Changes in ACTH challenge test characteristics in dairy cows changing their physiological status at different lactational stages and different feeding levels were not investigated in terms of repeatability yet. In 23 multiparous Holstein cows (10 cows fed a sole fresh herbage diet without concentrate, 13 cows fed with concentrate), three ACTH challenge tests were performed: once during pregnancy shortly prior to drying off ( T1), and in week 3 ( T2) and 8 ( T3) after parturition. Test characteristics were correlated to performance and metabolic parameters: DMI, BW, energy balance ( EB), plasma concentrations of free fatty acids ( NEFA) and beta-hydroxybutyrate ( BHB). Basal plasma cortisol concentrations were higher at T1 compared with T2 and T3 ( p < .05). The adrenal cortex sensitivity (expressed as total AUC ( AUCt) of cortisol response after ACTH application) was lowest at T2 compared with T1 and T3 ( p < .05). Ranking of the individual animals' responses was not repeatable between time points of the ACTH tests. Enhancing the energy deficiency during early lactation by omission of concentrate did not affect baseline cortisol concentrations in plasma, but decreased peak height at T2 ( p < .05). Baseline plasma cortisol concentrations were positively correlated with cortisol peak values after ACTH application, previous lactation performance, milk yield and BW ( p < .05). The AUCt was positively correlated with baseline cortisol concentrations, EB and DMI. Cortisol release after ACTH injection was lower in animals with high plasma concentrations of NEFA, BHB and with higher contents of fat and free fatty acids in milk ( p < .05). Cortisol peak height after ACTH administration was higher in cows with a more positive EB, higher DMI and lower plasma concentrations of NEFA and BHB. In dairy cows, hypocalcemia is caused by the sudden calcium demand by the mammary gland at the onset of lactation. Calcitriol (1,25-dihydroxy vitamin D; 1,25-VitD) increases the intestinal calcium absorption and the renal calcium reabsorption. Daylight contributes to the formation of 1,25-VitD, as it transforms 7-dehydrocholesterol into cholecalciferol, a 1,25-VitD precursor. Calving pens are usually set in quiet places where cows can stay calm and relaxed before parturition. However, those pens often have poor lighting conditions and therefore cows may become vitamin D deficient because of inadequate daylight exposure. Therefore, we have tested the hypothesis that direct daylight exposure supports the synthesis of 1,25-VitD and consequently attenuates the decline of calcium concentrations at parturition. Twenty Holstein dairy cows were randomly assigned to 2 experimental groups (daylight group, DL; and control group). Beginning on d -10 before expected parturition, both groups were placed in a standard indoor calving pen from 1700 h to 800 h. From 800 h to 1700 h cows from the DL group were moved into a contiguous open pen with direct access to daylight whereas controls remained at the standard indoor calving pen. After parturition both groups were permanently placed in an indoor calving pen until the end of the experimental period (d 30 postpartum). Blood samples were collected daily from d -10 prior to expected parturition through d 7 postpartum with an additional sample on d 30 postpartum. Milk yield was recorded at each milking during the whole experimental period. In the DL group, 25-hydroxyvitamin D (25-VitD), 1,25-VitD and total calcium concentrations around parturition were higher than in the controls. Higher parathyroid hormone concentrations were observed in the control group compared to the DL group at parturition. Pyridinoline concentrations did not differ between groups on d 1, and therefore it is assumed that the intensity of calcium transfer from the bones to the bloodstream was not affected by the increased daylight exposure. The measured plasma metabolites (β-hydroxybutyrate, fatty acids and glucose) as well as milk yield were not affected by the increased daylight exposure. In conclusion, the increased daylight exposure before parturition increased 25-VitD and 1,25-VitD concentrations, preventing a considerable decline of total calcium concentrations around parturition. Lactation evolved to be the core functional system of providing maternal care in mammalian species. The mammary gland provides an ideally composed nutrient source for the newborn. In addition, colostrum provides passive immunisation after birth, and each suckling process supports the establishment and maintenance of a close mother-offspring bonding. The importance of lactation for the survival of the offspring is represented by a high metabolic priority of