ÖZET Bu çalışmada, yeni türetilen ve karakteristik kök (ÇR) adi verilen bir indeks ile, çevre açismdan önemli bir kirletici grubu teşkil eden Poliklorlu Bifeniller'in (PCBs) sudaki çözünürlükleri, buhar basınçları ve oktanol-su oranı katsayıları gibi fizikokimyasal özellikleri arasındaki ilişki araştırılmıştır. Elde edilen sonuçlar, CR indeksinin, fizikokimyasal özelliklerin tahmin edilmesinde geçerli bir yapısal parametre olduğunu göstermiştir. Metod, yapısal çizit, moleküler topoloji ve yapı-etki ilişkilerine bagli olarak geliştirilmiştir. Bu metodla, 58 Poliklorlu Bifenil'in, çevredeki dağılımlarının kontrol edilmesinde önemli rol oynayan fizikokimyasal özellikleri hesaplanmıştır. Bu değerler, Mackay tarafından geliştirilen, maddenin bir fazdan kaçınım yatkınlığı ilkesine dayalı Fugasite Modeli 'nde veri olarak kullanılıp, PCB'lerin yüzde kaçının suda, toprakta, havada, canlıda ve sedimentte toplandığı ve bu kompartmanlardaki konsantrasyonları nispi olarak hesaplanmıştır. Sonuç olarak, Fugasite Modeli 'nin varsayımları dahilinde, Poliklorlu Bifeniller'in en fazla canlıda toplandığı ve klor sayısı arttıkça canlıdaki nispi konsantrasyonun da arttığı görülmüştür. via ABSTRACT Relationships between a newly developed index -which is based on topological characteristics of the molecules and called the Characteristic Root (CR) index- and the environmentally relevant physicochemical properties of the Poly chlorinated Biphenyls, namely, aqueous solubility, vapour pressure, Henry's Law Constants, and octanol-water partition coefficient are discussed. Correlations performed on the congeners clearly demonstrate that the CR index model accurately predicts the physicochemical properties of PCBs. High correlation coefficient (r = 0.998) was calculated for the linear one-parameter correlation between the CR index and molecular total surface area. So, the CR index was used in the prediction of properties dependent on total surface area. The quality and reliability of the correlations are shown to be high enough for environmental applications. Reported experimental physicochemical data of PCBs are tabulated. Recommended values are given for 58 of the 209 congeners. With the available data, it is possible to estimate what percentage of the chemical will be located in the soil, bottom sediments, water and air. The predicted and experimental physicochemical properties of PCBs were used to predict the environmental compartmentalization of these compounds in an evaluative environment using the fugacity approach. 144

Bu çalışmada, çeşitli azo boyaları ve aromatik amin türevlerinin S9 aktivasyonlu TA98 Salmonella typhimurium bakteri türü üzerindeki mutajenik aktivitesi kullanılarak kantitatif yapı - aktivite ilişkisi (KYAİ; (QSAR)) modeli geliştirilmiştir. Model geliştirme için gerekli tanımlayıcılar SPARTAN (v.10) ve DRAGON (v.7.0) yazılım programları ile elde edilmiştir. Tanımlayıcıların seçimi, QSARINS (v.2.2.3) yazılımında bulunan araçlar ile yapılmıştır. Tüm veri setinin % 80'i ve % 20'si sırası ile eğitim ve test setleri olacak şekilde farklı ayrımlar denenmiştir. 6 (CIC2, Chi_D/Dt, L/Bw, TDB09p, Mor28s and piPC08) tanımlayıcı ile nihai modeli oluşturulmuştur. Nihai modelin tahmin yeteneği, deneysel mutajenite verisi olmayan, halihazırda kullanılan 33 anyonik suda çözünür tekstil boyalarını (sekiz antrakinon ve 25 azo boyası) içeren harici bir veri seti kullanılarak test edilmiştr. Oluşturulan modelin uygulanabilirlik alanı (AD), hem aktivite hem de tanımlayıcıların oluşturduğu aralık gözönünde bulundurularak tanımlanmıştır. Önerilen KYAİ modelinin yapısal olarak dış setteki bileşiklerin %70 'ini kapsadığı görülmüştür. Halen kullanımda olan 22 adet antrakinon ve azo boyasının mutajenik aktiviteleri, önerilen QSAR modelinin uygulanabilirlik alanı içinde kalmıştır. Bu da mutajenik aktivitelerinin model tarafından iyi tahmin edildiklerini göstermiştir. En yüksek mutajenik aktivite gösteren, pamuk, elyaf boyama ve deri gölgemesi için kullanılmakta olan 6 antrakinon boyanın mutajenik aktivitelerinin sıralaması Asit Mavi 62>Asit Mavi 40>Asit Mavi 45>Asit Mavi 80>Asit Mavi 230>Asit Mavi 344 'tür. En düşük mutajenik aktiviteye sahip azo boya ise esas olarak pamuk, ipek, yün ve bunların karışımlı kumaş boyaması ve baskısı için kullanılmakta olan Direkt Turuncu 34' tür. Bu boya ayrıca deri ve kağıt gölgelemesi için de kullanılmaktadır. In the present study, the mutagenic activity of diverse structure of azo dyes and aromatic amine derivatives on TA98 Salmonella typhimurium bacterial strain with S9 activation was used to generate a quantitative structure – activity relationship (QSAR) model. The descriptors required for the model development were obtained by SPARTAN (v.10), DRAGON (v.7.0) software packages. The selection of descriptors was carried out by the tools implemented in QSARINS (v.2.2.3) software. Many division trials were performed on the dataset as training and test sets which comprise the 80% and 20% of the whole set, respectively. 6 descriptors (CIC2, Chi_D/Dt, L/Bw, TDB09p, Mor28s and piPC08) constitute the final model. The applicability domain (AD) of the generated QSAR model was defined by both the ranges of response and descriptors. The predictive ability of the final model was tested using an external dataset consisting of currently used 33 anionic water soluble textile dyes (eight anthraquinone and 25 azo dyes) with no experimental mutagenicity data. The proposed QSAR model had 70% structural coverage for the external set chemicals. The mutagenic activities of 22 current textile dyes belonging anthraquinone and azo dyes fell within the applicability domain of the proposed QSAR model which means that they were well predicted by the model. The order of 6 anthraquinone dyes which are mainly used for cotton, fiber dyeing and leather shading with the most mutagenic activity is: Acid Blue 62>Acid Blue 40>Acid Blue 45>Acid Blue 80>Acid Blue 230>Acid Blue 344. The least mutagenic azo dye is Direct Orange 34 which is mainly used for cotton, silk, wool and their blended fabric dyeing and printing, also can be used for leather and paper shading. 119

