Expanding on previous demonstrations of the therapeutic effects of adeno‐associated virus (AAV) carrying small‐hairpin RNA (shRNA) in downregulating the mechanistic target of rapamycin (mTOR) in in vivo retinal vascular disorders, vascular endothelial growth factor (VEGF)‐stimulated endothelial cells were treated with AAV2‐shmTOR to examine the role of mTOR inhibition in retinal angiogenesis. AAV2‐shmTOR exposure significantly reduced mTOR expression in human umbilical vein endothelial cells (HUVECs) and decreased downstream signaling cascades of mTOR complex 1 (mTORC1) and mTORC2 under VEGF treatment. Moreover, the angiogenic potential of VEGF was significantly inhibited by AAV2‐shmTOR, which preserved endothelial integrity by maintaining tight junctions between HUVECs. These data thus support previous in vivo studies and provide evidence that AAV2‐shmTOR induces therapeutic effects by inhibiting the neovascularization of endothelial cells., Inhibition of mTOR expression by AAV2‐shmTOR suppresses the proliferation and migration of endothelial cells stimulated by VEGF. mTOR is a major activator in endothelial cells under VEGF stimulation, and inhibition by AAV2‐shmTOR blocked the activation of downstream mTOR signaling molecules. Inhibition of the mTOR pathway consequently affected cell proliferation and the remodeling of tight junctions between endothelial cells.

Background Methamphetamine (METH) is the most widely used psychostimulant and has been known to exhibit reinforcing effects even after long abstinence. We showed the inhibitory effect of Korean Red Ginseng extract (RGE) on METH-induced addictive behaviors in animal models mimicking the human drug-use pattern. Methods We first investigated the effect of RGE on the acquisition of METH-induced dependence using self-administration and conditioned place preference (CPP) tests. Additionally, further experiments such as METH-induced motivational behavior and seeking behavior were conducted. To study the underlying mechanism, dopamine receptor, dopamine transporter, and N-methyl-D-aspartate receptor were assessed through Western blot analysis. Results Treatment with RGE significantly reduced METH-induced self-administration on a fixed-ratio 1 schedule of reinforcement. It could be also decreased a progressive ratio schedule, and inhibited METH-primed reinstatement. In CPP, RGE significantly prevented the development of METH-induced CPP. Moreover, RGE not only shortened the withdrawal period clearly, but also prevented the reinstatement of CPP. RGE treatment also reversed METH-induced overexpression of dopamine transporter, dopamine receptor D1, and NMDA receptor in the nucleus accumbens. Conclusion Our findings reflect that RGE has therapeutic potential to suppress METH-induced addictive behaviors by regulating dopaminergic and NMDAergic system., Graphical abstract Image 1

Purpose Translocator protein 18-kDa (TSPO) positron emission tomography (PET) is a valuable tool to detect neuroinflammed areas in a broad spectrum of neurodegenerative diseases. However, the clinical application of second-generation TSPO ligands as biomarkers is limited because of the presence of human rs6971 polymorphism that affects their binding. Here, we describe the ability of a new TSPO ligand, [18F]BS224, to identify abnormal TSPO expression in neuroinflammation independent of the rs6971 polymorphism. Methods An in vitro competitive inhibition assay of BS224 was conducted with [3H]PK 11195 using membrane proteins isolated from 293FT cells expressing TSPO-wild type (WT) or TSPO-mutant A147T (Mut), corresponding to a high-affinity binder (HAB) and low-affinity binder (LAB), respectively. Molecular docking was performed to investigate the interaction of BS224 with the binding sites of rat TSPO-WT and TSPO-Mut. We synthesized a new 18F-labeled imidazopyridine acetamide ([18F]BS224) using boronic acid pinacol ester 6 or iodotoluene tosylate precursor 7, respectively, via aromatic 18F-fluorination. Dynamic PET scanning was performed up to 90 min after the injection of [18F]BS224 to healthy mice, and PET imaging data were obtained to estimate its absorbed doses in organs. To evaluate in vivo TSPO-specific uptake of [18F]BS224, lipopolysaccharide (LPS)-induced inflammatory and ischemic stroke rat models were used. Results BS224 exhibited a high affinity (Ki = 0.51 nM) and selectivity for TSPO. The ratio of IC50 values of BS224 for LAB to that for HAB indicated that the TSPO binding affinity of BS224 has low binding sensitivity to the rs6971 polymorphism and it was comparable to that of PK 11195, which is not sensitive to the polymorphism. Docking simulations showed that the binding mode of BS224 is not affected by the A147T mutation and consequently supported the observed in vitro selectivity of [18F]BS224 regardless of polymorphisms. With optimal radiochemical yield (39 ± 6.8%, decay-corrected) and purity (> 99%), [18F]BS224 provided a clear visible image of the inflammatory lesion with a high signal-to-background ratio in both animal models (BPND = 1.43 ± 0.17 and 1.57 ± 0.37 in the LPS-induced inflammatory and ischemic stroke rat models, respectively) without skull uptake. Conclusion Our results suggest that [18F]BS224 may be a promising TSPO ligand to gauge neuroinflammatory disease-related areas in a broad range of patients irrespective of the common rs6971 polymorphism.

Zeolites are 3D aluminosilicate materials having subnanometer pore channels. The Lewis basic pores have charge-balancing cations, easily tuned to metallic ions as more chemically active sites. Among the ion-exchanged zeolites, Cu2+ ion-exchanged ZSM-5 (Cu-ZSM-5) is one of the most active zeolites with chemical interactions of Lewis basic compounds. Even though the chemical interactions of hydrocarbons with Cu2+ sites in Cu-ZSM-5 have been tremendously studied in the category of zeolite catalysts, it is not yet thoroughly investigated how such interactions affect the structural lattice of the zeolite. Hydrocarbons with different chemical properties and their relative size can induce lattice strain by different chemical adsorption effects on the Cu2+ sites. In this work, we investigate the internal deformation of the Cu-ZSM-5 crystal using Bragg coherent X-ray diffraction imaging during the adsorption of four hydrocarbons depending on the alkyl chain length, the existence of a double bond in the molecule, linear structure versus benzene ring structure, and so forth. In the three-carbon system (propane and propene), relatively weak chemical adsorption occurred at room temperature and 100 °C, whereas strong adsorption was observed over 150 °C. For the six-carbon system (n-hexane and benzene), strong strains evolved in the crystal by active chemical adsorption from 150 °C. The observations suggest that propene and propane adsorb at the Cu2+ sites from the outer shell to the center with increasing temperature. In comparison, n-hexane and benzene adsorb at both parts at the same temperature. The results provide the internal structural information for the lattice with the chemical interactions of hydrocarbons in the Cu-ZSM-5 zeolite and help to understand zeolite-based chemisorption or catalysis research.

