It has long been proposed that nuclear RNAs might play an important role in organizing the structure of the nucleus. Initial experiments performed more than 30 years ago found that global disruption of RNA led to visible rearrangements of nuclear organization. Yet, this idea remained controversial for many years, in large part because it was unclear what specific RNAs might be involved, and which specific nuclear structures might be dependent on RNA. Over the past few years, the contributions of RNA to organizing nuclear structures have become clearer with the discovery that many nuclear bodies are enriched for specific noncoding RNAs (ncRNAs); in specific cases, ncRNAs have been shown to be essential for establishment and maintenance of these nuclear structures. More recently, many different ncRNAs have been shown to play critical roles in initiating the three-dimensional (3D) spatial organization of DNA, RNA, and protein molecules in the nucleus. These examples, combined with global imaging and genomic experiments, have begun to paint a picture of a broader role for RNA in nuclear organization and to uncover a unifying mechanism that may explain why RNA is a uniquely suited molecule for this role. In this review, we provide an overview of the history of RNA and nuclear structure and discuss key examples of RNA-mediated bodies, the global roles of ncRNAs in shaping nuclear structure, and emerging insights into mechanisms of RNA-mediated nuclear organization.

Occurrence of biogenic amines in fresh fish and fishery products constitute a significant safety concern. Ingestion of histamine is identified as the causative agent of several food poisoning incidences on an annual basis. In addition, cadaverine, putrescine and tyramine have been recognized as potentiators of histamine poisoning. Accumulation of biogenic amines in fresh fish and fishery products has been mainly attributed to growth of bacteria possessing amino acid decarboxylase activity, which is facilitated by lack of hygienic conditions and strict temperature control during their storage. To this end, the effectiveness of traditional and modern approaches to control biogenic amine accumulation has been in the epicenter of intensive study. The aim of the present review article is to update and integrate current knowledge regarding the biogenic amine content of fresh fish and fishery products as well as the capacity of traditional and emerging control strategies.

Despite wide applications of noble metal-based catalysts in 5-hydroxymethylfurfural (HMF) oxidation, promoting the catalytic performance at low loading amounts still remains a significant challenge. Herein, a series of metal oxide modified MOx-Au/TiO2 (M = Fe, Co, Ni) catalysts with low Au loading amount of 0.5 wt% were synthesized. Addition of transition metal oxides promotes electron transfer and generation of the Auδ−–Ov–Ti3+ interface. A combination study reveals that the dual-active site (Auδ−-Ov-Ti3+) governs the catalytic performance of the rate-determining step, namely hydroxyl group oxidation. Auδ− site facilitates chemisorption and activation of O2 molecules. At the same time, Ov-Ti3+ site acts as the role of “killing two birds with one stone”: enhancing adsorption of both reactants, accelerating the activation and dissociation of H2O, and facilitating activation of the adsorbed O2. Besides, superoxide radicals instead of base is the active oxygen species during the rate-determining step. On this basis, a FDCA yield of 71.2% was achieved under base-free conditions, complying with the “green chemistry” principle. This work provides a new strategy for the transition metal oxides modification of Au-based catalysts, which would be constructive for the rational design of other heterogeneous catalysts.

Background Ibrutinib is an oral inhibitor of Bruton’s tyrosine kinase (BTK) that is FDA-approved for several lymphoproliferative disorders and chronic GVHD. Objective To characterize cutaneous eruptions arising from ibrutinib and highlight overlap with epidermal growth factor receptor inhibitor (EGFRi)-induced dermatologic adverse events (dAEs). Methods Single-center retrospective cohort of patients referred to the Skin Toxicities Program for management of cutaneous eruptions while taking ibrutinib. Results Among 19 patients, cutaneous eruptions manifested as facial-predominant papulopustular eruptions, petechiae or ecchymoses, photosensitivity, panniculitis, xerosis, and clinical staphylococcal overgrowth. The majority of patients were able to continue ibrutinib therapy with focused management of their cutaneous toxicities. Limitations This study represents cases at a single tertiary care center and is limited to patients referred for toxicity. Conclusions With the exception of petechiae, the cutaneous toxicities of ibrutinib overlap with those associated with selective EGFR inhibitors. We observed that these reactions can be successfully managed using approaches for EGFR inhibitor-induced cutaneous adverse events.

The study was conducted to evaluate the effect of Moringa olifera on the growth and gut health of Tilapia (Oreochromis niloticus). The feed having 30% crude protein was prepared as an experimental diet with 4%, 8% and 10% M. olifera leaf supplementation, respectively. The control diet was devoid of M. olifera leaves. The 10 weeks feeding trial was carried out on 60 fish in aquaria. Fish was fed @ 3% of body weight twice a day. Diet with the high level of inclusion of M. olifera leaves significantly increased the growth rate, Survival Rate (SR), Specific Growth Rate (SGR) and Feed Conversion Efficiency (FCE) in all treatment groups compared to the control group. Similarly, Feed Conversion Ratio (FCR) gradually decreased and found highly-significant. To check the gut health of the Tilapia, random samples were selected and dissected. Nutrient agar was used as culture media to check the growth of bacteria. Pour Plate Method was used for viable colonies count by colony counter. Through staining method, the different bacteria such as Escherichia coli, Salmonella, Shigella and Pseudomonas aeruginosa were identify abundantly in the intestine of control diet fish but less number present in treatment diets groups. These results showed that M. olifera leaves up to 10% of dietary protein can be used for Nile tilapia for significant growth and healthy gut microbiota of fish. Resumo O estudo foi conduzido para avaliar o efeito da Moringa olifera no crescimento e saúde intestinal da tilápia (Oreochromis niloticus). A ração com 30% de proteína bruta foi preparada como dieta experimental com 4%, 8% e 10% de suplementação de folhas de M. olifera, respectivamente. A dieta controle foi desprovida de folhas de M. olifera. O ensaio de alimentação de 10 semanas foi realizado em 60 peixes em aquários. O peixe pesava 3% do peso corporal duas vezes ao dia. A dieta com alto nível de inclusão de folhas de M. olifera aumentou significativamente a taxa de crescimento, taxa de sobrevivência (SR), taxa de crescimento de sobrevivência (SGR) e eficiência de conversão alimentar (FCE) em todos os grupos de tratamento em comparação com o grupo de controle. Da mesma forma, a taxa de conversão de alimentação (FCR) diminuiu gradualmente e foi considerada altamente significativa. Para verificar a saúde intestinal da tilápia, amostras aleatórias foram selecionadas e dissecadas. O ágar nutriente foi usado como meio de cultura para verificar o crescimento das bactérias. O método da placa de Verter foi usado para a contagem de colônias viáveis por contador de colônias. Através do método de coloração, diferentes como Escherichia coli, Salmonella, Shigella e Pseudomonas aeruginosa foram identificados abundantemente no intestino de peixes da dieta controle, mas em menor número nos grupos de dieta de tratamento. Esses resultados mostraram que M. olifera deixa até 10% da proteína dietética e pode ser usado para tilápia do Nilo para um crescimento significativo e microbiota intestinal saudável de peixes.

The lower lignin content in plants species with energy potential results in easier cellulose breakdown, making glucose available for ethanol generation. However, higher lignin levels can increase resistance to insect attack. The objective of this work was to evaluate the susceptibility of a bmr-6 biomass sorghum (a mutant genotype with a lower concentration of lignin) to important pests of energy sorghum, Diatraea saccharalis and Spodoptera frugiperda. Experiments were performed in the laboratory and greenhouse to evaluate the development of these pests on the biomass sorghum bmr hybrids BR007, BR008, and TX635 and their respective conventional near-isogenic genotypes (without the bmr gene). The lignin content was higher in non-bmr hybrids, but the evaluated insect variables varied between treatments, not being consistent in just one hybrid or because it is bmr or not. The lowest survival of S. frugiperda was observed in the BR008 hybrid, both bmr and non-bmr. The S. frugiperda injury scores on plants in the greenhouse were high (>7) in all treatments. For D. saccharalis, there was no difference in larval survival in the laboratory, but in the greenhouse, the BR007 hybrid, both bmr and non-bmr, provided greater survival. Due the need to diversify the energy matrix and the fact that greater susceptibility of the bmr hybrids to either pests was not found in this study, these results hold promise for cultivation of these biomass sorghum hybrids for the production of biofuels. Resumo O menor teor de lignina em espécies de plantas com potencial energético resulta na maior facilidade de quebra da celulose, disponibilizando glicose para geração de etanol. Porém, maiores teores de lignina representa um fator de resistência ao ataque de insetos. O objetivo deste trabalho foi avaliar como importantes pragas do sorgo energia, Diatraea saccharalis e Spodoptera frugiperda, se comportam quanto à alimentação e desempenho em sorgo bmr-6, um genótipo mutante com menor concentração de lignina. Foram realizados experimentos em laboratório e casa de vegetação, avaliando o desenvolvimento destas pragas nos híbridos de sorgo biomassa bmr 007, 008, TX635 e seus respectivos genótipos isogênicos convencionais (sem o gene bmr). O teor de lignina foi maior nos híbridos não bmr, mas nos parâmetros avaliados nos insetos, houve variação entre os tratamentos, não sendo consistente em apenas um híbrido e nem por ser ou não bmr. A menor sobrevivência de S. frugiperda foi verificada no híbrido BR008 tanto bmr quanto não bmr. As notas de injúria por S. frugiperda no sorgo em casa de vegetação foram altas (>7) em todos os tratamentos. Para D. saccharalis, não houve diferença significativa para a sobrevivência larval em laboratório, mas em casa de vegetação o híbrido BR007 tanto bmr quanto não bmr proporcionaram maior sobrevivência. Diante da necessidade de diversificar a matriz energética e o fato de que não foi comprovada neste estudo maior suscetibilidade dos híbridos bmr a ambas as pragas, estes resultados são promissores para o cultivo desses híbridos de sorgo biomassa para produção de biocombustíveis.

