We evaluated the differences between the supplementation of urea in rumen and/or abomasum on forage digestion, N metabolism and urea kinetics in cattle fed a low-quality tropical forage. Five Nellore heifers were fitted with rumen and abomasum fistulas and assigned to a Latin square design. The treatments were control, continuous infusion of urea in the abomasum (AC), continuous infusion of urea in the rumen, a pulse dose of urea in the rumen every 12 h (PR) and a combination of PR and AC. The control exhibited the lowest (P < 0·10) faecal and urinary N losses, which were, overall, increased by supplementation. The highest urinary N losses (P < 0·10) were observed when urea was either totally or partially supplied as a ruminal pulse dose. The rumen N balance was negative for the control and when urea was totally supplied in the abomasum. The greatest microbial N production (P < 0·10) was obtained when urea was partially or totally supplied in the abomasum. Urea supplementation increased (P < 0·10) the amount of urea recycled to the gastrointestinal tract and the amount of urea-N returned to the ornithine cycle. The greatest (P < 0·10) amounts of urea-N used for anabolism were observed when urea was totally and continuously infused in the abomasum. The continuous abomasal infusion also resulted in the highest (P < 0·10) assimilation of microbial N from recycling. The continuous releasing of urea throughout day either in the rumen or abomasum is able to improve N accretion in the animal body, despite mechanism responsible for that being different.

The literature lacks studies investigating the performance of supplemented replacement heifers grazing on intensively managed warm-season pasture. Our objective was to evaluate the effects of supplement composition (energetic or protein) on the performance, muscle development, thermogenisis, nutrient intake, and digestibility of replacement Holstein heifers grazing Mombaça grass. Eighteen Holstein heifers with an average age and initial body weight (BW) of 12.57 ± 2.54 mo and 218.76 ±47.6 kg, respectively, were submitted to a randomized block design, with six replicates on a rotational grazing system of Panicum maximum cv. Mombaça pasture. Treatments were: control (CON; mineral salt ad libitum); energy supplement (ENE; corn meal as supplement, 8% CP and 3.78 Mcal/kg DE); and protein supplement (PRO; corn and soybean meal, 25% CP and 3.66 Mcal/kg DE). Supplements were individually fed at 0.5% BW. The experiment lasted 120 days, subdivided into three periods. Titanium dioxide and indigestible neutral detergent fiber (iNDF) were used to estimate the intakes and digestibility of the nutrients. BW, wither height, thoracic circumference, body length, and ultrasound of ribeye fat thickness measurements were taken once per period. Body condition score (BCS) was assessed twice during the experiment. The MIXED procedure of SAS, including period as a repeated measure, was used and significance was declared at P ≤ 0.05. Dry matter intake (DMI), CP intake (CPI) and DE intake were greater in heifers fed PRO compared to CON and ENE. Heifers supplemented with ENE had the lowest DMI. Treatment affected pasture intake/BW; it was similar between PRO and CON heifers, and lower for the ENE treatment. A treatment × period interaction was observed for NDF intake (%BW), in which heifers fed PRO and CON had the greatest NDF intake and ENE had the lowest. The digestibility of DM was the greatest in PRO-supplemented heifers and the lowest in CON heifers. Heifers fed ENE had decreased CP digestibility compared to PRO and CON heifers. Average daily gain (ADG) and thoracic circumference gain were greatest in the PRO treatment. BCS was greater in PRO compared to CON and ENE heifers. Supplementing Holstein heifers at 0.5% BW using PRO supplementation resulted in better animal performance, primarily greater ADG, than feeding ENE or not supplementing (CON). The present study was conducted to evaluate the effects of high and low supplementation levels pre- and post-weaning on performance, endocrine, metabolic, and reproductive responses of Nellore heifers. Fifty Nellore heifers with 132 ± 9.9 kg average body weight (BW) and 138 ± 19 days of age were supplemented from 4 to 14 months. The heifers were distributed into five supplementation plans: HH-6 g/kg of BW of supplement pre- and post-weaning, HL-6 g/kg of BW of supplement pre-weaning and 3 g/kg post-weaning, LH-3 g/kg of BW pre-weaning and 6 g/kg of BW post-weaning, LL-3 g/kg of BW pre- and post-weaning, and CC-control, no supplementation. Interactions were not significant (P > 0.10). The level of supplement fed pre-weaning did not affect any of the performance variables evaluated at the end of the experiment (P > 0.10). There was a significant effect of supplementation and level of supplementation fed post-weaning on average daily gain (ADG) and final BW (P < 0.05). Overall ADG was also affected only by supplementation and level of supplement fed post-weaning (P < 0.05) with animals receiving 6 g/kg of BW post-weaning gaining more weight. Follicular diameter was greater in animals that received 6 g/kg of BW post-weaning (P < 0.05). In summary, performance, endocrine, metabolic, and reproductive variables evaluated in the current study were improved by the level of supplement fed post-weaning. Heifers receiving supplementation of 6 g/kg of BW post-weaning had greater responses, independent of the level received during the pre-weaning phase.