Precise control of the local electronic structure and properties of electrocatalysts is important for enhancing the multifunctionality and durability of electrocatalysts and for correlating the structure/chemistry with the catalytic properties. Herein, we report electronically coupled metallic hybrids of NiFe layered double hydroxide nanosheet/Ti3C2 MXene quantum dots deposited on a nitrogen‐doped graphene surface (LDH/MQD/NG) for high‐performance flexible Zn–air batteries (ZABs). As verified from the Mott–Schottky and Nyquist plots, as well as spectroscopic, electrochemical, and computational analyses, the electronic and chemical coupling of LDH/MQD/NG modulates the local electronic and surface structure of the active LDH to provide metallic conductivity and abundant active sites, leading to significantly improved bifunctional activity and electrocatalytic kinetics. The rechargeable ZABs with LDH/MQD/NG hybrids are superior to the previous LDH‐based ZABs, demonstrating a high power density (113.8 mW cm−2) and excellent cycle stability (150 h at 5 mA cm−2). Moreover, the corresponding quasi solid‐state ZABs are completely flexible and practical, affording a high power density of 57.6 mW cm−2 even in the bent state, and in real‐life operation of tandem cells for powering various electronic devices.

Bee venom is a medicinal product that is widely used in traditional therapies owing to its excellent anti-inflammatory activity. However, the use of bee venom has shown adverse effects. Therefore, there is a need for research that can remove the cytotoxicity of bee venom and enhance its efficacy. In this study, we hydrolyzed melittin, the main component of bee venom, and removed the other components to eliminate the toxicity of bee venom. To compare the efficacy of bee venom and detoxified bee venom, we examined their antioxidant effects using 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. In addition, cytotoxicity was confirmed in MCF 10A and RAW 264.7 cells, using 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt (MTS) assay. Detoxified bee venom showed a strong antioxidant activity and decreased a cytotoxicity in MCF 10A and RAW 264.7 cells. The anti-inflammatory activity of detoxified bee venom and bee venom were assessed by comparison of the expression of inflammatory cytokine mRNA and phosphorylation of IκBα in RAW 264.7 cells. Degranulation in RBL-2H3 cells was analyzed through β-hexosaminidase release assay to confirm the allergenic activity of bee venom and detoxified bee venom. Treatment of the detoxified bee venom inhibited inflammatory cytokine mRNA expression, IκBα phosphorylation, and β-hexosaminidase release. Taken together, the results indicated that compared to bee venom, detoxified bee venom exhibited decreased cytotoxicity and allergenicity and increased anti-inflammatory activity. In conclusion, detoxification of bee venom efficiently decreases the adverse effects, making it suitable for medicinal applications. Solid refuse fuels (SRF) and biomass contain significant amounts of KCl and NaCl which result in operation problems such as hard deposit formation in combustion. The effect of kaolinite in the decomposition of mineral chlorides in a commercial SRF-burning CFBC boiler is investigated. The injection of kaolinite with SRF in a commercial CFBC shows its effect on mineral chloride decomposition thus softens the hardness of the deposit on the heat exchanger tubes in the convection pass. By the reaction of molten salt with kaolinite, particles under 5 μm in the flue gas is significantly reduced. After 37 days operation with kaolinite use the PM 10 particle in flue gas is reduced originally 86 mg/Nm3 to 16 mg/Nm3, which is 81% reduction. The reaction of physical and chemical adsorption of metallic oxide component to kaolinite particle is also monitored by SEM images. This result is reflected to the particle side distribution of the fly ash in that the shift of the abundant particle size peak to a larger size cut after the use of the kaolinite.

Injectable hydrogels have been studied as drug delivery systems because of their minimal invasiveness and sustained drug release properties. Pluronic F127, consisting of poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) triblock copolymers, exhibits thermo-responsive properties and hence is injectable due to its rapid sol-gel transition. Unmodified Pluronic F127-based hydrogels, however, have limited long-term stability and controllable release of drugs entrapped within them. In this study, host-guest interactions between adamantane-conjugated Pluronic F127 (F127-Ad) and polymerized β-cyclodextrin (CDP) were employed to develop a hydrogel-based protein delivery system. Single or multiple adamantane units were successfully introduced at the termini of Pluronic F127 with a 100% conversion yield, and the synthesized F127-Ad polymer produced a physically crosslinked micelle-packing structure when mixed with CDP. As the number of adamantanes at the terminal ends of Pluronic F127 increased, the critical gelation concentration of F127-Ad/CDP hydrogel decreased from 15 to 6% (w/v). The F127/CDP hydrogel was able to maintain its structure even with lower polymer content, and its injectability improved with a reduction of the hydrogel viscosity. The long-term stability of F127/CDP hydrogels was evaluated in vitro and in vivo, and it was demonstrated that the subcutaneously injected hydrogel did not disintegrate for up to 30 d. Throughout the drug release test using gelatin and insulin as model drugs, it was demonstrated that their release rates could be regulated via complexation between the protein drugs and the β-cyclodextrin molecules inside the hydrogel. In conclusion, the F127-Ad/CDP hydrogel is expected to be a versatile protein delivery system with controllable durability and drug release characteristics. STATEMENT OF SIGNIFICANCE: Pluronic F127 is one of the widely studied polymeric materials for thermo-sensitive injectable hydrogels due to its high biocompatibility and rapid sol-gel transition. Since the Pluronic F127-based hydrogel has some limitations in its long-term stability and mechanical property, it is inevitable to modify its structure for the application to drug delivery. In this study, mono- or multi- adamantane-conjugated Pluronic F127s were synthesized and mixed with β-cyclodextrin polymers to form hydrogels with host-guest interaction-mediated micelle-packing structures. The host-guest interaction introduced into the hydrogel system endowed it a sustained protein drug release behavior as well as high durability in vitro and in vivo. By increasing the number of adamantane molecules at the end of the Pluronic F127, both the stability and injectability of the hydrogel could be also modulated.