The neuraminidase (NA) of influenza A and B viruses plays a distinct role in viral replication and has a highly conserved catalytic site. Numerous sialic (neuraminic) acid analogs that competitively bind to the NA active site and potently inhibit enzyme activity have been synthesized and tested. Four NA inhibitors are now licensed in various parts of the world (zanamivir, oseltamivir, peramivir, and laninamivir) to treat influenza A and B infections. NA changes, naturally occurring or acquired under selective pressure, have been shown to reduce drug binding, thereby affecting the effectiveness of NA inhibitors. Drug resistance and other drawbacks have prompted the search for the next-generation NA-targeting therapeutics. One of the promising approaches is the identification of monoclonal antibodies (mAbs) targeting the conserved NA epitopes. Anti-NA mAbs demonstrate Fab-based antiviral activity supplemented with Fc-mediated immune effector functions. Antiviral Fc-conjugates offer another cutting-edge strategy that is based on a multimodal mechanism of action. These novel antiviral agents are composed of a small-molecule NA inhibitor and an Fc-region that simultaneously engages the immune system. The significant advancements made in recent years further support the value of NA as an attractive target for the antiviral development.

Root-knot nematode Meloidogyne incognita is among the biotic factors which has greatly affected both the yield and the quality of the tomato crop. The egg parasitic nematode, Purpureocillium lilacinum (Pl) is considered as one of the most promising agents in controlling and overcoming this plant pathogen. The nematicidal effect of the native isolate Pl AUMC 10149 on second stage juvenile’s survival and egg hatching of M. incognita at different times of exposure was tested in vitro. The obtained data showed that Pl gave a maximum percentage of J2 mortality (97.6%) and egg hatching inhibition (79.8%) after 72 hours of exposure. The potentiality of Pl as well as Bio-Nematon to control M. incognita infecting tomato was conducted using different times of application in vivo. Nine treatments with five replicates were used for such bioagents compared with the nematicide Oxamyl. Each seedling was inoculated with 1000 J2s of nematode/pot and 10 mL of Pl (1x1010 CFU/mL) or Bio-Nematon spore suspension (1x108 CFU/mL) 10mL/pot. The results indicated that the most effective treatments in reducing nematode population, number of galls and egg masses of M. incognita in plant roots was performed with treatment by Pl pre-planting and post-infection with Pl (Rf 1.9) giving a significant enhancement in plant length (64.9%), fresh weight (72.52%) and shoot dry weight (163.41%) without negatively impacting environment. Therefore, the present study confirmed that using P. lilacinum AUMC 10149 can be used as a practical supplement to environmentally friendly disease management of root-knot nematodes in Egypt. Resumo O nematoide-das-galhas Meloidogyne incognita está entre os fatores bióticos que afetaram enormemente a produção e a qualidade da cultura do tomate. O nematoide parasita de ovos, Purpureocillium lilacinum (Pl), é considerado um dos mais promissores agentes no controle e superação desse fitopatógeno. O efeito nematicida do isolado nativo Pl AUMC 10149 na sobrevivência de juvenis de segundo estágio e na eclosão dos ovos de M. incognita em diferentes momentos de exposição foi testado in vitro. Os dados obtidos mostraram que o Pl deu um percentual máximo de mortalidade de J2 (97.6%) e inibição da eclosão dos ovos (79.8%) após 72 horas de exposição. A potencialidade de Pl e de Bio-Nematon para controlar M. incognita infectando tomate foi conduzida em diferentes tempos de aplicação in vivo. Nove tratamentos com cinco repetições foram usados para tais bioagentes em comparação com o nematicida Oxamyl. Cada muda foi inoculada com 1.000 J2s de nematoide / vaso e 10 mL de Pl (1×1010 CFU/mL). Ou suspensão de esporos Bio-Nematon (1×108 CFU/mL) 10mL/pot. Os resultados indicaram que os tratamentos mais eficazes na redução da população de nematoides, número de galhas e desovas de M. incognita nas raízes das plantas foram realizados com Pl pré-plantio e pós-infecção com Pl (Rf 1.9), dando um aumento significativo no comprimento da planta (64.9%), massa fresca (72.52%) e massa seca da parte aérea (163.41%) sem impactar negativamente o meio ambiente. Portanto, o presente estudo confirmou que o uso de P. lilacinum AUMC 10149 pode ser usado como um suplemento prático para o manejo ecologicamente correto de nematoides-das-galhas no Egito.

Mammals have a limited capacity to regenerate their tissues and organs. One of the mechanisms associated with natural regeneration is dedifferentiation. Several small molecules such as vitamin C and growth factors could improve reprogramming efficiency. In this study, the NTERA2-D1 (NT2) cells were induced towards differentiation (NT2-RA) with 10-5 M retinoic acid (RA) for three days and then subjected to various amounts of vitreous humor (VH). Results show that the growth rate of these cells was reduced, while this rate was partly restored upon treatment with VH (NT2-RA-VH). Cell cycle analysis with PI method also showed that the numbers of cells at the S phase of the cell cycle in these cells were increased. The levels of SSEA3 and TRA-1-81 antigens in NT2-RA were dropped but they increased in NT2- RA-VH to a level similar to the NT2 cells. The level of SSEA1 had an opposite pattern. Expression of OCT4 gene dropped after RA treatment, but it was recovered in NT2-RA-VH cells. In conclusion, we suggest VH as a potent mixture for improving the cellular reprogramming leading to dedifferentiation. Resumo Os mamíferos têm uma capacidade limitada de regenerar seus tecidos e órgãos. Um dos mecanismos associados à regeneração natural é a desdiferenciação. Várias moléculas pequenas, como vitamina C e fatores de crescimento, podem melhorar a eficiência da reprogramação. Neste estudo, as células NTERA2-D1 (NT2) foram induzidas à diferenciação (NT2-RA) com ácido retinóico (RA) 10-5 M por três dias e depois submetidas a várias quantidades de humor vítreo (VH). Os resultados mostram que a taxa de crescimento dessas células foi reduzida, enquanto essa taxa foi parcialmente restaurada após o tratamento com VH (NT2-RA-VH). A análise do ciclo celular com o método PI também mostrou que o número de células na fase S do ciclo celular nessas células estava aumentado. Os níveis de antígenos SSEA3 e TRA-1-81 em NT2-RA diminuíram, mas aumentaram em NT2-RA-VH a um nível semelhante ao das células NT2. O nível de SSEA1 teve um padrão oposto. A expressão do gene OCT4 diminuiu após o tratamento com AR, mas foi recuperado em células NT2-RA-VH. Em conclusão, sugerimos o VH como uma mistura potente para melhorar a reprogramação celular levando à desdiferenciação.

