A new indole diketopiperazine alkaloid, named penilline D (1), together with five known indole alkaloid analogues (2-5, 11), two meroterpenoids (6 and 12), and four butenolide derivatives (7-10), were isolated from the Antarctic fungus Penicillium sp. SCSIO 05705. Extensive spectroscopic analysis and electronic circular dichroism (ECD) calculation were used to elucidate the structure of penilline D (1), including its absolute configuration. All isolated compounds (1-12) were evaluated for their cytotoxic, antibacterial and enzyme inhibitory activities against acetylcholinesterase (AChE) and pancreatic lipase (PL). Among them, compound 5 exhibited moderate in vitro cytotoxic activity against the 143B cell line with IC50 value of 12.64 ± 0.78 μM. Compound 6 showed strong inhibitory activity against AChE with IC50 value of 0.36 nM (IC50 18.7 nM for Tacrine), while compounds 6 and 11 showed weak PL enzyme inhibitory activity. Furthermore, an in silico molecular docking study was also performed between 6 and AChE.

Euroticins C–E (1–3), three pairs of salicylaldehyde derivative enantiomers, were isolated from a marine-derived fungus Eurotium sp. SCSIO F452. They represent two types of salicylaldehyde derivatives bearing unprecedented highly constructed 6/6/6/6 tetracyclic structures. Their structures were established by comprehensive spectroscopic analyses, X-ray diffraction, and electronic circular dichroism calculations. Compounds (+)-1 and (−)-1 showed significant antioxidative activity and moderate cytotoxic activity.

Two pairs of salicylaldehyde derivative enantiomers, euroticins A and B (1 and 2), were isolated from a marine-derived fungus Eurotium sp. SCSIO F452. Compound 1 possesses a highly constructed 6/6/6/5/7 pentacyclic structure featuring an unprecedented 2,11-dioxatricyclo[5.3.1.04,8]undecane core. Compound 2 represents the first example of 6/6/6/6 tetracyclic salicylaldehyde derivative. Their structures were established by spectroscopic analyses, X-ray diffraction, and electronic circular dichroism (ECD) and 13C NMR calculations. Compounds (+)-2 and (-)-2 exhibited remarkable antioxidative activities.

To enlarge the chemical diversity of Eurotium sp. SCSIO F452, a talented marine-derived fungus, we further investigated its chemical constituents from a large-scale fermentation with modified culture. Four pairs of new salicylaldehyde derivative enantiomers, euroticins F-I (1–4), as well as a known one eurotirumin (5) were isolated and characterized. Compound 1 features an unprecedented constructed 6/6/6/5 tetracyclic structures, while 2 and 3 represent two new types of 6/6/5 scaffolds. Their structures were established by comprehensive spectroscopic analyses, X-ray diffraction, 13C NMR, and electronic circular dichroism calculations. Selected compounds showed significant inhibitory activity against α-glucosidase and moderate cytotoxic activities against SF-268, MCF-7, HepG2, and A549 cell lines.

A new butenolide derivative (±)-asperteretal F (1) and related congener (2) recently reported containing an unusual 2-benzyl-3-phenyl substituted lactone core, together with five known compounds (3–7) were isolated and characterized from the fungus Aspergillus terreus. SCSIO FZQ028 derived from a deep-sea sediment of South China Sea. Their chemical structures were established on the basis of 1D- and 2D-NMR spectroscopic data, and HR-ESI-MS analysis. Additionally, all the compounds were evaluated for the antioxidative activities against DPPH, cytotoxic activities against two tumor cell lines (SF-268 and HepG-2), and antimicrobial activities. Compounds 2-4, and 7 showed significant activities against DPPH with IC50 ranging from 5.89 to 10.07 μg/mL. Compounds 2 and 4 showed moderate antimicrobial activities against all four tested bacteria.

A new strategy was proposed for the electrochemical fabrication of manganese dioxide (MnO2) coatings on titanium (Ti) fibers. In order to achieve stable MnO2 coatings, a Ti wire was initially anodized in methanol/water containing fluoride ions for the in situ growth of nano/submicro-scale titania (TiO2) pyramids. Thereafter, silica nanoparticles (SiO2NPs) were electrophoretically deposited on the TiO2 coatings. Finally, the nanoparticle-like MnO2 coating was homogeneously formed on the Ti@TiO2@SiO2NPs fiber support in neutral electrolytic solution by cyclic voltammetry using polypyrrole as an organic binder. The resulting fiber showed good extraction selectivity and efficient extraction capability for aromatic compounds with large delocalized π-systems. For this purpose, this fiber was used for the selective enrichment of the studied polycyclic aromatic hydrocarbons (PAHs) coupled to high-performance liquid chromatography with ultraviolet detection. The linear calibration curves were obtained in the concentration range of 0.01–500 μg L−1 with correlation coefficients higher than 0.999 in all instances. The relative standard deviations (RSDs) for single fiber repeatability varied from 4.7% to 5.5% for intra-day and inter-day measurements (n = 5), and RSDs for the fiber-to-fiber reproducibility varied from 5.8% to 6.7% (n = 5). The developed method was successfully applied to the selective enrichment and sensitive determination of target PAHs in river water, rain water and wastewater samples with relative recoveries from 93.0% to 106%. Furthermore, the fabricated fiber was stable enough to perform the whole analytical sequence in practical applications.

Accumulation of toxic ions in leachate is one factor limiting bioleaching applications. The effect of fluoride ions on the growth of bioleaching microorganisms has been extensively emphasized. In this study, HF is found to be the toxic form of fluoride that affects the bacterial activity under acidic conditions. The added aluminum could compete with H+ to complex with F−, thus significantly decrease the concentration of HF and finally reduce the toxicity of fluoride to bacteria. When F−/Al3+ concentration ratio is 0.5:1.0, Fe2+ oxidation rate could reach 0.167 g·L−1·h−1, close to that of the biotic control group (0.195 g·L−1·h−1). The competitive complexation mechanism of fluoride by $${\text{AlF}}_{n}^{3 - n}$$ results in stability constants of $${\text{AlF}}_{n}^{3 - n}$$ complex (7.00) that are larger than those of HF (3.18). The F−/Al3+ concentration ratio in the medium could affect the speciation of $${\text{AlF}}_{n}^{3 - n}$$ complex. With the decrease in F−/Al3+ concentration ratio, the coordination numbers of $${\text{AlF}}_{n}^{3 - n}$$ decrease. Finally, the feasibility of fluoride detoxification by aluminum ion is verified. This work has meaningful implications for fluoride-containing bacterial bioleaching systems.

