Although fluorobis(phenylsulfonyl)methane (FBSM) and its cyclic analog 2-fluoro-1,3-benzodithiole-1,1,3,3-tetraoxide (FBDT) possess similar physicochemical properties, Shibata et al. found that FBSM failed to undergo nucleophilic monofluoromethylation of aldehydes regardless of the reaction conditions attempted (using various organic and inorganic bases). However, it was later discovered by Hu et al. that the nucleophilic monofluoromethylation could be accomplished by employing lithium hexamethyldisilazide (LiHMDS) as a base. Herein, we present an in-depth computational investigation into the intriguing effects of reagent structure and bases on the nucleophilic monofluoromethylation of aldehydes. The computations reveal the 1,4-diazabicyclo[2.2.2]octane (DABCO) catalyzed nucleophilic monofluoromethylation of benzaldehyde with acyclic FBSM is a thermodynamically unfavorable process mainly due to the destabilizing O•••O lone pair repulsions in FBSM product, whereas such repulsion could be largely avoided in FBDT product because of its constrained five-membered ring structure. Employing LiHMDS as a base cannot only facilitate the nucleophilic monofluoromethylation via Li–O interactions but also render the monofluoromethylation of benzaldehyde with FBSM thermodynamically favored.

Pregnane glycosides are exist in Asclepiadaceae, Apocynaceae, Malpighiaceae, Ranunculaceae and Zygophyllaceae. This is a class of substances with steroids as the mother nucleus and containing a glyosidic bond structure. Modern pharmacology has shown that these compounds are a class of compounds with different structures and multiple biological activities. It has a good development prospect in anti-cancer and anti-tumor directions. This article reviews the chemical structures and biological activities of 345 compounds from 1984 to 2019.

Hydrogen sulfide and biothiol molecules such as Cys and GSH acted important roles in many physiological processes. To simultaneously detect and distinguish them was quite necessary by a suitable fluorescent probe. A novel chemosensor 4-(4-(benzo[d]thiazol-2-yl)-2-methoxyphenoxy)-7-nitrobenzo[c][1,2,5]oxadiazole (BMNO) was designed to detect H2S/Cys/GSH using the combination of nitrobenzofurazan (NBD) and benzothiazole fluorophores linked by a facile ether bond. The probe BMNO was developed for simultaneous identification of H2S, Cys and GSH. Noticeably, the color changes (from colorless to light purple, light orange and light yellow) of probe BMNO solutions for sensing H2S, Cys and GSH could be observed by naked eyes, respectively. The probe BMNO exhibited high selectivity and sensitivity for H2S, Cys and GSH showing distinct optical signal with detection limit as low as 0.15 μM, 0.03 μM and 0.14 μM, respectively. The sensing mechanism was clarified by spectrum analysis and some controlled experiments. In addition, these outstanding properties of probe BMNO enabled its practical applications in detection H2S in beer, and in cell imaging for Cys and GSH as well.

The introduction of thianthrene as a linchpin has proven to be a versatile strategy for the C-H functionalization of aromatic compounds, featuring a broad scope and fast diversification. The synthesis of aryl thianthrenium salts has displayed an unusually high para regioselectivity, notably superior to those observed in halogenation or borylation reactions for various substrates. We report an experimental and computational study on the mechanism of aromatic C-H thianthrenation reactions, with an emphasis on the elucidation of the reactive species and the nature of the exquisite site selectivity. Mechanisms involving a direct attack of arene to the isolated O-trifluoracetylthianthrene S-oxide (TT+-TFA) or to the thianthrene dication (TT2+) via electron transfer under acidic conditions are identified. A reversible interconversion of the different Wheland-type intermediates before a subsequent, irreversible deprotonation is proposed to be responsible for the exceptional para selectivity of the reaction.

Acute pancreatitis (AP), an inflammatory disorder of the pancreas, is a complicated disease without specific drug therapy. (R)-4,6-dimethoxy-3-(4-methoxy phenyl)-2,3-dihydro-1H-indanone [(R)-TML104] is a synthesized analog of the natural product resveratrol sesquiterpenes (±) -isopaucifloral F. This study aimed to investigate the effect and underlying mechanism of (R)-TML104 on AP. The experimental AP model was induced by caerulein hyperstimulation in BALB/c mice. (R)-TML104 markedly attenuated caerulein-induced AP, as evidenced by decreased pancreatic edema, serum amylase levels, serum lipase levels, and pancreatic myeloperoxidase activity. In addition, (R)-TML104 significantly inhibited the expression of pancreatic chemokines C–C motif chemokine ligand 2 and macrophage inflammatory protein-2 and the infiltration of neutrophils and macrophages. Mechanistically, (R)-TML104 activated AMP-activated protein kinase and induced sirtuin 1 (SIRT1) expression. (R)-TML104 treatment markedly induced the SIRT1-signal transducer and activator of transcription 3 (STAT3) interaction and reduced acetylation of STAT3, thus inhibiting the inflammatory response mediated by the interleukin 6-STAT3 pathway. The effect of (R)-TML104 on SIRT1-STAT3 interaction was reversed by treatment with a SIRT1 inhibitor selisistat (EX527). Together, our findings indicate that (R)-TML104 alleviates experimental pancreatitis by reducing the infiltration of inflammatory cells through modulating SIRT1.

Synthetic chemistry is built around the formation of carbon–carbon bonds. However, the development of methods for selective carbon–carbon bond cleavage is a largely unmet challenge1–6. Such methods will have promising applications in synthesis, coal liquefaction, petroleum cracking, polymer degradation and biomass conversion. For example, aromatic rings are ubiquitous skeletal features in inert chemical feedstocks, but are inert to many reaction conditions owing to their aromaticity and low polarity. Over the past century, only a few methods under harsh conditions have achieved direct arene-ring modifications involving the cleavage of inert aromatic carbon–carbon bonds7,8, and arene-ring-cleavage reactions using stoichiometric transition-metal complexes or enzymes in bacteria are still limited9–11. Here we report a copper-catalysed selective arene-ring-opening reaction strategy. Our aerobic oxidative copper catalyst converts anilines, arylboronic acids, aryl azides, aryl halides, aryl triflates, aryl trimethylsiloxanes, aryl hydroxamic acids and aryl diazonium salts into alkenyl nitriles through selective carbon–carbon bond cleavage of arene rings. This chemistry was applied to the modification of polycyclic aromatics and the preparation of industrially important hexamethylenediamine and adipic acid derivatives. Several examples of the late-stage modification of complex molecules and fused ring compounds further support the potential broad utility of this methodology. Common aromatic rings, such as anilines, arylboronic acids and aryl halides, can be opened up and converted to alkenyl nitriles through carbon–carbon bond cleavage using a copper catalyst.