MiR-202-5p is identified as the maker microRNA (miRNA) of germplasm in fish. However, the role of miR-202-5p in gonadal development is still unclear. Here we investigated the expression patterns of miR-202-5p in Japanese flounder (Paralichthys olivaceus). Real-time PCR displayed that miR-202-5p was specifically expressed in bisexual gonads, and its transcription in the testis was relatively abundant than that in the ovary. Moreover, it was gradually increased in testis from stage II to stage IV. Fluorescent in situ hybridization (FISH) illustrated that miR-202-5p was co-localized with vasa (a marker gene of germ cells) in testis, and it was predominately expressed in spermatogonia and spermatocytes, implying that miR-202-5p might participate in spermatogenesis. Considering the important role of retinoic acid (RA) on regulating meiosis during spermatogenesis, we treated Japanese flounders by injecting all-trans retinoic acid (ATRA). MiR-202-5p in testis was shown to be up-regulated, and ccnd1, a key cell cycle regulator, was found to be down-regulated after ATRA treatment. Furthermore, ccnd1 was demonstrated to be a direct regulatory target of miR-202-5p using Dual-luciferase assay. In the primary testis cell from Japanese flounder, we further confirmed that ccnd1 was down-regulated by miR-202-5p. The results suggest that miR-202-5p exerts a crucial role in gonadal development by inhibiting ccnd1 in Japanese flounder.

Sluggish kinetics of methanol oxidation reaction (MOR) and alkaline hydrogen evolution reaction (HER) even on precious Pt catalyst impede the large-scale commercialization of direct methanol fuel cell (DMFC) and water electrolysis technologies. Since both of MOR and alkaline HER are related to water dissociation reaction (WDR), it is reasonable to invite secondary active sites toward WDR to pair with Pt for boosted MOR and alkaline HER activity on Pt. Mo2C and Ni species are therefore employed to engineer NiPt–Mo2C active site pairs, which can be encapsulated in carbon cages, via an in-situ self-confinement strategy. Mass activity of Pt in NiPt–Mo2C@C toward HER is boosted to 11.3 A/mgPt, 33 times higher than that of Pt/C. Similarly, MOR catalytic activity of Pt in NiPt–Mo2C@C is also improved by 10.5 times and the DMFC maximum power density is hence improved by 9-fold. By considering the great stability, NiPt–Mo2C@C exhibits great practical application potential in DMFCs and water electrolysers.

The studies results influence of the growth regulator Vermiyodis and sowing norms on wintering of winter oilseed rape plants and Mercedes hybrid on sod podzolic soils of the experimental field of Prykarpattia of the State Agricultural Experimental Station of the NASG region are presented. Studies have shown that the variants with the pre-sowing treatment off the winter variety Cheremosh seeds was carried out, on average over the years of research, at the sowing rate of 0600000 / ha plant death during overwintering was 4.7-5.7 pcs / m2 or 9, 5-10 , 8%, at a sowing rate of 0800000 / ha of germinating seeds - 5.7- 6.2 pieces / m2 or 8.5-9.3%, at a sowing rate of 1.0 million / ha of similar seeds - 8.7-9 , 3 pcs / m2 or 10.1- 10.9%. In the variants where the pre-sowing treatment of seeds was carried out with the Vermiyodis regulator at a dose of 5 l / t at a sowing rate of 08mln / ha, the plant death was 4.6-5.6%, which is 2.8-3.0% less than the norm sowing 1.0 million / ha of germinating seeds. In the variants where the pre-sowing treatment of seeds of the winter hybrid Mercedes was carried out with the growth regulator "Vermiodis" 5 l / t, the percentage of dead plants was 4.8-5.6% at the sowing rate of 0600000 / ha of germinating seeds, at the sowing rate of 0.8 million / ha - 6.1-7.3%, with a sowing rate of 1.0 million / ha of germinating seeds 8.1-8.6%, which is significantly less than the control and options, where presowing seed treatment was carried out with the growth regulator "Vermijodis". Studies have shown that the best overwintering, on average over the years of research, was in the rapeseed variety of winter hybrid Mercedes at a sowing rate of 0600000 / ha of germinating seeds, in the variety Cheremosh at a sowing rate of 0800000 / ha of germinating seeds for pre-sowing seed treatment.

IGS is abundant in polymorphism, which is widely used in the analysis of intraspecific genetic diversity and phylogenetic relationships among geographical populations. In this study, the 45S rDNA repeat unit of Saccharina japonica was obtained for the first time by BAC clone sequencing. The total length of 45S rDNA repeat unit of S. japonica was 8995 bp, including 5420 bp of 18s-5.8s-25s rDNA and 3575 bp of IGS (Intergenic Spacer), with the GC content of 51.4%. IGS was composed of 465 bp 3’-outer transcribed spacer (ETS), 874 bp 5’-ETS, and 2236 bp non transcribed spacer (NTS), with the GC content of 50.1%.Fiber-FISH (fiber-fluorescence in situ hybridization, fiber-FISH) analysis of 45S rDNA on the BAC molecule of female gametophytes of S. japonica illustrated that each fiber had at least five continuous moniliform hybridization signal points, indicating the distribution of 45S rDNA repeat unit on the bacterial artificial chromosome. This study provided a new candidate molecular marker for detecting intraspecific polymorphisms of S. japonica, and the successful fiber-FISH analysis of 45S rDNA on BAC molecule would contribute to the construction of the physical map and Map-based cloning of this kelp.

An 8-week feeding trial investigated the optimum dietary vitamin K3 requirements of largemouth bass. A total of 600 healthy fish (12.96 ± 0.07 g) were fed diets containing varying levels of vitamin K3 at 0.78 (K0), 5.80 (K5), 10.82 (K10), 15.84 (K15), and 20.85 (K20) mg/kg with four replicates per level. Results showed that dietary vitamin K3 enhanced growth, with weight gain rate and specific growth rate in K15 trials significantly higher than K0 trials (P 0.05). However, the supplementation of vitamin K3 significantly contributed to calcium accumulation in muscle tissues and vertebrae (P 0.05). The antioxidant capacity was improved with the addition of vitamin K3, which was inconsistent with the changes in malondialdehyde and MK-4 (a form of menaquinone) within liver and serum, suggesting that the toxicity of excessive artificial vitamin K3 may account for this difference. Using regression analysis, the appropriate dietary vitamin K3 levels within a largemouth bass diet was determined to be in the range of 9.93–15.22 mg/kg, which will provide a reference for the preparation of vitamin premix and artificial diet for largemouth bass.

Climate change has had large impacts on marine animals, including neon flying squid Ommastrephes bartramii (O. bartramii) in the North Pacific Ocean. O. bartramii statoliths from 2012, 2015, and 2016 were used to evaluate the variations in life cycle events. The relationship between mantle length and body weight showed significant differences between years and gender. The oldest squid was collected in 2016 at 271 days old, further proving that O. bartramii has nearly a 1-year life span. The hatching season ranged from December to May and the peak hatching time in 2015 was one-half month later than in 2012 and 2016. Growth rates varied with environmental factors such as sea surface temperature (SST) and chlorophyll a concentration (chl. a), indicating that higher SST and chl. a concentrations led to faster growth. An extreme El Nino event with lower SST in 2015 also led to younger age class and slower growth rates. The occurrence of differences in body size and growth rates between years, caused by the interannual variations of environmental factors, makes it necessary to use separate growth curves for different years when analyzing North Pacific O. Bartramii populations.

DNA introgressed from one species into another is typically deleterious at many genomic loci in the recipient species. It is therefore purged by selection over time. Here, we use mathematical modeling and whole-genome simulations to study the influence of recombination on the purging of introgressed DNA. We find that aggregate recombination controls the genome-wide rate of purging in the first few generations after admixture, when purging is most rapid. Aggregate recombination is quantified by, the average recombination rate across all locus pairs, and analogous metrics. It is influenced by the number of crossovers (i.e., the map length) and their locations along chromosomes, and by the number of chromosomes and heterogeneity in their size. A comparative prediction of our analysis is that species with fewer chromosomes should purge introgressed DNA more profoundly, and therefore should exhibit a weaker genomic signal of historical introgression. With regard to patterns across the genome, we show that, in heterogametic species with autosomal recombination in both sexes, more purging is expected on sex chromosomes than on autosomes, all else equal. The opposite prediction holds for species without autosomal recombination in the heterogametic sex. Finally, we show that positive genomic correlations between local recombination rate and introgressed ancestry, as recently observed in several taxa, are likely driven not by recombination’s effect in unlinking neutral from deleterious introgressed alleles, but rather by its effect on the rate of purging of the deleterious alleles themselves.Note on this versionAn earlier version of this manuscript had two parts: (1) Calculations of the variance of genetic relatedness between individuals with particular pedigree relationships, taking into account the randomness of recombination and segregation in their pedigree. Mosquitoes serve as the major vector transmitting malaria, dengue, yellow fever and several other diseases of human concern. Rising in mosquito-borne diseases and consequent fatalities throughout the world has made the management of mosquitoes of paramount importance. With the use of various insecticidal agents and their indiscriminate application in the fields for vector control; other issues such as multiple insecticide resistance, lethality to non-specific targets and adverse effects on human and environmental health have emerged making the situation more critical. Hence, the focus of researchers has diverted to the use of Insect Growth Regulators (IGRs) that affect the growth and development of the insects without inducing any appreciable toxic effects. The paper comprehensively reviews various IGRs and their potential use against insect pests and mosquito vectors. A special emphasis has been laid on the utilization of diflubenzuron, its larvicidal potency and growth regulatory effects against mosquitoes. The paper also delivers a detailed discussion on various approaches governing with the application of diflubenzuron, a chitin synthesis inhibitor, for its potent effects over a wide range of other insect species, low toxicity to humans, safety to other non-target animals, negligible deleterious environmental impact along with the possible development of resistance in the mosquitoes, thereby providing insights and the direction for the future in terms of the innovative and technological perspective. Keeping in view the role of multifarious mechanisms in the development of resistance; use of various synergistic compounds, such as hydrolase inhibitors - profenofos and S,S,S-tributyl phosphorotrithioate; glutathione S-transferase inhibitor – diethylmaleate; and oxidase inhibitor - piperonyl butoxide (PBO); has been recommended in combination with IGRs to enhance their efficacy, and reduce or reverse the resistance in target mosquitoes. Another compound, verapamil, has been found extremely efficient in imparting synergistic effect to diflubenzuron by inhibiting P-glycoproteins, a transporter of the insecticides causing their efflux from the cell. Recommendations have been made for safe and effective mosquito control measures, adequate policies and increased awareness about the mosquito-borne diseases among the masses. In addition, regular surveillance of mosquitoes is endorsed for the formulation of an efficient mosquito management strategy.

