One new lactone derivative helicascolide F (1), one new pyrrolidine derivative talaromydine (2), along with six known compounds (3-8) were isolated from the fungus Talaromyces assiutensis JTY2. The structure of the new compounds 1 and 2 was determined by 1D and 2D NMR as well as by HRESIMS. The inhibitory activity of all compounds against six phytopathogenic fungi and three cancer cell lines was evaluated.

Banana flowers are consumed as a vegetable and traditionally used for managing several health problems including joint pain, a symptom of bone loss. Osteoclasts are key effector cells responsible for bone loss. Some flavonoids in banana flowers, such as quercetin and quercitrin, have been shown to be able to inhibit osteoclastogenesis. Whether banana flowers can inhibit osteoclast formation is unknown. In this study, we prepared the ethyl acetate fraction (FFE-EA) of an ethanolic extract of fresh flowers of Musa nana. Using UPLC-MS/MS analyses, 76 polyphenols were identified in FFE-EA. In RANKL-stimulated RAW264.7 macrophages, FFE-EA inhibited osteoclastogenesis and osteoclastic bone resorption. Mechanistic studies revealed that FFE-EA suppressed NF-κB and MAPK pathways, and lowered mRNA levels of osteoclast formation/function-related genes. These findings suggest that flowers of M. nana could be a source for formulating functional food that benefits bone health.

IRAK-4 is a serine/threonine kinase that can bind to interleukin-1 receptor induced by interleukin-1. It plays a key role in the Toll-like receptor signaling pathway and is involved in innate and adaptive immune responses. In this study, piscine IRAK-4 significantly activated nuclear factor (NF)-κB signaling in grouper spleen cells. Grouper (Epinephelus coioides) IRAK-4 (EcIRAK-4) co-localized with EcMyD88 and did not impair EcMyD88-dependent NF-κB activation. Different doses of EcIRAK-4 caused different degrees of nuclear translocation of the transcription factor NF-κB p65 subunit, and it induced transcription of multiple pro-inflammatory cytokines. Using expression vectors of deletion domains or mutations at important sites of EcIRAK-4, we found that the EcIRAK-4 kinase domain is necessary for its signal transduction function. The conserved amino acid sites performed functions similar to those in mammals, and grouper-specific amino acids such as E339 also played important roles. These findings provide information about the functional characteristics of IRAK-4 in lower vertebrates.

Two new naphthalene-chroman coupling derivatives, cladonaphchroms A (1) and B (2), along with four known compounds (3-6) were obtained from EtOAc extract of the mangrove-derived fungus Cladosporium sp. JJM22. Their structures were elucidated by detailed analysis of comprehensive spectroscopic data. Compounds 1 and 2 exhibit antibacterial or antifungal activities.

Two new phenanthrene derivatives, 2,3,7-trihydroxy-5H-phenanthro[4,5-bcd] pyran (1) and 2,3-dihydroxy-7-methoxy-5H-phenanthro[4,5-bcd] pyran (2) together with four known phenanthrenes derivatives (3–6) were isolated from the aerial parts of Pholidota chinensis Lindl. The structure of the new compounds 1 and 2 was determined by 1D and 2D NMR, as well as HRESIMS. The inhibitory activities of all compounds against four cancer cell lines were evaluated. Compounds 1 and 3 showed the highest cytotoxicity against the MCF-7 cell lines (IC50 = 8.1 and 7.5 μM). The other compounds showed modest activities or were inactive.

We will study the spin-polarized transport through devices of single-ion magnets. A device consisted of the scanning tunneling microscopy (STM) Co tip, the single-ion magnet (Cp*) Ln (COT) (Cp* represents pentamethyl pentacadiene; Ln represents ErIII, DyIII, HoIII; COT stands for cyclooctatetraene), and the Au(111) substrate is proposed. We calculate the current curve for the parallel and anti-parallel configurations for the device of Er single-ion magnet, and find that the tunnel magnetoresistance (TMR) changes with bias − 34~24%, which indicates it is promising for the application in magnetic storage. After comparing the currents through devices of (Cp*) Ln (COT) single-ion magnets, we find that Er single-ion magnet has a better rectifying character than Dy and Ho single-ion magnet, but Ho single-ion magnet has an obvious negative differential conductance. By analysis of their transmission spectrums, these properties are well explained. The spin-polarized transport for the derivatives of Er single-ion magnets is also investigated; we find that they have better rectification and negative differential conductance features, and have potential application on multifunctional molecular devices.

IκB kinase (IKK) is the core regulator of the nuclear factor-κB (NF-κB) pathway, which is involved in cellular development and proliferation, as well as the inflammatory response. IKKα is an important subunit of the IKK complex. In this study, two IKKαs (EcIKKα-1 and -2) were characterized in E. coioides. Similar to IKKα of other species, EcIKKα-1 and -2 contained a kinase domain, a leucine zipper, a helix-loop-helix domain and a beta NF-κB essential modulator-binding domain. Sequence alignment indicated that EcIKKα-1 and -2 shared high degrees of sequence identity with IKKs from other species (about 63%-96%). EcIKKα-1 and -2 are widely expressed in all tissues, but have different expression profiles in normal groupers. Additionally, EcIKKα-1 and -2 responded rapidly to Cryptocaryon irritans infection at the local infection site (i.e., gill tissue), but there was no significant change in EcIKKα-2 expression. In GS cells, EcIKKα-1 was uniformly distributed in the cytoplasm, while EcIKKα-2 was observed uniformly both in the cytoplasm and nucleus. Both EcIKKα-1 and -2 were found to activate NF-κB, but the luciferase activity of EcIKKα-2 was twice that of EcIKKα-1. In addition, EcIKKα-1 and -2 can regulate the expression of immune-related cytokines (IL-1β, IL-6, IL-8, IL-12 [p35 subunit], and TNF-α). These findings should prove helpful to further elucidate the innate immunity function of IKKα in fish.

The middle Jurassic Zhiluo Formation in the Dongsheng is comprised of a big set of green sandstone/mudstone with most of uranium orebodies occurring in close proximity to its footwall. By synthesizing field observations, region analysis, data collected from previous coal and uranium borehole, a regional north-south geological profile across the entire orefield is conducted. Experiments on sandstone/mudstone including rock mineral identification, clastic micromorphology and element geochemistry were carried out. Information from the geological profile indicates that green sandstone/mudstone is widely present in a stable horizon with clear boundaries to the country rock. Microscopic observations and geochemical data on sandstone/mudstone exhibit similar mineral composition with almost identical slightly flat, minor Eu enriched, Ce depleted chondrite-normalized REE patterns. Furthermore, the green clay membrane of the clasts has a complex composition containing chlorite/smectite, green smectite, chlorite, and green kaolinite, with elements including Fe, Mg, Si, and Al. These above results indicate that the green sandstone/mudstone underwent resemble sedimentary diagenetic processes as the country rock without transformation by large-scale regional fluid, while the existence of Fe2+-rich membrane is the main factor to the green sandstone/mudstone. Further concentration of the pre-enrichment uranium during diagenetic process led to the final formation for uranium deposits. The above studies are conducive to enrich the metallogenic mechanism of sandstone type uranium deposits and could provide certain reference for uranium exploration and deployment.

