Macrocyclic oligoesters (MCOs) derived from biomass such as those prepared from alkylene alkanedioates and 2,5-furandicarboxylates are suitable compounds for ring opening polymerization (ROP) to produce high molecular weight bio-based polyesters. The synthetic procedures that have been recently developed to obtain these MCOs in good yields, their ROP either alone or together with other cyclic compounds, and their recovery by cyclodepolymerization of the final polyesters are the main topics covered in this review. Both organometallic compounds and enzymes such as Candida antarctica lipase B are the catalytic systems used for all reactions. This synthetic approach based on ROP of MCOs derived from renewable sources opens a green pathway for a circular economy in plastics.

Triblock/grafted poly(ether-ester-peptide) copolymers made or PEG, poly(globablide) and poly(L-phenylalanine) were synthesized by sequential ROP starting from diaminated PEG as first initiator. The aromatic functionality of L-phenylalanine inserted on the globalide units was utilized to impart the amphiphilicity required to drive their self-assembly in water and to provide aromaticity of potential value for the efficient loading of aromatic drugs. Depending of the method used for preparation, nanoparticles possessing diameters between 25 and 600 nm were obtained by self-assembling in aqueous medium.

In this communication we wish to report on the synthesis of PBFxSy copolyesters by ring opening polymerization (ROP) using either organometallic catalysts or enzymes. The ROP of macrocyclic oligomers (MCOs) has been demonstrated to offer significant advantages over the traditional polycondensation method. The synthesis of MCOs of butylene succinate c(BS)n is known from long whereas MCOs of butylene 2,5-furandicarboxylate (c(BF)n) have not been reported until very recently. In this work, the synthesis of c(BF)n and c(BS)n has been performed using high dilution condensation (HDC) and enzymatic cyclization, respectively. Mixtures of dimer to tetramer and dimer to nonamer were obtained for c(BF)n and c(BS)n respectively.

In this work we have prepared and compared the thermal and mechanical properties of aliphatic polyesters made from aliphatic diacids with different polymethylene chain lengths (2, 4, 6, 8) and three bicyclic sugar derived diols namely, dianhydro-1,4:3,6-D-glucitol (also known as isosorbide, Is), and two bicyclic diazetalized hexitols, 2,4:3,5-di-O-methylene- D-mannitol (Manx-diol) and -D-glucitol (Glux-diol) derived from D-mannose and D-glucose respectively (Fig. 1)

The ring opening polymerization (ROP) of these macrolactones mediated by alcohols as initiators and organic and metal-based catalysts has been reported before with satisfactory results [4,5]. In the present work, the polymerization of these macrolactones has been studied using amines as initiators. The ROP of both PDL and 6HDL was carried out in bulk at 100 oC using 1,5,7-triazabicyclo[4.4.0]dec-5-ene (TBD) as organocatalyst. Distinct types of amines were tested as initiators for the ROP of these macrolactones. The copolymerization reaction was also studied by ROP of PDL and 6HDL with an oligo-polyethylene glycol bis(3-aminopropyl) terminated and an amino-ended homopolypeptide (poly(¿-benzyl a,Lglutamate) as initiator.

Poly(ß,L-malic acid) (PMLA) was made to interact with the cationic anticancer drug Doxorubicin (DOX) in aqueous solution to form ionic complexes with different compositions and an efficiency near to 100%. The PMLA/DOX complexes were characterized by spectroscopy, thermal analysis, and scanning electron microscopy. According to their composition, the PMLA/DOX complexes spontaneously self-assembled into spherical micro or nanoparticles with negative surface charge. Hydrolytic degradation of PMLA/DOX complexes took place by cleavage of the main chain ester bond and simultaneous release of the drug. Poly (β-L-malic acid) (PMLA) is a biodegradable polymer and it has various important applications in the biomedical field. In the present work the structural and spectral characteristics of PMLA have been studied by methods of infrared, Raman spectroscopy and quantum chemistry. Electrostatic potential surface, optimized geometry, harmonic vibrational frequencies, infrared intensities and activities of Raman scattering were calculated by density functional theory (DFT) using oligomeric approach employing B3LYP with complete relaxation in the potential energy surface using 6-311++G (d, p) basis set. Based on results, we have discussed the correlation between the vibrational modes and the structure of the PMLA. A complete analysis of the experimental infrared and Raman spectra has been reported on the basis of wavenumber of the vibrational bands and potential energy distribution. The calculated HOMO and LUMO energies shows that charge transfer occur within the molecule. The calculated infrared and the Raman spectra of the polymer based on DFT calculations show reasonable agreement with the experimental results.

PMLA nanoparticles with diameters of 150–250nm are prepared, and their hydrolytic degradation is studied under physiological conditions. Degradation occurs by hydrolysis of the side chain methyl ester followed by cleavage of the main-chain ester group with methanol and L-malic acid as the final degradation products. No alteration of the cell viability is found after 1 h of incubation, but toxicity increases significantly after 3 d, probably due to the noxious effect of the released methanol. Anticancer drugs temozolomide and doxorubicin are encapsulated in the NPs with 20–40% efficiency, and their release is monitored using in vitro essays. Temozolomide is fully liberated within several hours, whereas doxorubicin is steadily released from the particles over a period of 1 month.

Esterification of microbial poly(malic acid) is performed with either ethanol or 1-butanol to obtain polymalate conjugates capable to form nanoparticles (100–350 nm). Degradation under physiological conditions takes place with release of malic acid and the corresponding alcohol as unique degradation products. The anticancer drugs Temozolomide and Doxorubicin are encapsulated in nanoparticles with efficiency of 17 and 37%, respectively. In vitro drug release assays show that Temozolomide is almost completely discharged in a few hours whereas Doxorubicin is steadily released along several days. Drug-loaded nano-particles show remarkable effectiveness against cancer cells. Partially ethylated poly(malic acid) nano-particles are those showing the highest cellular uptake.

Cyclic butylene furandicarboxylate (c(BF)n) and butylene isophthalate (c(BI)n) oligomers obtained by high dilution condensation reaction were polymerized in bulk at 200 ¿C with Sn(Oct)2 catalyst via ring opening polymerization to give homopolyesters and copolyesters (coPBFxIy) with weight average molar masses in the 60,000–70,000 g·mol-1 range and dispersities between 1.3 and 1.9. The composition of the copolyesters as determined by NMR was practically the same as that of the feed, and they all showed an almost random microstructure. The copolyesters were thermally stable up to 300 ¿C and crystalline for all compositions, and have Tg in the 40–20 ¿C range with values decreasing almost linearly with their content in isophthalate units in the copolyester. Both melting temperature and enthalpy of the copolyesters decreased as the content in butylene isophthalate units increased up to a composition 30/70 (BF/BI), at which the triclinic crystal phase made exclusively of butylene furanoate units changed to the crystal structure of PBI. The partial replacement of furanoate by isophthalate units decreased substantially the crystallizability of PBF

In this communication we wish to report on HyA ionic complexes prepared by coupling with a) choline-based surfactants (nACh·HyA complexes), and b) alkylphosphonium surfactants (nATMP·HyA complexes) with n being the number of carbons of the alkanoyl or alkyl side chain. nACh surfactants are synthesized from choline and fatty acids, and distinguished because they are fully biobased and biocompatible. The nATMP surfactants have been scarcely studied as counterions of polyacids in spite of being more stable to heat and less harmful than their alkylammonium analogs.

In this work, the synthesis of both c(BF)n and c(BT)n has been performed using the high dilution cyclization technique and these cycles have been then used for the preparation of the homopolyesters PBF and PBT, as well as of a series of poly(butylene 2,5-furandicarboxylate-co-terephthalate) (coPBFxTy) with a wide range of compositions (Fig. 1). Results are compared with those obtained from similar copolyesters prepared by melt polycondensation, which have been recently reported

