The efficiency of a simple mechanical extraction system as applied in veterinary drug analysis has been tested in the field of pesticide residue analysis. As a first application, the system was used for the extraction of organochlorine pesticides from vegetables. The convenience of this simple extraction system consists of performing mechanical extraction in disposable polyethylene-based extraction bags, reducing considerably manual operation and cross-contamination. For the tested compounds and matrix (lettuce), recoveries of the 10 organochlorine pesticides performed at two spiking levels (n = 4 each level and compound) ranging between 0.06 and 3.3 mg/kg were between 60 and 80%, with most standard deviations

Long-term exposure to fat-soluble xenobiotics is assessed by the concentration of DDE [1,1-dichloro-2,2-bis(p-chlorophenyl)ethyl-ene], the persistent metabolite of DDT, in subcutaneous fat, aspirated from the buttocks of breast cancer patients and age-matched controls, from five European centers collaborating in a case-control study on breast cancer. In such studies using sample material of living subjects only small amounts of samples can be made available for analysis. In this particular study the only sample material available for the analysis of DDE were aliquots of the aspirates that were originally analyzed for fatty acids. Due to the small sample quantities available, e.g. aliquots of 200–800 p. 1, on-line LC-GC is most convenient because a major part of the sample can be used in the analytical procedure. In the LC-GC procedure 50 μl of sample was injected on the LC column resulting in a 180 μl fraction containing the analytes of interest. The LC fraction was transferred to a capillary GC with electron capture detector by means of partially concurrent solvent evaporation. This way, sample handling is minimized thus reducing losses and preventing contamination. The feasibility of the on-line LC-GC system for the analysis of DDE is demonstrated with the analysis of 634 adipose tissue extracts in different series. The validity of this approach, using samples already analyzed for another parameter, with LC-GC is clearly demonstrated by the fact that in over 97% of the samples DDE can be quantified. Thus rendering a meaningful data-set for further epidemiological evaluation. The DDE levels found ranged from 0.99–3.13 μg/g adipose tissue.

A clean-up method for organochlorine compounds in fatty samples based on normal-phase liquid chromatography (NPLC) is described. To this end, an existing clean-up procedure which uses column switching for the separation of organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) from the fat matrix was simplified to a single silica LC column procedure. The use of an LC column packed with 3 microns silica enables complete fat/OCP separation in a total fraction volume of 12 ml, and results in a fully automated clean-up procedure that takes only 32 min per sample. The method showed average recoveries of 80-110% in the concentration range of 1-510 micrograms/kg, with relative standard deviations of less than 10%. Limits of determination were in the range of 0.5-50 micrograms/kg. The simplified approach has shown its potential for a variety of samples, such as milk, pork fat, animal feed and cod liver oil, showing its general applicability to fatty samples.

The advantages and drawbacks of coupled chromatographic techniques are discussed on the basis of selected applications of LC-LC and LC-GC in the analysis of pesticide residues applied to environmental and biological samples.

Normal-phase LC (NPLC) is a powerful method for the clean-up of fatty samples in the determination of organochlorine pesticides (OCPs). The injected sample deactivates the stationary phase and the triglyceride matrix therefore serves as a polarity modifier in the NPLC separation. Thus, the amount of sample injected is the key to both selectivity and sensitivity in matrixmodified LC coupled to capillary GC.

Off-line normal-phase LC has been used for the clean-up of compounds in our laboratory for several years. On-line coupling of this LC system, which typically yields 12 ml fractions, is not possible due to its large fraction volume. The maximum transfer volume in on-line LC-GC/ECD is approx. 300 μl. Therefore down-scaling of the LC system was attempted in order to reach these low fraction volumes. Miniaturization resulted in a 240 μ1 LC fraction containing the analytes of interest, which is transferred to GC/ECD via an on-column interface. Sensitivity requires that a minimum amount equivalent to 1–2 mg of sample should reach the GC detector; the selectivity is determined by the separation between the matrix and the last eluting target analyte. For the analysis of fatty samples, limitations were observed in the separation of dieldrin from triglycerides. Other organochlorine compounds, e.g. polychlorobiphenyls (PCBs), the DDT group, HCB and the HCHs can be analyzed with RSDs of 2–4 % (n = 10) at concentration levels of sub-μ/kg in milk fat using a 3 μm Hypersil silica 50 × 1.0 mm i.d. LC column.

The combination of automated solid-phase extraction (SPE) and large-volume introduction gas chromatography electron-capture detection (GC-ECD) is used for the determination of synthetic pyrethroids in surface and drinking water. The selectivity that is needed for the use of GC-ECD of environmental samples is achieved by addition of a polarity modifier to the sample. In this study, sufficient methanol was added to 19 ml of sample to obtain a level of 30% of modifier prior to SPE. The cartridge is washed with one additional ml of 30% methanol in water. After drying, the cartridge is eluted with toluene. GC-ECD analysis is performed by on-column introduction of 100 μl of the toluene extract. The whole procedure was automated by using an automated solid-phase extraction apparatus. Limits of detection (LODs) for the pyrethroids in surface water are in the range of 1 to 8 ng/l (signal-to-noise ratio=3); even lower limits of detection are possible for drinking water.

A method has been developed for the automated sample pretreatment of organochlorine pesticides (OCP's) and polychlorinated biphenyls (PCB's) in extracts of human milk. This work was part of a regular monitoring program presently carried out at our institute. In this program several hundreds of human milk samples have to be analyzed for the occurrence of PCB's and OCP's. With a normal bore straight phase HPLC system, utilizing column switching we are able to separate the fat from the compounds of interest and, moreover, complete separation of the PCB fraction from the OCP's can be achieved. Under the conditions used to separate the PCB fraction from intefering OCP's column-switching is essential since the retention times for the OCP's vary from 4 minutes for hexachlorobenzene (HCB) to more than 2 hours for dieldrin. 1 ml of an extract containing 45 mg of fat is injected on the first (pre)column, the fat is retained on this column and the early eluting HCB, the PCB fraction, and the DDT complex are transferred to the second (analytical) column. Compounds eluting later than p,p′-DDT are collected directly from the precolumn. Meanwhile, the PCB fraction is separated from the rest of the OCP's on the analytical column. Contrary to conventional gravity-controlled chromatography or solid phase extraction the clean-up process can be monitored on-line by UV-detection, thus rendering a fast and reliable optimization of the system. The OCP-fractions collected from the LC are pooled before they are transferred to a high resolution gas chromatograph equipped with a large volume option. The PCB-fraction is injected directly in a HRGC equipped with a concurrent solvent evaporation injection device. The limits of detection for the OCP's (HCB, α-,β- and γ-HCH, β-Hepo (heptachlorepoxide), dieldrin, p,p′-DDE, o,p′-DDT p,p′-DDT and TDE) and the PCB's investigated are at sub-ppb level (fat basis); the recoveries vary from 80 to 100%.
