Many corals depend upon the highly specialised and intricate relationship they form with Symbiodiniaceae algal symbionts. Porites lutea is a massive reef-building coral found throughout Southeast Asia that hosts these endosymbionts obligately. Yet despite the prevalence and importance of P. lutea as one of the most dominant corals here, its associated Symbiodiniaceae communities have not been precisely characterised. In this study, we used high-throughput DNA amplicon sequencing of the nuclear internal transcribed spacer 2 (ITS2) to characterise the diversity, community structure and biogeographic distribution of Symbiodiniaceae in P. lutea throughout Singapore and Peninsular Malaysia. Consistent with previous studies, we found that Cladocopium was the most dominant genus among all samples, and Cladocopium C15 was the most dominant type (or subclade) with 100% occurrence in all samples from every study site. Results also revealed numerous Symbiodiniaceae types associated with P. lutea that were previously undetected in Southeast Asia. Endosymbiont diversity and community variation are driven by a combination of site-specific mean monthly cloud cover and variance in monthly sea surface temperature. This study contributes baseline data toward understanding differences in Symbiodiniaceae assemblages hosted by P. lutea, shedding light on how they might be indicative of particular environmental conditions and coral responses.

In this study, the complete mitogenome sequence of massive coral, Porites lutea (Cnidaria: Poritidae), has been sequenced by next-generation sequencing method. The overall base composition of Porites lutea mitogenome is 26.0% for A, 13.3% for C, 23.0% for G and 37.8% for T and have high AT content of 63.7%. The assembled mitogenome, consisting of 18 646 bp, has unique 13 protein-coding genes (PCGs), seven transfer RNAs and two ribosomal RNAs genes. The Porites lutea mitogenome has the common mitogenome gene organization and feature of scleractinian coral. Among 13 PCGs, ND5 and COX1 genes are interrupted by group I intron (11 130 and 971 bp, respectively). There are 13 genes embedded in ND5 group I intron (tRNA-Glu, ND1, CYTB, tRNA-Met, ND2, ND6, ATP6, ND4, 12S rRNA, COX3, COX2, ND4L and ND3), and two genes embedded in COX1 group I intron (tRNA-Ile and tRNA-Pro). The complete mitogenome provides essential and important DNA molecular data for further phylogenetic and evolutionary analysis for stony coral.

The coral holobiont is a complex ecosystem consisting of coral animals and a highly diverse consortium of associated microorganisms including algae, fungi, and bacteria. Several studies have highlighted the importance of coral‐associated bacteria and their potential roles in promoting the host fitness and survival. Recently, dynamics of coral‐associated microbiomes have been demonstrated to be linked to patterns of coral heat tolerance. Here, we examined the effect of elevated seawater temperature on the structure and diversity of bacterial populations associated with Porites lutea, using full‐length 16S rRNA sequences obtained from Pacific Biosciences circular consensus sequencing. We observed a significant increase in alpha diversity indices and a distinct shift in microbiome composition during thermal stress. There was a marked decline in the apparent relative abundance of Gammaproteobacteria family Endozoicomonadaceae after P. lutea had been exposed to elevated seawater temperature. Concomitantly, the bacterial community structure shifted toward the predominance of Alphaproteobacteria family Rhodobacteraceae. Interestingly, we did not observe an increase in relative abundance of Vibrio‐related sequences in our heat‐stressed samples even though the appearance of Vibrio spp. has often been detected in parallel with the increase in the relative abundance of Rhodobacteraceae during thermal bleaching in other coral species. The ability of full‐length 16S rRNA sequences in resolving taxonomic uncertainty of associated bacteria at a species level enabled us to identify 24 robust indicator bacterial species for thermally stressed corals. It is worth noting that the majority of those indicator species were members of the family Rhodobacteraceae. Coral‐associated microorganisms play an important role in their host fitness and survival. A number of studies have demonstrated connections between thermal tolerance in corals and the type/relative abundance of Symbiodinium they harbor. More recently, the shifts in coral‐associated bacterial profiles were also shown to be linked to the patterns of coral heat tolerance. Here, we investigated the dynamics of Porites lutea‐associated bacterial and algal communities throughout a natural bleaching event, using full‐length 16S rRNA and internal transcribed spacer sequences (ITS) obtained from PacBio circular consensus sequencing. We provided evidence of significant changes in the structure and diversity of coral‐associated microbiomes during thermal stress. The balance of the symbiosis shifted from a predominant association between corals and Gammaproteobacteria to a predominance of Alphaproteobacteria and to a lesser extent Betaproteobacteria following the bleaching event. On the contrary, the composition and diversity of Symbiodinium communities remained unaltered throughout the bleaching event. It appears that the switching and/or shuffling of Symbiodinium types may not be the primary mechanism used by P. lutea to cope with increasing seawater temperature. The shifts in the structure and diversity of associated bacterial communities may contribute more to the survival of the coral holobiont under heat stress.