Microcystin-LR (MC-LR) is a hazardous hepatotoxin substance that threaten aquatic animals. Intestinal microbes can interact with and influence the health of their hosts. In this study, the shrimp Litopenaeus vannamei were exposed to 0.1 or 1 µg/L MC-LR for 72 h, and changes in intestinal microbial community and hepatopancreas transcriptome and metabolome were investigated. The results showed that both two doses of MC-LR increased intestinal microbial diversity and caused community composition variation by decreasing the abundances of putatively beneficial bacteria (Alloprevotella, Prevotella 1, Bacteroidales S24–7 group_norank and Lactobacillus). A large number of immune-related genes were differentially expressed in the hepatopancreas, including pattern recognition receptors and antimicrobial molecules; nutrient digestion and detoxification metabolism dysfunction were also induced. Furthermore, MC-LR altered hepatopancreas metabolic phenotypes, especially amino acid metabolism, the citric acid cycle, and linoleic acid and arachidonic acid metabolism. The changes in the intestinal bacteria were correlated with the immune factors and metabolic functions of their host. This study provides evidence for the toxicity of MC-LR on shrimp and identifies several biomarkers associated with the stress response.

Purpose: It is revealed that Xiaoyaosan could reduce glutamate level in the hippocampus of depressed rats, whose metabolism leads to the pathophysiology of depression. However, the underlying mechanism remains unclear. This study aims to explore the effect of Xiaoyaosan on glutamate metabolism, and how to regulate the excitatory injury caused by glutamate.Methods: Rats were induced by chronic unpredictable mild stress, then divided into control, vehicle (distilled water), Xiaoyaosan, fluoxetine, vehicle (DMSO), Xiaoyaosan + Ly294002 and Ly294002 groups. Ly294002 was microinjected into the lateral ventricular catheterization at 5 mM. Xiaoyaosan (2.224 g/kg) and fluoxetine (2.0 mg/kg) were orally administered for three weeks. The open field test (OFT), forced swimming test (FST), and sucrose preference test (SPT) were used to assess depressive behavior. The glutamate and corticosterone (CORT) levels were detected by ELISA. Western blot, immunochemistry or immunofluorescence were used to detect the expressions of NR2B, MAP2, PI3K and P-AKT/Akt in the hippocampal CA1 region. The mRNA level of MAP2, NR2B and PI3K were detected by RT-qPCR.Results: Compared to the rats in control group, body weight and food intake of CUMS rats was decreased. CUMS rats also showed depression-like behavior as well as down regulate the NR2B and PI3K/Akt signaling pathway. Xiaoyaosan treatments could increase food intake and body weight as well as improved time spent in the central area, total distance traveled in the OFT. Xiaoyaosan could also decrease the immobility time as well as increase the sucrose preference in SPT. Moreover, xiaoyaosan decreased the level of glutamate in the hippocampal CA1 region and serum CORT in CUMS rats. Furthermore, xiaoyaosan improved the expression of MAP2 as well as increased the expression of NR2B, PI3K and the P-AKT/AKT ratio in the hippocampal CA1 region in the CUMS rats.Conclusion: Xiaoyaosan treatment can exert the antidepressant effect by rescuing hippocampal neurons loss induced by the glutamate-mediated excitotoxicity in CUMS rats. The underlying pathway maybe through NR2B and PI3K/Akt signaling pathways. These results may suggest the potential of Xiaoyaosan in preventing the development of depression.

ObjectivesPatients with primary aldosteronism (PA) have cardiac remodeling due to hemodynamic and non-hemodynamic causes. However, component analysis of cardiac remodeling and reversal in PA patients is lacking. We investigated components of cardiac remodeling and reversal after adrenalectomy in patients with aldosterone-producing adenoma (APA).MethodsThis study prospectively enrolled 304 APA patients who received adrenalectomy and 271 with essential hypertension (EH). Clinical, biochemical and echocardiographic data were collected in both groups and 1 year after surgery in the APA patients. The hemodynamic and non-hemodynamic components of left ventricular (LV) remodeling were represented by predicted left ventricular mass index (LVMI) (pLVMI) and inappropriately excessive LVMI (ieLVMI, defined as LVMI-pLVMI).ResultsAfter propensity score matching, 213 APA and 213 EH patients were selected. APA patients had higher hemodynamic (pLVMI) and non-hemodynamic (ieLVMI) components of LV remodeling than EH patients. In multivariate analysis, baseline pLVMI was correlated with systolic blood pressure (SBP) and serum potassium, whereas ieLVMI was correlated with log plasma aldosterone concentration but not blood pressure. Post-operative echocardiography was available in 207 patents and showed significant decreases in both pLVMI and ieLVMI after adrenalectomy. In multivariate analysis, ΔpLVMI was correlated with SBP, ΔSBP, and pre-operative pLVMI, whereas ΔieLVMI was correlated with Δlog aldosterone-to-renin ratio (ARR) and pre-operative ieLVMI.ConclusionsThis study concluded that extensive cardiac remodeling in APA patients occurs through hemodynamic and non-hemodynamic causes. Adrenalectomy can improve both hemodynamic and non-hemodynamic components of LV remodeling. Regressions of pLVMI and ieLVMI were correlated with decreases in blood pressure and ARR, respectively.

A new chromene derivative, alternapyran (1), along with three known compounds (2–4), was isolated from the Alternaria sp. ZG22 obtained from a Dasymaschalon rostratum collected from the Hainan. The structure of the new compound 1 was elucidated by NMR and mass spectroscopic data. Compounds 2 and 3 showed broad spectrum antibacterial activity against four terrestrial pathogenic bacteria.

