In this study, mycelia of eight recently described species of Halophytophthora and H. avicennae collected in Southern Portugal were analysed for lipids and fatty acids (FA) content to evaluate their possible use as alternative sources of FAs and understand how each species FAs profile relates to their phylogenetic position. All species had a low lipid percentage (0.06% in H. avicennae to 0.28% in H. frigida ). Subclade 6b species contained more lipids. All species produced monounsaturated (MUFA), polyunsaturated (PUFA) and saturated (SFA) FAs, the latter being most abundant in all species. H. avicennae had the highest FA variety and was the only producer of γ-linolenic acid, while H. brevisporangia produced the lowest number of FAs. The best producer of arachidonic acid (ARA) and eicosapentaenoic acid (EPA) was H. thermoambigua with 3.89% and 9.09% of total FAs, respectively. In all species, palmitic acid (SFA) was most abundant and among the MUFAs produced oleic acid had the highest relative percentage. Principal component analysis (PCA) showed partial segregation of species by phylogenetic clade and subclade based on their FA profile. H. avicennae (Clade 4) differed from all other Clade 6 species due to the production of γ-linolenic and lauric acids. Our results disclosed interesting FA profiles in the tested species, adequate for energy (biodiesel), pharmaceutical and food industries (bioactive FAs). Despite the low amounts of lipids produced, this can be boosted by manipulating culture growth conditions. The observed interspecific variations in FA production provide preliminary insights into an evolutionary background of its production.

This work studied the potential biotechnological applications of a naviculoid diatom (IMA053) and a green microalga ( Tetraselmis marina IMA043) isolated from the North Adriatic Sea. Water, methanol, and dichloromethane (DCM) extracts were prepared from microalgae biomass and evaluated for total phenolic content (TPC) and in vitro antioxidant properties. Biomass was profiled for fatty acid methyl esters (FAME) composition. The DCM extracts had the highest levels of total phenolics, with values of 40.58 and 86.14 mg GAE/g dry weight (DW in IMA053 and IMA043, respectively). The DCM extracts had a higher radical scavenging activity (RSA) than the water and methanol ones, especially those from IMA043, with RSAs of 99.65% toward 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt (ABTS) at 10 mg/mL, and of 103.43% against 2,2-diphenyl-1-picrylhydrazyl (DPPH) at 5 mg/mL. The DCM extract of IMA053 displayed relevant copper chelating properties (67.48% at 10 mg/mL), while the highest iron chelating activity was observed in the water extract of the same species (92.05% at 10 mg/mL). Both strains presented a high proportion of saturated (SFA) and monounsaturated (MUFA) fatty acids. The results suggested that these microalgae could be further explored as sources of natural antioxidants for the pharmaceutical and food industry and as feedstock for biofuel production.

This work reports for the first time the nutritional profile, including proximate chemical composition, amino acids, fatty acids and minerals of Parastichopus regalis from the Mediterranean Sea (SE Spain). The studied species had a high moisture content, moderate protein and low lipid levels. The most abundant amino acids were glutamic acid, arginine and tyrosine. Polyunsaturated fatty acids, especially arachidonic acid, dominated the fatty acid profile. Iron, sodium, calcium and zinc were the most abundant mine rals. In general, P. regalis has a balanced nutritional quality suitable for human consumption.

The fatty acid (FA) composition of six macroalgae from the Cystoseira genus, namely Cystoseira compressa, Cystoseira humilis, Cystoseira tamariscifolia, Cystoseira nodicaulis, Cystoseira baccata and Cystoseira barbata, was determined. Polyunsaturated fatty acids (PUFA) corresponded to 29-46% of the total FA detected. C. compressa, C. tamariscifolia and C. nodicaulis stood out for their low PUFA/saturated fatty acid, low n-6 PUFA/n-3 PUFA ratios as well as favourable unsaturation, atherogenicity and thrombogenicity indices, suggesting a high nutritional value with potential applications in the nutraceutical industry.