Sivrisinek Aedes aegypti Zika, dengue ateşi, sarı humma, chikungunya artritine neden olanvirüsleri ileten ana vektör olarak bilinmektedir. Vektörün kontrolü bu tür salgınların yayılmasınıönlemek için önemli bir stratejidir. Bununla birlikte, vektör kontrolü, sivrisineklerin böceköldürücülere karşı artan direnci nedeniyle tehdit altındadır. Öte yandan, bu böcek öldürücülerin yoğunkullanımının çevresel etkileri endişe vericidir. Bu çalışmada, bitki kaynaklı bileşiklerin larvisidalaktivitesi, kantitatif bir yapı-aktivite ilişkisi (QSAR) analizine tabi tutulmuştur. Ekonomik İşbirliğive Kalkınma Örgütü (OECD) tarafından belirlenen kriterlerine uygun geçerli bir QSAR modeli,QSARINS 2.2.2 yazılımı kullanılarak üretilmiş, üretilen model hem dahili hem de harici olarakdoğrulanmıştır. Modelin harici tahmin özelliği deneysel veri içermeyen kimyasallarla test edilmiş,yapısal kapsamının %95.3 olduğu görülmüştür. En toksik ve en az toksik bitki bazlı larvisidlerbelirlenmiştir. Piperidin türevleri Aedes aegypti larvası üzerinde oldukça etkili bulunmuştur. Ayrıca,Piper nigrum bitkisi bitki bazlı larvisid kaynağı olarak dikkat çekici bulunmuştur. İlave olarak,güvenli bir larvisid önermek için larvisidlerin sucul sistemlerde yaşayan hedef olmayanorganizmalara toksisitesi, daha önce Çevre Bilimleri Enstitüsü, Ekotoksikoloji ve KemometriLaboratuvarı grubu tarafından geliştirilen üç temsili sucul türe (alg, balık ve planaryan) yönelik akuttoksisite ve sitotoksisite modelleri kullanılarak değerlendirilmiş ve üç organizma çeşidi için en toksiklarvisidler belirlenmiştir The mosquito Aedes aegypti is known as the main vector that transmits the viruses cause dengue,yellow fever, chikungunya epidemic arthritis, and Zika. Control of the vector is an important strategyto avoid disease propagation. However, vector control is threatened by the increasing resistance ofmosquitoes to insecticides. On the other hand, environmental impacts of the intense use of theseinsecticides is of great concern. In the present study, the larvicidal activity of plant-derivedcompounds was subjected to a quantitative structure-activity relationship (QSAR) analysis. A validQSAR model which fulfill the criteria set by the Organization for Economic Co-operation andDevelopment (OECD) was generated using QSARINS 2.2.2 software. The generated QSAR modelwas validated both internally and externally. The external predictivity of model was tested withchemicals with no experimental larvicidal data and it has 95.3% structural coverage. The most toxicand the least toxic plant-based larvicides were determined. Piperidine derivatives were found highlyeffective on Aedes aegypti larvae. Also, the fruit Piper nigrum was highlighted as a plant-basedlarvicide source. Additionally, in order to propose a safe larvicide the toxicity of larvicides to nontarget organism living in aquatic systems was evaluated by using previously generated acute toxicityand cytotoxicity models towards three representative aquatic species (algae, fish, and planarian) byInstitute of Environmental Sciences, Ecotoxicology and Chemometrics Lab group and the most toxiclarvicides are detected for these aquatic species. 123

Su kaynaklarının pestisitler tarafından yaygın olarak kontaminasyonu, oldukça büyük birendişe teşkil etmektedir. Bu nedenle, pestisitlerin algler, omurgasızlar ve balıklar dahil olmak üzere hedefte olmayan organizmalar üzerindeki olumsuz etkileri araştırılmalıdır. Bu çalışmada, sonzamanlarda TÜBİTAK tarafından açıklanan ekotoksikolojik veri eksikliği bulunan kimyasallarlistesinden seçilen 8 pestisit (asetamiprid, asetoklor, boscalid, diklofop, difenamid, gibberellik asit, iyoksinil ve 2,4,5-T) için tatlı su alg toksisitesi deneyi gerçekleştirilmiştir. Seçilen pestisitlerin Chlorella vulgaris'in büyüme hızı üzerindeki etkisi, OECD'nin 201 numaralı testine göre ölçülmüştür. 96 saatlik alg toksisitesi sırasıyla asetoklor>iyoksinil>diklofop>2,4,5-T olarakbulunmuştur. Boscalid'in toksisite değeri, çözünürlük sınırları içinde belirlenememiştir.Asetamiprid, difenamid ve gibberellik asit, IC50 değerleri > 100 mg /L olarak bulunmuştur. Deneysonuçları, literatür QSAR modellerinden test edilen kimyasalların tahmini toksisite değerleri ilekarşılaştırılmıştır. Ayrıca, toksisite değerleri bir alg-alg (Chlorella vulgaris - Pseudokirchneriella subcapitata) toksisite ilişkisinden tahmin edilmiştir. Düşük toksisite ve hidrofobisite ilişkisi, bu kimyasalların toksisitesini temel toksisite dışındaki farklı etki modlarında uyguladıklarını gösteren düşük bir ilişki bulunmuştur. Tüm kimyasallar için global yarı ömür ve kalıcılık biyobirikim ve toksisite endeksleri hesaplanmıştır. Boscalid ve asetoklor PBT özelliği olan kimyasallar olarak bulunmuştur. Risk değerlendirmesinde hesaplanan düşük toksik etki konsantrasyonları (LOEC, NOEC ve IC20) kullanılmıştır ve ACR verileri kimyasal konsantrasyonlarının alg için belirlenmiş olan güvenli seviyesinde