Despite N6-methyladenosine (m6A) is functionally important in various biological processes, its role and the underlying regulatory mechanism in the liver remain largely unexplored. In the present study, we showed that fat mass and obesity-associated protein (FTO, an m6A demethylase) was involved in mitochondrial function during hepatic ischemia–reperfusion injury (HIRI). We found that the expression of m6A demethylase FTO was decreased during HIRI. In contrast, the level of m6A methylated RNA was enhanced. Adeno-associated virus-mediated liver-specific overexpression of FTO (AAV8-TBG-FTO) ameliorated the HIRI, repressed the elevated level of m6A methylated RNA, and alleviated liver oxidative stress and mitochondrial fragmentation in vivo and in vitro. Moreover, dynamin-related protein 1 (Drp1) was a downstream target of FTO in the progression of HIRI. FTO contributed to the hepatic protective effect via demethylating the mRNA of Drp1 and impairing the Drp1-mediated mitochondrial fragmentation. Collectively, our findings demonstrated the functional importance of FTO-dependent hepatic m6A methylation during HIRI and provided valuable insights into the therapeutic mechanisms of FTO.

Temozolomide (TMZ) is a chemotherapeutic agent that has been the first-line standard of care for the aggressive brain cancer glioblastoma (GBM) since 2005. Though initially beneficial, TMZ- resistance is universal and second-line interventions are an unmet clinical need. Here we took advantage the mechanism of action of TMZ to target guanines (G) and investigated G-rich g- quadruplex (G4) and splice site changes that occur upon TMZ-resistance. We report TMZ-resistant GBM has guanine mutations that disrupt the G-rich DNA G4s and splice sites that lead to deregulated alternative splicing. These alterations create vulnerabilities, which are selectively targeted by either the G4 stabilizing drug TMPyP4 or a novel splicing kinase inhibitor of cdc2- like kinase. Finally, we show that the G4 and RNA-binding protein EWSR1 aggregates in the cytoplasm in TMZ-resistant GBM cells and patient samples. Together, our findings provide insight into targetable vulnerabilities of TMZ-resistant GBM and present cytoplasmic EWSR1 as a putative biomarker.TeaserTargeting temozolomide mutations in drug resistant glioma via g-quadruplex and splicing modulators with a putative biomarker.

Fluorine incorporation into organic molecules is expected to lower the pKa of neighboring functionality via its strong electron-withdrawing effect, and this strategy has been widely exploited in diverse disciplines. Herein, we report a striking anomalous α-fluorine substitution effect on the α-Csp3 -H acidity. We have experimentally measured the pKa values of a series of popular nucleophilic monofluoromethylating reagents α-fluoro(phenylsulfonyl)methane derivatives as well as their C-H analogues by Bordwell's overlapping indicator method in dimethyl sulfoxide solution. Contrary to expectations, we found that α-fluorine substituent does not generally enhance but rather weaken the α-Csp3 -H acidity of most (phenylsulfonyl)methane derivatives. DFT computations reproduce and provide insight into the anomalous α-fluorine effect. A correlation was identified between the C-H pKa of (phenylsulfonyl)methane derivatives and Mayr's nucleophilicity parameter (N) of the corresponding carbanions.

Pharmacological inhibition of vacuolar-type H+-ATPase (V-ATPase) by its specific inhibitor can abrogate tumor metastasis, prevent autophagy, and reduce cellular signaling responses. Bafilomycin A1, a member of macrolide antibiotics and an autophagy inhibitor, serves as a specific and potent V-ATPases inhibitor. Although there are many V-ATPase structures reported, the molecular basis of specific inhibitors on V-ATPase remains unknown. Here, we report the cryo-EM structure of bafilomycin A1 bound intact bovine V-ATPase at an overall resolution of 3.6-Å. The structure reveals six bafilomycin A1 molecules bound to the c-ring. One bafilomycin A1 molecule engages with two c subunits and disrupts the interactions between the c-ring and subunit a, thereby preventing proton translocation. Structural and sequence analyses demonstrate that the bafilomycin A1-binding residues are conserved in yeast and mammalian species and the 7’-hydroxyl group of bafilomycin A1 acts as a unique feature recognized by subunit c., Bafilomycin A1, a member of macrolide antibiotics and an autophagy inhibitor, serves as a specific and potent V-ATPases inhibitor. Here authors report the cryo-EM structure of bafilomycin A1-bound V-ATPase with six bafilomycin A1 molecules bound to the c-ring and reveal the molecular basis for Bafilomycin A1 inhibition of the V-ATPase.

Significance Adipocytes are the most abundant and perhaps most active components of the tumor microenvironment in obese individuals that potentiate breast tumorigenesis through secretory mechanisms. The modulation of adipocytes can be novel therapy targets for breast cancer. Here, we revealed a specific upregulation of adipocytic TAZ through the FFA/PPARγ axis in diet-induced adiposity. Adipocytic TAZ knockdown or deficiency in mice inhibits adipocyte-induced breast cancer proliferation and stemness through impaired expression and secretion of Resistin. Immunostaining in triple-negative breast cancer samples showed that higher adipocytic TAZ/Resistin expression associates with higher clinical stages and poorer survival, demonstrating promising therapeutic targets., Adipocytes have been implicated in breast tumor growth and stemness maintenance through secreted factors. However, the mechanisms by which these cytokines are regulated during diet-induced obesity and contribute to breast tumorigenesis remain largely unknown. Here we show that transcription cofactor TAZ in adipocytes is directly up-regulated by the free fatty acid/PPARγ axis upon dietary fat stimulation. TAZ knockdown alters the expression profile of a series of secreted proteins and attenuates the tumor-supporting function of adipocytes. Moreover, we identify Resistin, an adipose-derived hormone, as a functional downstream target of TAZ, which facilitates tumorigenesis, and its expression correlated with adipocyitc TAZ in triple-negative breast cancer samples. Further, Adiponectin-cre–mediated TAZ knockout in adipocytes mitigates breast tumor growth. Taken together, our findings highlight how diet-induced TAZ expression in adipocytes promotes tumorigenesis, suggesting promising cancer therapeutic targets.