Salt-tolerant plants, also known as halophytes, could provide a novel source of feedstock for biorefineries. After harvesting fresh shoots for food, the lignified fraction of Salicornia ramosissima J. Woods could be used to produce bioactive botanical extracts for high-value industries such as nutraceuticals, cosmetics, and biopharmaceuticals. The residual fraction after extraction can be further used for bioenergy or lignocellulose-derived platform chemicals. This work analysed S. ramosissima from different sources and growth stages. After pre-processing and extractions, the obtained fractions were analysed for their contents of fatty acids, pigments, and total phenolics. Extracts were also evaluated for their in vitro antioxidant properties and inhibitory effect towards enzymes related to diabetes, hyperpigmentation, obesity, and neurogenerative diseases. The ethanol extract from the fibre residue and the water extract from completely lignified plants showed the highest concentration of phenolic compounds along with the highest antioxidant potential and enzyme-inhibitory properties. Holothuria arguinensis aquaculture started to be developed in 2014, being the first sea cucumber species from Europe. However, some aspects of its aquaculture biotechnology, such diets, need to be assessed. This work aimed to evaluate seagrass debris of Zostera noltii and Cymodocea nodosa as food source for broodstock maintenance in tanks, during breeding periods. The given feed rations per tank were calculated as the 30% of the total sea cucumber biomass in each tank and reviewed each week. Then, feed rations of seagrass and sediment were calculated from this value, according to the following percentages: 40% sediment, 15% Z. noltii, 40% Z. noltii, 15% C. nodosa and 40% C. nodosa. H. arguinensis growth, feeding rate and nutritional value were assessed under these diets. H. arguinensis fed with 40% of Z. noltii showed the highest growth (specific growth rate = 0.09 ± 0.06%/day, absolute growth rate = 0.11 ± 0.07 g/day) increasing their final weight in 5.86 ± 3.57% in 57 days. However, the individuals fed with C. nodosa showed a negative growth. H. arguinensis showed a reduction in its feeding rate as the organic matter content in the diets increased. H. arguinensis did not show any important change on proximate composition, protein, lipid, mineral contents and fatty acids profile among the feeding groups, or in comparison with the individuals collected from wild habitat. Therefore, H. arguinensis could be fed with Z. noltii debris during tanks maintenance along breeding period, ensuring its growth and maintaining its nutritional profile. [ABSTRACT FROM AUTHOR]