This study investigates the effects of organic acid on the simultaneous absorption of SO2 and NO2 in the packed column. Organic acids, i.e., formic acid, acetic acid, and propionic acid, were used as additives. In the case of the absence of additives, when SO2 and NO2 were simultaneously absorbed into the limestone slurry, both SO2 and NO2 contributed to increasing the mutual absorption efficiency. In the simultaneous absorption of SO2 and NO2 with addition of additives, the SO2 removal efficiency appeared in the order of formic acid

High efficiency thermoelectric (TE) materials still require high thermopower for energy harvesting applications. A simple elemental metallic semiconductor, tellurium (Te), has been considered critical to realize highly efficient TE conversion due to having a large effective band valley degeneracy. This paper demonstrates a novel approach to directly probe the out-of-plane Seebeck coefficient for one-dimensional Te quantum wires (QWs) formed locally in the aluminum oxide layer by well-controlled electrical breakdown at 300 K. Surprisingly, the out-of-plane Seebeck coefficient for these Te QWs ≈ 0.8 mV/K at 300 K. This thermopower enhancement for Te QWs is due to Te intrinsic nested band structure and enhanced energy filtering at Te/AO interfaces. Theoretical calculations support the enhanced high Seebeck coefficient for elemental Te QWs in the oxide layer. The local-probed observation and detecting methodology used here offers a novel route to designing enhanced thermoelectric materials and devices in the future.

The aggregation and structural conversion of normal prion peptide (PrPC) into the pathogenic scrapie form (PrPSc), which can act as a seed to enhance prion amyloid fiber formation, is believed to be a crucial event in prionopathies. Previous research suggests that the prion monomer may play an important role in oligomer generation during disease pathogenesis. In the present study, extensive replica-exchange molecular dynamics (REMD) simulations were conducted to explore the conformational characteristics of the huPrP (125-160) monomer under the histidine tautomerism effect. Investigating the structural characteristics and fibrilization process is challenging because two histidine tautomers [Ne2-H (e) and Nδ1-H (δ)] can occur in the open neutral state. Molecular dynamics (MD) simulation outcomes have shown that the toxic eδ and δδ isomer (containing several and broader local minima) had the highest α-helix structures, with contents of 21.11% and 21.01%, respectively, and may have a strong influence on the organizational behavior of a monomeric prion. The amino acids aspartate 20 (D20)-asparagine 29 (N29) and isoleucine 15 (I15)-histidine 16 (H16), D20-arginine 27 (R27) as well as N29 formed α-helix with the highest probabilities in the δδ and eδ isomer, accordingly. On the basis of our findings, we propose the histidine tautomerization hypothesis as a new prion accumulation mechanism, which may exist to induce the formation of prion accumulates. Overall, our tautomerism hypothesis constitutes a promising perspective for enhancing understanding of prion disease pathobiology and may help in the design of a good inhibitor.

We demonstrate a flexible and stretchable supercapacitor assembled via straightforward interfacial gelation of reduced graphene oxide (rGO) with carbon nanotube (CNT) on a stretchable fabric surface. The difference between the redox potential of aqueous graphene oxide (GO) dispersion, prepared using a modified Hummers' method, and of a solid Zn plate, which was used as an external stimulus, induces a spontaneous reduction of GO flakes forming porous CNT–rGO hydrogel at the liquid–solid interface. With the aid of Zn, a macroporous and flexible CNT–rGO hydrogel was fabricated on a stretchable fabric platform using a facile fabrication method, and the CNT–rGO fabric composite was assembled into a supercapacitor to demonstrate its feasibility as a wearable electrode. The porous structure of the as-formed CNT–rGO fabric composite allows excellent electrolyte accessibility and ion transport that result in a fast charge/discharge rate up to 100 mV/s and a large areal capacity of 10.13 mF/cm2 at a discharge rate of 0.5 mA (0.1 mA/cm2). The inclusion of one-dimensional CNT as conductive bridges allows an excellent capacity retention of 95.2% after complete folding of the electrode and a capacity retention of 93.3% after 1000 bending cycles. Additional stretching test displayed a high capacity retention of 90.0% even at an applied strain as high as 50%, overcoming previous limitations of brittle graphene-based electrodes. This low-cost, lightweight, easy to synthesize, stretchable supercapacitor holds promise for next-generation wearable electronics and energy storage applications.

Early oligomerization of human islet amyloid polypeptide (hIAPP), which is accountable for β-cell death, has been implicated in the progression of type 2 diabetes mellitus. Some researches have shown the connection between hIAPP and Alzheimer's disease as well. However, the mechanism of peptide accumulation and associated cytotoxicity remains unclear. Due to the unique properties and significant role of histidine in protein sequences, here for the first time, the tautomeric effect of histidine at the early stages of amylin misfolding was investigated via molecular dynamics simulations. Considering Tau and Pi tautomeric forms of histidine (Tau and Pi tautomers are denoted as ϵ and δ, respectively), simulations were performed on two possible isomers of amylin. Our analysis revealed a higher probability of transient α-helix generation in the δ isomer in monomeric form. In dimeric forms, the δδ and δϵ conformations showed an elevated amount of α-helix and lower coil in comparison to the ϵϵ dimer. Due to the significant role of α-helix in membrane disruption and transition to β-sheet structure, these results may imply a noticeable contribution of the δ isomer and the δδ and δϵ dimers rather than ϵ and ϵϵ conformations in the early stages of diabetes initiation. Our results may aid in elucidating the hIAPP self-association process in the etiology of amyloidosis.

Transient receptor potential vanilloid member 1 (TRPV1) is a Ca2+-permeable cation channel that serves as the primary heat and capsaicin sensor in humans. Using cryo-EM, we have determined the structures of apo and capsaicin-bound full-length rat TRPV1 reconstituted into lipid nanodiscs over a range of temperatures. This has allowed us to visualize the noxious heat-induced opening of TRPV1 in the presence of capsaicin. Notably, noxious heat-dependent TRPV1 opening comprises stepwise conforma- tional transitions. Global conformational changes across multiple subdomains of TRPV1 are followed by the rearrangement of the outer pore, leading to gate opening. Solvent-accessible surface area analyses and functional studies suggest that a subset of residues form an interaction network that is directly involved in heat sensing. Our study provides a glimpse of the molecular principles underlying noxious physical and chemical stimuli sensing by TRPV1, which can be extended to other thermal sensing ion channels.

