Below-ground processes are crucial in determining the effects of plants on ecosystem function. The root-soil interface is a highly active zone due to root exudation and nutrient uptake. However, its role in determining effects of tree species and their interactions on the soil microbial community, ecosystem function and above-ground growth is less well known. We compared the effects of tree species monocultures and their mixture on rhizospheric microbial communities, specific functional genetic markers associated with processes in the nitrogen (N) cycle, and above-ground and below-ground growth and nutrient allocation. Two pairs of tree species were grown: Pseudotsuga menziesii and Alnus rubra; Acer pseudoplatanus and Quercus robur. Tree establishment altered soil microbial composition, but after 26 months differences amongst tree species and effects of species mixture were minor, suggesting functional redundancy in microbial communities. A greater abundance of fungi, bacteria, and specifically ammonia oxidising and denitrifying bacteria in the rhizospheric soil of the N-fixing A. rubra was the most notable trend. Mixing A. rubra with P. menziesii did produce overyielding: trees grown in mixture attained a two-fold greater (Relative Yield Total 2.03 ± 0.52) above-ground biomass than in a mixture predicted from trees grown in monoculture. We did not observe strong trends in overyielding for A. psuedoplatanus and Q. robur. Inclusion of the N-fixing species A. rubra in admixture with P. menziesii promoted N cycling, and decreased the C:N ratios of leaf, branch, and root tissues but not soil C:N ratio for P. menziesii. Given the observed overyielding in the A. rubra with P. menziesii mixtures, we explored potential mechanistic links between functional genetic markers for nitrification and ammonification, however we found no statistically significant effects attributable to these genetic markers. We found root area index was significantly lower in A. rubra monocultures than in admixture with P. menziesii. For both P. menziesii and A. rubra, the number of root tips was lower in mixture than monoculture, indicating physical partitioning of soil space as a result of growing in mixture. We documented additive and synergistic effects of tree species identity on above and belowground productivity, and rhizospheric microbial community development in these four tree species.

Tree species can influence rates of soil N transformations, but the question remains whether differences in N cycling rates are mirrored by the abundance of relevant functional genes. We studied whether the influence of tree species on soil N transformation processes and abundance of functional genes exist across two sites in British Columbia with different N availability. We used the 15 N pool-dilution method to estimate gross rates of ammonification and nitrification in forest floors of four conifers in a common garden experiment. The abundances of bacteria, fungi, nitrification (AOA amoA , AOB amoA ) and denitrification ( nirS , nirK ) genes were determined by qPCR. Western red cedar ( Thuja plicata) had the highest rates of gross ammonification and NH 4 + consumption, followed by Sitka spruce ( Picea sitchensis) , hemlock ( Tsuga heterophylla) , and Douglas-fir ( Pseudotsuga menziesii) ; all species showed net nitrate immobilization. Western red cedar forest floors had the greatest abundance of bacterial 16S genes and ammonia-oxidizing archaea amoA genes. This suggests that tree species foster different abundances of ammonification and denitrification functional groups. Differences in N transformation rates between the sites were related to site N status, as reflected in C:N ratios of the forest floor and microbial biomass, and were more closely tied to rates of N consumption rather than gross mineralization. Rates of most N transformation processes were related to microbial C:N ratio, indicating that the N status of microbes rather than their biomass or activity level determined the rates of N cycling. Ammonification rates were associated with forest floor and microbial biomass C:N ratio as well as bacterial and fungal abundances. Nitrification rates and denitrification gene abundance were associated with microbial biomass C:N ratios and AOA amoA gene abundance. The forest floor's genetic potential for denitrification was positively correlated with its nitrification potential as indicated by ammonia-oxidizer abundance. We conclude that tree species influenced forest floor N cycling and soil microbial gene abundances, and that functional genetics can be useful for exploring mechanistic links between tree species and nitrogen cycling processes.

