Background: Extreme terrestrial, analogue environments are widely used models to study the limits of life and to infer habitability of extraterrestrial settings. In contrast to Earth's ecosystems, potential extraterrestrial biotopes are usually characterized by a lack of oxygen., Methods: In the MASE project (Mars Analogues for Space Exploration), we selected representative anoxic analogue environments (permafrost, salt-mine, acidic lake and river, sulfur springs) for the comprehensive analysis of their microbial communities. We assessed the microbiome profile of intact cells by propidium monoazide-based amplicon and shotgun metagenome sequencing, supplemented with an extensive cultivation effort., Results: The information retrieved from microbiome analyses on the intact microbial community thriving in the MASE sites, together with the isolation of 31 model microorganisms and successful binning of 15 high-quality genomes allowed us to observe principle pathways, which pinpoint specific microbial functions in the MASE sites compared to moderate environments. The microorganisms were characterized by an impressive machinery to withstand physical and chemical pressures. All levels of our analyses revealed the strong and omnipresent dependency of the microbial communities on complex organic matter. Moreover, we identified an extremotolerant cosmopolitan group of 34 poly-extremophiles thriving in all sites., Conclusions: Our results reveal the presence of a core microbiome and microbial taxonomic similarities between saline and acidic anoxic environments. Our work further emphasizes the importance of the environmental, terrestrial parameters for the functionality of a microbial community, but also reveals a high proportion of living microorganisms in extreme environments with a high adaptation potential within habitability borders. As part of the Biology and Mars Experiment (BIOMEX; ILSRA 2009-0834), samples of the lichen Circinaria gyrosa were placed on the exposure platform EXPOSE-R2, on the International Space Station (ISS) and exposed to space and to a Mars-simulated environment for 18 months (2014-2016) to study: (1) resistance to space and Mars-like conditions and (2) biomarkers for use in future space missions (Exo-Mars). When the experiment returned (June 2016), initial analysis showed rapid recovery of photosystem II activity in the samples exposed exclusively to space vacuum and a Mars-like atmosphere. Significantly reduced recovery levels were observed in Sun-exposed samples, and electron and fluorescence microscopy (transmission electron microscope and field emission scanning electron microscope) data indicated that this was attributable to the combined effects of space radiation and space vacuum, as unirradiated samples exhibited less marked morphological changes compared with Sun-exposed samples. Polymerase chain reaction analyses confirmed that there was DNA damage in lichen exposed to harsh space and Mars-like environmental conditions, with ultraviolet radiation combined with space vacuum causing the most damage. These findings contribute to the characterization of space- and Mars-resistant organisms that are relevant to Mars habitability.

To ensure that scientific investments in space exploration are not compromised by terrestrial contamination of celestial bodies, special care needs to be taken to preserve planetary conditions for future astrobiological exploration. Significant effort has been made and is being taken to address planetary protection in the context of inner Solar System exploration. In particular for missions to Mars, detailed internationally accepted guidelines have been established. For missions to the icy moons in the outer Solar System, Europa and Enceladus, the planetary protection requirements are so far based on a probabilistic approach and a conservative estimate of poorly known parameters. One objective of the European Commission-funded project, Planetary Protection of Outer Solar System, was to assess the existing planetary protection approach, to identify inherent knowledge gaps, and to recommend scientific investigations necessary to update the requirements for missions to the icy moons.

The search for life beyond Earth involves investigation into the responses of model organisms to the deleterious effects of space. In the frame of the BIOlogy and Mars Experiment, as part of the European Space Agency (ESA) space mission EXPOSE-R2 in low Earth orbit (LEO), dried colonies of the Antarctic cryptoendolithic black fungus Cryomyces antarcticus CCFEE 515 were grown on martian and lunar analog regolith pellets, and exposed for 16 months to LEO space and simulated Mars-like conditions on the International Space Station. The results demonstrate that C. antarcticus was able to tolerate the combined stress of different extraterrestrial substrates, space, and simulated Mars-like conditions in terms of survival, DNA, and ultrastructural stability. Results offer insights into the habitability of Mars for future exploration missions on Mars. Implications for the detection of biosignatures in extraterrestrial conditions and planetary protection are discussed.