A novel mesophilic marine actinobacterial strain, designated as SCSIO 08198T, was isolated from a deep-sea sediment sample collected from the Indian Ocean. The strain was Gram-stain-positive, rod-shaped and salmon pink in colour. Good growth occurred on marine agar with 1–5 % (w/v) NaCl and incubation at 28 °C for more than a fortnight. The present study aimed to explore the role and mechanisms of proprotein convertase subtilisin/kexin type 9 (PCSK9) in the oxidized low-density lipoprotein (oxLDL)-induced pyroptosis of vascular endothelial cells. For this purpose, human umbilical vein endothelial cells (HUVECs) were incubated with oxLDL (100 µg/ml) for 24 h to induce pyroptosis, which was detected using PI/hoechst33342 double staining. The expression of pyroptosis-associated molecules was measured by western blot analysis and RT-qPCR. Reactive oxygen species (ROS) and membrane potential were examined through ROS probe and JC-1 staining, respectively. PCSK9 and mitochondrial ubiquinol-cytochrome c reductase core protein 1 (UQCRC1) protein were knocked down by small interfering RNA (siRNA). PCSK9 was overexpressed by lentivirus. The results revealed that oxLDL induced HUVEC injury, pyroptosis and inflammatory factor release, and upregulated the expression of PCSK9 protein in the HUVECs in a concentration-dependent manner. The silencing of PCSK9 expression with siRNA suppressed the oxLDL-induced damage to HUVECs, the release of inflammatory substances and the occurrence of pyroptosis. In addition, oxLDL inhibited UQCRC1 expression, promoted mitochondrial membrane potential collapse and damaged mitochondrial function; however, these processes were reversed by the silencing of PCSK9. PCSK9 overexpression induced the pyroptosis of HUVECs, the generation of ROS and the disorder of mitochondrial function by inhibiting UQCRC1. Therefore, PCSK9 mediates the oxLDL-induced pyroptosis of vascular endothelial cells via the UQCRC1/ROS pathway.

Advanced oxidation processes (AOPs) are important technologies for aqueous organics removal. Despite organic pollutants can be degraded via AOPs generally, high mineralization of them is hard to achieve. Herein, we synthesized a manganese oxide nanomaterial (H2-OMS-2) with abundant Bronsted-acid sites via ion-exchange of cryptomelane-type MnO2 (OMS-2), and tested its catalytic performance for the degradation of phthalate esters via peroxymonosulfate (PMS) activation. About 99% of dimethyl phthalate (DMP) at a concentration of 20 mg/L could be degraded within 90 min and 82% of it could be mineralized within 180 min over 0.6 g/L of catalyst and 1.8 g/L of PMS. The catalyst could activate PMS to generate SO4-˙ and ·OH as the dominant reactive oxygen species to reach complete degradation of DMP. Especially, the higher TOC removal rate was obtained due to the rich Bronsted-acid sites and surface oxygen vacancies on the catalyst. Kinetics and mechanism study showed that MnII/MnIII might work as the active sites during the catalytic process with a lower reaction energy barrier of 55.61 kJ/mol. Furthermore, the catalyst could be reused for many times through the regeneration of the catalytic ability. The degradation and TOC removal efficiencies were still above 98% and 65% after seven consecutive cycles, respectively. Finally, H2-OMS-2-catalyzed AOPs significantly reduced the organismal developmental toxicity of the DMP wastewater through the investigation of zebrafish model system. The present work, for the first time, provides an idea for promoting the oxidative degradation and mineralization efficiencies of aqueous organic pollutants by surface acid-modification on the catalysts.

The present trial was performed to reveal the regulatory effects of L-theanine on the levels of lipopolysaccharide (LPS) endotoxin within different biofluids, as well as relevant inflammatory responses of dairy cattle under heat stress conditions. Thirty lactating Chinese Holstein dairy cattle (189 ± 47 days in milk, and 2 ± 1 parities) were allocated in a completely randomized design to each of 3 dietary treatments: the control (CON, 0 g/d per cow L-theanine), the low L-theanine dosage treatment (LL, 16 g/d per cow L-theanine), and the high L-theanine dosage treatment (HL, 32 g/d per cow L-theanine). This trial consisted of 38 d (7 d for adaption and 31 d for data and sample collection), and sample collection for rumen liquid, blood plasma or serum, and milk were conducted on the d 27 and 38, respectively. SCSIO F452. Their structures and absolute configuration were determined by extensive spectroscopic analysis and electronic circular dichroism (ECD) calculations. All the new optical pure enantiomers [(+)-1, (-)-1, (+)-2, (-)-2] were evaluated for their cytotoxic and antioxidative activities. Compound (-)-1 exhibited weak cytotoxic activity.