olup olmadığının kontrolü bakımından değerlendirilmiştir. Widespread contamination of water bodies by pesticides is indeed a matter of great concern.Therefore, their adverse effects on non-target organisms including algae, invertebrates, and fishshould be investigated. In this study, freshwater algal toxicity assay was carried out for 8 pesticides (acetamiprid, acetochlor, boscalid, diclofop, diphenamid, gibberellic acid, ioxynil, and 2,4,5-T) selected from the list of chemicals with no ecotoxicological data recently announced by TUBITAK. The effect of selected pesticides on the growth rate of Chlorella vulgaris was measured using the Organisation for Economic Cooperation and Development (OECD) test number 201. The order of96-h algal toxicity is acetochlor>ioxynil>diclofop>2,4,5-T. The toxicity value for boscalid couldnot be determined in its solubility limit. Acetamiprid, diphenamid and gibberellic acid revealed IC50 values > 100 mg/L.The experimental results are compared to the predicted toxicity values of tested chemicals from the literature QSAR models. Also, the toxicity values were predicted from an algae-algae (Chlorella vulgaris -Pseudokirchneriella subcapitata) toxicity relationship. Low correlation was found between the toxicity and hydrophobicity implying that these chemicals exert their toxicity in different mode of action other than baseline toxicity. Global half-life, persistence bioaccumulation and toxicity (PBT) indices were calculated for all chemicals. Boscalid and diclofop were found to be PBT chemicals. The calculated low-toxic-effect concentrations (LOEC,NOEC and IC20) were used in risk assessment and Acute to Chronic Ratio (ACR) were evaluated tocontrol if the ACR values are within the safe level of chemicals set for algae. 117

Kimyasalların sucul organizmalara yönelik zararlılık ve risk değerlendirmesi Kimyasalların Kaydı, Değerlendirilmesi, İzni ve Kısıtlanması hakkındaki Avrupa Birliği Yönetmeliği (REACH) doğrultusunda önem taşımaktadır. Hayvan refahı ve sürdürülebilirlik gibi etik konular ile sucul toksisite veri gereksinimi göz önünde bulundurulduğunda, Kantitatif Yapı-Toksisitesi İlişkileri (KYTİ) gibi bilgisayarla modelleme yöntemleri çevresel toksisitelerin tahmin edilmesinde büyük önem taşımaktadır. Bu çalışmada, geometri optimizasyon yönteminin seçimine yönelik aktivite-bağımsız akılcı bir yaklaşım iyileştirilmiş KYTİ modellemesi için ilk defa önerilmiştir. Temsili üç sucul türe (su yosunları, balık, planarya) yönelik farklı KYTİ ve türler arası modeller önerilen akılcı yaklaşım kullanılarak geliştirilmiştir. Gökkuşağı alabalığı karaciğer hücre dizini sitotoksisitesi ve Dugesia japonica toksisitesi için KYTİ modelleri ile Daphnia magna ve D. japonica için türler arası kantitatif toksisite ilişkisi ilk kez raporlanmıştır. Endişe verici kimyasallar ve yüksek hacimde üretilen kimyasallar gibi çevresel açıdan önemli kimyasallara yönelik geliştirilen ve dikkate değer tahmin kapsamı sağlayan KYTİ modelleri ile literatüre katkıda bulunulmuştur. Ayrıca, Türkiye Bilimsel ve Teknolojik Araştırma Kurumu (TÜBİTAK) tarafından açıklanan `Ekotoksikolojik Verisi Olmayan Kimyasallar Listesi` nde belirtilen kimyasalların büyük bir çoğunluğu için tahmin edilen sucul toksisite ve sitotoksisite verileri ilk kez olarak sağlanmıştır. Geliştirilen modeller, bilimsel ve düzenleyici bir çerçevede kimyasalların toksisite değerlendirmesi, taranması ve önceliklendirilmesinde potansiyel araçlar olarak ümit vericidir. Environmental hazard and risk assessment of chemicals are crucial for aquatic species within the direction of the European Regulation on the Registration, Evaluation, Authorization and Restriction of Chemicals (REACH). In consideration of ethical concerns, animal welfare and sustainability, as well as the need for aquatic toxicity data, in silico models, such as validated quantitative structure-toxicity relationships (QSTRs), are of great importance. In the present study, an activity-independent rational approach towards selecting an optimal geometry optimization method for improved QSTR modeling was proposed for the first time. Different QSTR and interspecies models towards three representative aquatic species (algae, fish, and planarian) were developed using the rational approach recommended. QSTR models on the prediction of cytotoxicity to rainbow trout liver cell line (RTL-W1), toxicity to Dugesia japonica, and an interspecies quantitative toxicity relationship between Daphnia magna and D. japonica were reported for the first time. The presented QSTR models have contributed to the literature by providing notable prediction coverage for environmentally significant chemicals, such as contaminants of emerging concern and high production volume chemicals. Furthermore, the first predicted aquatic toxicity and cytotoxicity data were provided for a great majority of the chemicals addressed in `The List of Chemicals with no Ecotoxicological Data` announced by the Scientific and Technological Research Council of Turkey (TÜBİTAK). The developed models are promising as potential tools in toxicity assessment, screening and prioritization of chemicals in a scientific and regulatory frame. 216