Herein, we identify the unique trifunctional active sites of ReS2 and NiFe layered double hydroxide (NiFe-LDH) heteronanosheets (ReS2/NiFe-LDH) for integrated cascade Zn–air battery/electrolyzer systems. Along with the edge and surface sites of NiFe-LDH for both oxygen evolution reaction and oxygen reduction reaction activities, the unprecedented activity of the ReS2/NiFe-LDH heteronanosheets for the hydrogen evolution reaction emerges from the S–O bonds at the heterointerfaces, together with the strong coupling effect and vertical alignment of NiFe-LDH and ReS2. The outstanding trifunctional activities and a well understood mechanism ensure the use of ReS2/NiFe-LDH heteronanosheets for the development of integrated cascade battery/electrolyzer systems, in which electricity storage in the battery mode and H2 production in the electrolyzer mode are efficiently switched with high round-trip efficiency (61%) and Faraday efficiency (96%). The systems show great promise for cost-effective energy storage and H2 production applications ranging from the distribution in households to the assembly for electrical vehicles.

Thin filament pyrometry is used to measure the time-varying temperature field in a 1-m methanol pool fire. A digital camera with optical filters and zoom lens recorded the emission intensity of an array of 12-µm silicon–carbide filaments oriented horizontally at various heights across the steadily burning pool fire. A 50-µm-diameter thermocouple measured the temperature at locations corresponding to the filament positions. A correlation was developed between the local probability density functions of the thermocouple time-series measurements corrected for radiation and thermal inertia effects and the camera grayscale pixel intensity of the filaments. A regression analysis yields the local mean temperature and its variance. The time series of the temperature field is transformed into average values during consecutive phases of the fire’s puffing cycle, providing quantitative insight into the complex and dynamic structure of a turbulent fire.

Achieving high ionic conductivity, wide voltage window, and good mechanical strength in a single material remains a key challenge for polymer-based electrolytes for use in solid-state supercapacitors (SCs). Herein, we report cross-linked composite gel polymer electrolytes (CGPEs) based on multi-cross-linkable H-shaped poly(ethylene oxide)–poly(propylene oxide) (PEO-PPO) tetrablock copolymer precursors, SiO2 nanoparticles, and 1-ethyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide, an ionic liquid (IL). Self-standing CGPE membranes with a high IL content were prepared using in situ cross-linking reactions between the silane groups present in the precursor and the SiO2 surface. The incorporation of an optimal amount of SiO2 increased the cross-linking density of the resulting CGPE while reducing polymer-chain ordering and, consequently, increasing both ionic conductivity and mechanical strength. As a result, the CGPE with 0.1 wt % SiO2 exhibited a high ionic conductivity (2.22 × 10–3 S cm–1 at 25 °C), good tensile strength (453 kPa), and high thermal stability up to 330 °C. Finally, an all-solid-state SC assembled with the prepared CGPE showed a high operating voltage (3 V), a large specific capacitance (103.9 F g–1 at 1 A g–1), and excellent durability (94% capacitance retention over 10,000 charge/discharge cycles), which highlights its strong potential as a solid-state electrolyte for SCs.

The high cost and relative scarcity of platinum (Pt) restrict large-scale commercialization of fuel cells, which has spurred researchers to develop low-cost alternatives integrating with high hydrogen evolution reaction (HER) and oxygen reduction reaction (ORR) catalytic activity. Herein, we performed density functional theory (DFT) calculations to explore the electrocatalytic activity of graphyne nanotubes (GyNTs). Several GyNTs were found to be potential metal-free electrocatalysts, with both HER and ORR activity superior to Pt. Moreover, we revealed a linear relationship between the Gibbs free energy change of O2 adsorption (ΔGOOH) and binding energy of H adsorption (ΔEH), which could be attributed to the fact that both the C O bond of OOH adsorption and the C H bond of H adsorption are single bonds. Therefore, ΔEH is proposed as a general descriptor for the rational design of bifunctional graphyne materials toward HERs and ORRs. Our findings provide a simple strategy for the rational design of bifunctional materials.

Purpose To investigate the long-term progression of pericentral hydroxychloroquine retinopathy. Design Multicenter, retrospective cohort study. Participants Eighty eyes (60 with pericentral pattern) of 41 Korean patients with hydroxychloroquine retinopathy followed up for 2 years or more after drug cessation. Methods Patients were screened for hydroxychloroquine retinopathy using spectral-domain or swept-source OCT, fundus autofluorescence (FAF), and Humphrey visual field (VF) tests. Follow-up was divided into short-term (≤2 years) and subsequent periods, and progression was evaluated in each period and severity group. Retinopathy progression on OCT was defined as increased length of the ellipsoid zone defect, decreased distance from the fovea to the photoreceptor defects, or newly developed or enlarged retinal pigment epithelium defects. On FAF, progression was defined as an increase in the area of hyperautofluorescence or hypoautofluorescence. Functional progression was defined as a regression coefficient of less than 0 dB/year for mean deviation and more than 0 dB/year for pattern standard deviation, based on linear regression analysis of 3 or more VF tests. Structural and functional progression rates were calculated using the slopes of retinal thicknesses on the Early Treatment Diabetic Retinopathy Study grid and perimetric parameters over time, respectively. Main Outcome Measures Structural and functional progression of retinopathy. Results Approximately one third of eyes with early pericentral retinopathy showed limited progression during the short-term period after drug cessation, but they subsequently showed stable or improved photoreceptors. Most eyes with moderate pericentral retinopathy showed continuous progression, particularly when converted to the severe stage. Severe eyes showed progressive damage throughout the follow-up period. In all severity groups, the rates of retinal thinning decreased over time. In eyes with pericentral retinopathy showing progression, circumferential enlargement of retinal damage was prominent in earlier stages, whereas centripetal enlargement of the ring-shaped lesion was noted in advanced stages. Functional progression, noted in 58.7% of the pericentral eyes, corresponded with structural progression. Conclusions Pericentral hydroxychloroquine retinopathy showed severity-dependent progression. Moderate pericentral retinopathy usually progressed, but centripetal progression threatening the fovea was remarkable mostly in severe retinopathy. Our results suggest that early detection of retinopathy may minimize the risk of progression to foveal involvement in pericentral retinopathy.