No Mow May is a community science initiative popularized in recent years that encourages property owners to limit their lawn mowing practices during the month of May. The goal of No Mow May is to provide early season foraging resources for pollinators that emerge in the spring, especially in urban landscapes when few floral resources are available. We worked with the city council of Appleton, Wisconsin, USA. to allow No Mow May to take place in May 2020. Four hundred and thirty-five property owners registered for No Mow May in Appleton. We measured floral and bee richness and abundance in the yards of a subset of homes (N = 20) located near regularly mowed urban parks (N = 15) at the end of the month. We found that homes that participated in No Mow May had more diverse and abundant flora than regularly mowed green spaces throughout the city. No Mow May homes had three times higher bee richness and five times higher bee abundances than frequently mowed greenspaces. Using generalized linear models, we found that the best predictor of bee richness was the size of the designated unmowed area, and the best predictors of bee abundances were the size of the unmowed area as well as floral richness. While our findings cannot conclusively attribute increases in bee abundances and richness to the No Mow May efforts, our data does show that bee pollinators make use of no mow spaces as key floral resources during early spring in the upper midwestern United States. A post-No Mow May survey revealed that the participants were keen to increase native floral resources in their yards, increase native bee nesting habitat, reduce mowing intensities, and limit herbicide, pesticide, and fertilizer applications to their lawns. The No Mow May initiative educated an engaged community on best practices to improve the conservation of urban pollinators in future years.

1. Climate change is a world-wide threat to biodiversity and ecosystem structure, functioning and services. To understand the underlying drivers and mechanisms, and to predict the consequences for nature and people, we urgently need better understanding of the direction and magnitude of climate change impacts across the soil–plant–atmosphere continuum. An increasing number of climate change studies are creating new opportunities for meaningful and high-quality generalizations and improved process understanding. However, significant challenges exist related to data availability and/or compatibility across studies, compromising opportunities for data re-use, synthesis and upscaling. Many of these challenges relate to a lack of an established ‘best practice’ for measuring key impacts and responses. This restrains our current understanding of complex processes and mechanisms in terrestrial ecosystems related to climate change. 2. To overcome these challenges, we collected best-practice methods emerging from major ecological research networks and experiments, as synthesized by 115 experts from across a wide range of scientific disciplines. Our handbook contains guidance on the selection of response variables for different purposes, protocols for standardized measurements of 66 such response variables and advice on data management. Specifically, we recommend a minimum subset of variables that should be collected in all climate change studies to allow data re-use and synthesis, and give guidance on additional variables critical for different types of synthesis and upscaling. The goal of this community effort is to facilitate awareness of the importance and broader application of standardized methods to promote data re-use, availability, compatibility and transparency. We envision improved research practices that will increase returns on investments in individual research projects, facilitate second-order research outputs and create opportunities for collaboration across scientific communities. According to the pyrodiversity paradigm, a wide range of fire regimes is required to maintain biodiversity in fire-prone landscapes. However, the requisite level of pyrodiversity has seldom been tested and may actually be very low. Here, we examine the sensitivity of tropical savanna ants to variation in fire regimes using results from a long-term fire experiment near Darwin, Australia. Six experimental fire regimes, with varying fire frequency and seasonality, have been applied to 18 one-ha plots in three replicated blocks since 2004, with ants sampled prior to experimental burning and then annually after up to 2009. Our primary focus is on the extent to which different patterns of ant richness and composition are associated with each of the six treatment regimes, or whether there is such high overlap that differences only become apparent when experimental treatments are grouped to provide strongly contrasting fire regimes. When treating each of the six fire treatments separately, we were unable to detect a significant influence of fire on any ant community response variable. We were only able to detect significant ant responses when we grouped the experimental treatments into two contrasting fire frequency classes, low (burnt at most once over the 5 years) vs. high (burnt every 1 or 2 years). Even then, these responses were only evident after 3 years of fire treatment. Our findings demonstrate that ant communities have very high resilience in relation to fire, with differences evident only between strongly contrasting regimes. Such resilience appears to be characteristic of savanna ants throughout the world. Synthesis and applications. A large range of finely tuned fire regimes is unlikely to promote regional ant diversity. Rather, only very limited pyrodiversity (a combination of frequently and infrequently burnt areas) would appear to be sufficient for maintaining diversity at a landscape scale. It is important that fire management for biodiversity conservation focuses on the demonstrated requirements of target species, rather than be based on an assumption that ‘pyrodiversity begets biodiversity’.

Tree species influence soils through direct and indirect inputs above- and belowground through leaf litter and root inputs. Soil microbial communities can in turn influence tree growth and development through processes such decomposition and chemical transformation of nutrients in soils. In this chapter we will provide an overview of the mechanisms by which trees influence soil microbial communities and nitrogen cycling processes. Specifically, we explore the effects of tree species on ammonification and nitrification processes in forest floor soils, and relate those to functional genetic markers for ammonia-oxidation by archaea and bacteria (amoA AOA and AOB) bacterial denitrification (nirS and nirK). We will cover the use of complementary laboratory methods used to investigate these relationships, including the use of molecular techniques such as quantitative polymerase chain reaction (qPCR) to target gene abundances in soils, and 15N tracing experiments to understand the production and consumption of nitrogen. We will also address some of the benefits and drawbacks of these approaches, with special focus on the types of research questions that can be answered using these approaches. The chapter will wrap up with an example study in a common garden tree species trial in Vancouver, B.C., Canada, which demonstrated tree species effects on soil microbial communities and nitrogen cycling dynamics.

