Oxidative stress is the product or aetiology of various multifactorial diseases; on the other hand, the development of multifunctional compounds is a recognized strategy for the control of complex diseases. To this end, a series of benzothiazole derivatives was synthesized and evaluated for their multifunctional effectiveness as antioxidant, sunscreen (filter), antifungal and antiproliferative agents. Compounds were easily synthesized via condensation reaction between 2-aminothiophenols and different benzaldehydes. SAR study, particularly in position 2 and 6 of benzothiazoles, led to the identification of 4g and 4k as very interesting potential compounds for the design of multifunctional drugs. New benzofuranhydrazones 3-12 were easily prepared and assayed for their radical-scavenging ability. Hydrazones 3-12 showed different extent antioxidant activity in DPPH, FRAP and ORAC assays. Good antioxidant activity is related to the number and position of hydroxyl groups on the arylidene moiety. High antioxidant activity is showed by the 2-hydroxy-4-(diethylamino)benzylidene derivative 11. Furthermore, hydrazones 3-12 showed photoprotective capacities with satisfactory in vitro SPF as compared to the commercial PBSA sunscreen filter. The antiproliferative effects of the hydrazones 3-12 was tested on erythroleukemia K562 and Colo-38 melanoma human cells. All the compounds showed growth inhibition in the micromolar to sub micromolar concentration range. If taken together these results points to benzofuran hydrazones as potential multifunctional molecules especially in the treatment of neoplastic diseases being the good antioxidant properties of 5, 7 and 11 correlated to their high antiproliferative activity., (Copyright © 2018. An ever-increasing incidence of skin neoplastic diseases is registered. Therefore, it is important to protect the skin from the UV radiation that reaches the epidermis and dermis but also to block ROS generated by them. Our attention was attracted in developing new compounds provided with both UV filtering and antioxidant capacities. To this end, 2-phenyl-1H-benzimidazole-5-sulfonic acid (PBSA), a known UV filter, was selected as lead compound for its lack of antioxidant activity, high water solubility and good safety profile. PBSA was sequentially modified introducing hydroxyls on the phenyl ring and also substituting the functional group in position 5 of the benzimidazole ring. At the end of the synthetic study, a new, very potent class of antioxidants has been obtained. Surprisingly some of the developed molecules, while devoid of significant UV-filtering activity was endowed with potent UV-filtering booster capability if associated with known commercial UVB and UVA filters.

In this study, we have designed novel chromanyl derivatives that share with alpha-tocopherol a chromanyl head but differ in the lateral chain in: (i) length and saturation (FEBL-45, 50, 70), (ii) position of double bonds in Z or E (FEBL-50 and 53 and their respective 6-chromanyl methyl derivatives FEBL-161 and 162), or (iii) presence of additional antioxidant molecules, such as the catechol compound hydroxytyrosol (FEBL-80) or dopamine (FEBL-82, 95). The efficiency of these compounds in preventing free-radical-induced oxidative stress was investigated in isolated membranes as well as intact cells. The results of this study clearly show that all compounds synthesized were active in: (i) inhibiting AAPH- or tert-BOOH-induced lipid peroxidation in microsomes and (ii) preventing H2O2-induced ROS production, cell damage, and heat-shock protein expression in immortalized RAT-1 fibroblasts. Such effects were dose- and time-dependent. Independent of the kind of pro-oxidant used, differences in the antioxidant potency of these compounds were found in relation to the chemical structure with respect to the natural alpha-tocopherol: (1) The concomitant presence of a chromanyl head and an additional aromatic ring markedly increased the antioxidant potency of the molecule. In particular, FEBL-82 and FEBL-95, resulting from the molecular combination of trolox and dopamine, were much more potent than alpha-tocopherol, alpha-tocotrienol, and the other synthetic compounds. Moreover, they were also more potent than trolox and dopamine, used alone or in combination, suggesting synergistic cooperative interactions in the molecule between chromanyl and catechol moieties. (2) The length of the side chain affected the antioxidant properties of the molecule: FEBL-70, which displays a bulky squalene side chain, was less effective than the natural alpha-tocotrienol and the synthetic FEBL-45 and FEBL-50. (3) The presence of polyunsaturated double bonds in the side chain in the Z configuration (FEBL-53, FEBL-162) increased the antioxidant potency of the molecule with respect to the E configuration (FEBL-50, FEBL-161).