Hypervalent iodine-promoted dearomatization of phenols has received intense attention. This mini-review summarizes recent computational mechanistic studies of phenolic dearomatizations promoted by hypervalent iodine(III) reagents or catalysts. The first part of this review describes mechanisms of racemic dearomatization of phenols, paying special attention to the associative and dissociative pathways. The second part focuses on mechanisms and selectivities of diastereo- or enantio-selective dearomatization of phenols.

In this work, to overcome the fouling phenomenon of hydrophobic polymer membranes, polyvinylidene fluoride (PVDF) was blended with hydrophilic polycarbonate (PC) to prepare ultrafiltration membranes via the nonsolvent-induced phase separation method. The effects of PC content on membrane morphology, pore size distribution, and surface porosity were characterized and investigated by FE-SEM and image analyzer software. Solubility parameters calculated by molecular dynamics (MD) simulation showed that PVDF and PC are compatible and the results were confirmed by differential scanning calorimetry and wide angle X-ray diffractometry. The long-term chemical stability against NaOH and mechanical property before and after the abrasion test of the prepared membranes were also characterized by dynamic thermomechanical analysis. It was found that the hydrophilicity, water flux, abrasion resistance, and antifouling properties as the performance criteria of polymeric membranes were improved because of the presence of PC, and the separation efficiency of PVDF/PC membranes is much higher than that of the pristine PVDF membrane. The exemplary water filtration performances of these polymer membranes are harnessed here in this work to purify raw water polluted by natural organic matters, addressing the key environmental issue of water contamination.

Over 17 and 160 types of chemical modifications have been identified in DNA and RNA, respectively. The interest in understanding the various biological functions of DNA and RNA modifications has lead to the cutting-edged fields of epigenomics and epitranscriptomics. Developing chemical and biological tools to detect specific modifications in the genome or transcriptome has greatly facilitated their study. Here, we review the recent technological advances in this rapidly evolving field. We focus on high-throughput detection methods and biological findings for these modifications, and discuss questions to be addressed as well. We also summarize third-generation sequencing methods, which enable long-read and single-molecule sequencing of DNA and RNA modification.

We developed a synbiotic yogurt using monk fruit extract as a sweetener and investigated the effects of feeding the yogurt to rats with type 2 diabetes induced by streptozotocin and a high-fat diet. The rats fed the synbiotic yogurt showed greater blood glucose regulation and a significant decrease in insulin resistance and glycosylated hemoglobin compared with rats fed yogurt sweetened with sucrose, and they showed a remarkable improvement in short-chain fatty acid levels and gut microbiota status. Liver and kidney damage was also ameliorated in the rats fed the synbiotic yogurt. Immunohistochemistry analysis showed that the synbiotic yogurt inhibited β-cell loss compared with the control yogurt. Consuming the synbiotic yogurt helped to restore the islets of Langerhans. Our results indicated that monk fruit extract may be a good alternative to sucrose for synbiotic yogurt products in people with type 2 diabetes to delay the progression of diabetes and associated complications.

N-doped graphene (NG)/TiO2composites were prepared by a two-step hydrothermal method using HF as the surface etchant and urea as the nitrogen source. The morphology, structure, and bonding conditions of the NG/TiO2composites were characterized by X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), X-ray photoelectron spectroscopy (XPS), Raman spectroscopy, UV-visible diffuse reflectance spectroscopy (UV-Vis DRS), and photoluminescence (PL) spectroscopy. The effects of NG on the lifetime of photogenerated electron-hole pairs, adsorption capacity, and photocatalytic activity of the composite photocatalysts were also investigated. The photocatalytic activity was evaluated under ultraviolet light and sunlight irradiation. The recovery testing was completed under ultraviolet light irradiation. The results show that TiO2was uniformly loaded on the NG surface by chemical bonding. The introduction of NG effectively inhibited the recombination of photogenerated electron-hole pairs and improved both the adsorption capacity and photocatalytic activity of the composites. The 7 wt % NG/TiO2showed the best adsorption capacity of methyl orange (MO). The best photocatalytic activity occurred for 5 wt % NG/TiO2composites, and after four recovery tests, the photocatalytic degradation of MO under 60 min ultraviolet light irradiation exceeded 90%.

We present long-read Tet-assisted pyridine borane sequencing (lrTAPS) for targeted base-resolution sequencing of DNA methylation and hydroxymethylation in regions up to 10 kb from nanogram-level input. Compatible with both Oxford Nanopore and PacBio Single-Molecule Real-Time (SMRT) sequencing, lrTAPS detects methylation with accuracy comparable to short-read Illumina sequencing but with long-range epigenetic phasing. We applied lrTAPS to sequence difficult-to-map regions in mouse embryonic stem cells and to identify distinct methylation events in the integrated hepatitis B virus genome. A very broad acidity scale (≈40 pK units) for about 400 N-heterocyclic carbene precursors (NHCPs) with various backbones and electronic features, including imidazolylidenes, 1,2,4-triazolylidenes, cyclic diaminocarbenes (CDACs), diamidocarbenes (DACs), thiazolylidenes, cyclic (alkyl)(amino)carbenes (CAACs) and mesoionic carbenes (MICs), was established in DMSO by a well examined computational method. Varying the backbone structure or flanking N-substituents can have different extent of acidifying effects, depending on both the nature and number of substituent(s). The Gibbs energies (ΔGr s) for the reactions between the corresponding NHCs and CO2 were also calculated. There is a good linear correlation between the pKa s of most NHCPs and ΔGr s, suggesting that a greater basicity of NHC leads to a more stable NHC-CO2 adduct. Interestingly, the nearby asymmetric environment has virtually no differential effect on the acidities of the chiral NHCP enantiomers, but has a pronounced effect on the ΔGr values.