the mammary gland within the organism of the lactating animal. Therefore, animal breeding for high milk production has been quite successful, and modern breeding methods have allowed an enormous increase in the performance within only few generations of cows. Mainly in early lactation, most of the available nutrients are directed to the mammary gland, both those from feed, and those mobilised from body tissue. Therefore, milk production can be maintained at a high level despite a negative energy balance. However, the high metabolic load and mobilisation of body tissue requires adequate endocrine and metabolic regulation, which can be successful or less successful in individual animals, i.e. the dairy cow population consists of both metabolically robust and metabolically vulnerable dairy cows. While robust animals adapt adequately, vulnerable cows show often high plasma concentrations of non-esterified fatty acids and b-hydroxybutyrate, and are prone to various production-related diseases. In pasture- or forage-based feeding systems, an additional challenge is a limited availability of nutrients for milk production. Forage feeding without complementary concentrate leads to enormous metabolic disorders in high-yielding cows, but is tolerated in dairy cows with a moderate genetic-performance level. Fifty-one dairy cows with subclinical ketosis were used to investigate the effects of butafosfan alone or in combination with cyanocobalamin on metabolism. Treatments included i.v. injection of 10 ml/100 kg of body weight with butafosfan ( BUT) or combined cyanocobalamin with butafosfan ( BUTCO) at a similar concentration as in Catosal®. Control cows ( CON) received a 0.9% saline solution. Cows were injected on days 1-3 at 22.3 ± 0.7 days post-partum. Milk production and composition were not affected by the treatments. In plasma, CON cows had a significantly higher plasma NEFA concentration (0.59 ± 0.03 m m) across the study period than BUTCO cows (p < 0.05; 0.42 ± 0.03 m m), whereas the plasma NEFA concentration of BUT was intermediate (0.52 ± 0.03 m m) but not significantly different from CON. Both BUTCO and BUT cows had lower (p < 0.05) plasma BHBA concentrations (1.02 ± 0.06 m m and 1.21 ± 0.06 m m, respectively) across the study period than CON (1.34 ± 0.06 m m). Plasma glucose was not different between treatments, but plasma glucagon concentrations were consistently high in BUT compared to BUTCO and CON. Lowest post-treatment glucagon levels were observed in BUTCO. Hepatic mRNA abundance of liver X receptor α, a nuclear receptor protein involved in lipid metabolism, was higher in BUTCO compared to BUT and CON (p < 0.05) on day 7. Furthermore, on day 7, the mRNA abundance of beta-hydroxybutyrate dehydrogenase 2 was higher in BUTCO compared to BUT and CON (p < 0.01). In conclusion, injections of combined cyanocobalamin with butafosfan post-partum in early lactation ketotic dairy cows act on lipid metabolism with effects on plasma metabolites, most likely mediated via modified activity of key factors in the liver. Cortisol in dairy cows is released in an episodic manner underlying a circadian rhythm. The involvement of cortisol in numerous adaptive processes to cope with adverse conditions such as pain and inflammation is well characterized. Recent studies described contradictory effects of nutrition and metabolism on the secretory pattern of cortisol. However, up to now, the direct effects of single metabolites during various metabolic conditions without the profound endocrine changes around parturition on the glucocorticoid secretion in dairy cows have not been described. The objective of this study was to investigate the effects of long-term (56-h) manipulated metabolic states, that is, manipulated plasma concentrations of glucose and β-hydroxybutyrate (BHBA), on the release of cortisol in midlactation dairy cows. Besides the concentration of cortisol at defined time points, its pulsatile secretory pattern was studied in combination with an acute immune challenge through an intramammary lipopolysaccharide (LPS) challenge. Twenty-five midlactation dairy cows were randomly assigned to 1 of 4 treatments (hyperinsulinemic hypoglycemic clamp [HypoG], hyperinsulinemic-euglycemic clamp [EuG], continuous infusion of BHBA [HyperB], or infusion of saline solution for the control group [Control). Different metabolic states induced by infusion treatments affected the characteristics of cortisol secretion (elevation of baseline [the HypoG and HyperB treatments] and decreased peak length [the HypoG treatment]; P < 0.05), whereas amplitude, peak interval, height, peak area, area under the curve (AUC) above the baseline cortisol