This study has investigated the effects of six different unialgal diets ( Chaetoceros calcitrans , Platymonas helgolandica , Chlorella sp., Isochrysis galbana , Nannochloropsis oculata , and Pavlova viridis ) on the composition of fatty acids and sterols in juvenile ark shell Tegillarca granosa Linnaeus. The best feeding effects on the growth of shellfish were found in C. calcitrans, followed by I. galbana and P. viridis, whereas Chlorella sp. and N. oculata exhibited relatively poor effects. The fatty acid and sterol compositions in the six microalgae and the juvenile ark shell after feeding were analyzed, and 39 fatty acids and 18 sterols were identified. Although the results demonstrate a close correlation between the sterol compositions in algal species and juvenile ark shell, a similar correlation was not observed between fatty acids. In the juvenile ark shell fed microalgae, the ratio of total saturated fatty acids (SFA) rapidly decreases, whereas the proportion of total polyunsaturated fatty acids (PUFAs) increases considerably. The abundances of AA, EPA, and DHA increase most significantly in shellfish with better growth (fed C. calcitrans, I. galbana, and P. viridis). The number of sterol species is reduced, but the total sterol content in groups fed corresponding microalgae increases, and abundant plant sterols, instead of cholesterol, are accumulated in juvenile ark shell fed appropriate microalgae I. galbana and P. viridis. Therefore, to be more conducive to human health, I. galbana and P. viridis, of the six experimental microalgae, are recommended for artificial ark shell culture.

Ghrelin, glucagon-like peptide-1 (GLP-1), and peptide YY (PYY) are potent hormones mediating food intake according to the nutritional status in fish. However, limited information is available on these genes and their expression in response to nutrition in silver pomfret (Pampus argenteus). A comparison analysis revealed that ghrelin and GLP-1 were relatively conserved in marine fish. PYYa and PYYb shared a low identity and were clustered to different PYY branches. Ghrelin, pyya, and pyyb mRNAs were highly expressed in the brain, while glp-1 was highly expressed in the gills and liver. The ghrelin mRNA expression was relatively high an hour post-feeding and decreased after 3 to 72 h of fasting. The glp-1 mRNA expression was increased after 6 to 24 h of fasting. The pyya mRNA expression was increased after 72 h of fasting, while the pyyb expression remained stable during fasting. The diet with 14% lipid promoted the expressions of glp-1, pyya, and pyyb, but inhibited the ghrelin expression. The ghrelin expression in the intestine was increased after palmitic acid, oleic acid (OA), linoleic acid (LA), α-linolenic acid, eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA) incubation. OA decreased the expression of glp-1 and increased the expression of pyya. The expression of pyyb was downregulated by LA, DHA, and EPA. These results suggest the importance of ghrelin, glp-1, and pyy in coordinating food intake in response to fasting, dietary lipid concentration, and fatty acids in silver pomfret. Schizochytrium sp. is a kind of heterotrophic protist, rich in docosahexaenoic acid (DHA). To study the application value of Schizochytrium sp. in fish diet, a 20-day feeding experiment was initiated to evaluate its effect on growth, fatty acid contents, and the gut microbiota of the important economic fish, silver pomfret. In this study, the diets of the treatment group were added with 2% Schizochytrium sp. The fish in the treatment group gained more weight and had a higher growth rate than the control group. The levels of DHA, eicosapentaenoic acid, and polyunsaturated fatty acids in the edible tissue of fish were highly increased in the treatment group after 20 days. After feeding Schizochytrium sp., high-throughput 16S rRNA sequencing showed that the Proteobacteria, Firmicutes and Bacteroidetes were the phyla with the highest abundance, and at the genus and species level, we observed an increased abundance of probiotics. The results indicated that ingestion of Schizochytrium sp. could change the dominant microbiota population, which might lead to accelerated growth and improved unsaturated fatty acid content and fish health. This study investigated the expression of zebrafish (Danio rerio) fatty acid transporters, such as scavenger receptor CD36, plasma membrane fatty acid‐binding protein (FABPpm), and fatty acid transport protein family (FATPs), in responses to nutritional status and diet composition. For diet composition study, three isonitrogenous (43% crude protein) purified diets were formulated to contain 6%, 12% and 18% crude lipid levels. Meanwhile five isonitrogenous and isoenergetic purified diets were formulated to contain different lipid sources including fish oil (FO; rich in n‐3 HUFA), palmitic acid (PA; rich in C16:0), olive oil (OO; rich in C18:1n‐9), sunflower oil (SO; rich in C18:2n‐6) or perilla oil (PO; rich in C18:3n‐3) as the sole lipid source. Results