, cilia beating frequency, and mucin secretion. The results indicate that a unified 28‐day study can be used to measure several important safety endpoints in physiologically relevant human in vitro ALI airway cultures, including DNA damage, mutagenicity, and tissue‐specific general toxicity.

Background . Ceftazidime/Avibactam (C/A) and Ceftolozane/Tazobactam (C/T) are two novel antibacterial agents with a known efficacy on Gram-negative bacteria (GNB). Objectives . We aimed to describe the efficacy and safety of surgical management combined with C/A or C/T treatment for bone and joint infections (BJI). Patients and Methods . We conducted an observational, bi-centric study of patients treated with C/A or C/T for a BJI between May 2016 and June 2019. Failure was defined as a need for an unplanned additional antibiotic treatment or orthopedic surgery or death due to the BJI until the patient's latest visit. Results . Overall, 15 patients were included. Nine patients were treated with C/A, mainly for polymicrobial BJI due to multidrug-resistant (MDR) bacteria (Enterobacteriaceae n=7). Six patients were male; median age was 66 years-old, and median Charlson comorbidity index (CCI) was 5. It was the first septic episode on the site in 3/9 patients. Cure rate was 7/9 (median follow-up: 272 days). Two patients showed C/A-related confusion. Five patients were treated with C/T, for BJI involving MDR-Pseudomonas aeruginosa. Four patients were male; median age was 53 years-old, and median-CCI was 2. All patients had previous septic episodes on infectious site. Cure rate was 3/5 (median follow-up: 350 days). One patient was successfully treated by C/T then C/A for multistage spondylodiscitis. Conclusion . In our experience, C/A and C/T were two effective and safe options, even as salvage treatment of BJI due to MDR GNB despite the absence of label, but more data are warranted.

Background Multivariable baseline factor analysis across cabotegravir + rilpivirine clinical trials showed that HIV-1 subtypes A6/A1 and the presence of rilpivirine resistance-associated mutations (RAMs) were associated with an increased risk of virological failure of this dual therapy. The aim of this study was to describe the prevalence of genotypic baseline risk factors for cabotegravir + rilpivirine failure among ARV-naive patients. Patients and methods From 2010 to 2020, 4212 sequences from ARV-naive patients were collected from three large Parisian academic hospital genotypic databases. Cabotegravir and rilpivirine RAMs were defined according to the ANRS algorithm. Results Among 4212 ARV-naive patients, 38.6% were infected with subtype B, 32.4% with CRF02_AG (32.4%) and 5.1% with subtype A (85.5% being A6/A1 subtype). Overall, the presence of at least one cabotegravir or rilpivirine RAM was 16.2% and 14.3%, respectively. Considering genotypic resistance interpretation, using the ANRS algorithm, 0.74% (n = 31), 6.2% (n = 261) and 0.09% (n = 4) of sequences were resistant to cabotegravir, rilpivirine or both, respectively. The overall prevalence of L74I in integrase and E138A in RT was 13.0% and 3.2%, respectively, and stable over the decade. Thus, adding 183 subtype A6/A1 sequences to 244 sequences interpreted as resistant to rilpivirine led to 427 (10.1%) sequences combining both baseline virological risk factors for cabotegravir + rilpivirine dual-therapy failure. Conclusions Among large sequence databases, when adding prevalence of rilpivirine-resistant viruses and HIV-1 subtype A6/A1 sequences, 10.1% of patients would not be eligible for cabotegravir + rilpivirine dual therapy. These data re-emphasize the need for a pre-therapeutic genotypic resistance test to detect polymorphisms and transmitted drug resistance and to define HIV-1 subtype.