concentration (AUCb), and the total AUC (AUCt) were not different between infusion treatments. The induced inflammatory response due to the intramammary LPS challenge at simultaneously maintained infusion treatments diminished the pulsatile nature of cortisol release, whereas AUCb (and AUCt, respectively) was lowest for the HypoG treatment compared with the HyperB and Control treatments (P < 0.05). This study indicates that single metabolites (glucose and BHBA) and their availability or turnover (in case of glucose) have a different impact on the regulation of cortisol secretion resulting in changes of its pulsatile release. Furthermore, cortisol release during intramammary inflammation was found to be greater in the HyperB, EuG, and Control treatments compared with the HypoG treatment (P < 0.05). Control of metabolic pathways is a major task of the somatotropic axis and its constituents. Insulinlike growth-factor binding proteins (IGFBPs) bind IGF-I and -II and act as carriers and regulators of their activities in blood, body fluids and tissues. Over two periods of physiological adaptation, this study investigated the binding pattern of IGF-I to IGFBPs in the plasma of 50 multiparous Holstein dairy cows and identified relationships with the hepatic mRNA abundance of IGFBPs and plasma IGF-I during the lactational negative energy balance (NEB) and during a deliberately induced NEB by feed restriction. Period 1 lasted from week 3 antepartum (a.p.) to week 12 postpartum (p.p.) and period 2, the period of feed restriction, started at around 100 DIM and lasted for three weeks with a control (C) and a restricted group (R). Blood samples and liver biopsies were collected in week 3 a.p., and in weeks 1 and 4 p.p. of period 1 and in weeks 0 and 3 of period 2. For column chromatography of IGFBPs, plasma samples of all animals were pooled by group and time points of sampling. Plasma IGF-I dropped from week 3 a.p. to week 1 p.p. and thereafter increased until week 0 (period 2) and did not change up to week 3 of period 2. The binding of IGF-I to plasma IGFBP-1 and -2 increased in period 1 from week 3 a.p. to week 4 p.p., while at the same time it decreased for IGFBP-3. During period 2, the binding of IGF-I to plasma IGFBP-1 and -2 decreased for both groups, but less for R cows. In C cows, the IGF-I binding to IGFBP-3 in plasma increased from week 0 to week 3 of period 2, whereas R cows showed a slight decrease. In period 1, hepatic mRNA abundance of IGFBP-3 followed the plasma IGFBP-3 binding in contrast to the mRNA abundances of IGFBP-1 and -2. The latter increased from week 3 a.p. to week 1 p.p. and decreased afterwards whereas IGF-I binding to IGFBP-1 and -2 increased. In week 3 of period 2, the binding of IGF-I to IGFBP-1 and -2 and their hepatic mRNA abundance were higher in R cows compared to C cows. Hepatic mRNA abundance of IGF-I was consistently positively correlated with plasma IGF-I, especially pronounced during the NEBs in week 1 p.p. (period 1) and in week 3 (period 2) in R cows. While no distinct relation between mRNA abundance of IGFBP-1 and plasma IGF-I was evident, the mRNA abundance of IGFBP-2 was inversely related to plasma IGF-I over all experimental time points independent of treatment. The mRNA abundance of IGFBP-3 was particularly correlated with plasma IGF-I during the 2 experimental stages of a NEB. Obviously IGFBP-3, but not IGFBP-1 and -2, binding in plasma closely followed the respective pattern of hepatic mRNA abundance during the entire experimental period. The fact that changes in the different plasma IGFBPs during altering metabolic stages in different stages of lactation do not always strictly follow their mRNA abundance in liver suggests tissues other than the liver flexibly contributing to the IGFBP pool in plasma as well as a partially post-transcriptional regulation of IGFBP synthesis. Gene expression of adipose factors, which may be part of the mechanisms that underlie insulin sensitivity, were studied in dairy cows around parturition. Subcutaneous fat biopsies and blood samples were taken from 27 dairy cows in week 8 antepartum (a.p.), on day 1 postpartum (p.p.) and in week 5 p.p. In the adipose tissue samples, mRNA was quantified by real-time reverse transcription polymerase chain reaction for tumour necrosis factor alpha (TNFα), insulin-independent glucose transporter (GLUT1), insulin-responsive glucose transporter (GLUT4), insulin receptor, insulin receptor substrate 1 (IRS1), insulin receptor substrate 2 (IRS2), regulatory subunit of phosphatidylinositol-3 kinase (p85) and catalytic subunit of phosphatidylinositol-3 kinase. Blood plasma was assayed for concentrations of glucose, β-hydroxybutyric