A study was carried out at CSIR-CIMAP, RC. Hyderabad (India) to study the impact of coppicing time on the oil percent and its chemical composition of different cultivars of lemongrass using a randomized block design. The freshly harvested herb was used for the hydro-distillation for essential oil, and subsequent chemical analysis. The chemical components identification of the essential oil was performed by GC–MS. In the results coppicing time influenced significant changes in the essential oil content, essential oil yield, and its chemical composition especially, total citral, and geraniol content. The four top rankers varieties/cultivars of lemongrass for the essential oil yield were CIM-Shikhar > Krishna > CKP-25 > CIMAP-Suwarna; for the essential oil content CIM–Shikhar > Krishna > CIMAP-Suwarna > Praman; for total citral content Nima > CIM–Shikhar > Krishna > Pragati, and for the total geraniol content Himrosa > CN-5 > Tawirosa > Kalam, respectively. Therefore, we can recommend these varieties/cultivars for commercial cultivation in large areas under the semi-arid region of south India.

Abstract. Carotenoid intake is associated with low mortality and cancer risks; data on non-provitamin carotenoid intake is limited especially in Asians. We aimed to estimate carotenoid intake in Japanese adult women. Carotenoid content database comprises 196 food items, including 39 fruits, 87 vegetables and mushrooms, and 11 seaweeds, and was established using data from the literature and analyses of foods available in Japan. We surveyed the intake of these foods in Japanese women aged 21–56 years (n=109). Total intake of 7 carotenoids (mean±SD [range]) was 7,450±3,840 (1,160–21,300) μg/day; α-carotene, β-carotene, β-cryptoxanthin, lutein, zeaxanthin, lycopene, and fucoxanthin represented 4.3%, 23%, 3.4%, 15%, 2.0%, 39%, and 13% of total intake, respectively. Lutein intake was 1,132±686 (294–3,490) μg/day; its best sources were spinach, cucumber, chicken egg, green onion, and Chinese chives, representing 51% of total intake. Lutein can be obtained from a variety of sources. Thus, lutein intake levels did not vary widely among individuals and very few individuals consumed insufficient levels of lutein. Intake of zeaxanthin, lycopene, and fucoxanthin was 149±93 (2–479), 2,890±2,970 (0–17,100), and 980±1,230 (0–5,660) μg/day, respectively. Their intake required rich sources including chicken egg for zeaxanthin (52%); tomato products for lycopene (98%), and wakame seaweed for fucoxanthin (76%). The carotenoid content database including all food items consumed in Japan will be helpful for further investigations on carotenoid intake and its health benefits.

Fluoride contamination in groundwater is a major concern in many parts of India and all over the world. Researches paying attention for the removal of fluoride through the application of biosorbents prepared from different parts of plants are finding greater scope and importance. The present research work focuses on Senna auriculata L., flower petal biomass as biosorbent, and evaluated its feasibility for fluoride ion elimination from aqueous solutions. Batch experiments were conducted to remove fluoride under different experimental conditions have been optimized for the maximum removal of fluoride; 80% removal was observed at pH: 6, sorbent dosage: 0.25 g/100 mL, time of agitation: 90 min, and initial concentration of the fluoride ions: 5 mg/L. Characterization studies of the biosorbent revealed its favorability towards the sorption of fluoride. In the isothermal modeling studies, Langmuir isotherm model was obeyed by the biosorption process with R2 value of 0.98 and from a kinetic perspective, the biosorption of fluoride onto the biosorbent observed the pseudo-second-order reaction with R2 value of 0.98. The developed biosorbent has been applied to real field fluoride-contaminated water samples and found to be successful.

The changed phenolic acids (PAs) allelochemicals exuded by the roots induced by interspecific interactions is related to intercropping alleviates soil-borne disease. However, the presence of PAs in roots and root exudations and their rhizodeposition under intercropping are still unclear. Hydroponic and soil experiments of wheat, faba bean, and wheat intercropped with faba bean were conducted, and the major compositions and contents of PAs in roots, root exudations, and rhizospheric soil were determined. The results showed that ρ-hydroxybenzoic, vanillic, and syringic acids were the major components of PAs in roots, root exudations, and rhizospheric soil in a wheat and faba bean intercropping system. The compositions and percentages of PAs in roots of faba bean were altered when faba bean intercropped with wheat. The total exudation rate of PAs in root exudations was decreased by 30%–60% under the wheat and faba bean intercropping (W//F) system as compared to mono-cropped faba bean (MF). ρ-hydroxybenzoic acid was identified in the root exudation of both MF and mono-cropped wheat (MW), but not detected in the intercropping on 60 days after transplanting. Vanillic acid was only detected in the root exudation of MF on 30 days after transplanting. The rhizodepostion of vanillic and cumaric acid were decreased at both branching and pod setting stages in W//F as compared to MF. In conclusion, interspecific interaction changed the compositions and contents of PAs in faba bean roots and root exudations. W//F constrained vanillic acid exuded by roots and decreased vanillic and coumaric acid rhizodeposition by faba bean, which provides insight into root-soil interactions in the intercropping systems.

Introduction: A recent law (DCTO-2020-883-APN-PTE-Law No. 27,350. Regulation) passed in Argentina put an end to the ban imposed for the last 60 years on cannabis cultivation within the country. The law permits restricted access to cannabis derivatives for medicinal, therapeutic, and palliative use by individuals and communities, allowing self- and community-based cannabis production. This is cause for concern in view of the lack of quality controls for cannabis derivatives. The several varieties of cannabis grown in Argentina have different chemical profiles and are processed in a variety of ways-mostly by alcohol extraction or maceration at different temperatures and for different amounts of times-making the cannabinoid content of these preparations highly variable. Determining the characteristics of home- and community-grown cannabis products will facilitate the implementation of public policies conducive to their safety and improvement. Objective: The aim of this study was to determine the cannabinoid chemotypes used for therapeutic purposes in Argentina and evaluate whether the cannabinoids present in homemade derivatives are comparable to those in commercially available products. Materials and Methods: High performance liquid chromatography with ultraviolet and diode array detector (HPLC/UV-DAD) analysis of 436 samples (oils, resins, and inflorescences) was carried out to determine the identity and concentration of five cannabinoids: tetrahydrocannabinolic acid (THCA), tetrahydrocannabinol (THC), cannabidiolic acid (CBDA), cannabidiol (CBD), and cannabinol (CBN). From three different sources, the samples represent the type of medical cannabis preparations to which patients have access. Results: The results indicate that the medium-to-low cannabinoid concentration in a significant number of homemade oil samples is similar to that found in commercial products. Most of the samples have a THC/CBD ratio >1 or only contain THC. Acidic cannabinoids were detected in homemade preparations, but were not reported in package inserts of commercial products. Conclusions: Our results indicate that despite their considerable variability, homemade preparations as a whole show cannabinoid levels and profiles equivalent to the commercially available products commonly used for medicinal, therapeutic, and palliative purposes in Argentina.

