The utilization of full-fat high-oleic soybean meal in layer diets could lead to value-added poultry products. To test this idea, 336 hens were randomly assigned to 4 isonitrogenous (18.5% CP) and isocaloric (2,927 kcal/kg) formulated diets and fed the following diets for eight weeks: conventional control solvent-extracted defatted soybean meal (CON); extruded-expelled defatted soybean meal (EENO); full fat normal-oleic soybean meal (FFNO); or full fat high-oleic soybean meal (FFHO). Body weights (BW) were collected at week 0 and week 8. Eggs were collected daily, and the totals counted each week. Feed consumption was measured weekly, and egg quality was measured bi-weekly. Eggs were collected at wk 0 and wk 8 for fatty acid analysis. There were no significant treatment differences in any of the production parameters measured, BW, feed consumption, feed conversion ratio or egg production (P > 0.05). Eggshell strength was significantly greater in eggs produced from the EENO group as compared to the control (P < 0.01), while egg yolk color was significantly darker in eggs of the control and EENO treatment groups relative to the FFNO and FFHO treatments (P < 0.0001). Eggs produced by hens fed the FFHO diet had a 52% increase in monounsaturated n-9 oleic acid content (P < 0.0001) and reduced palmitic (P < 0.01) and stearic (P < 0.0001) saturated fatty acid levels as compared to the conventional controls. The aim of this study was to analyze and compare the Hg, Pb, Cd, and Mn levels in egg feed, soil, and water among laying hens, laying ducks, and free-grazing duck farms in contaminated and uncontaminated areas. This study revealed that the Hg concentration in the eggs of free-grazing ducks was significantly greater than that in the eggs of laying hens and ducks in both contaminated and uncontaminated areas. However, the Pb and Mn levels in the eggs of laying ducks and free-grazing ducks were significantly greater than those in the eggs of laying hens in the contaminated area. Unfortunately, the Hg, Pb, Cd, and Mn concentrations in the feed, soil, and water from these three farms in both areas were not significantly different (P > 0.05). Hg and Cd were confirmed to be enriched in the egg albumin fraction, while Pb and Mn were found mainly in the egg yolk. However, egg consumption from free-grazing duck farms was the riskiest to Hg, Pb, and Mn contamination in the contaminated area. Additionally, the ecological risk factor (ER) in the soil revealed that all the farms were at considerable to high environmental risk for Cd except for Hg and Pb. Although the potential ecological risk index (RI) indicated a moderate risk for all farms in both contaminated and uncontaminated areas, these results were not consistent with our hypothesis. Infections by gastrointestinal parasites are found in a variety of animals worldwide. For the diagnosis of such infections, the flotation method is commonly used to detect parasitic microorganisms, such as oocysts or eggs, in feces. Instead of adding a flotation solution after the final centrifugation step and using a cover slip to collect the parasites, the method using a wire loop for the recovery of the organisms has been reported as one of alternative methods. However, the recovery rates of microorganisms from the flotation method have not been analysed. In the present study, the utility of a flotation method with the use of a wire loop of 8 mm in diameter (the loop method) was evaluated using different numbers of E. tenella oocysts and Heterakis gallinarum eggs, and chicken fecal samples collected at the farms. Consequently, we found that the oocysts and eggs in tubes could be collected at a ratio of 2.00 to 3.08. The effect of the diet modification (soybean and lupine addition) on the content of protein and amino acids (AA) in eggs was studied. Both the sampling day and the diet influenced the total protein content. In albumen, the lowest protein content (10.6%) was noted after administering a diet containing 25% lupine; in the same egg the yolk contained the most proteins (16.7%). In the content of nonessential AA (NAA) in egg yolks, differences were noted only for cysteine, with its the highest content in the yolks of the control group. The stable content of essential yolk amino acids (EAA) was observed only for isoleucine, leucine, tryptophan and phenylalanine. The highest contents of EAA and NAA were recorded in the yolks of the control group (~47 and ~53 g/100 g of protein, respectively) and in the group with 25% additions of lupine (~42 and ~51 g/100 g of protein, respectively). AA with constant content in the tested albumens were methionine, tryptophan and alanine. The highest content of EAA (>~42 g/100 g of protein) and NAA (>~62 g/100 g of protein) were determined in albumen of eggs determined in the group with at least 20% additions of lupine. The highest content of EAA for humans delivered eggs from groups 4-6 (with the addition of soy into the diet ≤5%). The protein sources used in the hen diet significantly influenced the content of protein and individual AA in the produced eggs.

Omega-3 long-chain polyunsaturated fatty acids (LCPUFA) play critical roles in human development and health. Their intake is often effectively estimated solely based on seafood consumption, though the high intake of terrestrial animal-based foods with minor amounts of LCPUFA may be significant. Covalent adduct chemical ionization (CACI) tandem mass spectrometry is one approach for de novo structural and quantitative analysis of minor unsaturated fatty acids (FA), for which standards are unavailable. Here, CACI-MS and MS/MS are used to identify and quantify minor omega-3 LCPUFA of terrestrial animal foods based on the application of measured response factors (RFs) to various FA. American mean intakes of pork, beef, chicken, and eggs contribute 20, 27, 45, and 71 mg/day of docosahexaenoic acid (DHA), respectively. The estimated intake of omega-3 DHA, eicosapentaenoic acid, and docosapentaenoic acid from nonseafood sources is significant, at 164, 103, and 330 mg/day, greater than most existing estimates of omega-3 LCPUFA intake.

The demand for the use of fluralaner in an extra label manner is increasing due to lack of efficacious treatment to combat mites and bed bugs in the poultry industry in the United States. Fluralaner residue data in eggs is lacking and residues might cause risks to human health. The present study aimed to determine the depletion profiles of fluralaner in eggs and estimate the drug withdrawal interval in whole eggs by adopting the US Food and Drug administration tolerance limit method with single intravenous (0.5 mg/kg) or transdermal administration (average 58.7 mg/kg) in healthy shaver hens. Hens were treated intravenously or trans-dermally with fluralaner. The eggs were collected daily for 28 d for intravenous treated and for 40 d from the transdermal route group. Fluralaner concentrations in yolk and albumen were determined by mass spectrometry. The greater percentage of fluralaner was observed in yolk when compared to the albumen for both administration routes. Noncompartmental analysis was used to calculate the pharmacokinetic parameters in yolk, albumen and whole egg. The longest apparent half-life confirmed in yolk was 3.7 d for intravenous and 14.3 d for the transdermal route. The withdrawal intervals in whole egg for fluralaner following the intravenous and transdermal administration were 7 d and 81 d, respectively, with maximum residue limits (1.3 µg/g) at 13 d and 171 d, respectively, based on the limit of quantification (0.4 µg/g) from the analytical assay reported by EMA and APVMA., Competing Interests: DISCLOSURES The authors declare no conflicts of interest., (Copyright © 2024 The Authors. *1. In many countries, eggs are not refrigerated and must be stored at room temperature. The objective of this study was to explore the effects of dietary oregano oil (275 mg / kg; ORE) versus an unsupplemented control diet (CON) on laying hens on the shelf life and fatty acid profile of eggs.2. Treatments were randomly distributed into 10 pens containing 27 birds each. A total of 200 eggs were collected from both groups on the same day and were stored for either 0, 10, 21 and 35 d. At each storage time, egg yolks were analysed for fatty acid profile and lipid peroxidation.3. The main indicator of lipid peroxidation, malondialdehyde (MDA), was significantly lower in ORE eggs compared to CON eggs ( p  = 0.001). Storage time had a significant impact on MDA concentrations ( p  = 0.023), with the highest found after 35 d. Significant differences were found for individual fatty acids, saturated (SFA), monounsaturated (MUFA) and polyunsaturated fatty acids (PUFA). Palmitic acid, stearic acid, oleic acid, linoleic acid and arachidonic acid were significantly lower in ORE eggs compared to CON eggs ( p  < 0.05). Palmitoleic acid ( p  = 0.002), linolenic acid ( p  = 0.001) and docosahexaenoic acid (DHA, p  = 0.001) were significantly higher in ORE eggs.4. Storage only affected oleic, linolenic, linoleic, arachidonic and docosahexaenoic acids ( p  < 0.05). Total SFA, MUFA, n -6 and ratio of n -3 to n -6 ( n -3: n -6) PUFA were significantly higher in CON eggs ( p  < 0.05). The ratio of SFA to PUFA (SFA:PUFA, p  = 0.005) and total n -3 PUFA ( p  = 0.001) were significantly higher in ORE eggs.5. The n -3: n -6 ratio was significantly impacted by treatment ( p  = 0.021) and storage ( p  = 0.031) with no significant interaction. This ratio is important for human health indication and could lead to the development of designer eggs.