Self-assembly of hyperphosphorylated tau proteins into neurofibrillary tangles (NFT) is a hallmark of Alzheimer's disease. Previous studies suggest that the tau monomer may play an important role in NFTs formation in two general categories: inert (Mi) monomer and seed-competent (Ms) monomer. In the current study, replica-exchange molecular dynamics (REMD) were performed to investigate the effect of histidine tautomerism on the structures of a key fragment (R3) of tau protein and the transformation between different conformations. Based on the simulation results, we propose the histidine tautomerism hypothesis for tau protein misfolding. Histidine tautomerism greatly expands the conformational library, which triggers the emergence of conformations with higher aggregation tendency. Moreover, the conversions existing in both isomers and conformations may cause protein misfolding to occur more readily.

Gelatin/polyvinyl alcohol (PVA) (g/P) porous scaffolds with a glutaraldehyde/g ratio of 3 were synthesized by freeze/thaw and lyophilization to evaluate the effect of the g/P ratio (10/0 to 7/3) on the mechanical properties and cell growth behavior of the scaffolds. Fourier transform infrared spectroscopy results showed that the g/P scaffolds exhibited good crosslinking between gelatin and gelatin (imine), PVA and PVA (acetal group), and gelatin and PVA (imine). The pore size decreased gradually from 170±63 μm, to 140±60 μm, 129±64 μm, and 74 ±23 μm by varying the g/P ratio from 10/0, to 9/1, 8/2, and 7/3, respectively. As the g/P ratio was changed from 10/0 to 7/3, the compressive strength and swelling rate increased gradually from 150± 17 kPa to 290 ±28 kPa and 1268± 11% to 1640 ±35%, respectively. Among the scaffolds, 7/3 g/P scaffolds with tailored properties are suitable for wound healing.

In this research, various types of nitride additives were incorporated into titanium diboride attaining dense TiB2-based ceramics by field-assisted sintering technique. The addition of different types of nitride additives, namely Si3N4, BN, AlN, and TiN, significantly improved the sinterability of TiB2, achieving near fully dense ceramics. The X-ray diffraction analysis and microstructural evaluation confirmed the presence of the h-BN compound in all specimens. In the TiB2-Si3N4 ceramic, Si3N4 additive reacted with B2O3 oxide, in-situ generating h-BN, and SiO2 phases. Although the h-BN phase was produced in the TiB2-AlN specimen, the main proportion of AlN remained in the sample as an unreacted ex-situ phase. In terms of the TiB2-TiN ceramic, some of the nitrogen and boron atoms could leave the TiN and TiB2 crystalline structures, contributing to the in-situ formation of h-BN.

Retinal degenerative disorders, including age-related macular degeneration and retinitis pigmentosa (RP), are characterized by the irreversible loss of photoreceptor cells and retinal pigment epithelial (RPE) cells; however, the long-term effect of implanting both human induced pluripotent stem cell (hiPSC)-derived RPE and photoreceptor for retinal regeneration has not yet been investigated. In this study, we evaluated the long-term effects of hiPSC-derived RPE and photoreceptor cell transplantation in Pde6b knockout rats to study RP; cells were injected into the subretinal space of the right eyes of rats before the appearance of signs of retinal degeneration at 2–3 weeks of age. Ten months after transplantation, we evaluated the cells using fundus photography, optical coherence tomography, and histological evaluation, and no abnormal cell proliferation was observed. A relatively large number of transplanted cells persisted during the first 4 months; subsequently, the number of these cells decreased gradually. Notably, immunohistochemical analysis revealed that the hiPSC-derived retinal cells showed characteristics of both RPE cells and photoreceptors of human origin after transplantation. Functional analysis of vision by scotopic electroretinogram revealed significant preservation of vision after transplantation. Our study suggests that the transplantation of hiPSC-derived retinal cells, including RPE cells and photoreceptors, has a potential therapeutic effect against irreversible retinal degenerative diseases., Retinal disease: Cell transplants offer hope Cells with the potential to regenerate retinal cells could become a useful treatment for serious eye diseases such as age-related macular degeneration and retinitis pigmentosa. The retina has only limited self-regenerating potential. Joo Yong Lee and colleagues at the University of Ulsan in Seoul, South Korea, studied the long-term effects of implanting cells derived from human stem cells into the eyes of rats that had been genetically modified to act as a model of retinal degenerative disease. The implanted cells have the features of the photoreceptor cells that detect light and those cells that generate a layer of the retina called the retinal pigment epithelium. A substantial population of the cells persisted for at least four months, significantly preserving the animals’ vision. The results justify further exploration of the therapeutic possibilities.

Microporous organic polymer nanoparticles bearing tetraphenylethylene moieties (MOPTs) were prepared in the presence of poly(vinylpyrrolidone) (PVP). The PVP was entrapped into the microporous network of MOPT to form MOPT-P and played the roles of size control, porosity enhancement, and surface property management. MOPT materials without PVP showed superhydrophobicity with a water contact angle of 151°. In comparison, the MOPT-P showed excellent water compatibility. Moreover, due to the aggregation-induced emission property of tetraphenylethylene moieties, the MOPT-P showed emission and excellent emission-based sensing of nitrophenols in water with Ksv values in the range of 1.26 × 104 ∼ 3.37 × 104 M–1. It is noteworthy that the MOPT-P used water only as a sensing medium and did not require additional organic solvents to enhance water dispersibility of materials. The MOPT-P could be recovered and reused for the sensing at least five times.

Neuroinflammation—a common pathological feature of neurodegenerative disorders such as Alzheimer’s disease—is mediated by microglial activation. Thus, inhibiting microglial activation is vital for treating various neurological disorders. 7,3’,4’-Trihydroxyisoflavone (THIF)—a secondary metabolite of the soybean compound daidzein—possesses antioxidant and anticancer properties. However, the effects of 7,3’,4’-THIF on microglial activation have not been explored. In this study, antineuroinflammatory effects of 7,3’,4’-THIF in lipopolysaccharide (LPS)-stimulated BV2 microglial cells were examined. 7,3’,4’-THIF significantly suppressed the production of the proinflammatory mediators nitric oxide (NO), inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) as well as of the proinflammatory cytokine interleukin-6 (IL-6) in LPS-stimulated BV2 microglial cells. Moreover, 7,3’,4’-THIF markedly inhibited reactive oxygen species (ROS) generation. Western blotting revealed that 7,3’,4’-THIF diminished LPS-induced phosphorylation of extracellular signal-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), glycogen synthase kinase-3β (GSK-3β), and nuclear factor kappa B (NF-κB). Overall, 7,3’,4’-THIF exerts antineuroinflammatory effects against LPS-induced microglial activation by suppressing mitogen-activated protein kinase (MAPK) and NF-κB signaling, ultimately reducing proinflammatory responses. Therefore, these antineuroinflammatory effects of 7,3’,4’-THIF suggest its potential as a therapeutic agent for neurodegenerative disorders.

