Unter Spikelets versteht man kleine Depolarisationen mit einer Spike-ähnlichen Wellenform, die man in intrazellulären Ableitungen von verschiedenen Neuronentypen messen kann. In kortikalen Pyramidenzellen wurde ausgeprägte Spikelet-Aktivität nachgewiesen, die erheblich das Membranpotential beeinflussen kann (Crochet et al., 2004; Epsztein et al., 2010; Chorev and Brecht, 2012). Nichtsdestotrotz bleibt der Ursprung von Spikelets in diesen Neuronen unbekannt. In der vorgelegten Arbeit nutzte ich theoretische Modellierung um die Mechanismen von Spikelet-Erzeugung in Pyramidenzellen zu untersuchen. Zuerst sah ich die verschiedenen Hypothesen über den Ursprung von Spikelets durch. In der Literatur entdeckte ich zwei verschiedene Typen von Spikelets. Diese Arbeit konzentriert sich auf den häufiger vorkommenden Typ von Spikelets, welcher durch relativ große Amplituden gekennzeichnet ist. Die Eigenschaften dieser Spikelets passen am besten zu einem axonal Erzeugungsmechanismus. Im zweiten Kapitel widmete ich mich der Hypothese, dass somatische Spikelets axonalen Ursprungs mit somato-dendritischen Inputs hervorgerufen werden können. Ich identifizierte Bedingungen, die es erlauben ein Aktionspotential (AP) am Initialsegment vom Axon (AIS) zu initiieren, welches sich entlang des Axons ausbreitet, aber kein AP im Soma auslöst. Schließlich simulierte ich extrazelluläre Wellenformen von APs und Spikelets und verglich sie mit experimentellen Daten (Chorev and Brecht, 2012). Dieser Vergleich zeigte auf, dass die extrazellulären Wellenformen von Spikelets, die innerhalb einer Zellen am AIS erzeugt werden, gut zu den Daten passen. Zusammenfassend unterstützen meine Ergebnisse die Hypothese, dass Spikelets in Pyramidenzellen am AIS entstehen. Dieser Mechanismus könnte ein Mittel zum Energiesparen bei der Erzeugung von Output-APs sein. Außerdem könnte dadurch die dendritische Plastizität, die auf der Rückwärtspropagierung von APs beruht, reguliert werden., Spikelets are transient spike-like depolarizations of small amplitudes that can be measured in somatic intracellular recordings of many neuron types. Pronounced spikelet activity has been demonstrated in cortical pyramidal neurons in vivo (Crochet et al., 2004; Epsztein et al., 2010; Chorev and Brecht, 2012), influencing membrane voltage dynamics including action potential initiation. Nevertheless, the origin of spikelets in these neurons remains elusive. In thi thesis, I used computational modeling to examine the mechanisms of spikelet generation in pyramidal neurons. First, I reviewed the hypotheses previously suggested to explain spikelet origin. I discovered two qualitatively different spikelet types described in the experimental literature. This thesis focuses on the more commonly reported spikelet type, characterized by relatively large amplitudes of up to 20 mV. I found that the properties of these spikelets fit best to an axonal generation mechanism. Second, I explored the hypothesis that somatic spikelets of axonal origin can be evoked with somato-dendritic inputs. I identified the conditions allowing these orthodromic inputs to trigger an action potential at the axon initial segment, which propagates along the axon to the postsynaptic targets, but fails to elicit an action potential in the soma and the dendrites. Third, I simulated extracellular waveforms of action potentials and spikelets and compared them to experimental data (Chorev and Brecht, 2012). This comparison demonstrated that the extracellular waveforms of single-cell spikelets of axonal origin are consistent with the data. Together, my results suggest that spikelets in pyramidal neurons might originate at the axon initial segment within a single cell. Such a mechanism might be a way of reducing the energetic costs associated with the generation of output action potentials. Moreover, it might allow to control the dendritic plasticity by backpropagating action potentials.

