The transition period from late pregnancy to early lactation is a vital time of the lifecycle of dairy cows due to the marked metabolic challenges. Besides, the liver is the pivot point of metabolism in cattle. Nevertheless, the hepatic physiological molecular adaptation during the transition period has not been elucidated, especially from the metabolomics and proteomics view. Therefore, the present study aims to investigate the hepatic metabolic alterations in transition cows by using integrative metabolomics and proteomics methods. Gas chromatography quadrupole-time-of-flight mass spectrometry-based metabolomics and data-independent acquisition-based quantitative proteomics methods were used to analyze liver tissues collected from 8 healthy multiparous Holstein dairy cows 21 d before and after calving. In total, 44 metabolites and 250 proteins were identified as differentially expressed from 233 metabolites and 3,539 proteins detected from the liver biopsies during the transition period. Complementary functional analysis of different metabolites and proteins indicated the upregulated gluconeogenesis, tricarboxylic acid cycles, AA degradation, fatty acid oxidation, AMP-activated protein kinase signaling pathway, peroxisome proliferator-activated receptor signaling pathway, and ribosome proteins in postpartum dairy cows. In terms of the metabolites and proteins, glucose-6-phosphate, fructose-6-phosphate, carnitine palmitoyltransferase 1A, and phosphoenolpyruvate carboxykinase played a significant role in these pathways. The upregulated oxidative status may be accompanied by the pathways mentioned above. In addition, the upregulated glucagon and insulin signaling pathways also indicated the significant requirement for glucose in postpartum dairy cows. These outcomes, from the view of global metabolites and proteins, may present a better comprehension of the biology of the transition period, which can be helpful in further developing nutritional regulation strategies targeting the liver to help cows overcome this metabolically challenging time. Simple Summary: Dietary neutral detergent fiber (NDF) is commonly used as an indicator of the fiber content in animal diets, particularly in ruminant nutrition. NDF plays a crucial role in the digestive process of ruminant animals, as it affects feed intake, rumen fermentation, and nutrient utilization. The levels of NDF affect feeding behavior, rumen fermentation, and production performance during the period of peak lactation in dairy cows. To prove it, four Holstein dairy cows were subjected to a feeding experiment with varying NDF levels in their diets. The results indicated that increased NDF levels resulted in reduced dry matter intake, while the time spent eating and ruminating increased. Moreover, higher NDF levels led to increased pH value and acetate concentration, as well as changes in the proportions of certain bacteria in the rumen. Milk yield, protein percentage, and nitrogen efficiency decreased with increasing NDF levels, while milk fat percentage and milk urea nitrogen concentration increased. Notably, diets with 25% and 34% NDF had negative effects on feeding behavior, rumen fermentation, and production performance. However, the diet with 28% NDF was effective in enhancing the production performance compared to the one with 31% NDF. These findings offer a valuable strategy for optimizing the provision of dietary NDF to cattle, thereby enhancing their overall performance. This study investigated the impact of dietary neutral detergent fiber (NDF) levels (25.49%, 28.65%, 31.66%, and 34.65%, respectively) on the feeding behavior, rumen fermentation, cellulolytic bacteria, and production performance of dairy cows during peak lactation. A feeding experiment was conducted using four fistulated Holstein dairy cows (600 ± 25 kg) with days in milk (50 ± 15 days), employing a 4 × 4 Latin square design to assign the cows to four groups. The results demonstrated that increasing NDF levels in the diet had the following effects: (1) A linear decrease in dry matter intake (DMI), NDF intake, and physically effective NDF8.0 (peNDF8.0) intake; a linear increase in the average time spent eating and ruminating, as well as the time spent eating and ruminating per kilogram of dry matter (DM); a quadratic response in the time spent ruminating per kilogram of NDF and peNDF8.0. (2) A linear increase in average pH value, acetate concentration, and the proportions of Fibrobacter succinogenes and Ruminococcus flavefaciens among total bacteria; a linear decrease in ammonia