Creep feeding has been used to reduce calves' nutritional dependence on the cow, but research results under tropical conditions have not been conclusive about the effects on the cow. Therefore, this study was conducted to evaluate the effects of high and low supplementation levels for Nellore heifer calves on performance, milk production, and metabolic profile of their mothers. Fifty multiparous Nellore cows and their respective calves were used. The following treatments were evaluated: 0-control, no supplement was fed to calves; 3-calves received supplement in the amount of 3 g/kg of body weight (BW); 6-calves received supplement in the amount of 6 g/kg of BW. There was no significant effect of level of supplementation offered to offspring on cow BW, body condition score (BCS) and subcutaneous fat thickness (P > 0.05). Level of supplementation of heifer calves did not significantly affect milk production corrected to 4% of fat (P > 0.05). Fat, protein, lactose, and total solids of the milk also did not differ among supplementation strategies (P > 0.05). Level of supplement fed to calves had no effect on cows' glucose, total cholesterol, HDL, LDL, triglycerides, total protein, and albumin levels (P > 0.05), but cows nursing calves that did not receive supplement had lower level of serum urea N (SUN; P < 0.05). We conclude that creep feeding in the amounts of 3 or 6 g/kg of BW daily has no major impact on dams' performance and metabolism.

This study evaluated the efficiency of Lys utilization by growing steers. Five ruminally cannulated Holstein steers (165 ± 8 kg) housed in metabolism crates were used in a 6 × 6 Latin square design; data from a sixth steer was excluded due to erratic feed intake. All steers were limit fed (2.46 kg DM/d), twice daily, diets low in RUP (81% soybean hulls, 8% wheat straw, 6% cane molasses, and 5% vitamins and minerals). Treatments were 0, 3, 6, 9, 12, and 15 g/d of Lys continuously abomasally infused. To prevent AA other than Lys from limiting performance, a mixture providing all essential AA to excess was continuously abomasally infused. Additional continuous infusions included 10 g urea/d, 200 g acetic acid/d, 200 g propionic acid/d, and 50 g butyric acid/d to the rumen and 300 g glucose/d to the abomasum. These infusions provided adequate ruminal ammonia and increased energy supply without increasing microbial protein supply. Each 6-d period included 2 d for adaptation and 4 d for total fecal and urinary collections for measuring N balance. Blood was collected on d 6 (10 h after feeding). Diet OM digestibility was not altered ( ≥ 0.66) by treatment and averaged 73.7%. Urinary N excretion was decreased from 32.3 to 24.3 g/d by increasing Lys supplementation to 9 g/d, with no further reduction when more than 9 g/d of Lys was supplied (linear and quadratic, < 0.01). Changes in total urinary N excretion predominantly were due to changes in urinary urea N. Increasing Lys supply from 0 to 9 g/d increased N retention from 21.4 to 30.7 g/d, with no further increase beyond 9 g/d of Lys (linear and quadratic, < 0.01). Break-point analysis estimated maximal N retention at 9 g/d supplemental Lys. Over the linear response surface of 0 to 9 g/d Lys, the efficiency of Lys utilization for protein deposition was 40%. Plasma urea N tended to be linearly decreased ( = 0.06) by Lys supplementation in agreement with the reduction in urinary urea N excretion. Plasma concentrations of Lys linearly increased ( < 0.001), but Leu, Ser, Val, and Tyr ( ≤ 0.02) were linearly reduced by Lys supplementation, likely reflecting increased uptake for protein deposition. In our model, Lys supplementation promoted significant increases in N retention and was maximized at 9 g/d supplemental Lys with an efficiency of utilization of 40%.

