We explore the use of preparative size-exclusion chromatography (SEC) and high-performance liquid chromatography (HPLC) to purify quantum dots (QDs) after surface modification. In one example, in which Bio-Beads (S-X1) were used as the packing material for the preparative SEC column, CdSe QDs treated with a functional coumarin dye could be separated from the excess free dye by using tetrahydrofuran (THF) as the mobile phase. This column was unable to separate polymer-coated QDs from free polymer (M approximately 8000) because of the relatively low cutoff mass of the column. Here a preparative HPLC column packed with TOYOPEARL gel allowed the effective separation of polymer-bound QDs from the excess free polymer by using N-methyl-2-pyrrolidinone (NMP) as the mobile phase. When other solvents such as absolute ethanol, acetonitrile, THF, and THF-triethylamine mixtures were used as the eluent, QDs stuck to the column. While NMP was an effective medium to remove excess free polymer from the QDs, it was difficult to transfer the purified QDs to more volatile solvents and maintain colloidal stability.

Pulsed field gradient nuclear magnetic resonance (PFG NMR) experiments have been used to examine ligand exchange between poly(2-(N,N-dimethylamino)ethyl methacrylate) (PDMA) (Mn = 12,000, Mw/Mn = 1.20, Nn = 78) and trioctylphosphine oxide (TOPO) bound to the surface of CdSe/TOPO quantum dots (QDs). We show that PFG 1H NMR can quantify the displacement of TOPO by PDMA through its ability to differentiate signals due to TOPO bound to the QDs versus those from TOPO molecules free in solution. For CdSe QDs with a band edge absorption maximum at 558 nm (diameter 2.7 nm by transmission electron microscopy), we determined that, at saturation, 8 polymer chains on average displace greater than 90% of the surface TOPO groups. At partial saturation, with an average of 6 polymer chains/QD, each TOPO displaced requires 28 DMA repeat units. Assuming that one Me2N- group binds to a surface Cd2+ for each TOPO displaced, we infer that only about 3% of the DMA units are directly bound to the surface. The remaining groups are present as loops or tails that protrude into the solvent and increase the hydrodynamic diameter of the particles.

Control of the intermolecular aggregation of organic π‐conjugated molecules as chromophores is crucial for tuning their physical properties such as light absorption/emission, and energy and charge transfer. Lots of advances have been achieved in control of intermolecular aggregation of organic chromophores in solid states where an indefinitely large number of molecules are involved. However, much less understanding has been gained at a mesoscale of aggregates formed by well‐defined organization of a deterministic number of chromophores, which has been realized in natural photosynthetic systems but still remains rare in manmade materials. Here, we report both the kinetic and the thermodynamic control of the supramolecular aggregation of a near‐infrared cyanine dye, PPcy, and its derivatives confined in colloidal nanoparticles stabilized by surfactants in aqueous media. Our results demonstrate that both the aggregation number, the aggregation state and the optical properties of the PPcy chromophores are controllable through optimization of the alkyl and polymer chains tethered from PPcy, the effective concentration of the chromophore inside each particle, and the surfactants utilized to stabilize the colloids in water. Colloidal nanoparticles (NPs) containing near-infrared-fluorescent J-aggregates (JAGGs) of pyrrolopyrrole cyanines (PPcys) stabilized by amphiphilic block co-polymers were prepared in aqueous medium. JAGG formation can be tuned by means of the chemical structure of PPcys, the concentration of chromophores inside the polymeric NPs, and ultrasonication. The JAGG NPs exhibit a narrow emission band at 773 nm, a fluorescence quantum yield comparable to that of indocyanine green, and significantly enhanced photostability, which is ideal for long-term bioimaging. Near‐infrared fluorescent J‐aggregates were discovered in colloidal nanoparticles of pyrrolopyrrole cyanines stabilized by amphiphilic block co‐polymers in aqueous media. The formation of such colloidal‐state J‐aggregates can be tuned by the chemical structure and the concentration of the chromophores inside the polymeric nanoparticles, and can be further enhanced by ultrasonic treatment. These J‐aggregate nanoparticles with an average diameter of 30 nm, exhibit a narrow emission band at 773 nm, a fluorescence quantum yield comparable to that of indocyanine green, and significantly enhanced photostability that is ideal for long‐term bioimaging.