Summary Aim The aim of the study was to determine the clinical value of in vitro high-throughput drug sensitivity screening with primary hepatocellular carcinoma cells to select drugs for adjuvant chemotherapy. Methods This study included 162 patients who underwent hepatectomy from September 2013 to December 2016. The patients were divided into a drug sensitivity screening group and an empirical treatment group. High-throughput drug sensitivity screening using primary HCC cells was carried out and, based on the test results, effective drugs were selected for treatment. Patients in the empirical group were treated with commonly used drugs, according to the clinicians’ preferences. Clinical efficacy, i.e., disease-free survival (DFS) time, was compared between the two groups. Results Most patients with HCC showed extensive resistance to known chemotherapeutic drugs. However, bortezomib, regorafenib, sorafenib, romidepsin, hydroxycamptothecin and adriamycin + oxaliplatin showed strong anti-HCC activity in the sensitivity assay. Comparing clinical efficacy, the overall median DFS of patients in the drug sensitivity screening group was significantly better than that of patients in the empirical treatment group (17.00 ± 3.80 months vs. 9.00 ± 1.18 months, P = 0.001). Median DFS times in the TACE group were 9.00 ± 4.07 months vs. 7.00 ± 1.06 months (P = 0.014) and median DFS times in the oral drugs group were 16.80 ± 3.98 months vs. 10.00 ± 0.81 months (P = 0.024). Patients DFS was 69.4%, 62.5% at 1-, 2- years, respectively, for patients with drug sensitivity screening, and 48.5%, 37.8% at 1-, 2- years, respectively, for patients with empirical treatment. Conclusion High-throughput drug sensitivity screening can be successfully used to screen chemotherapeutic drugs for efficacy against HCC and the efficacious drugs can be used in postoperative adjuvant chemotherapy of HCC patients. This treatment paradigm is safe and reliable, and improves survival compared with empirical chemotherapy.

Summary The symmetrical configuration is attractive for electrochemical energy storage devices due to using one electrode material, but it is difficult to achieve for a rechargeable battery. Here, we propose an electrochemical activity modulation method to construct a quasi-symmetrical rechargeable battery with decoupled conversion reactions of the anionic/cationic counterparts of covalent transition metal sulfides. The modulated process contains potential window and electrolyte modulation, followed by a predesigned conversion reaction. For molybdenum disulfide (MoS2), after cathodic modulation the sulfur redox (S0 to S2−) inside MoS2 was activated as redox center of cathode, resulting in a capacity increase of one order of magnitude and high cycling stability. Meanwhile, an anodic-modulated MoS2 anode displayed high capacity due to the reversible conversion between Mox+ and Mo0. Coupling this cathode and anode, a quasi-symmetrical battery is fabricated and opens up a new direction for building high-energy electrochemical systems.

In this work, cellulose nanocrystal (CNC) was modified by an ureido-pyrimidinone (UPy) system based on quadruple hydrogen bondings, and CNC-UPy was obtained. Then, this powder was added into polyvinyl alcohol (PVA), and PVA/CNC-UPy composite membranes and hydrogels were prepared. Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), polarizing optical microscopy (POM) and particle size distribution (PSD) were used to characterize CNC-UPy. From the FTIR results, the characteristic peaks of NCO group sat 2270 cm&minus, 1 disappeared, indicating the successful synthesis of CNC-UPy. XRD results showed that the modification by UPy may change the structure of CNC and its degree of crystallinity was increased. PSD analysis showed that the particle size of CNC was increased and its size distribution became narrower after modification by UPy groups. The structure and properties of the composite membranes and hydrogels were studied by differential scanning calorimeter (DSC), thermogravimetric analysis (TGA) together with investigation of swelling, sustained release and self-healing performances. DSC curves depicted that the glass transition temperature, Tg, of different PVA membranes was increased with addition of different proportions of CNC-UPy. TGA data showed that the temperature of maximum weight loss rate was increased, which illustrated the enhanced thermal stability of PVA/CNC-UPy composites. Meanwhile, it was also revealed that the PVA/CNC-UPy composite hydrogels possess good self-healing and better sustained release behavior for the soil conditioner, fulvic acid (FA).

Seven new compounds, including four new chlorinated diphenyl ethers, namely chrysines A–D ( 1 – 4 ), one new dichlorinated xanthone, chrysoxanthone ( 5 ), dichloroorcinol ( 6 ), and one new benzeneacetic acid derivative, 3-isopentyl-4-hydroxy phenylacetic acid methyl ester ( 7 ), along with fourteen known compounds ( 8 – 21 ), were isolated from a deep-sea-derived fungus Penicillium chrysogenum SCSIO 41001. Their structures were determined by extensive spectroscopic methods and X-ray single-crystal diffraction analysis. All of the isolated compounds ( 1 − 21 ) were evaluated for their α-glucosidase inhibitory activity using PNPG method. Among them, nine compounds ( 2 , 3 , 5 , 6 , 8 , 9 , 13 , 17 , and 18 ) exhibited inhibitory activity against α-glucosidase with IC 50 values of 0.35, 0.20, 0.04, 0.16, 0.15, 0.09, 0.14, 0.14, and 0.12 mM, respectively (IC 50 0.28 mM for the positive control acarbose).

Background Lipoprotein(a) [LP(a)] is implicated as a common and independent risk factor for cardiovascular diseases. The therapeutic options currently available for reducing plasma LP(a) concentrations are limited. Diallyl disulphide (DADS), the main component of garlic, regulates lipid metabolism in hepatocytes and adipocytes through ERK1/2 signalling. This study aimed to assess the effect of DADS on apolipoprotein(a) [apo(a)] in HepG2 cells. We also determined the effects of DADS on apo(a) expression and secretion in HepG2 cells as well as the underlying mechanisms. Methods We examined the role of DADS on apo(a) expression in HepG2 cells by treating cell with different concentrations of DADS (10, 20, 40 and 80 μg/mL) for 24 h or treating cells with 40 μg/mL DADS for 0, 6, 12, 24 and 48 h. Then we used quantitative real-time PCR to analysis apo(a) mRNA levels, used Western blot to analysis apo(a) protein levels and used enzyme-linked immunosorbent assay to test apo(a) secreted levels. To farther determined the role of DADS, we applied Transfection of small interfering RNA to knockdown ELK-1levels and applied PD98059, a specific inhibitor of ERK1/2, to block ERK1/2 signal. Results The results show DADS inhibited apo(a) at both the mRNA and protein levels in HepG2 cells in a dose-dependent manner. DADS-mediated inhibition of apoa(a) expression in HepG2 cells was attenuated when the cells were cultured in medium containing PD98059 (ERK1/2 inhibitor) or were transfected with siRNAs against MEK1 or ELK-1. Overexpression of apo(a) yielded similar results. Conclusions This study reveals that DADS can downregulate apo(a) expression in a dose-dependent manner via the MEK-ERK12-ELK-1 pathway.