This study was conducted to determine the phenolic compounds that may be utilized as chemotaxonomic study to identify some phenolic compounds concentration that are useful for pharmaceutical purposes from 5 taxa belongs to the Brassicaceae family which are; Cardaria draba (L.) Desv., Lepidum aucheri Boiss., Sisymbrium irio L., Sisymbrium officinale (L.) Scop., and Sisymbrium septulatum DC. Leaves powder of these taxa were collected and analyzed by High-performance liquid chromatography (HPLC). The results identified 9 phenolic compounds, chlorogenic acid, cinnamic acid, ferulic acid, genstic acid, kaempferol, p-coumaric acid, protocatechuic acid, sinapic acid, and vanillic acid. The results showed that the genus Lepidium aucheri had the highest concentration of 4 phenolic compounds, cinnamic acid, genstic acid, p-coumaric acid, and sinapic acid with concentration of (37.2, 80.79, 62.91, and 37.10µg/ ml) respectively. On the other hand, the genus Cardaria draba showed the lowest concentration of 3 phenolic compounds, cinnamic acid, p- coumaric acid, and protocatechuic acid, and doesn't have 2 phenolic compounds, genstic acid and kaempferol. This variation in the phenolic concentrations might be due to the genotype and plant growth stage differences. Increasing fuel prices and depletion of conventional sources of energy has promoted research in non-conventional sources like biodiesel from non-edible seeds and industrial wastes. The present work focuses on the elicitation of biodiesel from the seeds of capparis spinosa L. The crude bio-oil extracted from the seeds of capparis spinosa L transesterified into biodiesel using calcium oxide as a heterogeneous catalyst derived from oyster shells. The oyster shell powder is characterized before and after calcination using XRD and the calcinated oyster shell is observed through SEM to analyze the size of the particle and morphology. The produced biodiesel was characterized by GC analysis and the fuel properties of biodiesel such as density, flash point, cloud point, pour point and kinematic viscosity, ash content, carbon residue was analyzed and compared with commercial diesel.

Black rice (Zizania aqatica) is famous for its high nutritional value. Other names given for this rice are forbidden rice, emperor’s rice and royal’s rice. Mainly this rice is originated from Asian countries. The richest country in cultivation of black rice is China (62%) followed by Sri Lanka (8.6 %), Indonesia (6.2%) and India holds the 4th placed with (5.1%) follows by different countries. It contains a high amount of antioxidant which is known as anthocyanin and tocopherols (which is one of the most powerful antioxidants and it is also known as vitamin E). Foods which consist of antioxidant are black in colour or dark purple. It is also rich in fibre, protein, carbohydrates, potassium and B vitamins respectively. Activity of “free radicals” in our body is naturally dominated by another group of chemical compounds called ‘antioxidant’ which are present in our regularly consumed food. Therefore, consuming of black rice in your regular diet can prevent many non-communicable diseases (NCD) like occurrence of cancer cells, atherosclerosis, hypertension, diabetes, osteoporosis, asthma, digestive health, reduces stroke risk level in women. Black rice is also use in developing a new novel food product.

Malaria, Zika virus, West Nile virus, Dengue fever, and Lyme disease are common causes of morbidity and mortality around the world. While arthropod bites may cause local inflammation and discomfort, a greater concern is the potential to develop deadly systemic infection. The use of insect repellents (IR) to prevent systemic infections constitutes a fundamental public health effort. Cost-effectiveness, availability, and high-efficacy against arthropod vectors are key characteristics of an ideal IR. Currently, numerous IRs are available on the market, with DEET (N,N-diethyl-3-methylbenzamide) being the most widely used. DEET has an excellent safety profile and remarkable protection against mosquitoes and various other arthropods. Other EPA-registered IR ingredients (permethrin, picaridin, IR3535, oil of lemon eucalyptus, oil of citronella, catnip oil, and 2-undecanone) are alternative IRs of great interest due to some having efficacies comparable to that of DEET. These alternative IRs possess low toxicity and favorable customer experiences in utilization (e.g., cosmetically pleasant, naturally occurring). This review summarizes currently available EPA-registered IRs: the origins, mechanisms of action, side effect profiles, and available formulations will be discussed. This review will enable the clinician to select the best IR option to meet patients' needs and provide the greatest protection from arthropod bites and sequelae.

India is an agricultural country. Nearly 70% of the populations thrive in rural areas, engaged in agriculture making the backbone of our economy. The Trichoderma genus is widely distributed and provides an alternative to chemical pesticides in crops and plants growth. Trichoderma species are free living and commonly found in soil and root ecosystems. Root colonization by Trichoderma also regularly promotes growth and development of plants. They also perform beneficial and significant role in enhancing crop productivity, fight against pathogens, resistant to abiotic stress and increase of nutrients uptake from soil. The present study was undertaken to investigate quantitative and qualitative estimation of phosphate solubilizer by using nineteen isolates of Trichoderma grown on Pikovskaya’s Agar and NBRIP media. It was found that all isolates were capable of differentially utilizing 5 g/L TCP in NBRIP broth. This was indicated by the soluble phosphate concentrations and increase in acidity of growth medium. Out of 19 isolates, all isolates were capable in phosphate solubilization. However, the P- solubilizing efficiency was found higher in case of the isolate number-59 and 89. While isolate number-102 was the strongest and isolate number-23 was the weakest P-solubilizer. Minimum Phosphate solubilization concentration was observed in 3rd day (0%) that simultaneously increased on 7th day (52.63%) followed by 9th day (100%) and remaining all days 13th, 17th and 21st days also observed 100% efficiency.

Microalgae-bacteria symbiotic association can provide many competitive advantages over the pure axenic culture of microalgae to obtain biofuel. This system significantly reduces an important technical constraint, including associated capital cost with the infrastructure required for maintaining pure microalgae culture. The current study aims to compare the symbiotic growth of Tetraselmis indica - Pseudomonas aeruginosa consortium with axenic growth of Tetraselmis indica in dairy wastewater (DWW). The extraction and optimization of chlorophyll-a were also investigated. The overall maximum chlorophyll-a recovery of 29.106 ± 0.045 mg L−1 was obtained through the sonification as extractant method with methanol as an extraction solvent. The chlorophyll-a content of consortium cultivated in DWW higher than the axenic pure microalgae culture (APMC). The maximum biomass yield of the consortium was achieved as 1454.88 mg L−1, which is much higher than APMC growth in DWW by 38.80%. On the 10th day, the consortium removed 87.49%, 83.76%, and 79.83% of chemical oxygen demand (COD), total dissolved nitrogen (TDN), and total dissolved phosphorus respectively. The higher biomass growth and wastewater nutrient removal exhibited by the symbiotic microalgae-bacteria consortium can be utilized in biofuel technology.

Endocrine disrupting compounds and in particular estrogenic substances have the ability to interact with the hormone system of organisms. Among them are not only synthetic but also natural substances that potentially stress the aquatic ecosystem. High human population densities such as around the Pearl River Estuary (PRE) are suspected of exerting significant anthropogenic pressure onto coastal areas. In this study, natural and synthetic estrogens as well as estrogen-like substances derived from plants and fungi were investigated in the PRE and at the adjacent northern shelf of the South China Sea. Maximum concentration of 3.6 ng L−1 for estrone (E1), 0.7 ng L−1 for 17α-ethinylestradiol (EE2), 12.9 ng L−1 for genistein (GEN), 11.9 ng L−1 for daidzein (DAI) and 1.9 ng L−1 for zearalenone (ZEN) were observed. While E1 and EE2 were detected in fresh and saltwater samples, GEN, DAI and ZEN were observed only at freshwater sampling sites. During the investigations, the analysis of 17β-estradiol (E2) and EE2 indicated a strong matrix dependence. Additionally, an estrogen screen observation showed estrogenic activity in form of estradiol equivalent quotients up to 0.18 ng L−1.