showed that liver CD36, FABPpm‐a, FABPpm‐b, FATP1‐a and FATP6, intestine CD36, FABPpm‐a and FABPpm‐b, and muscle CD36 and FATP1‐b were significantly increased during postprandial period. During starvation, liver CD36, FABPpm‐a, FABPpm‐b and FATP1‐a, intestine FABPpm‐a, FABPpm‐b, FATP4 and FATP6, and muscle FATP1‐b, FATP4 and FATP6 were significantly increased. The expression of some fatty acid transporters (including liver CD36 and FABPpm‐b, intestine FABPpm‐a, and muscle CD36, FABPpm‐a, FATP1‐a, FATP2 and FATP4) was up‐regulated by refeeding, while the expression of other fatty acid transporters (liver FABPpm‐a and intestine FATP1‐a, FATP4 and FATP6) was down‐regulated by refeeding. The expression of several fatty acid transporters (liver FATP2 and FATP4, and intestine CD36, FATP2 and FATP4) was significantly increased by high‐lipid diet, whereas CD36, FATP1‐b, FATP2 and FATP4 expression in the muscle were reduced by increasing dietary lipid level. Compared with FO group, the muscle of fish fed the diet with OO had a higher CD36 mRNA abundance, while the muscle of fish fed the diet with OO and PO had a higher FATP1‐b expression. Compared with the FO group, the hepatic FATP2 mRNA was significantly increased in the PA group, while the intestine FATP2 mRNA was significantly increased in the SO and PO groups. In addition, intestine FATP4 expression in the PA and OO groups was lower compared with the FO group. The results indicate that zebrafish fatty acid transporters play a central role in coordinating the mobilization of fuel substrates in responses to nutritional status and diet composition. A Gram-negative, motile, rod-shaped, aerobic bacterial strain with tufty polar flagella, JLT1363T, was isolated from the South China Sea. The bacteria formed yellow colonies on rich organic medium. The major cellular fatty acids present in JLT1363T were C18:1 ω7c and/or C18:1 ω6c (36.06%), C17:1 ω6c (17.04%), C14:0 2-OH (9.85%), and C16:0 (8.09%). The genome size was ~3.12 Mbps with a G+C content of 64.9%. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain JLT1363T fell within the genus Erythrobacter. The 16S rRNA gene sequence similarity between strain JLT1363T and the type strains of Erythrobacter species ranged from 95.0% (with Erythromicrobium ramosum) to 98.7% (with Erythrobacter lutimaris). The Average Nucleotide Identity (ANI) value between genome sequences of strain JLT1363T and Erythrobacter lutimaris KCTC 42109T was 82.2%. Strain JLT1363T lacked bacteriochlorophyll a, and the major polar lipids were sphingoglycolipids, diphosphatidylglycerol, phosphatidylcholine, and phosphatidylglycerol. Phylogenetic and phenotypic properties indicated that strain JLT1363T represents a novel species of the genus Erythrobacter, for which the name Erythrobacter nanhaiensis sp. nov. is proposed. Pyropia has a unique heteromorphic life cycle with alternation stages between thallus and conchocelis, which lives at different water temperatures in different seasons. To better understand the different adaptation strategies for temperature stress, we tried to observe comparative biochemical changes of Pyropia haitanensis based on a short term heat shock model. The results showed that: (1) At normal temperature, free-living conchocelis contains significantly higher levels of H2O2, fatty acid-derived volatiles, the copy number of Phrboh and Phhsp70 genes,the activities of NADPH oxidase and floridoside than those in thallus. The released H2O2 and NADPH oxidase activity of conchocelis were more than 7 times higher than those of thallus. The copy number of Phrboh in conchocelis was 32 times that in thallus. (2) After experiencing heat shock at 35°C for 30 min, the H2O2 contents, the mRNA levels of Phrboh and Phhsp70, NADPH oxidase activity and the floridoside content in thallus were all significantly increased. The mRNA levels of Phrboh increased 5.78 times in 5 min, NADPH oxidase activity increased 8.45 times in 20 min. (3) Whereas, in conchocelis, the changes in fatty acids and their down-stream volatiles predominated, significantly increasing levels of saturated fatty acids and decreasing levels of polyunsaturated fatty acids occurred, and the 8-carbon volatiles were accumulated. However, the changes in H2O2 content and expression of oxidant-related genes and enzymatic activity were not obvious. Overall, these results indicate that conchocelis maintains a high level of active protective apparatus to endure its survival at high temperature, while thallus exhibit typical stress responses to heat shock. The serum polar lipid metabolic changes for two common cage-cultured fishes, yellow coraker Pseudosciaena crocea and Japanese seabass Lateolabrax japonicus, after tropical storm attack have been studied by ultra-performance liquid chromatography-quadrupole-time of flight mass spectrometry (UPLC-qTOF-MS). The full scan mass spectrometry combined with principal component analysis (PCA) and orthogonal projections to latent structures discriminant analysis (OPLS-DA) indicated that yellow