This study investigated the effect of Ethylenediamine dihydroiodide (EDDI) on growth performance, immune function and intestinal health of meat ducks challenged with Avian pathogenic Escherichia coli (APEC). A total of 360 one-day-old Cherry Valley ducks with similar body weight were randomly allocated to 6 treatments (6 floor cages, 10 birds/cage). A 3 × 2 factor design was used with 3 dietary iodine levels (0, 8, 16 mg/kg in the form EDDI and whether APEC was challenged or not at 7-day-old ducks. The feeding period lasted for 20 d. The results showed that the addition of EDDI reduced APEC-induced decrease of the 20-d weight loss of meat ducks (P < 0.05), and alleviated the inflammatory response of liver tissue induced by APEC challenge in meat ducks. In terms of immune function, EDDI supplementation reduced the immune organ index and increased the immune cell count of meat ducks, reduced the level of endotoxins in the serum of meat ducks (P < 0.05), as well as inhibited the expression levels of liver and spleen inflammatory factors and TLR signaling pathway related genes induced by APEC (P < 0.05). In terms of intestinal health, EDDI inhibited APEC-induced decreases in ZO-3 genes expression and increases in IL-1β and TNF-α expression, increased relative abundance of beneficial bacteria in the cecum and content of metabolites. Pearson correlation analysis showed that there was a significant correlation between liver inflammatory factors and TLR4 signaling pathway genes, and there might be a significant correlation between intestinal microbial flora and other physiological indexes of meat ducks, which indicated that EDDI could reduce the damage to immune function and intestinal health caused by APEC challenge through regulating the structure of intestinal flora. Collectively, our findings suggest that the EDDI can promote growth performance, improve immune function and the intestinal barrier in APEC-challenged meat ducks, which may be related to the suppression of NF-κB signal., (Copyright © 2024 The Authors. Licorice flavonoids, a bioactive substance derived from glycyrrhiza, have been reported for many pharmacological properties and are beneficial to animal health. This study aimed to explore the effects of licorice flavonoids powder (LFP) on growth performance and intestinal health of piglets. A total of 96 weaned piglets were randomly assigned into four treatments and supplemented with 0, 50, 150 and 250 mg/kg LFP for 5 weeks. Dietary LFP supplementation tended to increase (p = 0.068) average daily gain (ADG) and reduce (p = 0.089) the feed intake/body gain (F/G) of piglets than that of the control group during 15-35 days; and concentrations of LFP supplementation reduced (p < 0.01) diarrhoea index during 14-35 days and 0-35 days. Piglets fed on diets supplied with LFP had a lower (p < 0.05) pH in caecum and colon. Dietary LFP supplementation increased (p < 0.01) the villi height and the ratio of villi height/crypt depth in duodenum, and reduced (p < 0.05) crypt depth in duodenum. Compared with the control group, 250 mg/kg LFP supplementation up-regulated (p < 0.05) the mRNA level of occludin (OCLN) in ileum. Meanwhile, dietary LFP supplementation down-regulated (p < 0.05) mRNA abundance of Interleukin (IL)-1β, IL-8 and induced nitrogen monoxide synthase (INOS) in duodenum. Dietary 150 mg/kg LFP supplementation down-regulated (p < 0.05) mRNA abundance of IL-1β and 250 mg/kg LFP up-regulated (p < 0.05) the expression of IL-10 in ileum. In summary, dietary LFP supplementation has a trend to improve the performance of weaning piglets, those improvements are accompanied by reduction in diarrhoea, enhancement of intestinal morphological structure, barrier function, immune function, and development. This study was conducted to investigate the influence of zinc (Zn) supplementation on growth