CONSPECTUS: The concept of pericyclic reactions and the explanation of their specificity through orbital symmetries introduced a new way of understanding reactions and looking for new ones. One of the 1965 Woodward-Hoffmann communications described "the (as yet unobserved) symmetry-allowed 6 + 4 combination", the prediction of a new field of "higher-order" cycloadditions, involving more than six electrons. Later these authors predicted exo-stereoselectivity for the [6 + 4]-cycloaddition. Chemists rushed to test this prediction (for the most part successfully). For more than half a century, chemists have hunted for additional higher-order cycloadditions. The application of catalysis within organic chemistry allows the accomplishment of previously unattainable reactions, including higher-order cycloadditions.The many examples of [8 + 2], [6 + 4], and cycloadditions of even higher electron-counts discovered since the Woodward-Hoffmann rules were introduced illustrate the difficulty in predicting which of these transformations will occur when two highly unsaturated molecules react. Periselectivity has been a challenge, and the development of enantioselective variants has been elusive. While progress was made, the rise of organocatalysis in asymmetric synthesis has led to a surge of interest in stereoselective versions of higher-order cycloadditions. Through organocatalytic activation of conjugated cyclic polyenes and heteroaromatic compounds, asymmetric [8 + 2]-, [6 + 4]-, and [10 + 4]-cycloadditions have been realized by our groups. In this century, [6 + 4]-cycloadditions have been found also to occur in enzyme-catalyzed reactions for the biosynthesis of spinosyn A, heronamide, and streptoseomycin natural products. A whole new class of enzymes, the pericyclases that catalyze pericyclic reactions, has been discovered.A remarkable aspect of these recent developments is the cross-disciplinary research involved: from organic synthesis to computational studies integrated with experimental studies of reaction mechanisms, intermediates, and dynamics, to understanding mechanisms of enzyme catalysis and engineering of enzymes.This Account describes how our groups have been involved in the expansion of the higher-order cycloaddition frontiers. We describe both the history and recent progress in higher-order cycloadditions, and how these advances have been made by our collaborative experimental and computational studies. Progress in asymmetric organocatalysis, incorporating enantioselective higher-order cycloadditions in organic synthesis, and the stereoselective synthesis of important scaffolds will be highlighted. Experimental progress and computational modeling with density functional theory (DFT) has identified ambimodal cycloaddition pathways and led to the realization that multiple products of pericyclic reactions are linked by common transition states. Molecular dynamic simulations have provided fundamental understanding of factors controlling periselectivity and have led to discoveries of a group of enzymes, the pericyclases, which catalyze pericyclic reactions such as [6 + 4]-cycloadditions.

Pyrophosphate synthetase-1(PRS-1) is a crucial enzyme that catalyzes the synthesis of phosphoribosylpyrophosphate (PRPP) with substrate: adenosine triphosphate (ATP) and ribose-5-phophate(R5P) in the de novo pathways of purine and pyrimidine nucleotide synthesis. Mutation in PRPS1 can result in a series of diseases of purine metabolism, which includes PRS-1 superactivity. The common clinical phenotypes are hyperuricemia and hyperuricosuria. We identified a novel missense mutation in X-chromosomal gene PRPS1 in a young Chinese woman while her mother has heterogeneous genotype and phenotype. A 24-year-old Chinese female patient suffered hyperuricemia, gout, and recurrent hyperpyrexia for more than 6 years, and then was diagnosed with hyperandrogenism, insulin resistance (IR), and polycystic ovary syndrome (PCOS). A novel missense mutation, c.521(exon)G>T, p.(Gly174Val) was detected by next-generation sequencing (NGS) and confirmed by Sanger sequencing in the patient and her parents. Interestingly, her mother has the same heterozygous missense mutation but without uric acid overproduction which can be explained by the phenomenon of the skewed X-chromosome inactivation. The substituted amino acid Val for Gly174 is positioned in the pyrophosphate (PPi) binding loop, and this mutation impacts the binding rate of Mg2+-ATP complex to PRS-1, thus the assembling of homodimer is affected by changed Val174 leading to the instability of the allosteric site. Our report highlights the X-linked inheritance of gout in females caused by mutation in PRPS1 accompanied with severe metabolic disorders and recurrent hyperpyrexia.

The purpose of this study was to prepare a dioscin nanosuspension (Dio-NS) that has a better distance and high solubility for oral administration and to evaluate its hepatoprotective effects. Optimal primary manufacture parameters, including shear time, shear speed, emulation temperature, pressure, and cycles of homogenization, were determined by single-factor experiments. The concentrations of dioscin, SDS, and soybean lecithin were optimized using the central composite design-response surface method, and their effects on the mean particle size (MPS) and particle size distribution of Dio-NS were investigated. Characterization of the Dio-NS formulations included examinations of the surface morphology and physical status of dioscin in Dio-NS, the stability of Dio-NS at different temperatures, in vitro solubility, and liver protective effect in vivo. Under optimal conditions, Dio-NS had an MPS of 106.72 nm, polydispersity index of 0.221, and zeta potential of −34.27 mV. Furthermore, the proportion of dioscin in Dio-NS was approximately 21.26%. The observation of particles with a spherical shape and the disappearance of crystalline peaks indicated that the physical and chemical properties of Dio-NS were altered. Furthermore, we observed that the dissolution of Dio-NS was superior to that of a physical mixture and Dio-GZF. Moreover, Dio-NS was demonstrated to have a protective effect against CCl4-induced acute liver damage in mice that was equivalent to that of silymarin (a positive control drug) at the same dose. The good hepatoprotective effect of our Dio-NS preparation can provide a theoretical basis for investigating its absorption mechanisms in the body.

Astaxanthin (AST) is a fat-soluble and non-vitamin A source of carotenoid that can quench reactive oxygen species and it has strong antioxidant and anti-inflammatory abilities. Herein, we have used H2O2 to establish a model of oxidative damage to RAW 264.7 cells and cells treated with vitamin C as the positive control group. The changes in metabolome were examined using 1H NMR and the results demonstrated that H2O2 treatment and various metabolic pathways such as amino acid, glucose, and glycerolipid metabolism were downregulated, which in turn affected citric acid cycle and energy status. AST could reverse downregulation of some of these metabolic pathways to a certain extent, and reduce cellular oxidative stress and death. The AST group differed from the vitamin C group in regulating d-glutamine, d-glutamic acid, pyruvate, and glycerolipid metabolism. The experimental results help to further understand the antioxidant effects of AST.