Continuing our investigations on inhibitors of ribonucleotide reductase (RNR), the crucial enzyme that catalyses the reduction of ribonucleotides to deoxyribonucleotides, we have now prepared and evaluated 5'-phosphonoacetic acid, amide and ester analogues of adenosine, uridine and cytidine with the aim to verify both substrate specificity and contribution to biological activity of diphosphate mimic moieties. A molecular modelling study has been conducted on the RNR R1 subunit, in order to verify the possible interaction of the proposed bioisosteric moieties. The study compounds were finally tested on the recombinant murine RNR showing a degree of inhibition that ranged from 350 microM for the UDP analogue 5'-deoxy-5'-N-(phosphon-acetyl)uridine sodium salt (amide) to 600 microM for the CDP analogue 5'-O-[(diethyl-phosphon)acetyl]cytidine (ester). None of the tested compounds displayed noteworthy cytostatic activity at 100-500 microM concentrations, whereas ADP analogue 5'-N-[(diethyl-phosphon) acetyl]adenosine (amide) and 5'-deoxy-5'-N-(phosphon-acetyl)adenosine sodium salt (amide) showed a moderate inhibitory activity (EC50: 48 microM) against HSV-2 and a modest inhibitory activity (EC50: 110 microM) against HIV-1, respectively.

Several lines of evidence suggest potential benefits by a combination of carotenoids and tocopherols in chronic diseases. Therefore, we have designed FeAOX-6, a novel antioxidant that combines into a single molecule the chroman head of tocopherols and a fragment of lycopene, consisting of a polyisoprenyl sequence of four conjugated double bonds. The ability of FeAOX-6 in inhibiting lipid peroxidation and reactive oxygen species (ROS) production induced by different sources of free radicals (t-BOOH, AAPH, and H2O2) in arachidonic acid solution and in isolated thymocytes was investigated. Its antioxidant efficiency was also compared with that of alpha-tocopherol, lycopene, and a mixture of the two antioxidants. The results strongly suggest that FeAOX-6 can act as a potent antioxidant in our models, by inhibiting malondialdehyde production and ROS generation in a dose- and a time-dependent manner. In the cell model, the compound also provides a higher antioxidant capacity than alpha-tocopherol and lycopene, alone or in combination, suggesting the possibility of an oxidative intramolecular cooperation.

The genus Hedyosmum (family: Chloranthaceae) represents an interesting source of natural active compounds, and the 45 species of this genus are widespread in Central and South America and to a lesser extent Southeast Asia (southern China and western Malaysia). Several species are traditionally used in folk medicine. However, the data made available in recent years have not been organized and compared. The present study is a critical assessment of the state-of-the-art concerning the traditional uses, the phytochemistry and the pharmacology of species belonging to the genus Hedyosmum to suggest further research strategies and to facilitate the exploitation of the therapeutic potential of Hedyosmum species for the treatment of human disorders. The present review consists of a systematic overview of scientific literature concerning the genus Hedyosmum published between 1965 and 2018. Moreover, an older text, dated from 1843, concerning the traditional uses of H. bonplandianum Kunth has also been considered. Several databases (Francis & Taylor, Google Scholar, PubMed, SciELO, SciFinder, Springer, Wiley, and The Plant List Database) have been used to perform this work. Sixteen species of the genus Hedyosmum have been mentioned as traditional remedies, and a large number of ethnomedicinal uses, including for the treatment of pain, depression, migraine, stomach-ache and ovary diseases, have been reported. Five species have been used as flavouring agents, tea substitutes or foods. Sesterterpenes, sesquiterpene lactones, monoterpenes, hydroxycinnamic acid derivatives, flavonoids, and neolignans have been reported as the most important compounds in these species. Studies concerning their biological activities have shown that members of the Hedyosmum genus possesses promising biological properties, such as analgesic, antinociceptive, antidepressant, anxiolytic, sedative, and hypnotic effects. Preliminary studies concerning the antibacterial, antioxidant, antiplasmodial, and antifungal activities of these plants as well as their cytotoxic activities against different tumour cell lines have been reported. Some active compounds from the Hedyosmum genus have been used as starting points for the innovative and bioinspired development of synthetic molecules. A critical assessment of these papers has been performed, and some conceptual and methodological problems have been identified regarding the materials and methods and the experimental design used in these studies, including a lack of ethnopharmacological research. The present review partially confirms the basis for some of the traditional uses of Hedyosmum species (mainly H. brasiliense) through preclinical studies that demonstrated their antinociceptive and neuroprotective effects. Due to promising preliminary results, further studies should be conducted on 13-hydroxy-8,9-dehydroshizukanolide and podoandin. Moreover, several essential oils (EOs) from this genus have been preliminarily investigated, and the cytotoxic and antibacterial activities of H. brasiliense and H. sprucei EOs certainly deserve further investigation. From the promising findings of the present analysis, we can affirm that this genus deserves further research from ethnopharmacological and toxicological perspectives. Image 1 [ABSTRACT FROM AUTHOR]