nitrogen (NH3-N) concentration, microbial crude protein (MCP), total volatile fatty acid (TVFA), propionate, butyrate, and lactate. (3) A linear decrease in milk yield, milk protein percentage, and nitrogen efficiency of dairy cows; a linear increase in milk fat percentage and milk urea nitrogen (MUN) concentration. Based on the combined results, it was found that diets with 25% and 34% NDF had detrimental effects on the feeding behavior, rumen fermentation, and production performance of dairy cows. However, the diet with 28% NDF showed superior outcomes in production performance compared to the one with 31% NDF. Simple Summary: No previous studies have focused on pain perception and management in intensively raised dairy cattle in China. The results of this survey study show that severely painful conditions such as parturition and fracture have the highest score in pain perception. Pain perception is highly associated with pain management willingness. To promote animal welfare and reduce unnecessary production losses, training in pain perception and management should be emphasised. Pain in dairy cattle is gaining attention globally. This study investigated the current attitudes of Chinese dairy practitioners to pain and its management in intensively raised dairy cattle. A total of 465 valid questionnaires with 26 painful conditions scored on numerical rating scales were collected from dairy practitioners. Data were analysed by descriptive statistics, analysis of variance, principal component analysis, and multivariate regression models. Dystocia was perceived as the most painful, while mild mastitis with milk changes only was perceived as the least painful. Respondents who agreed with the statement "pain management is worthwhile" tended to give a higher pain score. Young respondents (≤23 years old) and those from farms with ≤1000 cattle had lower pain scores for conditions with severe pain and low variability but higher pain scores for conditions with less severe pain and high variability, whereas highly educated respondents had consistently lower pain scores. As for pain management, older respondents (≥24 years old) tended to choose non-steroidal anti-inflammatory drugs, and farms with >1000 cattle were more likely to use analgesics. Simple Summary: The transition period (3 wks before to 3 wks after calving) is an important period in the lactation cycle of dairy cows. During this period, dairy cows undergo a series of physiological and metabolic changes due to the demands of pregnancy, parturition and postpartum lactation, which can easily lead to a negative energy balance (NEB). Therefore, the energy balance and nutritional regulation mechanism of dairy cows during the transition period are the focus of dairy cow nutrition and physiology research. However, crossbred cows have received less attention to date. Our study showed that feeding a prenatal concentrate mix at a rate of 0.6% of the body weight of Montbéliarde–Holstein crossbred cows has no negative effect on the performance and rumen fermentation of postpartum dairy cows and can satisfy the energy needs of dairy cows in the prepartum period. Furthermore, our data provide a theoretical basis for further revisions to the feeding standard and to alleviate the NEB of Montbéliarde–Holstein crossbred cows in China. This study was conducted to investigate the effect of three rates of prepartum dietary concentrate feeding on the milk performance, energy balance, and rumen fermentation of Montbéliarde–Holstein crossbred cows. Eighteen transition Montbéliarde–Holstein crossbred cows with similar days of gestation (258 ± 12 day) and body weights (622 ± 44 kg) were selected and randomly divided into three groups. In the prepartum period, the addition of concentrates accounted for 0.3% (low concentrate, LC), 0.6% (medium concentrate, MC), and 0.9% (high concentrate, HC) of the cow's body weight. The forage was corn stover, which was fed to the cows ad libitum with free access to water. Postpartum, all of the cows were fed a common lactation total mixed ration. The experimental period lasted from 21 days prepartum to 28 days postpartum. The energy balance (EB), net energy intake (NEI), and dry matter intake (DMI) of the HC group were greater than those of the other groups (p < 0.05). Likewise, the non-esterified fatty acids (NEFA), β-hydroxybutyric acid (BHBA), and total bilirubin (TBIL) in the blood of the LC group had significantly higher concentrations than they did in the other groups (p < 0.05). Moreover, the increase in the level of dietary concentrate