acid, non-esterified fatty acids (NEFA) and insulin. Plasma parameters followed a pattern typically observed in dairy cows. Gene expression changes were observed, but there were no changes in TNFα concentrations, which may indicate its local involvement in catabolic adaptation of adipose tissue. Changes in GLUT4 and GLUT1 mRNA abundance may reflect their involvement in reduced insulin sensitivity and in sparing glucose for milk synthesis in early lactation. Unchanged gene expression of IRS1, IRS2 and p85 over time may imply a lack of their involvement in terms of insulin sensitivity dynamics. The application of prestimulation results in enhanced milking performance compared with milking without prestimulation. In the present study oxytocin (OT) release and milking characteristics were investigated in 43 dairy cows after the application of various prestimulation routines by vibration stimulation lasting between 0 and 90 s. Additionally, different maximum pulsation vacuum settings during vibration stimulation were investigated. The actual degree of udder fill was calculated as a percentage of the estimated storage capacity. The amplitude of OT release, total milk yield, and stripping milk yield did not differ between prestimulation routines. Increased maximum pulsation vacuum during vibration stimulation resulted in milk flow during prestimulation, but did not negatively influence milking characteristics. The lag time from the start of teat stimulation until the start of milk ejection was negatively correlated with the degree of udder fill. This relationship was the reason for variations in optimal duration of prestimulation. The optimal duration of prestimulation to receive immediate and continuous milk flow at the start of milking was 90 s in udders containing small amounts of milk, whereas the optimal duration was only 20 s in well-filled udders. A short prestimulation enhances milking stall capacity when milking full udders, and a prolonged prestimulation reduces the total vacuum load on the teat when milking udders that are not full. The acyclic postpartum period in cows is prolonged due to frequent suckling when compared with twice daily milking. In conventional milking systems twice daily milking is common, while in automatic milking systems (AMS) three or more milkings are performed in early lactation. In this study, the hypothesis was tested that an increased milking frequency in AMS causes a delay in resumption of ovarian cyclicity postpartum and increases days open in German Fleckvieh cows (n = 124). Calvings were equally distributed throughout the 1-year experimental period. All cows were housed in one barn. Sixty-three cows were milked in an AMS (AC) and 61 cows were twice daily milked in a conventional milking parlour (PC). Forty-two AC and 36 PC remained in the herd until conception. After parturition, milk samples were analysed for progesterone twice weekly to record the ovarian activity until pregnancy was diagnosed. The first and the second ovulation were determined by the progesterone profile of each individual cow. The mean milking frequency during the first 3 weeks after parturition was higher in AC (3.17 ± 0.09 milkings/day) than in PC (milked twice daily). The individual mean milking frequency of AC for the first 3 weeks of lactation ranged from 2.0 to 5.9 daily milkings. The time span from parturition until the first and second ovulation did not differ between AC and PC. However, first ovulation was observed earlier in primiparous (23 ± 1 days) than in multiparous cows (29 ± 1 days,P < 0.05). Within AC no relationship between milking frequency and duration of the acyclic period was observed. Days open did not significantly differ between AC and PC and between primiparous and multiparous cows. Twenty-nine pairs of high-yielding dairy cows (HC; ≥45 kg/day reached at least once during lactation) and corresponding control cows (CC; with milk yields representing the average yield of the herds) were examined on 29 Swiss farms from March 1995 to September 1996. The hypotheses were tested that there are differences in feed intake, body-conformation traits, body weight (BW), body condition score (BCS), fertility status and disease incidence between HC and CC cows. Cows were studied 2 weeks before and at 5, 9, 13, 17 and 40 weeks post-partum. HC cows produced more energy-corrected milk (ECM) than CC cows (10 670 ± 321 kg in 293 ± 5 days and 8385 ± 283 kg in 294 ± 4 days, respectively; P ≤ 0.001) and yields in the first 100 days of lactation were greater in HC than in CC cows (46.2 ± 1.1 and 36.2 ± 1.0 kg ECM/day, respectively; P ≤ 0.001). Concentrate intake was greater (P ≤ 0.05) in HC than in CC