Total genomic DNA of eight colonies for each of the two species, Acropora muricata and Porites lutea, sampled on the west coast of Reunion Island, was isolated using DNeasy Blood & Tissue kit (Qiagen™) following the manufacturer’s instructions and sent to GenoScreen, Lille, France (www.genoscreen.fr).

Large-amplitude internal waves (LAIW) cause intense drops in sea water temperature (6–9 °C) and acidity (0.6 pH units) within minutes, posing a major disturbance factor for shallow water corals in the Andaman Sea. During periods of elevated temperatures, however, they may provide a vital source of relief to heat-stressed corals. The reef coral Goniastrea aspera is regarded as one of the most environmentally tolerant species on Indo-Pacific reefs. Its demise, following a severe bleaching event in the Andaman Sea in the north-eastern Indian Ocean in 2010, was surprising in view of the rapid recovery of co-existing species such as Porites lutea. Demographic studies of G. aspera at this site showed the population was mainly composed of large individuals, which recruited in the early 1990s. These results, and size-specific mortality observed in G. aspera, post-bleaching, suggest that factors, related to size and age, may have contributed to the coral’s marked decline.

Coral reefs are threatened throughout the world. A major factor contributing to their decline is outbreaks and propagation of coral diseases. Due to the complexity of coral-associated microbe communities, little is understood in terms of disease agents, hosts and vectors. It is known that compromised health in corals is correlated with shifts in bacterial assemblages colonizing coral mucus and tissue. However, general disease patterns remain, to a large extent, ambiguous as comparative studies over species, regions, or diseases are scarce. Here, we compare bacterial assemblages of samples from healthy (HH) colonies and such displaying signs of White Plague Disease (WPD) of two different coral species (Pavona duerdeni and Porites lutea) from the same reef in Koh Tao, Thailand, using 16S rRNA gene microarrays. In line with other studies, we found an increase of bacterial diversity in diseased (DD) corals, and a higher abundance of taxa from the families that include known coral pathogens (Alteromonadaceae, Rhodobacteraceae, Vibrionaceae). In our comparative framework analysis, we found differences in microbial assemblages between coral species and coral health states. Notably, patterns of bacterial community structures from HH and DD corals were maintained over species boundaries. Moreover, microbes that differentiated the two coral species did not overlap with microbes that were indicative of HH and DD corals. This suggests that while corals harbor distinct species-specific microbial assemblages, disease-specific bacterial abundance patterns exist that are maintained over coral species boundaries. published

Porites lutea in the Andaman Sea 17 and Diploastrea heliopora in the central Red Sea 2 , is negatively correlated with regional SST. However, these studies also show that the calcification response of corals to ocean warming is highly variable, both taxonomically and geographically. It is this variability in corals’ response to warming that has compelled investigations of the ability of corals to acclimatize and/or adapt to recent and predicted future ocean warming 3 , and whether their unique acclimatization and/or adaptive capacities can be used to improve coral reef management in the face of rapid global climate change 18 .

We explored variations in polyp morphology of four ecologically important massive coral species, Porites lutea (Milne-Edwards and Haime, 1860), Diploastrea heliopora (Lamarck, 1816), Favia speciosa (Dana, 1846), and Favia matthaii (Vaughan, 1918), across a depth gradient and among sites of differing turbidity within the Wakatobi Marine National Park, Indonesia. Increased polyp area with decreased light availability was found in all target species except F. speciosa, whereas polyp density only varied across sites or depth in D. heliopora, P. lutea, and F. speciosa. Variability in polyp morphology may reflect both an optimization of light capture and an increase in heterotrophic efficiency of the host to compensate for decreased photosynthesis with depth. The morphological variability demonstrated in these key species, which acts to optimize environmental suitability, may be a crucial attribute that has contributed to the success and abundance of these species within the Wakatobi Marine National Park.