In tropical regions, protein supplementation is a common practice in dairy and beef farming. However, the effect of highly degradable protein in ruminal fermentation and microbial community composition has not yet been investigated in a systematic manner. In this work, we aimed to investigate the impact of casein supplementation on volatile fatty acids (VFA) production, specific activity of deamination (SAD), ammonia concentration and bacterial and archaeal community composition. The experimental design was a 4×4 Latin square balanced for residual effects, with four animals (average initial weight of 280±10 kg) and four experimental periods, each with duration of 29 days. The diet comprised Tifton 85 (Cynodon sp.) hay with an average CP content of 9.8%, on a dry matter basis. Animals received basal forage (control) or infusions of pure casein (230 g) administered direct into the rumen, abomasum or divided (50 : 50 ratio) in the rumen/abomasum. There was no differences (P>0.05) in ruminal pH and microbial protein concentration between supplemented v. non-supplemented animals. However, in steers receiving ruminal infusion of casein the SAD and ruminal ammonia concentration increased 33% and 76%, respectively, compared with the control. The total concentration of VFA increased (P0.05) in species richness and diversity of γ-proteobacteria, firmicutes and archaea between non-supplemented Nellore steers and steers receiving casein supplementation in the rumen. However, species richness and the Shannon-Wiener index were lower (P<0.05) for the phylum bacteroidetes in steers supplemented with casein in the rumen compared with non-supplemented animals. Venn diagrams indicated that the number of unique bands varied considerably among individual animals and was usually higher in number for non-supplemented steers compared with supplemented animals. These results add new knowledge about the effects of ruminal and postruminal protein supplementation on metabolic activities of rumen microbes and the composition of bacterial and archaeal communities in the rumen of steers.

The effects of supplementation with different levels of crude protein on performance, intake and nutrient digestibility and efficiency of microbial protein synthesis in suckling beef calves on pasture were assessed. Fifty-five calves, with an average age of 100 days and an initial average body weight of 110 ± 7.5 kg and their respective dams, were used. The experimental design was completely randomised with five treatments and 11 replications. The experimental treatments for calves were as follows: control = calves received only mineral mixture; supplementation levels = calves received supplement containing 8, 19, 30 or 41% of crude protein (CP, at a rate of 0.5% of body weight (BW)). The cows received only mineral mixture ad libitum. Supplemented calves had higher (P < 0.1) average daily gain (ADG). Protein levels showed a quadratic effect (P < 0.1) on average daily gain (ADG) of calves. There was no difference in total dry matter (DM) intake (P > 0.1). However, intake of dry matter forage (DMF) presented cubic profiles (P < 0.1), with CP levels in the supplements. Supplementation increased (P < 0.1) the digestibility of nutrients, except for the digestibility of neutral detergent fibre. Supplementation increased (P < 0.1) the production of microbial nitrogen and N losses in urine. It can be concluded that multiple supplementations optimise the performance of beef calves on creep feeding. The intake of supplements with CP levels between 8 and 30% partially replaces of the pasture ingested by calves and increases the digestibility of the diet.

