The aim of this study was to conduct a screening of potential therapeutic compounds found in the Atractylodes macrocephala rhizoma essential oil (AO) and explore its mechanism of action in the treatment of ulcerative colitis (UC). An inflammation cell model was employed in conjunction with phospho-antibody array technology to explore potential therapeutic compounds of AO and their anti-inflammatory and antioxidant effects. Furthermore, we assessed their efficacy and mechanisms of action in treating dextran sulfate sodium (DSS)-induced colitis in mice. Via the screening process, we identified atractylone (ATR) as the primary active compound in AO. It has been demonstrated that ATR can both decrease the levels of tumor necrosis factor (TNF)-α and reactive oxygen species (ROS) and increase the expression of adhesion proteins such as claudin, ZO-1, and occludin in vitro. Moreover, ATR has been shown to improve UC symptoms in vivo. Via a non-targeted metabolomics analysis of colon tissue, we identified 57 distinct metabolites that responded to ATR treatment. Subsequent analysis of the metabolic pathways revealed that the action of ATR was primarily focused on the amino acid metabolism pathway. In summary, ATR may alleviate the symptoms of UC by regulating multiple signaling pathways. Essential oils (EOs) have stability problems, including volatility, oxidation, photosensitivity, heat sensitivity, humidity sensitivity, pH sensitivity, and ion sensitivity. A drug delivery system is an effective way to stabilize EOs, especially due to the protective effect of polymeric drug carriers. Polysaccharides are frequently employed as drug carrier materials because they are highly safe, come in a variety of forms, and have plentiful sources. Interestingly, the EO drug delivery system is based on the biomimetic concept since it corresponds to the structure of plant tissue. In this paper, we associate the biomimetic plant-like structures of the EO drug delivery system with the natural forms of EO in plant tissues, and summarize the characteristics of polysaccharide-based drug carriers for EO protection. Thus, we highlight the research progress on polysaccharides and their modified materials, including gum arabic, starch, cellulose, chitosan, sodium alginate, pectin, and pullulan, and their use as biomimetic drug carriers for EO preparations due to their abilities and potential for EO protection. In this study, star anise (Illicium verum) essential oils (SAEOs) were extracted by hydrodistillation (HD), ethanol solvent extraction (ESE), supercritical CO2 (SCD) and subcritical extraction (SE) via electronic nose (E-nose), gas chromatography-mass spectrometry (GC-MS), and GC-ion mobility spectrometry (GC-IMS). GC-MS and GC-IMS were used to identify the volatile compounds, and GC-MS was also used to determine their concentrations. Principal component analysis (PCA) and linear discriminant analysis (LDA) were used to visualise volatile compounds and differentiate samples. The results showed that anethole and limonene were the main volatile compounds in SAEOs extracted using the four methods and their components were similar, albeit in different proportions. In addition, the fingerprints of their volatile components were established via E-nose and GC-IMS analyses. In general, GC-MS, GC-IMS, and E-nose combined with PCA and LDA analysis could accurately distinguish SAEOs extracted using different extraction methods, and GC-IMS was identified as the most suitable method because of its accuracy and rapidity. Essential oils (EOs) have good natural antioxidant and antimicrobial properties; however, their volatility, intense aroma, poor aqueous solubility, and chemical instability limit their applications in the food industry. The encapsulation of EOs in β-cyclodextrins (β-CDs) is a widely accepted strategy for enhancing EO applications. The complexation of cinnamon essential oil (CEO) with five types of β-CDs, containing different substituent groups (β-CD with primary hydroxyl, Mal-β-CD with maltosyl, CM-β-CD with carboxymethyl, HP-β-CD with hydroxypropyl, and DM-β-CD with methyl), inclusion process behaviors, volatile components, and antioxidant and antibacterial activities of the solid complexes were studied. The CEOs complexed with Mal-β-CD, CM-β-CD, and β-CD were less soluble than those complexed with DM-β-CD and HP-β-CD. Molecular docking confirmed the insertion of the cinnamaldehyde benzene ring into various β-CD cavities via hydrophobic interactions and hydrogen bonds. GC-MS analysis revealed that HP-β-CD had the greatest adaptability to cinnamaldehyde. The CEO encapsulated in β-, Mal-β-, and CM-β-CD showed lower solubility but better control-release characteristics than those encapsulated in DM- and HP-β-CD, thereby increasing their antioxidant and antibacterial activities. This study demonstrated that β-, Mal-β-, and CM-β-CD were suitable alternatives for the encapsulation of CEO to preserve its antioxidant and antibacterial activities for long-time use. Compared with traditional liquid–liquid embedding method and solid–liquid embedding method of inclusion complexes of β‐cyclodextrin (β‐CD) inclusion of essential oil to form stable properties, the gas–liquid embedding method was applied to encapsulate vaporized illicium verum essential oil (IvEO), with β‐CD as wall materials so that core and wall materials molecules are in active state during complexing process. At optimal conditions with a mass ratio of 1:10, temperature of 80°C, time of 1 h, the β‐CD‐IvEO inclusion complexes (β‐CD‐IvEO‐ICs) had an encapsulation efficiency (EE) of 84.55 ± 2.31%. Fourier transform infrared spectroscopy (FTIR) revealed the encapsulation of IvEO into inclusion