Polymeric ionogels, cross-linked gels swollen by ionic liquids (ILs), are useful vehicles for the release and storage of molecular solutes in separation, delivery, and other applications. Although rapid solute diffusion is often critical for performance, it remains challenging to predict diffusivities across multidimensional composition spaces. Recently, we showed that water (a neutral solute) diffuses through alkyl-methylimidazolium halide ILs by hopping between hydrogen bonding sites on relatively immobile cations. Here, we expand on this activated hopping mechanism in two significant ways. First, we demonstrate that water diffuses through poly(ethylene glycol)diacrylate ionogels via the same mechanism at a reduced rate. Second, we hypothesize that the activation energy barrier can be determined from relatively simple 1H NMR chemical shift measurements of the proton responsible for H-bonding. This relationship enables water's diffusivity in ionogels of this class to be predicted quantitatively, requiring only (1) the composition-dependent diffusivity and Arrhenius behavior of a single IL and (2) 1H NMR spectra of the ionogels of interest. High-throughput microfluidic Fabry-Perot interferometry measurements verify prediction accuracy across a broad formulation space (four ILs, 0 ≤ xH2O ≤ 0.7, 0 ≤ ϕPEGDA ≤ 0.66). The predictive model may expedite IL-material screening; moreover, it intimates a powerful connection between solute mobility and hydrogen bonding and suggests targets for rational design.

Strong and well-engineered interfaces between dissimilar materials are a hallmark of natural systems but have proven difficult to emulate in synthetic materials, where interfaces often act as points of failure. In this work, curing reactions that are triggered by exposure to different wavelengths of visible light are used to produce multimaterial objects with tough, well-defined interfaces between chemically distinct domains. Longer-wavelength (green) light selectively initiates acrylate-based radical polymerization, while shorter-wavelength (blue) light results in the simultaneous formation of epoxy and acrylate networks through orthogonal cationic and radical processes. The improved mechanical strength of these interfaces is hypothesized to arise from a continuous acrylate network that bridges domains. Using printed test structures, interfaces were characterized through spatial resolution of their chemical composition, localized mechanical properties, and bulk fracture strength. This wavelength-selective photocuring of interpenetrating polymer networks is a promising strategy for increasing the mechanical performance of 3D-printed objects and expanding light-based additive manufacturing technologies.

Copper-zinc superoxide dismutase (SOD1) is a major antioxidant metalloenzyme that protects cells from oxidative damage by superoxide anions (O2-). Structural, biophysical, and other characteristics have in the past been compiled for mammalian SOD1s and for the highly homologous fungal and bovine SOD1s. Here, we characterize the biophysical properties of a plant SOD1 from tomato chloroplasts and present several of its crystal structures. The most unusual of these structures is a structure at low pH in which tSOD1 harbors zinc in the copper-binding site but contains no metal in the zinc-binding site. The side chain of D83, normally a zinc ligand, adopts an alternate rotameric conformation to form an unusual bidentate hydrogen bond with the side chain of D124, precluding metal binding in the zinc-binding site. This alternate conformation of D83 appears to be responsible for the previously observed pH-dependent loss of zinc from the zinc-binding site of SOD1. Titrations of cobalt into apo tSOD1 at a similar pH support the lack of an intact zinc-binding site. Further characterization of tSOD1 reveals that it is a weaker dimer relative to human SOD1 and that it can be activated in vivo through a copper chaperone for the SOD1-independent mechanism.

The Mizoroki-Heck reaction is one of the most known and best studied catalytic transformations and has provided an outstanding driving force for the development of catalysis and synthetic applications. Three out of four classical Mizoroki-Heck catalytic cycle intermediates contain Pd-C bonds and are well known and studied in detail. However, a simple palladium hydride (which is formed after the product-releasing β-H-elimination step) is a kind of elusive intermediate in the Mizoroki-Heck reaction. In the present study, we performed a combined theoretical and mass spectrometry (MS) study of palladium hydride complexes [PdX2H]- (X = Cl, Br, and I), which are reactive intermediates in the Mizoroki-Heck reaction. Static and molecular dynamic calculations revealed that these species have a T-shaped structure with a trans-arrangement of halogen atoms. Other isomers of [PdX2H]- are unstable and easily rearrange into the T-shaped form or decompose. These palladium hydride intermediates were detected by MS in precatalyst activation using NaBH4, Et3N, and a solvent molecule as reducing agents. Online MS monitoring allowed the detection of [PdX2H]- species in the course of the Mizoroki-Heck reaction.