Three new tricyclic cyclopiazonic acid (CPA) related alkaloids asperorydines N-P (1–3), together with six known compounds (4–9) were isolated and characterized from the fungus Aspergillus flavus SCSIO F025 derived from the deep-sea sediments of South China Sea. The structures including absolute configurations of 1–3 were deduced from spectroscopic data, X-ray diffraction analysis, and electronic circular dichroism (ECD). All compounds were evaluated for the antioxidative activities against DPPH, cytotoxic activities against four tumor cell lines (SF-268, HepG-2, MCF-7, and A549), and antimicrobial activities. Compound 9 showed significant radical scavenging activities against DPPH with an IC50 value of 62.23 μM and broad-spectrum cytotoxicities against four tumor cell lines with IC50 values ranging from 24.38 to 48.28 μM. Furthermore, compounds 4–9 exhibited weak antimicrobial activities against E scherichia coli, and compound 9 also showed antibacterial activity against Bacillus thuringiensis, Micrococcus lutea, Staphylococcus aureus, Bacillus subtilis, Methicillin resistant Staphylococcus aureus.

The therapeutic effect of metformin (Met) on atherosclerosis was studied here. Effects of methionine and Met on the induction of inflammatory response and H2 S expression in peritoneal macrophages were evaluated. Enzyme-linked immunosorbent assay, immunohistochemistry assay, western blot, and quantitative reverse transcription polymerase chain reaction were conducted to observe the levels of cystathionine γ-lyase (CSE), DNA methyltransferases 1 (DNMT1), DNMT3a, DNMT3b, tumor necrosis factor (TNF- α), interleukin 1b (IL-1β), and hydrogen sulfide (H 2 S). Luciferase and bisulfite sequencing assays were also utilized to evaluate the CSE promoter activity as well as the methylation status of CSE in transfected cells. Methionine significantly elevated Hcy, TNF-a, H 2 S, and IL-1β expression while decreasing the level of CSE in C57BL/6 mice. In contrary, co-treatment with Methionine and Met reduced the detrimental effect of Methionine. Homocysteine (Hcy) decreased H 2 S expression while promoting the synthesis of IL-1β and TNF-α in THP-1 and raw264.7 cells. Treatment of THP-1 and raw264.7 cells with methionine and Met reduced the activity of methionine in dose dependently. Moreover, Hcy increased the expression of DNMT and elevated the level of methylation in the CSE promoter, whereas the co-treatment with methionine and Met attenuated the effects of Hcy. Methionine significantly decreased plasma level of CSE while increasing the severity of inflammatory responses and plasma level of Hcy, which in turn suppressed H 2 S synthesis and enhanced DNA hypermethylation of CSE promoter to promote the pathogenesis of atherosclerosis. In contrary, co-treatment with methionine and Met reduced the detrimental effect of methionine.

The objective of the present study was to investigate the alterations of lipopolysaccharide (LPS) endotoxin across four different biofluids (i.e., rumen fluid, blood plasma, milk, and urine) and LPS-related inflammation responses in Holstein dairy cows under heat stress through the regulation of dietary cation-anion difference (DCAD). Eight lactating Holstein dairy cows were randomly assigned in a 2 × 2 crossover design to either a control DCAD diet (CON: 33.5 mEq/100 g DM) or a high DCAD diet (HDCAD: 50.8 mEq/100 g DM), and each experimental period lasted for 21 days. Among them, new compounds chrysopyrones A and B (1 and 2) displayed obvious inhibitory activities against PTP1B with IC50 values of 9.32 and 27.8 μg/mL, respectively. SCSIO F452. 1 and 2, occurring as a pair of diastereomers, both presented a hexahydropyrrolo[2,3-b]indole skeleton. Their chemical structures, including absolute configurations, were elucidated by 1D and 2D NMR, HRESIMS, quantum chemical calculations of electronic circular dichroism, and single crystal X-ray diffraction experiments. Most isolated compounds were screened for antioxidative potency. Compounds 3, 5, 6, 7, 9, 10, and 12 showed significant radical scavenging activities against DPPH with IC50 values of 13, 19, 4, 3, 24, 13, and 18 µM, respectively. Curcumin, a traditional Chinese derivative from the rhizomes of Curcuma longa, is beneficial to health by modulating lipid metabolism and suppressing atherogenesis. A key part of atherosclerosis is the failure of macrophages to restore their cellular cholesterol homeostasis and the formation of foam cells. In this study, results showed that curcumin dramatically increased the expression of ATP-binding cassette transporter 1 (ABCA1), promoted cholesterol efflux from THP-1 macrophage-derived foam cells, and reduced cellular cholesterol levels. Curcumin activated AMP-activated protein kinase (AMPK) and SIRT1, and then activated LXRα in THP-1 macrophage-derived foam cells. Inhibiting AMPK/SIRT1 activity by its specific inhibitor or by small interfering RNA could inhibit LXRα activation and abolish curcumin-induced ABCA1 expression and cholesterol efflux. Thus, curcumin enhanced cholesterol efflux by upregulating ABCA1 expression through activating AMPK-SIRT1-LXRα signaling in THP-1 macrophage-derived foam cells. This study describes a possible mechanism for understanding the antiatherogenic effects of curcumin on attenuating the progression of atherosclerosis.

A new three-phase solvent system, n-hexane–acetonitrile–dichloromethane–water–ethyl acetate (5:5:1:5:1.5, v/v/v/v/v) was developed for the high-speed counter-current chromatographic (HSCCC) separation and purification of five bioactive constituents, syringic acid (1), vomifoliol (2), vanillic acid (3), 6-hydroxy-2-benzoxazolinone (4), and 2-benzoxazolinone (5), from Acanthus ilicifolius.

