Melilotus albus, which contains abundant pharmacologically active coumarins, is usually used as a rotation crop and green manure worldwide. Abscisic acid (ABA) is a crucial plant hormone that plays an important role in plant stress responses. There is a paucity of information about the ABA signaling pathway and its regulatory network in M. albus. Here, we performed a comparative physiological and transcriptomic analysis to assess the response of M. albus to exogenous ABA. Physiological analysis revealed that proline (Pro), soluble protein and H2O2 content after ABA treatment 3 h significantly increased by 14.0%, 12.0% and 32.4% compared with 0 h in M. albus. A total of 19,855 differentially expressed genes (DEGs) were identified under ABA treatment, including 13,392 in shoots and 15,471 in roots. We obtained two modules that were significantly correlated with the ABA treatment (the darkorange module was positively correlated at 24 h in the shoot, brown2 module positively correlated at 3 h in the root) by weighted correlation network analysis (WGCNA). KEGG enrichment analysis showed that genes within two modules were primarily enriched in protein synthesis and metabolism, secondary metabolites, purine and pyrimidine metabolism, and phenylalanine, tyrosine and tryptophan biosynthesis. GO enrichment analysis indicated that genes within two modules were primarily enriched in energy substance metabolism. These pathways were mainly associated with abiothic stress, which indicated that exogenous application of ABA activated the stress resistance system of M. albus. The hub gene 4CL1 (4-Coumarate: CoA ligase 1) was translated and expressed in yeast, resulting in enhanced salt and ABA tolerance in the transgenic yeast. Overexpression of Ma4CL1 in M. albus improved the salt resistance of the transgenic plants. In order to explore the characteristics of sugar beet (Beta vulgaris L.) AP2s (BvAP2s) in response to drought stress hormone abscisic acid (ABA), genome-wide identification, and the phylogeny, gene structure and promoter precursor analysis of the BvAP2s were employed to predict their potential functions. It is shown that there are a total of 13 BvAP2 genes in the Beta vulgaris. Based on the primary amino acid sequence, the BvAP2s can be further subdivided into euAP2, euANT and basalANT. In addition, cis-acting element analysis showed that BvAP2s contained several abiotic stress-related elements, including those associated with ABA and drought stress. Roots are the first to perceive stress signals, and ABA-treated beetroot transcriptome and downstream gene prediction of BvAP2s revealed that BVRB_4g074790, BVRB_6g128480 and BVRB_7g179610 may play an important role involved in ABA signaling pathways during the stress response by regulating downstream GRAM genes, LEAs and U-boxes. Additionally, quantitative real-time polymerase chain reaction (qRT-PCR) further confirmed the downregulation of these three BvAP2s in response to ABA induction in sugar beet roots. This study investigates the role of abscisic acid (ABA) in bolstering drought resistance in plants, employing "Panjiang Sophora viciifolia" as the subject. A simulated drought scenario was created using polyethylene glycol (PEG-6000) to examine the impact of varying drought intensities (0%, 5%, 20% PEG) and ABA concentrations (0, 10, 50, 100, 200 mg·L−1) on the germination and physiological parameters of Sophora viciifolia. The results showed that in the absence of ABA, the germination rate (GR), germination potential (GP), and germination index (GI) of S. viciifolia seeds initially increased and then decreased with escalating PEG-induced drought stress. At PEG-induced drought stress levels of 5% and 20%, the activities of peroxidase (POD) and catalase (CAT), along with the malondialdehyde (MDA) content, were significantly higher than in the control (CK) (p < 0.05). In response to drought stress, S. viciifolia seeds adapted by modulating germination behavior, augmenting the content of osmoregulatory substances, and boosting the activity of protective enzymes. The addition of ABA markedly enhanced GR, GE, GI, activities of POD, superoxide dismutase (SOD), and CAT, as well as the levels of MDA and proline (Pro) under drought conditions (p < 0.05). Relative to CK, low ABA concentrations (10–100 mg·L−1) resulted in increased GR, GP, GI, POD, SOD, CAT, MDA, and Pro levels; whereas, at a higher concentration (200 mg·L−1), although GR, GP, and GI decreased, POD, SOD, CAT, MDA, and Pro levels increased. Through principal component analysis and membership function comprehensive evaluation, it was determined that administering 50 mg·L−1 ABA was most effective in enhancing drought resistance in S. viciifolia seedlings. This study comprehensively investigates the physiological and molecular regulatory mechanisms of Camellia oleifera seedlings under drought stress with a soil moisture content of about 30%, where exogenous abscisic acid (ABA) was applied via foliar spraying at concentrations of 50 µg/L, 100 µg/L, and 200 µg/L. The results demonstrated that appropriate concentrations of ABA treatment can regulate the physiological state of the seedlings through multiple pathways, including photosynthesis, oxidative stress response, and osmotic balance, thereby aiding in the restructuring of their drought response strategy. ABA treatment