Late Quaternary carbonate dissolution was evaluated by using planktonic foraminifera indices perfect test ratio (PTR) of Goborotalia menardii, G. menardii fragmentation index (MFI), percentage of total resistant species (RSP) and sediment calcium carbonate (CaCO3) content in five sediment cores collected from two diverse oceanographic settings of the Indian Ocean. The cores collected from a depth range of 2000 and 4250 m of the Andaman Sea, a marginal sea in the Northern Indian Ocean and a seamount rise in the Central Indian Basin (CIB) in the abyssal Indian Ocean depict the intensity of selective carbonate dissolution in the glacial-interglacial cycles of the late Quaternary period. The assemblages from the Andaman Sea show little or no dissolution (deepest core AAS 11, water depth: ~3000 m), whereas cores collected from the CIB shows moderate to very high dissolution (SVBC 37, water depth of ~4000 m and core BC 37, water depth of ~4250 m). Excellent carbonate preservation in the Andaman Sea is reflected in the high order of species diversity consisting of thirty-five species. In contrast, selective dissolution in the CIB resulted in poor diversity with just about twenty-three species. The well-preserved planktonic foraminifera assemblages provide a better paleoceanographic record in the Andaman Sea. The carbonate dissolution proxies in the Andaman Sea point towards excellent preservation during Marine Isotope Stage 2 (MIS 2), especially during the deglacial period (Heinrich event 1) attributed to enhanced ventilation and deepening of carbonate compensation depth (CCD). Cores from the CIB demonstrate that the dissolution does not uniformly modify an assemblage due to the differential susceptibility of each species. Selective dissolution is an important issue in interpreting foraminiferal assemblages for climatic and environmental changes. The study reveals that the relationship between G. menardii abundances and its fragments in a single core is an effective parameter to understand the dissolution as its ratio is not affected by climatic or related environmental changes. Utmost care should be taken while using species variation as an evidence for climatic or related environment change from the CIB. The present investigation observes that the MFI can trace deep-sea carbonate dissolution quantitatively at both CIB and the Andaman Sea locations.

Background: Cyclooxygenase (COX) and Lipoxygenase (LOX) enzymes catalyze the production of pain mediators like Prostaglandins (PGs) and Leukotrienes (LTs), respectively from arachidonic acid. Introduction: The COX and LOX enzyme modulators are responsible for the major PGs and LTs mediated complications like asthma, osteoarthritis, rheumatoid arthritis, cancer, Alzheimer’s disease, neuropathy and Cardiovascular Syndromes (CVS). Many synthetic Nonsteroidal Anti- Inflammatory Drugs (NSAIDs) used in the treatment have serious side effects like nausea, vomiting, hyperacidity, gastrointestinal ulcers, CVS, etc. Methods: The natural inhibitors of pain mediators have great acceptance worldwide due to fewer side effects on long-term uses. The present review is an extensive study of the advantages of plantbased vs synthetic inhibitors. Results: These natural COX and LOX inhibitors control inflammatory response without causing side-effect-related complicacy. Conclusion: Therefore, the natural COX and LOX inhibitors may be used as alternative medicines for the management of pain and inflammation due to their less toxicity and resistivity.

Background: The establishment of strategy to inhibit the virus replication is an attractive means in combating SARS-CoV-2 infection. Objective: We studied phyto-compounds from Strychnos nux-vomica (a poisonous plant) against SARS-CoV-2 RNA-dependent RNA polymerase by computational methods. Method: Molecular docking, molecular dynamics (MD) simulation and energetics calculations were employed to elucidate the role of the phyto-compounds. Results: Ergotamine with a binding free energy of -14.39 kcal/mol showed a promising capability in terms of binding affinity and the interaction to conserved motifs, especially the SDD signature sequence. The calculated dissociation constants for ATP, ergotamine, isosungucine and sungucine were 12 μM, 0.072 nM, 0.011 nM and 0.152 nM, respectively. The exhibited kd by these phyto-compounds reflected tens of thousands fold potency as compared to ATP. The binding free energies of sungucine and isosungucine were much lower (-13.93 and -15.55 kcal/mol, respectively) compared to that of ATP (-6.98 kcal/mol). Conclusion: Sharing the same binding location as that of ATP and having high binding affinities, Ergotamine, Isosungucine, Sungucine and Strychnine N-oxide could be effective in controlling the SARS-CoV-2 virus replication by blocking the ATP and inhibiting the enzyme function.

Aging is associated with functional decline and structural deteriorations of the myocardium and plays a crucial role in cardiovascular ailments. Decalepis hamiltonii (Dh) is well known medicinal plant used in traditional medicine and exhibits potent antioxidant properties. Therefore, the current study was proposed to assess the cardioprotective potential of Dh extract and its bioactive constituents [Ellagic acid (EA) and 4‑hydroxyisophthalic acid (4-HIA)] against isoproterenol (ISO)-induced region-specific myocardial damage in aging rats. Male albino Wistar rats aged 22 months were administered orally with Dh extract (100mg/kg BW), EA (50 mg/kg BW), and 4‑HIA (30 mg/kg BW) for one month before inducing MI using ISO (150 mg/kg BW) for the consecutive last two days. The ISO group exhibited a significant increase in hemodynamic parameters, cardiac marker enzymes, serum lipids, and oxidative stress markers. A significant reduction in region-specific antioxidant enzymes levels and histopathological alterations were observed in ISO-treated rats. Whereas, pretreatment of EA, 4-HIA, and Dh extract reduced the cardiac dysfunction biomarkers and mitigated oxidative stress by enhancing antioxidant enzymes and restoring histopathological changes in ISO-treated rats. The Overall findings give insights into the therapeutic applications of Dh extract and its compounds in ameliorating cardiovascular disorders through its antioxidant potential.

Roselle (Hibiscus sabdariffa L.) is recognized as a valuable food crop due to its nutraceutical potential, rich pigment content, and medicinal properties. However, there is limited information on planting dates and suitable genotypes for roselle cultivation in arid and semi-arid regions. The objective of this field study was to investigate the influence of five planting dates (March, April, May, June, and July) on morphological and phytochemical characteristics of different roselle genotypes. Among genotypes, eight (Jiroft, Dalgan, Bampoor, Iranshahr, Nikshahr, Roodbar, Saravan, and Qaleganj) were Iranian landraces, and HA and HS-24 were originated from Ghana and Bangladesh, respectively. Planting date significantly influenced the number of branches, bolls and seeds per plant, sepal fresh weight, calyx and biomass yields, and harvest index in tested roselle landraces. The greatest morphological growth, fresh sepal weight (50.6 g plant−1) and calyx yield (1519 kg ha−1) were observed with the roselle planted in early May. Moreover, amounts of chlorophyll, flavonoids and antioxidant remained higher in roselle when planted between April–May. The number of branches/plant was found to be an important determinant of calyx yield (r = 0.707) in roselle. Dalgan resulted in the greatest growth and yields within a comparatively shorter period of time compared to other tested landraces. Both principal component analysis (PCA) and cluster analysis also indicated that Dalgan landrace possessed most suitable morphological and phytochemical traits among tested landraces, and therefore, it could be planted adopted for extensive cultivation settings in the arid and semi-arid regions.

Ashwagandha (W. somnifera L) is a popular ethnomedicinal herb endowed with potential pharmacotherapeutic properties. In this study the ethanol extract (EE) and water extract (WE) of ashwagandha roots were qualitatively analysed using high performance liquid chromatography (HPLC), the results showed EE containing withanoside IV (a), physagulin D (b), 27-hydroxywithanone (c), withanoside V (d), withaferin A (e), withastramonolide (f), withanolide A (g), withanone (h), and withanolide B (i) as major bioactive compounds. Antioxidant potential was studied using four in vitro free radical scavenging assays and EE displayed higher antioxidant effect compared to WE for all assays. The EE also showed enzyme inhibitory effect against lipoxygenase (5-LOX) >cyclooxygenases-1 (COX-1) compared to cyclooxygenases-2 (COX-2) and to that of WE. Furthermore, EE demonstrated a potential cytotoxic effect on proliferation of A549 cells in vitro, showing IC50 values, 394.8 (24 h), 171.9 (48 h) and 126.1 (72 h) μg/mL, respectively. EE extract also exhibited induction of apoptosis in dose dependent manner confirmed with the production of ROS in A549 cells through flow cytometric analysis. The obtained results demonstrated that ashwagandha EE with high concentration of bioactive compounds could act as potential drug candidates against pro-inflammatory enzymes involved in development of inflammation and progression of lung cancer. Further, extensive research is needed to investigate the ashwagandha bioactive compounds responsible for treating inflammation, oxidative stress and lung cancer.