Micro-organisms on the eggshell surface of affect the quality of the egg. Sometimes, these microbes even pose a serious threat to the health of the egg's consumer. Bacterial 16S rDNA and fungal internal transcribed spacer region were sequenced to analyze the microbial diversity on the shell surface of the eggs collected from 4 distinct regions of China: Guyuan (GY; 1.5 million hens), Langfang (LF; 0.1 million hens), Beihai (BH; 1.2 million hens), and Dongguan (DG; 0.2 million hens). The results showed a higher bacterial and fungal abundance on the eggs collected from the northern and southern China farms, respectively. The dominant bacterial phylum detected across all egg samples was Firmicutes. In addition, the shell surfaces of the DG and LF samples harbored abundant levels of Proteobacteria. The dominant fungal phyla detected across all egg samples were Ascomycota and Basidiomycota. The bacterial compositions on eggshell surfaces differed significantly across all geographic regions, and the fungal composition differed significantly between samples collected from the southern and northern farms (P < 0.05). The abundance and composition of microbial colonies on the eggshell surface varied based on their geographical location (climate and environment) and farming scale (management). The chicken egg, an excellent natural source of proteins, has been an overlooked native biomaterial with remarkable physicochemical, structural, and biological properties. Recently, with significant advances in biomedical engineering, particularly in the development of 3D in vitro platforms, chicken egg materials have increasingly been investigated as biomaterials due to their distinct advantages such as their low cost, availability, easy handling, gelling ability, bioactivity, and provision of a developmentally stimulating environment for cells. In addition, the chicken egg and its by-products can improve tissue engraftment and stimulate angiogenesis, making it particularly attractive for wound healing and tissue engineering applications. Evidence suggests that the egg white (EW), egg yolk (EY), and eggshell membrane (ESM) are great biomaterial candidates for tissue engineering, as their protein composition resembles mammalian extracellular matrix proteins, ideal for cellular attachment, cellular differentiation, proliferation, and survivability. Moreover, eggshell (ES) is considered an excellent calcium resource for generating hydroxyapatite (HA), making it a promising biomaterial for bone regeneration. This review will provide researchers with a concise yet comprehensive understanding of the chicken egg structure, composition, and associated bioactive molecules in each component and introduce up-to-date tissue engineering applications of chicken eggs as biomaterials.

Poultry products such as meat and eggs are rich sources of proteins, vitamins, and minerals. It is a good indicator of healthy food. Keeping in view, the present study is designed to evaluate the prevalence of toxic heavy metals (lead, nickel, cadmium, and chromium) in chicken eggs and feed. For this purpose, five samples of egg and feed were collected from five different commercial markets in Skardu City. Each sample was prepared using the wet digestion method and analyzed using a flame atomic absorption spectrophotometer. The results showed that lead, nickel, and chromium were present in varying amounts in the feed and egg, with nickel being the most concentrated metal, followed by lead and chromium in egg samples, while the feed samples showed the highest concentration of chromium followed by lead and nickel. However, concentrations of selected heavy metals except cadmium were all above the permissible limit of the World Health Organization. The self-organizing map-artificial neural network is employed for the identification of patterns of heavy metals in chicken feed and egg samples. The lower left neurons of the maps showed higher heavy metal concentrations found in samples taken from Bazar, whereas the rest of the samples showed varied concentrations. A comparison of feed and egg concentrations showed that nickel concentration was lower in feed samples than in egg samples. The lead concentration decreased in eggs except in the Krasmathang feed sample. It is inevitable to obtain chicks from breeders of different ages in broiler fattening. However, breeder age has an effect on eggs. There is no detailed study on the changes in mineral levels in eggs and bones of these effects. Therefore, the aim of this study examines how mineral levels of shell, albumen, egg yolk, and bone, embryonic development and bone characteristics change according to the breeder age. A total of 370 fresh hatching eggs were obtained from Ross-308 breeders at 2 different ages (25 to 60-week-old). Egg, embryo, yolk sac and chick characteristics were determined. Morphometric parameters of bones of embryos at 19 days of age and chicks at hatch were examined. Mineral levels of albumen, yolk, shell and bones of embryo and chick were determined. Egg weight, embryo weight, embryo length, chick weight and chick length were higher in old breeder than those of younger one. Femur length, width and Seedor index of tibia, weight, width and Seedor index of metatarsus were found to be high in embryos at 19 days of old breeder than young breeder. There were differences in relative femur weight, tibia length and relative metatarsus weight of chicks at hatch. The mineral levels were high in the femur, tibia and metatarsus of embryos and chicks, in parts of the egg of old breeder. K, Ca, Mg, P, Fe and Mn levels in the eggshell were decreased during incubation. Ca and Mg levels were decreased in the yolk during incubation. K, Ca, Mg, P, Fe, Zn and Mn levels were found to be high in chick bones than those of bones of embryos at 19 days of age. The total embryonic death was found to be high in the fertile eggs of younger breeder group. No differences were observed in fertility, hatchability of fertile and set eggs according to the age of breeders. The levamisole maximum residue limit for edible fat, kidney, and muscle of chickens is 0.01 mg/kg. However, no maximum residue limit has been established for eggs. In the present study, the pharmacokinetic profile and levamisole residue in the eggs from laying hens were investigated using ultra-performance liquid chromatography-tandem mass spectrometry. A single dose of levamisole (30 mg/kg) was administered via the intramuscular or oral route, and an additional egg residue study was performed with 300 or 600 mg/kg commercial LEV drug (30 or 60 mg/kg as levamisole) orally. The limit of quantification was 0.0056 μg/mL and 0.0015 mg/kg for plasma and eggs, respectively. The plasma concentration was below the limit of quantification 10 and 12 h after intramuscular and oral administration, respectively. The half-life of the absorption phase was comparable between the intramuscular and oral routes, which was approximately 1 h, and the mean maximum concentration value was significantly higher in intramuscular (2.29 ± 0.30 μg/mL) than in oral (1.45 ± 0.38 μg/mL) route. The relative oral bioavailability after intramuscular administration was 92.3%. In the egg residue study, dose-dependent area under concentration and maximum concentration were observed after single oral administration of 30 and 60 mg/kg egg residue, and the calculated withdrawal period for both 30 and 60 mg/kg groups based on the positive list system standard (0.01 mg/kg) was 7 d after the treatment., (Copyright © 2023 The Authors. Studies from our laboratory over the past decade have yielded new information with regard to the dietary enrichment of eggs and poultry meat with omega-3 (n-3) polyunsaturated fatty acids (PUFA) but have also generated a number of unanswered questions. In this review, we summarize the novel findings from this work, identify knowledge gaps, and offer possible explanations for some perplexing observations. Specifically discussed are: 1) Why feeding laying hens and broilers an oil rich in stearidonic acid (SDA; 18:4 n-3), which theoretically bypasses the putative rate-limiting step in the hepatic n-3 PUFA biosynthetic pathway, does not enrich egg yolks and tissues with very long-chain (VLC; ≥20 C) n-3 PUFA to the same degree as obtained by feeding birds oils rich in preformed VLC n-3 PUFA; 2) Why in hens fed an SDA-rich oil, SDA fails to accumulate in egg yolk but is readily incorporated into adipose tissue; 3) How oils rich in oleic acid (OA; 18:1 n-9), when co-fed with various sources of n-3 PUFA, attenuates egg and tissue n-3 PUFA contents or rescues egg production when co-fed with a level of docosahexaenoic acid (DHA; 22:6 n-3) that causes severe hypotriglyceridemia; and 4) Why the efficiency of VLC n-3 PUFA deposition into eggs and poultry meat is inversely related to the dietary content of α-linolenic acid (ALA; 18:3 n-3), SDA, or DHA., (Copyright © 2023 The Authors. Piling, a dense cluster of hens, is a behavior of major concern to the cage-free egg industry. It can cause large numbers of mortalities at which point it is considered smothering. The aim of this study was to quantify whether piling can also have consequences on production and non-smothering mortalities, which have not previously been described. Additionally, we aimed to describe characteristics of piling behavior relevant to for management. Video footage from 12 flocks of laying hens was analyzed for piling behavior across 3 wks. Production and mortality data were accessed using an integrated online management system. Bayesian linear mixed-effect models were used for formal statistical testing of the relationships between piling and production. Accounting for some missing data, a total of 252 d amounting to approximately 15,624 h were analyzed for the presence or absence of piling behavior, which we believe constitutes the largest analysis of piling behavior in the scientific literature to date. All flocks observed exhibited piling behavior even if they had no history of smothering. On average, flocks piled more than 4 times per day for around 44 min per event and the peak piling time occurred between 1300 and 1359. We found that the number of piling events was associated with a reduction in the number of eggs produced the next day; based on the average of 4 piles per day this amounted to 7.35 fewer eggs per 1,000 birds per day. Contrary to our hypothesis, we found total piling duration per day was positively associated with fewer Grade B eggs, with a decrease of 0.74 Grade B eggs for every hour of piling per day. No relationship was observed between piling and mortality not attributed to smothering. We discuss possible causes and explanations for these results including birds' response to stress, crowd dynamics, and daily rhythms. 1. This study developed a comprehensive sensory evaluation system that consisted of descriptions corresponding to United States Department of Agriculture photos to evaluate overall acceptability, albumen and yolk appearances and odours. It determined physiochemical parameters of eggs stored at 7°C (7W12 and 7U12 for washed and unwashed, respectively) for 12 weeks and stored at 25°C (25W4 and 25U4 for washed and unwashed, respectively) for four weeks.2. Throughout storage, there was a general downward trend in Haugh units (HU) and yolk index and an upward trend in air cell size, weight loss and S -ovalbumin content were observed ( P  < 0.05). The 25W4 and 25U4 egg quality rapidly deteriorated from grade AA (HU 81.7) to grade B after two weeks (HU 46.5 and 49.6), whereas 7W12 and 7U12 eggs remained grade A after 12 weeks (HU 67.3 and 66.9). High correlations were observed between the sensory and physiochemical parameters ( i.e ., R 2  = 0.93, 0.93, 0.88 and 0.94 for albumen appearance, yolk appearance, sensorial odour and overall acceptability, respectively, with HU in 25W4 eggs).3. Eggs stored at 25°C and classified into 'premium', 'class I', and 'class II' on the basis of their HU had estimated shelf life of 0.5, 1.5 and 2.5 weeks, while shelf lives of 4, 9 and 15 weeks were estimated for 7°C-stored premium, class I and II eggs, respectively.4. In conclusion, distinct HU requirements for eggs of different quality classes under two storage temperatures need to be established. Incorporating sensory evaluation with conventional physiochemical analyses is promising to assess and estimate egg quality changes. Further research work about the influences of different storage temperatures and possible temperature fluctuations during storage on egg quality changes is needed.