effectively activated the antioxidant system by reducing stomatal conductance and moderately inhibiting the photosynthetic rate, thus alleviating oxidative damage caused by drought stress. Additionally, ABA treatment promoted the synthesis of osmotic regulators such as proline, maintaining cellular turgor stability and enhancing the plant's drought adaptability. The real-time quantitative PCR results of related genes indicated that ABA treatment enhanced the plant's response to the ABA signaling pathway and improved disease resistance by regulating the expression of related genes, while also enhancing membrane lipid stability. A comprehensive evaluation using a membership function approach suggested that 50 µg/L ABA treatment may be the most-effective in mitigating drought effects in practical applications, followed by 100 µg/L ABA. The application of 50 µg/L ABA for 7 h induced significant changes in various biochemical parameters, compared to a foliar water spray. Notably, superoxide dismutase activity increased by 17.94%, peroxidase activity by 30.27%, glutathione content by 12.41%, and proline levels by 25.76%. Background: Low temperatures during flowering severely affect crop growth and yield. Methods: This experiment was conducted under potted conditions with LXD 4 and TJH as test materials and pre-sprayed with exogenous ABA at a concentration of 20 mg·L-1 at flowering stage, respectively and then with an average of 15℃ condition for 5 days. Result: The results showed that foliar spraying exogenous ABA effectively inhibited the increase of MDA content and relatively significantly improved the photosynthetic parameters such as Gs and Tr and alleviated the loss of yield factors caused by chilling stress. Compared with spraying water, spraying exogenous ABA can significantly increase the yield per pot of LXD 4 by 6.72~17.77% and TJH by 6.41~37.04% under low temperature conditions. In summary, foliar spraying exogenous ABA ameliorated the effects of chilling stress on the physiological characteristics of adzuki bean leaves and improved yield as a result of improved antioxidant defense mechanisms which impeded lipid peroxidation. Therefore, a strategy is needed to enhance the tolerance of crops to this heavy metal or reduce its accumulation in crops. Abscisic acid (ABA) plays an active role in plants' response to abiotic stress. The application of exogenous ABA can reduce Cd accumulation in shoots of some plants and enhance the tolerance of plants to Cd; therefore, ABA may have good application prospects. In this paper, we reviewed the synthesis and decomposition of ABA, ABA-mediated signal transduction, and ABA-mediated regulation of Cd-responsive genes in plants. We also introduced physiological mechanism underlying Cd tolerance because of ABA. Specifically, ABA affects metal ion uptake and transport by influencing transpiration and antioxidant systems, as well as by affecting the expression of metal transporter and metal chelator protein genes. Drought at the grain filling stage of wheat will cause premature leaf senescence, thus leading to considerable loss of wheat yield. Therefore, this paper aims to establish a cultivation technology for strong drought resistance, delayed senescence, and yield improvement based on the analysis of hormones homeostasis obtained by applying chemical control substances. Experiments were conducted with two genotypes of wheat. Four water irrigation treatments were applied to impose the water deficit, including well-watered control treatment (WW), mild water deficit (MiWD), moderate water deficit (MoWD), and severe water deficit (SWD). Exogenous abscisic acid (ABA) was sprayed on the plants at the anthesis stage of the wheat. As a result, exogenous ABA reduced initial senescence rate (r0), total duration of chlorophyll (Chltotal), rapid senescence phase (Chlloss), and the accumulated temperature at an inflection point (M) but improved the persistence phase (Chlper) of flag leaves under all of the four treatments. However, exogenous ABA produced inconsistent effects on photoassimilate relocation and grain weight under different treatments. It produced positive regulatory effects on grain weight under WW, MiWD, and MoWD treatments. On the one hand, spraying ABA during the persistence phase of flag leaves reduced the ratios of zeatin to gibberellin (Z/GA3), spermine to spermidine (Spm/Spd), and salicylic acid to ABA (SA/ABA), which prolonged active photosynthesis by stimulating high level of proline (Pro) and increased the activities of antioxidant enzymes, such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX). Therefore, drought tolerance was enhanced, and more photosynthetic assimilates were accumulated. On the other hand, the rapid senescence phase and the transport rate of assimilates into grains were accelerated, resulting in higher grain weight, yield, and water use efficiency (WUE). However, under SWD treatment, exogenous ABA improved the ratio of SA/ABA, leading to low Pro content and low antioxidant enzyme activity of flag leaves in the rapid loss phase. Meanwhile, drought resistance declined and the transport duration of assimilates into grains was shortened, thus making photosynthetic assimilates redundant. Therefore, exogenous ABA can lead to the reduction in grain weight, yield, and WUE of wheat under SWD treatment.