performance, intestinal development and intestinal barrier function in Pekin ducks. A total of 480, one-day-old male Pekin ducks were divided into 6 groups with 8 replicates: 0 mg/kg Zn, 0 mg/kg Zn +0.5 mg/kg lipopolysaccharide (LPS), 30 mg/kg Zn, 30 mg/kg Zn +0.5 mg/kg LPS, 120 mg/kg Zn, 120 mg/kg Zn +0.5 mg/kg LPS. The duck primary intestinal epithelial cells (DIECs) were divided into 6 groups: D-Zn (Zinc deficiency, treated with 2 µmol/L zinc Chelator TPEN), A-Zn (Adequate Zinc, basal medium), H-Zn (High level of Zn, supplemented with 20 µmol/L Zn), D-Zn + 20 µg/mL LPS, A-Zn + 20 µg/mL LPS, H-Zn + 20 µg/mL LPS. The results were as follows: in vivo, with Zn supplementation of 120 mg/kg reduced LPS-induced decrease of growth performance and intestine damage (P < 0.05), and increased intestinal digestive enzyme activity of Pekin ducks (P < 0.05). In addition, Zn supplementation also attenuated LPS-induced intestinal epithelium permeability (P < 0.05), inhibited LPS-induced the expression of proinflammatory cytokines and apoptosis-related genes (P < 0.05), as well as reduced LPS-induced the intestinal stem cells mobilization of Pekin ducks (P < 0.05). In vitro, 20 µmol/L Zn inhibited LPS-induced expression of inflammatory factors and apoptosis-related genes (P < 0.05), promoted the expression of cytoprotection-related genes, and attenuated LPS-induced intestinal epithelium permeability in DIECs (P < 0.05). Mechanistically, 20 µmol/L Zn enhanced tight junction protein markers including CLDN-1, OCLD, and ZO-1 both at protein and mRNA levels (P < 0.05), and also increased the level of phosphorylation of TOR protein (P < 0.05) and activated the TOR signaling pathway. In conclusion, Zn improves growth performance, digestive enzyme activity, and intestinal barrier function of Pekin ducks. Importantly, Zn also reverses LPS-induced intestinal barrier damage via enhancing the expression of tight junction proteins and activating the TOR signaling pathway., (Copyright © 2021 The Authors. The current study was conducted with a mathematical model to investigate the appropriate iron supplementation by analyzing growth performance, hematological parameters, nutrient utilization, organ development, and Fe-containing enzyme activity in Pekin ducks. A total of 1120 ducks (1-day-old; 50.62 ± 0.20 g) were randomly allotted to seven groups with eight replicates of 20 ducks per replicate. Ducks were fed either a low-iron diet or basal diets supplemented with 0, 30, 60, 90, 120, and 150 mg Fe/kg from iron sulfate, respectively. The results showed that iron deficiency decreased body weight (BW), average daily gain (ADG), average daily feed intake (ADFI), red blood cell count (RBC), hemoglobin (Hb), hematocrit (Hct), mean cell volume (MCV), the apparent utilization of energy and dry matter, the weight of duodenum, jejunum, and ileum but increased feed conversion ratio (F/G) (P < 0.05). Meanwhile, birds in low-iron groups showed an increase in oxidative stress evidenced by the decreased catalase (CAT) activities and production of malonaldehyde (MDA) (P < 0.05). On the contrary, diets supplemented with iron significantly improved the growth performance, in different ages, and achieved the best values in group 4 which supplemented 60 mg Fe/kg (P < 0.05). Also, iron supplementation increased the apparent utilization of energy and dry matter but decreased the utilization of iron in ducks at 35 days of age (P < 0.05). Furthermore, the hematological and intestine Fe-containing enzyme activities were improved with iron up to an optimal level (P < 0.05) at 14 and 35 days. In conclusion, iron deficiency impaired growth performance, physiological indexes, nutrient utilization, and antioxidant system, while dietary 71.25~82.80 mg/kg iron for starter ducks and 75.00~89.41 mg/kg iron for grower ducks were suggested to improve growth performance.

