The large use of fish meal/fish oil in carnivorous fish feeds is the main concern regarding environmental sustainability of aquaculture. Here, we evaluated the effects of an innovative diet, designed to be (1) environmentally sustainable by lowering the marine protein content while being (2) cost effective by using sustainable alternative raw materials with acceptable cost and produced on an industrial scale, on growth performance, gut microbiota composition, health and welfare of European sea bass (Dicentrarchus labrax), a key species of the Mediterranean marine aquaculture, reared in sea cages. Results show that the specific growth rate of fish fed the low marine protein diet was significantly lower than those fed conventional diet (0.67% vs 0.69%). Fatty acid profile of fillets from fish fed a low marine protein diet presented significant lower n-6 and higher n-3 content when compared to conventional ones. Then, a significant increase in the abundance of Vibrio and reduction of Photobacterium were found in the gut of fish fed with the low marine protein diet but effects on sea bass health needs further investigation. Finally, no major health and welfare alterations for fish fed the low marine protein diet were observed, combined with a potential slight benefit related to humoral immunity. Overall, these results suggest that despite the low marine protein diet moderately affects growth performance, it nevertheless may enhance environmental and economic sustainability of the sea bass aquaculture. Within the thermal tolerance range of fish, metabolism is known to escalate with warming. Rapid thermic changes also trigger a series of physiological responses, including activation of the stress axis, producing cortisol. Fish have adapted to their environment by producing a low level of plasmatic cortisol when unstressed (basal), so that thriving in their natural temperature should not impact their basal cortisol levels. Yet, surprisingly, little is known on how temperature affects cortisol within and between fish species. Here, we conducted a phylogenetic meta‐analysis to (1) test whether temperature can explain the differences in basal cortisol between species and (2) evaluate the role of temperature on differences in cortisol levels between individuals of a same species. To do this, we retrieved basal plasma cortisol data from 126 studies, investigating 33 marine and freshwater fish species, and correlated it to water temperature. Intra‐species variability in basal plasma cortisol levels was further investigated in two species: the European sea bass Dicentrarchus labrax and the Nile tilapia Oreochromis niloticus. Factors such as life stage, sex and weight were also considered in the analyses. Overall, our phylogenetic analysis revealed a clear positive correlation between basal cortisol level and the temperature at which the fish live. The role of temperature has also been confirmed within D. labrax, while it failed to be significant in O. niloticus. In this paper, the influence of habitat, life stage, sex and weight on basal plasma cortisol levels is also discussed. Since some abiotic parameters were not included in the analysis, our study is a call to encourage scientists to systematically report other key factors such as dissolved oxygen or salinity to fully depict the temperature‐cortisol relationship in fishes. Bolder individuals have greater access to food sources and reproductive partners but are also at increased risk of predation. Boldness is believed to be consistent across time and contexts, but few studies have investigated the stability of this trait across variable environments, such as varying stress loads or long periods of time. Moreover, the underlying molecular components of boldness are poorly studied. Here, we report that boldness of 1154 European sea bass, evaluated using group risk-taking tests, is consistent over seven months and for individuals subjected to multiple environments, including a chronically stressful environment. Differences in risk-taking behaviour were further supported by differences observed in the responses to a novel environment test: shy individuals displayed more group dispersion, more thigmotaxic behaviour and lower activity levels. Transcriptomic analyses performed on extreme phenotypes revealed that bold individuals display greater expression for genes involved in social and exploration behaviours, and memory in the pituitary, and genes involved in immunity and responses to stimuli in the head kidney. This study demonstrates that personality traits come with an underpinning molecular signature, especially in organs involved in the endocrine and immune systems. As such, our results help to depict state–behaviour feedback mechanisms, previously proposed as key in shaping animal personality. [ABSTRACT FROM AUTHOR]

Background: Telemetry applied to aquatic organisms has recently developed greatly. Physiological sensors have been increasingly used as tools for fish welfare monitoring. However, for the technology to be used as a reliable welfare indicator, it is important that the tagging procedure does not disrupt fish physiology, behaviour and performance. In this communication, we share our medium-term data on stress physiological profile and growth performance after surgical tag implantation in two important marine fish species for European aquaculture, the sea bream (Sparus aurata) and the European sea bass (Dicentrarchus labrax). Results: Blood samples after surgical tag implantation (46 days for the sea bream and 95 days for the sea bass) revealed no differences between tagged and untagged fish in cortisol, glucose and lactate levels, suggesting that the tag implantation does not induce prolonged stress in these species. Moreover, the specific growth rates were similar in the tagged and untagged fish of both species. Conclusion: Surgical tag implantation does not have medium-term consequences for the stress physiology and growth performance of these two marine fish species in a controlled environment. These observations support the use of accelerometer tags as valuable tools for welfare monitoring in aquaculture conditions. This study also shows that tagged fish can be sampled during experiments and considered a representative portion of the population, as they display growth and physiological parameters comparable to those of untagged fish. Coping styles consist of a coherent set of individual physiological and behavioral differences in stress responses that are consistent across time and context. Such consistent inter-individual differences in behavior have already been shown in European