When synthetic polymers are married to proteins or peptides, the resulting bioconjugates can synergistically combine the properties of the individual components and overcome their separate limitations. This Thesis is focused on the study of hybrid copolymers based on polypeptides and polymacrolactones. Block and graft copolymers have been synthesized by making use of the ring opening polymerization method (ROP) mainly and extensively characterized including both their chemical structure and their structure in the solid state. The self-assembly properties of the new copolymers have been preliminary examined regarding their potential application as nanocarriers for pharmaceutical compounds. This Thesis initially reports the ROP of w-pentadecalactone (PDL) using different amino-ended initiators and assisted by either organic or enzymatic catalysts. This method was then extended for the ROP of PDL using bisamino-ended poly(ethylene glycol) (PEG) for the preparation of poly(w-pentadecalactone)-b-poly(ethylene glycol)-b-poly(wpentadecalactone) [PPDLx-PEG-PPDLx] triblock copolymers. These amphiphilic ABA-type copolymers were able to selfassemble in water to form nanoparticles with diameters between 100 and 200 nm. Hybrid copolymers of poly(ester-peptide) or poly(ether-ester-peptide) type exhibiting different architectures (e.g. diblock, triblock, graft or triblock/grafted) respectively, were then synthesized using as building blocks: poly(w-pentadecalactone), poly(globalide) (PGl), PEG as well as polypeptides derived from the L-glutamic acid (Glu), L-lysine (Lys), L-alanine (Ala) and L-phenylalanine (Phe) a-amino acids. The hybrid copolymers were synthesized through several stages depending on the desired architecture. The first stage in the preparation of these copolymers was the synthesis of macroinitiators from PDL or PGl containing either an amino group at the end of the chain or multiple amine groups along their polymeric chain. In the second stage, such macroinitiators were used to trigger the polymerization of the a-amino acid N-carboxyanhyrides (NCA) with the COOH group of L-glutamic acid and NH2 of L-lysine duly protected as g-benzyl-L-glutamate (BLG) and eNcarbobenzoxy-L-lysine (ZLL) respectively. Some copolymers containing BLG or ZLL units were treated with acids to render copolymers bearing the amino acids residues with their COOH or NH2 functionalities in the free form. All of the synthesized copolymers were fully characterized through GPC and NMR spectroscopy. The thermal properties were studied by TGA and DSC techniques. The conformation adopted by the peptide-based copolymers in the solid-state was assessed by FTIR, and their crystalline structure was examined by X-ray diffraction using synchrotron radiation in most cases. The conformation in aqueous solution of water-soluble copolymers containing Glu or Lys residues in the free form was explored by circular dichroism. The self-assembly behavior in aqueous medium of all the amphiphilic copolymers was investigated with the purpose of obtaining nanoparticles with the appropriated diameters required for their application as biomedical nanocarriers. The nanoparticles were duly characterized by light scattering and SEM and TEM microscopies. Block and graft copolymers were able to load doxorubicin and release it under pH control. Copolymers containing L-lysine were shown to be able of condensing DNA. The potential of these copolymers as DDS of anticancer drugs and vectors for transfection have been evidenced. Los polipéptidos o proteínas obtenidos de manera natural son considerados como polímeros altamente refinados. Cuando los polímeros sintéticos se unen a proteínas o polipéptidos, los sistemas bioconjugados que se obtienen pueden sinérgicamente combinar las propiedades de sus componentes individuales y mejorar las propias limitaciones que tienen por separado. La proteína o el elemento polipeptídico puede impartir propiedades bifuncionales al bioconjugado, mientras que el polímero sintético puede mejorar la estabilidad proteica, la solubilidad y la biocompatibilidad. Esta tesis está enfocada en el estudio de copolímeros híbridos basados en polipeptidos y polimacrolactonas. Copolímeros tipo bloque e injerto fueron sintetizados utilizando principalmente la polimerización por apertura de anillo y extensamente caracterizados tanto su estructura química, como su estructura en estado sólido. Las propiedades de auto-agregación de los nuevos copolímeros han sido anteriomente examinadas respecto a su potencial aplicación como nanotransportadores de compuestos farmacéuticos. Esta Tesis inicialmente reporta la homopolimerización de w-pentadecalactona (PDL) usando diferentes iniciadores aminoterminados mediante el uso de catalizadores tanto orgánicos como enzimáticos. Este se extiende a la ROP de PDL usando poli(etilén glicol) bisamino-terminado (PEG) para la preparar copolímeros tribloque poli(w-pentadecalactona)-b-poli(etilén glicol)-b-poli(w-pentadecalactona) [PPDLx-PEG-PPDLx]. Estos copolímeros de tipo ABA fueron capaces de auto-agregarse en agua para formar nanopartículas con diámetros entre 100 y 200 nm. Por otra parte, sistemas híbridos de tipo poli(éster-péptido) o poli(éter-éster-péptido) que presentan distintas arquitecturas (por ejemplo dibloque, tribloque, injerto, o tribloque-injertado) respectivamente, se sintetizaron utilizando como bloques de construcción derivados de macrolactonas (w-pentadecalactona), globalida) y a-amino ácidos (ácido L-glutámico (Glu), Llisina (Lys), L-alanina (Ala) y L-fenilalanina (Phe) así como poli(etien glicol) telequélico. Los copolímeros híbridos fueron sintetizados en varias etapas dependiendo de cuál fuese la arquitectura deseada. La primera etapa fue la preparación de los macroiniciadores a partir de PDL o PGl conteniendo en su estructura ya sea un grupo amino en el extremo de la cadena, o múltiples grupos aminos a lo largo de la cadena polimérica. En la segunda etapa, los macroiniciadores fueron utilizados en la polimerización de a-amino ácidos N-carboxianhídridos (NCA), con los grupos COOH del ácido L-glutámico y el grupo NH2 de la L-lisina apropiadamente protegidos como g-bencil-L-glutamato (BLG) y eN-carbobenzoxi-L-lisina (ZLL) respectivamente. Para los copolímeros que contienen bloques peptídicos de BLG o ZLL, las funcionalidades COOH o NH2 fueron regeneradas bajo condiciones ácidas, para producir así los copolímeros conteniendo el amino ácido en su forma libre. Todos los copolímeros sintetizados fueron completamente caracterizados mediante GPC y espectroscopia de RMN. Las propiedades térmicas fueron estudiadas por las técnicas de TGA y DSC. La conformación adoptada por los copolímeros en el estado sólido fue estudiada por FTIR, y su estructura cristalina fue analizada mediante difracción de rayos X usando radiación sincrotrón en la mayoría de los casos. La conformación en solución acuosa de los copolímeros solubles en agua, que contienen residuos de Glu o Lys, fue analizada por dicroísmo circular. Se estudió el comportamiento de todos los copolímeros para auto-agregarse en agua obteniéndose partículas con diámetros del orden nanométrico, como se demostró por DLS así como también por SEM y TEM, las cuales son apropiadas para ser aplicadas en biomedicina. Las nanopartículas de copolímeros dibloque y de injerto conteniendo ácido L-glutámico fueron capaces de incorporar doxorubicina y efectuar su liberación bajo control por medio del pH. Por otro lado, los copolímeros dibloque y de injerto con bloques conteniendo L-lisina mostraron la habilidad de condensar el ADN, demostrando así su potencial uso como vectores en transfección. Aplicat embargament des de la data de defensa fins el dia 1/3/2019 In recent years, the increasing public concern about the environmental pollution caused by persistent plastic wastes has stimulated the interest in replacing synthetic polymers by biopolymers. The term biopolymer refers to polymers that are renewable or biodegradable or both. Biopolymers are capable of bio-assimilation at accelerated rates so they are biocompatible with the environment. Furthermore, their sustainability is of exceptional interest; the renewable origin of biopolymers makes them inexhaustible in contrast with the uncertain accessibility at medium-term of synthetic polymers produced from fossil sources. Nevertheless, biopolymers often have inferior properties compared to commodity polymers. Modification is a way to improve properties and achieve property combinations required for specific applications. Therefore the synthesis, characterization and property evaluation of new biopolymers derivatives are essential tasks that have to be done for the development of new materials able to replace the traditional plastics in areas such as industrial, medical, food, consumer products, and pharmaceutical applications. In the present Thesis, the chemical modification of two kinds of carboxylic biopolymers has been studied to respond to the necessity of creating new biopolymer derivatives with advanced properties at reasonable cost. Poly (gamma-glutamic acid) (PGGA) and hyaluronic acid (HyA) were the biopolymers selectedin this Thesis for their capacity to form stable ionic complexes with cationic surfactants to generate stable materials with new properties. These complexes are currently object of intensive research in our group due to their outstanding features. They are easily prepared and they tend to be selfassembled in well-ordered amphiphilic structures able to respond reversibly to thermal effects. This behaviour is of high scientific interest and also of practical relevance in the design of medical devices for thermally and chemically controlled store and delivery of drugs. The main goal of this Thesis is the preparation of ionic complexes of the two mentioned polyacids using different cationic surfactants depending on the desired final properties. The first part of the work is devoted to provide physicochemical knowledge of the structure and properties of alkyltrimethylphosphonium surfactants which have potential interest for novel applications. Then these surfactants were coupled to both PGGA and HyA to obtain the respective ionic complexes with biocide activity and thermal stability higher than those made by their ammonium analogs. PGGA complexes, abbreviated as nATMP·PGGA, have high interest as food preserving and packaging applications displaying as main advantage the edibility of the polymer and the possibility of improving their basic properties through blending with nanoclays. On the other hand, HyA complexes, nATMP·HyA, are useful to obtain HyA derivatives with antimicrobial activity. In addition, the preparation of nanoparticles of nATMP·HyA with antimicrobial properties was feasible using the ionotropic gelation method. The s econd part of the Thes is was devoted to the preparation of “greener” complexes of hyaluronic acid. For this regard, alkanoylcholine surfactants were used to prepare the ionic complexes nACh·HyA. These complexes constitute a highly promising biocompatible/biodegradable platform for the design of systems suitable for drug transport and targeting delivery in anticancer chemotherapy because it was demonstrated that non-cytotoxic nanoparticles can be prepared from these systems. The third part of the Thesis is dedicated to the preparation of biocompatible antimicrobial complexes using as cationic surfactant one of the most potent food preservative agents that is known today, that is the ethyl alpha-N-lauroyl L-arginate chloride surfactant (also known as LAE). These complexes (LAE·PGGA and LAE·HyA) are shown to be potential candidates to develop antimicrobial materials. En els últims anys s’ha produït un augment de la preocupació sobre la contaminació produïda pels plàstics persistents que ha estimulat l’interès per reemplaçar els polímers sintètics pels biopolímers. El terme biopolímer es refereix als polímers que son o bé renovables o biodegradables, o ambdós. A més, és de excepcional interès la seva sostenibilitat; l’origen renovable dels biopolímers els fa inesgotables al contrari dels polímers sintètics, que al ser produïts a partir de fonts fòssils presenten una accessibilitat incerta a mig termini. No obstant, els biopolímers presenten de vegades unes propietats inferiors si se'ls comparem amb els polímers tradicionals. La modificació és una via per millorar les propietats i arribar a les combinacions de propietats requerides per a aplicacions específiques. Per tant, la síntesi, caracterització i avaluació de propietats de nous derivats de biopolímers són tasques essencials que s’han de realitzar pel des envolupament de nous materials capaços de reemplaçar els plàstics tradicionals en sectors com ara la indústria química, medicina, alimentació, productes de consum i farmàcia. En la present Tes i, s’ha estudiat la modificació química de dos tipus de biopolímers carboxílics per respondre a la necessitat de crear derivats de biopolímers amb propietats avançades i a un preu raonable. L’àcid poli(gamma-glutámic) (PGGA) i l’àcid hialurònic (HyA) van ser seleccionats per la seva capacitat per formar complexes iònics estables amb surfactants catiònics per generar materials estables amb noves propietats. Aquests complexes es preparen fàcilment i tendeixen a autoassemblar-se en estructures amfifíliques ordenades, capaços de respondre reversiblement a efectes tèrmics. Aquest comportament és d’elevat interès científic i de rellevància pràctica en el disseny de dispositius mèdics per l’emmagatzematge i alliberament de fàrmacs de manera tèrmica i químicament controlada. El principal objectiu d’aquesta Tes i es la preparació de complexes iònics dels dos poliàcids mencionats utilitzant diferents surfactants catiònics depenent de les propietats finals desitjades. La primera part del treball està dedicada a proporcionar el coneixement físic-químic de l’estructura i propietats dels surfactants d’alquiltrimetilfos foni, els quals tenen un interès potencial per a noves aplicacions. Posteriorment, aquests surfactants foren acoblats als dos biopolímers, PGGA i HyA, per obtenir els respectius complexes iònics amb propietats biocides i de major estabilitat tèrmica que aquells complexes preparats pels seus anàlegs d’amoni. Els complexes de PGGA, tenen un alt interès com a preservadors d’aliments i aplicacions d’envasat tenint com a principal avantatge la comestibilitat d’aquest polímer i la possibilitat de millorar les seves propietats bàsiques mesclant aquests compostos amb nanoargiles. Per altra banda, els complexes de HyA, són útils per obtenir derivats de l’àcid hialurònic amb activitat antimicrobiana. A més, s’ha dut a terme la preparació de nanopartícules de nATMP·HyA amb propietats antimicrobianes utilitzant el mètode “ionotropic gelation”. La segona part de la Tesi es va focalitzar en la preparació de complexes sostenibles d’àcid hialurònic. Per a realitzar-ho es van utilitzar surfactants d’alcanoilcolines per preparar els complexes iònics nACh·HyA. Aquests complexes constitueixen una prometedora plataforma biocompatible/biodegradable pel disseny de sistemes capaços de transportar i alliberar de forma específica els fàrmacs anticancerígens de quimioteràpia. La tercera part de la Tesi està dedicada a explorar la preparació de complexes biocompatibles amb propietats antimicrobianes utilitzant com a surfactant catiònic un dels preservants alimentaris més potents que es coneixen actualment, el LAE. Aquests complexes (LAE·PGGA i LAE·HyA) són candidats potencials per desenvolupar films comestibles amb propietats microbianes i materials d’àcid hialurònic amb propietats microbianes respectivament