A kombucha multimicrobial culture (KMC) was exposed to simulated Mars-like conditions in low-Earth orbit (LEO). The study was part of the Biology and Mars Experiment (BIOMEX), which was accommodated in the European Space Agency's EXPOSE-R2 facility, outside the International Space Station. The aim of the study was to investigate the capability of a KMC microecosystem to survive simulated Mars-like conditions in LEO. During the 18-month exposure period, desiccated KMC samples, represented by living cellulose-based films, were subjected to simulated anoxic Mars-like conditions and ultraviolet (UV) radiation, as prevalent at the surface of present-day Mars. Postexposure analysis demonstrated that growth of both the bacterial and yeast members of the KMC community was observed after 60 days of incubation; whereas growth was detected after 2 days in the initial KMC. The KMC that was exposed to extraterrestrial UV radiation showed degradation of DNA, alteration in the composition and structure of the cellular membranes, and an inhibition of cellulose synthesis. In the "space dark control" (exposed to LEO conditions without the UV radiation), the diversity of the microorganisms that survived in the biofilm was reduced compared with the ground-based controls. This was accompanied by structural dissimilarities in the extracellular membrane vesicles. After a series of subculturing, the revived communities restored partially their structure and associated activities.

As a part of the European Space Agency mission "EXPOSE-R2" on the International Space Station (ISS), the BIOMEX (Biology and Mars Experiment) experiment investigates the habitability of Mars and the limits of life. In preparation for the mission, experimental verification tests and scientific verification tests simulating different combinations of abiotic space- and Mars-like conditions were performed to analyze the resistance of a range of model organisms. The simulated abiotic space- and Mars-stressors were extreme temperatures, vacuum, and Mars-like surface ultraviolet (UV) irradiation in different atmospheres. We present for the first time simulated space exposure data of mosses using plantlets of the bryophyte genus Grimmia, which is adapted to high altitudinal extreme abiotic conditions at the Swiss Alps. Our preflight tests showed that severe UVR 200-400nm irradiation with the maximal dose of 5 and 6.8 × 10 5 kJ·m -2 , respectively, was the only stressor with a negative impact on the vitality with a 37% (terrestrial atmosphere) or 36% reduction (space- and Mars-like atmospheres) in photosynthetic activity. With every exposure to UVR 200-400nm 10 5 kJ·m -2 , the vitality of the bryophytes dropped by 6%. No effect was found, however, by any other stressor. As the mosses were still vital after doses of ultraviolet radiation (UVR) expected during the EXPOSE-R2 mission on ISS, we show that this earliest extant lineage of land plants is highly resistant to extreme abiotic conditions.

BIOMEX (BIOlogy and Mars EXperiment) is an ESA/Roscosmos space exposure experiment housed within the exposure facility EXPOSE-R2 outside the Zvezda module on the International Space Station (ISS). The design of the multiuser facility supports-among others-the BIOMEX investigations into the stability and level of degradation of space-exposed biosignatures such as pigments, secondary metabolites, and cell surfaces in contact with a terrestrial and Mars analog mineral environment. In parallel, analysis on the viability of the investigated organisms has provided relevant data for evaluation of the habitability of Mars, for the limits of life, and for the likelihood of an interplanetary transfer of life (theory of lithopanspermia). In this project, lichens, archaea, bacteria, cyanobacteria, snow/permafrost algae, meristematic black fungi, and bryophytes from alpine and polar habitats were embedded, grown, and cultured on a mixture of martian and lunar regolith analogs or other terrestrial minerals. The organisms and regolith analogs and terrestrial mineral mixtures were then exposed to space and to simulated Mars-like conditions by way of the EXPOSE-R2 facility. In this special issue, we present the first set of data obtained in reference to our investigation into the habitability of Mars and limits of life. This project was initiated and implemented by the BIOMEX group, an international and interdisciplinary consortium of 30 institutes in 12 countries on 3 continents. Preflight tests for sample selection, results from ground-based simulation experiments, and the space experiments themselves are presented and include a complete overview of the scientific processes required for this space experiment and postflight analysis. The presented BIOMEX concept could be scaled up to future exposure experiments on the Moon and will serve as a pretest in low Earth orbit.

The multi-user facility EXPOSE-E was designed by the European Space Agency to enable astrobiology research in space (low-Earth orbit). On 7 February 2008, EXPOSE-E was carried to the International Space Station (ISS) on the European Technology Exposure Facility (EuTEF) platform in the cargo bay of Space Shuttle STS-122 Atlantis. The facility was installed at the starboard cone of the Columbus module by extravehicular activity, where it remained in space for 1.5 years. EXPOSE-E was returned to Earth with STS-128 Discovery on 12 September 2009 for subsequent sample analysis. EXPOSE-E provided accommodation in three exposure trays for a variety of astrobiological test samples that were exposed to selected space conditions: either to space vacuum, solar electromagnetic radiation at >110 nm and cosmic radiation (trays 1 and 3) or to simulated martian surface conditions (tray 2). Data on UV radiation, cosmic radiation, and temperature were measured every 10 s and downlinked by telemetry. A parallel mission ground reference (MGR) experiment was performed on ground with a parallel set of hardware and samples under simulated space conditions. EXPOSE-E performed a successful 1.5-year mission in space.