The effects of supplementation of beef calves on weight gain, intake and digestibility of pasture, milk production and composition of their dams, and behavior of the pair cow-calf were assessed. Fifty-five beef cows with an initial average body weight of 449 ± 8 kg and their respective offspring, with an initial average body weight of 138 + 3 kg and aged between 90 and 150 days, were used. Animals were submitted to an experimental period of 112 days. The experimental treatments consisted of: control = mineral mixture only, plan 1 = high protein and high carbohydrate multiple supplement, plan 2 = high protein and low carbohydrate multiple supplement, plan 3 = low protein and high carbohydrate multiple supplement, and plan 4 = low protein and low carbohydrate multiple supplement. About 25 and 12.5 % of the protein requirements were supplied by the high and low protein supplements, respectively, and 15 and 7.5 % of total digestible nutrient requirements by the high and low carbohydrate supplements, respectively. Grazing behavior, performance, milk production, milk composition, intake, and digestibility of the cows were not affected (P > 0.05) by the supplementation of the calves. The supplemented calves had greater (P < 0.05) performance (as measured by final body weight, average daily gain, and final body condition score), intake and idle time, and lower (P < 0.05) grazing time, but supplementation did not affect (P > 0.05) suckling time and suckling frequency. It can be concluded that supplementation affects the behavior and feed intake of calves. However, it does not affect the suckling time and suckling frequency of calves. Additionally, performance, milk production, nutritional characteristics, and behavior of their dams are not affected.

The performance of female calves in creep feeding under different strategies of supplementation and milk production, intake, and digestibility of grazing Nellore and crossbred cows (Nellore × Holstein) during the dry-rainy transition season were assessed. Forty-four female beef suckling calves, with initial age between 90 and 150 days and average initial body weight of 117.7 ± 4.3 kg, and their respective dams (24 Nellore and 20 crossbred) with average initial body weight of 417.5 ± 8.3 kg, were used. The experimental treatments consisted of: control group--mineral mixture only; strategy 1--supplementation from 112 days prior to weaning (0.375 kg/animal/day); strategy 2--supplementation from 112 days prior to weaning, in increasing amounts of 0.15, 0.30, 0.45, and 0.60 kg/animal/day through the four experimental periods, respectively; and strategy 3--supplementation from 56 days prior to weaning (0.750 kg/animal/day). Calves from strategy 1 had greater (P < 0.05) average daily gain (0.672 kg/day) than control animals (0.582 kg/day) and greater (P < 0.05) efficiency of supplement use than the other groups. Crossbred cows produced more milk than Nellore cows (P < 0.05). Crossbred cows presented greater (P < 0.05) dry matter intake (DMI) than Nellore cows. However, no differences were found (P > 0.05) for nutrient digestibility among genetic types. It can be concluded that strategies of supplementation that present an equitable distribution of supplement provides greater weight gain in suckling female beef calves. Crossbred cows produce more milk and present greater DMI than Nellore cows. There are no differences in the nutrient digestibility between Nellore and Nellore × Holstein crossbred cows.

The effects of supplementation with nitrogenous compounds on intake, digestibility, and microbial protein synthesis in cattle fed low-quality tropical forage were assessed. Five rumen fistulated crossbred Holstein x Gir heifers were used, with initial average live weight of 180 +/- 21 kg. Signal grass (Brachiaria decumbens) hay (48.6 g kg(-1) of crude protein (CP), on a dry matter (DM) basis) was used as roughage. Five treatments were defined according to nitrogen supplementation level (0, 20, 40, 60, and 80 g of CP kg(-1) above the CP level of the hay). A mixture of urea, ammonium sulfate, and albumin at the ratios of 4.5:0.5:1.0, respectively, was used as nitrogen source. The experiment consisted of five experimental periods, according to a 5 x 5 Latin square design. The average CP contents in the diets were 51.9, 71.1, 86.0, 116.7, and 130.2 g kg(-1), on a DM basis. A quadratic effect was detected (P < 0.10) of the CP levels in the diets on DM and neutral detergent fiber intake (kg/day), with maximum response at the levels of 102.4 and 100.5 g CP kg(-1) DM, respectively. The average daily concentration of rumen ammonia nitrogen showed increasing linear pattern (P < 0.01) as function of CP levels in the diet, with estimated value of 9.64 mg dL(-1) equivalent to the maximum DM intake. Microbial nitrogen flow in the intestine was linearly and positively related (P < 0.01) with the CP levels in the diet.