The development of efficient electrocatalysts for the CO2 reduction reaction (CO2 RR) remains a challenge. Demonstrated here is a NiSn atomic-pair electrocatalyst (NiSn-APC) on a hierarchical integrated electrode, which exhibits a synergistic effect in simultaneously promoting the activity and selectivity of the CO2 RR to formate. The NiSn atomic pair consists of adjacent Ni and Sn, each coordinated with four nitrogen atoms (N4 -Ni-Sn-N4 ). The as-prepared NiSn-APC displays exceptional activity for the CO2 RR to formate with a turnover frequency of 4752 h-1 , a formate productivity of 36.7 mol h-1 gSn -1 and an utilization degree of active sites (57.9 %), which are superior to previously reported single-atomic catalysts. Both experimental data and density-functional theory calculations verify the electron redistribution of Sn imposed by adjacent Ni, which reduces the energy barrier of the *OCHO intermediate and makes this potential-determining step thermodynamically spontaneous. This synergistic catalysis provides a successful paradigm for rational design and preparation of atomic-pair electrocatalysts with enhanced performance.

To investigate the effects of their surface recovery and optical properties, extremely small sized (12 µm × 12 µm mesa area) red AlGaInP micro light emitting diodes ($$\upmu$$ μ LED) were fabricated using a diluted hydrofluoric acid (HF) surface etch treatment. After the chemical treatment, the external quantum efficiencies (EQEs) of $$\upmu$$ μ -LED at low and high injection current regions have been improved by 35.48% and 12.86%, respectively. The different phenomena of EQEs have a complex relationship between the suppression of non-radiative recombination originating from the etching damage of the surface and the improvement of light extraction of the sidewalls. The constant enhancement of EQE at a high injection current it is attributed to the expansion of the active region’s sidewall surface area by the selective etching of AlInP layers. The improved EQE at a low injection current is related to the minimization of the surface recombination caused by plasma damage from the surface. High-resolution transmission electron microscopy (HR-TEM) revealed physical defects on the sidewall surface, such as plasma-induced lattice disorder and impurity contamination damage, were eliminated using chemical treatment. This study suggests that chemical surface treatment using diluted HF acid can be an effective method for enhancing the $$\upmu$$ μ -LED performance.

Thermochemical heat storage is a promising energy storage method and has received much attention in recent years. To improve the design and application of such systems, a better understanding of the characteristics of thermochemical heat storage is required. In this study, we analyze the heat release characteristics of thermochemical heat storage considering various operating parameters, such as charging temperature, humidity, and material. Three different thermochemical materials are considered herein, namely, zeolite 13X, zeolite 5A, and silica gel. The temperature profile at the inlet and outlet of the storage chamber, and that inside the chamber packed with thermochemical material, were measured. The pressure drop across the chamber was measured as well. The results indicate that the heat release characteristics change with position and time. Furthermore, the temporal curves of released heat are significantly affected by charging temperature and humidity. Zeolite 13X rapidly released a large amount of heat within a short period of time, whereas silica gel gradually released a small amount of heat, over a longer duration of time. The pressure drop across the chamber was found to vary with the bead size of the thermochemical material. Thus, the operating parameters including charging temperature, humidity, and material, have a significant effect on the characteristics of heat release. The data obtained herein can serve as a reference for the design of thermochemical storage systems.

The development of activatable photosensitizers to allow for the reversible control of singlet oxygen (1O2) production for photodynamic therapy (PDT) faces great challenges. Fortunately, the flourishing field of supramolecular biotechnology provides more effective strategies for activatable PDT systems. Here, we developed a new reversible PDT on a switch that controls the 1O2 generation of self-assembled albumin nanotheranostics in vitro and in vivo. A new molecular design principle of aggregation-induced self-quenching photochromism and albumin on-photoswitching was demonstrated using a new asymmetric, synthetic diarylethene moiety DIA. The photosensitizer porphyrin and DIA were incorporated as building blocks in a glutaraldehyde-induced covalent albumin cross-linking nanoplatform, HSA-DIA-porphyrin nanoparticles (NPs). More importantly, the excellent photoswitching property of DIA enables the resultant nanoplatform to act as a facile, switchable strategy for photodynamic-immunotherapy.

The durability of the membrane electrode assembly (MEA) is one of the important requirements for the successful commercialization of anion exchange membrane fuel cells (AEMFCs). While chemical stabilities of the components (the catalyst, membrane, and ionomer) have been assessed by ex situ tests, the degradation mode of the components in a single-cell is still not fully understood. Herein, the degradation of a MEA based on an ionomer having a polycarbazole main chain and a trimethyl ammonium side group was systematically investigated for a single-cell level under constant current conditions. The degradations of the catalyst, membrane, and ionomer were compared by in situ electrochemical and ex situ post-mortem analyses. The catalyst and ionomer in the cathode showed significant degradation, whereas the membrane was relatively durable, suggesting that the formation of a strong alkaline environment in the cathode catalyst layer is a major cause of the performance fade. The use of an alkaline-stable ionomer with an N,N-dimethylhexylamine group, which is designed based on density functional theory (DFT) calculations, in the cathode catalyst layer leads to 87% durability improvement. This work enhances the understanding of the key factors affecting the durability of AEMFCs under real operation and suggests practical strategies toward higher durability.