Globally mature forests appear to be increasing in biomass density. There is disagreement whether these increases are the result of increases in CO2 concentrations or a legacy effect of previous land-use. Recently, it was suggested that a threshold of 450 years should be used to define mature forests and that many forests increasing in biomass may be younger than this. However, the study making these suggestions failed to account for interactions between forest age and climate. Here we revisit the issue to identify: (1) how climate and forest age control global forest biomass density and (2) whether we can set a threshold age for mature forests. Using data from previously published studies we modelled the impacts of forest age and climate on biomass density using linear mixed effects models. We examined the potential biases in the dataset by comparing how representative it was of global mature forests in terms of its distribution, the climate space it occupied and the ages of the forests used. Biomass density increased with forest age, mean annual temperature and annual precipitation. Importantly the effect of forest age increased with increasing temperature, but the effect of precipitation decreased with increasing temperatures. The dataset was biased towards Northern hemisphere forests in relatively dry, cold climates. The dataset was also clearly biased towards forests

The decline of Tsuga canadensis (eastern hemlock)?a foundation tree species?due to infestation by Adelges tsugae (hemlock woolly adelgid) or its complete removal from a stand by salvage logging dramatically affects associated faunal assemblages. Among these assemblages, species composition (richness and abundance) of ants increases rapidly as T. canadensis is lost from the stands. Because ants live and forage at the litter-soil interface, we hypothesized that environmental changes caused by hemlock loss (e.g., increased light and warmth at the forest floor, increased soil pH) and shifts in ant species composition would interact to alter soil ecosystem variables. In the Harvard Forest Hemlock Removal Experiment (HF-HeRE), established in 2003, T. canadensis in large plots were killed in place or logged and removed to mimic adelgid infestation or salvage harvesting, respectively. In 2006, we built ant exclosure subplots within all of the canopy manipulation plots to examine direct and interactive effects of canopy change and ant assemblage composition on soil and litter variables. Throughout HF-HeRE, T. canadensis was colonized by the adelgid in 2009, and the infested trees are now declining. The experimental removal of T. canadensis from the canopy was associated with an increase in the rate of cellulose decomposition by >50%, and exclosure of ants from subplots directly reduced their soil nitrate availability by 56%. Partial least squares path models revealed sequential interactive effects prior to adelgid infestation: canopy change (as a proxy for associated environmental changes) altered both decomposition and ant assemblage structure; changes in ant assemblage structure and decomposition rates altered nitrogen availability. The results illustrate that biotic changes directly associated with decline of T. canadensis can have cascading effects on ecosystem nutrient availability and cycling.

Ecological studies aim to better understand the distribution and abundances of organisms. Yet ecological works often are subjected to unintentional biases thus an improved framework for hypothesis testing should be used. Double-blind ecological studies are rare but necessary to minimize sampling biases and omission errors and improve the reliability of research. We used a double-blind design to evaluate associations between nests of red wood ants(Formica rufa,RWA) and the distribution of tectonic faults. We randomly sampled two regions in western Denmark to map the spatial distribution of RWA nests. We then calculated nest proximity to the nearest active tectonic faults. Red wood ant nests were eight times more likely to be found within 60 meters of known tectonic faults than were random points in the same region but without nests. This pattern paralleled the directionality of the fault system, with NNE-SSW faults having the strongest associations with RWA nests. The nest locations were collected without knowledge of the spatial distribution of active faults thus we are confident that the results are neither biased nor artefactual. This example highlights the benefits of double-blind designs in reducing sampling biases, testing controversial hypotheses, and increasing the reliability of the conclusions of research.