This work makes a comparative evaluation of the biochemical profile of three sea commercial cucumber species (Holothuria mammata, H. polii and H. tubulosa) caught from different locations of the Mediterranean Sea (SE Spain). All species had high levels of moisture (from 73.6% in H. mammata to 81.2% in H. tubulosa), crude ash (from 9.61% in H. mammata to 14.7% in H. tubulosa) and protein (3.01% in H. tubulosa to 11.1% in H. mammata). They also had a low fat content, from 0.21% in H. tubulosa to 0.55% in H. mammata. Holothuria polii had intermediate values between the other two species, for all considered variables. All species had adequate protein/lipid ratios (H. mammata, 20:1; H. polii, 23:1; H. tubulosa, 14:1) and low lipid levels, enriched in omega-3 polyunsaturated fatty acids, especially arachidonic acid. The fatty acid profile suggests that H. polii is feeding on sediments more influenced by terrestrial inputs than the remaining species. Holothuria mammata and H. tubulosa are feeding on marine food sources mainly, but also with some terrestrial influence. The most abundant amino acids detected were alanine, arginine, glutamic acid, and glycine. All species had similar contents of essential amino acids (EAA) and ratios of EAA/non-essential amino acids. Holothuria tubulosa had a high content of toxic metals including Cr, Pb and Ni. Marine invertebrates contain nutritionally important elements that make them suitable to be included in the human diet, and they are considered an outstanding source of secondary metabolites with multiple biotechnological uses. The sea hareBursatella leachiiis an invasive species in Mar Menor that can be managed through the control of its reproduction and/or by massive biomass removal. In this context, this work evaluated the possible use of biomass fromB. leachiiin the food and cosmetics industries. Biomass was appraised for nutritional properties, and extracts were prepared and evaluatedin vitrofor antioxidant and anti-inflammatory capacity, cholinesterase and tyrosinase inhibition, and for total contents in different phenolic groups. It had a moderate moisture level, low ash and fat levels, and an adequate protein amount and protein/lipid ratio. The fatty acids (FA) profile was mainly saturated, dominated by palmitic acid. The methanol extract had the highest acetylcholinesterase inhibition. The acetone extract had the strongest capacity to scavenge 1,1-diphenyl-2 picrylhydrazyl (DPPH) radical, reduce iron, inhibit tyrosinase, decrease nitric oxide production in lipopolysaccharide (LPS)-stimulated microglia cells, and the upmost levels of total phenolics, flavonoids, and tannins. This work reports for the first time the nutritional profile and antioxidant potential of the edible sea cucumber Holothuria arguinensis from the North-eastern Atlantic. H. arguinensis has high levels of protein, with the amino acids profile dominated by alanine, glycine and proline and low lysine/arginine ratios. Its carbohydrate and energetic contents are also low as well as the total lipid levels, although its lipid profile is rich in polyunsaturated fatty acids (PUFA), especially arachidonic, eicosapentaenoic and docosahexaenoic acids. In addition, H. arguinensis has high levels of calcium. The water and ethanol extracts show ability to scavenge free radicals and to chelate copper and iron ions. Our results indicate that H. arguinensis has a balanced nutritional quality suitable for human consumption. The isolation of lipid-rich autochthonous strains of microalgae is a crucial stage for the development of a microalgae-based biofuel production plant, as these microalgae already have the necessary adaptations to withstand competition, predation and the temperatures observed at each production site. This is particularly important in extreme climates such as in Saudi Arabia. Resorting to fluorescence activated cell sorting (FACS) we screened for and isolated several microalgal strains from samples collected from the Red Sea. Relying on the fluorescence of BODIPY 505/515 (4,4-difluoro-1,3,5,7- tetramethyl-4-bora-3a,4a-diazasindacene) and growth performance, four promising candidates were identified and the total lipid content and fatty acid profile was assessed for biofuels production. Selected isolates were classified as chlorophytes, belonging to three different genera: Picochlorum, Nannochloris and Desmochloris. The lipid contents were assessed microscopically by means of BODIPY 505/515-associated fluorescence to detect intracellular lipid bodies, which revealed several lipid drops in all selected strains. This result was confirmed by lipid gravimetric determination, which demonstrated that all strains under study presented inner cell lipid contents ranging from 20% to 25% of the biomass dry weight. Furthermore, the fatty acid methyl esters profile of all strains seems ideal for biodiesel production due to a low degree of polyunsaturated fatty acid methyl esters and high amount of palmitic and oleic acids. Simple Summary: Marine invertebrates are a rich source of natural products with multiple commercial applications. However, little is known about the biotechnological potential of Sabella spallanzanii and Microcosmus squamiger. Aimed at filling this gap, this work evaluated such species for biochemical, antioxidant and metal chelating properties. Both species had interesting nutritional profiles in terms of crude protein and fat, among others. Regarding amino acids, S. spallanzanii and M. squamiger were found to be rich in arginine and taurine, respectively, while the most abundant minerals were sodium, calcium, and potassium, in both species. Methanol extracts of both species had antioxidant and metal chelating capacity. Fucoxanthinol and fucoxanthin were the major carotenoids in the M. squamiger dichloromethane extract. The fatty acid (FA) profile was strongly influenced by the type of extract. Saturated FA were higher than unsaturated ones in methanol extracts, while unsaturated FA prevailed in the dichloromethane extracts. Low n-6/n-3 ratios were obtained. Our results suggest both species as sustainable sources of bioactive products with several applications, for example, as food ingredients, as fish feed, and as ingredients for veterinary products. Sabella spallanzanii and Microcosmus squamiger were profiled for proximate composition, minerals, amino acids, fatty acids (FA), carotenoids, radical scavenging activity on the 2,2-diphenyl-1- picrylhydrazyl (DPPH) radical, oxygen radical absorbance capacity (ORAC) and iron and copper chelating properties. Microcosmus squamiger had the highest level of moisture and crude protein, S. spallanzanii was enriched in crude fat and ash. Both species had similar levels of carbohydrates and energy. There was a prevalence of arginine and glycine in S. spallanzanii, and of taurine in M. squamiger. The most abundant minerals in both species were Na, Ca, and K. The methanol extract of S. spallanzanii had metal chelating properties towards copper and iron, while the methanol extract of M. squamiger was able to chelate copper. M. squamiger extracts had similar ORAC values. Fucoxanthinol and fucoxanthin were the major carotenoids in the M. squamiger dichloromethane extract. Saturated FA were more abundant than unsaturated ones in methanol extracts, and unsaturated FA prevailed in the dichloromethane extracts. Palmitic acid was the predominant FA in methanol extracts, whereas eicosapentaenoic (EPA) and dihomo-γ-linolenic acids were the major compounds in dichloromethane extracts. Halophytic plants can provide an economical and environmentally sustainable source of products for human and animal feeding, in the context of the increase of worldwide emergent semi-arid landscapes. This work reports a comprehensive evaluation of the qualitative and quantitative composition of fatty acids (FA) of nineteen Mediterranean halophytes collected in southern Portugal, with the purpose of establishing their possible uses as food and feed. For FA determination, lipids and free FA were converted to the corresponding fatty acid methyl esters (FAME) and analyzed by GC-MS. Beta maritima had the highest FAME levels (7.3 mg/g DW) while Suaeda vera had the lowest content (1.0 mg/g DW). The most common saturated fatty acid (SFA) across all studied species was palmitic acid. The most prevalent monounsaturated fatty acid (MUFA) was oleic acid. Polyunsaturated fatty acids (PUFA) levels were led by linoleic acid. Less common FAMEs were also detected, namely eicosadienoic and hexadecatrienoic acids. Cotula coronopifolia, Phragmites australis and Suaeda vera displayed the best FA nutritional profiles. These species also showed bioactivities relevant for both human and animal health according to the literature and thus, collectively with this study, they could be further explored as food and feed.
