The Atrato watershed is a rainforest that supports exceptional wildlife species and is considered one of the most biodiversity-rich areas on the planet, currently threatened by massive gold mining. Aimed to protect this natural resource, the Constitutional Court of Colombia declared the river subject to rights. The objective of this study was to quantify trace elements in sediments and fish from Atrato watershed, assessing their environmental and human health risk. Forty-two trace elements were quantified using ICP-MS. Thirty-one elements increased their concentration downstream the river. Concentration Factors (CF) suggest sediments were moderately polluted by Cr, Cu, Cd, and strongly polluted by As. Most stations had Cr (98%) and Ni (78%) concentrations greater than the Probable Effect Concentration (PEC) criteria. Together, toxic elements generate a Pollution Load Index (PLI) and a Potential Ecological Risk Index (RI) that categorized 54% of the sediments as polluted, and 90% as moderate polluted, respectively. Hemiancistrus wilsoni, a low trophic guild fish species, had the greater average levels for Ni, Cu, As and Cd, among other elements. Rubidium and Cs showed a positive correlation with fish trophic level, suggesting these two metals biomagnify in the food chain. The Hazard Quotient (HQ) for As was greater than 1 for several species, indicating a potential risk to human health. Collectively, data suggest gold mining carried out in this biodiversity hotspot releases toxic elements that have abrogated sediment quality in Atrato River, and their incorporation in the trophic chain constitutes a large threat on environmental and human health due to fish consumption. Mercury (Hg) is a heavy metal known as one of the most toxic elements on the planet. The importance of Hg on living organisms resides on its biomagnification ability. Artisanal gold extraction activities release substantial amounts of this metal, polluting the ecosystems. To assess the impact of gold mining in Las Orquideas National Natural Park (Colombia), total Hg (T-Hg) levels were evaluated from 37 bird and 8 small rodent species collected at two sites within the boundaries of the Natural Park (Abriaqui and Frontino municipalities) that have experienced some gold-extraction history. The mean concentration of T-Hg in bird feathers from both sites was 0.84 ± 0.05 μg/g fw. Differences between species were found according to diet. Total Hg levels were greater on insectivorous (1.00 ± 0.08 μg/g fw), followed by nectarivorous (0.73 ± 0.07 μg/g fw) and frugivorus (0.57 ± 0.09 μg/g fw) species. These Hg levels were greater than those found in feathers from a control sample belonging to the species Penelope perspicax (0.53 ± 0.03 μg/g fw), a frugivorous species living at the Otun Quimbaya Fauna and Flora Sanctuary, a forest without known gold mining. Mercury concentrations in the livers of small rodents were greater in specimens from Frontino (0.15 ± 0.01 μg/g fw) than those from Abriaqui (0.11 ± 0.01 μg/g fw), but levels were not different between species. These results indicate that Hg in birds depends mainly on their diet, but geographical location may affect Hg concentration in rodents. Moreover, Hg sources in natural parks of Colombia may not rely solely on gold mining, atmospheric deposition, among others factors, could be influencing its accumulation in biota.

Perchlorate is an inorganic ion widespread in the environment, generated as a natural and anthropogenic pollutant, with known endocrine disruption properties in the thyroid gland. Nonetheless, there are few reports of its ecotoxicological impact on wildlife. The aim of this study was to evaluate the adverse effects of KClO 4 exposure on different cell lines, HEK, N2a, and 3T3, as well as in ecological models such as Vibrio fischeri, Pseudokirchneriella subcapitata, Daphnia magna, and Eisenia fetida. Perchlorate exhibited similar toxicity against tested cell lines, with LC 50 values of 19, 15, and 19 mM for HEK, N2a, and 3T3, respectively; whereas in V. fischeri, the toxicity, examined as bioluminescence reduction, was considerably lower (EC 50 = 715 mM). The survival of the freshwater algae P. subcapitata was significatively impaired by perchlorate (LC 50 = 72 mM), and its effect on the lethality in the crustacean D. magna was prominent (LC 50 = 5 mM). For the earthworm E. fetida, the LC 50 was 56 mM in soil. In this organism, perchlorate induced avoidance behavior, weight loss, and decreased egg production and hatchling, as well as morphological and histopathological effects, such as malformations, dwarfism, and necrosis. In conclusion, perchlorate toxicity varies according to the species, although E. fetida is a sensitive model to generate information regarding the toxicological impact of KClO 4 on biota.