had no significant effect on the rumen fermentation parameters (p > 0.05), and the total intestinal digestibility of dry matter (DM), crude protein (CP), and ether extract (EE) in the HC group was significantly higher (p < 0.05) than it was in the other groups during the prepartum period. In conclusion, the administration of the MC diet in the prepartum period had no negative effect on the performance and rumen fermentation of postpartum dairy cows and can satisfy the energy needs of prepartum dairy cows. This study investigated the effect of moderate risk level (8 µg/kg) AFB1 in diet supplemented with or without adsorbents on lactation performance, serum parameters, milk AFM1 content of healthy lactating cows and the AFM1 residue exposure risk in different human age groups. Forty late healthy lactating Holstein cows (270 ± 22 d in milk; daily milk yield 21 ± 3.1 kg/d) were randomly assigned to four treatments: control diet without AFB1 and adsorbents (CON), CON with 8 μg/kg AFB1 (dry matter basis, AF), AF + 15 g/d adsorbent 1 (AD1), AF + 15 g/d adsorbent 2 (AD2). The experiment lasted for 19 days, including an AFB1-challenge phase (day 1 to 14) and an AFB1-withdraw phase (day 15 to 19). Results showed that both AFB1 and adsorbents treatments had no significant effects on the DMI, milk yield, 3.5% FCM yield, milk components and serum parameters. Compared with the AF, AD1 and AD2 had significantly lower milk AFM1 concentrations (93 ng/L vs. 46 ng/L vs. 51 ng/L) and transfer rates of dietary AFB1 into milk AFM1 (1.16% vs. 0.57% vs. 0.63%) (p < 0.05). Children aged 2–4 years old had the highest exposure risk to AFM1 in milk in AF, with an EDI of 1.02 ng/kg bw/day and a HI of 5.11 (HI > 1 indicates a potential risk for liver cancer). Both AD1 and AD2 had obviously reductions in EDI and HI for all population groups, whereas, the EDI (≥0.25 ng/kg bw/day) and HI (≥1.23) of children aged 2–11 years old were still higher than the suggested tolerable daily intake (TDI) of 0.20 ng/kg bw/day and 1.00 (HI). In conclusion, moderate risk level AFB1 in the diet of healthy lactating cows could cause a public health hazard and adding adsorbents in the dairy diet is an effective measure to remit AFM1 residue in milk and its exposure risk for humans. Simple Summary: Nowadays, the lifetime milk production of dairy cows, as well as the fat and protein contents of milk, has reached an unprecedented high. These improvements pose threats and challenges to animal health and welfare due to metabolic stress. The cows, during the high-yielding period, are especially susceptible to metabolic diseases such as digestive alterations, rumen acidosis, and lameness. This study assessed the effects of Saccharomyces cerevisiae culture (SC), a food supplement, on ruminal pH, volatile fatty acid (VFA), inflammatory cytokines, and the performance of high-yield dairy cows. The results show that supplementing high-yield lactating cows with the SC of 100 g/d increases milk yield, milk fat content, and milk lactose content, but does not affect protein content. SC supplementation affects overall ruminal VFA concentration and induces a significantly greater ruminal pH. It has the potential to enhance the rumen microbial growth and decrease the inflammation response. Our research suggests that SC supplementation has a positive effect on the productivity and health of dairy cows. High-yield dairy cows with high-concentrate diets are more prone to experiencing health problems associated with rumen microbial imbalance. This study assessed the effects of Saccharomyces cerevisiae culture (SC), a food supplement, on ruminal pH, volatile fatty acid (VFA), inflammatory cytokines, and performance of high-yield dairy cows. Forty Holstein cows with similar characteristics (e.g., milk yield, days of milk, and parity) were randomly divided into two groups: an experimental group fed the basal ration supplemented with the SC of 100 g of SC per cow per day (hour, SC group), and a control group fed the same basal ration diet without SC (i.e., CON group). On average, the supplementation of SC started at 73 days of lactation. The experimental period lasted approximately 70 days (from 18 January to 27 March 2020), including 10 days for dietary adaptation. Milk yield was recorded daily. Rumen fluid and milk samples were collected after 2 h of feeding in the morning of day 0, 15, 30, and 60. The data showed that rumen pH increased (p < 0.05) when cows were provided with SC. On average, the cows in the SC group produced 1.36 kg (p < 0.05) more milk per day than those