Bu çalışmada, beş kantitatif yapı-toksisite ilişkisi (QSTR) modeli geliştirilmiştir. Çeşitli kimyasalların Poeciliopsis lucida hepatokarsinom hücre dizisi (PLHC-1) kullanılarak üç farklı test - etoksiresorufin-O-deetilaz enzimi (EROD), neutral red (NR) ve 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolyum bromit (MTT) ile sitotoksisitesi ölçülmüş ve bu veriler kullanılarak üç QSTR modeli geliştirilmiştir. EROD, NR and MTT testleri sırasıyla enzimatik aktivite, lizozomal hasar ve mitokondriyal aktiviteyi göstermektedir. Diğer iki QSTR modeli ise akvaryum balığı (GFS), Carassius auratus'un pul dokusuna ve fathead minnow (FHM), Pimephales promelas'ın epitel hücre dizisine ait NR verisi kullanılarak geliştirilmiştir. Modellerdeki tanımlayıcılar QSARINS 2.2.1 yazılımında bulunan araçlarla seçilmiştir. Tüm QSTR modelleri Ekonomik İşbirliği ve Kalkınma Örgütü'nün (OECD) belirlediği ilkelerle uyumlu olacak şekilde geliştirilmiş ve dâhili ve harici olarak doğrulanmıştır. pEC50,EROD[PLHC-1] modeli bir tanımlayıcı, pEC50,NR[GFS] modeli iki ve pEC50,NR[PLHC-1], pEC50,NR[FHM] ve pEC50,MTT[PLHC-1] modellerinin her biri üçer tanımlayıcılıdır ve bu tanımlayıcılar DRAGON 6.0 yazılımı ile hesaplanan tanımlayıcılardır. Geliştirilen QSTR modellerinin tahmin performansı, farklı yapıda ve deneysel sitotoksisite verisi bulunmayan kimyasalların oluşturduğu birer harici set ile test edilmiştir. pEC50,NR[PLHC-1], pEC50,NR[GFS], pEC50,MTT[PLHC-1], pEC50,EROD[PLHC-1] ve pEC50,NR[FHM] modellerinin harici set kimyasallarını yapısal olarak kapsama yüzdeleri sırasıyla %98.6, %95.0, %98.3, %92.1 ve %89.1 olarak bulunmuştur. Fathead minnow modeli dışında, harici set kimyasalları için deneysel in vivo ve tahmin edilen in vitro değerleri arasında orta veya güçlü korelasyon gözlemlenmiştir. Bu çalışmada geliştirilen QSTR modelleri, çeşitli kimyasalların akut balık toksisitelerinin değerlendirilmesinde bir hızlı öntarama ve önceliklendirme sağlayabilir ve kapsamlı toksisite testlerine olan ihtiyacı azaltabilir. In the present study, five Quantitative Structure-Toxicity Relationship (QSTR) models were generated. Three QSTR models were developed for the cytotoxcity (pEC50) of diverse chemicals to Poeciliopsis lucida hepatocarcinoma cell line (PLHC-1) measured with three assays, namely ethoxyresorufin-O-deethylase enzyme (EROD), neutral red (NR) and 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) assays. EROD, NR and MTT assays reflects enzymatic activity, lysosomal and mitochondrial activity, respectively. The other two QSTR models were generated using the NR data for goldfish (GFS), Carassius auratus, scale tissue and fathead minnow (FHM), Pimephales promelas, epithelial cell line. Descriptors appearing in each model were selected via the tools implemented in QSARINS 2.2.1 software. All QSTR models were generated in line with the Organization of Economic Co-operation Development (OECD) principles and validated both internally and externally. pEC50,EROD[PLHC-1] model had one descriptor, pEC50,NR[GFS] had two and each of pEC50,NR[PLHC-1], pEC50,NR[FHM] and pEC50,MTT[PLHC-1] models had three descriptors from DRAGON. The external predictivity of the generated QSTR models were tested using structurally diverse chemicals with no experimental cytotoxicity data. Structural coverage of the pEC50,NR[PLHC-1], pEC50,NR[GFS], pEC50,MTT[PLHC-1], pEC50,EROD[PLHC-1] and pEC50,NR[FHM] models for the external set compounds were 98.6%, 95.0%, 98.3%, 92.1% and 89.1%, respectively. A moderate/strong correlation was observed between the experimental in vivo and predicted in vitro values for the external set chemicals, except fathead minnow. The generated QSTR models may provide an initial, rapid screening and prioritization of these diverse chemicals for the acute fish toxicity assessment and reduce the need for extensive in vivo toxicity testing. 245

Bu çalışmada, suda bulunan çeşitli organik bileşiklerin klor ihtiyaçları kantitatif yapı-özellik ilişkileri modeli kullanılarak incelenmiştir. Model geliştirmek için gerekli olan tanımlayıcılar SPARTAN (v.10), DRAGON (v.6.0), and ADMET (v.8.0) yazılımları kullanılarak elde edilmiştir. Tanımlayıcıların seçimi QSARINS (v.2.2.1) yazılımında bulunan araçlarla yapılmıştır. Veri setinin sırasıyla %80 i eğitim seti ve %20 si test seti olacak şekilde çok sayıda ayrımı denenmiştir. Oluşturulan modeller Ekonomik İş birliği ve Kalkınma Örgütü'nün belirlediği kriterlere uygun olur ve içsel ve dışsal olarak doğrulanmıştır. DRAGON (v.6.0) yazılımdan gelen 6 adet tanımlayıcı ve ADMET (8.0) yazılımdan gelen bir adet tanımlayıcı en son modeli oluşturmaktadır. Bu tanımlayıcılar geometri-topoloji-atom ağırlıkları birleşiminden oluşan tanımlayıcılar, ağırlıklı bütünsel yapıya bağlı değişebilen tanımlayıcılar, bilgi indeksi tanımlayıcıları, moleküler özelliklere bağlı tanımlayıcılar, basit yapısal tanımlayıcılar, 2 boyutlu öz ilinti ve 2 boyutlu atom çiftlerinden oluşmaktadır. Elde edilen son modelin tahmin kapasitesi klor tüketim verisi bulunmayan çok çeşitli ilaç ve kişisel bakım malzemesini içeren bir dışsal veri set kullanılarak test edilmiştir. Önerilen kantitatif yapı-özellik ilişkileri modeli harici kimyasalları yapısal olarak %91 ini kapsamaktadır. Oluşturulan modelin uygulanabilirlik alanı tanımlayıcı aralığı yaklaşımı ile de kesin olarak tanımlanmıştır. Oluşturulan modelin tahmin kapasitesi test edilen çok sayıda ilaç ve kişisel bakım malzemesi için güvenilir bulunmuştur. Test edilen ilaç ve kişisel bakım ürünleri içerisinde en yüksek klor ihtiyacıyla öne çıkan grup antibiyotiklerdir. The generated models were validated internally and externally in line with the Organization of Economic Co-operation and Development (OECD) principles. Six descriptors from DRAGON (v.6.0) and one descriptor from ADMET (v.8.0) constitute the final model. These descriptors stem from various blocks including GETAWAY, WHIM, information indices, molecular properties, simple constitutional, 2D autocorrelation and 2D atom pairs. The predictive ability of the final model was tested using an external data set consisting of various pharmaceuticals and personal care products (PPCPs) with no experimental chlorine demand data. The proposed QSPR model covers structurally 91% of the external chemicals. The AD of the generated model was also strictly defined by the range of descriptors' approach. The predictive ability of the generated model was found to be reliable for most of the tested PPCPs. Antibiotics are the highlighted pharmaceuticals among the tested PPCPs with the highest chlorine demand. 108