Curcumin possesses medicinal properties that are beneficial in various diseases, such as heart disease, cancer, and type 2 diabetes mellitus (T2 DM). It has been proposed that pancreatic beta cell dysfunction in T2 DM is promoted by oxidative stress caused by NADPH oxidase over-activity. The aim of the present study was to evaluate the efficacy of curcumin as a protective agent against high glucose/palmitate (HP)-induced islet cell damage and in streptozotocin (STZ)-induced DM rats. INS-1 cells were exposed to HP with or without curcumin. Cell proliferation, islet cell morphological changes, reactive oxygen species production, superoxide dismutase and catalase activity, insulin levels, NADPH oxidase subunit expression, and the expression of apoptotic factors by INS-1 cells were observed. Our results show that curcumin can effectively inhibit the impairment of cell proliferation and activated oxidative stress, increase insulin levels, and reduce the high expression of NADPH oxidase subunits and apoptotic factors induced by HP in INS-1 cells. The STZ-induced DM rat model was also used to determine whether curcumin can protect islets in vivo. Our results show that curcumin significantly reduced pathological damage and increased insulin levels of islets in STZ-induced DM rats. Curcumin also successfully inhibited the high expression of NADPH oxidase subunits and apoptotic factors in STZ-induced DM rats. These results suggest that curcumin is able to attenuate HP-induced oxidative stress in islet cells and protect these cells from apoptosis by modulating the NADPH pathway. In view of its efficiency, curcumin has potential for translation applications in protecting islets from glucolipotoxicity.

Steam methane reforming (SMR) via thermal catalytic approach is one of the dominant sources of industrial hydrogen, however it proceeds with slow response and low specific productivity. Here we demonstrate a plasma catalytic SMR for distributed hydrogen production, for which warm plasma by gliding arc discharge initiates the reaction, followed by Ni-based catalyst in a heat-insulated reactor without extra heating. In terms of the plasma alone process, specific energy input (SEI), steam/CH4 ratio (S/C) and total inlet flow rate (Ft) contribute to the methane conversion. In parallel, SEI and S/C account for the decrease in C2Hx selectivity hence the increase in selectivity of CO and CO2, while with Ft all the selectivity is approximately constant. The reaction pathway represented by the selectivity can be influenced by SEI and S/C rather Ft. To utilize the heat and active species with the reaction in plasma zone, Ni/CeO2/Al2O3 catalyst bed is coupled. For the coupled process, the conversion approaches the thermodynamic equilibrium values, with the favorable dismissed C2Hx selectivity thus the complete selectivity to CO and CO2. The coupled process was maintained steady for six hours, and the methane conversion of 90% at total hydrogen (t-H2) production rate of 2.7 SLM is achieved under optimum conditions of SEI, S/C, Ft and gas hourly space velocity (GHSV) of 110 kJ/mol, 3, 3 SLM and 18,000 ml·g−1 h−1. Compared to 59% and 2.3 kW h/Nm3 of the plasma alone process, such a coupled process achieves the energy efficiency (of methane to t-H2) of 75% and the low energy cost of 1.5 kW h/Nm3. Consequently, our approach of plasma catalytic SMR features the merits of rapid response, compact system and high specific productivity, which can be anticipated for the emerging needs of distributed hydrogen generation.

Efficient hydrogen production from methanol steam reforming is carried out in a heat-insulated warm plasma-catalytic (WPC) reactor. Methanol steam reforming in warm plasma and warm plasma-catalytic cases is investigated. For warm plasma alone, methanol mainly converts via pyrolysis reaction and its conversion linearly increases with specific energy input (SEI). The arc channel temperature and electron density of warm plasma are respectively measured to be around 2500 K and 3 × 1014 cm−3, based on optical emission spectra. To take advantage of energy from plasma, Fe-Cu/γ-Al2O3 catalyst for methanol steam reforming and water gas shift is placed after the warm plasma. Compare with plasma alone, methanol conversion in WPC case is nearly double therefore the energy cost decreases from 1.71 kW h/Nm3 to 0.85 kW h/Nm3. Energy efficiency of 84% and H2 selectivity of 98% with methanol conversion of 94% are achieved in this warm plasma-catalytic reactor.

As a promising plasmonic photocatalyst under visible-light irradiation, Au/TiO2 nanocatalyst commonly suffers from risk of Au nanoparticles aggregation and relatively long activation period by using the conventional calcination activation method. We report here that a short-term O2 plasma treatment enables the Au/TiO2 catalyst to exhibit higher visible-light photocatalytic activity for formaldehyde (HCHO) oxidation compared with the calcination activated sample. X-ray photoelectron spectroscopy characterization suggests that Au/TiO2 catalyst activated by O2 plasma achieves metallic Au content of 72.4% and surface oxygen species content of 29.8%, which not only lead to rapid reduction of cationic Au in HCHO oxidation but also facilitate the preliminary oxidation of HCHO to dioxymethylene and formate on catalyst surface. UV–vis diffuse reflectance spectra, photoluminescence spectra and diffuse reflectance infrared Fourier transform spectra measurements indicate that O2 plasma activated Au/TiO2 catalyst presents an enhanced visible-light absorption and surface charge transfer efficiency in HCHO oxidation, which accelerates the formation of superoxide (O2−) and OH radicals via hot electron transfer mechanism under visible-light irradiation, remarkably promoting formate oxidation to carbonate, carbonate decomposition and thus HCHO oxidation.

The smut fungus Ustilago esculenta infects Zizania latifolia and induces stem expansion to form a unique vegetable named Jiaobai. Although previous studies have demonstrated that hormonal control is essential for triggering stem swelling, the role of hormones synthesized by Z. latifolia and U. esculenta and the underlying molecular mechanism are not yet clear. To study the mechanism that triggers swollen stem formation, we analyzed the gene expression pattern of both interacting organisms during the initial trigger of culm gall formation, at which time the infective hyphae also propagated extensively and penetrated host stem cells. Transcriptional analysis indicated that abundant genes involving fungal pathogenicity and plant resistance were reprogrammed to maintain the subtle balance between the parasite and host. In addition, the expression of genes involved in auxin biosynthesis of U. esculenta obviously decreased during stem swelling, while a large number of genes related to the synthesis, metabolism and signal transduction of hormones of the host plant were stimulated and showed specific expression patterns, particularly, the expression of ZlYUCCA9 (a flavin monooxygenase, the key enzyme in indole-3-acetic acid (IAA) biosynthesis pathway) increased significantly. Simultaneously, the content of IAA increased significantly, while the contents of cytokinin and gibberellin showed the opposite trend. We speculated that auxin produced by the host plant, rather than the fungus, triggers stem swelling. Furthermore, from the differently expressed genes, two candidate Cys2-His2 (C2H2) zinc finger proteins, GME3058_g and GME5963_g, were identified from U. esculenta, which may conduct fungus growth and infection at the initial stage of stem-gall formation.