Backyard poultry hens are becoming very popular as pets, and thus the demand for treating the individual chicken is increasing. Few basic diagnostic techniques commonly used in small animal practice have been evaluated in this species. At the moment, there is no study in backyard hens describing radiographic measurements of internal organs contrary to psittacine birds or birds of prey. Moreover, the effect of egg laying on these measurements has not been studied in avian species even though it could affect radiographic measurements depending on the stage of egg formation. This is of particular concern in laying hens since they are able to lay on a daily basis. Thirteen adult hens and 3 juvenile Rhode Island red hybrid hens ( Gallus gallus domesticus ) were used to evaluate which organs can be reliably measured and to provide preliminary reference values for clinically healthy chickens. Additionally, whole body radiographs were collected every 2 hours over 24 hours in 5 adult hens to study the evolution of these measurements throughout egg formation. Organs that could be clearly delimitated on radiographs were measured, and the only organs that could be reliably measured were the heart and liver silhouettes. These measurements were significantly higher in adult compared to juvenile hens ( P = 0.024). Among the different organ ratios, heart width: total liver width ratio was the only significantly different measurement and was higher in juvenile hens ( P = 0.024). Hepatic silhouette measurements and ratios were found to increase over time ( P < 0.02) but did not follow the progress of egg mineralization. Eggshell thickness was found to be a promising parameter to evaluate the stage of egg formation, and thus should be taken into account when trying to establish reference intervals for radiographic measurements of internal organs in laying hens.