sea bass (Dicentrarchus labrax), but the associated mechanisms are still poorly understood. Here, we combine physiological measurements with individual behavioral responses in order to characterize coping styles in fish. Fish were tagged and placed in a tank for group risk-taking tests (GRT) at 8 months of age to evaluate boldness using the proxy latency of leaving a sheltered area towards an open area. A subsample of these fish were individually challenged 16 months later using an open field test (OFT), in which the boldness was assessed after being placed in a shelter within an open arena. Latency to exit the shelter, time spent in the shelter, and distance travelled were recorded for this purpose. The blood and brain were then collected to evaluate plasma cortisol concentration and neurotransmitter levels (dopamine, norepinephrine, serotonin, and related metabolites), as well as brain transcription of key genes involved in stress axis regulation (gr1, gr2 , mr, crf), neurogenesis (neurod1, neurod2, pcna), and neuronal development (egr1). Fish acting bolder in the GRT were not necessarily those acting bolder in the OFT, highlighting the relatively low consistency across different types of tests performed with a 16-months interval. There was, however, a significant correlation between stress markers and boldness. Indeed, mRNA levels of mr, crf, gr2, egr1 , and neurod2 , as well as norepinephrine levels were higher in shy than bold fish, whereas brain serotonergic activity was lower in shy fish. Overall, our study highlights the fact that boldness was not consistent over time when testing context differed (group vs. alone). Stress modifies energy allocation in fishes by redirecting energy from growth and reproduction to coping mechanisms. However, these adjustments become inappropriate when the challenge consists of sustained or repeated stressors, with the animal entering a maladaptive state. Capacities to cope with additional threats are then altered and compromise survival. The characterization of the responses to chronic stress in fishes helps better understanding the physiological limits in an aquaculture or ecological context. Here, we investigate the coping capacities of European sea bass to multiple and diverse stressors applied over a 3-weeks period. Multiple behavioural (group dispersion and swimming activity) and physiological responses (blood cortisol, osmoregulatory mechanisms, stress-related gene expression, etc.) were evaluated in resting fish or in fish exposed to additional challenges. Resilience to the chronic stress protocol was evaluated 4 months after the end of the chronic stress. Chronically stressed individuals showed reduced growth, lower cortisol response, increased chloride and sodium concentration in the plasma and modified gill gene expression translating osmoregulatory dysfunctions. Chronic stress had no significant effect on plasmatic calcium, lysozyme concentration and osmotic pressure. Increased thigmotaxic behaviour was observed in a new environment behavioural test. Four months after the chronic stress, no significant difference was observed in growth performances and in plasma parameters. Altogether, gills and more generally osmoregulatory functions were found to be the most sensitive to the chronic stress, while only limited changes in growth, activity of the HPI axis, immunity and swimming behaviour were observed when assessed individually. Three groups of European sea bass (Dicentrarchus labrax) were fed for seven months, with either a conventional diet or two different organic diets, which contain organic vegetables and a natural antioxidant compound. The two organic diets differed themselves in terms of raw proteins, fish oil, and lipid contents. Sea bass welfare condition was assessed in relation to these three diets, using 16 different indicators. These were: swimming activity (recovery test, muscle activity), haematological and serological stress indicators (haematocrit, haemoglobin, red-blood-cell count, cortisol, glucose, lactate), aspecific immunity parameter (lysozyme), indicators of exposure to organic contaminants (7-ethoxyresorufin-O-deethylase and glutathione-S-transferase), and growth parameters (weight gain, specific growth rate, feed conversion ratio, protein efficiency ratio, and hepato-somatic index). Most of these parameters individually did not give consistent responses, but their integration can provide an accurate evaluation of the fish welfare conditions among the three diet experimental groups. The multiparametric approach outlined a comprehensive picture of sea bass physiological state. The principal component analysis and the multi-criteria-decision-analysis were found to be useful tools for an integrated fish welfare assessment, highlighting that the best welfare condition was achieved in the experimental group fed with the protein-rich organic diet. In aquaculture, fish are exposed to unavoidable stressors that can be detrimental for their health and welfare. However, welfare in farmed fish can be difficult to assess, and, so far, no standardized test has been universally accepted as a welfare indicator. This work contributes to the establishment of behavioural welfare indicators in a marine teleost in response to different water quality acute stressors. Groups of ten fish were exposed to high Total Ammonia Nitrogen concentration (High TAN, 18 mg.L-1), Hyperoxia (200 % O 2 saturation), Hypoxia (20 % O 2 saturation), or control water quality (100% O 2 saturation and TAN < 2.5 mg.L-1) over 1 hour. Fish were then transferred in a novel environment for a group behaviour test under the same water quality conditions over 2 hours. Videos were recorded to assess thigmotaxis, activity and group cohesion. After this challenge, plasma cortisol concentration was measured in a subsample, while individual behavioural response was measured in the other fish using novel tank diving test. Prior to this study, the novel tank diving test was validated as a behavioural challenge indicative of anxiety state, by using nicotine as anxiolytic drug. Overall, all stress conditions induced a decrease in activity and thigmotaxis and changes in group cohesion while only fish exposed to Hypoxia and High TAN conditions displayed elevated plasma cortisol concentrations. In post -stress condition, activity was still affected but normal behaviour was recovered within the 25 minutes of the test duration. Our work suggests that the activity, thigmotaxis and group cohesion are good behavioural indicators of exposure to degraded water quality, and could be used as standardized measures to assess fish welfare.