Bu çalışmada, 2,3,7,8-tetraklorodibenzo-p-dioxin/2,3,7,8-tetraklorodibenzofuran (TCDD/TCDF)-benzeri etkiler gösteren halojenlenmiş aromatik hidrokarbonların toksik etkileri kantitatif yapı-toksisite ilişkileri ile incelenmiştir. Bu doğrultuda, bu kimyasalların Aril Hidrokarbon Reseptörü'ne (AhR) bağlanma afinitelerinden yararlanılmıştır. Modellerde kullanılan tanımlayıcılar SPARTAN 10, DRAGON 6.0 ve ADMET 8.0 yazılımları kullanılarak oluşturulmuş ve tanımlayıcı seçimi QSARINS (v.2.2.1) yazılımı kullanılarak yapılmıştır. Veri setleri, eğitim ve test setlerine ayrılmıştır. Eğitim setleri, her iki modelde de veri setlerinin % 81'inden oluşmuştur. Oluşturulan modeller, Ekonomik İşbirliği ve Kalkınma Örgütü'nün belirlediği ilkelere uygun olacak şekilde dâhili ve harici olarak doğrulanmıştır. TCDD'ye göre normalize edilen veri setinin modeli, DRAGON 6.0 ve ADMET 8.0'dan elde edilen altı tanımlayıcılıdır. In the present study, the toxic potencies of xenobiotics such as halogenated aromatic hydrocarbons inducing 2,3,7,8-tetrachlorodibenzo-p-dioxin/2,3,7,8-tetrachlorodibenzofuran (TCDD/TCDF)-like effects were investigated by quantitative structure-toxicity relationships using their aryl hydrocarbon receptor (AhR) binding affinity data. The descriptor pool was created using SPARTAN 10, DRAGON 6.0 and ADMET 8.0 software and the descriptors were selected using QSARINS (v.2.2.1) software. Data sets were divided into training and test sets. The training sets were comprised of 81% of the complete data set for both models. The generated models for AhR of chemicals with TCDD/F-like effects were internally and externally validated in line with the Organization of Economic Co–operation and Development principles. TCDD-based model had six descriptors from DRAGON 6.0 and ADMET 8.0. TCDF-based model had seven descriptors from DRAGON 6.0 These descriptors were from various blocks including Weighted Holistic Invariant Molecular, Moriguchi Descriptors, and 2D and 3D descriptors blocks. The predictive ability of the generated models was tested for about 1000 diverse group of chemicals from polychlorinated/brominated biphenyls, dioxins/furans, ethers, polyaromatic hydrocarbons with fused heterocyclic rings (i.e. phenoxathiins, thianthrenes and dibenzothiophenes), and polyaromatic hydrocarbons (i.e. halogenated napthalenes and phenanthrenes) with no AhR data. For the external set chemicals, the structural coverage of the generated models was 95.55% and 89.37% for TCDD/F-like chemicals, respectively. 206

Registration, Evaluation, Authorization and Restriction of CHemicals (REACH) regülasyonu risk belirleme amaçları için alg toksisite verileri gerektirmektedir. Kantitatif Yapı-Toksisite İlişkileri (KYTİ) eksik verileri tamamlamada kabul edilir araçlardır. Bu nedenle, toksisite verisi olmayan kimyasalların alg üzerindeki toksik etkilerinin deneysel ve bilgisayarla modelleme yoluyla çalışılması çok değerli bilgi sağlayacaktır. Bu çalışma ile elde edilecek bilgi sucul ekosistemlerin korunmasına bilimsel bir taban oluşturacaktır. Bu çalışmada, nitro, kloro, metoksi ve metil eklenmiş fenol ve anilin türevlerinin Chlorella vulgaris'e olan etkileri 96 saatlik alg toksisite deneyleriyle belirlenmiştir. Toksisite modellemesi için bu verinin daha önceden laboratuvarımızdan raporlanmış veri ile birleştirilmesi tek kaynaktan yüksek kalitede alg toksisite verisini mümkün kılmıştır. Sonrasında, akut toksisite ve düşük toksik etki konsantrasyonlarının tahmini için modeller geliştirilmiş ve OECD ilkelerine dayanılarak doğrulanmıştır. Release of emerging pollutants such as pesticides, phthalates, and substituted phenols and anilines is detrimental threat for the aquatic environment. Registration, Evaluation, Authorization and Restriction of CHemicals (REACH) regulation requires algal toxicity data for regulatory risk assessment purposes. Quantitative Structure–Toxicity Relationships (QSTRs) are well accepted tools for data gap-filling. Therefore, studying the toxic effects of chemicals on algae via experimental and in silico methods would provide invaluable information for the chemicals with no toxicity data; and the knowledge gained through this study forms a scientific basis towards the protection of aquatic ecosystems. In the present study, the 96-h algal toxicity tests were performed with nitro-, chloro-, methoxy-, and methyl- substituted phenols and anilines to Chlorella vulgaris. Merging these data with the previously reported toxicity data of our laboratory enabled a high quality single source algal toxicity data for toxicity modeling. Consequently, models for the prediction of acute toxicity and low-toxic-effect concentrations were developed and verified based on the principles of OECD. Interspecies models were also developed using algae-algae and algae ciliate toxicity data. Developed models displayed decent predictivity and have a high potential to assess the toxicity of untested phenols and anilines on C. vulgaris within the applicability domain of models. 171