We have developed a method for complete dissolution of whole eggs in formic acid that provides a new approach to analyzing egg biomolecules. As expected from prior work with extracted lipids, phosphatidylcholine represents the most abundant 31 P NMR signal. A simplified methanol/chloroform partitioning method for separating the dissolved egg solution into metabolites, lipids and protein was performed and after ultra-high mass resolution and tandem MS fragmentation analyses several phosphatidylcholine molecules containing different fatty acid chain lengths as well as number and position of double bonds was detected. The MS based proteomic analysis further revealed 6 Gallus sequences annotated as 'uncharacterized' because they show no sequence homology with any other protein found in nature and thus, may represent proteins uniquely evolved to perform functions specific to chickens. This experiment was conducted in the poultry field of the College of Agriculture, University of Diyala, and the Biochemistry Laboratory in the Department of Animal Production to show the effect of using tartaric and salicylic acids on the chemical composition of eggs produced by Lohman chickens raised at high temperatures, in which 210 laying hens of brown Lohman strain were used. Sixty weeks were divided into seven treatments: T1, the control treatment, T2 and T3, in which 0.2% of tartaric and salicylic acid were added to drinking water, respectively, and T4 added to it a mixture of organic acids at a rate of 0.4% to the drinking water, and treatments T5 and T6 were added. It contained 0.2% of tartaric and salicylic acid to the ration, respectively, and T7 was added to the ration of 0.4% of the two organic acids to the diet formulation. At the age of 75 weeks, eggs were taken randomly from the treatments and at a rate of 5 eggs/treatment to know the effect of the treatments on the chemical composition of the eggs produced, and the results of the statistical analysis showed It is proved in Appendix 1 that there was no significant effect ( P ≥0.05) of the different treatments on the studied traits, which included the percentage of moisture, protein, fat, carbohydrates and ash in each of the whites and yolks. This study aimed to investigate the abundance of microorganisms and quality of eggs washed with different washings (tap water, 0.03% calcium hypochlorite solution, 0.25% hydrogen peroxide solution, or 1% sodium percarbonate solution) and unwashed for 28-day storage. The results showed that the washing significantly decreased the abundance of microorganisms in all cases. Washing with one of the three alkaline sterilizing agent solutions significantly inhibited the deterioration of egg quality (evidenced by lower weight loss, air cell depth, albumen pH, yolk pH, and total volatile base nitrogen, but higher Haugh unit and yolk index) during storage, while washing with tap water showed opposite effects. The texture profile analysis and high-resolution scanning electron microscopy observation showed that all washings had slight negative effects on eggshell quality (eggshell breaking strength and microstructure), and washing with the alkaline sterilizing agent solution had no additional effects. The occurrence of egg proteins in products containing spent fowl manufactured under current practices was studied to assess the risk these food products may pose to egg-allergic consumers and to determine if Precautionary Allergen Labelling (PAL) was recommended. Spent fowl slaughtering and processing operations in 2 Canadian facilities were observed. Raw hen pieces (n = 134), coming from 2 facilities, and intermediate and processed products containing spent fowl (n = 57), coming from one facility, were analyzed using ELISA. All samples tested positive for egg proteins. Raw pieces were tested using a qualitative method (i.e., swabbing); estimated egg proteins concentrations suggest the presence of highly contaminated samples (>600 mg/kg in 2 hen wing samples). Swabbing was found to be efficient for rapid detection of eggs in raw hen pieces, but not for quantification. A comparison between swab and grind results showed that egg proteins concentration is underestimated by at least a factor 2 for whole carcasses and a factor 10 for breast, wings and drumsticks, when using the swab protocol. For intermediate and processed products, quantitative measurements indicate that egg protein levels were below 16 mg/kg. Additionally, 88 water samples from chiller tanks were analyzed and indicate that this step could be the cause of the global contamination observed with an increase in egg protein concentrations overtime during the production schedule. The avian egg perivitelline layer (PL) is a proteinaceous structure that encloses the egg yolk. It consists of the inner and the outer perivitelline layers (IPL and OPL, respectively) that are assumed to play distinct roles in bird reproduction. To gain insight into their respective function, we analyzed the proteome of IPL and OPL in chicken unfertilized eggs after mechanical separation, using a GeLC-MS/MS strategy. Of the 412 proteins identified, 173 proteins were uniquely recovered in IPL and 98 proteins in OPL, while 141 proteins were identified in both sublayers. Genes coding the most abundant proteins were shown to be expressed either in the liver/ovary (IPL formation) or in the oviduct (OPL formation), but rarely in both. The presence of oviduct-specific proteins (including LYZ, VMO1, AvBD11, PTN, OVAL and LOC10175704) in IPL strongly suggests that they participate in the physical association of IPL to OPL, whose tight attachment was further evidenced by analyses of IPL/OPL interfaces (by scanning electron microscopy). Functional annotation of identified proteins revealed functions associated with fertilization and early development for IPL, while OPL would rather participate in egg defense and embryogenesis. Collectively, our data highlight the complementary functions of IPL and OPL that are major determinants of bird reproductive success. SIGNIFICANCE: The present study unveils for the first time the individual proteomes of the two sublayers composing the chicken egg perivitelline layer (PL), which allowed to assign their respective putative biological roles in avian reproduction. The combination of proteomics with gene expression and ultrastructural analyses provides insightful data on the structure and biochemistry of the avian PL. The functional annotation of PL proteins highlights the multifaceted biological functions of this structure in reproduction including fertilization, embryonic development, and antimicrobial protection. The aim of this research was to investigate the effects of laying hen genotype and age on eggshell cuticle deposition. A total of 4,320 brown eggs were obtained from 3 modern hen strains (A, B, and C), currently used worldwide for commercial egg production, at different intervals of age (20-30, 40-50, and 60-70 wk). Four samplings of 120 randomly collected eggs were carried out for each genotype/interval of age. Eggs were individually weighed and cuticle blue staining was used to assess quality and degree of cuticle coverage. On each egg, the eggshell color profile was assessed before and after staining using the CIE L*a*b* system and these values were used to calculate ΔE* ab . A 4-point scale visual score (VS) system was also applied to estimate the degree of cuticle coverage after staining (0 = no coverage, 1 = partial coverage, 2 = total coverage - low degree, 3 = total coverage - high degree). The effects of genotype and age and their interaction on eggshell color attributes were assessed by means of factorial ANOVA, while omnibus Chi-Square and Chi-squared Automatic Interaction Detector algorithm were applied for the analysis of VS data. Overall, both genotype and age affected the eggshell color profile as well as the degree of cuticle coverage. Hen strain A showed better cuticle deposition in comparison with B and particularly C one, being ΔE* ab values significantly higher. The VS evaluation revealed that eggs with impaired cuticle coverage degree increased with the hen age (23, 34, and 37%, respectively for 20-30, 40-50, and 60-70 wk; P < 0.05). However, a significant interaction between genotype and age was observed: transition from early to late hen age resulted in a significantly different pattern of ΔE* ab changes in each genotype. The classification tree analysis confirmed that the hen genotype has a greater effect than the age on cuticle deposition. Chicken meat and hen egg are very popular foodstuffs around the world and highly consumed as curry, fast food, processed food, etc. assuming a promising source of protein. In the present study, the concentrations of Pb, Cd, Cr, As, Hg, Mn, Fe, and Zn in nationally representative samples of chicken meat and hen egg were determined and found in the range of 0.03-2.73, 0.01-0.015, 0.025-0.67, 0.04-0.06, 0.01-0.015, 0.15-0.63, 2.50-38.6, and 1.02-19.4 mg/kg-fw, respectively. The results demonstrated that only Pb exceeded the maximum allowable concentration (MAC) for dietary food. Multivariate statistical analyses depicted that anthropogenic activities were the major source of heavy metals in the investigated foodstuffs. Human health risks associated with the dietary intake of these metals through the consumption of chicken meat and hen egg were evaluated in terms of estimated daily intake (EDI), non-carcinogenic risk of individual heavy metal by target hazard quotient (THQ), total target hazard quotient (TTHQ) for combined metals, and carcinogenic risk (CR) for lifetime exposure. The calculated values of EDI, THQ, TTHQ, and CR were below their respective permissible benchmarks indicating the safe consumption of the investigated foodstuffs with respect to heavy metal contamination., (© 2021. Using a variety of genetic resources, the aim of this study is to see how genetic background affects egg traits in chickens. Three different chicken genotypes (a commercial Brown layer, BOR; 2 Japanese indigenous breeds, NGY and YKD) were investigated effects on genotype in 10 external and internal egg quality traits along with 20 yolk and albumen free amino acid traits. Significant effects on genotype in 10 external and internal egg quality traits and 18 yolk and 17 albumen amino acid traits were found (P < 0.05). In sizes and weights of egg and eggshell redness, there were significant differences among all combinations of genotype (BOR > NGY > YKD). In 14 yolk (Asn, Ser, Gln, Gly, His, Arg, Ala, Pro, Tyr, Val, Met, Leu, Phe, and Lys) and 8 albumen amino acid traits (Gln, Gly, His, Arg, Val, Ile, Leu, and Lys), BOR was significantly higher than NGY and YKD, while the opposite relations were seen in 2 amino acid traits (Cys and GABA). Moreover, phenotypic correlation analyses revealed that positive correlations among amino acid traits within each yolk and albumen were broadly seen (0.30 < r < 0.98, P < 0.05). However, there are almost no phenotypic correlations in amino acids between yolk and albumen in BOR and NGY, but negative correlations in YKD, which implying a potential use of untapped genetic resources for modifying amino acid balance. Non-destructive testing (NDT) for eggshell faults is highly important for the egg industry, as cracked eggs account for around 3% of total production. The most commonly used method at present, candling, is labor intensive, while computer vision systems are expensive and complicated. In this paper, we present a simple, yet efficient, novel method for eggshell crack detection by acoustic spectroscopy. Altogether, 693 sound recordings were evaluated by different classification methods. The results show a cross-validated 2.1% total classification error, with only 0.87% false positive rate, which is the crucial metric for fresh eggs. Adapting the developed method to an industrial setting may lead to a reliable, fast and cost-effective detection method.