Following an European Space Agency announcement of opportunity in 1996 for "Externally mounted payloads for 1st utilization phase" on the International Space Station (ISS), scientists working in the fields of astrobiology proposed experiments aiming at longterm exposure of a variety of chemical compounds and extremely resistant microorganisms to the hostile space environment. The ESA exposure facility EXPOSE was built and an operations' concept was prepared. The EXPOSE experiments were developed through an intensive pre-flight experiment verification test program. 12 years later, two sets of astrobiological experiments in two EXPOSE facilities have been successfully launched to the ISS for external exposure for up to 1.5 years. EXPOSE-E, now installed at the balcony of the European Columbus module, was launched in February 2008, while EXPOSE-R took off to the ISS in November 2008 and was installed on the external URM-D platform of the Russian Zvezda module in March 2009.

The evolution of the solar system and the origin of life remain some of the most intriguing questions for humankind. Addressing these questions experimentally is challenging due to the difficulty of mimicking environmental conditions representative for Early Earth and/or space conditions in general in ground-based laboratories. Performing experiments directly in space offers the great chance to overcome some of these obstacles and to possibly find answers to these questions. Exposure platforms in Low Earth Orbit (LEO) with the possibility for long-duration solar exposure are ideal for investigating the effects of solar and cosmic radiation on various biological and non-biological samples. Up to now, the Exobiology and space science research community has successfully made use of the International Space Station (ISS) via the EXPOSE facility to expose samples to the space environment with subsequent analyses after return to Earth. The emerging small and nanosatellite market represents another opportunity for astrobiology research as proven by the robotic O/OREOS mission, where samples were monitored in-situ , i.e. in Earth orbit. In this framework, the European Space Agency is developing a novel Exobiology facility outside the ISS. The new platform, which can host up to four different experiments, will combine the advantages of the ISS (long-term exposure, sample return capability) with near-real-time in-situ monitoring of the chemical/biological evolution in space. In particular, ultraviolet–visible (UV–Vis) and infrared (IR) spectroscopy were considered as key non-invasive methods to analyse the samples in situ. Changes in the absorption spectra of the samples developing over time will reveal the chemical consequences of exposure to solar radiation. Simultaneously, spectroscopy provides information on the growth rate or metabolic activities of biological cultures. The first quartet of experiments to be performed on-board consists of IceCold, OREOcube and Exocube (dual payload consisting of ExocubeChem and ExocubeBio). To prepare for the development of the Exobiology facility, ground units of the UV–Vis and IR spectrometers were studied, manufactured and tested as precursors of the flight units. The activity led to a modular in-situ spectroscopy platform able to perform different measurements (e.g. absorbance, optical density, fluorescence measurements) at the same time on different samples. We describe here the main features of the ground model platform, the verification steps, results and approach followed in the customization of commercial–off-the-shelf (COTS) modules to make them suitable for the space environment. The environmental tests included random and shock vibration, thermal vacuum cycles in the range −20 °C to +40 °C and irradiation of the components with a total dose of 1800 rad (18 Gy). The results of the test campaign consolidated the selection of the optical devices for the Exobiology Facility. The spectroscopic performance of the optical layout was tested and benchmarked in comparison with state-of-the-art laboratory equipment and calibration standards showing good correlation. This includes spectra of samples sets relevant for the flight experiments and a performance comparison between the SPECTROModule ground model and state-of-the-art laboratory spectrometers. Considering the large number of samples and different types of optical measurements planned on-board the ISS, the main outcome was the implementation of an LED-photodiode layout for the optical density and fluorescence measurements of IceCold (42 samples) and ExocubeBio (111 samples); while the UV–Vis spectrometer will be mainly focused on the change of the absorption spectra of the 48 samples of OREOcube.The ExocubeChem samples (in total 48) will be analysed by infrared spectroscopy. The ground platform supports the establishment of analogue research capabilities able to address the long-term objectives beyond the current application. • Biological samples shall be exposed to solar and cosmic radiation. • UV/VIS and IR spectroscopy are used for in-situ measurements in space. • Commercial off-the shelf (COTS) spectrometers were tested and verified for use in space. • A scientific test campaign proved the predicted results. • SPECTROmodule is the precursor mission of the Exobiology facility accommodated outside the International Space Station.