Summary Reducing greenhouse–gas (GHG) emissions in oil and gas production could provide several benefits, including energy conservation, cost reduction, and economic returns. The direct–emissions measurements and reduction–potential evaluation are the prerequisites to achieve an effective reduction goal in GHG emissions. On the basis of the survey of production processes and related parameters, we identified and measured a series of GHG–emissions sources. The emissions sources were measured, including production processes and leakage–prone facilities such as dehydrators, boilers, heaters, associated gas–treatment plants, light–hydrocarbon–recovery units, storage tanks, and gas flaring. A series of leakage–detection/measurement instruments was applied as well, such as flow samplers, impeller flowmeter, gas detectors, and gas–flow probes. On the basis of the measured emissions data, we then used a simulation model to evaluate the specific forms, sources, and reduction potentials of the GHGs. The measured GHG emissions showed that evaporation and flashing losses from storage tanks were the largest source, accounting for 86% of the total methane (CH4) emissions and 42% of the total GHG emissions. The contribution of CH4 emissions from heaters and boilers during incomplete combustions was less than 1% of the total CH4 emissions and approximately 16% of the total GHG emissions. When controlling technology on storage–tank losses was applied, CH4 emissions could be reduced by 81.7% and the GHG emissions could be reduced by 39.9%. Furthermore, such controlling technologies also presented substantial economic benefits through the recovery of fuel gas. In this study, the recovery potentials of various GHG–emissions sources were analyzed. In addition, a preliminary cost/benefit analysis was performed per the emissions categories, reduction potentials, and the feasibility of reduction technologies. Finally, the probability of the application of such reduction technologies was evaluated.

Four mononuclear lanthanide complexes containing 4′-phenyl-2,2′:6′,2″-terpyridine (ptpy), [Ln(NO3)3(ptpy)(H2O)] (Ln = Eu (1), Gd (2), Tb (3), Dy (4)), were solvothermally synthesized and characterized via elemental analysis, infrared spectroscopy, thermogravimetric analysis, single-crystal X-ray diffraction, and powder X-ray diffraction. Hirshfeld surfaces and the solid-state luminescence properties of the complexes were investigated. The 3-D Hirshfeld surface and 2-D fingerprint plots show that the main interactions are the O⋯H/H⋯O intermolecular interactions in 1–4. Solid-state luminescence investigation reveals that GdIII complex 2 displays a ligand-centered emission and the EuIII, TbIII and DyIII complexes 1, 3 and 4 show the characteristic lanthanide-centered luminescence upon UV excitations. The EuIII and TbIII complexes exhibit red (CIE: 0.6549, 0.3447) and green (CIE: 0.3760, 0.5412) luminescence in the solid state with quantum yields of 16.8% and 0.8% and lifetimes of 0.545 and 0.043 ms, respectively. Density functional theory (DFT) calculations were conducted to unravel the HOMO–LUMO energy gaps of the structures of ptpy and complexes 1 and 3.

Pyrolysis of waste tire is attracting widespread interest recently because of the great potential in waste valorization treatment. In this study, the thermal decomposition behavior, pyrolysis mechanism and products distribution of waste bicycle tire were investigated by means of TG-FTIR and Py-GC/MS. According to the TG results, two main pyrolysis stages of waste bicycle tire were observed, which were considered as the decomposition of rubber (285–531 °C) and further pyrolysis of pyrolytic products (663–847 °C), respectively. The volatiles release characteristics and FTIR spectrum at different temperatures exhibited good consistency with pyrolysis behavior. A detail information of pyrolysis products was analyzed by Py-GC/MS, which mainly include gaseous, alkenes and aromatics. The pyrolysis mechanism of waste bicycle tire was belong to free radical reaction, and the possible further pyrolysis pathway of D-limonene and styrene was also presented. Moreover, the products distribution under different pyrolysis final temperatures and heating rates conditions were summarized. Thus, this study could enhance our understanding on more specific details for the pyrolysis process of waste tire to some extent.

Two new sesquiterpenes, litseachrandaevanes C and D (1 and 2), together with five known sesquiterpenes (3 − 7), were isolated from the stems of Fissistigma glaucescens (Hance) Merr. Their structures were elucidated using comprehensive spectroscopic methods. The inhibitory effect of all compounds on the proliferation of primary synovial cells was evaluated. Compound 3 showed inhibitory effect on the proliferation of synoviocytes, with an IC50 value of 12.5 μM.

Six dinuclear lanthanide(III) nitrato complexes [Ln(NO(3))(3)(H(2)O)](2)(μ-tppz) (where tppz = 2,3,5,6-tetra(2-pyridyl) pyrazine and Ln(III) = Nd (1), Sm (2), Eu (3), Gd (4), Tb (5), and Dy (6)) with bis-tridentate N-heterocyclic 2,3,5,6-tetra(2-pyridyl)pyrazine as bridging ligand have been solvothermally synthesized and characterized via elemental analysis, infrared spectroscopy, thermogravimetric analysis, single-crystal X-ray diffraction, and powder X-ray diffraction. The 3-D Hirshfeld surface and 2-D fingerprint plots show that the main interactions in 1–6 are the O⋯H/H⋯O intermolecular interactions with relative contributions of about 62%. Although the poor lanthanide(III)-centered luminescence properties clearly point to the efficiency of nonradiative quenching processes (presence of water molecules in the coordination sphere of the lanthanide(III) ions), the ligand tppz is better suited to sensitize the lanthanide(III)’s emissions of Eu(III) and Nd(III) than Sm(III), Tb(III), and Dy(III). Finally, the magnetic data of Dy(III) comple×6 reveals antiferromagnetic coupling between Dy(III) ions.

Elevated serum aldosterone promotes arterial hypertension, cardiac hypertrophy, and diastolic dysfunction. However, the effect of elevated aldosterone levels on cardiac mitochondria remains unclear. We used primary cultures of mouse cardiomyocytes to determine whether aldosterone has direct effects on cardiomyocyte mitochondria, and aldosterone-infused mice as a preclinical model to evaluate the impact of aldosterone in vivo. We show that aldosterone suppressed mtDNA copy number and SOD2 expression via the mineralocorticoid receptor (MR)-dependent regulation of NADPH oxidase 2 (NOX2) and generation of reactive oxygen species (ROS) in primary mouse cardiomyocytes. Aldosterone suppressed cardiac mitochondria adenosine triphosphate production, which was rescued by N-acetylcysteine. Aldosterone infusion for 4 weeks in mice suppressed the number of cardiac mitochondria, mtDNA copy number, and SOD2 protein expression. MR blockade by eplerenone or the administration of N-acetylcysteine prevented aldosterone-induced cardiac mitochondrial damage in vivo. Similarly, patients with primary aldosteronism had a lower plasma leukocyte mtDNA copy number. Plasma leukocyte mtDNA copy number was positively correlated with 24-hour urinary aldosterone level and left ventricular mass index. In conclusion, aldosterone suppresses cardiac mitochondria in vivo and directly via MR activation of ROS pathways.