Bakteriyel selülozların çevre, tıp, gıda ve elektronik gibi çeşitli alanlarda geniş ölçekte kullanımı söz konusudur. Bu çalışmada, iki karbon kaynağının bakteriyel selüloz üretimi için uygunluğu test edilmiştir. Nişasta bakımından zengin bir materyal olan kasava unu, nişasta içeriğinin glikoza hidrolizinden sonra bir karbon kaynağı olarak kullanılmak üzere test edilmiştir. Nişastanın mikroalg biyokütlesinin değerli bileşenlerinden biri olmasından dolayı, Chlorella vulgaris'ten sağlanan nişastanın hidrolizi ile elde edilen glikoz da bakteriyel selüloz üretiminde kullanılmak üzere test edilmiştir. Algin nişasta içeriği kasava ununa göre oldukça düşük olduğundan, nişasta içeriğini arttırmak amacıyla C. vulgaris üzerinde birkaç stres koşulu denenmiştir. Bildiğimiz kadarıyla bu çalışma, kasava unu ve alg nişastası hidrolizatlarının Komagataeibacter hansenii tarafından bakteriyel selüloz üretiminde karbon kaynağı olarak kullanıldığı ilk çalışmadır. Glikoz (kontrol olarak), kasava unu ve alg nişastasından elde edilen bakteriyel selüloz (kuru ağırlık olarak) sırası ile 1.202±0.005 g.L-1, 1.138±0.004 g.L-1 ve 1.104±0.002 g.L-1 şeklindedir. Karakterizasyon çalışmalarının sonuçları, kasava unu ve alg nişastasının kullanımı ile üretilen bakteriyel selülozların, glikozdan elde edilen bakteriyel selüloz ile benzer morfolojik ve kimyasal özelliklere sahip olduğunu göstermiştir. Bacterial celluloses have a wide range of applications in many areas such as environmental, medical, food and electronics. In the present study, the suitability of two carbon sources were tested for BC production. Cassava flour, as a starch-rich material, was tested to be used as a carbon source after the hydrolysis of its starch content to glucose. Since starch is among the valuable components of microalgal biomass, glucose obtained by the hydrolysis of starch from Chlorella vulgaris was also tested to be used in BC production. While algae has a lower starch content than cassava flour, several stress conditions were attempted to increase the starch content of C. vulgaris. To the best of our knowledge, the present study is the first report on the use of hydrolysates of cassava flour and algal starch as carbon sources in the production of BC by Komagataeibacter hansenii. BC yields on dry weight basis were 1.202±0.005 g.L-1, 1.138±0.004 g.L-1, and 1.104±0.002 g.L-1 from glucose (as control), cassava flour, and algal starch, respectively. The results of the characterization studies have revealed that the morphological and chemical characteristics of the BCs produced from cassava flour and algal starch were similar to the BC produced from glucose. 87

Ekonomik ve çevresel faktörler kimyasalların toksisitelerinin belirlenmesi konusundaki çalışmaların büyük bir çoğunluğunu hesaplama yöntemlerine yöneltmiştir. Pestisitler, hedef olmayan canlılar üzerindeki zararlı etkileri nedeniyle, kimyasallar içinde önemli bir yer tutarlar. Bu çalışmada, pestisit de içeren 91 organik kimyasalın tatlı su algi Chlorella vulgaris'e ve 34 pestisitin Oncorhynchus mykiss'e olan toksisiteleri Counter Propagation Neural Network (CPNN) ve Çoklu Doğrusal Regresyon (MLR) ile modellendi. Analizler Dragon 5.4, Spartan 06 ve Codessa 2.2 programları kullanılarak hesaplanan yaklaşık 1500 tanımlayıcı ile yapıldı. Ek olarak, daha önceki QSPR/QSTR çalışmalarda önemli bir tanımlayıcı olduğu kanıtlanan Karakteristik Kök İndisi (CRI)'ni de kullandık. Tanımlayıcı seçimi Heuristic Yöntem'le yapıldı. Economical and environmental considerations for assessing toxicity of chemicals have led to a considerable amount of studies on the computational techniques. Pesticides allocate a significant part in these chemicals, mainly for their toxic effects on nontarget organisms. In the present study, the toxicities of 91 organic compounds including pesticides to freshwater algae, Chlorella vulgaris; and the toxicities of a set of 34 pesticides to Oncorhynchus mykiss were modeled employing Counter Propagation Neural Network (CPNN) and Multiple Linear Regression (MLR). The analyses were performed with about 1500 descriptors calculated using Dragon 5.4, Spartan 06, and Codessa 2.2 software. Additionally, we used the Characteristic Root Index (CRI) which was proved to be a significant descriptor in previous QSPR/QSTR studies. Descriptor selection was made by Heuristic Method. Kohonen network was used for splitting the data set into training and test sets. Linear and nonlinear 3, 4 and 5-descriptor models were compared according to their statistics such as squared correlation coefficient and Root Mean Squared Error (RMSE). All models were validated externally by using test sets. BLTD48 from Dragon, electrophilicity from Spartan, and the CRI appeared to be significant for the developed QSTR models of Chlorella vulgaris. Oncorhynchus mykiss model underscores the Dragon descriptors. The statistical quality of the models for Chlorella vulgaris is compared to those of the previously published models using the same experimental data and found to be superior to those models. Oncorhynchus mykiss models are compared to literature models in terms of chemical classes, mechanism of action, and statistical tools and fits. Linear and nonlinear methods were found to be comparable for both species. 96