in the CON group. Milk fat content of cows in the SC group was also higher (4.11% vs. 3.96%) (p < 0.05). Compared with the CON group, the concentration of acetic acid in the rumen fluid of dairy cows in the SC group was significantly higher (p < 0.05). There were no differences (p > 0.05) found in milk protein content and propionic acid between groups. The SC group had a tendency increase in butyric acid (p = 0.062) and total VFA (p = 0.058). The result showed that SC supplementation also enhanced the ratio between acetic and propionic. Most of the mean inflammatory cytokine (IL-2, IL-6, γ-IFN, and TNF-α) concentrations (p < 0.05) of the SC group were lower than CON group. This study demonstrated that high-yield cows receiving supplemental SC could produce more milk with higher fat content, have higher rumen acetate, and potentially less inflammatory cytokines. Simple Summary: The main challenge faced by cows during the first three weeks after calving is the sudden increase in nutrient demand for milk production while dry matter intake and nutrient supply lag. Although metabolic hormone changes are a normal adaptive process in high yielding cows, failure to adapt to this challenge can lead to disease states and affect production and reproductive efficiency. Therefore, a smooth transition from prepartum to postpartum period for optimal dairy cow health and performance. Whether rumen-protected niacin supplementation could be a useful additive was controversial. This work tested whether the widespread use of rumen-protected niacin in multiparous Holstein dairy cows could be justified. Extensive studies about rumen-protected niacin (RPN) supplementation on dairy cows in early-lactation have been done, but the effects of RPN on changes in dry matter intake (DMI), milk production, feed digestibility, and fecal bacterial community were conflicting. The aim of this study was to investigate them affected by RPN in postpartum cows. Multiparous Holstein dairy cows (n = 12, parity = 3.5 ± 0.5, body weights = 740 ± 28 kg) were divided into two groups supplemented with either 0 (CON) or 20 g/d RPN (RPN). Our results showed that RPN supplementation increased DMI and milk production of cows during the first three weeks after calving (p < 0.05). The concentrations of neuropeptide Y and orexin A were significantly higher in RPN group than that in the CON group during postpartum period (p < 0.05). The apparent total-tract digestibility of nutrients was similar between the CON and RPN groups at 2 weeks after calving (p > 0.05). The 16S rRNA gene sequencing analysis showed that RPN had no impact on the alpha and beta diversity, although 4 genera were changed in cow feces at 14 days after calving. Simple Summary: Cassava (Manihot esculenta) residue is a by-product of cassava processing. Although it contains residual nutrients, it is highly perishable. Decayed cassava residue pollutes the environment and leads to major losses in feed. If cassava residue could be utilized as a dairy cow feedstuff, these problems could be solved. Our study showed that cassava residue is a good alternative to concentrate in the feed of Holstein cows. Furthermore, our data demonstrate the efficacy of the application of cassava residue as a feed for dairy cows and could help solve the shortage of feed resources in China. The feeding value of replacing concentrate with cassava (Manihot esculenta) residue in the feed of Holstein cows was confirmed using an in vitro gas test. The treatments consisted of 0% (control, CON), 5%, 10%, 15%, 20%, 25%, and 30% inclusion of cassava residue in fermentation culture medium composed of buffer solution (50 mL) and filtrated rumen fluid (25 mL). The parameters analyzed included the kinetics of gas production and fermentation indexes. Forty-eight hours later, there were no significant differences on in vitro dry matter disappearance (IVDMD), pH, and microbial crude protein (MCP) content among treatments (p > 0.05). However, the "cumulative gas production at 48 h" (GP48), the "asymptotic gas production" (A), and the "maximum gas production rate" (RmaxG) all increased linearly or quadratically (p < 0.01). The GP48 was significantly higher in the 25% treatment compared to the other treatments, except for the 30% (p < 0.01). The A was significantly larger in the 25% treatment compared to the other treatments, except for the 20% and 30% (p < 0.01). The RmaxG was distinctly larger in the 25% treatment compared to other treatments (p < 0.01); moreover, the "time at which RmaxG is reached" (TRmaxG) and the "time at which