Donax denticulatus is a key mollusk for the ecology of sandy beaches, serving as a controller of organic matter, microorganisms, and as bioindicator of heavy metals pollution. The goal of this study was to characterize some ecological aspects of D. denticulatus and its relationship with the content of heavy metals in their tissue, in three beaches of the Caribbean coast of Colombia. The results showed the study populations were different in terms of morphological characteristics and density (Berrugas-Sucre < Cartagena-Bolívar < Riohacha-Guajira), but not in sexual proportion; although density was clearly related to beach occupancy by tourists. Analysis of metals revealed tissue concentrations varied depending on the location (Greater means: Hg = 0.018 ± 0.004 in Riohacha; Pb = 0.110 ± 0.060 in Berrugas and Cd = 0.040 ± 0.010 µg/g in Cartagena). No relationships were found between morphometric variables and heavy metals content. Principal components analysis highlighted Riohacha for presenting differences respecting to Bocagrande and Berrugas in terms of physicochemical water parameters such as pH, temperature, salinity, dissolved oxygen and conductivity. Results suggest tourism rather than environmental pollution could be a sensitive factor for biota survival in Caribbean beaches.

The Magdalena River, the main river of Colombia, receives contaminated effluents from different anthropogenic activities along its path. However, the Magdalena River is used as drinking water source for approximately 30 million inhabitants, as well as a major source of fish for human consumption. Only a few studies have been conducted to evaluate the environmental and toxicological quality of the Magdalena River. To evaluate sediment toxicity, wild-type and GFP transgenic Caenorhabditis elegans were exposed to methanolic extracts, and effects on lethality, locomotion, growth, and gene expression were determined based on fluorescence spectroscopy. These biological and biochemical parameters were correlated with measured pollutant concentrations (PAHs and trace elements), identifying patterns of toxicity along the course of the river. Effects on lethality, growth, and locomotion were observed in areas influenced by industrial, gold mining, and petrochemical activities. Changes in gene expression were evident for cyp-34A9, especially in the sampling site located near an oil refinery, and at the seaport, in Barranquilla City. Body bend movements were moderately correlated with Cr and As concentrations. The expression of mtl-1, mtl-2, hsp-6, and hsp-70 were significantly associated with Pb/U, Pb, Sr, and As/Sr/Pb/U, respectively. Interestingly, toxicity of methanolic as well as aqueous extracts were more prone to be dependent on Cd, Zn, and Th. In general, ecological risk assessment showed sediments display low environmental impact in terms of evaluated metals and PAHs. Different types of waste disposal on the Magdalena River, as a result of mining, domestic, agricultural, and industrial activities, incorporate toxic pollutants in sediments, which are capable of generating a toxic response in C. elegans.

Mercury (Hg) is a global contaminant posing severe risks to human health worldwide. The aim of this study was to assess the levels of total Hg (T-Hg) in human hair and fish in the Caqueta River, at the Colombian Amazon, as well as to determine fish consumption-based risks for T-Hg ingestion. T-Hg levels were measured using a direct mercury analyzer. The overall mean T-Hg level in hair for humans in the Caqueta River sample (n = 200) was 17.29 ± 0.61 μg/g (1.2 to 47.0 μg/g). Ninety-four percent of the individuals had hair T-Hg concentrations greater than the WHO threshold level (5 μg/g), and 79 % displayed levels higher than 10 μg/g. Average Hg concentrations in fish varied between 0.10-0.15 μg/g and 0.10-1.60 μg/g, for noncarnivorous and carnivorous species, respectively. Based on the maximum allowable fish consumption rate for adults, most carnivorous species should be avoided in the diet, as their target hazard quotient ranged from 2.96 up to 31.05, representing a risk for Hg-related health problems. In the light of existing evidence for elevated Hg levels in the indigenous population of the Colombian Amazon, carnivorous fish should be restricted as part of the diet, and breastfeeding should be reduced to protect children health. Most importantly, gold mining activities directly on rivers demand immediate attention from the national government to avoid extensive damage on the environment and human health.