Background The long-term prognosis of Henoch-Schönlein purpura (HSP) depends on the severity of renal involvement, and crescent formation is considered an important risk factor for poor prognosis of Henoch-Schönlein purpura nephritis (HSPN). The objective of this study was to evaluate factors affecting crescent formation in children with HSPN. Methods Demographic factors, clinical characteristics, and laboratory data of children with HSPN with or without crescents were retrospectively analyzed. Univariate and multivariate logistic regression analyses were used to determine the risk factors of crescent formation in HSPN. Results A total of 191 children with HSPN were enrolled in the study. There were 107 (56%) males and 84 (44%) females, with a median age of 7 years (range: 2 years–15 years). International Study of Kidney Disease in Children (ISKDC) grading was used to divide subjects into two groups: those without glomerular crescent formation (ISKDC grades I–II, n = 146 cases) and those with glomerular crescent formation (ISKDC grades III–V, n = 45 cases). Logistic regression analysis showed that higher urinary white blood cell (WBC) count (OR = 3.300; 95% CI, 1.119–9.739; P = 0.0306) and higher urinary microalbumin/creatinine ratio (ACR) (OR = 25.053; 95% CI, 1.354–463.708; P = 0.0305) were independent risk factors for the formation of crescents in HSPN. The area under the receiver operating characteristic curve of urinary WBC and ACR were 0.753 and 0.698 respectively, with the Hosmer and Lemeshow goodness-of-fit test (P = 0.0669, P > 0.05). Conclusion These results suggest that higher urinary WBC count and ACR should be strictly monitored for children with HSPN. Adequate clinical intervention for these risk factors may limit or prevent renal crescent formation.

Many experiments show that vitrification significantly reduces the fertilization capacity of mammalian oocytes, restricting the application of vitrified oocytes. It has been proven that the JUNO protein plays a vital role in mammalian oocytes fertilization. However, little information is available about the effects of vitrification on the JUNO protein and the procedure to protect it in bovine oocytes. Here, the present study was designed to investigate the effect of vitrification on the JUNO protein level in bovine oocytes. In this study, MII oocytes were treated with cholesterol-loaded methyl-β-cyclodextrin (CLC; 0, 10, 15, 20 mM) for 45 min before vitrification and methyl-β-cyclodextrin (MβCD; 0, 2.25, 4.25, 6.25 mM) for 45 min after thawing (38-39°C). Then, the expression level and function of JUNO protein, cholesterol level in the membrane, the externalization of phosphatidylserine, sperm binding capacity and the developmental ability of vitrified bovine oocytes were examined. Our results showed that vitrification significantly decreased the JUNO protein level, cholesterol level, sperm binding capacity, development ability, and increased the promoter methylation level of the JUNO gene and apoptosis level of bovine oocytes. Furthermore, 15 mM CLC + 4.25 mM MβCD treatment significantly improved the cholesterol level and increased sperm binding and development ability of vitrified bovine oocytes. In conclusion, the combination treatment of cholesterol-loaded methyl-β-cyclodextrin and methyl-β-cyclodextrin significantly improves the fertilization capacity of vitrified bovine oocytes by protecting fertilization protein JUNO.

The harmful effects of microcystin-LR (MC-LR) stress are unavoidable in shrimp culture. Arachidonic acid (AA) is a fatty acid that regulates immune responses in aquatic animals. In this study, we investigated the effects of dietary AA on on growth, immunity and intestinal microbiota of Litopenaeus vannamei under microcystin-LR stress. The shrimp were fed a control diet (non-supplemented with AA) or a AA diet (supplemented with 8.0 g/kg AA) for 56 days, followed by an acute MC-LR stress for 72 h. The results showed that dietary AA improved the growth and feed utilization of the shrimp. Microcystin-LR exposure increased the mRNA expressions of reactive oxygen species modulator 1 (ROMO1), glutathione peroxidase (GPx), caspase-3 (Casp-3), NADPH-cytochrome P450 reductase (NCPR) and sulfotransferase (SULT), but decreased that of catalase (CAT) and cytochrome C (Cytc). Dietary AA supplementation reversed the expressions of ROMO1, CAT, GPx and NCPR to the control level, but still maintained the higher levels of Cytc, Casp-3 and SULT than the control and MC-LR stress groups. Dietary AA could not effectively reverse the changes of intestinal microbial diversity, but it could improve intestinal microbial composition variation induced by MC-LR stress. Specially, dietary AA increased the relative abundances of beneficial bacteria Bacteroides and Lactobacillus, and reversed the changes of pathogenic bacteria Vibrio and Photobacterium induced by MC-LR stress to the control level. The changes of intestinal bacteria were correlated with immune gene expression. These results revealed that dietary AA had a positive effect on L. vannamei resistance to MC-LR stress by modulating immune response and intestinal microbial composition.

The giant freshwater prawn, Macrobrachium rosenbergii (M. rosenbergii) as an important freshwater aquaculture species with high commercial value, exhibited unsynchronized growth. However, the potentially metabolic mechanism remains unclear. In this study, we used liquid chromatography tandem mass spectrometry (LC-MS/MS) to investigate the hepatopancreatic metabolic profiles of twenty giant freshwater prawns between the fast-growing group and slow-growing group. In the metabolomics assay, we isolated 8,293 peaks in positive and negative iron mode. Subsequently, 44 significantly differential metabolites were identified. Functional pathway analysis revealed that these metabolites were significantly enriched in three key metabolic pathways. Further integrated analysis indicated that glycerophospholipid metabolism and aminoacyl-tRNA biosynthesis have significant impact on growth performance in M.rosenbergii. Our findings presented here demonstrated the critical metabolites and metabolic pathways involved in growth performance, moreover provided strong evidence for elucidating the potentially metabolic mechanism of the unsynchronized growth in M. rosenbergii.

Background Disorders of the metabolism and absorption of vitamin B12 can lead to decrease in activity of methionine synthetase and methylmalonate coenzyme A mutase (MMUT), which results in increased levels of methylmalonic acid and homocysteine in blood and urine. Often, combined methylmalonic acidemia (MMA) and homocysteinemia is misdiagnosed due to a lack of specific symptoms. The clinical manifestations are diverse, but proteinuria as the initial presentation is rare. Case presentation Two cases of MMA with homocysteinemia in children are reported. Proteinuria were a primary presenting symptom, followed by anemia and neurologic symptoms (frequent convulsions and unstable walking, respectively). Screening of amino acids and acyl carnitine in serum showed that the propionyl carnitine:acetylcarnitine ratio increased. Profiling of urinary organic acids by gas chromatography–mass spectrometry revealed high levels of methylmalonic acid. Homocysteine content in blood was increased. Comprehensive genetic analyses of peripheral blood-derived DNA demonstrated heterozygous variants of methylmalonic aciduria type C and homocystinuria (MMACHC) and amnionless (AMN) genes in our two patients, respectively. After active treatment, the clinical manifestations in Case 1 were relieved and urinary protein ceased to be observed; Case 2 had persistent proteinuria and was lost to follow-up. Conclusions Analyses of the organic acids in blood and urine suggested MMA combined with homocysteinemia. In such diseases, reports of renal damage are uncommon and proteinuria as the initial presentation is rare. Molecular analysis indicated two different genetic causes. Although the pathologic mechanisms were related to vitamin B12, the severity and prognosis of renal lesions were different. Therefore, gene detection provides new insights into inherited metabolic diseases.