croaker underwent significant chemico-physiological changes during the recovery process, whereas Japanese seabass did not show such noticeable time-dependent consistent metabolites change patterns. Further identification of the metabolite biomarkers showed the increase of phosphatidylcholine with high unsaturated fatty acid and lysophospholipids, and the decrease of phosphatidylcholine with saturated fatty acids and plasmologens, which indicated the need of energy supplement and successive stressful inflammation. The increase of taurocholic acid and decrease of cortol could be regarded as the physiological alleviation measure during the recovery period. This is the first metabolomic study to tackle the fish physiological response for the complex environmental changes, and demonstrated that lipidomics is an effective analytical tool for predicting the stress resistance of fish to ultra uncontrolled environmental stress. The razor clam Sinonovacula constricta is known for its richness in long-chain polyunsaturated fatty acids (LC-PUFA, C ≥ 20). Previously, we demonstrated that it possesses a complete LC-PUFA biosynthetic pathway. However, the mechanisms by which it senses the LC-PUFA pool to regulate their biosynthesis remain unclear. Here, we presented the LC-PUFA sensor UBXD8 as a critical molecule in this intriguing process. The S. constricta UBXD8 (Sc UBXD8) shared all characteristic features of its mammalian counterpart and exhibited high mRNA levels in digestive tissues, suggesting its functional role in this bivalve species. By purification of Sc UBXD8 protein in vitro , we discovered its ability to sense unsaturated fatty acids (UFA, C ≥ 14) but not saturated ones, as evidenced by polymerization detection. Furthermore, the intensity of Sc UBXD8 polymerization increased progressively with longer acyl chain lengths, greater unsaturation degrees, and higher UFA concentrations. Exceptionally, for those located at the same node in LC-PUFA biosynthetic pathway, Sc UBXD8 displayed a stronger sensitivity to n-6 UFA compared to n-3 UFA. These results suggested a critical role for Sc UBXD8 in balancing fatty acids composition and ratio of n-6/n-3 UFA in S. constricta. Moreover, the UAS domain was confirmed essential for Sc UBXD8 polymerization. Through knockdown of Sc Ubxd8 gene in vivo , there were significant shifts in expression patterns of genes related to LC-PUFA biosynthesis, concurrently influencing fatty acids compositions. These results suggested that Sc UBXD8 likely plays a regulatory role in LC-PUFA biosynthesis, possibly through the INSIG-SREBP pathway. Understanding the mechanisms of lipid transport and metabolism in fish is crucial to enhance dietary lipid utilization. Here, fatty acid translocase (CD36) gene was characterized in silver pomfret (Pampus argenteus). The open reading frame of silver pomfret cd36 gene was 1395 bp, encoding 464 amino acids. The silver pomfret CD36 protein contained typical transmembrane regions and N-glycosylation modification sites, and was localized to the cytomembrane. The cd36 gene was ubiquitously expressed in all tested tissues, with the highest expression observed in brain tissue. In vivo , both fasting and short-term high-fat feeding could increase cd36 expression in intestinal tissue. In vitro , cd36 expression was induced by palmitic acid, oleic acid, linolenic acid, eicosapentaenoic acid (EPA), and docosahexaenoic acid treatment in intestinal tissue. Furthermore, dual-luciferase reporter assay results indicated that peroxisome proliferator-activated receptor gamma (PPARγ) could enhance cd36 promoter activity, and the co-expression of cd36 and pparγ was observed in EPA-incubated intestine, suggesting that EPA may regulate the expression of cd36 via PPARγ to maintain the homeostasis of intestinal lipid metabolism in silver pomfret. These results highlighted the crucial role of CD36 in silver pomfret, and suggested that the cd36 expression may be regulated by PPARγ. As an unusual economically important aquaculture species, Sinonovacula constricta possesses high levels of long-chain polyunsaturated fatty acids (LC-PUFA). Previously, our group identified fatty acyl desaturases (Fad) with Δ5 and Δ6 activities in S. constricta , which was the first report of Δ6 Fad in a marine mollusc. Here, we further successfully characterize elongases of very long-chain fatty acids (Elovl) in this important bivalve species, including one Elovl2/5, two Elovl4 isoforms (a and b) and a novel Elovl (c) with Elovl4 activity. In addition, we also determined the desaturation activity of S. constricta Δ6 Fad toward 24:5n-3 to give 24:6n-3, a key intermediate in docosahexaenoic acid (DHA) biosynthesis. Therefore, S. constricta is the first marine mollusc reported to possess all Fad and Elovl activities required for LC-PUFA biosynthesis via the 'Sprecher pathway'. This finding greatly increases our understanding of LC-PUFA biosynthesis in marine molluscs. Phytoplankton composition is an important factor affecting the growth and physiological