As one of the main phytohormones, abscisic acid (ABA) has been recently studied to regulate the fruit qualities during the post-harvest storage. However, the mechanism of the ABA affected the texture changes of post-harvest fruits remained unclear. In this study, the mature fruits of astringent persimmon 'Gongcheng-shuishi' with firm flesh had been treated with 300 mg/L ABA. Compared to the control group, the decreasing of fruit firmness as well as color development had been enhanced by exogenous ABA during post-harvest storage. A total of 300 differentially expressed genes (DEGs) had been isolated by means of Weighted Gene Co-Expression Network Analysis (WGCNA) and predicted to be associated with the changes of fruit firmness and color index. Among these DEGs, DkPG6 , DkXTH13 , DkBG11/12 showed highly correlated with cell wall metabolism, and DkPSY2 with the carotenoid accumulation. There were totally 12 differentially expressed transcript factors (DETFs) among these 300 DEGs. As transcript activator and repressor respectively, DkZAT4 (ZAT-LIKE TRANSCRIPTION FACTOR) and DkTCP4 (TCP TRANSCRIPTION FACTOR) could bind to and regulate the transcript activity of DkBG11 promoter significantly based on the results of dual luciferase assay and electrophoretic mobility shift assay. [Objective] In order to provide theoretical basis for exploring the physiological mechanism of adzuki bean under flooding stress and the application effect of exogenous ABA, we studied the effects of flooding stress at seedling stage on physiological and yield of adzuki bean, and the alleviating effect of exogenous ABA on stress. [Method] The pot experiment in the research was conducted, and the crop cultivar included Longxiaodou 4 (LXD 4) and Tianjinhong (TJH). The treatment of flooding and foliar spraying ABA in seedling period of adzuki bean was applied. The lasting time of flooding stress in the experiment was 5 days. The purpose was to analyze the effect of ABA pretreatment against flooding stress, on the accumulation of reactive oxygen species (ROS) in adzuki bean roots, membrane lipid peroxidation damage, antioxidant system, and yield, and mitigating effects of exogenous ABA [ Result] The physiological indexes in root of adzuki bean in flooding stress were changed during seedling stage. The content of H2O2 and MDA, praline, soluble sugar and soluble protein were significantly increased, the same as SOD, POD and CAT activities. Flooding treatment for 5 days resulted in a significant decrease in the output of LXD 4 in a single pot by 6.18% to 8.40%, and that of TJH decreased by 4.97% to 9.91%. Exogenous ABA has the effect of resisting flooding stress, can effectively increase the praline, soluble sugar and soluble protein content in the root of adzuki bean. At the same time, it can significantly reduce the content of H2O2 and MDA. In addition, it can significantly increase the activity of SOD, POD and CAT, the value of SOD/ POD. Foliar spraying exogenous ABA increased the yield of LXD 4 significantly by 1.50% to 6.05% after 3 days of flooding, and increased the yield of TJH significantly by 3.00% to 4.46% after 4 days of flooding, respectively. [Conclusion] flooding stress caused H2O2 accumulation and increased membrane lipid peroxidation in adzuki bean roots, inducing protective enzyme activity and increasing osmotic adjustment substance content to resist the stress, and flooding stress eventually led to a decline in yield. Exogenous ABA can increase the protective enzyme activity and the content of osmotic adjustment substances, improve the antioxidant capacity, reduce the damage caused by flooding stress, and increase the yield of adzuki bean, relatively. Abscisic acid (ABA) plays an important role in regulating activities of antioxidant enzymes, hormone levels, accumulation of reactive oxygen species