These complexes constitute a highly promising biocompatible/biodegradable platform for the design of systems suitable for drug transport and targeting delivery in anticancer chemotherapy because it was demonstrated that non-cytotoxic nanoparticles can be prepared from these systems. The third part of the Thesis is dedicated to the preparation of biocompatible antimicrobial complexes using as cationic surfactant one of the most potent food preservative agents that is known today, that is the ethyl alpha-N-lauroyl L-arginate chloride surfactant (also known as LAE). These complexes (LAE·PGGA and LAE·HyA) are shown to be potential candidates to develop antimicrobial materials. En els últims anys s’ha produït un augment de la preocupació sobre la contaminació produïda pels plàstics persistents que ha estimulat l’interès per reemplaçar els polímers sintètics pels biopolímers. El terme biopolímer es refereix als polímers que son o bé renovables o biodegradables, o ambdós. A més, és de excepcional interès la seva sostenibilitat; l’origen renovable dels biopolímers els fa inesgotables al contrari dels polímers sintètics, que al ser produïts a partir de fonts fòssils presenten una accessibilitat incerta a mig termini. No obstant, els biopolímers presenten de vegades unes propietats inferiors si se'ls comparem amb els polímers tradicionals. La modificació és una via per millorar les propietats i arribar a les combinacions de propietats requerides per a aplicacions específiques. Per tant, la síntesi, caracterització i avaluació de propietats de nous derivats de biopolímers són tasques essencials que s’han de realitzar pel des envolupament de nous materials capaços de reemplaçar els plàstics tradicionals en sectors com ara la indústria química, medicina, alimentació, productes de consum i farmàcia. En la present Tes i, s’ha estudiat la modificació química de dos tipus de biopolímers carboxílics per respondre a la necessitat de crear derivats de biopolímers amb propietats avançades i a un preu raonable. L’àcid poli(gamma-glutámic) (PGGA) i l’àcid hialurònic (HyA) van ser seleccionats per la seva capacitat per formar complexes iònics estables amb surfactants catiònics per generar materials estables amb noves propietats. Aquests complexes es preparen fàcilment i tendeixen a autoassemblar-se en estructures amfifíliques ordenades, capaços de respondre reversiblement a efectes tèrmics. Aquest comportament és d’elevat interès científic i de rellevància pràctica en el disseny de dispositius mèdics per l’emmagatzematge i alliberament de fàrmacs de manera tèrmica i químicament controlada. El principal objectiu d’aquesta Tes i es la preparació de complexes iònics dels dos poliàcids mencionats utilitzant diferents surfactants catiònics depenent de les propietats finals desitjades. La primera part del treball està dedicada a proporcionar el coneixement físic-químic de l’estructura i propietats dels surfactants d’alquiltrimetilfos foni, els quals tenen un interès potencial per a noves aplicacions. Posteriorment, aquests surfactants foren acoblats als dos biopolímers, PGGA i HyA, per obtenir els respectius complexes iònics amb propietats biocides i de major estabilitat tèrmica que aquells complexes preparats pels seus anàlegs d’amoni. Els complexes de PGGA, tenen un alt interès com a preservadors d’aliments i aplicacions d’envasat tenint com a principal avantatge la comestibilitat d’aquest polímer i la possibilitat de millorar les seves propietats bàsiques mesclant aquests compostos amb nanoargiles. Per altra banda, els complexes de HyA, són útils per obtenir derivats de l’àcid hialurònic amb activitat antimicrobiana. A més, s’ha dut a terme la preparació de nanopartícules de nATMP·HyA amb propietats antimicrobianes utilitzant el mètode “ionotropic gelation”. La segona part de la Tesi es va focalitzar en la preparació de complexes sostenibles d’àcid hialurònic. Per a realitzar-ho es van utilitzar surfactants d’alcanoilcolines per preparar els complexes iònics nACh·HyA. Aquests complexes constitueixen una prometedora plataforma biocompatible/biodegradable pel disseny de sistemes capaços de transportar i alliberar de forma específica els fàrmacs anticancerígens de quimioteràpia. La tercera part de la Tesi està dedicada a explorar la preparació de complexes biocompatibles amb propietats antimicrobianes utilitzant com a surfactant catiònic un dels preservants alimentaris més potents que es coneixen actualment, el LAE. Aquests complexes (LAE·PGGA i LAE·HyA) són candidats potencials per desenvolupar films comestibles amb propietats microbianes i materials d’àcid hialurònic amb propietats microbianes respectivament