biochemical characteristics of filter-feeding bivalves. With the increasing trend in dinoflagellate biomass and blooms in mariculture areas, how the physio-biochemical traits and seafood quality of the mariculture organism are affected by the dinoflagellates, especially those at nonfatal levels, is not well understood. Different densities of two Karlodinium species, namely K. veneficum (KV) and K. zhouanum (KZ), mixed with high quality microalgal food Isochrysis galbana was applied in feeding manila clam Ruditapes philippinarum in a 14-day temporary culture, to comparatively study how the critical biochemical metabolites such as glycogen, free amino acids (FAAs), fatty acids (FAs), volatile organic compounds (VOCs) in the clam were affected. The survival rate of the clam showed dinoflagellate density and species specificity. The high-density KV group inhibited survival to 32% lower than that of the pure I. galbana control, respectively, while KZ at low concentrations did not significantly affect the survival compared with the control. In the high-density KV group, the glycogen and FAA contents decreased (p < 0.05), indicating that energy and protein metabolism were significantly affected. Amount of carnosine (49.91 ± 14.64 to 84.74 ± 8.59 μg/g of muscle wet weight) was detected in all the dinoflagellate-mixed groups, while it was not present in the field samples or in the pure I. galbana control, showing that carnosine participated in the anti-stress activities when the clam was exposed to the dinoflagellates. The global composition of FAs did not significantly vary among the groups. However, contents of the endogenous C 18 PUFA precursors linoleic acid and α-linolenic acid significantly decreased in the high-density KV group compared to all the other groups, indicating that high density of KV affected the metabolisms of fatty acids. From the results of the changed VOC composition, oxidation of fatty acids and degradation of free amino acids might occur in the clams exposed to dinoflagellates. The increased VOCs, such as aldehydes, and decreased 1-octen-3-ol probably produced a more fishy taste and reduced food flavor quality when the clam was exposed to the dinoflagellates. This present study demonstrated that the biochemical metabolism and seafood qulity of the clam were affected. However, KZ with moderate density in the feed seemed to be beneficial in aquaculture for increasing the content of carnosine, a high-valued substance with multiple bioactivities. Fatty acid and sterol changes in razor clam Sinonovacula constricta (Lamarck 1818) reared at different salinities were investigated. In one experiment, juvenile S. constricta with shell length of 1.63 ± 0.11 mm were firstly cultured at 13 psu for 10 days, and then cultured at 8, 13, 18, and 23 psu for another 5 days. In another experiment, adult S. constricta with shell length of 15.54 ± 0.80 mm were cultured at 13 and 23 psu for 40 days. On the 25th day, half of the S. constricta cultured at 13 psu were transferred to 23 psu for another 15 days. The results showed that higher salinity significantly decreased growth rate and crude lipid content in S. constricta . The proportions of long chain-polyunsaturated fatty acids (LC-PUFAs, C ≥ 20, double bonds ≥ 3) were significantly increased in S. constricta cultured at higher salinity as compared with those at lower salinity. The proportion of cholesterol was firstly increased and then significantly decreased with increasing salinity. The proportion of brassicasterol was higher in S. constricta cultured at higher salinity, while proportions of other plant sterols showed no obvious change trends. Interestingly, when S. constricta were around 60 days old with shell length of 32.06 ± 1.13 mm, the inhibitory effect of high culture salinity on growth rate was weakened, but the nutritional value was significantly improved. The results of this study suggest an optimal culture mode for S. constricta , i.e. if S. constricta were firstly cultured at lower salinity (10–15 psu) and then exposed to higher salinity (20–25 psu) for a short period of time before harvest, a combination of both optimal production and nutritional value would be achieved. The successful care and breeding of intertidal shellfish is determined by the variety and quality of the microalgae used as feed. Practice has found obvious feeding selectivity in different intertidal shellfish for diet microalgae, so screening for microalgal species with high food value is a priority for research related to the culture of hatchery shellfish. In this work, the effects of four different microalgae, Chaetoceros calcitrans , Isochrysis galbana , Nannochloropsis oculata and Diacronema viridis , on the growth of juvenile ark shells ( Tegillarca granosa Linnaeus) was studied. The microalgal diets were applied in three different types of treatments: a mixture of all four microalgae, single species, and binary combinations of C. calcitrans or I. galbana with the other individual species. The best feeding effects were