under stress. To understand the effects of exogenous ABA treatment on flooding tolerance at seedling stage in adzuki bean (Vigna angularis), the water-sensitive (WS) variety ‘Tianjinhong’ (TJH) and the water-tolerant (WT) variety ‘Longxiaodou 4’ (LXD 4) were foliar sprayed exogenous ABA with 20 μM, followed by flooding stress for 5 days under pot conditions. Our results revealed that under flooding stress at seedling stage, the contents of H2O2 and MDA, proline and soluble protein in adzuki bean leaves significantly increased, the contents of ABA, IAA and SA significantly increased, and the activities of SOD, POD and CAT also significantly increased. Flooding treatment for 5 days resulted in a significant decrease in single pot yield of LXD 4 by 8.40% and TJH by 9.91%. Foliar spray of 20 μM exogenous ABA could resist flooding stress, considerably increased the contents of proline and soluble protein, significantly decreased the contents of H2O2 and MDA, significantly increased the activities of SOD and POD, increased the contents of ABA and SA, and significantly inhibited the increase of IAA content in adzuki bean leaves. Spraying exogenous ABA significantly increased the yield of ‘LXD’ 4 by 6.95% after 4 days of flooding treatment, and ‘TJH’ by 4.46%. To sum up, there were significant differences in physiological stress among different varieties of adzuki bean under flooding stress, and spraying exogenous ABA could effectively alleviate the effects of flooding stress on physiology and yield of adzuki bean. The results of this study provided a theoretical basis for further studying the physiological mechanism of adzuki bean resisting flooding stress at seedling stage and improving the yield of adzuki bean under flooding stress. Thus, foliar spraying exogenous ABA effectively improved submergence tolerance by enhancing the activity of protective enzymes and osmoregulation. These results provided novel insights and were expected to aid in the development of more effective stress resistance cultivation methods in adzuki bean production.

To explore the role of abscisic acid (ABA) in modulating the response of plant water relation characteristics to progressive drought under ambient (a[CO2], 400 ppm) and e[CO2] (800 ppm) growth environments, two tomato (Solanum lycopersicum) genotypes, Ailsa Craig (AC) and its ABA-deficient mutant (flacca), were grown in pots, treated with or without exogenous ABA, and exposed to progressive soil drying until all plant available water in the pot was depleted. The results showed that exogenous ABA application improved leaf water potential, osmotic potential, and leaf turgor and increased leaf ABA concentrations ([ABA]leaf) in AC and flacca. In both genotypes, exogenous ABA application decreased stomatal pore aperture and stomatal conductance (gs), though these effects were less pronounced in e[CO2]-grown AC and gs of ABA-treated flacca was gradually increased until a soil water threshold after which gs started to decline. In addition, ABA-treated flacca showed a partly restored stomatal drought response even when the accumulation of [ABA]leaf was vanished, implying [ABA]leaf might be not directly responsible for the decreased gs. During soil drying, [ABA]leaf remained higher in e[CO2]-grown plants compared with those under a[CO2], and a high xylem sap ABA concentration was also noticed in the ABA-treated flacca especially under e[CO2], suggesting that e[CO2] might exert an effect on ABA degradation and/or redistribution. Collectively, a fine-tune ABA homeostasis under combined e[CO2] and drought stress allowed plants to optimize leaf gas exchange and plant water relations, yet more detailed research regarding ABA metabolism is still needed to fully explore the role of ABA in mediating plant physiological response to future drier and CO2-enriched climate.

