This visual feedback may be scaled or otherwise distorted by unpredictable perturbations. Although adjustments to visual feedback scaling have been studied before, the underlying brain activation differences between upscaling (visual feedback larger than real movement) and downscaling (feedback smaller than real movement) are currently unknown. Brain activation differences between upscaling and downscaling might be expected because within-trial adjustments during upscaling require corrective backwards accelerations, whereas correcting for downscaling requires forward accelerations. In this behavioural and fMRI study we investigated adjustments during up- and downscaling in a target-directed finger flexion-extension task with real-time visual feedback. We found that subjects made longer and more complete within-trial corrections for downscaling perturbations than for upscaling perturbations. The finger task activated primary motor (M1) and somatosensory (S1) areas, premotor and parietal regions, basal ganglia, and cerebellum. General scaling effects were seen in the right pre-supplementary motor area, dorsal anterior cingulate cortex, inferior parietal lobule, and dorsolateral prefrontal cortex. Stronger activations for down- than for upscaling were observed in M1, supplementary motor area (SMA), S1 and anterior cingulate cortex. We argue that these activation differences may reflect differing online correction for upscaling versus downscaling during finger flexion-extension. 13 p.

Peripheral encoding of movement kinematics has been well-characterized, but there is little understanding of the relationship between movement kinematics and associated brain activation. We hypothesized that kinematics of passive movement is differentially represented in the sensorimotor network, reflecting the well-studied afferent responses to movement. A robotic forefinger manipulandum was used to induce passive kinematic stimuli and monitor interaction force in 41 healthy participants during whole-brain functional magnetic resonance imaging (fMRI). Levels of forefinger displacement amplitude and velocity were presented in flexion and extension. Increases in velocity were linearly associated with activation in contralateral primary somatosensory cortex (S1), bilateral secondary somatosensory cortex (S2), primary motor cortex, and supplementary motor area. No difference in activation was found for direction of the finger movement. Unexpectedly, S1 and S2 activation decreased nonlinearly with increasing displacement amplitude. We conclude that while straightforward relations were found with velocity, the complex neural representation of displacement amplitude suggests a more nuanced relationship between peripheral responses to kinematic stimuli and sensorimotor network activity. Here we present a novel, systematic characterization of the whole-brain response to passive movement kinematics.

In neurorehabilitation, longitudinal assessment of arm movement related brain function in patients with motor disability is challenging due to variability in task performance. MRI-compatible robots monitor and control task performance, yielding more reliable evaluation of brain function over time. The main goals of the present study were first to define the brain network activated while performing active and passive elbow movements with an MRI-compatible arm robot (MaRIA) in healthy subjects, and second to test the reproducibility of this activation over time. For the fMRI analysis two models were compared. In model 1 movement onset and duration were included, whereas in model 2 force and range of motion were added to the analysis. Reliability of brain activation was tested with several statistical approaches applied on individual and group activation maps and on summary statistics. The activated network included mainly the primary motor cortex, primary and secondary somatosensory cortex, superior and inferior parietal cortex, medial and lateral premotor regions, and subcortical structures. Reliability analyses revealed robust activation for active movements with both fMRI models and all the statistical methods used. Imposed passive movements also elicited mainly robust brain activation for individual and group activation maps, and reliability was improved by including additional force and range of motion using model 2. These findings demonstrate that the use of robotic devices, such as MaRIA, can be useful to reliably assess arm movement related brain activation in longitudinal studies and may contribute in studies evaluating therapies and brain plasticity following injury in the nervous system.

Isometric compensation of predictably frequency-modulated low forces is associated with corticomuscular coherence (CMC) in beta and low gamma range. It remains unclear how the CMC is influenced by unpredictably modulated forces, which create a mismatch between expected and actual sensory feedback. We recorded electroencephalography from the contralateral hand motor area, electromyography (EMG), and the motor performance of 16 subjects during a visuomotor task in which they had to isometrically compensate target forces at 8% of the maximum voluntary contraction with their right index finger. The modulated forces were presented with predictable or unpredictable frequencies. We calculated the CMC, the cortical motor alpha-, beta-, and gamma-range spectral powers (SP), and the task-related desynchronization (TRD), as well as the EMG SP and the performance. We found that in the unpredictable condition the CMC was significantly lower and associated with lower cortical motor SP, stronger TRD, higher EMG SP, and worse performance. The findings suggest that due to the mismatch between predicted and actual sensory feedback leading to higher computational load and less stationary motor state, the unpredictable modulation of the force leads to a decrease in corticospinal synchrony, an increase in cortical and muscle activation, and a worse performance.