Elevating levels of environmental lead (Pb) results in serious hazards to health of animals and human beings. In this study, daily diet with three different levels of Pb (Pb nitrate at doses of 1, 10, and 100 mg/kg body weight) were fed to ISA Brown layers. It showed that the kidney and liver have the relatively high Pb concentration (2.34 and 0.51 ppm) after culture, while the meat has the Pb concentration as low as 0.07 ppm (lower than the standard of Codex Alimentarius). It was also confirmed that egg laying worked as a potential pathway for hens to excrete Pb as Pb concentrations in eggshell and yolk increased from 0.10 to 3.11 ppm. However, the Pb concentration in egg white remains at a safe level (<0.10 ppm). Furthermore, even the intake of low dose Pb can cause a decline of bone mineral density and bone strength. Raman spot and mapping analysis indicated that carotenoids content in humerus from the hens of high dose group increased significantly, which hence can be applied as an indicator for resist stress. The degradation of bone quality will further damage the health of laying hens. Therefore, Pb exposure not only toxifies organs and reduces physiological features (e.g., body weight and laying rate) instantly, but also hurts poultry via degrading bone quality in long term. The appearance, size, and weight of poultry meat and eggs are essential for production economics and vital in the poultry sector. These external characteristics influence their market price and consumers' preference and choice. With technological developments, there is an increase in the application and importance of vision systems in the agricultural sector. Computer vision has become a promising tool in the real-time automation of poultry weighing and processing systems. Owing to its noninvasive and nonintrusive nature and its capacity to present a wide range of information, computer vision systems can be applied in the size, mass, volume determination, and sorting and grading of poultry products. This review article gives a detailed summary of the current advances in measuring poultry products' external characteristics based on computer vision systems. An overview of computer vision systems is discussed and summarized. A comprehensive presentation of the application of computer vision-based systems for assessing poultry meat and eggs was provided, that is, weight and volume estimation, sorting, and classification. Scaffoldin, an S100 fused-type protein (SFTP) with high amino acid sequence similarity to the mammalian hair follicle protein trichohyalin, has been identified in reptiles and birds, but its functions are not yet fully understood. Here, we investigated the expression pattern of scaffoldin and cornulin, a related SFTP, in the developing beaks of birds. We determined the mRNA levels of both SFTPs by reverse transcription polymerase chain reaction (RT-PCR) in the beak and other ectodermal tissues of chicken ( Gallus gallus ) and quail ( Coturnix japonica ) embryos. Immunohistochemical staining was performed to localize scaffoldin in tissues. Scaffoldin and cornulin were expressed in the beak and, at lower levels, in other embryonic tissues of both chickens and quails. Immunohistochemistry revealed scaffoldin in the peridermal compartment of the egg tooth, a transitory cornified protuberance (caruncle) on the upper beak which breaks the eggshell during hatching. Furthermore, scaffoldin marked a multilayered peridermal structure on the lower beak. The results of this study suggest that scaffoldin plays an evolutionarily conserved role in the development of the avian beak with a particular function in the morphogenesis of the egg tooth.

To create high-quality eggs by using different breed and feed materials, we investigated free amino acid contents and taste sensor traits using two chicken breeds (Rhode Island Red; RIR and Australorp; AUS) fed two feeds (mixed and fermented feeds). Two-way ANOVA revealed significant breed and feed main and interaction effects on albumen bitterness and a significant interaction effect on yolk bitterness. Albumen from RIR fed mixed feed and AUS fed fermented feed was higher bitterness, whereas yolk from those groups was lower bitterness. Significant breed effects were detected in four albumen amino acid traits (His, Met, Ile, and Lys) and a yolk His, whereas significant feed effects were found in 15 albumen amino acid traits (Asp, Glu, Ser, His, Gly, Thr, Ala, Tyr, Val, Met, Trp, Ile, Leu, Lys, and Pro) and a yolk cystine trait. Compared to albumen amino acids, yolk amino acids had limited effects by breed and feed. The present results suggest that genetic and nutritional factors can alter not only amino acid contents but also sensor values of bitterness, indicating that selecting the combination of breed and feed enable us to make amino acids enriched and taste added designer eggs in future., (© 2021 Japanese Society of Animal Science.)

Whole-genome sequencing (WGS) is a valuable tool in research on foodborne pathogens. In this study, a total of 143 isolates of Salmonella serotypes Enteritidis, Typhimurium, and Heidelberg sourced from eggs and chickens were analyzed for their antimicrobial resistance profiles using WGS data. The isolates carried high rate of genes resistant to aminoglycoside (70.63%), tetracycline (26.57%), fosfomycin (25.17%), sulfonamides (23.78%), and β-lactamases (15.38%); and aadA was the most frequently observed antimicrobial resistance gene (ARG). Antimicrobial resistance varies by Salmonella serotypes, with Salmonella enterica serovar Enteritidis (Salmonella ser. Enteritidis) isolates being highly resistant to aminoglycoside (particularly streptomycin); Salmonella ser. Typhimurium more resistant to aminoglycoside, tetracycline, and sulfonamides; and Salmonella ser. Heidelberg more resistant to aminoglycoside and fosfomycin. Salmonella ser. Typhimurium isolates presented more varieties of ARG than Salmonella ser. Enteritidis and Salmonella ser. Heidelberg. Our data showed that 5 isolates of Salmonella ser. Typhimurium and Salmonella ser. Heidelberg contained ARG resistant to ≥ 5 antimicrobials. In addition, 23 Salmonella isolates carried ARG resistant to 4 antimicrobials., (Copyright © 2020. Fusarium mycotoxins are fungal contaminants found in different crops intended for human and animal consumption. Due to the co-occurrence of several of mycotoxins, the present study aimed at examining the transfer of these toxins into tissues of broiler chickens and eggs of laying hens fed contaminated diets. After an adaptation period, the chickens were fed contaminated diets containing mg/kg levels of deoxynivalenol (DON), enniatins (ENN A, A 1 , B, B 1 ) and beauvericin (BEA) and high μg/kg levels of HT-2 toxin (HT-2), T-2 toxin (T-2) and zearalenone (ZEN) during a repletion period of two weeks, followed by a depletion period of two weeks. DON, ZEN, T-2 and HT-2 were not carried out into the skin and the liver of broiler chickens. ENN B (20.5 ± 6.6 μg/kg) and BEA (162 ± 55 μg/kg) were found in the liver, while in the skin their respective concentrations were 50 ± 17 μg/kg and 120 ± 16 μg/kg during the first week of the repletion period. Carry-over rates into liver and skin were higher for BEA (1.6% and 1.2%, respectively) than for ENNs (0.1 and 0.4%, respectively). During the depletion period, ENNs and BEA were eliminated from the skin and the liver. ENN B, ENN B 1 and BEA were carried over into eggs at 0.1%, 0.05% and 0.44% upon 2-3 days of feeding the contaminated diet, respectively. These transfers were fully eliminated 9-10 days after feeding the control diet again. These results indicate the transfer of ENN B, ENN B 1 and BEA from feed to chicken offal, meat products and eggs at a very low degree, thus marginally contribute to the total dietary intake of these fusariotoxins for consumers. Nevertheless, taking precautionary measures in the field, harvest, transport and storage of the raw materials is required to keep the mycotoxin concentration in feed below the safe levels., (Copyright © 2020 Elsevier Ltd. Different strains of commercial laying hens have been molded by varying selection pressures, impacting their production, health, and behavior. Therefore, assumptions that all laying hen strains use the given resources within aviary systems similarly and maintain equal health and performance may be false. We investigated interactions among patterns of aviary resource use by 2 strains of white and 2 strains of brown laying hens (4 units per strain, 144 hens per unit) with daily egg production, location of egg laying, keel fractures, and footpad damage across the lay cycle. Hens' distribution among resources (litter, nest, wire floor, ledge, and perch) was recorded during light and dark periods at 28, 54, and 72 wk of age. Daily egg production and location were recorded, and 20% of hens per unit were randomly selected and assessed for keel bone damage, foot health, and plumage quality. Production and health risks associated with hens' resource use were assessed using multivariable regression. During the day, more brown hens occupied wire floors, while larger numbers of white hens were on perches and litter. More brown hens were on lower-tier wire floors in the dark, while more white hens occupied top tiers. Brown hens laid more eggs outside nests, showed lower incidence of keel fractures, and had better plumage quality than white hens. White hens had higher odds of keel fractures (4.2) than brown hens. Odds of keel fractures were 3.7 and 5.7 times higher at 54 and 72 wk than at 28 wk in all strains (P ≤ 0.05). Occupying the upper tier at night increased odds of keel fractures by 5.4 times. Occupying perches was associated with lower odds of foot lesions and poor plumage quality in all strains across the lay cycle (P ≤ 0.05). Finally, white hens were associated with lower odds of non-nest laying (0.76), whereas higher nest use by brown hens resulted in higher odds of non-nest egg laying (1.56) across the lay cycle (P ≤ 0.05). Methionine (Met), an essential amino acid in poultry diets, when overdosed may cause hyperhomocysteinemia, which is mainly a trigger for cardiovascular diseases in humans. Homocysteine is neutralized (remethylated) in the presence of folic acid (FA), which also plays an important role in hematopoiesis and participates in the synthesis of DNA, and its deficiencies may result in the development of neural tube defects. One of the basic tools in studying the impact of both xenobiotics and nutrients on the animal organism is hematological analysis. Thus, the aim of this study was to determine the effect of in ovo supplementation with Met and FA on the hematological parameters of broiler chickens. On the 17th day of incubation, embryonated eggs (Ross 308) were injected with 5 or 25 mg of Met per egg (M5 and M25), 3 and 15 mg of FA per egg (F3 and F15), or a mixture of these 2 compounds (M5/F3 and M25/F15). The broilers were reared in accordance with welfare regulations and fed with commercial diets ad libitum. Blood samples were collected on the first, seventh, and 35th day of rearing (D1, D7, and D35), and complete hematological analysis was performed. The observed changes in red blood cell parameters probably result from physiological changes occurring during bird growth. Mean erythrocyte volume decreased with the age of chickens in the control, M5, and M25 groups, but not in those supplied with FA. Among supplemented groups, the number of white blood cells on D1 was lower only in group M5 than in the sham (C) group. The analysis of leukograms showed no significant differences between the groups. Comparing D1 with D7 in the group injected with a higher dose of Met and FA (MF25/15), a statistically significant increase in the percentage of lymphocytes and a significant decrease in the percentage of heterophils were observed. In addition, in the group injected with a higher FA dose (F15), there was statistically significant reduction in the percentage of eosinophils and a significant increase in the percentage of monocytes at day 7 compared with day 1. A video received by faculty at North Carolina State University's Prestage Department of Poultry Science revealed a live parasite inside a chicken egg. The parasite was identified as an oviduct fluke (Prosthogonimus macrorchis), a trematode with a three-host life cycle: the primary host, a galliform bird, then an aquatic snail, and finally a dragonfly larva or adult consumed by the infected bird. The egg was from a "backyard flock" with access to a watercourse. No other instances of this parasite were seen in eggs from the flock. The presence of this parasite inside an egg suggests that the worms had migrated above the shell gland in the oviduct to be incorporated inside the egg. Currently, the occurrence of an oviduct fluke inside an egg in the United States is rare. Such parasites are not found in eggs from caged layers because those birds do not have access to watercourses. This case reinforces the view that parasites requiring intermediate hosts may become more common in birds reared under free-range conditions.