The Magdalena River is the most important river in Colombia, supplying over 70% of the population of fish and drinking water, and it also is the main river transportation way of the country. It receives effluents from multiple sources along its course such as contaminant agricultural and industrial discharges. To evaluate the toxicity profile of Magdalena River sediments through endpoints such as survival, locomotion, and growth, wild type strains of Caenorhabditis elegans were exposed to aqueous extracts of the sediments. To identify changes in gene expression, GFP transgenic strains were used as reporter genes. Physiological and biochemical data were correlated with metal concentration in the sediments, identifying patterns of toxicity along the course of the river. Levels of some metals such as Cd, Cu, and Ni were above TEC and PEC limits. Effects in survival, growth, and locomotion were observed in most of the samples, and changes in gene expression were evident in the genes mtl-2, sod-4, and gst-1 using fluorescence expression. Cadmium and lead were the metals which were primarily associated with sediment toxicity, and the sampling sites with the highest increased expression of stress response genes were Barrancabermeja and Girardot. Extraction, transport and utilization of coal spread out coal dust. Nowadays, Colombia is an important producer of this mineral in South America, being the Santa Marta area one of the largest coal exporting ports in the country. The aim of this work was to assess the pollutants levels and toxicity of shoreline sediments from this place. 16 PAHs and 46 elements were measured in nine locations during dry and rainy seasons. HepG2 cells were exposed to 1% sediment extracts and mRNA expression evaluated for selected genes. PAHs levels were greater during the rainy season. The highest ∑PAHs (89.9 ng g(-1)) appeared at a site located around 300 m far from the coast line at close proximity to the area where coal is loaded into cargo vessels for international shipments, being naphthalene the most abundant PAH. At Santa Marta Bay port, ∑PAHs were 62.8 ng g(-1) and 72.8 ng g(-1) for dry and rainy seasons, respectively, with greatest levels for fluoranthene. Based on sediment standards, most stations have poor condition regarding Cr, but moderate contamination on Cu, Pb and Zn. Sediments from the port and coal transport sites, the most polluted by PAHs and metals, induced CYP1A1 and NQO1 during the dry season. Gold mining is responsible for most Hg pollution in developing countries. The aims of this study were to assess the levels of total Hg (T-Hg) in human hair, fish, water, macrophyte, and sediment samples in the gold mining district of San Martin de Loba, Colombia, as well as to determine fish consumption-based risks for T-Hg ingestion. T-Hg levels were measured by electrothermal atomization and atomic absorption spectroscopy. The overall mean T-Hg level in hair for humans in the mining district of San Martin de Loba was 2.12 μg/g, whereas for the reference site, Chimichagua, Cesar, it was 0.58 μg/g. Mean T-Hg levels were not different when considered within localities belonging to the mining district but differed when the comparison included Chimichagua. T-Hg levels in examined locations were weakly but significantly associated with age and height, as well as with fish consumption, except in San Martin de Loba. High T-Hg concentrations in fish were detected in Pseudoplatystoma magdaleniatum, Caquetaia kraussii, Ageneiosus pardalis, Cyrtocharax magdalenae, and Triportheus magdalenae, whereas the lowest appeared in Prochilodus magdalenae and Hemiancistrus wilsoni. In terms of Hg exposure due to fish consumption, only these last two species offer some guarantee of low risk for Hg-related health problems. Water, floating macrophytes, and sediments from effluents near mining sites also had high Hg values. In mines of San Martin de Loba and Hatillo de Loba, for instance, the geoaccumulation index (I(geo)) for sediments reached values greater than 6, indicating extreme pollution. In short, these data support the presence of a high Hg-polluted environment in this mining district, with direct risk for deleterious effects on the health of the mining communities.