The objective of this study was to evaluate the effects of supplements with different crude protein (CP) contents on grazing cattle intake, digestibility, ruminal fermentation pattern, and nitrogen (N) metabolism characteristics during the rainy season. Five ruminal and abomasal cannulated Holstein×Zebu steers (296 kg body weight, BW) were used in a 5×5 Latin square design. The animals grazed five signal grass paddocks (0.34 ha). The five treatments evaluated were: Control (no supplement) and 1.0 g of supplement/kg BW with 0, 330, 660, and 1,000 g of CP/kg as-fed. The supplement was composed of starch, soybean meal, urea, and ammonium sulphate. There was a positive linear effect (p≤0.033) of the CP content in the supplements on the organic matter (OM), CP, and digested OM intakes. The provision of supplements did not increase (p≥0.158), on average, total and ruminal digestibilities of OM and CP. However, the increase in CP content in the supplements caused a positive linear effect (p≤0.018) on ruminal digestibilities of OM and CP. Additionally, a quadratic effect of the CP contents of the supplements were observed (p = 0.041) for the ruminal digestibility of neutral detergent fiber corrected for ash and protein, with the highest estimate obtained with the CP content of 670 g/kg. The supply of supplements increased (p<0.001) the ruminal ammonia N concentration, which also changed linearly and positively (p<0.001) according to increase in CP content in the supplements. The apparent N balance and relative N balance (g/g N intake) were not, on average, changed (p≥0.164) by the supplements supply. However, both showed a tendency of a linear increase (p≤0.099) with increasing supplement CP content. The supplements increased (p = 0.007) microbial N production in the rumen, which also changed linearly and positively (p = 0.016) with increasing supplement CP content. In conclusion, protein supplementation in grazing cattle during the rainy season, while stimulating voluntary forage intake, results in higher efficiency of N utilization when compared to energy supplementation. The aim of this study was to evaluate the effects of ruminal and abomasum protein supplementation in cattle consuming tropical forages on nutritional characteristics, fibrolytic enzymatic activity, in vitro degradation of insoluble fiber, and the ruminal microbial community profile. Two experiments were sequentially conducted and differed only in the forage quality that was offered: medium-quality forage (MQF, 78.4 g of crude protein, CP/kg of dry matter, DM) and high-quality forage (HQF, 98.6 g CP/kg DM). Four Nellore young bulls fitted with rumen and abomasum cannulas were utilized, and the following supplementation schemes were evaluated: control (only forage), daily supply of 230 g of additional CP (casein) into the rumen, daily supply of 230 g of additional CP into the abomasum, and supply of 115 g of additional CP into the rumen and 115 g of additional CP into the abomasum. Experiments were conducted using a 4 × 4 Latin square design where two sequential squares were used, one for MQF and other for HQF. The improvement in forage quality increased (P < 0.05) intake, fiber digestibility, and nitrogen retention. Supplementation, independently of site (rumen, abomasum or both), increased (P < 0.05) nitrogen balance in the rumen and in the animal’s body. Ruminal supplementation increased (P < 0.05) rumen ammonia nitrogen concentration and consequently in vitro degradation rate of neutral detergent fiber. Enzymatic activity was not affected (P > 0.05) by forage quality. However, in disagreement with data of degradation and digestibility, decreases in enzymatic activity were observed mainly for post-ruminal supplementation. The improvement of forage quality increased (P < 0.05) the richness and diversity associated with liquid fractions from ruminal fluid, although there was no effect on solid fractions. The improvement in forage quality and protein supplementation increases nitrogen accretion in the animal body. Well-defined associations cannot be established between production characteristics and enzymatic activities, and rumen microbial diversity in cattle fed tropical forages and receiving protein supplementation. Abstract: Sugar cane is widely used in an in natura forage in tropical countries, but