Sufficiently strong molecular magnets are used in small modern electronic and spintronic devices. Diradical organic magnetic molecules (OMMs) are promising options due to their lightness, flexibility, and the low energy required for their synthesis. In this field, the coupler is a dominating component, though acetylene, the shortest coupler, has not been studied for this use. In this study, we conceptualize several model systems using an acetylene link to couple different monoradicals. We simulated their magnetic strength using the broken symmetry method via density functional theory calculations. Our results show that the acetylene (–CC–) coupler gives much larger J values than the ethylene (–CC–) coupler. Acetylene coupling a phenoxyl (PO) and a nitronyl nitroxide (NN) diradical is the strongest ferromagnetic diradical OMM, and acetylene coupling a PO bisradical is the strongest anti-ferromagnetic diradical OMM. Moreover, we found that the magnetic strength is dependent on the coupler length and torsion, but is independent of the bending. Since only a very small energy barrier is needed, it is feasible to use torsion to tune a diradical OMM's magnetic strength. Our results are also beneficial for the rational design and synthesis of diradical OMMs.

To achieve highly efficient organic photovoltaics (OPVs), control of the molecular orientation is one of the prime important factors, for the interfacial dipole orientation and energy offset at the donor/acceptor (D/A) interface influence the device performance. We examine the effect of the planar-shape molecular orientation on the D/A interface, by comparing two different metal phthalocyanine molecules under identical process conditions. From the noticeable changes in the near edge X-ray absorption fine structure and ionization potential, copper phthalocyanine (CuPc) on a thin CuI layer reveals a strong change in molecular orientation compared to zinc phthalocyanine (ZnPc). To account for the different templating effect between CuPc and ZnPc, Cu d–d orbital coupling is evidenced as for the interaction between CuI and CuPc. The apparent change of the CuPc orientation reveals a relatively large energy offset at the CuPc/C60 D/A interface and increased open-circuit voltage in the corresponding OPV device. The lying-down orientation of CuPc on CuI or face-on geometry at the C60/CuPc interface induces strong electron–electron coupling and long-lived charge transfer exciton states, which is directly related to better charge separation and correspondingly good OPV performance.

Background/Aims: Calcium channel blockers (CCBs) are the most widely pre scribed medication for patients with vasospastic angina (VA). However, few studies have compared the prognosis of VA patients who are prescribed different CCBs. Methods: We enrolled 2,960 patients who received provocation test prospectively in 11 university hospitals in Korea. We divided 1,586 patients received four major CCBs into two groups: a first generation CCB (diltiazem and nifedipine) group and a second generation CCB (amlodipine and benidipine) group. Primary out come was time to events of composite of death from any cause, acute coronary syndrome (ACS) and symptomatic arrhythmia during 3-year follow-up. We also compared the effect of each CCB on the control of angina symptoms. Results: There was no difference of the primary outcome among the two groups with a cumulative incidence rate of 5.4%, 2.9%, and a person-month incidence rate of 2.33 and 1.26, respectively (hazard ratio [HR], 0.54; 95% confidence interval [CI], 0.25 to 1.17; p = 0.120, as reference with the 1st generation CCBs). The incidence of ACS was significantly lower in 2nd generation CCBs group with a person-month incidence rate of 1.66 vs. 0.35 (HR, 0.22; 95% CI, 0.05 to 0.89; p = 0.034). Use of benidipine showed a significant better control of angina symptom compared with diltiazem for 3 years (odds ratio, 0.17; 95% CI, 0.09 to 0.32; p < 0.0001 at 3rd year). Conclusions: The first and second generation CCB groups did not differ in terms of composite outcome occurrence. However, the ACS incidence rate was signifi cantly lower in the users of the 2nd generation CCBs.

We first observed the spin-to-charge conversion due to both the inverse Rashba-Edelstein effect (IREE) and inverse spin-Hall effect in a holey multilayer molybdenum disulfide (MoS2) intermediate layer in a Pt/YIG structure via LSSE measurements under nonequilibrium magnetization. We found an enhancement of approximately 238%, 307%, and 290% in the longitudinal spin Seebeck effect (LSSE) voltage, spin-to-charge current, and thermoelectric (TE) power factor, respectively, compared with the monolayer MoS2 interlayer in a Pt/YIG structure. Such an enhancement in the LSSE performance of Pt/holey MoS2/YIG can be explained by the improvement of spin accumulation in the Pt layer by induced spin fluctuation as well as increased additional spin-to-charge conversion due to in-plane IREE. Our findings represent a significant achievement in the understanding of spin transport in atomically thin MoS2 interlayers and pave the way toward large-area TE energy-harvesting devices in two-dimensional transition metal dichalcogenide materials.

Histidine tautomerism is considered a crucial component that affects the constitutional and accumulation characteristics of the tau267-312 monomer in the neutral condition, which are connected with the pathobiology of Alzheimer's disease (AD). Interpreting the organizational characteristics and accumulation procedure is a challenging task because two tautomeric conformations (the Ne-H or Nδ-H tautomer) can occur in the open neutral condition. In the current work, replica-exchange molecular dynamics (REMD) simulations were performed to investigate the structural properties of the tau267-312 monomer considering the histidine tautomeric effect. Based on the simulation outcomes, the histidine 268 (H268) (δ)-H299 (δ) (δδ) isomer had the highest β-sheet content with a value of 26.2%, which acquires a sheet-governing toxic conformer with the first abundant conformational state of 22.6%. In addition, δδ displayed notable antiparallel β-sheets between lysine 8 (K8)-asparagine 13 (N13) and valine 40 (V40)-tyrosine 44 (Y44) as well as between K32-H33 and V40-Y44 (β-meander supersecondary structure), indicating this tautomeric isomer may exist to stimulate tau oligomerization. Furthermore, H299 was found to play an essential role in the structural stabilization of the δδ isomer compared with H268. The present research will aid in obtaining insight into the organizational and accumulation properties of tau protein in the presence of histidine tautomerism to control AD.

Oxygen evolution (OER) and oxygen reduction (ORR) reactions are the key electrocatalytic redox couple for advanced energy storage/conversion, including rechargeable metal-air batteries and regenerative fuel cells. Heteroatom doped carbon catalysts propose a promising candidate for such purposes along with the superior durability and cost-effectiveness. Unfortunately, exact identification of the catalytic site as well as the critical role of dopants is still controversial in the catalytic mechanism. Here we present bifunctional catalytic site of nitrogen pair-doped graphene nanoribbons for precisely switchable OER and ORR. Pyrazolated N2-edges of graphene nanoribbon serve as switchable dual-functional active sites for OER/ORR with efficient activities and extraordinary durability. Theoretical calculation reveals genuine catalytic mechanism originating from the electrochemical potential-dependent molecular absorption and conversion at the atomic level dopant site. This judiciously controllable bifunctional electrocatalytic activity of dopant catalyst fundamentally addresses the interference between ORR and OER and attains highly stable rechargeable metal-air battery with long-term stability.