Eggs from egrets (Egretta thula) were collected from Cartagena Bay and Totumo Marsh, two sites at the north of Colombia with different pollution background, and measured their morphometric parameters as well as total mercury (T-Hg) and calcium levels in eggshell. Statistically significant differences were observed for egg weight and size between the two sampling locations. T-Hg and calcium concentrations in eggshell were greater in eggs from Cartagena Bay, the industrial site, compared to Totumo Marsh, a non-industrial location. The opposite was observed for eggshell thickness (3.6% less in the bay). Pearson correlation analysis showed eggshell T-Hg negatively correlated with eggshell weight in eggs from the marsh (R=-0.795, P<0.006), but not from the bay (R=0.387, P=0.269), probably suggesting greater susceptibility to Hg in birds from the non-polluted site. In short, results suggest eggs from E. thula at Cartagena Bay have greater T-Hg concentrations and less eggshell thickness than those from Totumo Marsh., (Copyright © 2013 Elsevier Ltd. Perfluorinated compounds are widespread pollutants of toxicological importance that have been detected in environmental matrices. However, little is known on their distribution in South America. In this study, distribution of perfluorooctanesulfonate (PFOS), perfluorooctanoic acid (PFOA), perfluorohexanesulfonate (PFHxS), and perfluorooctanesulfonamide (PFOSA) was determined in the bile of mullet, Mugil incilis, and in tissues of pelicans (Pelecanus occidentalis) collected from North Colombia. Analysis was performed by HPLC mass spectrometry after ion-pair extraction. PFOS was found in all bile samples and PFOA and PFHxS were detected at lower frequency. Average concentrations of PFOS, PFOA, and PFHxS in bile of fish from Cartagena Bay, an industrialized site, and Totumo marsh, a reference site, were 3673, 370, 489 and 713, 47.4, 1.27 ng/mL, respectively. PFOS concentrations in pelican organs decreased in the order of spleen>liver>lung>kidney>brain>heart>muscle. These results suggest, for the first time, that perfluorinated compounds are also found in wildlife from Latin American countries.

Abstract: Coal is a mixture of chemicals with the capacity of promoting biochemical changes that may lead to DNA damage. In this study, the comet assay in peripheral blood cells, and the micronucleus test in blood smears were used to evaluate potential genotoxic effects derived from exposure to coal mining activities on wild populations of Mus musculus and Iguana iguana. Four locations from Colombia were evaluated: La Loma and La Jagua de Ibirico, two municipalities located near coal mining fields at the Department of Cesar; and Valledupar and Arjona, cities used as reference sites, both localized at least 100 and 200km far from the mines, respectively. Compared to Valledupar and Arjona, animals collected in close proximity to coal mining areas showed highest percentages of DNA damage for both species, evidencing that living around coal mining fields may result in an increase of DNA lesions in blood cells of rodents and reptiles. The results for micronucleus test were conflicting. Mice from Arjona had greater number of cells with micronucleus than those from the other studied locations, probably as a result of infection found by blood parasites. In summary, it was demonstrated that animals living around coal mining areas have a greater chance of having DNA damage, as measured by the comet assay, than those from sites far from the coal dust source. Mugil incilis (lisa) is an important commercial fish species in many countries, living along the coasts of the western Atlantic Ocean. It has been used as a model organism for environmental monitoring and ecotoxicological investigations. Nevertheless, available genomic and transcriptomic information for this organism is extremely deficient. The aim of this study was to characterize M. incilis hepatic transcriptome using Illumina paired-end sequencing. A total of 32,082,124 RNA-Seq read pairs were generated utilizing the HiSeq platform and subsequently cleaned and assembled into 93,912 contigs (N50 = 2,019 bp). The analysis of species distribution revealed that M. incilis contigs had the highest number of hits to Stegastes partitus (13.4%). Using a sequence similarity search against the public databases GO and KEGG, a total of 7,301 and 16,967 contigs were annotated, respectively. KEGG database showed genes related to environmental information, metabolism and organismal system pathways were highly annotated. Complete or partial coding DNA sequences for several candidate genes associated with stress responses/detoxification of xenobiotics, as well as housekeeping genes, were employed to design primers that were successfully tested and validated by RT-qPCR. This study presents the first transcriptome resources for Mugil incilis and provides basic information for the development of genomic tools, such as the identification of RNA markers, useful to analyze environmental impacts on this fish Caribbean species.