cows (7.6 ± 0.5 and 5.7 ± 0.5 kg/day, respectively) and dry matter intakes (measured in week 5 of lactation over 3 days on six farms) were greater in HC than in CC cows (24.0 ± 1.1 and 20.3 ± 1.1 kg/day, respectively; P ≤ 0.001). HC cows were taller than CC cows (wither heights 143.3 ± 0.8 and 140.1 ± 0.8 cm, respectively; P ≤ 0.01). Although BW in HC cows was greater than in CC cows throughout the study, differences and decreases of BW during lactation were not significant. BCS at the end of pregnancy and decrements during lactation were similar in HC and CC cows. Fertility parameters were similar in HC and CC cows. Incidences of mastitis, claw and feet problems, hypocalcemia/downer cow syndrome, ovarian cysts and abortions were similar in HC and CC cows, but there were more indigestion problems in HC than in CC cows. Omitting the dry period (DP) generally reduces milk production in the subsequent lactation. The aim of this study was to evaluate the effect of dietary energy source—glucogenic (G) or lipogenic (L)—and energy level—standard (std) or low—on milk production; energy balance (EB); lactogenic hormones insulin, insulin-like growth factor 1 (IGF-1), and growth hormone (GH); and lactation curve characteristics between wk 1 and 44 postpartum in cows after a 0-d or 30-d DP. Cows (n = 110) were assigned randomly to 3 transition treatments: a 30-d DP with a standard energy level required for expected milk yield [30-d DP(std)], a 0-d DP with the same energy level as cows with a 30-d DP [0-d DP(std)], and a 0-d DP with a low energy level [0-d DP(low)]. In wk 1 to 7, cows were fed the same basal ration but the level of concentrate increased to 6.7 kg/d for cows fed the low energy level and to 8.5 kg/d for cows fed the standard energy level in wk 4. From wk 8 postpartum onward, cows received a G ration (mainly consisting of corn silage and grass silage) or an L ration (mainly consisting of grass silage and sugar beet pulp) with the same energy level contrast (low or std) as in early lactation. Cows fed the G ration had greater milk, lactose, and protein yields, lower milk fat percentage, greater dry matter and energy intakes, and greater plasma IGF-1 concentration compared with cows fed the L ration. Dietary energy source did not affect EB or lactation curve characteristics. In cows with a 0-d DP, the reduced energy level decreased energy intake, EB, and weekly body weight gain, but did not affect milk production or lactation curve characteristics. A 30-d DP resulted in a greater total predicted lactation yield, initial milk yield after calving, peak milk yield, energy intake, energy output in milk, days to conception [only when compared with 0-d DP(low)], plasma GH concentration [only when compared with 0-d DP(std)], and decreased weekly body weight gain compared with a 0-d DP. A 30-d DP decreased both the increasing and the declining slope parameters of the lactation curve and the relative rate of decline in milk yield (indicating greater lactation persistency) compared with a 0-d DP, and decreased plasma insulin and IGF-1 concentration, and EB. In conclusion, feeding a G ration after wk 7 in milk improved energy intake and milk production, but did not affect EB compared with an L ration. For cows without a DP, a reduced dietary energy level did not affect milk production and lactation curve characteristics, but did decrease EB and weekly body weight gain. This study tested the hypothesis that adrenaline has different effects on blood plasma glucose, lactate and nonesterified fatty acid (NEFA) concentrations in early and mid lactation and that the feeding of crystalline fat (consisting of triglycerides) and free fatty acids (FFA), compared with a starch-rich ration, modifies the effects of adrenaline in dairy cows. Glucose, lactate and NEFA responses to intravenously infused adrenaline [15 μg/kg × (min) for 10 min] were studied in weeks 9 and 19 of lactation in cows whose diets were supplemented with crystalline fat or FFA or a starch-rich ration. Concentrations of glucose, lactate and NEFA rapidly increased up to 15 min after the start of adrenaline infusions. Glucose responses to adrenaline were comparable in weeks 9 and 19 of lactation, whereas the lactate responses were smaller, and those of NEFA were greater in week 9 than in week 19 of lactation. As the feeding of crystalline fat, FFA or a starch-rich ration did not modify the effects of adrenaline on glucose, lactate or NEFA, the target tissue sensitivity and/or responsiveness to adrenaline had obviously not been changed. The transition from the nonlactating to the lactating state represents a critical period for dairy cow lipid metabolism because body