Three new indolyl diketopiperazine derivatives, penillines A and B (1 and 3), isopenilline A (2), together with seven known alkaloid compounds (E)-3-(1H-imidazole-4-yimethylene)-6-(1H-indl-3-ylmethyl)-2,5-piperazinediol (4), penilloid A (5), meleagrin (6), neoxaline (7), questiomycin A (8), N-(2-hydroxyphenyl)-acetamide (9), 2-benzoxazolinone (10), were isolated from the Antarctic soil-derived fungus Penicillium sp. SCSIO 05705. The new structures of 1–3 were elucidated on the basis of 1D and 2D NMR, mass spectra, CD spectra, and quantum chemical calculations. All the isolated compounds were tested for their antiviral (H1N1 and H3N2), antituberculosis, antibacterial, and cytotoxic activities. Among them, compounds 6–8 had significant in vitro cytotoxicities against the K562, MCF-7, A549, U937, Hela, DU145, HL60, and HT29 cell lines, with IC50 values ranging from 2.73 to 17.7 μM. In addition, compound 8 showed potent antituberculosis activity with an MIC value of 3.91 μM, and the inhibition effect was close to the positive control INH (isoniazid, MIC 2.04 μM). A possible biogenesis pathway for compounds 1–7 was proposed.

Increased levels of asymmetric dimethylarginine (ADMA) have been observed in patients with acute ischemic stroke. We aimed to investigate the correlation between ADMA and ischemic stroke, and evaluate the effect of supplementation of folic acid and vitamin B12 on concentrations of ADMA. Patients were randomized into intervention and non-intervention groups within 3 days after symptom onset. Intervention group patients were treated with folic acid (5mg daily) and vitamin B12 (500 μg twice daily) for 12 weeks. ADMA and homocysteine (Hcy) concentrations were measured before treatment (baseline) and 2 and 12 weeks after treatment. The laboratory measures were also collected from healthy controls. Eighty five subjects were enrolled in this study, from whom 72 with complete baseline and follow-up laboratory data were included in the present analysis. Lipoprotein(a)[Lp(a)] is a risk factor for coronary heart diseases. However, the metabolism of this protein remains poorly understood. Efficient and specific drugs that can decrease high plasma levels of Lp(a) have not been developed yet. Vitamin C is responsible for maintaining the catalytic activity of a group of iron and 2-oxoglutarate (2OG)-dependent dioxygenases and induces the generation of 5-hydroxymethylcytosine (5hmC) via Ten-eleven translocation (Tet) dioxygenases. In addition, It has been reported vitamin C deficiency induces atherosclerosis and increases Lp(a) and apo(a) plasma levels in Lp(a)+ mice. However, the mechanism is still unclear. In this study, we investigated the effects of vitamin C on apo(a) expression and the possible molecular mechanism of vitamin C that influences apolipoprotein(a) [apo(a)] biosynthesis in HepG2 cells. Results showed that vitamin C significantly inhibited the expression and secretion levels of apo(a). Vitamin C can also increase ELK1 expression and hydroxymethylation of ELK1 promoter and the globle DNA in HepG2 cells. In addition, the effects of vitamin C inhibiting the apo(a) expression were attenuated by ELK1siRNA and Tet2siRNA. These results suggested vitamin C down-regulate apo(a) expression via Tet2-dependent DNA demethylation in HepG2 cells.

In order to strengthen the monitoring and control of the groundwater quality, the groundwater of Hetang and Longtan water source regions in Beihai city was taken as example to evaluate the groundwater quality. PH, Total dissolved solids, Chloride, Iron, Nitrate, Ammonia- nitrogen, Cyanide, Mercury, Cadmium, Lead and so on were selected as the main factors of water quality evaluation. Single component and comprehensive evaluation methods were used to analyze the class and change trend of groundwater quality. The results showed that groundwater quality of Longtan was slightly better that of Hetang. F value of these two water source regions varied from 2.14 to 2.19 and met the classⅡ, which showed that the groundwater quality of Beihai City was good.

The aim of the present study was to investigate the attenuation of endothelial cell senescence by H2S and to explore the mechanisms underlying the anti-aging effects of H2S. Senescence was induced in human umbilical vein endothelial cells (HUVECs) by incubation in 25 µmol/l H2O2 for 1 h. Senescence-associated β-galactosidase (SA-β-gal) activity was examined to determine the effects of H2S on senescent HUVECs. The results indicated that SA-β-gal activity in the H2O2-treated HUVECs was 11.2 ± 1.06%, which was attenuated in the NaHS group. Pretreatment with nicotinamide (NAM), a sirtuin 1 (SIRT1) inhibitor, inhibited the reduction in senescence associated with H2S. Immunoblot analyses revealed that SIRT1 levels in senescent HUVECs treated with NaHS (60 µM) were indistinguishable from controls; however, analyses of SIRT1 activity indicated that SIRT1 enzyme activity was enhanced. In addition, we found that H2S improves the function of senescent HUVECs. The present study demonstrated that H2S protects against HUVEC senescence, potentially through modulation of SIRT1 activity. Furthermore, this study establishes a novel endothelial protective effect of H2S.

Atherosclerosis is an inflammatory disease of large and medium sized arteriole walls that is precipitated by elevated levels of low-density lipoprotein (LDL) cholesterol in the blood. However, the mechanisms that lead to the initiation of atherosclerosis are not fully understood. In this study, endothelial cells (ECs) were incubated with LDL for 24 h, and then the lipid was detected with Oil Red O staining and cholesterol ester was assayed with high-performance liquid chromatography (HPLC). F-actin was examined by fluorescence microscopy and the viscoelasticity of ECs was investigated using the micropipette aspiration technique. Then, a parallel-plate flow chamber device was used to observe the adhesion and retention of ECs under shear stress. The results demonstrated that elevated LDL significantly increased the cellular lipid content and induced the rearrangement of cytoskeletal F-actin. The initial rapid deformability (l/K 1 + l/K 2) was reduced by elevated cellular LDL levels, while membrane viscosity (μ) was increased by LDL accumulation. After treatment with 150 mg L(-1) LDL for 24 h, the adhesion of ECs under fluid shear stress was significantly decreased (p < 0.05). These results suggested that LDL induced cellular lipid accumulation and cytoskeleton reorganization which increased the cellular stiffness and decreased the adhesion of ECs.