This study aimed to determine the effects of dietary spermine supplementation on the inflammatory response and immune function of the thymus and spleen in piglets. Eighty suckling piglets were randomly assigned to receive adequate nutrients supplemented with spermine (0.4 mmol/kg body weight) or restricted nutrient intake supplemented with normal saline for 7 h or 3, 6 and 9 days in pairs. Regardless of treatment time, spermine supplementation decreased (p < 0.05, compared with the controls) the following: (1) tumour necrosis factor α (TNF-α), interleukin (IL)-1β, 2 and 6, and interferon (IFN)-γ levels in serum; (2) gene expression of cluster of differentiation 8 and integrin beta-2 in the thymus and spleen and the lymphocyte function-associated antigen 1 in the thymus; (3) mRNA levels of TNF-α, IL 1β, 2, 6, and 12, IFN-γ and inducible nitric oxide synthase in the thymus and spleen, as well as IL-8 in the spleen; and (4) eukaryotic IF4E-binding protein 1, Janus kinase 2, signal transducer and activator of transcription 3, and nuclear factor-kappa B P65 gene transcriptions in the thymus and spleen. By contrast, spermine supplementation increased (p < 0.05) the following: (1) immunoglobulin M, IL-10, and transforming growth factor β1 gene expression, as well as (2) relative mRNA levels of mammalian target of rapamycin (mTOR) and ribosomal protein S6 kinase 1 in the thymus and spleen. These effects were also observed upon prolonged spermine administration (p < 0.05). In summary, dietary spermine supplementation can alleviate inflammatory response, enhance the immune function and regulate the gene expression of signalling molecules related to inflammation.

N-Carbamylglutamate (NCG), an effective precursor of arginine (ARG), can enhance ARG synthesis, increase intestinal growth, and improve reproductive performance. However, the antioxidant effect of NCG remains largely unknown. This study aims to survey the effects of ARG and NCG supplementation on the antioxidant statuses of the liver and plasma in rats under oxidative stress. Rats were fed for 30 days with one of the three iso-nitrogenous diets: basal diet (BD), BD plus 1% ARG, and BD plus 0.1% NCG. On day 28, half of the rats fed with BD were intraperitoneally injected with 12 mg per kg body weight of diquat (diquat group) and the other half was injected intraperitoneally with sterile 0.9% NaCl solution (control group). The other diet groups also received an intraperitoneal injection of 12 mg per kg body weight of diquat, as follows: diquat + 1% ARG (DT + ARG), and diquat + 0.1% NCG (DT + NCG). Rat liver and plasma samples obtained 48 h after diquat injection were analyzed. Results indicated that diquat significantly affected the plasma conventional biochemical components (relative to the controls), which were partially alleviated in both the DT + ARG and DT + NCG groups (P < 0.05). Diquat also significantly decreased the glutathione (GSH) content (by 30.0%), and decreased anti-superoxide anion (ASA; by 13.8%) and anti-hydroxyl radical (AHR; by 38.9%) abilities in the plasma, and also decreased catalase (CAT) activity both in the liver (by 17.5%) and plasma (by 33.4%) compared with the control group. By contrast, diquat increased the malondialdehyde (MDA) content (by 23.0%) in the plasma (P < 0.05) compared with the control group. Relative to those of the diquat group, higher CAT activity and GSH content were noted in the plasma of the DT + ARG group and in the liver of both DT + ARG and DT + NCG groups (P < 0.05). Furthermore, the DT + ARG group exhibited significantly enhanced plasma ASA activity (P < 0.05). The DT + NCG group showed significantly improved total antioxidant capacity (T-AOC) in the liver and plasma (P < 0.05). Increased GSH content and elevated ASA and AHR activities were also found, but the MDA content in the plasma was depleted (P < 0.05). Compared with the DT + ARG group, the DT + NCG group showed increased liver and plasma T-AOC, enhanced plasma AHR activity, increased liver ASA activity, and decreased plasma MDA content (P < 0.05). Overall, supplementation of 1% ARG and 0.1% NCG can partially protect the liver and plasma from oxidative stress. Furthermore, compared with 1% ARG, 0.1% NCG more effectively alleviated oxidative stress.