reserves have to be mobilized to meet the increasing energy requirements for the initiation of milk production. The purpose of this study was to provide a comprehensive overview on cholesterol homeostasis in transition dairy cows by assessing in parallel plasma, milk, and hepatic tissue for key factors of cholesterol metabolism, transport, and regulation. Blood samples and liver biopsies were taken from 50 multiparous Holstein dairy cows in wk 3 antepartum (a.p.), wk 1 postpartum (p.p.), wk 4 p.p., and wk 14 p.p. Milk sampling was performed in wk 1, 4, and 14 p.p. Blood and milk lipid concentrations [triglycerides (TG), cholesterol, and lipoproteins], enzyme activities (phospholipid transfer protein and lecithin:cholesterol acyltransferase) were analyzed using enzymatic assays. Hepatic gene expression patterns of 3-hydroxy-3-methylglutaryl-coenzyme A (HMGC) synthase 1 (HMGCS1) and HMGC reductase (HMGCR), sterol regulatory element-binding factor (SREBF)-1 and -2, microsomal triglyceride transfer protein (MTTP), ATP-binding cassette transporter (ABC) A1 and ABCG1, liver X receptor (LXR) α and peroxisome proliferator activated receptor (PPAR) α and γ were measured using quantitative RT-PCR. Plasma TG, cholesterol, and lipoprotein concentrations decreased from wk 3 a.p. to a minimum in wk 1 p.p., and then gradually increased until wk 14 p.p. Compared with wk 4 p.p., phospholipid transfer protein activity was increased in wk 1 p.p., whereas lecithin:cholesterol acyltransferase activity was lowest at this period. Total cholesterol concentration and mass, and cholesterol concentration in the milk fat fraction decreased from wk 1 p.p. to wk 4 p.p. Both total and milk fat cholesterol concentration were decreased in wk 4 p.p. compared with wk 1 and 14 p.p. The mRNA abundance of genes involved in cholesterol synthesis (SREBF-2, HMGCS1, and HMGCR) markedly increased from wk 3 a.p. to wk 1 p.p., whereas SREBF-1 was downregulated. The expression of ABCA1 increased from wk 3 a.p. to wk 1 p.p., whereas ABCG1 was increased in wk 14 p.p. compared with other time points. In conclusion, hepatic expression of genes involved in the biosynthesis of cholesterol as well as the ABCA1 transporter were upregulated at the onset of lactation, whereas plasma concentrations of total cholesterol, phospholipids, lipoprotein-cholesterol, and TG were at a minimum. Thus, at the gene expression level, the liver seems to react to the increased demand for cholesterol after parturition. Whether the low plasma cholesterol and TG levels are due to impaired hepatic export mechanisms or reflect an enhanced transfer of these compounds into the milk to provide essential nutrients for the newborn remains to be elucidated. Negative energy balance in dairy cows in early lactation has been associated with increased inflammation and oxidative stress in these cows. The objective of this study was to evaluate the effects of dry period (DP) length and dietary energy source on inflammatory biomarkers and oxidative stress in dairy cows. Holstein-Friesian dairy cows (60 primiparous and 107 multiparous) were assigned randomly to a 3 x 2 factorial design with 3 DP length (0, 30, or 60 d) and 2 early lactation rations (glucogenic or lipogenic). Cows were fed a glucogenic or lipogenic ration from 10 d before the expected calving date. Blood was collected in wk -3, -2, -1, 1, 2, and 4 relative to calving. Dry period length affected inflammatory biomarkers and oxidative stress, especially in wk 1 and 2 after calving. Cows with a 0-d DP had higher levels of ceruloplasmin, cholesterol, and reactive oxygen metabolites, and they tended to have higher haptoglobin levels compared with cows with a 30- or 60-d DP. Cows with a 0-d DP had a lower plasma paraoxonase and bilirubin in the first 2 wk after calving and a lower liver functionality index compared with cows with a 60-d DP. Cows of parity >3 fed a glucogenic ration had higher cholesterol levels compared with cows of parity >3 fed a lipogenic ration. No interaction between DP length and ration was present for inflammatory biomarkers or oxidative stress variables. Plasma bilirubin levels for cows with a 0-d DP were negatively related to energy balance and metabolic status in these cows. Moreover, occurrence of clinical health problems (fever, mastitis, metritis, and retained placenta) was 41, 27, and 30% for cows with 0-, 30-, and 60-d DP, respectively. High levels of ceruloplasmin, cholesterol, and reactive oxygen metabolites in cows with 0-d DP were related to the occurrence of health problems in these cows. In conclusion, omitting the DP increased levels of ceruloplasmin, cholesterol, and reactive oxygen metabolites, and decreased