Polibromlu ve poliklorlu difenil eterler (PBDEs/PCDEs) gibi Polihalojenli bileşikler çevredeki dayanıklıkları, yüksek hidrofob özellikleri ve canlılardaki biyobirikimlerinden dolayı önemli bir kimyasal gruplardır. n-oktanol/hava (log Koa), n-oktanol/su (log Kow) dağılım katsayısı gibi fizikokimyasal ve toksisite gibi özellikleri, bu bileşiklerin çevresel akıbeti ve davranışlarını daha iyi anlayabilmek için önem taşımaktadır.Bu çalışmada, PBDE için log Koa ve log RBA, PBDE ve PCDE için log Kow modelleri Heuristic yöntem ve Çoklu Doğrusal Regresyon (MLR) kullanılarak Kantitatif yapı-aktivite/Kantitatif yapı-özellik ilişkisi (QSAR/QSPR) ile oluşturuldu. Polyhalogenated compounds (PHCs) such as polybrominated and polychlorinated diphenyl ethers (PBDEs/PCDEs) are an important chemical group because of their environmental persistence, high hydrophobicity, and bioaccumulation in humans. Their physico-chemical properties such as n-octanol/air partition coefficient (log Koa) and n-octanol/water partition coefficient (log Kow) and toxicities are of fundamental importance to gain a better understanding of the environmental fate and behavior of these compounds.In this study, several Quantitative Structure-Activity Relationship/Quantitative Structure-Property Relationship (QSAR/QSPR) models were developed on log Koa of PBDEs, log Kow of PBDEs and PCDEs, and the aryl hydrocarbon receptor relative binding affinity (log RBA) of PBDEs by employing Heuristic Method (HM) and Multiple Linear Regression (MLR). Descriptors used were from DRAGON 5.4, SPARTAN 06, and CODESSA 2.2 software and the Characteristic Root Index (CRI) program. All the best models were internally validated for their performance using the leave-one-out procedure and scrambling of the responses. External validation was provided by splitting the data sets into training and test sets either using random division in terms of property modeled or Kohonen network considering the size of the data sets.Of the models developed log Koa and log Kow models were validated externally by using test sets. log RBA model could not be validated externally because of a lack of RBA data. EHOMO and Eaq from SPARTAN, and the CRI appeared to be significant descriptors for the developed log Kow models of PBDEs/PCDEs. The CRI also appeared to be an important parameter in modeling log Koa of PBDEs. The statistical quality of all the models for polyhalogenated diphenyl ethers is compared to those of the previously published models using the same experimental data and found to be superior to those models. All the QSAR/QSPR models were developed taking into account the OECD principles for validation, for regulatory purposes, of QSAR. This implied internal and external validations, the analysis of the applicability domain (AD) and, when possible, a mechanistic interpretation of the models. 147

Bu çalısmada, boya banyosu ve üç azo boya çözeltisinin, Reaktif Siyah 5, ReaktifSarı 37 ve Reaktif Turuncu 69, tuzlu su algi D. tertiolecta ile renk giderimi arastırıldı vealgin ek enerji ve karbon kaynagı olarak glikoz ve gliserol varlıgında boyayı kullanmakapasitesi tartısıldı. Boyaların renginin giderilmesi, her boya çözeltisinin maximum dalgaboyundaki absorbanslarının azalması takip edilerek belirlendi. In this study, the decolorization of Dye Bath (DB), and the three hydrolyzed dyes,namely Reactive Black 5 (RB-5), Reactive Orange 69 (RO-69) and Reactive Yellow 37(RY-37) by marine microalgae Dunaliella tertiolecta, in a batch culture was investigatedand the ability to utilize the azo dyes in the presence of co-substrates namely glucose andglycerol as an additional carbon and energy sources was discussed. Decolorization of dyeswas determined by monitoring the decrease in the intensity of absorption at the maximumwavelength (lmax) of each of the dye solution. The color change and pH values of theculture media were monitored during the growth period of algae exposed to dyes in thepresence and absence of glucose and glycerol.The individual and combined effects of DB, RB-5, RO-69 and RY-37 in thepresence and absence of glucose and glycerol on the growth of Dunaliella tertiolecta werealso examined. Algal growth was followed through optical density (OD750) measurementsof the culture medium. Furthermore, algal growth was examined with differentconcentrations of glucose and glycerol.D. tertiolecta was found to be quite effective in the decolorization of all dyes. Atlow dilutions (30% and 60% (v/v)) of all DB, RB-5, RO-69 and all dilutions of RY-37 canbe removed without co-substrate by the algae. However, addition of both concentrations ofglucose and glycerol has some beneficial effects such as increasing the decolorizationefficiency or decreasing decolorization period of all dyestuffs except RY-37. Atconcentrations beyond the IC25 values of dyes, decolorization achievement is decreasingdue to the different structure and complexity of dyes. Considering the average 8-d IC25 andSC20 values expressed as % dilution (v/v), the order of toxicity was found as DB>RB-5>RO-69>RY-37. Glucose and glycerol increased the stimulatory effect of all the testedchemicals on the growth of algae at a low concentration range, whereas the inhibitoryeffect at a high concentration range decreased. Glucose and glycerol did not affect theinitial pH values and pH-changing trend of the medium containing dyestuffs, significantly. 181