Chronic heat stress (CHS) compromised the immunity and spleen immunological function of pigs, which may associate with antioxidant suppression and splenocyte apoptosis and splenic inflammation. Selenium (Se) exhibited antioxidant function and immunomodulatory through selenoprotein. Thus, this study aimed to investigate the protective effect of dietary hydroxy-selenomethionine (Selisso®, SeO) on chronic heat stress (CHS)-induced porcine splenic oxidative stress, apoptosis and inflammation. Growing pigs were raised in the thermoneutral environment (22 ± 2 °C) with the basal diet (BD), or raised in hyperthermal conditions (33 ± 2 °C) with BD supplied with 0.0, 0.2, 0.4 and 0.6 mg Se/kg SeO for 28 d, respectively. The results showed that dietary SeO supplementation recovered the spleen mass and enhanced the splenic antioxidant capacity of CHS growing pigs. Meanwhile, SeO activated the Nrf2/Keap1 signal, downregulated p38, caspase 3 and Bax, inhibited the activation of NFκb and STAT3, and enhanced the protein expression level of GPX1, GPX3, GPX4, SELENOS and SELENOF. In summary, SeO supplementation mitigates the CHS-induced splenic oxidative damages, apoptosis and inflammation in pigs, and the processes are associated with the activation of Nrf2/Keap1 signal and the suppression of NFκb, p38(MAPK) and STAT signal. This study was carried out to evaluate the effects of supplemental zinc methionine (Zn–Met) on growth performance, immune function, and intestinal health of meat ducks challenged with avian pathogenic Escherichia coli (APEC). A total of 480 1-day-old Cherry Valley male ducks were randomly assigned to 8 treatments with 10 replicates, each replicate containing 10 ducks. A 4 × 2 factor design was used with four dietary zinc levels (0, 30, 60, 120 mg Zn/kg in the form Zn–Met was added to the corn–soybean basal diet) and challenged with or without APEC at 8-days-old ducks. The trial lasted for 14 days. The results showed that a dietary Zn–Met supplementation significantly increased body weight (BW) of 14 days and BW gain, and decreased mortality during 7–14-days-old ducks (p < 0.05). Furthermore, dietary 30, 60, 120 mg/kg Zn–Met supplementation noticeably increased the thymus index at 2 days post-infection (2 DPI) and 8 DPI (p < 0.05), and 120 mg/kg Zn–Met enhanced the serum IgA at 2 DPI and IgA, IgG, IgM, C3 at 8 DPI (p < 0.05). In addition, dietary 120 mg/kg Zn–Met supplementation dramatically increased villus height and villus height/crypt depth (V / C) of jejunum at 2 DPI and 8 DPI (p < 0.05). The TNF- α and IFN- γ mRNA expression were downregulated after supplemented with 120 mg/kg Zn–Met in jejunum at 8 DPI (p < 0.05). Moreover, dietary 120 mg/kg Zn–Met supplementation stimulated ZO-3 , OCLN mRNA expression at 2 DPI and ZO-2 mRNA expression in jejunum at 8 DPI (p < 0.05), and improved the MUC2 concentration in jejunum at 2 DPI and 8 DPI (p < 0.05). At the same time, the cecal Bifidobacterium and Lactobacillus counts were increased (p < 0.05), and Escherichia coli counts were decreased (p < 0.05) after supplemented with Zn–Met. In conclusion, inclusion of 120 mg/kg Zn–Met minimizes the adverse effects of APEC challenge on meat ducks by improving growth performance and enhancing immune function and intestinal health. Tryptophan can alleviate stress and improve intestinal health, but the precise mechanism has not been fully elucidated. This study aimed to examine the effects of tryptophan supplementation on antioxidant status, inflammation, endoplasmic reticulum (ER) stress, apoptosis, and pyroptosis signaling pathway in the intestine of piglets after Escherichia coli lipopolysaccharide (LPS) challenge. Thirty-two weaning piglets were allotted to four treatments including: non-challenged control, LPS-challenged control, LPS + 0.2% tryptophan and LPS + 0.4% tryptophan. On day 35 of feeding, piglets were injected intraperitoneally with 100 μg/kg of body weight LPS or saline. Among the LPS-challenged pigs, tryptophan supplementation improved intestinal morphology as indicated by greater villus height, villus area and smaller crypt depth, and antioxidant status, and decreased the mRNA expression and concentration of proinflammatory cytokines. Moreover, tryptophan downregulated the expression of ER stress (ER oxidoreductase-1α, ER oxidoreductase-1β, glucose-regulated protein-78, activating transcription factor 6, C/EBP homologous protein), apoptosis (B-cell lymphoma-2, BCL2-associated X protein, caspase 3), and pyroptosis signaling pathway (nucleotide-binding oligomerization domain-like receptor protein 3, caspase 1, gasdermin-D, apoptosis-associated speck-like protein containing a CARD). Collectively, tryptophan supplementation can contribute to gut health by improving antioxidant status and alleviating inflammation, ER stress, apoptosis, and pyroptosis in the intestine of piglets after lipopolysaccharide challenge.