The fertilization capacity of sex-sorted sperms is seriously decreased, which inhibits its wide application. However, little information is still available about the effect of vitamin C (VC) and lycopene (Lyc) on the fertilization capacity of sex-sorted bull sperm. In this study, the washing medium and fertilization medium of sex-sorted sperm from three bull individuals were supplemented with different concentrations of VC (0, 1 × 10-3 , 1 × 10-4 , 1 × 10-5 , 1 × 10-6 M) or Lyc (0, 1 × 10-4 , 1 × 10-5 , 1 × 10-6 , 1 × 10-7 ). After washing twice and incubation for 1.5 hr, the malondialdehyde (MDA) level, phosphatidylserine (PS) translocation, membrane potential (Δψm) and IVF (in vitro fertilization) ability of sex-sorted sperm were investigated. For the sex-sorted sperm of bulls A, B and C, 1 × 10-3 M VC or 1 × 10-4 M Lyc treatment significantly decreased their MDA levels and PS translocation and increased their Δψm levels and cleavage rates after IVF. When blastocysts were concerned, 1 × 10-4 M Lyc significantly improved the blastocyst rates and their IFN-tau expression of bulls A and C. In conclusion, supplementation of 1 × 10-3 M VC or 1 × 10-4 M Lyc in washing and fertilization medium contributed greatly to improving the fertilization capacity of sex-sorted bull sperm during IVF procedure.

An epizootic outbroke in offshore cage-cultured Trachinotus ovatus, in Hainan provinces of China, causing massive mortality. Necropsy, histopathology and bacteriology experiments were carried out to identify the cause of this disease. The most prominent lesions happened in the brain, spleen and kidney, and a gram-positive bacterium was isolated from diseased fish. Through analysis of 16s rRNA and biochemical profiles, we identified this isolate as a strain of Streptococcus iniae, and named it HN strain. Artificial infection experiments were carried out, the HN strain can cause similar symptom with an LD50 value of 8.9 × 103 colony-forming units (CFU), which confirmed the isolated HN strain was virulent for T. ovatus. The HN strain is sensitive to all of the antimicrobial drugs tested, some of them, such as florfenicol and norfloxacin may be used to control the disease.

The aim of this study was to test the effects of five different concentrations (0, 10−3, 10−4, 10−5, and 10−6 M) of resveratrol (Res) supplementation in bull sperm washing and fertilisation medium on levels of reactive oxygen species (ROS), phosphatidylserine (PS) externalisation, mitochondrial membrane potential (Δψm), ATP and malondialdehyde (MDA), acrosomal integrity, blastocyst rate, and blastocyst quality after in vitro fertilisation (IVF). The results for sex-sorted sperm from three bulls showed: (1) ROS and MDA levels in 10−3 M and 10−4 M Res groups were significantly lower than those of controls (P −3 M and 10−4 M Res groups were significantly higher than those of controls (P −4 M Res group were significantly higher than those of controls (P −4 M Res supplementation in washing and fertilisation medium of sex-sorted bull sperm significantly decreased ROS, PS externalisation, and MDA, and protected mitochondrial function and acrosomal integrity, thereby increasing blastocyst percentage and quality following IVF.

Scuteformoids A-J, ten previously undescribed neo- clerodane diterpenoids along with one known analogue, were isolated from petroleum ether soluable fraction of the whole plants of Scutellaria formosana . The differences among these compounds are the substituents and stereochemistry at C-13. Their structures were elucidated by 1D and 2D NMR experiments, and the absolute configurations of Scuteformoids A, C, E, F, and I were further confirmed by single-crystal X-ray diffraction. Scuteformoids A, C, D, F, H, and I were evaluated for their inhibitory effects against HIV lytic replication and cytotoxic activities. All of them showed weak anti-HIV activities, with EC 50 values ranging from 48.24 to 79.17 μg/mL.

Two novel aporphine-derived alkaloids, aporaloids A and B (1 and 2), together with eight known biogenetically related alkaloids (3−10), two known isoquinoline alkaloids (3 and 4), and six known aporphinoid alkaloids (5–10) were isolated from the stems of Fissistigma glaucescens. Their structures were elucidated using comprehensive spectroscopic methods. Compounds 1 and 2 represent the rare example of a six-membered lactone ring aporphine-derived alkaloids from natural products. The inhibitory activities of all compounds against four cancer cell lines were evaluated. Aporaloids A and B (1 and 2) showed broad spectrum cytotoxic activities.

Four new alkaloids, nonialkaloids A–D (1–4) and six known analogues (5–10) were isolated from the noni juice. Among the new compounds, 1 and 2 are indole alkaloids with a seven-membered fused N-heterocyclic ring, 3 and 4 are quaternary ammonium derivatives. The structures were elucidated by extensive NMR and MS analysis, while the absolute configurations of the stereogenic carbons were established based on quantum-chemical electronic circular dichroism calculations or the modified Mosher's method. All the isolates were tested for α-glucosidase inhibitory activities. Compounds 1 and 3 displayed potent inhibitory activity against α-glucosidase with the IC50 values of 413.7 and 364.4 μM, respectively.

The phytochemical investigation of the stems of Fissistigma maclurei Merr. led to the isolation of a new eudesmane-type sesquiterpene 6α,12-dihydroxy-4(15)-eudesmen- 1-one (1), together with seven known eudesmane-type sesquiterpenes (2–8) and four known aporphine-type alkaloids (9–12). The structures of these compounds were elucidated using comprehensive spectroscopic methods. All compounds were isolated from the F. maclurei for the first time. The chemotaxonomic significance of the isolates was also discussed.