In recent years, as a result of climate change, the interest in sustainable materials has increased dramatically. The replacement of compounds of petrochemical origin by bio-based has spread to all areas of chemistry. Nowadays there is a development of sustainable polymers based in 2,5-furanedicarboxylic acid (FDCA) as it may compete in performance with the ones based on terephthalic acid (PTA). On the other hand, the entropically driven ring opening polymerization (ED-ROP) is an attractive synthesis route whose interest has grown in this last decade because the reaction is athermal, no byproducts are generated and polymers of high molecular weight can be obtained in short reaction times. This Thesis reports the synthesis and characterization of cyclic oligomers of FDCA with different diols, such as 1,4-butanediol (c(BF)n), ethylene glycol (c(EF)n), resorcinol (c(RF)n) and isomannide (c(ImF)n) using different synthetic routes including high dilution condensation (HDC) and cyclodepolymerization (CD). The cyclic oligomers of butylene isophthalate (c(BI)n) and butylene terephthalate (c(BT)n) have also been synthesized by HDC. Finally, the cyclic oligomers of butylene succinate (c(BS)n) and ethylene succinate (c(ES)n) were obtained by enzymatic cyclization reaction (EC). The furanic cyclic oligomers were homopolymerized and copolymerized with the aforementioned cyclic oligoesters as well as with epsilon-caprolactone (e-CL). The polymerization reactions were catalyzed with tin octanoate (SnOct2), except in the case of homopolyesters and copolyesters with aliphatic units in which the Candida antarctica lipase B (CALB) was used. The molecular weights of the polymers obtained ranged between 25,000 and 80,000 g / mol. The copolymers presented a random microstructure with the exception of the caprolactone derivatives, which presented a blocky microstructure. Differential scanning calorimetry (DSC) studies revealed that PEF, PBF, PBT, PBI, PBS, PES, and their copolymers with less than 30% of aliphatic comonomer, turned out to be crystalline, on the contrary, copolymers with close compositions to 50:50 were usually amorphous. On the other hand, the PRF and the PImF were amorphous and the incorporation of their units in other copolyesters restricted the their crystallinity. In addition, the melting temperature (Tm) and the glass transition temperature (Tg) were intermediate to those of the homopolyesters, with a practically linear tendency in the second case. The crystallizability of most copolyesters was also studied, which allowed obtaining different kinetic parameters through the application of the Avrami equation. Thermogravimetric analysis (TGA) revealed that most polyesters and copolyesters were thermally stable up to 400 oC. The studies of hydrolytic and enzymatic degradability showed the high resistance to the degradation of PBF, PEF and PRF, which decreased markedly with the incorporation of aliphatic units such as BS, ES, CL or ImF in the copolyesters. This thesis summarizes the main aspects in the synthesis of cyclic oligomers of FDCA and its homopolymerization and copolymerization with different cyclic oligomers via ED-ROP. According to the results obtained, polyesters based on furanic derivatives could be considered as a viable alternative to polyesters of petrochemical origin for industrial applications., En estos últimos años, como consecuencia del cambio climático, ha aumentado extraordinariamente el interés por materiales sostenibles. El reemplazo de los compuestos de origen petroquímico por naturales se ha extendido a todos los ámbitos de la química. Se están desarrollando polímeros sostenibles basados en ácido 2.5-furandicarboxílico (FDCA) ya que pueden competir en prestaciones con los basados en el ácido tereftálico (PTA). Por otro lado, la polimerización entrópica por apertura de anillo (ED-ROP) es una vía de síntesis atractiva cuyo interés ha crecido en esta última década debido a que la reacción es atérmica, no se generan subproductos y se puede obtener polímeros de alto peso molecular en cortos tiempos de reacción. En esta Tesis se reporta la síntesis y caracterización de oligómeros cíclicos de FDCA con diferentes dioles, tales como 1,4- butanodiol (c(BF)n), etilenglicol (c(EF)n), resorcinol (c(RF)n) e isomannide (c(ImF)n) usando diferentes rutas sintéticas incluyendo la condensación con alta dilución (HDC), la ciclodepolimerización (CD). También se han sintetizado mediante HDC los oligómeros cíclicos de butilén isoftalato (c(BI)n) y butilén tereftalato (c(BT)n). Finalmente, se han obtenido los oligómeros cíclicos de butilén succinato (c(BS)n) y etilén succinato (c(ES)n) por reacción de ciclación enzimática (EC). Los oligómeros cíclicos furánicos se homopolimerizaron y copolimerizaron con los oligoesteres cíclicos mencionados anteriormente así como con la epsilon-caprolactona (e-CL). Las reacciones de polimerización fueron catalizadas con el octanoato de estaño (SnOct2), excepto en el caso de los homopoliésteres y copoliésteres con unidades alifáticas en los que se utilizó la enzima lipasa B de Cándida antárctica (CALB). Los pesos moleculares de los polímeros obtenidos oscilaron entre 25,000 y 80,000 g/mol. Los copolímeros presentaban una microestructura al azar con la excepción de los derivados de caprolactona, los cuales presentaron una microestructura en bloques. Los estudios de calorimetría diferencial de barrido (DSC) revelaron que el PEF, PBF, PBT, PBI, PBS, PES, y sus copolímeros con menos del 30% de comonómero alifático, resultaron ser cristalinos, por el contrario, los copolímeros con composiciones cercanas al 50:50 resultaron ser por lo general amorfos. Por otro lado, el PRF y el PImF eran amorfos y la incorporación de sus unidades en otros copoliésteres restringió la cristalinidad de los mismos. Además la temperatura de fusión (Tm) y la temperatura de transición vítrea (Tg) eran intermedias a las de los homopoliésteres, con una tendencia prácticamente lineal en el segundo caso. También se estudió la cristalizabilidad de la mayoría de los copoliésteres, lo cual permitió la obtención de diferentes parámetros cinéticos mediante la aplicación de la ecuación de Avrami. El análisis termogravimétrico (TGA) reveló que la mayoría de los poliésteres y copoliésteres eran térmicamente estables hasta 400 oC. Los estudios de degradabilidad hidrolítica y enzimática mostraron la alta resistencia a la degradación del PBF, PEF y PRF, la cual decrecía notablemente con la incorporación de unidades alifáticas tales como el BS, ES, CL o ImF. La presente Tesis resume los principales aspectos en la síntesis de oligómeros cíclicos de FDCA y su homopolimerización y copolimerización con diferentes oligómeros cíclicos vía ED-ROP. De acuerdo con los resultados obtenidos, los poliésteres basados en derivados furánicos podrían considerarse como una alternativa viable a los poliésteres de origen químico.