Multimodal, genome-wide characterization of epigenetic and genetic information in circulating cell-free DNA (cfDNA) could enable more sensitive early cancer detection, but it is technologically challenging. Recently, we developed TET-assisted pyridine borane sequencing (TAPS), which is a mild, bisulfite-free method for base-resolution direct DNA methylation sequencing. Here, we optimized TAPS for cfDNA (cfTAPS) to provide high-quality and high-depth whole-genome cell-free methylomes. We applied cfTAPS to 85 cfDNA samples from patients with hepatocellular carcinoma (HCC) or pancreatic ductal adenocarcinoma (PDAC) and noncancer controls. From only 10 ng of cfDNA (1 to 3 ml of plasma), we generated the most comprehensive cfDNA methylome to date. We demonstrated that cfTAPS provides multimodal information about cfDNA characteristics, including DNA methylation, tissue of origin, and DNA fragmentation. Integrated analysis of these epigenetic and genetic features enables accurate identification of early HCC and PDAC.

A straightforward one-pot method for the synthesis of unreported pyrido-[2,1-a]isoindolones in excellent yield is described. Two novel isoindolones were synthesized and fully characterized. The alkyl substituents on the pyridine play an important role in the outcome of the reaction. The mechanism, investigated through DFT calculations, features an unprecendented intramolecular cyclization reaction involving a carboxylic acid activated by tosyl chloride and an electron-poor pyridinic nitrogen. This protocol completes the known strategies to obtain functionalized isoindolones.

Programmed cell death 1 (PD-1) blockade is considered contraindicated in liver transplant (LT) recipients due to potentially lethal consequences of graft rejection and loss. Though post-transplant PD-1 blockade had already been reported, pre-transplant use of PD-1 blockade has not been thoroughly investigated. This study explores the safety and efficacy of neoadjuvant PD-1 blockade in patients with hepatocellular carcinoma (HCC) after registration on the waiting list. Seven transplant recipients who underwent neoadjuvant PD-1 blockade combined with lenvatinib and subsequent LT were evaluated. The objective response rate (ORR) and disease control rate (DCR) was 71% and 85% according to the mRECIST criteria. Additionally, a literature review contained 29 patients were conducted to summarize the PD-1 blockade in LT for HCC. Twenty-two LT recipients used PD-1 inhibitors for recurrent HCC. 9.1% (2/22) and 4.5% (1/22) recipients achieved complete remission (CR) and partial remission (PR), respectively; 40.9% (9/22) recipients had progressive disease (PD). Allograft rejection occurred in 45% of patients. In total, seven patients from our center and three from the literature used pretransplant anti-PD-1 antibodies, eight patients (80%) had a PR, and the disease control rate was 100%. Biopsy-proven acute rejection (BPAR) incidence was 30% (3 in 10 patients), two patients died because of BPAR. This indicated that neoadjuvant PD-1-targeted immunotherapy plus tyrosine kinase inhibitors (TKI) exhibited promising efficacy with tolerable mortality in transplant recipients under close clinical monitoring.

The Diels–Alder reactions of tropolone and its conjugate base with N-methylmaleimide have been explored computationally and experimentally. Previous studies of the [4+2] cycloaddition under basic conditions show that both endo- and exo-products are obtained in similar, but variable amounts. Density functional theory (ωB97X-D) explorations of potential energy surfaces, and molecular dynamics trajectories show that the reaction involves an ambimodal transition state for the reaction of the ammonium tropolonate with N-methylmaleimide, and that similar amounts of endo- and exo-products are obtained. The thermal reaction, studied experimentally in detail here for the first time, is predicted to form the endo-adduct through an ambimodal transition state. The exo-adduct can be formed from the same transition state, but requires a hydrogen shift, that hinders this reaction dynamically. Longer reaction times give a small excess of the exo-product, which is thermodynamically more stable.

Background Parkinson's disease is a common neurodegenerative problem. Pramipexole (PPX) plays protective role in Parkinson's disease. Nevertheless, the mechanism of PPX in Parkinson's disease-like neuronal injury is largely uncertain. Methods 1-methyl-4-phenylpyridinium (MPP+)-stimulated neuronal cells and 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced mice were used as the model of Parkinson's disease. MPP+-induced neuronal injury was assessed via cell viability, lactic dehydrogenase (LDH) release and apoptosis. microRNA-96 (miR-96) and BCL2/adenovirus E1B 19 kDa interacting protein 3 (BNIP3) abundances were examined by quantitative reverse transcription polymerase chain reaction (qRT-PCR) or Western blotting. Mitophagy was tested by Western blotting and immunofluorescence staining. MPTP-induced neuronal injury in mice was investigated via behavioral tests and TUNEL. Results PPX alleviated MPP+-induced neuronal injury via increasing cell viability and decreasing LDH release and apoptosis. PPX reversed MPP+-induced miR-96 expression and inhibition of mitophagy. miR-96 overexpression or BNIP3 interference weakened the suppressive role of PPX in MPP+-induced neuronal injury. miR-96 targeted BNIP3 to inhibit PTEN-induced putative kinase 1 (PINK1)/Parkin signals-mediated mitophagy. miR-96 overexpression promoted MPP+-induced neuronal injury via decreasing BNIP3. PPX weakened MPTP-induced neuronal injury in mice via regulating miR-96/BNIP3-mediated mitophagy. Conclusion PPX mitigated neuronal injury in MPP+-induced cells and MPTP-induced mice by activating BNIP3-mediated mitophagy via directly decreasing miR-96.