Beyond climatic conditions, qualitative performance is led by the intrinsic characteristics of the genotype. The aim of this study was to investigate the relationship between vine water status and exogenous abscisic acid (ABA) application on berry composition of the cultivars Cannonau, Merlot and Sangiovese. The experiment, carried out in 2016 and 2017, consisted of comparing two levels of irrigation treatments, full irrigation versus a non-irrigation treatment. Within each treatment, two sub-treatments were set up: (i) 4 mL L-1 of exogenous ABA applied at veraison to clusters only and subsequently repeated after six days; (ii) a control (untreated vines). The application of di erent irrigation regimes confirmed that the response to water stress is highly cultivar-dependent. Berry composition was influenced di erently among cultivars by water stress. In terms of metabolites, positive influences were observed with Cannonau. No significant e ects were observed by spraying exogenous ABA directly on grapes. Moreover, no significant interactions were found between the application of water stress and ABA. Exogenous ABA application did not appear to be a viticultural practice capable of influencing must composition in environments characterized by severe environmental conditions such as heat and drought. Thellungiella salsuginea is highly tolerant to abiotic stress, while its a close relative Arabidopsis thaliana is sensitive to stress. This characteristic makes T. salsuginea an excellent model for uncovering the mechanisms of abiotic stress tolerance. Abscisic acid (ABA) plays essential roles in plant abiotic and biotic stress tolerance.To test the changes in gene expression of T. salsuginea under ABA treatment, in this study, the transcriptomes of T. salsuginea roots and leaves were compared in response to exogenously application of ABA.The results showed that ABA treatment caused different expression of 2,200 and 3,305 genes in leaves and roots, respectively, compared with the untreated control. In particular, genes encoding transcription factors such as WRKY, MYB, NAC, GATA, ethylene‐responsive factors (ERFs), heat stress transcription factors, basic helix‐loop‐helix, PLATZ and B3 domain‐containing family members were enriched. In addition, 49 and 114 differentially expressed genes were identified as ABA‐regulated genes, separately in leaves and roots, respectively, which were related to biotic and abiotic stresses. The expression levels of some genes were validated by qRT‐PCR. Different responses of genes to ABA treatment were discovered in T. salsuginea and A. thaliana.This transcriptome analysis expands our understanding of the role of ABA in stress tolerance in T. salsuginea. The purpose of this study was to investigate the physiological responses of drought-tolerant and drought-susceptible soybean genotypes to exogenous abscisic acid (ABA) application during progressive soil drying at seedling stages. Five-day old soybean seedlings were transplanted into PVC tubes filled with soil mixture. Seedlings were watered daily with similar water volumes until second trifoliate leaves emerged, and thereafter soil drying with or without exogenous ABA application was imposed. Half of the seedlings of each genotype were left for regular watering as control plants. Soil water status declined significantly over seven days of withholding water supply for both genotypes. Leaf expansion rate, stomatal conductance (gs), leaf water potential (ψw), and relative water content of leaves (%RWC) declined significantly under soil drying as well as soil drying with ABA application, compared to their values for well-watered soybean genotypes. However, a drought-tolerant genotype (C12) responded more rapidly than a drought-susceptible genotype (C08) after imposition of soil drying and soil drying with exogenous ABA. In addition, application of exogenous ABA to water-restricted soybeans resulted in higher %RWC and ψw in the drought-tolerant than in the drought-susceptible genotype. Compared to the drought-susceptible genotype, the drought-tolerant genotype was more responsive to exogenous ABA application, resulting in a higher root-to-shoot ratio.