In this century, the major use of synthetic polymers have been as replacements for more traditional materials, particulary in packaging. Today the packaging industry is by far the major user of plastics. Another interesting application of these materials is drug delivery systems. Polymers have played an integral role in the advancement of drug delivery technology by providing controlled release of therapeutic agents in constant doses over long periods, cyclic dosage and tunable release of both hydrophilic and hydrophobic drugs. Modern advances in drug delivery are now predicated upon the rational design of polymers tailored for specific cargo and engineered to exert distinct biological functions. Aliphatic polyesters such as poly(L-lactic acid), poly(butylene succinate), and polyhydroxyalkanoates among others, constitute primary examples of bio-based polymers that distinguish by being fully renewable and displaying partial or total biodegradability. This Ph.D. Thesis is devoted to the synthesis of aliphatic random and block polyesters from renewable resources with application for packaging and drug delivery. The main goal of this project is to develop new bio-based polymers with similar or even improved properties compared to those of conventional plastics obtained from non-renewable sources. The two cyclic acetals, 2,3-di-O-methylene-L-threitol and dimethyl 2,3-di-O-methylene-L-threarate, were used for the synthesis of two series of PBS copolyesters differing in which unit, butylene or succinate, was replaced, in addition of the corresponding parent homopolyesters. 2,4:3,5-di-O-methylene-D-glucitol was used for the synthesis of PBS copolyesters by melt polycondensation. Three series of polyalkanoates (adipates, suberates and sebacates) were synthesized using as monomers three sugar-based bicyclic diols derived from D-glucose and D-mannose. ABA triblock copolyesters were synthesized by ROP of L-lactide in solution initiated by telechelic D-glucose- and L-tartaric-based polyester macroinitiators. The synthesized polyesters were characterized by nuclear magnetic resonance (NMR) spectroscopy, gel permeation chromatography (GPC) and viscosimetry. Thermal properties were analyzed by differential scanning calorimetry (DCS) and thermogravimetry (TGA). Crystalline structure of polyesters was studied by X-ray and its mechanical properties were evaluated as well. Hydrolytic degradation and biodegradation assays were followed by GPC and NMR. Nanoparticles made from triblock copolyesters were characterized by scanning electron microscopy (SEM) and dynamic light-scattering (DLS)., En este siglo, el mayor empleo que se les ha dado a los polímeros sintéticos ha sido como sustitutos de materiales más tradicionales, en particular, materiales para envase y embalaje. Hoy en día la industria de envasado es, por mucho, la mayor consumidora de plásticos. Otra aplicación interesante de estos materiales son los sistemas de administración de fármacos. Los polímeros han jugado un papel fundamental la tecnología de administración de fármacos al proporcionar un medio de liberación controlada de agentes terapéuticos en dosis constantes durante largos periodos; la dosificación cíclica y la liberación sintonizable de fármacos hidrofílicos e hidrofóbicos. Actualmente, los avances modernos en la administración de fármacos se basan en el diseño racional de polímeros para la liberación de dosis controladas que modifican distintas funciones biológicas de forma específica. Los poliésteres alifáticos tales como el poli(ácido L-láctico), el poli(succinato de butileno) y los polihidroxialcanoatos, entre otros, constituyen ejemplos primarios de polímeros de base biológica que se distinguen por ser completamente renovables y presentar biodegradabilidad parcial o total. Este trabajo de doctorado está dedicado a la síntesis de poliésteres alifáticos al azar y de bloques a partir de recursos renovables con aplicación para el envasado y la administración de fármacos. El objetivo principal de este proyecto es desarrollar nuevos polímeros biológicos con propiedades similares o incluso mejores en comparación a los plásticos convencionales obtenidos a partir de fuentes norenovables. Los dos acetales cíclicos, 2,3-di-O-metileno-L-treitol y dimetil 2,3-di-O-metileno-L-trearato, se usaron para la síntesis de dos series de copoliésteres de PBS que difieren en qué unidad, butileno o succinato, se reemplazó, además de los homopoliésteres originales correspondientes. Se usó 2,4:3,5-di-O-metileno-D-glucitol para la síntesis de copoliésteres de PBS mediante policondensación en estado fundido. Se sintetizaron tres series de polialcanoatos (adipatos, suberatos y sebacatos) usando como monómeros tres dioles bicíclicos base azúcar derivados de D-glucosa y D-manosa. Los copoliésteres tribloques ABA se sintetizaron mediante ROP de L-láctido en solución iniciada por macroiniciadores de poliester telequelico basados en D-glucosa y L-tartárico. Los poliésteres sintetizados se caracterizaron por espectroscopia de resonancia magnética nuclear (RMN), cromatografía de permeación en gel (GPC) y viscosimetría. Las propiedades térmicas se analizaron mediante calorimetría de barrido diferencial (DCS) y termogravimetría (TGA). La estructura cristalina de los poliésteres se estudió mediante rayos X y sus propiedades mecánicas también se evaluaron. La degradación hidrolítica y los ensayos de biodegradación fueron seguidos por GPC y NMR. Las nanopartículas hechas de copoliésteres tribloque se caracterizaron por microscopía electrónica de barrido (SEM) y dispersión de luz dinámica (DLS).

A polymer containing small amounts of ionic groups either along the polymer backbone chains or as pendant groups is defined as ionomer. As originally proposed by Eisenberg, the interaction between ionic groups leads to the formation of multiplets containing a small number of ion pairs, and also to ionic clusters, which constitute a second phase made of many multiplets as well as portions of the hydrocarbon chains. These ionic structures have been shown to act as strong electrostatic cross-links, which provide altered physical properties, such as enhanced mechanical properties and high melt viscosity, among others. Both aromatic polyesters as aliphatic are polymeric materials widely used today. Their most familiar applications are in clothing, food packaging and water and carbonated soft drinks bottles. Major part of biodegradable synthetic polymers are polyesters, and in particular, aliphatic polyesters. Monomers for the latter can be synthesized from renewable resources. Biodegradable polyesters play a key role in medical applications due to their biodegradability and versatile synthesis able to afford tailored properties; they are currently employed as biomaterials for medical purposes such as surgical sutures, scaffolds, screws and reinforcing plates as well as controlled release drug carriers, since they are biocompatible and nontoxic. This thesis is focused to the chemical modification of aromatic and aliphatic polyesters through the incorporation of various concentration and different ionic groups. We synthesized poly(hexamethylene terephthalate) and poly(butylene succinate)-based ionomers and investigated the effects of the ionic substitution on physical properties, crystallization rates and hydrodegradability of these polyester ionomers. In addition, we also studied the influence of the ionic interaction with nanoclays in the formation of ionomers-based nanocomposites. This thesis embodies a multidisciplinary task work that includes synthesis, spectroscopy characterization, evaluation of thermal and mechanical properties, hydrolytic degradation and nanocomposites preparation: the synthesis procedure of copolyesters was made in all cases by polycondensation in melt-phase. Different conditions of temperature, catalyst and time required were used for each family of copolyesters depending on both the stability and the reactivity of the comonomers involved in the synthesis. The copolyesters synthesized incorporated sulfonated, ammonium, trimethylamonium and tributylphosphonium ionic groups into their chains. The hydrolytic degradation results suggest that the insertion of sulfonated groups in the copolyesters make them more hydrophilic allowing better solvation and an easier attack of water on the ester groups. Nanocomposites with nanoclays were obtained by using different approaches for mixing and in some cases by making use of compatibilizers. Octadecyltrimethylammonium, 1,12-dodecylenediammonium salts and a terpolyester ionomer were used for compatibilization. The clay was used either unmodified or modified with alkylammonium soaps. Results obtained by the different procedures were compared and the effect of the compatibilizers on the mixing efficiency and composite properties were evaluated. Characterization was carried out by 1H and 13C nuclear magnetic resonance (NMR) spectroscopy, FTIR spectroscopy, gel permeation chromatography (GPC), differential scanning calorimetry (DSC), and thermogravimetry analysis (TGA). The changes in crystal structure upon copolymerization were followed by X-ray diffraction (XRD)., Un polímero que contiene pequeñas proporciones de grupos iónicos, ya sea a lo largo de la cadena principal como en los grupos laterales, es definido como ionómero. Según lo propuesto originalmente por Eisenberg, la interacción entre los grupos iónicos conduce a la formación de multipletes que contienen un grupo reducido de pares iónicos, y también a la formación de clusters, los cuales constituyen una segunda fase compuesta de muchos multipletes, así como de porciones de cadena hidrocarbonada. Estas estructuras iónicas han demostrado actuar como enlaces electrostáticos fuertes entrecruzados, los cuales producen modificacion en las propiedades físicas tales como una mejora de las propiedades mecánicas y un incremento en la viscosidad en fundido, entre otras. Tanto los poliésteres aromáticos como los alifáticos son materiales poliméricos usados ampliamente en la actualidad. Sus aplicaciones más conocidas van desde el campo textil hasta el envasado de alimentos y líquidos como el agua o bebidas carbónicas. La mayoría de los polímeros sintéticos biodegradables son poliésteres alifáticos. Estos poliésteres suelen ser biocompatibles y no tóxicos y juegan un papel clave en aplicaciones médicas debido a su síntesis versátil capaz de proporcionar propiedades a medida. Actualmente se vienen empleando como biomateriales en suturas quirúrgicas, implantes óseos porosos, tornillos y placas de refuerzo, así como portadores de fármacos de liberación controlada. Esta tesis está enfocada en la modificación química de poliésteres aromáticos y alifáticos mediante la incorporación de diferentes grupos iónicos y con diversas concentraciones. Se han sintetizado ionómeros en base al poli(hexametilen tereftalato) y poli(butilen succinato) y se han investigado los efectos de la sustitución iónica sobre las propiedades físicas, velocidades de cristalización e hidrodegradabilidad de estos poliésteres. Además, se ha estudiado la influencia de las interacciones iónicas con nanoarcillas en la formación de nanocompuestos a base de ionómeros. Esta tesis supone un trabajo multidisciplinar que incluye síntesis, caracterización espectroscópica, evaluación de propiedades térmicas y mecánicas, estudio de la degradación hidrolítica y preparación de nanocompuestos; el proceso de síntesis de los poliésteres ha sido realizado en todos los casos mediante policondensación en estado fundido. Se han utilizado condiciones diferentes de temperatura, catalizador y tiempo para cada familia de poliésteres dependiendo de la estabilidad y de la reactividad de los comonomeros involucrados en la síntesis. Los copoliésteres sintetizados incorporaron en sus cadenas grupos iónicos sulfonados, amonio, trimetilamonio y tributil fosfonio. Los resultados de la degradación hidrolítica sugieren que la inserción de grupos sulfonados en los poliésteres, los hace más hidrofílicos permitiendo una mejor solvatación y un ataque más fácil del agua a los grupos éster. Los nanocompuestos con nanoarcillas fueron obtenidos mediante diferentes metodologías de mezclado y en algunos casos haciendo uso de compatibilizadores. Para la compatibilización se usaron el bromuro de octadeciltrimetilamonio, el dicloruro de 1,12-dodecildiamonio y un ionómero terpoliéster. Se empleó la arcilla montmorillonite tanto sin modificar como modificada con grupos alquilamonio. Se compararon los resultados obtenidos mediante los diferentes procedimientos y se evaluaron los efectos de los compatibizadores en la eficacia de mezclado y en las propiedades de los composites. La caracterización se llevó a cabo mediante resonancia magnética nuclear de 1H y 13C (NMR), espectroscopia de infrarrojo, cromatografía de exclusión molecular (GPC), calorimetría diferencial de barrido (DSC), y análisis de termogravimétrico (TGA). Los cambios en la estructura cristalina debidos a la copolimerización fueron seguidos mediante difracción de rayos X (XRD).