Der Hippocampus ist eine von zwei Gehirnregionen, in der zeitlebens kontinuierlich neue Nervenzellen gebildet werden. Er spielt eine wichtige Rolle bei der Gedächtniskonsolidierung und wird mit der funktionellen Entstehung neurodegenerativer Erkrankungen in Verbindung gebracht. Strukturveränderungen im erwachsenen Gehirn, die mit einer Depression einhergehen, sind laut Literatur auf einen geringen Serotoninspiegel und reduzierte hippocampale Neurogenese zurückzuführen. Selektive Serotonin-Wiederaufnahmehemmer (SSRI) erhöhen die Serotoninkonzentration im synaptischen Spalt und üben einen positiven Effekt auf die adulte Neurogenese aus. In der vorliegenden Arbeit wird untersucht, wie Veränderungen in der Serotonin (5-HT)-Neurotransmission durch einmalige oder chronische Gaben von Fluoxetin und speziellen Agonisten und Antagonisten für die Serotoninrezeptoren 5-HT1a und 5-HT2 in der erwachsenen Maus die Proliferation und Differenzierung von neugebildeten Nervenzellen im Gyrus dentatus beeinflussen. Die Ergebnisse zeigen, dass ein konträres Agieren beider Rezeptoren zu einem ausgewogenen Serotoninspiegel führt. 5-HT1a- und 5-HT2c-Rezeptoren haben einen Einfluss auf das Überleben neugebildeter Nervenzellen, wobei sie unterschiedliche Entwicklungsstadien innerhalb der adulten Neurogenese kontrollieren. Die vorliegende Arbeit bekräftigt außerdem, dass die chronische Gabe von Fluoxetin die adulte Neurogenese steigert. The hippocampus as one region with ongoing neurogenesis throughout life contributes to the formation of long-term memory and has also been implicated in the pathology of major depression. Studies suggest that depression might be due to decreased levels of serotonin and reduced neurogenesis in the adult brain and that the beneficial effects of selective serotonin reuptake inhibitors would require adult hippocampal neurogenesis. Here, I investigated how modulation of serotonergic neurotransmission by acute and chronic treatment with the antidepressant fluoxetine, and selective serotonin receptor agonists and antagonists in adult mice influences precursor cell activity during development. I focused on 5-HT1a and 5-HT2 receptors as major mediators of serotonin action. The present findings suggest that an opposed action of 5-HT1a and 5-HT2c receptor subtypes result in a balanced regulation of serotonin levels in the dentate gyrus. Both receptors differentially affect intermediate cell stages in adult hippocampal neurogenesis and play an important role in the survival of newly generated neurons. Furthermore, this study confirms that chronic fluoxetine treatment increases adult neurogenesis. In conclusion, the latency of onset of fluoxetine action can be explained by a balanced interplay of 5-HT1a and 5-HT2c receptor subtypes.

Apolipoprotein E (ApoE) ist eine verbreitet vorkommende Komponente der Plasmalipoproteine und spielt eine Schlüsselrolle bei Lipidtransport und Cholesterin-Homöostase über den Low Density Lipoprotein Rezeptor. Im ZNS wird ApoE hauptsächlich von Astrozyten synthetisiert und sekretiert. ApoE-Isoformen haben unterschiedliche Wirkung auf eine Zahl von pathologischen Prozessen, die der Alzheimerschen Krankheit zugrunde liegen. Um die Rolle des ApoE für die Alzheimersche Krankheit zu erhellen, ist es wichtig Kenntnisse über seine Regulation zu erlangen. Ein Ziel dieser Arbeit war zu untersuchen, ob die „Second-Messenger“-Signalpfade „Adenylatcyclase/Proteinkinase A (PKA)“ und/oder „Phospholipase C/Proteinkinase C (PLC/PKC)“ in die Regulation der astrozytären ApoE Sekretion eingreifen. Hierfür wurden primäre hippocampale Astrozytenkulturen von Ratten mit verschiedenen Analoga, Rezeptoragonisten und Neurotransmittern, die diese Signalpfade beeinflussen, inkubiert. Dibutyryl-cAMP (cAMP-Analogon) erhöhte die ApoE Sekretion. Auch Rezeptoragonisten des Adenylatcyclase/PKA-Signalwegs beeinflussten die ApoE Sekretion. Isoproterenol (beta-Adrenorezeptoragonist) erhöhte die ApoE Sekretion, während Clonidine (alpha-2 Adrenorezeptoragonist) sie senkte. Der PKC-Aktivator Phorbol 12-Myristat 13-Acetat senkte die ApoE Sekretion und kehrte die dibutyryl-cAMP-vermittelte Erhöhung der ApoE Sekretion um. Arterenol (alpha-1 Adrenorezeptoragonist) und Serotonin (Neurotransmitter) erhöhten die ApoE Sekretion, wohingegen Carbachol (Acetylcholiner muskarinischer Rezeptoragonist) die ApoE Sekretion senkte. Es wird gezeigt, dass die verwendeten Substanzen einen von der ApoE Sekretion verschiedenen Einfluss auf die Sekretion des Nervenwachstumsfaktors (NGF) haben. Dies legt die Vermutung nahe, dass die beobachteten Ergebnisse nicht auf einen generellen Effekt der Proteinsynthese zurückzuführen sind. Es kann gefolgert werden, dass die astrozytäre ApoE Sekretion von Faktoren beeinflusst werden kann, die die intrazelluläre Konzentration von cAMP verändern oder die PKC aktivieren. Das zweite Ziel der Arbeit war zu untersuchen ob Amyloid Fragmente einen Einfluss auf die astrozytäre ApoE Sekretion haben. Senile Amyloid-Plaques in Alzheimer-Gehirnen zeigen eine ApoE-Immunreaktivität, Astrozyten die diese Plaques umgeben dagegen nicht. Es wird gezeigt, dass gealtertes fibrilläres Amyloid (1-40) die ApoE Sekretion erhöht. Das sekretierte ApoE wird durch die, die Zellen umgebenden Amyloid-Konglomerate, gebunden. Die verwendeten Amyloid Fragmente beeinflussten nicht die Menge des sekretierten basischen Fibroblastenwachstumsfaktors. Dies legt nahe, dass die beobachteten Ergebnisse nicht auf einen generellen Effekt der Proteinsynthese zurückzuführen sind., Apolipoprotein E (apoE) is an abundant component of plasma lipoproteins that plays a key role in lipid transport and cholesterol homeostasis via the low density lipoprotein receptor. In the CNS, apoE is synthesised and secreted especially by astrocytes. ApoE isoforms have different effects on a number of pathological processes underlying Alzheimer’s disease. Therefore, understanding the regulated synthesis of apoE is important for determining its role in Alzheimer’s disease. One aim of this work was to examine whether the second-messenger-pathways „adenylyl cyclase/proteinkinase A (PKA)“ and/or „phospholipase C/proteinkinase C (PLC/PKC) are involved in the regulation of apoE secretion in astrocytes. Therefore rat primary hippocampal astrocyte cultures were incubated with various analogues, receptor agonists and neurotransmitters which influence these pathways. Dibutyryl-cAMP (cAMP analogue) increased the apoE secretion. ApoE secretion was also modulated by receptor agonists of the adenylyl cyclase/PKA pathway. Isoproterenol (beta-adrenoceptor agonist) enhanced, while Clonidine (alpha 2-adrenoceptor agonist) decreased, the secreted apoE. In contrast, the PKC activator phorbol 12-myrisate 13-acetate decreased the apoE secretion. It also reversed the effects of dibutyryl-cAMP. Arterenol (alpha 1-adrenoceptor) and serotonin (neurotransmitter) enhanced, whereas carbachol (acetylcholine muscarinic receptor agonist) deceased secreted apoE. It is shown, that the used substances have different effects on the secretion of the nerve growth factor (NGF) as compared to apoE secretion, suggesting that the results obtained were unlikely to be due to a general effect on protein synthesis. It can be concluded that actrocytic apoE production can be regulated by factors that affect cAMP intracellular concentration or activate PKC. The second aim of this work was to examine whether amyloid fragments have an effect on the apoE secretion of astrocytes. Senile amyloid plaques in Alzheimer’s disease brains show apoE immunoreactivity, astrocytes which surround them do not. It is shown, that aged, fibrillic amyloid (1-40) increases apoE secretion. The secreted apoE is bound to the surrounding amyloid conglomerates. The used amyloid fragments did not increase or decrease basic fibroblast growth factor secretion, suggesting that the results obtained were unlikely to be due to a general effect on protein synthesis.