Effects of elevated seawater temperature show high spatial heterogeneity and variation within and among coral species. The objective of this study was to investigate how two coral species, Porites lutea and Galaxea fascicularis, from two high latitude reefs differently exposed to chronic disturbance, respond to elevated seawater temperatures. Corals were collected from reefs nearshore (i.e. subjected to high sediment load, higher chlorophyll α concentrations, turbidity etc.) and offshore (i.e. less exposed). The corals were exposed in the lab to gradually increasing temperatures (25.5-33.5 °C) for 72 h after which they were allowed to recover to ambient temperature (25.5 °C) for 24 h. Production and respiration were measured after 24, 48, 72 and 96 h. The results show that P. lutea from nearshore reefs suffered an initial decrease in gross primary production/respiration (GP/R) ratio after 24 h, after only a moderate temperature increase (+2 °C, from 25.5 to 27.5 °C), while there was no difference in GP/R ratio between heat-exposed and controls the other days, indicating that the chronic disturbance in the nearshore reef had no effect on their thermotolerance. Furthermore, P. lutea from the offshore reef showed a decrease in GP/R ratio both after 24 h and 72 h (33.5 °C) of exposure. In comparison, G. fascicularis showed a decrease in GP/R ratio after 48 h, 72 h and 96 h of exposure for the nearshore corals. Also, after 72 h these corals had withdrawn their polyps. There were no differences between heat-treated and controls for the offshore G. fascicularis. This implies that the chronically disturbed G. fascicularis had lower thermotolerance when exposed to a temperature increase. This study, hence, shows that the response of corals to elevated seawater temperature varies with species and environmental background history.

The metabolic plasticity of the two mounding coral species Porites lutea (Milne-Edwards and Haime, 1860) and Diploastrea heliopora (Lamarck, 1816) was investigated in the Similan Islands (Thailand), an offshore Andaman Sea island group subjected to large amplitude internal waves (LAIW). Nutrient concentrations were highly correlated with LAIW intensity and contributed to 3- and 10-fold higher symbiont densities in P. lutea and D. heliopora, respectively, along with elevated pigment concentrations, protein content, host tissue, and symbiont biomass. The comparison of LAIW-exposed and LAIW-sheltered island faces, and LAIW-intense and LAIW-weak years suggests a species-specific metabolic plasticity to LAIW, where D. heliopora benefits more from increased nutrient and organic matter availability than P. lutea. The ubiquitous LAIW in Southeast Asia and beyond may provide so far unexplored clues to coral acclimatization to disturbances on various scales, and hence, a potential key to coral resilience to climate change.

International audience; Recent surveys conducted on Reunion Island coral reefs revealed an atypical manifestation of black band disease on the main framework building coral, Porites lutea. This BBD manifestation (PorBBD) presented a thick lighter-colored band, which preceded the typical BBD lesion. Whilst BBD aetiology has been intensively described worldwide, it remains unclear if corals with apparently similar lesions across coral reefs are affectedby the same pathogens. Therefore, a multidisciplinary approach involving field surveys, gross lesion monitoring, histopathology and 454-pyrosequencing was employed to provide the first comprehensive characterization of this particular manifestation.Surveys conducted within two geomorphological zones over two consecutive summers and winters showed spatial and seasonal patterns consistent with those found for typical BBD. Genetic analyses suggested an uncharacteristically high level of Vibrio spp. bacterial infection within PorBBD. However, microscopic analysis revealed high densities of cyanobacteria, penetrating the compromised tissue as well as the presenceof basophilic bodies resembling bacterial aggregates in the living tissue, adjacent to the bacterial mat. Additionally, classical BBD-associated cyanobacterial strains, genetically related to Pseudoscillatoria coralii and Roseofilum reptotaenium were identified and isolated and the presence of sulfate-reducers or sulfide-oxidizers such as Desulfovibrio and Arcobacter, previously shown to be associated with anoxic microenvironmentwithin typical BBD was also observed, confirming that PorBBD is a manifestation of classical BBD.