Summary 1. Direct and indirect consequences of global warming on ecosystem functions and processes mediated by invertebrates remain understudied but are likely to have major impacts on ecosystems in the future. Among animals, invertebrates are taxonomically diverse, responsive to temperature changes, and play major ecological roles which also respond to temperature changes. 2. We used a mesocosm experiment to evaluate impacts of two warming treatments (+3� 5 and +5 °C, set-points) and the presence and absence of the ant Formica subsericea (a major mediator of processes in north temperate ecosystems) on decomposition rate, soil movement, soil respiration and nitrogen availability. 3. Replicate 19-L mesocosms were placed outdoors in lathe houses and continuously warmed for 30 days in 2011 and 85 days in 2012. Warming treatments mimicked expected temperature increases for future climates in eastern North America. 4. In both years, the amount of soil displaced and soil respiration increased in the warming and ant presence treatments (soil movement: 73–119%; soil respiration: 37–48% relative to the control treatments without ants). 5. Decomposition rate and nitrogen availability tended to decrease in the warmest treatments (decomposition rate: � 26 to � 30%; nitrate availability: � 11 to � 42%). 6. Path analyses indicated that ants had significant short-term direct and indirect effects on the studied ecosystem processes. These results suggest that ants may be moving more soil and building deeper nests to escape increasing temperatures, but warming may also influence their direct and indirect effects on soil ecosystem processes.

Tsuga canadensis (Eastern Hemlock) forests are experiencing widespread mortality due to the invasive insect Adelges tsugae (Hemlock Woolly Adelgid). This study sought to document the community response to HWA across latitude. I selected two sensitive response groups—plants and ants— to use as biological indicators of ecosystem change to monitor differences along a natural gradient of Eastern Hemlock mortality among 3 forest types: relatively healthy Eastern Hemlocks, dead or dying Eastern Hemlocks, and hardwood stands. I sampled understory vegetation, ants, and soils at each site and compared sites using a linear mixed-model to discern the best predictors of species density. I also compared analyses of variances across forest types among response variables. There was an average two-fold increase in understory vegetation species density between Eastern Hemlock and hardwood forests; ant species density was not influenced by forest type. Climate change affects communities both directly and indirectly via changes in interspecific interactions. One such interaction that may be altered under climate change is the ant-plant seed dispersal mutualism common in deciduous forests of eastern North America. As climatic warming alters the abundance and activity levels of ants, the potential exists for shifts in rates of ant-mediated seed dispersal. We used an experimental temperature manipulation at two sites in the eastern US (Harvard Forest in Massachusetts and Duke Forest in North Carolina) to examine the potential impacts of climatic warming on overall rates of seed dispersal (using Asarum canadense seeds) as well as species-specific rates of seed dispersal at the Duke Forest site. We also examined the relationship between ant critical thermal maxima (CTmax) and the mean seed removal temperature for each ant species. We found that seed removal rates did not change as a result of experimental warming at either study site, nor were there any changes in species-specific rates of seed dispersal. There was, however, a positive relationship between CTmax and mean seed removal temperature, whereby species with higher CTmax removed more seeds at hotter temperatures. The temperature at which seeds were removed was influenced by experimental warming as well as diurnal and day-to-day fluctuations in temperature. Taken together, our results suggest that while temperature may play a role in regulating seed removal by ants, ant plant seed-dispersal mutualisms may be more robust to climate change than currently assumed. Red spruce (Picea rubens) populations experienced a synchronous rangewide decline in growth and vigor starting in the 1960s, likely caused by climate change and a combination of environmental disturbances. However, it is not yet known if populations continue to decline or have recovered. Red spruce growing near its southern range margin in Massachusetts is a species of concern, in light of the vulnerability to climate change. This study uses population data from 17 permanent plots coupled with tree-ring data to examine radial growth rates, determine the growth-climate relationship, and document disturbance events. Red spruce at these plots ranged from 90 to 184 years old, and comprised 15 to 29 m(2)/ha basal area. Red spruce seedlings and saplings were common at plots with previously high overstory spruce abundance, indicating it could return to a more dominant position under favorable growing conditions. However, permanent plot measures over a 50 year time span did not indicate any consistent trends for changes in basal area or density for red spruce or other woody species. Climate data show that mean annual minimum, maximum, and summer temperatures have increased over the last 100 years. Dendroclimatological analyses indicated that red spruce growth was sensitive to both temperature and precipitation. Prior to the 1960s, spruce at these sites showed a positive response to precipitation; however after a multi-year drought in the 1960s showed an increasingly negative correlation with precipitation. There has been a negative growth response to regional warming, as spruce radial growth was mostly constrained by increasing temperatures, potentially coupled with the associated increasing drought-dress. I suggest the change in climate response is potentially due to a physiological threshold response to increasing temperatures, which may cause spruce to continue to decline or be lost from the lower elevation sites, while the high elevation sites has a persistent spruce population.