Background: Avian eggs have a proteinaceous cuticle. The quantity of cuticle varies and the deposition of a good cuticle in the uterus (Shell-gland) prevents transmission of bacteria to the egg contents., Results: To understand cuticle deposition, uterus transcriptomes were compared between hens with i) naturally good and poor cuticle and, ii) where manipulation of the hypothalamo-pituitary-gonadal-oviduct axis produced eggs with or without cuticle. The highest expressed genes encoded eggshell matrix and cuticle proteins, e.g. MEPE (OC-116), BPIFB3 (OVX-36), RARRES1 (OVX-32), WAP (OVX-25), and genes for mitochondrial oxidative phosphorylation, active transport and energy metabolism. Expression of a number of these genes differed between hens laying eggs with or without cuticle. There was also a high expression of clock genes. PER2, CRY2, CRY1, CLOCK and BMAL1 were differentially expressed when cuticle deposition was prevented, and they also changed throughout the egg formation cycle. This suggests an endogenous clock in the uterus may be a component of cuticle deposition control. Cuticle proteins are glycosylated and glycosaminoglycan binding genes had a lower expression when cuticle proteins were deposited on the egg. The immediate early genes, JUN and FOS, were expressed less when the cuticle had not been deposited and changed over the egg formation cycle, suggesting they are important in oviposition and cuticle deposition. The uterus transcriptome of hens with good and poor cuticle deposition did not differ., Conclusions: We have gained insights into the factors that can affect the production of the cuticle especially clock genes and immediate early genes. We have demonstrated that these genes change their expression over the period of eggshell formation supporting their importance. The lack of differences in expression between the uterus of hens laying eggs with the best and worse cuticle suggest the genetic basis of the trait may lie outside the oviduct.

Single-aged caged layer hen flocks were monitored for Salmonella over the course of their lifetime. Chicks from both flocks were Salmonella negative at hatch and remained negative during rearing. Pullets were transported to production farms at 15 weeks of age. Pre-population dust swabs collected from both production sheds had a high percentage of Salmonella positive samples (80 and 90%). Flocks were sampled at regular intervals until 70-72 weeks of age. The proportion of Salmonella positive samples and mean load detected on eggs was low on both farms. Analysis of dust samples revealed that Salmonella persisted in dust over 8 weeks. Dust total moisture content and water activity appears to influence bacterial persistence. On egg grading equipment, only suction cups prior to egg washing were Salmonella positive (mean proportion Salmonella positive samples 0.13 ± 0.07; mean load of 18.6 ± 12.31 MPN/ml). An egg washing experiment demonstrated that while washing reduced the total Salmonella load from eggshell surfaces, no effect was observed for shell pores. Salmonella enterica serovar Enteritidis is the leading cause of salmonellosis in people, and modeling of infections in chickens is used to identify intervention strategies. A review of 80 manuscripts encompassing 119 experiments indicated that the mean dose of infection was 10 8 CFU per bird. Experiments of less than 10 6 CFU were primarily conducted in immature birds. To address a lack of information on the impact of low dosages on the hen at lay, two experiments were conducted in triplicate. Experiment A addressed issues associated with vaccination; thus, hens were infected intramuscularly at 10 3 , 10 5 , and 10 7 CFU. For Experiment B, which was focused more on colonization and invasion, hens were infected orally with 5 × 10 3 CFU with 4 strains from different genomic clades. Samples from liver, spleen, ovarian pedicle, and paired ceca in both experiments were cultured 5, 6, 7, and 8 days postinfection. Eggshell microbiome taxa were assessed in Experiment B. Results indicated that dosages of 10 3 CFU in both experiments produced enough positive samples to be used within models. The intramuscular route resulted in approximately twice as many positive samples as the oral route. The kinetics of infection appeared to differ between low and high dosages suggestive of a J-curve response. These results could impact risk assessments if the hen at lay has a nonlinear response to infectious dose.