Actinobacteria is a ubiquitous major group in coral holobiont. The diversity and spatial and temporal distribution of actinobacteria have been rarely documented. In this study, diversity of actinobacteria associated with mucus, tissue and skeleton of Porites lutea and in the surrounding seawater were examined every three months for 1 year on Luhuitou fringing reef. The population structures of the P. lutea-associated actinobacteria were analyzed using phylogenetic analysis of 16S rRNA gene clone libraries, which demonstrated highly diverse actinobacteria profiles in P. lutea. A total of twenty-five described families and ten unnamed families were determined in the populations, and 12 genera were firstly detected in corals. The Actinobacteria diversity was significantly different between the P. lutea and the surrounding seawater. Only 10 OTUs were shared by the seawater and coral samples. Redundancy and hierarchical cluster analyses were performed to analyze the correlation between the variations of actinobacteria population within the divergent compartments of P. lutea, seasonal changes, and environmental factors. The actinobacteria communities in the same coral compartment tended to cluster together. Even so, an extremely small fraction of OTUs was common in all three P. lutea compartments. Analysis of the relationship between actinobacteria assemblages and the environmental parameters showed that several genera were closely related to specific environmental factors. This study highlights that coral-associated actinobacteria populations are highly diverse, and spatially structured within P. lutea, and they are distinct from which in the ambient seawater.

Coral diseases are characterized by microbial community shifts in coral mucus and tissue, but causes and consequences of these changes are vaguely understood due to the complexity and dynamics of coral-associated bacteria. We used 16S rRNA gene microarrays to assay differences in bacterial assemblages of healthy and diseased colonies displaying White Plague Disease (WPD) signs from two closely related Caribbean coral species, Orbicella faveolata and Orbicella franksi. Analysis of differentially abundant operational taxonomic units (OTUs) revealed strong differences between healthy and diseased specimens, but not between coral species. A subsequent comparison to data from two Indo-Pacific coral species (Pavona duerdeni and Porites lutea) revealed distinct microbial community patterns associated with ocean basin, coral species and health state. Coral species were clearly separated by site, but also, the relatedness of the underlying bacterial community structures resembled the phylogenetic relationship of the coral hosts. In diseased samples, bacterial richness increased and putatively opportunistic bacteria were consistently more abundant highlighting the role of opportunistic conditions in structuring microbial community patterns during disease. Our comparative analysis shows that it is possible to derive conserved bacterial footprints of diseased coral holobionts that might help in identifying key bacterial species related to the underlying etiopathology. Furthermore, our data demonstrate that similar-appearing disease phenotypes produce microbial community patterns that are consistent over coral species and oceans, irrespective of the putative underlying pathogen. Consequently, profiling coral diseases by microbial community structure over multiple coral species might allow the development of a comparative disease framework that can inform on cause and relatedness of coral diseases. published

The capacity of corals for repairing partial mortality is a fundamental determinant of reef resilience. This capacity was assessed in the major reef-building coral Porites lutea by monitoring the regeneration of artificially induced lesions of standard size (330 ± 50 mm2, 3 mm deep) in 4 shallow reef flat populations at Réunion Island, under different environmental conditions related to sites and seasons, during a period without positive temperature anomalies. An exponential decay model with an asymptote described the lesion regeneration through 14 experiments. In spite of fast initial lesion regeneration, limited capacity for repair in this massive coral was indicated, as only 18% of the inflicted lesions healed completely within 6 to 9 mo. Lesion regeneration was fastest and most complete in the cooling and cool seasons, and may be impaired during the warming and hot seasons. Both solar radiation and seawater temperature contributed to seasonal changes in regeneration capacity, although they had opposite effects. While high radiation during the warming season decreased lesion regeneration rate, potentially through reduction of the photosynthetic efficiency of zooxanthellae, high temperature boosted it but also increased the amount of lesion area that could not be regenerated. Study sites were characterised by different temperature and radiation regimes, but these parameters alone could not explain all site effects on lesion regeneration capacity. Additional factors, most probably chronic stress caused by inputs of organic matter and run-off from land, may further compromise the regeneration capacity of corals and the resilience of coral populations.

