In the present study, the membrane lipid composition of corals from a region with tidally induced upwelling was investigated. The coral community is subject to strong temperature oscillations yet flourishes as a result of adaptation. Glycerophosphocholine profiling of the dominant pocilloporid coral, Seriatopora caliendrum, was performed using a validated method. The coral inhabiting the upwelling region shows a definite shift in the ratio of lipid molecular species, covering several subclasses. Mainly, the coral possesses a higher percentage of saturated, monounsaturated and polyunsaturated plasmanylcholines and a lower percentage of polyunsaturated phosphatidylcholines. Higher levels of lyso–plasmanylcholines containing saturated or monounsaturated fatty acid chains were also revealed in coral tissue at the distal portion of the branch. Based on the physicochemical properties of these lipids, we proposed mechanisms for handling cellular membrane perturbations, such as tension, induced by thermal oscillation to determine how coral cells are able to spontaneously maintain their physiological functions, in both molecular and physical terms. Interestingly, the biochemical and biophysical properties of these lipids also have beneficial effects on the resistance, maintenance, and growth of the corals. Lipids are a source of metabolic energy, as well as essential components of cellular membranes. Although they have been shown to be key players in the regulation of cell proliferation in various eukaryotes, including microalgae, their role in the cell cycle of cnidarian-dinoflagellate (genus Symbiodinium) endosymbioses remains to be elucidated. The present study examined the effects of a lipid synthesis inhibitor, cerulenin, on the cell cycle of both cultured Symbiodinium (clade B) and those engaged in an endosymbiotic association with the sea anemone Aiptasia pulchella. In the former, cerulenin exposure was found to inhibit free fatty acid (FFA) synthesis, as it does in other organisms. Additionally, while it also significantly inhibited the synthesis of phosphatidylethanolamine (PE), it did not affect the production of sterol ester (SE) or phosphatidylcholine (PC). Interestingly, cerulenin also significantly retarded cell division by arresting the cell cycles at the G0/G1 phase. Cerulenin-treated Symbiodinium were found to be taken up by anemone hosts at a significantly depressed quantity in comparison with control Symbiodinium. Furthermore, the uptake of cerulenin-treated Symbiodinium in host tentacles occurred much more slowly than in untreated controls. These results indicate that FFA and PE may play critical roles in the recognition, proliferation, and ultimately the success of endosymbiosis with anemones. [ABSTRACT FROM AUTHOR]

A new polyoxygenated briarane-type diterpenoid, briarenol A ( 1 ), has been isolated from a gorgonian coral belonging to the genus Briareum . The structure, including the relative stereochemistry of briarane 1 , was elucidated by the interpretation of spectral data. Abstract: Three polyoxygenated briaranes, including two new compounds, excavatoids A (1) and B (2), and a known metabolite, briaexcavatin I (3), were isolated from the cultured octocoral Briareum excavatum. Moreover, the wild type B. excavatum, collected off southern Taiwan coast, yielded two new 5,6-epoxybriaranes, excavatoids C (4) and D (5). The structures of new compounds 1, 2, 4, and 5 were determined by spectroscopic methods and the structure of 1 was further confirmed by X-ray diffraction data analysis. The X-ray structure for briaexcavatin I (3) was also reported for the first time. Excavatoid A (1) is the first briarane which possesses six hydroxy groups and a 17-methoxy group. Excavatoid C (4) is the first 12,13-secobriarane which possesses a novel pentacyclic skeleton with an ɛ-lactone. Excavatoid D (5) displayed moderate inhibitory effects on superoxide anion generation and elastase release by human neutrophils. [Copyright &y& Elsevier]