Hedgehog signaling is essential for vertebrate development, however, less is known about the negative regulators that influence this pathway during the differentiation of cell fates. Using the mouse P19 embryonal carcinoma cell model, Suppressor of Fused (SUFU), a negative regulator of the Hedgehog pathway, was investigated during retinoic acid-induced neural differentiation. We found Hedgehog signaling was activated in the early phase of neural differentiation and became inactive during terminal differentiation of neurons and astrocytes. SUFU, which regulates signaling at the level of GLI, remained relatively unchanged during the differentiation process, however SUFU loss through CRISPIR-Cas9 gene editing resulted in decreased cell proliferation and ectopic expression of Hedgehog target genes. Interestingly, SUFU-deficient cells were unable to differentiate in the absence of retinoic acid, but when differentiated in its presence they showed delayed and decreased astrocyte differentiation; neuron differentiation did not appear to be affected. Retinoic acid-induced differentiation also caused ectopic activation of Hh target genes in SUFU-deficient cells and while the absence of the GLI3 transcriptional inhibitor suggested the pathway was active, no full-length GLI3 was detected even though the message encoding Gli3 was present. Thus, the study would indicate the proper timing and proportion of glial cell differentiation requires SUFU, and its normal regulation of GLI3 to maintain Hh signaling in an inactive state.

Dittrichia viscosa L., is a perennial plant belonging to the Asteraceae family, and this study was performed to investigate the chemical composition of its extract, using gas chromatography–mass spectrometry (GC–MS). Total phenol (TPC), flavonoid (TFC), and tannins contents (TTC), were quantified using colorimetric methods in two extracts (EtOH and ACE). The antioxidant activity was evaluated using DPPH scavenging, phosphomolebdenum test (TAC) and ferric reducing power assay (FRAP). The antimicrobial activity was determined against six nosocomial pathogens: Bacillus subtilis, Staphylococcus aureus, Salmonella enterica, Escherichia coli, Candida albicans, Aspergillus niger, using disc diffusion method and microdilution assay. The ACE and EtOH extracts had similar TPC: 151.18 ± 1.57 and 127.09 ± 15,81 mg GAE/ g DW. TFC & TTC recorded were also closely matched. The chemical composition revealed the presence of 18 phytochemical compounds with a total of 99.91%, where trimethylsilyl-meso-inositol (20.54%) was the major compound, followed by 5(4H)-Thebenidinone (16.80%). Both extracts showed high radical scavenging activity with an IC50 equal to 12.54 ± 0.2 μg/mL for EtOH, and 7.84 ± 0.1 μg/mL for ACE in DPPH test. In the FRAP test, we recorded an EC50 of 6.37 ± 0,012 mg/mL for EtOH, and 6 ± 0.022 mg/mL for ACE. The ACE presented higher antioxidant capacity (253.52 ± 2.98 mg AAE/g) compared to EtOH (189.14 ± 4,86 mg AAE/g) in the TAC assay. The higher inhibition zone was observed on B. subtilus (13 ± 0.1 mm) for EtOH, and the ACE was more effective on S. enterica (13.3 ± 0.08 mm). All the microbial strains were sensitive for both extracts, with MICs ranging from 0.93 mg/mL to 15 mg/mL.

Ginger (Zingiber officinale Rosc.) is well known as distinctive spice with considered therapeutic values. In the current work we aimed to investigate cytotoxic effect, phytochemical screening and genetic analysis of ginger callus and their rhizome. Cytotoxicity of ginger rhizome and callus extracts was examined against some human cancer cell lines HT29, HT116 and MCF-7 by MTT method. Phytochemicals were screened by thin layer chromatography (TLC) and genetic analysis was performed by ISSR markers. Ginger rhizome principle components, 6-gingerol and 6-shogoal were used as standards to detect their presence in callus. Results showed that, rhizome extracts displayed a strong cytotoxicity towards all tested cancer cells with the highest activity (IC50 20.4 ± 3.0 µg/mL; P 100 µg/mL in all examined cancer cell lines. TLC chromatograms of rhizome and callus extracts revealed the presence of phenols and terpenes, whereas 6-gingerol and 6-shogaol were only detected in rhizome extracts. Genetic analysis by ISSR markers revealed occurrence some genetic changes in DNA of callus compared to that of rhizome with total polymorphism 23.07%. In conclusion, ginger cytotoxicity to HT29, HT116 and MCF-7 cancer cells was mainly associated with the presence of 6-shogaol and 6-gingerol in extracts, which appeared as strong cytotoxic effect in rhizome compared to non-cytotoxic effect was evaluated in the callus extracts. Therefore, further studies on the callus elicitation and precursors feeding to induce biosynthesis of ginger bioactive molecules are recommended.

Ionic liquids (ILs) have been widely used in the pretreatment of biomass. However, the effects of residual ILs on the enzymolysis and fermentation of biomass are still unknown. Therefore, a large quantity of water-washing is usually followed after biomass pretreatment to eliminate the inhibition of residual ILs on subsequent hydrolysis and fermentation steps. In this work, the effect of choline glycine ([Ch][Gly]) concentration on the activity of cellulase and Bacillus sp. strain P38 was systematically investigated to explore the impacts of residual ILs on enzymolysis and fermentation. The results confirmed that the activities of them were almost not inhibited in low concentrations (less than 0.5 wt%) of [Ch][Gly]. Under optimal pretreatment conditions, the maximum cellulose digestibility was 99.23%. Enzymatic hydrolysate was suitable for l -lactic acid fermentation without appreciable inhibition, and the highest sugar-acid conversion rate of 96.33% was obtained by simplified detoxification. This work provides an economic route to produce fermentable sugar and l -lactic acid, which shows an industrial application prospect in lignocellulosic biorefinery.

Rational design and facile preparation of low-cost and efficient catalysts for the selective converting of biomass-derived monosaccharides into high value-added chemicals is highly demanded, yet challenging. Herein, we first demonstrate a N doped defect-rich carbon (NC-800-5) as metal-free catalyst for the selective oxidation of d -xylose into d -xylonic acid in alkaline aqueous solution at 100 °C for 30 min, with 57.4% yield. The doped graphitic N is found to be the active site and hydroxyl ion participating in the oxidation of d -xylose. Hydroxyl ion and d -xylose first adsorb on NC-800-5 surface, and the aldehyde group of d -xylose is catalyzed to form germinal diols ion. Then, C–H bond break to yield carboxylic group. Furthermore, NC-800-5 catalyst shows high stability in recycled test.

Paraquat is one of the most widely used nonselective herbicides in agriculture. Due to its wide use, paraquat resistant weeds have emerged and is becoming a potential threat to agriculture. The molecular mechanisms of paraquat resistance in weeds remain largely unknown. Physiological studies indicated that the impaired translocation of paraquat and enhanced antioxidation could improve paraquat resistance in plants. However, the detoxification of paraquat via active metabolism by plants has not been reported to date. Here we report that an activated expression of At1g01600 encoding the P450 protein CYP86A4 confers paraquat resistance as revealed by the gain-of-function mutant paraquat tolerance 11D (pqt11D), in which a T-DNA with four 35S enhancers was inserted at 1646 bp upstream the ATG of At1g01600. The paraquat resistance can be recapitulated in Arabidopsis wild type by overexpressing AtPQT11 (At1g01600), while its knockout mutant is hypersensitive to paraquat. Moreover, AtPQT11 also confers paraquat resistance in E. coli when overexpressed. We further demonstrate that AtPQT11 has P450 enzyme activity that converts paraquat to N-demethyl paraquat nontoxic to Arabidopsis, therefore detoxifying paraquat in plants. Taken together, our results unequivocally demonstrate that AtPQT11/ CYP86A4 detoxifies paraquat via active metabolism, thus revealing a novel molecular mechanism of paraquat resistance in plants and providing a means potentially enabling crops to resist paraquat.

Introduction The incidence of infections caused by aerobic actinomycetes is increasing. Recent changes in taxonomy and the variability in susceptibility patterns among species make necessary a proper identification and antibiotic susceptibility testing. Material and methods Fifty-three strains of aerobic actinomycetes were identified by MALDI-TOF MS using the VITEK MS Mycobacterium/Nocardia kit (bioMerieux, France) in a tertiary hospital in Spain during a six-year period. Antimicrobial susceptibility testing of the isolates was performed using the Sensititre Rapmycoi microdilution panel (Thermo Fisher Scientific, Massachusetts, USA). Results Forty strains of Nocardia spp. were identified in the study, being N. farcinica and N. cyriacigeorgica the most prevalent ones. All isolates were susceptible to linezolid and the resistance to amikacin was only observed in one isolate of Gordonia sputi. Resistance to cotrimoxazole was only found in five isolates. Conclusions Routine identification and antimicrobial susceptibility testing of aerobic actinomycetes is advisable for an efficient identification of species and effective treatment.