Three series of aromatic copolyesters derived from poly(ethylene terephthalate) (PET), poly(butylene terephthalate) (PBT) and poly(hexamethylene terephthalate) (PHT) have been synthesized by melt polycondensation in which the terephthalate and oxyalkylene units have been partially or totally replaced by monocyclic and bicyclic diacids and diols obtained by derivatization of renewable monomers such as tartaric acid and D-glucose respectively. Another series of aliphatic copolyesters derived from poly(butylene sebacate) has been prepared by both melt polycondensation and enzymatic polymerization in solution where both the sebacate and butylene units have been partially or totally replaced by the bicyclic diacid or diol derived from D-glucose. All series of polyesters and copolyesters were characterized by FTIR and NMR in solution. The chemical composition of the copolyesters was determined by 1H NMR. On the other hand the microstructure, determined by 13C NMR was observed to be at random for all series. The molecular weights determined by intrinsic viscosimetry and gel permeation chromatography (GPC) were high but decreasing when the content of the sugar monomer increased. The thermal properties were evaluated by differential scanning calorimetry (DSC) and thermogravimetric analysis. It was observed that the thermal stability was not affected when the oxyalkylene unit was the replaced unit, but decreased slightly when the terephthalate unit was replaced one. The new copolyesters showed higher glass transition temperatures than the parent poly(alkylen terephthalate)s, being superiors for the copolyesters obtained from the bicyclic derivative of D-glucose than for the monocyclic derivative of tartaric acid. All copolyesters were less crystalline than the parent homopolyesters, with both melting and enthalpies decreasing with the increased content of the sugar units. The crystalline structure determined by X-ray diffraction was similar to the PET, PBT and PHT polyesters. Finally it was studied the hydrolytic and enzymatic degradability by weight loss, molecular weight determinations and scanning electron microscopy (SEM). All new copolyesters showed enhanced degradability in water and biodegradability., En esta Tesis se han sintetizado mediante reacción de policondensación en masa tres series de copoliesteres aromáticos derivados del poli(etilen tereftalato) (PET), poli(butilen tereftalato) (PBT) y poli(hexametilen tereftalato) (PHT) en los que se han sustituido parcial o totalmente las unidades tereftalicas o las unidades oxialquilénicas por diácidos y dioles cíclicos y bicíclicos obtenidos por derivatización de monómeros de origen natural como son el ácido tartárico y la D-glucosa. También se han sintetizado mediante policondensación en masa y en disolución por vía enzimática una serie de copoliesteres alifáticos derivados del poli(butilen sebacato) en los que las unidades oxibutilénicas y sebácicas se han reemplazado por dioles y diácidos bicíclicos derivados de la D-glucosa. Todas las series se han caracterizado espectroscópicamente mediante FTIR y RMN en disolución. Mediante RMN de 1H se ha determinado la composición de los copolímeros. Por otro lado mediante RMN de 13C se ha estudiado la microestructura de los mismos. Dicho estudio reveló que todos los copolímeros eran al azar. Los pesos moleculares determinados mediante cromatografía de permeabilidad en gel y viscosimetría capilar resultaron ser aceptables, aunque en todos los casos disminuían a medida que aumentaba el contenido del monómero de origen renovable. Las propiedades térmicas se han evaluado mediante calorimetría diferencial de barrido (DSC) y análisis termogravimétrico. Se ha observado que la estabilidad térmica no se ve apenas afectada cuando la sustitución se realiza sobre la unidad oxialquilénica y que por otro lado desciende cuando es la unidad tereftálica la sustituida. Los nuevos copolímeros presentaron temperaturas de transición vítrea superiores, observándose que este aumento era superior en los copolímeros que incorporaban el diol o el diácido bicíclico derivado de la D-glucosa que en los que incorporaban el diol o el diácido monociclico derivado del ácido tartárico. Los copolimeros eran por lo general menos cristalinos, menos cristalizables y con temperaturas de fusión inferiores. La estructura cristalina de los mismos determinada mediante difracción de rayos-X era semejante a la de los homopolimeros PET, PBT y PHT. Se ha estudiado la degradabilidad hidrolitica y la biodegradabilidad de todos ellos mediante medidas de pérdida de peso, pesos moleculares, microscopía electrónica de barrido (SEM), observándose un aumento de ambas propiedades para los nuevos copolímeros.

Polymers are becoming preferred materials in biomedical applications because of their vast diversity of properties, functionalities and applications. Properties as mechanical strength, stability against degradation, biocompatibility and biodegradability, among others, have been attractive for different medical applications. One of the most interesting applications of these materials is drug delivery systems. Biodegradable polymers and copolymers are the preferred materials for the manufacture of a variety of devices for temporal applications in medicine and pharmacology; these biodegradable polymers can be chemically synthesized or biologically produced. Biotechnological polymers have attracted much attention because two main advantages; first, they are produced from renewable resources; and second, as they are biologically produced they are biocompatible, biodegradable and bioresorbable materials. Thus, modification of biotechnological polymers to obtain specific properties or functionalities is a good strategy for the development of promising biomedical materials. In this Thesis the water soluble biotechnological polymer poly(ß,L-malic acid) (PMLA) was modified to change its hydrophilic character to produce non water-soluble polymers capable of forming particulate systems for drug encapsulation and controlled release. PMLA is a polyester-3 with a pendant carboxylic group; it is biocompatible, biodegradable and bioresorbable. The carboxylic side group can be substituted in order to modify the properties of the polymer. The polymer as polyelectrolyte, is also water-soluble. Different strategies were used for polymer derivatization: direct esterification and amidation through the activation of the carboxylic side groups with carbodiimides; ionic complex formation with a cationic drug (Doxorubicin); and esterification with aliphatic long chains by a two step method employing thiol-ene click reactions. Obtained PMLA derivatives resulted in hydrophobic or amphiphilic polymers, which were appropriated for nanoparticle formation, either by emulsion solvent evaporation or by precipitation dialysis techniques. Derivatives physicochemical characterization was made by 1H and 13C nuclear magnetic resonance (NMR) spectroscopy, gel permeation chromatography (GPC) and differential scanning calorimetry. Hydrolytic degradation was followed by GPC and 1H NMR, while particles were observed by scanning electron microscopy and their size and surface charge characterized by dynamic light scattering and ζ-potential measurement. Assays of drug encapsulation and release were also performed and cytotoxicity tests were done on cancer cell lines. Nanoparticles (100-300 nm aprox.) were obtained from all PMLA derivatives, except for the ionic complex which formed microparticles. Nanoparticles showed potential as drug delivery systems since they were able to encapsulate the anticancer drugs Temozolomide and Doxorubicin, as well as the model drugs Theophylline and Carbamazepine. Drug release was assessed under physiological conditions; the release rate was found to depend on encapsulation method, drug and polymer derivative. Hydrolytic degradation assays showed that free malic acid and the organic compound derived from the reagent used for modification were the last products of aqueous degradation of PMLA derivatives. Cytotoxicity tests demonstrated the low toxicity of the synthesized derivatives. Results generated in this Thesis suggest that the biotechnological polymer PMLA is a material of interest as a platform for the design and development of biodegradable drug delivery systems with potential in the therapy of diseases considered today challenging for pharmacologic treatment.

In the present work was carried out the surface modification of microfibril and cellulose whiskers. Two different techniques were tested, one of them was the liquid phase modification and the other one, gas phase modification. Five reactants were tested, palmitoyl chloride, acetyl chloride, sebacoyl chloride and two anhydrides allow to functionalize the surface of the cellulose, granting it the hydrophobic character required. The liquid phase modification was made just with palmitoyl chloride, to perform this reaction was hard to find the conditions (not available information, no previous work) and many things to do to carry out the reaction, one of that was the exchange of solvent, from water to DMF; residual water and the size of the particles was the parameter to help to decide whether the exchange had worked. From the results we can say that the right temperature to accomplish this type of modification is 90 ° C and the time can change between 24 and 96hours, always taking care of the amount of pyridine to add. The gas phase modification was made with the five reactants, in all cases (except with maleic anhidride) the cellulose was modified. Was possible to find two reactants that just modify the surface of the crystal, sebacoyl chloride and ASA anhidride, the last one is environmentally friendly; in these cases the DSg was lower than DSsurface.