Modern pharmacological studies have shown that Fructus Tribuli can improve sexual function and treat cardiovascular diseases. In this study, we focused on comparing the pharmacokinetics of crude Fructus Tribuli (CFT) and stir-fried Fructus Tribuli (SFT) to further clarify the changes in chemical composition in vivo. The quantitation of six analytes was performed in a triple quadrupole mass spectrometer using the multiple reaction monitoring mode. Separation was performed on a Halo® C18 column using 0.05% formic acid and 5 μmol/L sodium formate in water, and 0.05% formic acid and 5 μmol/L sodium formate in acetonitrile as the mobile phase. The selectivity, precision, accuracy, extraction recovery, matrix effect and stability of the method were fully validated. Compared with the crude group, the parameters Cmax and AUC0-t of terrestroside B and terrestrosin K increased significantly (P < 0.05), but the Cmax and AUC0-t of polianthoside D, terrestrinin D, tribuluside A and terrestrosin D were decreased, terrestrosin D being especially decreased (P < 0.05), after oral administration of SFT extract. These results showed that the developed method was suitable for pharmacokinetic analysis of the six steroid saponins of CFT and SFT in rat plasma, and can be used to facilitate future clinical studies.

Difluorocarbene has important applications in pharmaceuticals, agrochemicals and materials, but all these applications proceed using just a few types of reaction by taking advantage of its intrinsic electrophilicity. Here, we report a palladium-catalysed strategy that confers the formed palladium difluorocarbene (Pd=CF2) species with both nucleophilicity and electrophilicity by switching the valence state of the palladium centre (Pd(0) and Pd(II), respectively). Controllable catalytic difluorocarbene transfer occurs between readily available arylboronic acids and the difluorocarbene precursor diethyl bromodifluoromethylphosphonate (BrCF2PO(OEt)2). From just this simple fluorine source, difluorocarbene transfer enables access to four types of product: difluoromethylated and tetrafluoroethylated arenes and their corresponding fluoroalkylated ketones. The transfer can also be applied to the modification of pharmaceuticals and agrochemicals as well as the one-pot diversified synthesis of fluorinated compounds. Mechanistic and computational studies consistently reveal that competition between nucleophilic and electrophilic palladium difluorocarbene ([Pd]=CF2) is the key factor controlling the catalytic difluorocarbene transfer.

We report a general and efficient approach to construct achiral and chiral gem-difluoroallylic amines from a masked difluorodiazo reagent (PhSO2CF2CHN2) and readily available imines. This facile protocol takes advantage of the phenylsulfonyl and diazo moieties as efficient activating and directing groups to assist difluoroalkyl incorporation and facilitate the chemodivergent and stereoselective formation of gem-difluoroallylic amines.

A transition-metal-free direct trifluoromethylthiolation of the ipso-carbon of lithium aryl boronates with trifluoromethanesulfenate under mild conditions was described. In addition, late-stage site-selective C-H borylation/trifluoromethylation and C-Cl borylation/trifluoromethylthiolation of biologically active molecules was developed. Initial mechanistic study suggested that the Li+ cation plays a vital role by coordinating to the oxygen atom of an aryl boronate complex and the oxygen of the reagent, thus allowing the aryl group to directly attack the trifluoromethylthio group of the trifluoromethylthiolating reagent.

Hydrogen and fuel cell technologies are being deployed to increase energy reliability and environmental performance. On-board hydrogen production via thermocatalytic reforming of liquid fuels is not a new concept, but has not been realized previously due to mismatches in rates and sizes of the necessary reactors. Here we demonstrate an advanced on-board hydrogen production by warm plasma chain catalysis for fuel-cell electric vehicles (FCEVs) with high specific productivity and transient response. Methanol, air and steam reactants as two specific feeds are fed into the reactor without soot formation and catalyst sintering. Oxidative pyrolysis reaction (OPR) by plasma, steam reforming (SR), and pyrolysis at high temperature, and SR and water gas shift (WGS) at medium temperature over catalysts occur subsequently for the coupled process. The heat with OPR is efficiently utilized by the subsequent endothermic reactions. Hydrogen selectivity of 99% (59 vol%, 4.1 SLM) with methanol conversion of 92% is achieved. The CO concentration of 7 vol% approaches the thermodynamic equilibrium value of 6.65 vol%. A fascinating match of energy efficiency of 91% (of methanol to hydrogen) and the hydrogen atom utilization of 88% is stressed. It is attributed to the chain catalysis of (oxidative) pyrolysis and WGS in sequence (as well as SR), which is initiated by warm plasma. Such a coupled process can be scaled up and used for wide applications from FCEVs and hydrogen refueling stations.

The mesoporous carbon nanocube@mesoporous silica (mCNC@mSiO2) has a potential application as drug carriers because of its large surface area and low toxicity and biocompatibility. Hence, a novel double mesoporous material of mCNC@mSiO2 was prepared via a soft-template method. The mCNC@mSiO2 exhibited a uniform cubic morphology with large surface area of 545.9 m2 g–1 and average pore size of 7.94 nm. The drug capacity for mCNC@mSiO2 was 0.78 mg at concentration of 0.5 mg/mL for 24 h loading. Furthermore, after modified by poly(acrylic acid) (PAA), the mCNC@mSiO2@PAA drug capacity had changed from 0.78 to 1.009 mg at the same concentration as a cancer drug carrier due to electrostatic interaction between carboxyl and 5-fluorourail (5-Fu). The findings may show potential for MOF-derived carbon materials for bioscience applications.

In the present study, a process for selective removal of arsenic from Cu–As-containing filter cakes using Na2CO3 leach was examined. When decreasing the weight percentage of arsenic in Cu–As-containing filter cakes, environmental problems can be eliminated while valuable metals will be recycled. The effect of α(Na2CO3) (where α represents excess coefficient), temperature, L/S ratio and leaching time were investigated. the optimal leaching conditions were achieved as follows: α(Na2CO3) = 2.7, L/S ratio 6 : 1 (mL/g), temperature 75°C, leaching time 3 h, where the leaching efficiency were 96.47% for As, 0.41% for Cu with final As and Cu concentrations of 11.72 and 0.02 g/L respectively. Further analysis showed that the Cu content in the leach residue increased to 51.42% with CuS as the major Cu phase. Notably, only 0.92% of As remained in the leach residue. The leaching solution was treated by sodium persulfate oxidation and arsenic precipitation which eliminated As 99%, leaving 0.5 mg/L As in the solution. Therefore, the proposed process is a viable method for the effective separation of Cu and As in Cu–As-containing filter cakes. Broadly, the results of this paper provide a reference for the treatment of arsenic sulfide from nonferrous metallurgy cyclic leaching process.