Hydration of the amphiphilic diblock oligomer C

Recognizing the importance of succinic acid as one of the most relevant platform molecules, the present study assesses the production of succinic acid from waste apple slurry/pomace as a sustainable and renewable feedstock. The assessment has been undertaken by incorporating technical and economic considerations in the analysis while also comparing two succinic acid production scenarios. The aforementioned considerations have been applied by utilizing process simulation results, generated from Aspen Plus software, as input data to undertake economic assessments using classic economic correlations. Employing well-defined system boundaries, scenarios to produce bio-succinic acid with either bioelectricity (scenario 1) or biogas (scenario 2) as co-products were therefore comparatively accessed. This study was able to demonstrate that waste pomace as a feedstock has the potential to generate low-cost succinic acid, while scenario 2 constituted the preferred pathway overall. This study results may provide valuable information to policy makers, to enable better decisions regarding the viability, design and execution of large-scale bio-succinic acid production projects based on waste apple pomace as the preferred feedstock.

Introduction: Diabetes mellitus (DM) is a group of metabolic diseases characterized by hyperglycemia resulting from defects of insulin secretion and/or increased cellular resistance to insulin. Abrusprecatoriushas been used for the treatment of various ailments including diabetes mellitus. This study is aimed at evaluating the possible anti-diabetic effect of the methanol fraction 2 (F2) of the seeds of Abrusprecatoriusin alloxan-induced diabetic Wistar albino rats. Methods: The methanol extract of the seeds of A. precatorius Linn Fabaceae was fractionated by Sephadex G15. Diabetes was induced by a single intraperitoneal administration of 150 mg/kg bodyweight of alloxan. Acute toxicity (LD50) study was done using Lorke’s method. The antidiabetic activities of Fraction 2 (F2) and the biochemical parameters were investigated using standard diagnostic methods. Results: The value of the LD50 is 529.2 mg/kg bw. Familial renal hypouricemia is a rare genetic disorder characterized by a defect in renal tubular urate reabsorption. Some patients present with exercise-induced acute kidney injury and nephrolithiasis. Type II is caused by mutations in the SLC2A9 gene. Here, we report the case of a young patient who developed acute kidney injury after exercise secondary to familial renal hypouricemia type II. The same mutation was found in other asymptomatic members of his family. We review the medical literature on this condition. This case highlights the importance of considering uric acid disorders in the work-up of acute kidney injury after exercise.

A new family of protic ammonium ionic liquids (ILs) with various inorganic anions was synthesized from bio-derived 5-HMF. Starting with cellulose biomass, a complete preservation of the C₆ unit was achieved throughout the synthetic sequence (no carbon loss). Evaluation of green metrics showed a significant advantage of the developed bio-derived pathway to access ILs from a natural renewable source, depending on feasible routes to 5-HMF manufacturing. The reduced number of synthetic steps and availability of the starting materials were the key advantages. Experimental physicochemical and biological studies, as well as computational modeling revealed a unique multifunctional intrinsic organization of these bio-derived ILs. The nature of interactions between the cations and anions of the novel ILs was mapped at the molecular level. The substituents in the cationic core and the nature of the original building blocks had a prominent impact on cytotoxicity of the novel ILs. The obtained results suggest possible sustainable applications of the least toxic ILs, while the regulation of biological activity of the ILs via the corresponding structural adjustments can find biological and medicinal applications. The 5-HMF-derived IL with a sulfate anion demonstrated potentially useful properties in dissolution of microcrystalline cellulose.

The Diels–Alder reaction of β-fluoro-β-nitrostyrenes with cyclic 1,3-dienes was investigated. A series of novel monofluorinated norbornenes was prepared in up to 97% yield. The reaction with 1,3-cyclohexadiene permits the preparation of monofluorinated bicyclo[2.2.2]oct-2-enes. The kinetic data of the reactions with 1,3-cyclopentadiene and 1,3-cyclohexadiene were used to calculate activation parameters. Furthermore, the synthetic utility of the cycloadducts obtained was demonstrated.