The intestinal microbiota plays an essential role in the metabolism and immune competence of chickens from the first day after hatching. In modern production systems, chicks are isolated from adult chickens, instead hatching in a clean environment. As a result, chicks are colonized by environmental bacteria, including potential pathogens. There is a need to investigate methods by which chicks can be exposed to a more appropriate microbial community at hatching. Such methods must be easy to apply in a hatchery and produce consistent results. The development of the intestinal microbiota of chicks hatched from eggs sprayed with dilute adult cecal content during incubation was observed at 0, 3, 7, and 14 days posthatching (dph) across two experiments. High-throughput Illumina sequencing was performed for the V4 hypervariable region of the 16S rRNA gene. A topical treatment of dilute adult cecal content was sufficient to transplant spore-forming bacteria such as Lachnospiraceae and Ruminococcaceae However, this treatment was not able to transplant other taxa that are considered to be core elements of the chicken cecal microbiota, such as Bacteroidaceae , Lactobacillaceae , Bifidobacteriaceae , and Burkholderiaceae The topical treatment significantly altered the microbiota of chicks immediately posthatching and accelerated the normal development of the microbiota with earlier colonization by Ruminococcaceae in the cecum and "Candidatus Arthromitus " in the ileum. The effect of the treatment on the cecal microbiota was maximal at 3 dph but diminished over time. IMPORTANCE Over the last 60 years poultry production has intensified in response to increased demand for meat. In modern systems, chicks hatch without contacting chickens and their gut bacteria. Consequently, they are colonized by environmental bacteria that may cause disease. The normal bacteria that live in the gut, or intestinal microbiota, play an important role in the development of the immune system. Therefore, it is essential to find easy ways to expose chicks to the more appropriate bacteria at hatching. This experiment investigated whether spraying eggs with adult cecal contents was sufficient to transfer an adult microbiota to chicks. Our findings show that spore-forming bacteria were transplanted, but other members of the microbiota were not. The objective of the current study was to evaluate increasing levels of manganese hydroxychloride (MHC) in 45-wk-old white leghorn laying hens, using yolk and shell manganese (Mn) content as a potential marker for Mn concentration. A total of 80, 45-wk-old white leghorns were assigned to 6 dietary treatments, each consisting of 14 individually caged laying hens, with the exception of the reference diet containing 10 individually caged laying hens. The experiment consisted of a reference diet that contained 70 ppm of supplemental inorganic Mn in the form of Mn oxide and 5 experimental treatments each containing 0, 15, 30, 60, and 90 ppm supplemental MHC. Experimental birds were subjected to a 21 D depletion phase in which no supplemental Mn was included in the diet; however, during this time reference fed birds were fed the control diet (70 ppm Mn). After the 21 D depletion phase, the depleted birds were fed experimental diets for a 35 D evaluation period. Yolk and shell Mn content were analyzed at the end of the depletion phase and during the experimental phase on day 5, 10, 15, 25, and 35. During the experimental phase, Mn was replenished in the yolk and shell in all experimental treatments containing supplemental Mn; however, dose and time impacted the rate of replenishment. The yolk tended to be more sensitive to variations in Mn level as increases in Mn inclusion significantly (P < 0.05) increased concentration. These data demonstrate the ability to deplete and replenish Mn, and the use of egg yolk Mn concentration as measurement for determining changes in dietary Mn. Background: Eggs have been considered an important component of the human diet for centuries. There is currently a wide assortment of eggs available to consumers, varying in terms of weight (from S to XL), housing system (0 to 3), diet, packaging, etc. Although the egg&rsquo;s nutritional value is most important to consumers, their perceptions are unfortunately generally based not on facts, but on popular opinion. Most commercial lines of laying hens used in Poland are of foreign origin. However, some farms use commercial lines from Polish breeding farms. Therefore, research was undertaken to determine selected characteristics of eggs obtained from hens of different origins raised in an intensive system, i.e. industrial production of eggs., Methods: The research material consisted of eggs from Hy-Line Brown and Messa 43 hens from large-scale production. The eggs were obtained from hens at the ages of 35, 40 and 45 weeks. The analysis included selected external and internal characteristics of eggs laid during the study period. The eggs were evaluated immediately after laying, and the following quality parameters were determined: egg weight, shell weight, shell thickness, white weight, yolk weight, and pH of white and yolk. Chemical analysis of dry matter, total protein, crude fat and crude ash content in the whites and yolks was performed. The cholesterol content and fatty acid profile in the yolk were analysed as well. A statistical analysis of the results was performed., Results: The analyses revealed significant differences in the weight and morphological composition of eggs depending on the genotype of the hens. Compared to eggs obtained from Hy-Line Brown hens, eggs from Messa 43 hens had significantly lower shell thickness, greater weight between 35 and 45 weeks of age,&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; a larger proportion of shell, and a smaller proportion of albumen. Yolks and whites of eggs from Hy-Line Brown hens contained significantly more dry matter and total protein than those of Messa43 chickens, while the cholesterol content in the yolks of both groups was similar. The fatty acid profile of the egg yolk was similar irrespective of the origin of the hens., Conclusions: Significant differences in egg weight and the parameters of morphological components of eggs were found between the experimental groups. Genotype did not influence cholesterol content or the fatty acid profile in the yolk.

The purpose of the current study was to investigate the effect of choline as a means of increasing docosahexaenoic acid (22:6 n-3, DHA) deposition in egg yolks of hens fed a high-DHA microalgae product. Fifty-six, 26-wk-old, White Leghorn hens were kept in individual cages and randomly allocated to 1 of 4 dietary treatments, each with 7 replicate groups of 2 hens (n = 7 per treatment). The experimental diets were corn and soybean meal based, with 0% microalgae (control), 1% microalgae and no additional choline chloride (Alg), and Alg plus choline chloride at 0.1% (Ch0.1) and 0.2% (Ch0.2). The feeding trial lasted 16 wk. The data were fit as a general linear mixed model to generate least square means in response to diet. Variables measured multiple times during the study were fit as repeated measures. Using orthogonal contrasts, Alg was compared to control, and Ch0.1 and Ch0.2 were compared separately to Alg. Ch0.1 increased hen day egg production (P < 0.05) and Haugh unit (P < 0.05), and reduced feed conversion ratio (P < 0.05) compared to Alg, but Ch0.2 did not. Alg increased egg DHA (P < 0.001), phosphatidylethanolamine (P < 0.05), and phosphatidylcholine (P < 0.001) compared to control, but Ch0.1 or Ch0.2 had no effect compared to Alg (P > 0.05). In the liver, Alg increased lipid peroxidation products compared to control (P < 0.01), and Ch0.1 reduced them compared to Alg (P < 0.01). Both Ch0.1 and Ch0.2 increased hepatic concentrations of γ- (P < 0.05; P < 0.001) and α-tocopherol (P < 0.01; P < 0.001), and Ch0.1 increased γ-tocopherol concentration in eggs compared to Alg (P < 0.05). Vitamin D insufficiency is prevalent worldwide. Recently, we showed that exposure of laying hens to sunlight or artificial ultraviolet B (UVB) light is an efficient strategy to increase the vitamin D content in eggs. In the current study, using 2 different chicken genotypes and stocking densities, we addressed the question of whether different UVB-emitting regimes work under real indoor housing conditions in a floor system or in furnished cages. Here, we found a 3.7-fold increase in the egg vitamin D content in Lohmann Selected Leghorn hens and a 4.2-fold increase in Lohmann Brown hens after UVB exposure for 6 h/d. The data further reveal that UVB exposure under high stocking density is equally effective compared to that at low stocking density. The different light regimes were not associated with changes in the behavior of these animals. Backyard layer chickens may be exposed to a variety of metals in the environment, including lead. The potential public health concerns associated with lead exposure prompted us to systematically screen liver samples from backyard layers submitted to the diagnostic laboratory to estimate the prevalence of lead exposure. Over a period of 1 y, we tested 1,476 chicken livers, of which 45 were found to have lead concentrations of 0.9-41 µg/g. The lead-positive cases were investigated by follow-up questions to the bird owners on the environment, general management of the flock, and egg consumption of family members. Lead concentrations in 14 pooled egg samples were determined, and a conservative estimate of daily exposure of family members to lead was made based on egg consumption. In some cases, estimated daily lead intake exceeded the recommended limits for lead consumption in children. Analysis of feed, water, and environmental samples did not identify a source of exposure in most cases. Only 34% of owners of lead-positive birds submitted eggs or environmental samples, indicating a lack of interest or financial concerns. In most cases, neither the case history nor postmortem findings were indicative of lead intoxication; without systematically testing all birds, some cases could have been missed. Our study highlights the need for backyard chicken owners, veterinarians, and public health personnel to be aware of the risk of lead exposure and undertake preventive and surveillance measures.