complexes, differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA) demonstrated the enhanced thermal stability of IvEO after embedding. Furthermore, the reducing power and 2‐phenyl‐4,4,5,5‐tetramethylimidazoline‐1‐oxyl‐3‐oxide (PTIO)‐scavenging capacity displayed certain capacity of antioxidation in a short time but stronger antioxidative activities as reaction time was extended. The diameter of growth zone (DGZ) indicated stronger antibacterial activity of β‐CD‐IvEO‐ICs against Escherichia coli, Bacillus subtilis, Staphylococcus epidermidis, and Staphylococcus aureus. Essential oils (EOs), which are plant-oriented anti-biofilm agents, are extensively encapsulated by cyclodextrins to overcome their aqueous solubility and chemical instability, and achieve slow release during long-term storage. However, the biological activities of EOs decreased after initial encapsulation in CDs. In this study, modified- β -cyclodextrins (β -CDs) were screened as wall materials to maintained the initial anti-biofilm effect of pure CEO. The inhibitory and bactericidal activities of CEO encapsulated in five types of β -CDs with different substituents (primary hydroxyl, maltosyl, hydroxypropyl, methyl, and carboxymethyl) against Staphylococcus aureus biofilm were evaluated. Crystal violet assay and 3D-View observations suggested that CEO and its inclusion complexes (CEO-ICs) inhibited Staphylococcus aureus biofilm formation through the inhibition of colonising spreading, exopolysaccharide synthesis, and cell surface properties. Molecular docking revealed the causes of the decrease in the anti-biofilm effect after encapsulation, and quantitative structure-activity relationship assays provided MIC and MBIC prediction equation for modified- β -cyclodextrins inclusion complexes. Maltosyl- β -CD was screened as the best wall material to retained the anti-biofilm activities as pure cinnamon essential oil in initial stage, and its inclusion complexes can effectively inhibited biofilm formation in milk. This study provides a theoretical guidance for the selection β -CDs to encapsulate CEO as plant-oriented anti-biofilm agents to inhibit bacterial biofilm formation. Atractylodes macrocephala Koidz. has been used as an invigorating spleen drug for eliminating dampness and phlegm in China. According to recent researches, different processing methods may affect the drug efficacy, so we collected A. macrocephala from the Zhejiang Province, produced with different processing methods, crude A. macrocephala (CA) and bran-processed A. macrocephala (BA), then analyzed its essential oils (EOs) by GC/MS. The results showed 34 components representing 98.44% of the total EOs of CA were identified, and 46 components representing 98.02% of the total EOs of BA were identified. Atractylone is the main component in A. macrocephala. Compared with CA, BA has 46 detected compounds, 28 of which were identical, and 6 undetected compounds. Pharmacodynamic results revealed that the EOs of CA and atractylone exhibited more effective anticancer activity in HepG2, MCG803, and HCT-116 cells than the EOs of BA; while the EOs of BA exhibited simple antiviral effect on viruses H3N2, both the EOs and atractylone show anti-inflammatory activity by inhibiting the lipopolysaccharide (LPS)-induced nitric oxide (NO) production in ANA-1 cells. Insomnia occurs frequently in modern society, and its common symptoms include difficulty in falling asleep and decreased sleep quality and time, memory, and attention. With the advantages of having few side-effects and reduced drug-dependence, a compound traditional Chinese medicine (TCM) prescription called Huaxiang Anshen Decoction (HAD) has been widely used in clinical practice in China mainly for primary insomnia treatment. Although the effects of volatile oils from TCM herbs have been increasingly reported, volatile oils in HAD are conventionally neglected because of its preparation process and clinical usage. Therefore, exploring the anti-insomnia effects of volatile oils from HAD is of great importance. The sedative and hypnotic effects of the conventional aqueous extracts, the volatile oils from HAD, and their combinations were investigated. The main components in HAD volatile oils (HAD-Oils), were analyzed through gas chromatography-mass spectrometry (GC-MS). The HAD volatile oil inclusion complex (HAD-OIC) was prepared with β-cyclodextrin, and characterized. P -chlorophenylalanine (PCPA) was used to induce insomnia mice model and the test groups of HAD aqueous extract (HAD-AE), HAD-OIC and their combination (AE-OIC). An open field test was used in evaluating the mice's activities, and the levels of 5-hydroxytryptamine (5-HT) in mice sera, glutamate (Glu) in the hypothalamus, and γ-aminobutyric acid (γ-GABA) and dopamine (DA) in the brain tissues were assayed by enzyme-linked immunosorbent assay (ELISA). A total 74 components in HAD-Oil were determined by GC/MS, and cyperenone (20.46%) and α-cyperone (10.39%) had the highest relative content. The characterization results of the physical phase showed that volatile oils were successfully encapsulated by β-cyclodextrin and HAD-OIC was produced. The average encapsulation rates of cyperenone and α-cyperone were 79.93% and 71.96%, respectively. The results of pharmacology study showed that all the test groups increased the body weight and decreased voluntary activity when compared with the model group (P < 0.05). The HAD-AE, HAD-OIC, and AE-OIC groups increased the levels of 5-HT in the sera and DA and Glu/γ-GABA in the brains, and AE-OIC groups showed better performance than the other test groups. HAD-Oil exerts sedative and hypnotic effects, which are increased when it is used with HAD-AEs.