Esta tesis doctoral está dirigida al desarrollo de poliuretanos a partir de dioles derivados de carbohidratos. Los poliuretanos son polímeros muy versátiles que pueden ser usados en una amplia variedad de aplicaciones que van desde espumas de relleno a artículos técnicos. Actualmente, existe un interés creciente por parte de la industria del poliuretano en la incorporación de productos procedentes de fuentes renovables. Dioles derivados de grasas naturales y aceites así como otros dioles de producción biotecnológica como el 1,3-propanodiol y ácido poliláctico han sido utilizados en la síntesis de estos polímeros. Hasta el momento, algunos poliuretanos derivados de sacáridos han sido descritos aunque ninguno de ellos ha alcanzado nivel industrial; incluyendo polímeros derivados de lignina polihidroxilada o dioles cíclicos como isosorbide y gulonolactonas. La Tesis describe la preparación de poliuretanos lineales a partir de dioles cíclicos derivados de carbohidratos, específicamente ácido tartárico y alditoles derivados del ácido tartárico, y arabino- y xilopiranosa. Además, otros dioles naturales derivados de la glucosa pero con estructura cíclica, 1,4:3,6-dianhidrosorbitol y 2,4:3,5-di-O-metilen-D-glucitol fueron usados. Todo los dioles usados en esta Tesis son accesibles vía síntesis química o son directamente comerciales, y por lo tanto son una buena opción para la introducción de productos naturales en la química del poliuretano. La incorporación de estas unidades hidrofílicas produce poliuretanos más susceptibles a la hidrólisis al ser incubados en soluciones neutras, ácidas o básicas. El primer bloque de esta Tesis está dedicado a la síntesis, caracterización y estudio de degradabilidad hidrolítica de poliuretanos derivados de alditoles con los grupos secundarios protegidos como éter de metilo, éter de bencilo o acetal. La hidrólisis total o parcial de los éteres bencílicos o los grupos acetal en el poliuretano es factible mediante hidrogenólisis o mediante tratamiento con ácidos, respectivamente, estos métodos proporcionan una ruta conveniente para la obtención de poliuretanos con grupos hidroxilo laterales, que son sumamente hidrofílicos, aumentando así la hidrodegrabilidad del poliuretano. El segundo bloque describe la síntesis de poliuretanos a partir de ésteres del ácido tartárico. En este caso, los tartratos se polimerizan directamente con los diisocianatos, obteniéndose poliuretanos con ésteres alquílicos o bencílicos laterales. La degradabilidad hidrolítica de estos poliuretanos fue estudiada mediante incubación en medio ácido o básico, observándose un aumento de la hidrodegradabilidad respecto a los poliuretanos lineales no funcionalizados. El tercer bloque está dedicado al estudio de poliuretanos segmentados. Debido al creciente interés por la introducción de extensores de cadena derivados de recursos renovables, y también con el objetivo de realzar la hidrodegradabilidad de los poliuretanos resultantes, dos dioles derivados de la glucosa fueron usados para la obtención de poliuretanos segmentados con segmento flexible de policaprolactona. Se estudiaron sus propiedades térmicas, mecánicas e hidrolíticas. Finalmente, el último bloque está dedicado al fenómeno de la estereocomplejación. Se describe el método de preparación, la formación, evidenciada por diferentes técnicas espectroscópicas, y la cristalizabilidad del estereocomplejo formado por dos politartaramidas enantioméricas., The presented Thesis is addressed to the development of polyurethanes made from carbohydrate derived diols. Polyurethanes are extremely versatile polymers that can be used in a wide variety of applications going from massive filling foams to highly technical items. The preparation of polyurethanes from renewable feedstocks is currently receiving increasing attention in the polyurethane industry. Diols derived from naturally-occurring fats and oils as well as biotechnologically produced diols like 1,3-propanediol and hydroxyl end-caped poly(lactic acid) have been recently introduced in the synthesis of these polymers. A fair number of polyurethanes containing units derived from saccharides have been reported so far although none of them has reached industrialization; the selection embraces polymers made from complex polyhydroxylated lignin fragments to cyclic diols such as isosorbide and gulonolactones. The Thesis describes the preparation of linear polyurethanes from acyclic carbohydrate derived diols, specifically tartaric acid and alditols from tartaric acid, and arabino- and xylopyranose. Additionally, other natural diols from glucose but with cyclic structure, 1,4:3,6-dianhydrosorbitol and 2,4:3,5-di-Omethylene-D-glucitol were used. All diols used in this Thesis are accessible via chemical synthesis or are directly commercially available, and therefore they are a good option to introduce natural products in the polyurethane chemistry. The incorporation of these hydrophilic units can also render polyurethanes more susceptible to hydrolysis. The first block of this Thesis is dedicated to the synthesis, characterization and hydrodegradability of polyurethanes from alditols having all secondary hydroxyl groups protected as methyl ether, benzyl ether or acetal. Since total or partial splitting of the benzyl ethers or acetal groups after polymerization was feasible by hydrogenolysis or treatment by acids, respectively, these methods provided a suitable route to polyurethanes bearing pendant hydroxyl groups, which are highly hydrophilic and more susceptible to hydrolysis. The second block describes the synthesis of tartrate derived polyurethanes. In this case, tartrates were directly reacted with diisocyanates, and polyurethanes with different alkyl or benzyl ester side groups were obtained. The hydrodegradability of these polyurethanes was enhanced by incubating them in acid or basic buffers that hydrolysate the side alkyl esters, or by hydrogenolysis of the benzyl esters; in both cases polyurethanes bearing carboxylic side groups much more hydrolysable than their parent polyurethanes were prepared. The third block is dedicated to the study of segmented polyurethanes. Because there is a growing interest in introducing new chain extenders derived from renewable resources as an alternative to oil-based monomers, and also with the objective of enhancing the hydrodegradability of the resulting polyurethanes, two different carbohydrate derived monomers and their corresponding segmented polyurethanes were used as chain extenders in segmented polyurethanes with soft segment of polycaprolactone. Finally, the last block is dedicated to the phenomenon of stereocomplexation. The preparation method, the formation, evidenced by different spectroscopic techniques, and crystallizability of a polytartaramide stereocomplex is described. In addition, the exploration of stereocomplexation in other systems is presented.

In the rapidly changing scientific world, contributions of scientists and engineers are leading to major new solutions of significant medical problems. Biology and medicine are beginning to reduce the problems of disease to problems of molecular science, and are creating new opportunities for treating and curing disease. Such advances are coupled closely with advances in biomaterials and are leading to a variety of approaches for relieving suffering and prolonging life.Of particular interest is the central position that materials especially biopolymers, have taken in the development of novel treatments over the last 30 years. Biopolymers are used in many biomedical and pharmaceutical preparations, they play a central role in extracorporeal devices; from contact lenses to kidney dialyzers, are essential components of implants; from vascular grafts to cardiac pacemakers, and are the basis of controlled drug delivery technology; from hydrogels to micro and nanoparticles.In the last few years, novel synthetic techniques have been used to impart desirable chemical, physical, and biological properties to biomaterials. Materials have either been synthesized directly, so that desirable chain segments or functional groups are built into the material, or indirectly, by chemical modification of existing structures to add desirable segments or functional groups.Polymeric biomaterials can be produced by copolymerizations of conventional monomers to achieve nearly monodisperse polymers. It is possible to produce polymers containing specific hydrophilic or hydrophobic entities or biodegradable repeating units.One interesting strategy to develop multitalented biomaterials is the biosynthesis of polymers because these macromolecules have several advantages like: biological resorbability, hydrodegradability, absence of immunogenicity, and appropriately high molecular mass. The chemical derivatizability of these polymers allows the formation of several products with a wide range of properties.