An efficient pathway toward a novel class of trifluoromethyl building blocks was elaborated. The reaction of α-CF3-enamines with arylaldehydes resulted in direct synthesis of α,β-diaryl-CF3-enones isolated in up to 93% yield as E-isomers. The possible reaction mechanism was proposed using the Zimmerman-Traxler model. The reaction of α,β-diaryl-CF3-enones with hydrazines opens a novel pathway to trifluoromethylated pyrazolines. Oxidation of pyrazolines with DDQ opened access to totally regioselective preparation of 3-CF3-pyrazoles isolated in high yield. Using this strategy, 4-arylated derivatives of known drugs Celebrex, Mavacoxib, and SC-560 can be synthesized.

Biomass is a renewable source of valuable feedstock for the chemical industry of the future. A promising approach to the utilization of valuable components of biomass is the synthesis of monomers and polymers, if the overall technology is designed for a clean cycle without pollution of the environment with newly created polymers. In this work, we have developed a methodology for the recycling of polymers based on biomass and calcium carbide. First, we modified a series of biomass-derived terpene alcohols with calcium carbide followed by polymerization of the isolated vinyl ethers. Then, to study the recycling potential, the obtained polymers were subjected to pyrolysis at moderate temperatures (200–450 °C). The pyrolysis products were analyzed using TGA-MS, GC-MS, and NMR, and it was found that the polymers can be transformed quite easily. The products of the pyrolysis consisted of the starting terpenols, as well as the corresponding non-toxic ketones or aldehydes: up to 87% of the starting alcohol or up to 100% of the total sum of alcohol + aldehyde or alcohol + ketone (GC-yields). Then, the reaction mixture was hydrogenated and resulted in the formation of starting alcohol only. According to the studied pathway of polymers re-building, a terpene fragment attached to the main polyethylene chain through an oxygen atom promotes the transformation of the obtained polymers. Thus, the products of pyrolysis are environmentally friendly and can be reused in the further synthesis of monomers. The developed system has shown a unique assembling/disassembling ability and advances the concept of reusable bio-derived high value-added materials.

Operations with nucleic acids are among the main means of studying the mechanisms of gene function and developing novel methods of molecular medicine and gene therapy. These endeavours usually imply the necessity of nucleic acid storage and delivery into eukaryotic cells. In spite of diversity of the existing dedicated techniques, all of them have their limitations. Thus, a recent notion of using ionic liquids in manipulations of nucleic acids has been attracting significant attention lately. Due to their unique physicochemical properties, in particular, their micro-structuring impact and tunability, ionic liquids are currently applied as solvents and stabilizing media in chemical synthesis, electrochemistry, biotechnology, and other areas. Here, we review the current knowledge on interactions between nucleic acids and ionic liquids and discuss potential advantages of applying the latter in delivery of the former into eukaryotic cells.

To monitor deficient or excessive intakes of biologically available fluoride (F), various biological samples have been tested for use as biomarkers of human exposure to F. Most such studies have concerned children and often have only involved measurement of F in 1 or 2 types of sample. The present study investigated the relationships of F concentrations in biomarkers of F exposure; including plasma, saliva, hair, finger- and toenails, and daily urinary F excretion (UFE) with the total daily F intake (TDFI) of adults. TDFI was assessed in 60 healthy adults, aged ≥20 years; 31 lived in a low-F water area (LFA, 0.04 mg F/L) and 29 in a high-F water area (HFA, 3.05 mg F/L) of Nigeria. All volunteers provided at least 1 biomarker sample from the above list and completed a questionnaire to evaluate F intake from the diet and toothpaste ingestion. TDFI, UFE and F concentrations of biomarkers were statistically significantly higher in the HFA than in the LFA. There were strong statistically significant positive correlations between TDFI and UFE (ρ = 0.730, p < 0.001); plasma F (ρ = 0.729, p < 0.001); fasting whole saliva F (ρ = 0.653, p < 0.001) and hair F (ρ = 0.603, p < 0.001). The statistically significant positive correlations between TDFI and fingernail F (ρ = 0.502, p < 0.001) and between TDFI and toenail F (ρ = 0.556, p < 0.001) were moderate. In conclusion, this study has indicated the usefulness of 24-h UFE as well as F concentration in plasma, fasting whole saliva and hair as biomarkers of contemporary or sub-chronic F exposure in groups of adults. However, they do not appear to have the necessary sensitivity to predict F exposure in individuals.