Kynurenic acid (KYNA), an antagonist of the α7 nicotinic acetylcholine receptor and the N-methyl-D-aspartate receptor, and 3-hydroxykynurenine (3-HK), a generator of reactive oxygen species, are neuroactive metabolites of the kynurenine pathway of tryptophan degradation. In the mammalian brain as elsewhere, both compounds derive from a common bioprecursor, L-kynurenine (L-KYN). Recent studies in rats demonstrated that D-kynurenine (D-KYN), a metabolite of the bacterial amino acid D-tryptophan, can also function as a bioprecursor of brain KYNA. We now investigated the conversion of systemically administered D-KYN to KYNA in mice and also explored the possible production of 3-HK in the same animals. Thirty min after an injection of D-KYN or L-KYN (30 mg/kg, i.p.), newly produced KYNA and 3-HK were recovered from plasma, liver, forebrain and cerebellum in all cases. Using a new chiral separation method, 3-HK produced from D-KYN was positively identified as D-3-HK. L-KYN was the more effective precursor of KYNA in all tissues and also exceeded D-KYN as a precursor of brain 3-HK. In contrast, D-KYN was more potent as a precursor of 3-HK in the liver. The production of both KYNA and 3-HK from D-KYN was rapid in all tissues, peaking at 15–30 min following a systemic injection of D-KYN. These results show that biosynthetic routes other than those classically ascribed to L-KYN can account for the synthesis of both KYNA and 3-HK in vivo. This new insight may be of significant physiological or pathological relevance.

A new triterpenoid saponin, 3-O-β-D-glucopyranosyl-(1→2)-β-D-glucopyranosyl subprogenin D (1), together with six known triterpenoids: subprogenin D (2), abrusgenic acid (3), triptotriterpenic acid B (4), abruslactone A (5), abrusogenin (6) and abrusoside C (7) were isolated from the leaves and stems of Abrus precatorius. Their structures were elucidated on the basis of physical and NMR analysis, respectively. Compounds 5 and 6 showed moderate cytotoxicity against MCF-7, SW1990, Hela, and Du-145 cell lines. Compounds 1, 2 and 4 were isolated from this plant for the first time.

Recent evidence indicates that hydrogen sulfide (H2S) exerts an antiatherogenic effect, but the mechanism is unclear. Formation of macrophage-derived foam cells is a crucial event in the development of atherosclerosis. Thus, we explore the effect of H2S on the formation of macrophage-derived foam cells. Incubation of monocyte-derived macrophages with oxidized LDL (oxLDL) alone caused significant increases both in intracellular lipids revealed by Oil-red O staining and in intracellular total cholesterol (TC) and esterified cholesterol (EC) concentrations assessed by high-performance liquid chromatography. Sodium hydrosulfide (NaHS, an H2S donor) remarkably abrogated oxLDL-induced intracellular lipid accumulation, and attenuated TC and EC concentrations and EC/TC ratio, whereas dl-propargylglycine (PPG) (a H2S-generating enzyme cystathionine gamma lyase inhibitor) exacerbated lipid accumulation and augmented TC and EC concentrations and EC/TC ratio. Incubation of 1,1′-dioctadecyl-3,3,3′,3′-tetramethylindocarbocyanine perchlorate (DiI)-oxLDL led to lipoprotein binding and uptake of macrophages, which was blunted by NaHS, but enhanced by PPG. Furthermore, OxLDL markedly induced CD36, scavenger receptor A (SR-A) and acyl-coenzyme A:cholesterol acyltransferase-1 (ACAT-1) expressions in macrophages, which was suppressed by NaHS (50–200 μmol/L). Finally, the down-regulations of TC and EC concentrations as well as CD36 and ACAT-1 expressions by NaHS were suppressed by glibenclamide, a KATP channel blocker, but facilitated by PD98059, an extracellular signal-regulated kinases 1 and 2 (ERK1/2) inhibitor. These results suggested that H2S inhibits foam cell formation by down-regulating CD36, SR-A and ACAT1 expressions via the KATP/ERK1/2 pathway in human monocyte-derived macrophages.

The electrochemical process of galena in a pH 12.8 buffer solution was investigated using chronoamperometry and chronopotentiometry. To establish kinetic parameters on the surface of galena in the diethyldithiocarbamate solution, the exchange current density and the dependence of current density on reaction time were determined. Experimental results demonstrate that the exchange current density of galena is 1.585×10−2 A/m2 in the diethyldithiocarbamate-free solution. In the diethyldithiocarbamate solution, the thickness of lead diethyldithiocarbamate adsorbed on the surface of galena is 3.28 molecular layers, the diffusion coefficient of diethyldithiocarbamate on the surface of galena electrodes is 1.13×10−10 m2/s, and the exchange current density of galena is 0.45 A/m2. Lead diethyldithiocarbamate on the surface of galena is firmly adsorbed.

A new natural product, iso-α-cyclopiazonic acid (1), together with its isomer α-cyclopiazonic acid (2); three mycotoxins: aflatoxin B1 (AFB1) (3), aflatoxin Q1 (AFQ1) (4), and O-methylsterigmatocystin (OMST) (5); two diketopiperazine alkaloids: ditryptophenaline (6) and 3-[(1H-indol-3-yl)methyl]-6-benzylpiperazine-2,5-dione (7), were isolated from the marine-derived fungus Aspergillus flavus. Their structures were determined by analysis of spectroscopic data. The cytotoxicities of compounds 1 and 2 were studied using HL-60, MOLT-4, A-549, and BEL-7402 cell lines.