A neutral nitrogen radical-mediation strategy, wherein the existing N-H moiety of substrates serves as a neutral nitrogen radical precursor to enable room-temperature intermolecular radical difunctionalization of styrenes under photoredox catalysis, is reported. The reaction shows high functional group tolerance and substrate scope with respect to both components, giving the corresponding products with generally good yields. Preliminary control experiments and DFT calculations are performed to explain the reaction mechanism.

Humification of Sphagnum occurs in peat bogs and locks up carbon as peat. Useful indicators and methods of humification can reflect the process of peat formation to some extent. Scanning electron microscopy (SEM), transmission electron microscopy (TEM), UV/Vis spectra and excitation-emission matrix (EEM) spectra coupled with parallel factor (PARAFAC) analysis were applied to characterize the humification of Sphagnum, peat, dissolved organic matter (DOM) and chlorophyll with depth. The TEM results showed that chlorophyllous cells were almost completely degraded under the brown layer (BL), while Sphagnum hyaline cells demonstrated little degradation, and their structure could be observed even in peat. EEM spectra coupled with PARAFAC analysis is a good way to study chlorophyll in peat, and chlorophyll can be a useful indicator in humus studies. The peat had a relatively high chlorophyll content, which was negatively correlated with fulvic-like components and positively correlated with easily biodegradable organic matter. The parameters of samples from different layers fell into four groups, i.e., fulvic-like components, humification, humic-like components and/or protein-like components, easily biodegradable organic matter, and a combination or transition of the fluorescent substance of Sphagnum and humic-like components. The collapsed and defoliated layer (CDL) may be the layer in which the process of humification begins, and the humification of the grass root layer (GRL) was similar to that of peat. The degree of humification gradually increased from peat at 0–5 cm (P0-5) to peat at 10–15 cm (P10-15) and had no regular trend with depth from peat at 15–20 cm (P15-20) to peat at 25–30 cm (P25-30).

Bisulfite sequencing has been the gold standard for mapping DNA modifications including 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC) for decades1–4. However, this harsh chemical treatment degrades the majority of the DNA and generates sequencing libraries with low complexity2,5,6. Here, we present a bisulfite-free and base-level-resolution sequencing method, TET-assisted pyridine borane sequencing (TAPS), for detection of 5mC and 5hmC. TAPS combines ten-eleven translocation (TET) oxidation of 5mC and 5hmC to 5-carboxylcytosine (5caC) with pyridine borane reduction of 5caC to dihydrouracil (DHU). Subsequent PCR converts DHU to thymine, enabling a C-to-T transition of 5mC and 5hmC. TAPS detects modifications directly with high sensitivity and specificity, without affecting unmodified cytosines. This method is nondestructive, preserving DNA fragments over 10 kilobases long. We applied TAPS to the whole-genome mapping of 5mC and 5hmC in mouse embryonic stem cells and show that, compared with bisulfite sequencing, TAPS results in higher mapping rates, more even coverage and lower sequencing costs, thus enabling higher quality, more comprehensive and cheaper methylome analyses. A new method using mild chemical reactions and enzymatic oxidation allows nondestructive sequencing of 5-methylcytosine and 5-hydroxymethylcytosine with base-level resolution.

A series of p-tert-butyldihomooxacalix[4]arenes bearing phenolic hydroxyls and thiourea moieties were prepared to investigate their anion binding behavior. These compounds showed F− binding according to 1H NMR titration experiments. Among them, dihomooxacalix[4]arenes bearing phenyl and benzyl thiourea (2a and 2b) showed multiple interactions with added F−, including N–H⋯F, O–H⋯F hydrogen bonds and “through-space” C–H⋯F− interactions between F− and the attached aromatic segment of thiourea. A conformational change of the dihomooxacalix[4]arene cavity was observed as well and proved by theoretical calculations. The dihomooxacalix[4]arene bearing methyl thiourea (2c) also showed the formation of N–H⋯F, O–H⋯F hydrogen bonds, but less conformational change of the dihomooxacalix[4]arene cavity according to 1H NMR titration results. The bis-dihomooxacalix[4]arene (3) showed clear N–H⋯F hydrogen bond formation upon addition of F− as characterized by 1H NMR spectroscopy.

We propose a novel Power to Synthesis Gas (P2SG) approach, composed of two high-efficiency and renewable electricity-driven units, i.e., plasma catalytic reforming (PCR) and water electrolysis (WE), to produce high-quality syngas from CH4, CO2 and H2O. As WE technology is already commercial, we mainly focus on the PCR unit, consisting of gliding arc plasma and Ni-based catalyst, for oxidative dry reforming of methane. An energy efficiency of 78.9% and energy cost of 1.0 kWh/Nm(3) at a CH4 conversion of 99% and a CO2 conversion of 79% are obtained. Considering an energy efficiency of 80% for WE, the P2SG system yields an overall energy efficiency of 79.3% and energy cost of 1.8 kWh/Nm(3). High-quality syngas is produced without the need for post-treatment units, featuring the ideal stoichiometric number of 2, with concentration of 94.6 vol%, and a desired CO2 fraction of 1.9 vol% for methanol synthesis. The PCR unit has the advantage of fast response to adapting to fluctuation of renewable electricity, avoiding local hot spots in the catalyst bed and coking, in contrast to conventional catalytic processes. Moreover, pure O-2 from the WE unit is directly utilized by the PCR unit for oxidative dry reforming of methane, and thus, no air separation unit, like in conventional processes, is required. This work demonstrates the viability of the P2SG approach for large-scale energy storage of renewable electricity via electricity-to-fuel conversion.

Two type of hollow graphene microspheres were investigated for the adsorption performance of gaseous formaldehyde. Hollow gently reduced graphene oxide microspheres (HgRGOS) were synthesized by using the polystyrene microspheres as sacrificial template and gently reducing graphene oxide with NH2NH2. Hollow amine modified graphene microspheres (DETA-HgRGOS) were synthesized by DETA-treatment following the before-mentioned steps. The materials were characterized by scanning electron microscopy, transmission electron microscopy, X-ray diffractometry, Fourier transform infrared and X-ray photoelectron spectroscopies. This was followed by formaldehyde adsorption tests. The results showed that HgRGOS and DETA-HgRGOS were characterized to have adsorption capacities of 24.54 and 17.18 mg/g. The amine-modified material can interacts with HCHO more easily than the material eith carboxyl and epoxy groups on the surface. The reaction mechanism was discussed according to FTIR and XPS analysis.