[Extract] One of the largest and oldest single hermatypic coral colonies known is a massive Porites coral located on the southwest corner of Ta'u island (14°15S, 169°30W) in the Manu'a group of American Samoa. The coral is believed to be Porites lutea based on colony and corallite morphology; however, Porites taxonomy is notoriously difficult and recent genetic work has revealed surprising levels of cryptic diversity (Forsman et al.2009). Small (1 cm) cores were collected for genetic and microscopic work, and to determine skeletal density. DNA sequence for mitochondrial (COI, COIII) and nuclear (ITS region) markers were deposited in GenBank (accession numbers ITS-FJ416523-6, mt COI-FJ423967, and mt control FJ427369), and skeletal voucher specimens have been deposited at the Bishop Museum (SC4164).

Exposure of the scleractinian coral Porites lutea to elevated iron concentrations leads to a loss of zooxanthellae (symbiotic algae) from the coral tissues. This response is very marked in corals not regularly exposed to high environmental concentrations of iron. For corals exposed to daily run-off from an enriched iron effluent, the response is diminished, suggesting that these animals may have developed a tolerance to the metal. In the field, corals from a polluted site exhibited significantly higher levels of iron in their tissues than did those from an unpolluted location.

Significant differences were found in the extent to which massive coral species at Enewetak are excavated by boring organisms: Goniastrea retiformis, 7.9%; Porites lutea, 2.5%; and Favia pallida, 1.2%. While polychaetes constituted the most abundant and diverse group of coral associates, clionid sponges accounted for approximately 70–80% of skeletal damage. Clionid boring rates are initially very high but burrowing ceases when a particular burrow size (˜0.6 cm) or distance from the surface (≦2 cm) is reached. Most coral skeletal excavation occurs within 2 cm of a dead surface. Therefore, bioerosional damage to corals depends primarily on the amount of skeletal surface not covered by live coral tissue. Damage to skeletons is inversely correlated with colony size but is not correlated with coral growth rates or water depth. Massive corals have a potential escape in size from catastrophic bioerosion. Models relating 1) coral growth forms to skeletal density and stability in currents, 2) resistance of coral skeletons to breakage by water movement and suspended rubble, and 3) dead surface area on coral heads to bioerosional damage and consequent probability of detachment from the substrate, are proposed.

Alternate dense and less-dense skeletal bands in massive corals have been used for many years to record the history of growth in species such as Porites lutea and Montastrea annularis, based on the assumption that one dense band and one less-dense band is equivalent to a year's growth. This report demonstrates that specimens of Porites lutea Edwards and Haime (collected from the same neighbourhood in Phuket, Thailand, from November 1983 through November 1984) produce skeletal bands asynchronously and that one year's growth in corals from certain sites may regularly consist of four bands of varying density. The annual banding pattern observed at all sites includes the deposition of a dense band in response to high sedimentation loads and, probably, reduced light levels.

Heads of the reef-building coral Porites lutea Milne Edwards & Haime are occasionally separated or torn loose from the substratum by bioerosion or mechanical stresses. Detached heads may survive and colonize down-slope or down-stream soft bottoms, forming incipient reefs in habitats unsuitable for colonization by larval settlement. One important result of this process is reef consolidation and growth on the lagoonward edge of truncated, inter-island reef flats on atolls. In some cases, portions of the detached colonies remain attached at the original site; thus there is asexual colony multiplication.

Linear and mass growth rates are compared for the massive coral species Favia pallida (Dana), Goniastrea retiformis (Lamarck), and Porites lutea Milne Edwards & Haime at Enewetak Atoll. Marshall Islands. Goniastrea retiformis is the densest of the three species and has an intermediate growth rate; Porites lutea grows more rapidly. All three grow indeterminately at a declining rate with increasing depth. The high-density portion of annual band couplets is produced during the late summer and fall when water temperatures are highest and possibly the availability of light is reduced, and the low density portion is formed during periods of seasonally lower water temperatures and possibly higher availability of light. A similar pattern is found in three massive coral species from Belize. The high density portions of annual bands account for a greater proportion of linear and mass growth in deeper water and in corals with relatively slow growth rates. I predict that linear growth rates will be highest where conditions are most favorable for deposition of the low density portion. Geographical patterns of coral density banding based on the literature are discussed and a model is proposed relating the interplay of light availability and water temperature to the production of high and low density skeletal bands.
