Heat waves occurring during droughts can have a devastating impact on yield, especially if they happen during the flowering and seed set stages of the crop cycle. Global warming and climate change are driving an alarming increase in the frequency and intensity of combined drought and heat stress episodes, critically threatening global food security. Because high temperature is detrimental to reproductive processes, essential for plant yield, we measured the inner temperature, transpiration, sepal stomatal aperture, hormone concentrations and transcriptomic response of closed soybean flowers developing on plants subjected to a combination of drought and heat stress. Here, we report that, during a combination of drought and heat stress, soybean plants prioritize transpiration through flowers over transpiration through leaves by opening their flower stomata, while keeping their leaf stomata closed. This acclimation strategy, termed 'differential transpiration', lowers flower inner temperature by about 2-3°C, protecting reproductive processes at the expense of vegetative tissues. Manipulating stomatal regulation, stomatal size and/or stomatal density of flowers could serve as a viable strategy to enhance the yield of different crops and mitigate some of the current and future impacts of global warming and climate change on agriculture., (© 2022 The Authors. Historically, extended droughts combined with heat waves caused severe reductions in crop yields estimated at billions of dollars annually. Because global warming and climate change are driving an increase in the frequency and intensity of combined water-deficit and heat stress episodes, understanding how these episodes impact yield is critical for our efforts to develop climate change-resilient crops. Recent studies demonstrated that a combination of water-deficit and heat stress exacerbates the impacts of water-deficit or heat stress on reproductive processes of different cereals and legumes, directly impacting grain production. These studies identified several different mechanisms potentially underlying the effects of stress combination on anthers, pollen, and stigma development and function, as well as fertilization. Here we review some of these findings focusing on unbalanced reactive oxygen accumulation, altered sugar concentrations, and conflicting functions of different hormones, as contributing to the reduction in yield during a combination of water-deficit and heat stress. Future studies focused on the effects of water-deficit and heat stress combination on reproduction of different crops are likely to unravel additional mechanisms, as well as reveal novel ways to develop stress combination-resilient crops. Episodes of prolonged drought coupled with heat waves (i.e. drought and heat combination) can have a devastating impact on agricultural production and crop yield. It is therefore not surprising that improving tolerance to drought and heat combination has been a major goal for breeders and biotech companies. Although much is known about the physiological and molecular responses of vegetative tissues to a combination of drought and heat stress, less is known about the impact of this stress combination on yield and different yield components. Here, we used a meta-analysis approach to synthesize results from over 120 published case studies of crop responses to combined drought and heat stress. Our findings reveal that drought and heat stress combination significantly impacts yield by decreasing harvest index, shortening the life cycle of crops, and altering seed number, size and composition. Furthermore, these impacts are more severe when the stress combination is applied during the reproductive stage of plants. We further identify differences in how legumes and cereals respond to the stress combination and reveal that utilizing C3 or C4 metabolism may not provide an advantage to plants during stress combinations. Plants grown in natural environment are regularly subjected to different combinations of abiotic stresses. Recent studies revealed that citrus plants subjected to a combination of severe drought and high temperatures displayed specific physiological, hormonal, molecular and metabolic responses. In the present study, we have performed a long-term experiment combining moderate drought and heat in Cleopatra mandarin to evaluate the impact of the stress-sequence, intensity and duration. Our results support previous observation of high sensitivity of Cleopatra mandarin to abiotic stresses that include high temperatures. In this sense, a combination of drought and heat stress negatively impacts Cleopatra seedlings independently of the drought intensity. However, some responses to combined drought and heat depend on drought intensity, especially those involved in stomatal regulation. The intricate natural environment, abiotic stress combinations and global climatic changes increase the complexity of studying plant responses to stress factors in the laboratory. Under field conditions crops are routinely subjected to a number of different abiotic stress factors simultaneously. Recent studies revealed that the response of plants to a combination of different abiotic stresses is unique and cannot be directly extrapolated from simply studying each of the different stresses applied individually. These studies have also identified specific regulatory transcripts, combinations of metabolites and proteins, and physiological responses that are unique to specific stress combinations, highlighting the importance of studying abiotic stress combination in plants. Here we describe the interactions between drought and other abiotic stresses with emphasis on drought and heat stress. We compile new data about the different molecular, physiological and metabolic adaptations of different plants and crops to this stress combination and we highlight the importance of reactive oxygen species (ROS) metabolism and stomatal responses for plant acclimation to drought and heat stress