Baclofen was illegally used in veterinary clinical medicine as a growth-promoting agent. To date, few methods have been developed for the monitoring of baclofen in animal tissues. In this study, a sensitive and efficient liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to identify and quantify baclofen in the muscle, liver, kidney, and fat of swine was developed and validated. Baclofen was extracted from tissues with ammonium acetate buffer (pH 5.2) and isolated with isopropyl-ethyl acetate (4:6, v/v). Then, a solid phase extraction using MCX cartridge was used to clean up the extracts. The elution was evaporated to dryness and reconstituted with water/methanol (90:10 v/v). All samples were determined by LC-MS/MS system through positive ionization in a multiple reaction monitoring (MRM) mode. The proposed method was validated by evaluation of specificity, linearity, recovery, accuracy, precision, LOD, and LOQ values according to Commission Decision 2002/657/EC. Estimated limit of quantification for baclofen in the muscle, liver, kidney, and fat of this method was 1.00 μg/kg, respectively. The antioxidant of reduced glutathione (GSH) is the most abundant thiol in cells for the maintenance of the intracellular redox balance. The study aimed to assay the effect of sperm treatment with GSH before incubation with oocytes on the development potential of embryos obtained by in vitro fertilization (IVF). Also the mitochondrial membrane potential (ΔΦm), plasma membrane integrity (viability), DNA fragmentation, reactive oxygen species (ROS) content, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities, methane dicarboxylic aldehyde (MDA) level as indices of lipid peroxidation in sex-sorted and unsorted sperm from three bulls were investigated using flow cytometry and enzyme-labeled instrument individually. Our untargeted analysis of metabolomics throughout the course of pathogenic development gave us an unprecedented high-resolution view of major shifts in metabolism that occur in the topmost fungal pathogen that infects rice, wheat, barley, and millet. Guided by these metabolic insights, we demonstrated their practical application by using two different small-molecule inhibitors of sphingolipid biosynthesis enzymes to successfully block the pathogenicity of M. oryzae. Our study thus defines the sphingolipid biosynthesis pathway as a key step and potential target that can be exploited for the development of antifungal agents. Furthermore, future investigations that exploit such important metabolic intermediates will further deepen our basic understanding of the molecular mechanisms underlying the establishment of fungal blast disease in important cereal crops., The blast fungus initiates infection using a heavily melanized, dome-shaped infection structure known as the appressorium, which forcibly ruptures the cuticle to enter the rice leaf tissue. How this process takes place remains not fully understood. Here, we used untargeted metabolomics analyses to profile the metabolome of developing appressoria and identified significant changes in six key metabolic pathways, including early sphingolipid biosynthesis. Analyses employing small molecule inhibitors, gene disruption, or genetic and chemical complementation demonstrated that ceramide compounds of the sphingolipid biosynthesis pathway are essential for normal appressorial development controlled by mitosis. In addition, ceramide was found to act upstream from the protein kinase C-mediated cell wall integrity pathway during appressorium repolarization and pathogenicity in rice blast. Further discovery of the sphingolipid biosynthesis pathway revealed that glucosylceramide (GlcCer) synthesized by ceramide is the key substance affecting the pathogenicity of Magnaporthe oryzae. Our results provide new insights into the chemical moieties involved in the infection-related signaling networks, thereby revealing a potential target for the development of novel control agents against the major disease of rice and other cereals.

Pyricularia oryzae (synonym Magnaporthe oryzae) is a plant pathogen causing major yield losses in cultivated rice and wheat. The P-type ATPases play important roles in cellular processes of fungi, plants, and animals via transporting specific substrates through ATP hydrolysis. Here, we characterized the roles of a P5-ATPase, Spf1, in the development and virulence of P. oryzae. Deletion of SPF1 led to decreased hyphal growth and conidiation, delayed spore germination and appressorium formation, reduced penetration and invasive hyphal extension, and attenuated virulence. Appressorium turgor, however, was not affected by deletion of SPF1. The co-localization of Spf1-GFP and an endoplasmic reticulum (ER) marker protein, Lhs1-DsRed2, indicated that Spf1 is an ER-localized P5-ATPase. An ER stress factor, 0.5 μg/ml tunicamycin (TUNI), inhibited the growth of ∆spf1, but another ER stress factor, 5 mM dithiothreitol (DTT), promoted the growth of ∆spf1. Treatment with chemicals for oxidative stress (5 mM H2O2 and 0.8 mM paraquat) also promoted the growth of ∆spf1. Gene expression assays showed that unfolded protein response (UPR) components KAR2, OST1, PMT1, ERV29, PDI1, SCJ1, SEC61, a Ca2+ channel-related P-type ATPase gene PMR1, and a calcineurin-dependent transcription factor CRZ1 were significantly up-regulated in ∆spf1, suggesting activation of UPR in the mutant. These lines of experimental evidence indicate that SPF1 is involved in some basal ER mechanisms of P. oryzae including UPR pathway and responses to ER related stresses, therefore, affecting fungal development and virulence. However, the detailed mechanism between Spf1 and virulence still awaits future researches.

A novel cyclopentenone derivative, talarocyclopenta A (1), a new phenolicethers derivative, talarocyclopenta B (2) and a new itaconic acid derivative, talarocyclopenta C (3) together with four known itaconic acid derivatives (4-7) were isolated from the Talaromyces assiutensis JTY2. Their structures were elucidated by the detailed analysis of comprehensive spectroscopic data. Among them, talarocyclopent (1) is the first represent an unusual type of cyclopentenone derivative, possessing a cyclopentenone unit, a 2-butanone unit and a 3-hydroxybutyric acid unit. All isolated compounds were evaluated for their anti-inflammatory and antibacterial activities. Compounds 1-4 showed inhibitory activities against the nitric oxide (NO) production induced by lipopolysaccharide in mouse macrophage RAW 264.7 cells in vitro. Compound 2 showed broad spectrum antibacterial against six terrestrial pathogenic bacteria.

Objectives/Scope: Greenhouse gas emission (carbon dioxide and methane, etc.) during shale gas production could decide whether it is a cleaner energy source than coal. However, the methane and carbon dioxide emission from flowback process during shale gas production remains unclear. The withdrawn of fracture water both from underground and surface water, and the relatively high gas pressure from the wellhead have made flowback water highly potential as a significant greenhouse gas emission source. Methods, Procedures, Process: In this study, the estimation of greenhouse emission potential was provided based on the flowback water samples collected from shale production sites of China. The buffer capacity of carbon dioxide was estimated based on the parameters measured including temperature, pressure, conductivity, salinity, dissolved inorganic carbon (DIC), pH, chloride etc. The possible methane emission was also estimated based on the solution/dissolution curve from gas-water equilibrium during the flowback process. Results, Observations, Conclusions: The results showed that: the relatively high salinity of the flowback water has provided a considerable buffer capacity for carbon dioxide. The water residence time in the water tank/pond largely determines air-water greenhouse flux. When the flowback water pass through the three-phase separator to the water outlet, the dissolution capacity of methane reduced sharply dropped. By switching to a higher water level in the separator, methane emission could be reduced. However, a higher water vapor ratio may be detected in the gas phase, as well. Besides the fugitive methane emission from pneumatic pumps, valves, etc., greenhouse gas emission from flowback water is a significant source which needs to be reduced in future shale gas production. Novel/Additive Information: The determination of greenhouse gas emission from flowback could better defined the life cycle of shale gas, while provide baseline analysis for the development of greenhouse gas control technology during flowback water treatment and three-phase separator optimization.