‘Dongnong Dongmai1 (Dn1)’ is a cultivar of wheat (Triticum aestivum L.) which may endure‒30°C in field with a reviving rate of 85%. ABA may enhance plant resistance. Aiming to reveal Dn1 coldresistant mechanism after exogenous ABA (exo-ABA) treated (control: non-ABA treated), some transcription factors (TFs) were selected and detected. Seedlings were prayed with exo-ABA at three-leaf stage, subsequently their tilling nodes were respectively sampled when it was at 5, 0,-10 and-25°C in field. Six TF genes (TabZIP1, TaWABI5, TaMYB1, TaMYB80, TaNAC2 and TaWRKY80) were selected and their expression at different low temperature was detected, the results showed that: in non-ABA treated tilling nodes, the expression of six TF genes all reached their peak level at-10°C; in exo-ABA treated tilling nodes, the expression of five TF genes (TabZIP1, TaWABI5, TaMYB80, TaNAC2 and TaWRKY80) reached their peak level at-25°C, and particularly the expression level of TabZIP1 was the highest in five TFs. In order to further defined the biological function of TabZIP1, the physical and chemical properties, protein secondary structure and the evolutionary tree was analyzed and predicted by some bioinformatics software or online program; TabZIP1 was localized in nuclear of tobacco (Nicotiana tabacum), which was observed by laser scanning confocal microscope (LSCM). This study initially exposed some TFs in Dn1 were upregulated to respond to the coldstress after exo-ABA treated, which indicated that a serious cold-responsive genes could be transcripted with the TFs assistance, and the results provided new theoretical basis to reveal the cold-reponsive mechanism of winter wheat. To alleviate the inhibition on plant growth and productivity caused by acid rain, it is crucial to clarify the regulating mechanism of ABA on adaptation of plants to acid rain. Here, we studied the effects of exogenously applied ABA on nutrients uptake of rice roots under simulated acid rain (SAR) stress from physiological, biochemical and molecular aspects. Compared to the single SAR treatment (pH 4.5 or 3.5), exogenous 10 μM ABA alleviated the SAR-induced inhibition of root growth by balancing endogenous hormones (abscisic acid, indole-3-acetic acid, gibberellic acid and zeatin), promoting nutrient uptake (nitrate, P, K and Mg) in rice roots, and increasing the activity of the plasma membrane H+-ATPase by up-regulating expression levels of genes (OSA2, OSA4, OSA9 and OSA10). However, exogenous 100 μM ABA exacerbated the SAR-caused inhibition of root growth by disrupting the balance of endogenous hormones, and inhibiting nutrient uptake (nitrate, P, K, Ca and Mg) through decreasing the activity of the plasma membrane H+-ATPase. These results indicate that proper concentration of exogenous ABA could enhance tolerance of rice roots to SAR stress by promoting nutrients uptake and balancing endogenous hormones. Peroxisomal enoyl-CoA delta isomerase2 (PECI2) is one of the key enzymes that has critical role in lipid metabolism and plant development during salt stress. Seven out of ten tobacco plants overexpressing human PECI2 (HsPECI2) with PTS1-sequence showed hypersensitivity to salt. Under salt-stress, T2 transformed plants (HsPECI2) displayed reduced primary root, delayed shoot-growth, and visibly smaller rosette leaves turning pale yellow as compared to the pKYLX71 vector control plant. Also, we found altered reactive oxygen species (ROS) levels and reduced catalase activity in 100mM sodium chloride (NaCl) treated HsPECI2 transformed plant compared with the pKYLX71 counterpart. ESI-MS/MS data showed that the polar lipids were differentially modulated upon salt treatment in HsPECI2 transformed and pKYLX71 plants as compared with the respective untreated counterpart. Notably, the levels of monogalactosyldiacylglycerol and phosphatidic acid varied significantly, whereas phosphatidylcholine, phosphatidylserine and digalactosyldiacylglycerol contents were moderately upregulated. In parallel, abscisic acid (ABA) responsiveness assay confirmed insensitivity of HsPECI2 transformed plant towards ABA. Abscisic acid (ABA) plays an important role in regulating photosynthesis under stress. To understand the differential function of exogenous ABA in the regulation of drought tolerance between two rice ( Oryza sativa L.) genotypes, upland rice (UR, resistant to drought stress) and lowland rice (LR, susceptible to drought stress), photosynthetic parameters, chlorophyll fluorescence parameters, and the expression of chloroplast and ABA biosynthesis-related genes were investigated under 15 % polyethylene glycol (PEG) and exogenous ABA (60 µM) treatments. In both rice lines, most of the photosynthetic parameters, chlorophyll fluorescence parameters, and chloroplast and ABA biosynthesis-related gene transcript levels were rapidly reduced by PEG stress, with the exception of up-regulated levels of OsPsbA, OsNCED3, OsNCED4, and OsZEP in LR and OsNCED3, OsNCED4, and OsZEP in UR. Moreover, a rapid stress-responsive regulation mechanism was found in UR according to the