An efficient synthesis of 2-pyridine derived 4-azido-1,2,3-triazoles was elaborated using the corresponding dichlorodiazadienes as precursors. The reaction of the prepared 2-azine substituted diazadienes with sodium azide permits the preparation of target triazoles isolated in up to 92% yield. Subsequent thermal cyclization was studied. Elimination of molecular nitrogen promoted the cyclization of nitrene at the azine nitrogen. As a result, a family of 2H-[1,2,3]triazolo[4′,5′:3,4]pyrazolo[1,5-a]pyridin-5-ium-4-ides was prepared. The synthesized compounds are members of a new heterocyclic system. Moreover, these compounds are new attractive blue light emitting molecules.

Device-level applications of organic electrolytes unavoidably imply extensive contact with the environment. Despite their excellent scientific potential, ionic liquids (ILs) cannot be approved for practical usage until their life cycle and impact on the environment are assessed. In this work, we carried out the first large-scale study on the mechanisms of the cytotoxic action of various classes of ionic liquids, including imidazolium, pyridinium, pyrrolidinium, ammonium, and cholinium ILs (25 in total). We determined the biological effect of these ILs in seven cell lines of various origins (HEK293 (human embryonic kidney), U937 (human myeloid leukemia), Jurkat (human T-cell leukemia), HL60 (human acute promyelocytic leukemia), K562 (human chronic myelogenous leukemia), A549 (human alveolar adenocarcinoma), and A2780 (human ovarian carcinoma)). The induction of apoptosis in cells upon treatment with the majority of the ILs tested was subsequently demonstrated. The new data suggest that ILs trigger the mitochondrial pathway of apoptosis due to the dissipation of the mitochondrial membrane potential and release of cytochrome c from mitochondria into the cytoplasm. The obtained results corroborate the earlier reported data on the cytotoxic effects of ILs, providing new insight into the detailed mechanisms of IL cytotoxicity. In addition, the first illustrative guide to be employed for designing ILs with targeted biological activity is compiled. As a possible link between the electrochemical behavior of ILs and their biological activity, the relationship between IL cytotoxicity and the electrophoretic mobility of IL cations is assessed.

Recently, the dynamic nature of the metal-NHC bond has been proposed and the key role of chemical evolution in changing the nature of catalytically active sites is now an emerging topic. A comparative analysis of the ketone α-arylation reaction with aryl halides, catalyzed by M/NHC complexes, was carried out in the present study and showed a fundamental difference in the behavior of the catalytic system for M = Ni and Pd. In situ evolution of Ni/NHC complexes with cleavage of the Ni-NHC bond leads to complete deactivation of catalytic systems, regardless of the nature of the aryl halide ArX (X = Cl, Br, I). However, upon Pd/NHC catalysis, the cleavage of the Pd-NHC bond causes deactivation only in the case of aryl chlorides. In the reactions of more active aryl iodides and aryl bromides, NHC-disconnected Pd species, formed as a result of the chemical transformation of Pd/NHC complexes, can provide effective catalysis in the arylation reaction under study. New catalytic systems based on Pd/NHC and Ni/NHC complexes generated in situ from stable imidazolium salts, IPr·HCl and IPr*OMe·HCl, and Pd(OAc)2 (0.1 mol%) or NiCl2Py2 (5 mol%) were developed for the selective α-arylation of methylaryl ketones (Pd-catalysis) and other ketones less prone to aldol-crotonic condensation (Ni-catalysis). The present study has shown that the different effects of the metal-NHC bond cleavage should be taken into account for the efficient choice and optimization of catalytic systems to carry out arylation reaction with various aryl halides.