Macrophages play an important role in aldosterone-induced myocardial fibrosis, in which the first key steps are macrophage recruitment and infiltration. We hypothesized that IL-6 may be a key mediator of aldosterone-induced macrophage recruitment and infiltration. To test this hypothesis, we designed cell studies with a human monocytic cell line THP-1 that with monocyte/macrophage functions to explore the signaling pathway of aldosterone-induced macrophage infiltration, and further investigated the phenomenon and consequent pathway in aldosterone-infused mice studies. The results showed that aldosterone induced the expression of IL-6 via mineralocorticoid receptors, and enhanced THP-1 cell migration and infiltration. Further experiments using a protease array and siRNA revealed that expressions of MMP-1 and MMP-9 were associated with aldosterone-induced macrophage infiltration. In addition, aldosterone-induced MMP-1 and MMP-9 expressions were mediated via cyclooxygenase-II and prostaglandin E2/EP-2 and EP-4 receptors. In aldosterone-infused mice, mRNA expressions of MMP-1, MMP-9 and COX-2 in peripheral blood monocytic cells were significantly increased. Moreover, the number of mouse macrophage-restricted F4/80 protein-positive cells in the myocardium was significantly higher in the aldosterone-infused mice compared with control mice. The increase in F4/80-positive cells in the myocardium was suppressed in the aldosterone-infused mice with the aldosterone antagonist eplerenone or anti-IL-6 antibody treatment. In conclusion, interleukin-6 played an important role in aldosterone-induced macrophage recruitment and infiltration in the myocardium.

Antimicrobial peptides (AMPs) are small proteins showing broad-spectrum antimicrobial activity that have been known to be powerful agents against a variety of pathogens (bacteria, fungi and viruses). In this study, the effects of AMPs from Bacillus subtilis on Epinephelus coioides were examined. E. coioides were fed with diets containing AMPs (0, 100, 200, 400 or 800 mg/kg) for four weeks. Results showed that the levels of total protein (TP), albumin (ALB), alanine aminotransferase (ALT), aspartate aminotransferase (AST), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and blood glucose (GLU) and lipopolysaccharide (LPS) in the serum of E. coioides changed than those of the control group; compared to the control group, the levels of total antioxidant capacity (T-AOC), superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA) and lysozyme (LZM) levels in E. coioides fed with different dosages AMP diets were also different; in addition, the mRNA expression of tumor necrosis factor alpha (TNF-α), interleukin-1-beta (IL-1β), and heat shock protein 90 (Hsp90) in the tissues of E. coioides were measured, the three genes in the tissues examined were significantly upregulated. The results demonstrated that diets containing AMPs can enhance the antioxidant capacity and innate immune ability of E. coioides, indicating that AMPs might be a potential alternative to antibiotics in E. coioides.

Four new cyclohexene derivatives cladoscyclitols A–D (1–4) and one new ribofuranose phenol derivative 4-O-α-D-ribofuranose-2-pentyl-3-phemethylol (5) were obtained from the EtOAC extract of the mangrove-derived endophytic fungus Cladosporium sp. JJM22. The structures were elucidated by extensive NMR and MS analysis, while the absolute configurations of the stereogenic carbons were established based on quantum-chemical electronic circular dichroism calculations or comparison of the optical rotations with those of related compounds. Compounds 2 and 5 displayed potent inhibitory activity against α-glucosidase with the IC50 values of 2.95 and 2.05 μM, respectively.

Studying pyrolysis behavior of saturate component is significant to understand the pyrolysis behaviors of coal, as the component can produce volatile matters. The combined utilization of TG-FTIR and Py-GC/MS provides a feasible strategy for investigating pyrolysis behaviors of saturate component. The results of TG-FTIR show that the pyrolysis of saturate component contains one stage. The yield of coal-char at different heating rates is quite low (0.04–0.50 wt%) and the values are close to each other, which indicates that the pyrolysis of saturate component has almost no coal-char and the saturate component mainly consists of volatile substances. Furthermore, the change in absorbance of typical compounds in FTIR indirectly reflects the variety in the content of compounds. The detailed information of pyrolysis products is analyzed by Py-GC/MS, which shows that alkanes and alkenes are dominant. Moreover, the change in the content of alkanes and alkenes is opposite. The pyrolysis mechanism of saturate component belongs to free radical reaction. The possible pyrolysis pathway of saturate component is presented and the appearance of hexane and hexene is also verified. This research provides a methodological model for exploring other group components and reveals more specific details for the pyrolysis of coal.

This study uses selected granite samples to evaluate the effect of redox conditions expressed as molar ferric-ferrous iron oxide ratio, Fe3+# [=Fe2O3/(FeO + Fe2O3)], on estimation of crystallization pressure (P) of granites using the Qtz-geobarometer. The results indicate that calculated P decreases significantly with increasing Fe3+# for calc-alkaline, metaluminous and peraluminous granites, but much less so for peralkaline and evolved granites. For a calc-alkaline, metaluminous or peraluminous granite sample of a fixed silica content, the calculated P values are a polynomial function of Fe3+#. The difference in P for Fe3+# between 0.1 (reduced) and 1.0 (oxidized) is as high as 437 MPa (or 4.37 kb), much higher than the uncertainty of the geobarometer. However, peralkaline and evolved granite samples display much less difference in P for Fe3+# between 0.1 and 1.0, essentially within the uncertainty. Therefore, redox conditions need to be considered when using Qtz-geobarometry to estimate P of calc-alkaline, metaluminous and peraluminous granites, particularly those bearing hornblende with relatively low silica contents, but such corrections may not be required for peralkaline and highly evolved granites. A practical procedure is proposed to calculate Fe3+# from whole-rock geochemical data to obtain improved P estimation, and a Microsoft Excel spreadsheet program is provided. In addition to constraining the emplacement depth, the program can also be used to evaluate the redox conditions using ferric-ferrous iron oxide ratios, both of which are important for mineralization associated with granite intrusions.