more rapid and strong up-regulation of three ABA biosynthesis-related genes in UR than in LR. Under PEG stress, exogenous ABA application significantly enhanced the recovery of the net photosynthetic rate ( P), stomatal conductance ( G), and transpiration rate ( T) in UR, with increased expression of OsPsbD1, OsPsbD2, OsNCED2, OsNCED3, OsNCED4, and OsNCED5. Background Abscisic acid (ABA) can regulate the expressions of many stress-responsive genes in plants. However, in defense responses to pathogens, mounting evidence suggests that ABA plays variable roles. Little information exists about genome-wide gene expression in ABA responses in tomato (Solanum lycopersicum L.), a model fruit crop plant. Results Global transcriptome profiles of tomato leaf responses to exogenous ABA were generated using Illumina RNA-sequencing. More than 173 million base pair reads were mapped onto the tomato reference genome and the expression pattern differences between treated and control leaves were assessed. In total, 50,616 transcripts were generated. Among them, 42,583 were functionally annotated in the NCBI non-redundant database and 47,877 in the tomato genome reference. Additionally, 31,107 transcripts were categorized into 57 functional groups based on Gene Ontology terms, and 14,371 were assigned to 310 Kyoto Encyclopedia of Genes and Genomes pathways. In both the ABA treatment and control samples, 39,671 transcripts were available to analyze their expressions, of which 21,712 (54.73%) responded to exogenous ABA. Of these transcripts, 2,787 were significantly differently expressed genes (DEGs). Many known and novel ABA-induced and -repressed genes were found. Exogenous ABA can influence the ABA signaling pathway with PYR/PYL/RCARs-PP2Cs-SnRK2s as the center. Eighteen PYL genes were detected. A large number of genes related to various transcription factors, heat shock proteins, pathogen resistance, and the salicylic acid, jasmonic acid, and ethylene signaling pathways were upregulated by exogenous ABA. Conclusions The results indicated that ABA has the potential to improve pathogen-resistance and abiotic stress tolerance in tomato. This study presents the global expression analysis of ABAregulated transcripts in tomato and provides a robust database for investigating the functions of genes induced by ABA. During one growing season, the effects of enhanced ultraviolet-B (UV-B) radiation, exogenous abscisic acid (ABA) and their combination on biomass accumulation, gas exchange, endogenous ABA, the concentration of UV-absorbing compounds, antioxidant system and on the carbon (C) and nitrogen (N) content and C/N ratio were investigated in two contrasting Populus cathayana Rehd. populations, originating from high and low altitudes in south-west China. Exogenous ABA was sprayed to the leaves, and enhanced UV-B treatments were applied using a square-wave system to expose the seedlings to ambient (1×) or twice ambient (2×) doses of biologically effective UV-B radiation (UV-BBE). Enhanced UV-B radiation significantly decreased height, basal diameter, total leaf area, total biomass, net CO2 assimilation rate (A), stomatal conductance (g s ), transpiration rate (E) and carbon (C) content in leaves, and significantly increased the activities of superoxide dismutase (SOD) and guaiacol peroxidase (GPx), and the contents of hydrogen peroxide (H2O2) and malonaldehyde (MDA), as well as the accumulation of UV-absorbing compounds and endogenous ABA concentrations among different organs in both populations. In contrast, exogenous ABA induced a significant decrease in A and significant increases in the activities of SOD and GPx, in the content of H2O2 and MDA, and in the endogenous ABA concentrations. Compared with the low altitude population, the high altitude population was more tolerant to enhanced UV-B and exogenous ABA. Significant interactions between UV-B and ABA were observed in A, E, and in the activities of SOD and GPx, as well as in endogenous ABA in the leaves and roots of both populations. The soybean ( Glycine max) Heihe No. 23 is sensitive to imbibitional chilling injury. Polyethylene glycol (PEG) treatment can improve chilling tolerance of soybean seeds to a certain extent. The changes of hydrolytic ATPase in plasma membranes and H+-pumping responses in soybean seeds were investigated during PEG treatments. Effects of exogenous calcium and exogenous ABA on the hydrolytic ATPase were also examined in order to understand the mechanism of chilling resistance. Highly purified plasma membranes were isolated by 6.0% aqueous two-phase partitioning from soybean seeds, as judged by the sensitivity of hydrolytic ATPase to sodium vanadate. PEG treatment resulted in a slight increase of the hydrolytic ATPase activity in 12 h. Then the activity decreased gradually, but still higher than the control. The H+-pumping activity increased steadily during PEG treatment. Exogenous calcium had both activating and inhibiting effects on the hydrolytic ATPase, but the activity was inhibited in soybean seeds treated with exogenous ABA.