We present chemical depth profiling studies on mineralogical inclusions embedded in amygdale calcium carbonate by our Laser Ablation Ionisation Mass Spectrometer designed for in situ space research. An IR femtosecond laser ablation is employed to generate ions that are recorded by a miniature time-of-flight mass spectrometer. The mass spectra were measured at several locations on the sample surface and yield chemical depth profiles along the depth length of about 30 mm. The presence of oxides and sulphides within inclusion material allows us to derive elemental abundance calibration factors (relative sensitivity coefficients, RSCs) for major and minor elements. These are obtained from the atomic intensity correlations performed on the depth profiling data. With the RSCs corrections the quantitative analysis of more complex mineralogical phases within the inclusion is conducted by correlating atomic abundance fractions in ternary diagrams, typically used in geology. The spatial resolution of the depth profiles was sufficient to study chemically distinct micrometre-sized objects, such as mineralogical grains and thin layers of minerals including micrometre-sized filamentous structures. The method presented here is well-suited for the quantitative chemical analyses of highly heterogeneous materials where the ablation condition can vary locally with the material composition making the application of standard reference materials less accurate. The presented method is developed to distinguish between abiotic and biological material while searching for micrometre-sized extinct or extent life forms on the surfaces of Solar System bodies.

C–H functionalization in the area of fine organic synthesis is dominated by noble metal catalysts, which represent the most expensive and least sustainable options. Sustainable C–H functionalization may involve Ni catalysts. However, Ni(0) complexes are unstable under regular conditions and are more difficult to obtain as compared to Pd(0) or Rh(I). In the present study, a facile method for Ni0/NHC-catalyzed C–H alkylation and alkenylation of heteroarenes with alkenes and internal alkynes is presented. This method relies on the in situ generation of Ni0/NHC complexes from air-tolerant bench-stable precursors, Ni(Cp)2, NHC·HCl salts and sodium formate. The optimized catalytic system demonstrates broad substrate scope and high selectivity (>60 products were obtained in up to 99% isolated yield). The approach represents a user-friendly alternative for air-sensitive and labile (NHC)Ni0 and Ni(COD)2 precatalysts or complexes. The intermediates involved in the catalytic system were investigated and possible decomposition routes were mapped with NMR and ESI-MS. Rational control over the catalyst decomposition pathways further strengthens the sustainability of the procedure.

Complexes of Pd(II) with NHC ligands can suffer facile decomposition in the presence of alkali metal hydroxides, alkoxydes and other strong oxygen-containing bases via the reductive elimination of the NHC and Pd-coordinated base anion, the so-called O–NHC coupling. O–NHC coupling can represent a serious problem for the stability of Pd/NHC catalytic systems in numerous practically important reactions conducted in the presence of bases. In the present study, a new approach to stabilizing the Pd–NHC bond against cleavage by strong bases was developed. The approach relies on the installation of an NH–acidic RNH substituent at position 3 of the triazole ring of the 1,2,4-triazol-5-ylidene ligand. A series of new Pd/NHCs containing RNH substituents (R = Ac, Ph, alkyl) in triazole NHC ligands were synthesized. These complexes undergo reversible deprotonation of the RNH group in strong alkaline media and demonstrate superior stability of the Pd–NHC bond, significantly higher than complexes of similar structure without the RNH group. DFT calculations revealed that the anionic Pd/NHC complex containing an N-deprotonated acetamido group (R = Ac) is more kinetically stable against O–NHC coupling and less prone to lose NHC via heterolytic dissociation of the Pd–NHC bond than the neutral complex. The new complexes with RNH-functionalized NHC ligands were tested as precatalysts in the Suzuki–Miyaura coupling of p-tolyl bromide with phenylboronic acid in the presence of KOH and revealed more than 2 times higher TONs than similar complexes without the RNH group or ligandless Pd system.

The mechanism of the C–N cross-coupling reaction, catalyzed by palladium complexes with N-heterocyclic carbene ligands (Pd/NHC), was evaluated in detail at the molecular and nanoscale levels. For the first time, the formation of a “cocktail”-type catalytic system was proven for the Buchwald–Hartwig reaction. The unique ability of the Pd/NHC system to generate several types of catalytic centers (Pd complexes, clusters and nanoparticles) and the involvement of complementary pathways (homogeneous and heterogeneous) were discovered to take place in a “one pot” manner directly in the reaction vessel. Access to various catalytic centers from a single and readily available Pd/NHC complex is the key to designing a universal catalytic system with adaptive tuning capability.