levels of bilirubin and paraoxonase in plasma, independent of ration, compared with cows with a 60-d DP. These contrasting effects of DP length on inflammatory status could be explained in part by the improved energy balance and occurrence of health problems in these cows, but was not related to increased somatic cell count in cows with a 0-d DP. Cows with a 0-d DP had better energy balance, but also had higher levels of oxidative stress compared with cows with a 60-d DP. Milking characteristics differ between the 4 quarters of a dairy cow udder. In particular, milking time is mostly prolonged in hind quarters compared with front quarters because of the usually higher amount of stored milk. The standard milking routine (STDMR) in both conventional and automatic milking systems (AMS) consists of teat preparation of all 4 quarters, followed by attachment of the 4 teat cups, regardless of the distribution of milk between quarters. In the current study, an alternative teat preparation and milking routine (ALTMR) in AMS was tested, which consisted of cleaning and starting the milking of hind teats before cleaning and attachment of front teats. The hypothesis was based on the fact that hind quarters have usually a longer milking time than front quarters. Starting the milking of hind quarters while the front teats are being cleaned may reduce the difference in the end of milking between front and hind quarters and thus reduce total milking time. Both routines were tested on 5 Swedish dairy farms equipped with AMS in a 4-wk experiment in which treatments were alternated weekly. Total milk yield did not differ between treatments. Machine-on time (MOT) was longer in ALTMR than in STDMR because the difference in milking time between hind and front quarters was less than the time needed to prepare the front teats. However, the longer MOT in ALTMR was compensated by a shorter total preparation time, including the attachment of the first teat cup, as only the hind teats (instead of all 4 teats) were cleaned before milking was started. This resulted in a similar total milking time from start of cleaning of the first quarter until the end of milking of the last quarter in both treatments. Because of the prolonged MOT, average milk flow rate was lower in ALTMR than STDMR. Peak flow rate was higher in ALTMR than STDMR, but only in teat cups 1 (first attached, hind quarter) and 3 (third attached, front quarter), whereas main milk flow was higher in ALTMR than STDMR in both front quarters. In conclusion, splitting teat cleaning and the start of milking between hind and front quarters does not prolong total milking time, including teat cleaning. The partially positive effect on peak and main milk flow indicates that the ALTMR is a suitable milking routine in AMS. The objective of the current study was to evaluate the effect of dry period (DP) length on milk yield, energy balance (EB), and metabolic status in cows fed a lipogenic or glucogenic diet in the second year after implementation of DP and dietary treatments. Holstein- Friesian dairy cows (n = 167) were assigned randomly to 1 of 3 DP lengths (0, 30, or 60 d) and 1 of 2 early lactation diets (glucogenic or lipogenic) for 2 consecutive years. Results of the first year were reported previously. In the second year, 19 cows in the 0-d DP group were attributed to a new group (0→67 d DP) because these cows had a milk yield of <4 kg/d at least 30 d before expected calving date and were dried off. Milk yield was recorded and EB was calculated from wk -8 to 9 relative to calving. Blood samples were taken weekly from wk -3 to 8 relative to calving. Liver samples were taken in wk -2, 2, and 4 relative to calving. At the onset of lactation, cows with a 0-d or 0→67-d DP had greater body condition score (BCS) than cows with a 60-d DP. During the first 9 wk, cows with a 0- or 30-d DP produced 5.0 and 4.3 kg less milk per day, respectively, but had similar EB compared with cows with a 60-d DP. Cows with a 0- or 30-d DP produced additional milk precalving, which could compensate milk yield losses in the first 9 wk postcalving. Cows with a 0-d DP did not have milk yield losses or improve EB in the second year as much as in the first year. Cows with a 0-d DP had greater plasma insulin and insulin-like growth factor-I (IGF-I) and lower liver triacylglycerol concentrations than cows with other DP lengths. Cows with a 0→67-d DP had lower EB, and greater plasma free fatty acids (FFA) and β-hydroxybutyrate (BHB) concentrations than cows with other DP lengths. Feeding a glucogenic diet increased plasma glucose, IGF-I, and insulin concentrations, and decreased plasma FFA, BHB, and urea concentrations compared