Three phosphate-solubilizing fungi, identified as Penicillium expansum, Mucor ramosissimus, and Candida krissii, were isolated from phosphate mines (Hubei, People’s Republic of China) and characterized. All the isolates demonstrated diverse levels of phosphate-solubilizing capability in National Botanical Research Institute’s phosphate growth medium containing rock phosphate as sole phosphate source. Acidification of culture medium seemed to be the main mechanism for rock phosphate solubilization. Indeed, citric acid, oxalic acid, and gluconic acid were shown to be present in the culture medium inoculated with these isolates. Moreover, the isolates produced acid and alkaline phosphatases in culture medium, which may also be helpful for RP solubilization. A strong negative correlation between content of soluble phosphorus and pH (r = – 0.89; p < 0.01) in culture medium was observed in this study. All the isolates promoted growth, soil available phosphorus, phosphorus, and nitrogen uptake of wheat seedling in field soil containing rock phosphate under pot culture conditions, thus demonstrating the capability of these isolates to convert insoluble form of phosphorus into plant available form from rock phosphate, and therefore hold great potential for development as biofertilizers to enhance soil fertility and promote plant growth.

The optimization for rock phosphate (RP) solubilization by phosphate-solubilizing fungi ( Candida krissii , Penicillium expansum and Mucor ramosissimus ) isolated from phosphate mines (Hubei, PR China) was investigated. The content of soluble phosphorus (P) released by these isolates was tested on the National Botanical Research Institute's phosphate growth medium (NBRIP) containing RP as sole P source. Results showed that the optimum conditions were at temperature, 32 °C; shaking speed, 160 rpm; RP concentration, 2.5 g l −1 ; EDTA concentration, 0.5 mg ml −1 . The content of soluble P gradually decreased with a larger particle size of RP with all the isolates. The content of soluble P was the highest when the initial pH for RP solubilization was 5.5 in the medium inoculated with C. krissii , which was different from that of 7.0 in the medium inoculated with P. expansum and 7.5 in the medium inoculated with M. ramosissimus . All the isolates showed lower tolerance with increasing concentration of silver ion (Ag + ). Moreover, the highest content of soluble P was achieved when released by C. krissii at 20 of C:N ratio, P. expansum at 40 of C:N ratio and M. ramosissimus at 30 of C:N ratio.

Six new ergosterols, including 3beta-hydroxyl-(22E, 24R)-ergosta-5,8,22-trien-7,15-dione (1), 3beta-hydroxyl-(22E, 24R)-ergosta-5,8,14,22-tetraen-7-one (2), 3beta,15beta-dihydroxyl-(22E, 24R)-ergosta-5,8(14),22-trien-7-one (3), 3beta,15alpha-dihydroxyl-(22E, 24R)-ergosta-5,8(14),22-trien-7-one (4), 3beta-hydroxyl-(22E, 24R)-ergosta-5,8(14),22-trien-7,15-dione (5), and 5alpha,8alpha-epidioxy-23,24(R)-dimethylcholesta-6,9(11),22-trien-3beta-ol (6), have been isolated from the marine-derived fungus Rhizopus sp., along with four known ones (7-10). The structures of the new compounds were determined on the basis of extensive spectroscopic data. All compounds were evaluated for their cytotoxic activities on P388, A549, HL-60, and BEL-7420 cell lines by the MTT and SRB methods.

Blumea balsamifera oil (BBO) is a main extract obtained from Blumea balsamifera (L.) DC (Ainaxiang) leaves, which are widely used as a traditional medicine by the Miao and Li Nations to promote skin trauma or burn injury healing. This study was initiated to investigate the healing efficacy in deep second-degree burn model in rats. The rats were treated by BBO for 21 consecutive days. The rate of healing, scabs dropped time and re-epithelialization time were observed every three days for 21 days after burn injury. The samples were collected from different treated rats by sacrificing the animals on the 1st, 2nd, 5th, 9th, 14th, and 21st day post-burn creation. Then, the water content of burn tissue was measured. Plasma interleukin-1 (IL-1) and tumor necrosis factor-alpha (TNF-α) levels were evaluated, and the tissue expressions of basic fibroblast growth factor (bFGF), vascular endothelial growth factor (VEGF), and transforming growth factor-beta (TGF-β) were determined along with skin histopathology. The results showed that the water content of tissue was significantly reduced, the scabs dropped time shortened, and healing accelerated after treatment with BBO in the burn injury rats. Furthermore, the expressions of growth factors were significantly increased in the tissue, however, the levels of inflammatory factors on plasma decreased. This study confirms the efficacy of BBO consumption on burn injuries.

Tissue culture, SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and spectra analysis were used for studying the aggregation mechanism of protoplasts from Bryopsis hypnoides Lamouroux and the discrepancy between the protoplast-regenerated plants and the wild type. The aggregation of protoplasts from B. hypnoides was observed in natural seawater and artificial seawater with different pH values, and the location and mechanism of the materials causing the aggregation were also studied. Results showed that the protoplasts could aggregate into some viable spheres in natural seawater and subsequently grow into mature individuals. Aggregation of the protoplasts depended exclusively upon the pH value (6-11), and the protoplasts aggregated best at pH 8-9. Some of the extruded protoplasts were separated into two parts by centrifugation: the pellet (PO) and the supernatant (PL). The PO could aggregate in artificial seawater (pH 8.3) but not in PL. No aggregation was found in PO cultured in natural seawater containing nigericin, which can dissipate the proton gradients across the membrane. These experiments suggest that the aggregation of protoplasts is proton-gradient dependent and the materials causing the aggregation were not in the vacuolar sap, but located on the surface or inside the organelles. Furthermore, the transfer of the materials across the membrane was similar to Delta pH-based translocation (Delta pH/TAT) pathway that occurs in the chloroplasts of higher plants and bacteria. Obvious discrepancies in both the total soluble proteins and the ratio of chlorophyll a to chlorophyll b between the regenerated B. hypnoides and the wild type were found, which may be related to the exchange of genetic material during aggregation of the organelles. In the process of development, diatom Amphora coffeaeformis Agardh attached to the protoplast aggregations, retarding their further development, and once they were removed, the aggregations immediately germinated, which showed that diatoms can affect the development of other algae.