Magnesium in biogenic aragonite skeletons is widely used to infer the geochemical conditions under which the minerals precipitated. However, because molecular speciation of Mg2+ in the aragonite crystal structure remains uncertain, questions are often posed about the reliability and utility of Mg as a paleoenvironment proxy. In this study, we investigated the Mg2+ coordination in abiogenic aragonite by applying first-principles density functional theory (DFT). We report for the first time a Mg2+-site disordered structure model of aragonite: substituted Mg2+ is five-fold coordinated and induces substantial disorder in the bond distances that depends on the relative positions of Mg2+ sites. Using the site disorder model, DFT qualitatively reproduces the key features of experimental Mg K-edge X-ray absorption near-edge structure (XANES) spectra of aragonite. DFT also resolves a long-standing conflict between experimental and theoretical studies pertaining to the enrichment of 26Mg in aragonite relative to calcite. The Boltzmann-averaged 26Mg/24Mg reduced partition function ratios were calculated in the order of aqueous Mg2+ > Mg-doped aragonite > Mg-doped calcite. The disorder structure model is likely applicable to the substitution of cations with radii smaller than the radius of Ca2+ ions in aragonite. Our study provides atomistic insight into metal substitution effects on the metal content and isotope fractionation in biogenic aragonite, which are critical for developing a reliable paleoenvironment proxy.

Purpose: In this study, we investigated the biological activities of Diphylleia grayi extract as a natural cosmetic ingredient. Methods: The antioxidant activities of Diphylleia grayi extract was measured through 2,2-diphenyl-1-(2,4,6-trinitrophenyl)- hydrazinyl (DPPH), 2,2’-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) scavenging assay and Fluorescence recovery after photobleaching (FRAP) assay. We further investigated whether Diphylleia grayi extract could reduce the intracellular reactive oxygen species (ROS) using dichlorofluorescin diacetate (DCF-DA) fluorescence. To investigate the whitening effect, we conducted tyrosinase inhibition assay and L-DOPA oxidation inhibition assay. Furthermore, we performed collagenase inhibition assay to evaluate the anti-wrinkle activity of Diphylleia grayi extract. Results: Diphylleia grayi extract showed strong antioxidant activities on DPPH, ABTS radical scavenging assay. Furthermore, Diphylleia grayi extract decreased the intracellular ROS on skin normal cells. Diphylleia grayi extract has also inhibition activities of tyrosinase, L-DOPA oxidation and collagenase. Conclusion: Therefore, Diphylleia grayi extract was identified as a promising natural cosmetic ingredient which has antioxidant, whitening and anti-wrinkle properties.

Summary Background Evidence from numerous randomised clinical trials suggest that shorter-term antimicrobial therapy is as effective as—and has other advantages over—longer-term antimicrobial regimens at achieving symptomatic cure for acute uncomplicated cystitis. Nevertheless, not all shorter regimens are adopted in clinical guidelines. This study was done to reappraise the treatment duration of each antibiotic in current guidelines for acute uncomplicated cystitis to investigate whether the regimen lengths of guideline approved antibiotics could be reduced. Methods We systematically searched the PubMed, Embase, and Cochrane Library databases for relevant publications from inception of the databases until Dec 31, 2019. Only randomised clinical trials of women with acute uncomplicated cystitis that assessed antibiotic therapy and reported clinical or microbial response outcome values were included. A network meta-analysis was done and the quality of evidence of all of the included studies was rated. Clinical response was the primary outcome, defined as the complete disappearance of all baseline symptoms at the test-of-cure visit. Bayesian hierarchical random-effects model for dichotomous outcomes was used to compare the efficacy of each antibiotic treatment regimen directly and indirectly. This systematic review is registered in PROSPERO, CRD42018093529. Findings Overall, 61 randomised clinical trials—which included 20 780 patients—were assessed in our systematic review. For the third-generation and fourth-generation fluoroquinolones, a 3-day regimen had similar effect to a single-dose regimen for clinical response (risk ratio [RR] 0·994 [95% credible interval 0·939–1·052] vs 1·024 [0·974–1·083]), with moderate quality of evidence. For pivmecillinam, 5-day and 7-day regimens were similar to a 3-day regimen for clinical response, with moderate quality of evidence (RR 1·041 [0·910–1·193] for the 5-day regimen and 1·095 [0·999–1·203] for the 7-day regimen). Meanwhile, for third-generation cephalosporins and amoxicillin and clavulanate, there was no difference between single-dose and 3-day regimens, but quality of evidence supporting this conclusion was low. For second-generation quinolones and co-trimoxazole, single-dose regimen was less effective than 3-day regimen in clinical response, with moderate quality of evidence. Interpretation Treatment duration of the third-generation and fourth-generation quinolones and pivmecillinam could be shorter than the currently recommended regimens for acute uncomplicated cystitis. For other antibiotics, shorter duration of regimens could be considered, but further research is needed because of the low quality of supporting evidence. Funding None.

The core-shell-structured gold-polyaniline (Au-PANI) nanocomposite delivered a CO2 -to-CO conversion efficiency of 85 % with a high current density of 11.6 mA cm-2 . The polyaniline shell facilitated CO2 adsorption, and the subsequent formation of reaction intermediates on the gold core contributed to the high efficiency observed.

Most cerebellar ataxias (CAs) are incurable neurological disorders, resulting in a lack of voluntary control by inflamed or damaged cerebellum. Although CA can be either directly or indirectly related to cerebellar inflammation, there is no suitable animal model of CA with neuroinflammation. In this study, we evaluated the utility of an intracerebellar injection of lipopolysaccharide (LPS) to generate an animal model of inflammatory CA. We observed that LPS administration induced the expression of pro-inflammatory molecules following activation of glial cells. In addition, the administration of LPS resulted in apoptotic Purkinje cell death and induced abnormal locomotor activities, such as impaired motor coordination and abnormal hindlimb clasping posture. Our results suggest that intracerebellar LPS administration in experimental animals may be useful for studying the inflammatory component of CA.