the maximum rate of substrate degradation is reached" (TRmaxS) were significantly higher in the 25% treatment than the CON, 20%, and 30% treatments (p < 0.01). Additionally, the content of ammonia-N (NH3-N) in all treatments showed linearly and quadratically decreases (p < 0.01), whereas total volatile fatty acid (VFA), iso-butyrate, butyrate, and iso-valerate contents changed quadratically (p = 0.02, p = 0.05, p = 0.01, and p = 0.02, respectively); all of these values peaked in the 25% treatment. In summary, the 25% treatment was associated with more in vitro gas and VFA production, indicating that this cassava residue inclusion level may be used to replace concentrate in the feed of Holstein cows. Simple Summary: Paper mulberry (Broussonetia papyrifera; PM) is a type of roughage rich in bioactive substances, such as phenolics and flavonoids, which are beneficial for animal health. This study evaluated the apparent digestibility of PM silage in Holstein dairy cows and its effect on the milk production, antioxidant capacity, and fecal bacteria composition of the animals. The results showed that the PM silage had no significant influence on the milk yield, apparent digestibility, and fecal bacteria composition of dairy cows. However, diets with PM silage can enhance the antioxidant and immune capacity of dairy cows, mainly due to the bioactive substance in PM. Today, faced with a shortage of feedstuff resources in ruminants, PM can be a useful feed resource for ruminants. Paper mulberry (Broussonetia papyrifera; PM) is an excellent and extensive type of roughage in Asia. This study aimed to evaluate the effects of PM silage on the milk production, apparent digestibility, antioxidant capacity, and fecal bacteria composition in Holstein dairy cows. Forty-five lactating Holstein dairy cows with a similar milk yield and parity were selected and randomly assigned to three groups. The control group was fed a non-PM silage diet, and the PM-treated groups were fed 4.5 and 9.0% PM silage supplementary diets for 28 days. Then, treatment groups were fed diets containing 13.5 and 18.0% PM silage for the next 28 days, respectively. PM silage increased the milk urea nitrogen and decreased the somatic cell count (p < 0.05), but did not affect the dry matter intake, milk yield, apparent digestibility, and energy balance of dairy cows. PM silage can enhance the blood total antioxidant capacity, superoxide dismutase, and immune globulin content (p < 0.05). The PM silage significantly decreased the relative abundance of the genera Ruminococcaceae UCG-013 and Tyzzerella-4 (p < 0.05). In conclusion, PM silage enhanced the antioxidant capacity and immunity of dairy cows, but did not influence the milk yield, dry matter digestibility, and fecal bacteria composition. Simple Summary: High mortality and involuntary culling rates cause great economic losses to the dairy industry around the world, and the survival of dairy calves and replacement heifers is paramount in modern dairy breeding. However, little has been done to genetically improve mortality rates of dairy calves and replacement heifers in Chinese Holstein cattle. In this study, we investigated population parameters (descriptive statistics) of mortality rates of dairy calves and replacement heifers and risk factors affecting mortality and involuntary culling rates in Chinese Holstein cattle. The mortality rate of dairy calves and replacement heifers from day 3 to 60, 61 to 365, and 366 to first calving was 5.5%, 7.4%, and 8.7%, and an unfavorable increasing trend has been observed in the Chinese Holstein population. Health events associated with digestive and respiratory or circulatory systems were the main reasons for deaths. Herd-birth year, birth season, and dam parity had significant effects on survival. Our findings will help farmers to better manage dairy calves and replacement heifers and highlight the need to include these survival traits as part of the national genetic evaluation schemes. Three survival traits were defined in dataset 2, which consisted of females born from 1999 to 2018. A binomial logistic regression was used to analyze the risk factors on the survival traits. The mortality rate of dairy calves and replacement heifers from day 3 to 60, 61 to 365, and 366 to first calving was 5.5%, 7.4%, and 8.7%, and an unfavorable increasing trend was observed. Health events associated with digestive and respiratory or circulatory systems were the main death reasons. Herd-birth year, birth season, and dam parity had significant effects on survival traits.