EHOMO ve µ ilişiklik yüzünden aynı modelde yer almamıştır. BCF'in tahminedilmesinde yalnızca CRI'ın kullanıldığı modelin korelasyon katsayısı r = 0.850 iken, 48h-EC50 modelinin korelasyon katsayısı r = 0.926 olmuş ve modelde tanımlayıcı olarak CRIve ELUMO yer almıştır. BCF için elde edilen ve yalnızca CRI'ın kullanıldığı modelde(model 6), veri sayısı 18 ile sınırlı olduğu için modelin güvenilirliği önce klasik testlerlekontrol edilmiş, daha sonra literatürdeki yine CRI'ya bağlı başka bir model (model 7) ilekarşılaştırılmıştır. Jackknife testlerinin sonuçları geliştirilen beş modelin de istatistikselolarak güvenilir olduğunu göstermiştir. Similarly, QSPR models have also been established for the biologicalproperties including 50% effective inhibition concentration (48h-EC50) to algae andbioconcentration factor (BCF) in fish for substituted benzenes having endocrine disruptingproperties. The parameters used to develop these QSPR models were topology basedCharacteristic Root Index (CRI) and semi-empirical molecular descriptors, namely -energies of the highest occupied and the lowest unoccupied molecular orbital (EHOMO andELUMO), and dipole moment (µ).The best fit equation found by ?forward multiple linear regression? showed that thetopology based CRI was the most important parameter for the modeling of S, KOW andBCF. For KOW a two-parameter equation including the CRI and EHOMO was obtained with acorrelation coefficient of r = 0.992, whereas a three-parameter equation including the CRI,ELUMO and µ was obtained for both solubility and Henry?s Law Constant with a correlationcoefficient of r = 0.986 and r = 0.933, respectively. EHOMO and µ didn?t appear in the samemodel because of the collinearity. The CRI and ELUMO descriptors were used in regressionsof 48h-EC50 with a correlation coefficient of r = 0.926 while only the CRI had appeared asa descriptor for the prediction of BCF with a correlation coefficient of r = 0.850. Since thenumber of BCF data is limited to 18 for the CRI-based QSPR model (model 6), reliabilityof this model was first checked by classical tests, and then it was compared with the otherCRI-based model (model 7) reported in the literature. The results of modified jackknifetests indicated that the five models were statistically robust. Mean deviation of calculatedvalues from experimental data amounted to 0.27, 0.17, 0.28, 0.19 and 0.29 log units for theS, KOW, H, 48h-EC50, and BCF, respectively. The developed models have been used topredict the S, KOW, H, 48h-EC50, and BCF of those compounds where there are noexperimental measurements. 109

Bu çalışmada, çevre açısından önemli üç metalin (bakır, kurşun ve selenyum), tek hücreli yeşil yosun olan Dunaliella tertiolecta'da. biyoakümülasyon değerlerinin karşılaştırılması yapılmıştır. Bu metallerin log fazındaki Dunaliella tertiolecta hücrelerinde 24 saatteki biyoakümülasyon değerleri, bakır için 0.25-1.5 mg/L, kurşun için 0.5-1.0 mg/L ve selenyum için 0.5-1.67 mg/L konsantrasyon aralığında ve iki farklı pH (8 ve 6) değerinde ölçülmüştür. Metal çiftlerinin biyoakümülasyon değerleri pH 8' de ölçülmüş ve metal çiftlerinin biyoakümülasyona olan muhtemel etkilerine de bakılmıştır. Biyoakümülasyon değerleri kullanılarak bu üç metalin biyokonsantrasyon faktörleri (BCF) hesaplanmıştır. Yosunda biriken metaller asitle muamele edilerek alınmıştır. Bakır ile kurşunun yosundaki miktarları atomik absorpsiyon spektrofotometresi ile, selenyumun (Se4+) yosundaki miktarı ise spektroflorimetre ile ölçülmüştür. Elde edilen sonuçlar, biyoakümülasyonun hem metale, hem de konsantrasyona bağlı olarak değiştiğini göstermiştir. Çalışılan konsantrasyon aralığında üç metalin biyoakümülasyon ve BCF değerlerinin sırası Pb > Cu > Se şeklinde bulunmuştur. Düşük pH'da, bakırın biyoakümülasyon ve BCF değerlerinin düştüğü fakat kurşun ve selenyumun biyoakümülasyon ve BCF değerlerinin değişmediği görülmüştür. Üç metalin değişik konsantrasyonlardaki kombinasyonları ile yapılan metal çifti çalışmaları, (1) bakır ve kurşun arasında antagonistik bir etki olduğunu (2) bakır ve selenyum arasında etkileşme olmadığım ve (3) kurşun ve selenyum arasında da etkileşme olmadığını göstermiştir. In this work, a comparative study of bioaccumulation of environmentally significant two essential metals (copper and selenium) and one nonesssential metal (lead) has been performed with the unicellular green alga Dunaliella tertiolecta. Individual bioaccumulation of these metals in exponentially growing cells of Dunaliella tertiolecta was investigated at 0.25-1.5 mg/L copper, 0.5-1.0 mg/L lead and 0.5-1.67 mg/L selenium concentrations for 24 hour exposure time at pH values of 8 and 6. Bioaccumulation of metal pairs was measured at pH value of 8 and the possible effects of mixed metals on bioaccumulation were also investigated. Bioconcentration factors (BCF) of these metals were calculated from bioaccumulation. Total metal bioaccumulated in algae was extracted by acid treatment. Total copper and lead concentrations were detected by atomic absorption spectrophotometer, whereas total selenium concentration (Se4+) was measured spectrofluorometrically. It was found that bioaccumulation varied both as a function of metal and concentration. Within the studied concentration range, the order of both bioaccumulation and BCF values of three metals was found as Pb > Cu > Se. When pH decreased, bioaccumulation and BCF values significantly decreased for Cu, but bioaccumulation and BCF values did not change for Pb and Se. Metal pair studies in which any combination of the three metals with each other indicated (1) antagonism between copper and lead (2) no interaction between copper and selenium and (3) no interaction between lead and selenium. 62