with a lipogenic diet, independent of DP length. In conclusion, omitting the DP or feeding a glucogenic diet improved metabolic status in early lactation of the second year after implementation of DP length and dietary treatments, although effects of omitting the DP were less pronounced in the second year than in the first year. To assess the regulation of mammary cell activity, survival, and proliferation by prolactin (PRL), 5 Holstein cows in early lactation received daily i.m. injections of 1 mg of quinagolide, a suppressor of PRL release, for 9 wk, whereas 4 control cows received the vehicle (water) only. During the last week of treatment, one udder half was milked once a day (1x) and the other twice a day (2x). Mammary biopsies were harvested 1 wk before and 4 and 8 wk after the start of quinagolide treatment. The quinagolide injections reduced milk yield and resulted in lower levels of κ-casein and a-lactalbumin mRNA in the mammary biopsies at wk 4 compared with the control cows. In the mammary tissue of the quinagolide-treated cows at wk 8 of treatment, cell proliferation (as determined by proliferating cell nuclear antigen labeling) was lower and apoptosis (as determined by the terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling assay) was higher than in the mammary tissue of the control cows. During differential milking, mammary epithelial cells (MEC) were extracted from the milk by centrifugation and purified by immunocytochemical binding to allow variations in the levels of mammary transcripts to be observed. After 9 wk of treatment, levels of a-lactalbumin and κ-casein mRNA were lower in the MEC isolated from milk of the quinagolide-treated cows. This effect was associated with lower PRL receptor mRNA levels and a tendency toward lower viability in the milk-isolated MEC from the 2x-milked glands. The decrease from 2x milking to 1x milking also downregulated a-lactalbumin and κ-casein transcripts in the milk-isolated MEC. Viability was higher for the MEC collected from the 1x-milked udder halves compared with the 2x-milked halves. In conclusion, the reduction in milk yield after chronic administration of the PRL-release inhibitor quinagolide is associated with a reduction in mammary cell activity, survival, and proliferation in lactating dairy cows. The effect of somatic cell count (SCC) and milk fraction on milk composition, distribution of cell populations, and mRNA expression of various inflammatory parameters was studied. Therefore, quarter milk samples were defined as cisternal (C), first 400 g of alveolar (A1), and remaining alveolar milk (A2) during the course of milking. Quarters were assigned to 4 groups according to their total SCC: 1) <12 x 10³/mL, 2) 12 to 100 x 10³/mL, 3) 100 to 350 x 10³/mL, and 4) >350 x 10³/mL. Milk constituents of interest were SCC, fat, protein, lactose sodium, and chloride ions as well as electrical conductivity. Cell populations were classified into lymphocytes, macrophages, and neutrophils (PMN). The mRNA expression of the inflammatory factors tumor necrosis factor-α, interleukin-lβ, cyclooxygenase-2, lactoferrin, and lysozyme was measured via real-time, quantitative reverse transcription PCR. Somatic cell count decreased from highest levels in C to lowest levels in A1 and increased thereafter to A2 in all groups. Fat content increased from C to A2 and with increasing SCC level. Lactose decreased with increasing SCC level but remained unchanged during milking. Concentrations of sodium and chloride, and electrical conductivity increased with increasing SCC but were higher in C than in A1 and A2. Protein was not affected by milk fraction or SCC level. The distribution of leukocytes was dramatically influenced by milk fraction and SCC. Lymphocytes were the dominating cell population in group 1, but the proportion of lymphocytes was low in groups 2, 3, and 4. Macrophage proportion was highest in group 2 and decreased in groups 3 and 4, whereas that of PMN increased from group 2 to 4. The content of macrophages decreased during milking in all SCC groups whereas that of PMN increased. The proportion of lymphocytes was not affected by milk fraction. The mRNA expression of all inflammatory factors showed an increase with increasing SCC but minor changes occurred during milking. In conclusion, milk fraction and SCC level have a crucial influence on the distribution of leukocyte populations and several milk constituents. The surprisingly high content of lymphocytes and concomitantly low mRNA expression of inflammatory factors in quarters with SCC <12 x 10³/mL indicates a different and possibly reduced readiness of the immune system to respond to invading pathogens.