the adoption of silage methods facilitates the preservation of its nutritional value and improves the logistics of its use. To explain differences in performance using alternative forages, it is important to conduct studies that evaluate the various digestion sites for the nutrients provided in diets. However, considering that the collection of omasal digesta is quite laborious and requires the use of a vacuum pump, reticular sampling has been suggested as a promising alternative. Thus, the objective of this study was to evaluate the intake and ruminal digestibility obtained from samples of digesta collected in the reticulum and omasum of cattle fed different diets. Five rumen-fistulated crossbred cattle with an average initial live weight of 336±16.6kg were used, being distributed in a 5×5 Latin square design. Five diets were evaluated, which contained 60% forage and 40% concentrate on dry matter basis using different forages: maize silage (CS); sugar cane in natura (SCIN); sugar cane silage (SCS0%); sugar cane silage treated with 0.4% calcium oxide (SCS0.4%) or 0.8% calcium oxide (SCS0.8%) on wet basis. The percentage of crude protein (CP) in all of the forages was corrected to 11% based on dry matter (DM) using a mixture of urea/ammonium sulfate (9:1). Six collections of reticular and omasal digesta were obtained over three days at 12h intervals. To calculate the flow of reticular and omasal nutrients, a double marker system was employed, using cobalt–EDTA and indigestible neutral detergent fiber (NDFi) as markers. The reticular and omasal digesta were similar (P>0.05) to estimate ruminal digestibility of DM, organic matter (OM), CP, neutral detergent fiber (NDF) and non-fiber carbohydrates (NFC). However, the ruminal digestibility of ether extract (EE) and the intestinal digestibility of CP and EE differed (P<0.05) between sampling sites. The results indicate that the omasal digesta is more suitable than the reticular digesta for measuring the ruminal digestion of diet components. [Copyright &y& Elsevier]

Abstract: This trial was conducted to evaluate if the effect of condensed tannin (CT) is associated with a true protein source on intake, ruminal and total digestibility, ruminal digestion rate, protein efficiency, and microbial efficiency in beef steers fed high concentrate diet (87% of DM). Four Bos indicus steers (407±12kg of BW) fitted with rumen cannula were assigned to a 4×4 Latin square design, arranged in a 2×2 factorial arrangement. Treatments consisted of either inclusion (0.4% of DM) or exclusion of condensed tannin (CT) from quebracho extract (76% of CT) with or without the use of soybean meal (SBM) as source of true protein. The level of inclusion was calculated to provide a daily intake of 1g/10kg of BW of CT. Intake of DM and nutrients was not affected (P >0.10) by CT inclusion. However, there was an effect (P <0.10) of CT inclusion on ether extract digestibility. An interaction (P <0.10) was observed between CT and SBM on ruminal digestibility and digestion rate of crude protein (CP): when mixed with soybean meal, CT decreased the ruminal digestibility and, consequently, reduced the digestion rate of CP. Intake of CP increased (P <0.10) with the inclusion of SBM. No differences in DM passage rate were observed (P >0.10) among treatments. Effects of the interaction (P <0.10) between CT and SBM were observed on flux of rumen undegradable protein (RUP), metabolizable protein (MP), and on the ratio MP:CP. In the presence of soybean meal, the addition of CT increased (P <0.10) the flux of RUP and MP, and improved the ratio MP:CP. The yield of microbial protein on the abomasum and the microbial efficiency did not differ among treatments (P >0.10). There was no difference (P >0.10) on the pH, VFA, and ruminal ammonia (N-NH3) with the addition of condensed tannin. The N-NH3 increased and the ruminal pH decreased with the inclusion of soybean meal (P <0.10). The utilization of condensed tannin as an additive in beef cattle diets with high level of concentrate and soybean meal as a source of true protein implies positive effects on crude protein utilization, decreasing digestion rate and ruminal digestibility of crude protein and consequently increasing the levels of metabolizable protein, with no changes in the ruminal fermentation parameters.