Background: Stroke is ranked on top of the persistent ailments associated with the brain. The incidence of ischemic stroke is rapidly rising along with intensified mortality as well as permanent disability conditions. However, tissue plasminogen activator (tissue-type plasminogen activator) is the only FDA-approved treatment for the treatment of stroke with decreased success rate. Eriocitrin, a flavonoid compound of Citrus limon with incredible therapeutic values, was active in the current investigation to explore the neuroprotective capability in arterial occlusion-provoked cerebral ischemic reperfusion injury (CIRI) in mouse replica. Materials and Methods: The neurological defects were considered through the standard scoring method and the expression status of apoptotic proteins, i.e., nuclear factor kappa B (NF-ƙB), caspase-3, and caspase-9, was examined via western blotting test. The expression of neuronal nitric oxide synthase (nNOS) was scrutinized through real-time polymerase chain reaction and standard ELISA kits were employed to examine the status of the proinflammatory markers. Results: The arterial occlusion provoked cerebral ischemic animals displayed the severely elevated status of inflammatory markers namely interleukin-6 (IL-6), IL-1β and tumor necrosis factor α, and the expression statuses of apoptotic proteins, as well as nNOS, were also uplifted while comparing to the sham subjected animals. Amazingly, eriocitrin (100 mg/kg) treatment revealed the declined inflammatory markers level, besides, the expression patterns of apoptotic proteins and nNOS were also reduced noticeably in the cerebral ischemic-reperfusion-operated animals. Primary aldosteronism has been associated with myocardial fibrosis, and is the most common cause of secondary hypertension. We previously showed that aldosterone can induce the secretion of galectin-3. The aim of this study was to investigate the association between myocardial fibrosis and plasma galectin-3 level in patients with primary aldosteronism. We prospectively analyzed 11 patients with aldosterone-producing adenoma (APA) who received adrenalectomy from December 2006 to October 2008, and 17 patients with essential hypertension as controls. Levels of plasma galectin-3 were determined in both groups, and both groups underwent echocardiography with cyclic variations of integrated backscatter (CVIBS) to characterize tissue initially and 1 year after surgery in the APA group. Diastolic blood pressure, concentration of plasma aldosterone and aldosterone-renin ratio were significantly higher, and serum potassium level and plasma renin activity significantly lower in the APA group compared to the controls. In addition, left ventricular mass index was significantly higher and CVIBS significantly lower in the APA group (7.3±2.0 vs 9.2±1.7 dB, p=0.015). Furthermore, the concentration of plasma galectin-3 was significantly higher in the APA group (2.1±0.9 vs 1.1±0.6 ng/mL, p=0.005) compared to the controls. CVIBS was correlated to plasma galectin-3 level. In the APA group, CVIBS increased significantly (7.3±2.0 to 9.2±2.4 dB, p=0.032) and plasma galectin-3 decreased (2.1±0.9 to 1.2±0.6, p=0.049) 1 year postadrenalectomy. The patients with APA had increased myocardial fibrosis, and this was associated with a higher plasma galectin-3 level. Both increased myocardial fibrosis and plasma galectin-3 level recovered at least partially after adrenalectomy. Trial registration number 200611031R; Results.

Selenophosphate synthetase (SPS) was initially detected in bacteria and was shown to synthesize selenophosphate, the active selenium donor. However, mammals have two SPS paralogues, which are designated SPS1 and SPS2. Although it is known that SPS2 catalyses the synthesis of selenophosphate, the function of SPS1 remains largely unclear. To examine the role of SPS1 in mammals, we generated a Sps1-knockout mouse and found that systemic SPS1 deficiency led to embryos that were clearly underdeveloped by embryonic day (E)8.5 and virtually resorbed by E14.5. The knockout of Sps1 in the liver preserved viability, but significantly affected the expression of a large number of mRNAs involved in cancer, embryonic development and the glutathione system. Particularly notable was the extreme deficiency of glutaredoxin 1 (GLRX1) and glutathione transferase Omega 1 (GSTO1). To assess these phenotypes at the cellular level, we targeted the removal of SPS1 in F9 cells, a mouse embryonal carcinoma (EC) cell line, which affected the glutathione system proteins and accordingly led to the accumulation of hydrogen peroxide in the cell. Furthermore, we found that several malignant characteristics of SPS1-deficient F9 cells were reversed, suggesting that SPS1 played a role in supporting and/or sustaining cancer. In addition, the overexpression of mouse or human GLRX1 led to a reversal of observed increases in reactive oxygen species (ROS) in the F9 SPS1/GLRX1-deficient cells and resulted in levels that were similar to those in F9 SPS1-sufficient cells. The results suggested that SPS1 is an essential mammalian enzyme with roles in regulating redox homoeostasis and controlling cell growth.

In this study, mild acid hydrolysis using γ-valerolactone (GVL)/water system integrated with enzymatic hydrolysis was carried out for the enhancement of enzymatic saccharification efficiency. The quantitative analysis of soluble carbohydrates and structural characterizations of solid residues were conducted. Results showed that the soluble carbohydrates in the water-phase were mainly composed of monomers and oligomers from xylose and glucose, while the contents of which were depended on the ratio of GVL to water. Moreover, the inhibitors were hardly detected due to the moderate pretreatment severity. Compared with the untreated feedstock, the yields of enzymatic hydrolysis from pretreated samples increased by two-fold with the mixture of 80/20 GVL/H2O. Combined with the amount of glucose (14.6%) dissolved in the water-phase, over 92.6% of glucose in cotton stalk was released and recovered. Based on the comprehensive analysis, treatment with GVL/H2O system provided us a more effective approach for sugar production from biomass.

Vitrification of oocytes has been shown to be closely associated with increased levels of reactive oxygen species (ROS) and apoptotic events. However, little information is available the effect of melatonin on the ROS levels and apoptotic events in vitrified oocytes. Therefore, we studied the effect of melatonin on ROS and apoptotic events in vitrified bovine oocytes by supplementing vitrification solution or in vitro maturation (IVM) and vitrification solution with 10(-9) m melatonin. We analyzed the ROS, mitochondrial Ca(2+) (mCa(2+) ) and membrane potential (ΔΨm), externalization of phosphatidylserine (PS), caspase-3 activation, DNA fragmentation, mRNA expression levels of Bax and Bcl2 l1, and developmental potential of vitrified bovine oocytes. Vitrified bovine oocytes exhibited increased levels of ROS, mCa(2+) , Bax mRNA, and caspase-3 protein and higher rates of PS externalization and DNA fragmentation, and decreased ΔΨm and Bcl2 l1 mRNA expression level. However, melatonin supplementation in vitrification solution or IVM and vitrification solution significantly decreased the levels of ROS, mCa(2+) , Bax mRNA expression, and caspase-3 protein, and PS externalization and DNA fragmentation rates, and increased the ΔΨm and Bcl2 l1 mRNA expression level in vitrified oocytes, resulting in an increased developmental ability of vitrified bovine oocytes after parthenogenetic activation. The developmental ability of vitrified oocytes with melatonin supplementation in IVM and vitrification solution was similar to that of fresh ones. This study showed that supplementing the IVM and vitrification medium or vitrification medium with 10(-9) m melatonin significantly decreased the ROS level and inhibited apoptotic events of vitrified bovine oocytes, consequently increasing their developmental potential.