Se han obtenido y caracterizado una serie de poli(a-n-alquil g-glutamato)s (PAAG-n) con n³12 y de poli(a-n-alquil b,L-aspartato)s (PAALA-n), mediante transesterificación con n-alcoholes y se ha estudiado la influencia que ejerce el grupo lateral sobre la estructura y propiedades de estos polímeros. Los polímeros se han caracterizado mediante FTIR, RMN de 1H y 13C y análisis elemental. La composición enantiomérica se ha determinado midiendo rotaciones específicas de los polímeros hidrolizados y/o mediante HPLC. Los pesos moleculares se han evaluado por viscosimetría y/o SEC/LS. La caracterización térmica se ha llevado a cabo mediante DSC, TGA y DMTA. El análisis estructural se ha realizado mediante las técnicas de DIR, 13C-CP/MAS-RMN, difracción de rayos X y de electrones. Los PAAG-n se han obtenido a partir del ácido poli(g-glutámico) (PGGA) de biosíntesis, de dos composiciones enantioméricas diferentes, uno enriquecido en el isómero D y otro de composición cercana a la racémica. El procedimiento general está basado en dos reacciones sucesivas (esterificación y transesterificación) y ha resultado especialmente útil para la preparación de ésteres de cadena larga difíciles de obtener por otros métodos. Las conversiones han sido completas no observándose racemización significativa. Los PAAG-n con n³14 forman una estructura bifásica supramolecular en láminas. Las cadenas principales adoptan una conformación helicoidal tipo hélice a y se alinean en láminas con las cadenas laterales interdigitadas formando una microfase entre las láminas. La distancia interlaminar es proporcional a n. Para estos polímeros se han encontrado las transiciones de fase A®B y B®C, a temperaturas T1 y T2. La transición A®B es reversible y tiene lugar a temperaturas entre 20 y 80 ºC. La transición B®C se ha observado para los PAAG-n con n³18 a temperaturas superiores a los 100 ºC. En la fase A las cadenas laterales están cristalizadas en una red hexagonal y dispuestas perpendicularmente a la cadena principal. En la fase B las cadenas laterales están desordenadas pero más o menos estiradas. La fase C presenta las cadenas laterales totalmente desordenadas. No se han observado efectos termocrómicos al calentar las muestras desde temperatura ambiente hasta temperaturas superiores a T2 cuando se observan bajo luz polarizada.Los PAALA-n y el PIBLA (poli(a-isobutil b,L-aspartato)) se han obtenido por transesterificación del PABLA (poli(a-bencil b,L-aspartato)). Resultó difícil conseguir conversiones completas debido a la existencia de reacciones de imidación. Los PAALA-n obtenidos mediante transesterificación mostraron temperaturas de descomposición similares a las de los PAALA-n obtenidos mediante polimerización. Las transiciones a y b (DMTA) se observan en los PAALA-n de cadenas cortas e intermedias a temperaturas más elevadas que las descritas para los PAAG-n con el mismo valor de n. La Ta disminuye a medida que aumenta n. La Tb aumentó ligeramente con n para los PAALA-n con n£ 8. La relajación g se observa para los PAALA-n con n³6.

El trabajo realizado en la Tesis tiene como objetivo general el estudio estructural de nylons fundamentalmente mediante la difracción de rayos-x y la microscopía electrónica. Los nylons estudiados se agrupan en tres familias:a) Nylons n con elevado número de metilenos: nylon 12 y nylon 16. El nylon 16 ha sido sintetizado y caracterizado por primera vez.b) Nylons 2/n: nylon 2/12 y nylon 2/16. Síntesis y caracterización de ambos productos siguiendo el método general de síntesis de nylons 2/n.c) Nylons (n, n+2): Nylons 4,6, 6,8, 8,10 3,5 y 5,7. Se ha realizado la síntesis de todos ellos excepto del nylon 4,6 el cuál se ha estudiado a partir de una muestra comercial (DSM, Holanda).Se han determinado las condiciones experimentales apropiadas para la preparación selectiva de las formas alfa y del nylon 12 en monocristales y películas. Los datos cristalográficos registrados para la fase y están en total acuerdo con la estructura descrita en la literatura. Los datos obtenidos para la fase alfa nos llevan a proponer paraesta forma una estructura triclínica basada en un modelo de empaquetamiento de láminas progresivo.Se ha llevado a cabo la síntesis del nylon 16 mediante policondensación térmica del ácido 16- aminohexadecanoico y efectuado su caracterización físico química. Presenta una Tf de 164 C. El análisis estructural se ha realizado sobre cristales crecidos en disolución y se determina una forma y.Se han sintetizado las copoliamidas 2/12 y 2/16 por condensación de ésteres activos y posterior postpolicondensación en estado sólido. Presentan Tf a 230 y 215 C respectivamente. El nylon 2/12 presenta dimorfismo mientras que en el 2/16 sólose ha observado la forma II; la forma II tiene una estructura pseudohexagonal con cadenas en conformación de hélices 6 y puentes de hidrógeno dirigidos en tres direcciones a 120 ; la forma I es minoritaria y sólo se observa bajo condiciones especiales.

El presente trabajo se ha centrado en la síntesis, caracterización y estudio estructural de poli(alfa-alquil-beta-L-aspartato)s con grupos laterales ramificados y cíclicos. Se han sintetizado los monómero: (S)-4-alcoxicarbonil-2-azetidinonas cuya síntesis se basa en la transesterificación de la beta-lactama del éster bencílico del ácido L-aspártico con el correspondiente alcohol. Se han establecido las condiciones óptimas de la polimerización aniónica por apertura del anillo de lactama, haciendo especial énfasis en el papel que ejerce la polaridad del solvente. En concreto el estudio se ha efectuado sobre la obtención del poli(alfa-isobutil-beta-L-aspartato). Se ha investigado la influencia de las condiciones de polimerización sobre la microestructura de los poli(alfa-isobutil-beta-D,L-aspartato)s obtenidos a partir de mezclas de monómeros enantiómeros. Se ha estudiado la conformación en disolución y la estructura cristalina en estado sólido, mediante difracción de rayos X, de los estereocopolímeros en relación con los polímeros ópticamente puros. Se han sintetizado una serie de poli(alfa-alquil-beta-L-aspartato)s nuevos con grupos alquilo ramificados y cíclicos, según las condiciones óptimas de polimerización establecidas para el derivado isobutílico. Se ha efectuado el análisis térmico de todos los polímeros y el estudio de la conformación en disolución por resonancia magnética nuclear y por dicroismo circular, detectando las transiciones de conformación. Mediante la técnica de difracción de rayos X se ha analizado el comportamiento estructural de la familia de poli(alfa-alquil-beta-L-aspartato)s sintetizada, estableciendo la comparación con los miembros de esta familia con grupos laterales lineales. Se ha llevado a cabo el análisis térmico dinamomecánico de los poli(alfa-alquil-beta-L-aspartato)s mediante la técnica de DMTA. Se han determinado las transiciones características para cada miembro y se ha establecido la correlación entre la relajación estructural de los polímeros y la constitución de la unidad repetitiva. Por último, se ha realizado el estudio de la degradación por ultrasonidos de los poli(-alquil--L-aspartato)s y de ciertos poliglutamatos, de forma que se han establecido las características generales de la degradación, así como la influencia que puede ejercer la estructura de la cadena principal y lateral sobre los resultados de dicha degradación.

Con la finalidad de obtener nuevos materiales de altas prestaciones a partir de recursos naturales, la memoria de la tesis "Síntesis y caracterización de poliamidas derivadas del ácido tartárico", presentada por Jordi Bou Serra, describe nuevos polímeros preparados a partir de derivados de carbohidratos. Se presentan cinco nuevas clases de poliamidas preparadas con ácido tartárico, un compuesto natural y biodegradable que se obtiene fácilmente tomando algunas frutas y vegetales como materia prima. Los comonómeros son diversos compuestos hidrocarbonatos (diaminas y diácidos) La tesis presenta la síntesis orgánica de todos estos materiales, así como su caracterización química (análisis elemental, espectro IR, espectro RMN de H y de 13C, rotación óptica específica, dicroísmo circular, puntos de fusión y tamaño molecular. Se han evaluado algunas propiedades de interés tecnológico, como su higroscopicidad, propiedades térmicas, capacidad de cristalizar y formación de piezas como fibras o películas. Cabe remarcar también que de algunas poliamidas (PxDMLT) se han determinado sus propiedades mecánicas y eléctricas, con resultados prometedores.Se detallan los resultados de todas estas técnicas de caracterización y propiedades, así como los procedimientos seguidos.También es interesante resaltar la poliamida PELYTA, que se ha preparado a partir del ácido tartárico y el aminoácido lisina, sustancias ambas de origen natural.De la tesis se han publicado cinco trabajos en publicaciones científicas de alto nivel y varias comunicaciones a congresos nacionales e internacionales. El director de la tesis ha sido el Dr Sebastián Muñoz Guerra., With the purpose of obtaining new materials of high performances from natural resources, the memory of the thesis "Synthesis and characterization of polyamides derived from the tartaric acid", presented by Jordi Bou Serra, describes new polymers prepared from carbohydrate derivatives. There appear five new classes of polyamides prepared with tartaric acid, a natural and biodegradable compound that is obtained easily taking some fruits and vegetables as a raw material. The comonomers are diverse hydrocarbon compounds (diamines and diacids)The thesis presents the organic synthesis of all these materials, as well as their chemical characterization (elementary analysis, IR spectra, H and 13C NMR spectra, optical specific rotation, circular dicroism, melting points and molecular size). There have been evaluated some properties of technological interest, like their hygroscopicity, thermal properties, capacity to crystallize and formation of pieces like fibers or films. It is important to note that of someone polyamides (PxDMLT) have determined their mechanical and electrical properties, with promising results.Details of the results of all these techniques of characterization and properties are presented, as well as the followed procedures.Also it is interesting to highlight the polyamide PELYTA, which has been prepared from tartaric acid and lysine (an amino acid) substances both of natural origin.Of the dissertation five works have been published in scientific publications of high level and several communications to national and international congresses.