observed with the mixture of all four microalgae, and binary algal diets were second best. When fed with single-species diets, the most favorable results were observed with C. calcitrans followed by I. galbana . Single-species diets of D. viridis or N. oculata were the least beneficial. Fatty acid and sterol compositions of the microalgae and juvenile ark shells were analyzed by gas chromatography–mass spectrometry (GC–MS) after feeding, and 32 fatty acids and 12 sterols were identified. The results of a principal components analysis (PCA) of the fatty acid and sterol compositions in juvenile ark shells, using the SIMCA-P + software package, revealed that these contents were affected by their microalgal diets. In the PCA score scatter plots for the first and third components, the samples fed with the full mixture of microalgae were grouped near the samples fed with binary-species diets of C. calcitrans and I. galbana . The samples fed with binary-species diets of C. calcitrans and N. oculata or D. viridis were located near the samples fed with only C. calcitrans , and the samples fed with binary-species diets of I. galbana and N. oculata or D. viridis were near the samples fed with only I. galbana . The samples fed with only N. oculata or D. viridis were located far from the other samples. These results demonstrate that the lipid compositions of juveniles fed with binary-species diets were similar to those of the shells fed with single microalgae, which had a more favorable feeding effect on the shell. This study highlights the feeding selectivity of the shells, and the results shown in the PCA scatter plot based on the fatty acid and sterol compositions demonstrate that the high-quality microalgae candidates for use in hatchery culture can be selected by comparing samples fed with single and mixed microalgae. Statement of relevance This study demonstrated that the fatty acids and sterols of the shellfish were largely determined by its diet microalgae. Based on the characteristics of the lipid composition of the microalgae candidates, by using GC–MS and principal components analysis (PCA), a precise method can be applied in shellfish hatchery culture when screening the potential microalgal diets of high quality. Plant invasions in coastal ecosystems may substantially alter benthic community composition and food web structure. We combined analyses of stable isotopes and fatty acids (FA) to examine the effects of two non-indigenous plants (Spartina alterniflora and Kandelia candel) on the food sources of three deposit feeders (Assiminea brevicula , Cerithidea cingulata and Uca arcuata) and a carnivorous opportunistic predator (Periophthalmus modestus) in Yueqing Bay, China. A wide range of differences was found in δ13C and δ15N among the macrobenthos. The deposit feeders in K. candel mangrove forest (KC) were markedly more 13C-depleted than those in S. alterniflora marsh (SA) and unvegetated tidal flat (TF), unlike the carnivorous feeder. PERMANOVA showed that FA composition significantly differed among the macrobenthos. Diatom FA markers were higher in tissues of the deposit feeders than in the carnivorous feeder, and slightly lower in macrobenthos from the KC and SA habitats as compared with the TF. In contrast, vascular plant FA markers of macrobenthos were higher in the KC and SA than in the TF, indicating the contribution of mangrove and Spartina detritus to the consumers. Sinonovacula constricta is the first marine mollusc demonstrated to possess all fatty acyl desaturase (Fad) and elongase of very long chain fatty acids (Elovl) activities required for long-chain polyunsaturated fatty acids (LC-PUFA) biosynthesis via the Sprecher pathway, providing a good opportunity to investigate the essential fatty acids (FA) for its larval development. In the present study, we analyzed the Fad and Elovl expressions, FA compositions, and the feed effects of three representative microalgae (Chlorella sp. , Chaetoceros calcitrans and Isochrysis galbana) in S. constricta at early developmental stages. The results showed that S. constricta Fad and Elovl were all highly expressed in zygotes, which might occur from maternal transfer. Compared to transcripts of Elovl4 activity, Elovl2/5 was only expressed relatively high in zygotes, indicating that C ≥ 22 LC-PUFA were more important than C ≤ 20 LC-PUFA for S. constricta larval development. The zygotes accumulated abundant FA from parents, especially eicosapentaenoic acid (EPA), but with small amount of docosahexaenoic acid (DHA). Meanwhile, the trochophore larva fed with single or mixed microalgae with I. galbana (rich in DHA) exhibited significantly higher growth and survival rate compared to those fed with single Chlorella sp. (rich in 18:2n-6 and 18:3n-3) and C. calcitrans (rich in EPA) or both of them. Those findings suggested that the highly expressed Fad and Elovl might be largely responsible for DHA biosynthesis, but this capability was limited to meet the developmental requirement and the dietary DHA was necessary.