Background: Colorectal cancer (CRC) is the third most common malignancy worldwide. Matrine can act as a potential antitumor drug, and its antitumor activities have been tested in various cancers, including CRC. However, the effect of matrine and the related mechanisms on CRC cells remains poorly defined. Materials and Methods: CRC cells were treated with different concentrations of matrine, and then MTT, flow cytometric, and transwell assays were used to assess cell proliferation, apoptosis, invasion, and migration. MiR-10b-5p and Phosphatase and tensin homolog (PTEN) expression levels were measured by quantitative real-time polymerase chain reaction and western blot assay. The binding interaction of miR-10b-5p and PTEN were predicted by TargetScan and verified by a dual-luciferase reporter and RIP assay. The effect of matrine, miR-10b-5p, and PTEN on CRC cell proliferation, apoptosis, migration, and invasion was detected by MTT, flow cytometric, and transwell assays severally. Results: Matrine notably restrained proliferation, invasion, and migration and boosted apoptosis of CRC cells, as well as downregulated miR-10b-5p expression and upregulated PTEN protein level. PTEN was a direct target of miR-10b-5p in CRC cells. MiR-10b-5p knockdown and matrine treatment inhibited cell proliferation, migration, and invasion and induced apoptosis, and reintroduction of si-PTEN partly regained the inhibiting effect. Besides, MiR-10b-5p knockdown and matrine treatment repressed CRC growth in vivo. Conclusion: Matrine could suppress proliferation, migration, and invasion and induce apoptosis of CRC cells via the miR-10b/PTEN pathway, providing the potential molecular mechanism of matrine in blocking CRC progression.

Background Klebsiella pneumoniae is a gram-negative opportunistic pathogen that causes diseases mostly in immunocompromised individuals. Recently, hypervirulent K. pneumoniae strains also cause severe disease in healthy individuals. Capsular polysaccharide (CPS) is the major virulence determinant in hypervirulent K. pneumoniae and protects the cell against the bactericidal activity of the immune system. Gallic acid (GA), a natural phenolic compound, is known to exhibit wide spectrum antibacterial activity; however, its effect on hypervirulent K. pneumoniae remains largely unresolved. We aimed to identify the effects of GA on CPS biosynthesis in hypervirulent K. pneumoniae. Methods Antibacterial activity of GA was evaluated by counting colonies. CPS amount was determined by glucuronic acid content. The transcriptions of cps gene cluster were measured by quantitative real time PCR (qRT-PCR) and the β-galactosidase activity. The effect of GA on the resistance of K. pneumoniae to streptonigrin (SNG), an iron-activated antibiotic, was evaluated. The effect of GA on the resistance of K. pneumoniae to serum killing and phagocytosis by macrophages was observed. Results GA inhibited the growth and CPS biosynthesis in K. pneumoniae. GA may affect the iron availability in K. pneumoniae, thus possibly repressing the cps transcription. In addition, GA reduced the resistance of K. pneumoniae to serum killing and enhanced its susceptibility to phagocytosis. Conclusion GA possesses bactericidal activity and inhibits the CPS biosynthesis in hypervirulent K. pneumoniae, thereby facilitating pathogen clearance by the host immune system. Therefore, GA may represent a promising strategy for the prevention or treatment of patients with hypervirulent K. pneumoniae infections.

New indigenous bacteria, extracted from soil samples that had been collected from different parts of Iran, were introduced to improve dune sand using microbially induced carbonate precipitation (MICP) as an environmental engineering method. In order to isolate urease-producing bacteria from local alkaline soils, a series of standard tests was conducted to evaluate the urease performance and potential of calcium carbonate (calcite) sedimentation. Based on their properties, the bacteria with the best performance were identified by 16S ribosomal ribonucleic acid sequencing. To achieve a better understanding of the performance of the selected isolates, Sporosarcina pasteurii was employed as the control sample. The geotechnical behaviour of bio-mediated sand was studied through a series of geotechnical experiments, including unconfined compressive strength (UCS), constant-head permeability and wind erosion tests. The influence of freeze–thaw cycles and the durability of specimens on the UCS was also investigated. It is worth noting that sand MICP-treated by Bacillus sp. UTMC 2623 gave a performance relatively equal to that of the control sample during all UCS tests. Scanning electron microscopy images and X-ray diffraction were also used to observe calcite precipitation formation using Bacillus sp. UTMC 2623, which indicated precipitation of calcite throughout the sand column.

Clove basil (Ocimum gratissimum L.) is an aromatic, perennial herb belonging to the family Lamiaceae. The plant is indigenous to tropical areas especially India and West Africa. In Nigeria, it is found in the Savannah and coastal areas. The whole herbs of the plant contain essential oils and it is cultivated for various purposes. The plant is a rich source of eugenol in its essential oil. It is also used in the preparation of tea and infusions. It is also used in the treatment of fungal infections, fever, cold, and cough. Eugenol (4-allyl-2-methoxy phenol) is a phenolic compound from the class of phenylpropanoids. It is used in the food industry as a preservative, mainly due to its antioxidant property, and flavoring agent. The fresh herb × winter season/environment produced the highest essential oil 2.07 mL/plot with essential oil content 0.41% followed by fresh herb × summer season/environment (M1S2) = 1.68 mL. For the eugenol content, the autumn season/environment was found highly favorable = 74.52% (leaves = 69.024, stem = 74.531, and in inflorescence = 80.012) followed winter season/environment 72.30% (leaves = 71.841, stem 69.389, inflorescence 76.042, mean = 72.42%). These seasons are recommended for harvesting to obtain the optimum benefit for the quality essential oil yield.

This is a case of a young woman who developed neurological and psychiatric symptoms 3 days after resection of an immature teratoma. She was diagnosed with anti-NMDA receptor encephalitis via positive serum antibody titres, which was later confirmed with cerebrospinal fluid antibody titres. Given her cancer diagnosis, she underwent treatment with bleomycin, etoposide and cisplatin chemotherapy in addition to 5 days of high-dose steroids (1 g of intravenous methylprednisolone) for the encephalitis. This treatment regimen led to significant clinical improvement 3 weeks after completion of one cycle of chemotherapy.

Objectives. To assess cannabis and alcohol involvement among motor vehicle crash (MVC) fatalities in the United States. Methods. In this repeated cross-sectional analysis, we used data from the Fatality Analysis Reporting System from 2000 to 2018. Fatalities were cannabis-involved if an involved driver tested positive for a cannabinoid and alcohol-involved based on the highest blood alcohol concentration (BAC) of an involved driver. Multinomial mixed-effects logistic regression models assessed cannabis as a risk factor for alcohol by BAC level. Results. While trends in fatalities involving alcohol have remained stable, the percentage of fatalities involving cannabis and cannabis and alcohol increased from 9.0% in 2000 to 21.5% in 2018, and 4.8% in 2000 to 10.3% in 2018, respectively. In adjusted analyses, fatalities involving cannabis had 1.56 (95% confidence interval [CI] = 1.48, 1.65), 1.62 (95% CI = 1.52, 1.72), and 1.46 (95% CI = 1.42, 1.50) times the odds of involving BACs of 0.01% to 0.049%, 0.05% to 0.079%, and 0.08% or higher, respectively. Conclusions. The percentage of fatalities involving cannabis and coinvolving cannabis and alcohol doubled from 2000 to 2018, and cannabis was associated with alcohol coinvolvement. Modulation of proteasome function by pharmacological interventions and molecular biology tools is an active area of research in cancer biology and neurodegenerative diseases. Curcumin (diferuloylmethane) is a naturally occurring polyphenol that affects multiple signaling pathways and known to modulate PKC activities. The therapeutic significance of curcumin is often considered to reside in its anti-inflammatory, antioxidant, anti-angiogenic, or anti-apoptotic properties. However, recent research suggests that the therapeutic efficacy of curcumin may be due to its activity as a potent inhibitor of the proteasome. In this study, we show that both curcumin and its synthetic polyphenolic derivative (didemethylcurcumin), CUIII modulated proteasome activity in a biphasic manner. Curcumin and CUIII increased proteasome activity at nanomolar concentrations but inhibited proteasome activity at micromolar concentrations. Curcumin was more effective than CUIII in relative proteasome activity increase at nanomolar concentrations. Also, curcumin was more effective than CUIII in relative proteasome activity inhibition at micromolar concentration. The docking study was conducted on the 20S proteasome catalytic subunit. Estimated Kd values for curcumin and didemethylcurcumin are 0.0054μM and 1.3167μM, respectively. These values correlate well with the results of the effectiveness of modulation by curcumin compare to CUIII. The small size of CUIII makes its dock to the narrow cavity of the active site, but the binding interaction is not strong compare to curcumin. This study suggests that curcumin and its didemethyl derivative can be used to modulate proteasome activity. This communication implicates the reason why curcumin and its didemethyl derivative can be used to two different disease mechanisms, neurodegeneration, and cancer.