combination. Background: In natural environments, several adverse environmental conditions occur simultaneously constituting a unique stress factor. In this work, physiological parameters and the hormonal regulation of Carrizo citrange and Cleopatra mandarin, two citrus genotypes, in response to the combined action of high temperatures and water deprivation were studied. The objective was to characterize particular responses to the stress combination., Results: Experiments indicated that Carrizo citrange is more tolerant to the stress combination than Cleopatra mandarin. Furthermore, an experimental design spanning 24 h stress duration, heat stress applied alone induced higher stomatal conductance and transpiration in both genotypes whereas combined water deprivation partially counteracted this response. Comparing both genotypes, Carrizo citrange showed higher phostosystem-II efficiency and lower oxidative damage than Cleopatra mandarin. Hormonal profiling in leaves revealed that salicylic acid (SA) accumulated in response to individual stresses but to a higher extent in samples subjected to the combination of heat and drought (showing an additive response). SA accumulation correlated with the up-regulation of pathogenesis-related gene 2 (CsPR2), as a downstream response. On the contrary, abscisic acid (ABA) accumulation was higher in water-stressed plants followed by that observed in plants under stress combination. ABA signaling in these plants was confirmed by the expression of responsive to ABA-related gene 18 (CsRAB18). Modulation of ABA levels was likely carried out by the induction of 9-neoxanthin cis-epoxicarotenoid dioxygenase (CsNCED) and ABA 8'-hydroxylase (CsCYP707A) while conversion to ABA-glycosyl ester (ABAGE) was a less prominent process despite the strong induction of ABA O-glycosyl transferase (CsAOG)., Conclusions: Cleopatra mandarin is more susceptible to the combination of high temperatures and water deprivation than Carrizo citrange. This is likely a result of a higher transpiration rate in Carrizo that could allow a more efficient cooling of leaf surface ensuring optimal CO2 intake. Hence, SA induction in Cleopatra was not sufficient to protect PSII from photoinhibition, resulting in higher malondialdehyde (MDA) build-up. Inhibition of ABA accumulation during heat stress and combined stresses was achieved primarily through the up-regulation of CsCYP707A leading to phaseic acid (PA) and dehydrophaseic acid (DPA) production. To sum up, data indicate that specific physiological responses to the combination of heat and drought exist in citrus. In addition, these responses are differently modulated depending on the particular stress tolerance of citrus genotypes.

Drought and heat stresses are two of the most frequent environmental factors that take place simultaneously in the field constraining global crop productivity. Metabolism reconfiguration is often behind the adaptation of plants to adverse environmental conditions. Carrizo citrange and Cleopatra mandarin, two citrus genotypes with contrasting ability to tolerate combined heat and drought conditions, showed different metabolite patterns. Increased levels of phenylpropanoid metabolites were observed in Cleopatra in response to stress, including scopolin, a metabolite involved in defense mechanisms. Tolerant Carrizo accumulated sinapic acid and sinapoyl aldehyde, direct precursors of lignins. Finally, Cleopatra showed an accumulation of flavonols and glycosylated and polymethoxylated flavones such as tangeritin. The activation of flavonoid biosynthesis in Cleopatra could be aimed to mitigate the higher oxidative damage observed in this genotype. In general, limonoids were more severely altered in Cleopatra than in Carrizo in response to stress imposition. To conclude, all metabolite changes observed in Cleopatra suggest the activation of energy metabolism along with metabolic pathways leading to the accumulation of photoprotective and antioxidant secondary metabolites, oriented to mitigate the damaging effects of stress. Conversely, the higher ability of Carrizo to retain a high photosynthetic activity and to cope with oxidative stress allowed the maintenance of the metabolic activity and prevented the accumulation of antioxidant metabolites. As a result of global warming and climate change, the number and intensity of weather events such as droughts, heat waves, and floods are increasing, resulting in major losses in crop yield worldwide. Combined with the accumulation of different pollutants, this situation is leading to a gradual increase in the complexity of environmental factors affecting plants. We recently used the term 'multifactorial stress combination' (MFSC) to describe the impact of three or more stressors occurring simultaneously or sequentially on plants. Here, we show that a MFSC of six different abiotic stressors (high light, heat, nitrogen deficiency, paraquat, cadmium, and salinity) has a negative impact on the growth, photosystem II function, and photosynthetic activity of mature tomato plants. We further reveal a negative correlation between proline accumulation and the increasing number of stress factors combined, suggesting that proline could have an adverse effect on plants during MFSC. Our findings further indicate that alterations in hormonal levels and stomatal responses are stress/stress combination-dependent, and that a tomato mutant deficient in jasmonic acid accumulation is more sensitive to high light and its combinations with salinity and/or paraquat. Taken together, our study reveals that the effects of MFSC on tomato plants are broad, that photosynthesis and proline accumulation are especially vulnerable to MFSC, and that jasmonic acid is required for tomato acclimation to MFSCs involving high light, salinity and paraquat. • Multifactorial stress combination (MFSC) negatively impacts plants. • MFSC results in specific physiological and hormonal changes in tomato. • Jasmonic acid is involved in tomato tolerance to MFSC.