This study was conducted to determine the optimum level of home-made selenium-enriched yeast (SeY) in the diet of broiler breeder hens and to compare the effects of this product with sodium selenite (SS) or Selemax (SM) on their productive and reproductive performance. A total of 150 broiler breeder hens were divided to six groups and hens in each group were received a basal diet containing no selenium (CG), 0.15, 0.30, 0.45 mg SeY/kg diet (SeY-0.15, SeY-0.30 and SeY-0.45, respectively), 0.30 mg SM/kg diet or 0.30 mg SS/kg diet for 15 successive weeks. The results showed that egg weight and production and hatchability rate were higher in SeY-0.45 compared to other groups (P < 0.05). Also, SeY-0.45 group led to lower embryonic mortality rate compared to CG and SS groups. Fertility rate and chick quality parameters were not affected by selenium supplementation during this period (P > 0.05). In conclusion, the dietary supplementation of home-made selenium, as an organic selenium source, can be used to improve the productive and reproductive performance in aged broiler breeder hens at 0.45 mg/kg feed., (Copyright © 2018. Contamination of eggs with Salmonella enterica is a significant risk factor contributing to foodborne disease. Periods of peak egg contamination were identified by conducting longitudinal environmental and egg sampling in 7 layer flocks until they were 50 weeks of age. A total of 714 environmental samples and 8958 eggs were cultured using standard methods for the detection of salmonellae. Pooled egg contamination with Salmonella Typhimurium or Salmonella Infantis was detected at a true prevalence (TP) of 0.002 (95% CI = 0.001, 0.004) or 0.005 (95% CI = 0.004, 0.007), respectively. S. Typhimurium and S. Infantis were detected in individual egg components; in shell rinse at a TP of 0.014 (95% CI = 0.005, 0.038), in shell and membrane at a TP of 0.01 (95% CI = 0.003, 0.032), and in albumen and yolk content at a TP of 0.007 (95% CI = 0.001, 0.027). The concentration of salmonellae in all fractions was <1 CFU/mL. The TP of Salmonella enterica in eggs was highest at the onset of lay. Higher egg prevalence was associated with a lower body weight, higher egg production, higher egg weight and mass than the breed standard for age, and poorer feed conversion efficiency. Soya bean is the main protein source in poultry feed but rising prices make an alternative protein source necessary. Insects, such as the black soldier fly (Hermetia illucens), may be an attractive solution for hens, although little information is available on their effect on egg quality. The present study aims to fill this gap by testing the effect of 100% replacement of soya bean with H. illucens larva meal in the diet of Lohmann Brown Classic laying hens for 21 weeks. At the end of the trial, the eggs were characterized for parameters such as weight, colour, proximate composition of albumen and yolk, and content of carotenoids, tocopherols and cholesterol. The fatty acid profile of yolks was also determined. Hens fed the insect-based diet produced eggs (HIM group) with a higher proportion of yolk than the group fed the soya bean-based diet (SBM group). HIM was associated with redder yolks (red index 5.63 v. 1.36) than SBM. HIM yolks were richer in γ-tocopherol (4.0 against 2.4 mg/kg), lutein (8.6 against 4.9 mg/kg), β-carotene (0.33 against 0.19 mg/kg) and total carotenoids (15 against 10.5 mg/kg) than SBM yolks. The fatty acid composition of HIM yolks was almost identical to that of SBM yolks. Finally, HIM yolks contained 11% less cholesterol than SBM yolks. These results suggest that H. illucens larva meal is a suitable total substitute for soya bean meal in the diet of Lohmann Brown Classic laying hens. A sustainable alternative to the plant protein source therefore seems feasible.

A 1.5-year-old Polish hen was presented with a history of watery droppings and poor vent tone. Results of diagnostic tests revealed blood lead at levels considered to be toxic. Chelation therapy was started with calcium ethylenediaminetetraacetate. The hen was laying eggs before, during, and after chelation therapy. Eggs were tested for the presence of lead by combining yolk and albumen together. Before chelation therapy, the level of lead in the egg tested was 14 μg. Two days after the end of chelation therapy, results of a second blood lead test revealed a drop to nontoxic levels. No lead was detected in the combined yolks and albumen of eggs collected 7-11 days after the end of chelation therapy. Four weeks after the end of chelation therapy, no lead was identified in the shells of tested eggs.

Aims: To determine the plasma disposition and concentrations of ivermectin (IVM) in eggs produced by laying hens following S/C, oral and I/V administration., Methods: Twenty-four laying hens, aged 37 weeks and weighing 1.73 (SD 0.12) kg were allocated to three groups of eight birds. The injectable formulation of IVM was administered either orally, S/C, or I/V, at a dose of 0.2 mg/kg liveweight, following dilution (1:5, v/v) with propylene glycol. Heparinised blood samples were collected at various times between 0.25 hours and 20 days after drug administration. Eggs produced by hens were also collected daily throughout the study period. Samples of plasma and homogenised egg were analysed using HPLC., Results: Maximum concentrations of IVM in plasma and mean residence time of IVM were lower after oral (10.2 (SD 7.2) ng/mL and 0.38 (SD 0.14) days, respectively) than after S/C (82.9 (SD 12.4) ng/mL and 1.05 (SD 0.24) days, respectively) administration (p<0.01). The time to maximum concentration and elimination half-life were shorter following oral (0.14 (SD 0.04) and 0.23 (SD 0.11) days, respectively) than S/C (0.25 (SD 0.00) and 1.45 (SD 0.45) days, respectively) administration (p<0.01). IVM was first detected in eggs 2 days after treatment in all groups and was detected until 8 days after oral and I/V administration, and until 15 days after S/C administration. Peak concentrations of IVM were 15.7, 23.3 and 1.9 µg/kg, observed 2, 5 and 4 days after I/V, S/C and oral administration, respectively., Conclusions and Clinical Relevance: The low plasma bioavailability of IVM observed after oral administration in laying hens could result in lower efficacy or subtherapeutic plasma concentrations, which may promote the development of parasitic drug resistance. Due to high IVM residues in eggs compared to the maximum residue limits for other food-producing animal species, a withdrawal period should be necessary for eggs after IVM treatment in laying hens.

Lomefloxacin (LOM) and pefloxacin (PEF) are synthetic antibiotics that have been used in the treatment of infectious diseases in both human and animals. In the People's Republic of China, the use of LOM and PEF in livestock has been prohibited because of the concern that the residues of these drugs may pose a risk to public health. Despite this prohibition, these drugs are still being used in the poultry industry illegally, and so far there has been no systematic study of the persistence of LOM and PEF residues in chickens. In this study, laying hens were treated with a daily dose (10 mg/kg of body weight) of LOM or PEF for five consecutive days, and the drug residues in various tissues and eggs were determined over a 15-day period after the last drug administration. The highest LOM and PEF residual concentrations were found in the tissues 4 h after the last drug administration, and concentrations gradually decreased over time. Plasma had the lowest and liver had the highest residual concentrations throughout the 15-day study period. At the end of the 15 days, 3.64 ± 0.74 μg/kg LOM and 1.78 ± 0.28 μg/kg PEF were detected in the liver, with slightly lower residual concentrations in the kidney. No LOM or PEF residue was detected in the ovarian follicle, plasma, and muscle at the end of the 15 days. In eggs, the depletion rate of LOM was slower than that of PEF. LOM and PEF residues were detected in whole eggs for up to 10 and 8 days, respectively, after drug administration ceased. These findings suggest that the liver and, to a lesser extent, the kidney may be the sites where LOM or PEF residues would persist. This information can be a reliable reference for governmental agencies with respect to the screening of LOM and PEF residues in food products derived from laying hens.

Organochlorine pesticides such as dichlorodiphenyltrichloroethane (DDT) have been used in agriculture and for disease control purposes over many decades. Reports suggest that DDT exposure may result in a number of adverse effects in humans. In the KwaZulu-Natal Province of South Africa, DDT is sprayed annually in homes (indoor residual spraying) to control the mosquito vector of malaria. In the northern part of the Province, samples of free-range chicken meat (n = 48) and eggs (n = 13), and commercially produced chicken meat (n = 6) and eggs (n = 11), were collected and analysed. Of the free-range chicken meat samples, 94% (45/48) contained DDTs (ΣDDTs median 6.1 ng/g wet weight [ww], maximum 79.1 ng/g ww). Chicken egg contents were also contaminated (ΣDDTs in free-range eggs median 9544 ng/g ww, maximum 96.666 ng/g ww; and in commercial eggs median 1.3 ng/g ww, maximum 4.6 ng/g ww). The predominant DDT congener detected was p,p'-DDE in both free-range meat (>63%) and eggs (>66%), followed by p,p'-DDT and then p,p'-DDD. Based on estimated daily intake values, calculated human risk ratio (carcinogenic) values were >1 for DDTs detected in both free-range chicken products. Consumption of free-range eggs poses a particularly high health risk.