Although practical catalytic transformations involving aryl chlorides are difficult to implement, they are highly desirable since the starting compounds are inexpensive and readily available. Retarded oxidative addition of aryl chlorides to palladium catalyst as compared to aryl bromides and aryl iodides is typically taken for granted as an explanation for the overall inefficiency of the process. The comparative experimental study and analysis reported herein suggest that oxidative addition cannot be considered the sole reason of the observed low reactivity of aryl chlorides. Other factors were found to play an important role in influencing the reactivity of aryl halides. The present findings suggest that a substantial revision of catalyst design principles is necessary for successful transformations of aryl chlorides.

α-CF3-enamines can be prepared by the reaction of pyrrolidine with the corresponding haloalkenes. The prepared enamines react with 2-nitrobenzaldehydes to give ortho-nitro-substituted α,β-diaryl-CF3-enones highly stereoselectively in up to 88% yield. Subsequent reduction of the nitro-group by an Fe–AcOH system promotes intramolecular cyclization to afford 2-CF3-3-arylquinolines in up to 99% isolated yield. High synthetic utility of all synthetic steps of the sequence was shown. A one-pot procedure was developed to give the target trifluoromethylated quinolines directly from enamines or haloalkenes.

Dried leaves of Eucalyptus camaldulensis were finely grinded and fractionated by sieving into four granulometric classes ( 355 μm). The obtained powder fractions were used for essential oil (EO) extraction by hydrodistillation and their phytochemical profile and in vitro antioxidant activities were evaluated. The mother powder (unsieved powder) was used for comparison. Particle size exerted a significant influence (p < 0.05) on the phytochemical composition and in vitro antioxidant properties of the EOs. This study reports substantial improvement in the process for oxidising α-pinene, using environmentally friendly H2O2 at high atom economy (∼93%) and selectivity to α-pinene oxide (100%). The epoxidation of α-pinene with H2O2 was catalysed by tungsten-based polyoxometalates without any solvent. The variables in the screening parameters were temperatures (30–70 °C), oxidant amount (100–200 mol%), acid concentrations (0.02–0.09 M) and solvent types (i.e., 1,2-dichloroethane, toluene, p-cymene and acetonitrile). Screening the process parameters revealed that almost 100% selective epoxidation of α-pinene to α-pinene oxide was possible with negligible side product formation within a short reaction time (∼20 min), using process conditions of a 50 °C temperature in the absence of solvent and α-pinene/H2O2/catalyst molar ratio of 5 : 1 : 0.01. A kinetic investigation showed that the reaction was first-order for α-pinene and catalyst concentration, and a fractional order (∼0.5) for H2O2 concentration. The activation energy (Ea) for the epoxidation of α-pinene was ∼35 kJ mol−1. The advantages of the epoxidation reported here are that the reaction could be performed isothermally in an organic solvent-free environment to enhance the reaction rate, achieving nearly 100% selectivity to α-pinene oxide.

Introduction. Osteoarthritis (OA) is generally a progressive disease that affects synovial joints, resulting in abnormalities to articular cartilage subchondral bone, synovium, and adjacent soft tissues. Aim. The purpose of this work was to examine the histological changes in knee cartilage and bone following the administration of two different chondroitin sulfate products in two experimental OA models in rats. Material and methods. OA was induced in rats by either a single injection of mono-iodoacetate or four once-weekly injections of dexamethasone. 70 adult rats were included: 30 received mono-iodoacetate, 30 received dexamethasone and the 10 remaining controls received no injection. Samples of knee bone and cartilage were then analyzed histologically. Results. Animals with OA that received CS had significantly less inflammation, improved motor activity, and better analgesia compared with those that did not receive CS, with little difference between products. Histologically, both products reduced the signs of OA and resulted in the activation of regenerative processes of cartilage and bone and stimulation of proliferation and formation of amorphous material. Conclusion. These results substantiate the importance of using high-quality pharmaceutical-grade CS to ensure optimal efficacy and safety of the final product in patients with OA.