A new flavanone glycoside, (2S)5,7,4'-trihydroxyflavanone-8-C-beta-D-(6aEuro(3)-O-acetyl)glucopyranoside (1), together with six known flavonoids, isohemiphloin (2), vitexin (3), cirsimaritin (4), hispidulin (5), apigenin (6), and eupatorin (7), was isolated from the leaves and stems of Abrus precatorius. Their structures were elucidated on the basis of physical and spectral analysis. Rotamers exist for compounds 1, 2, and 3. Compounds 1-3, 6, and 7 were isolated from this plant for the first time.

A new kind of polyacrylamide derivate, hydroximic polyacrylamide (HPAM), was prepared from polyacrylamide (PAM) with hydroxylamine hydrochloride in KOH/alcohol solution. Based on colorimetric method in the measurement of C7–9 alkyl hydroximic acid, the substitution degree of HPAM was determined by selecting octyl hydroximic acid as a primary standard substance. The effects of temperature, the amount of hydroxylamine and potassium hydroxide, and the reaction time on the reaction are discussed. HPAM with 5.78% substitution of degree was acquired by optimizing the reaction conditions. IR spectra revealed that tautomerism appears in the HPAM, and that HPAM exists in both the forms of hydroxamic type and hydroximic type, and the latter type can easily lead to the forming of intramolecular hydrogen bond.

Oxidized low‑density lipoprotein (ox‑LDL) can increase the expression of adipophilin and the accumulation of intracellular lipid droplets. However, the detailed mechanisms remain to be fully elucidated. The present study aimed to investigate the mechanism underlying the effect of ox‑LDL on the expression of adipophilin and the accumulation of intracellular cholesterol esters. The results revealed that ox‑LDL increased the activation of protein kinase C α (PKCα), expression of adipophilin and acyl‑coenzymeA: cholesterol acyltransferse 1 (ACAT1) and increased accumulation of intracellular cholesterol esters. In addition, PKCα siRNA abrogated ox‑LDL‑induced adipophilin, expression of ATAC1 and accumulation of cholesterol esters. Furthermore, ox‑LDL increased the accumulation of intracellular cholesterol esters and expression of ACAT1, and this effect were reversed by transfection with adipophilin siRNA. Taken together, these results demonstrated that ox‑LDL induces the accumulation of cholesterol esters, which is mediated by the PKCα‑adipophilin‑ACAT1 pathway.

FGF21, a member of the fibroblast growth factor superfamily, is an important endogenous regulator of systemic glucose and lipid metabolism. Elevated serum FGF21 levels have been reported in subjects with coronary heart disease and carotid artery plaques. However, whether FGF21 is associated with atherosclerotic diseases remains unclear. In this study, the effects of FGF21 on cholesterol efflux in THP1 macrophage-derived foam cells and the underlying mechanisms were investigated. THP1 macrophage-derived foam cells were incubated with 0, 25, 50, 100, 200, and 400 ng/mL of FGF21 for varying time periods (0, 6, 12, and 24 h). Cholesterol efflux onto apoA-1 was assessed by high-performance liquid chromatography assays, while change in ABCA1 expression was analyzed by western blot and real-time quantitative PCR. Incubation was performed with the ERK1/2-specific inhibitor PD98059, PPARγ-specific inhibitor GW9662, and LXRα siRNA. Our results show that FGF21 promotes cholesterol efflux and ABCA1 expression in THP1 macrophage-derived foam cells in a dose- and time-dependent manner. In addition, inhibition of ERK1/2 or PPARγ, or knockdown of LXRα attenuated FGF21-mediated promotion of ABCA1 expression and cholesterol efflux. These results demonstrate that FGF21 can promote cholesterol efflux by upregulating ABCA1 through the ERK1/2-PPARγ-LXRα pathway in THP1 macrophage-derived foam cells.

We report here for the first time, preparation of the ilmenite titanate MgTiO 3 thin films on Si and SrTiO 3 substrates by atmospheric pressure metalorganic chemical vapor deposition (APMOCVD) technique. Magnesium acetylacetonate [Mg(acac) 2 ] and titanium isopropoxide [TIP] were used as the Mg and Ti precursors, and O 2 was the oxidizing gas. The deposited films were investigated using scanning electron microscopy, X-ray energy dispersion analysis and X-ray diffraction technique. The experimental results showed that the films deposited on silicon substrate were single-phase randomly oriented MgTiO 3 , and on SrTiO 3 substrate, only a (0 1 2) orientation was observed. The MgTiO 3 films, prepared on SrTiO 3 (1 0 0) substrates at a growth temperature of 650°C, were transparent in the visible and exhibited a sharp absorption edge at 380 nm in the ultraviolet. The birefringence of the MgTiO 3 films was also measured at room temperature by the cross-correlation method.

A Gram-stain-positive actinobacterium, designated strain SCSIO 15020T, was isolated from sediment of the South China Sea, and characterized by using a polyphasic approach. The temperature range for growth was 24–60 °C, with optimal growth occurring at 50 °C. The pH range for growth was 6–10 (optimum pH 8–9). The NaCl concentration range for growth was 0–5 % (w/v). The peptidoglycan type was A4α. Polar lipids contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannoside and an unknown polar lipid. The major menaquinone was MK-8(H4); MK-7(H4) was present as a minor component. The major fatty acids (>5 %) were anteiso-C15 : 0, iso-C15 : 0 and iso-C16 : 0. The DNA G+C content of strain SCSIO 15020T was 73.2 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain SCSIO 15020T belonged to the genus Georgenia , with the closest neighbours being Georgenia muralis 1A-CT (96.3 % similarity), Georgenia thermotolerans TT02-04T (95.7 %) and Georgenia ruanii YIM 004T (95.6 %). Based on evidence from the present polyphasic study, strain SCSIO 15020T is considered to represent a novel species of the genus Georgenia , for which the name Georgenia sediminis sp. nov. is proposed. The type strain is SCSIO 15020T ( = DSM 25884T = NBRC 108941T).