Background. In healthy subjects, observation of actions activates a motor network similar to that involved in the performance of the observed actions. Movement observation could perhaps be applied to functionally sustain brain motor functions when efferent motor commands and proprioceptive feedbacks are disconnected. Objective. The authors examined whether observation-induced brain activation is preserved in people with chronic complete spinal cord injury (SCI). Methods. Nine patients and 12 healthy subjects underwent behavioral assessment and functional magnetic resonance imaging. The SCI patients attempted to perform dorsal and plantar flexions of the right foot, and the controls also executed the same movement. Both groups observed subsequent video clips showing the same movement. Results. In the SCI group, attempted and observed foot movements activated a common observation–execution network including ventral premotor, parietal cortex, and cerebellum as in healthy subjects. Conclusions. Long after onset of complete SCI, the brain maintains its ability to respond to task-specific visual inputs, which suggests preservation of motor programs. This persistence might be a prerequisite for repair strategies of the spinal cord that rely on appropriate activation of the brain to try to restore limb function. The preserved cortical network may offer an additional motor rehabilitation approach for people with SCI.

The objective of this study is to better understand the role of proprioception in handwriting and test earlier conclusions stating that the automated shaping of letters was not impaired by the removal of visual control in deafferentation. To this aim we compared the performance of the deafferented patient GL, who suffers from a complete loss of cutaneous and proprioceptive sensation, with that of eight healthy age- and sex-matched subjects. The word "Parallele", written within a short sentence with and without visual control, was quantified using a digital writing tablet. Three of the 13 analyzed parameters were strikingly different in patient GL compared to healthy subjects, both with and without vision: increase of number of pen touches, increase in number of inversions in velocity, and decrease of mean stroke frequency. The changes in these three parameters indicate a strong impairment in automated behaviour in the absence of proprioception and touch. This impairment is also supported by the significantly longer movement duration, which is also significantly increased by the removal of visual control. The present study provides for the first time a quantification of handwriting in a completely deafferented patient and reveals the central role of proprioception for the storage, updating, and maintenance of skilled motor programs. The fact that the deficits are already present with visual feedback suggests that the role of vision in handwriting is only secondary.

Recently, we studied corticomuscular coherence (CMC) in a visuomotor task and showed for the first time gamma-range (30–45 Hz) CMC during isometric compensation of a periodically modulated dynamic force. We speculated that for the control of such forces, the sensorimotor system resonates at gamma-range frequencies to rapidly integrate the visual and proprioceptive information and produce the appropriate motor command. In this study, we tested the role of the proprioceptive afferent feedback on gamma-range CMC by comparing the deafferented patient GL to six age- and sex-matched subjects during the performance of a visuomotor force task consisting of isometric compensation of static and dynamic forces applied on the finger. Patient GL presented no significant gamma-band CMC during dynamic force. Instead, she had only beta-range CMC as in the static force condition; concurrently, her performance was significantly worse than that of the controls in both conditions. This gives support to the conclusions of our previous paper and suggests that proprioceptive information is mandatory in the genesis of gamma-band CMC during the generation and control of dynamic forces.

Phantom limbs are traditionally conceptualized as the phenomenal persistence of a body part after deafferentation. Previous clinical observations of subjects with phantoms of congenitally absent limbs are not compatible with this view, but, in the absence of experimental work, the neural basis of such “aplasic phantoms” has remained enigmatic. In this paper, we report a series of behavioral, imaging, and neurophysiological experiments with a university-educated woman born without forearms and legs, who experiences vivid phantom sensations of all four limbs. Visuokinesthetic integration of tachistoscopically presented drawings of hands and feet indicated an intact somatic representation of these body parts. Functional magnetic resonance imaging of phantom hand movements showed no activation of primary sensorimotor areas, but of premotor and parietal cortex bilaterally. Movements of the existing upper arms produced activation expanding into the hand territories deprived of afferences and efferences. Transcranial magnetic stimulation of the sensorimotor cortex consistently elicited phantom sensations in the contralateral fingers and hand. In addition, premotor and parietal stimulation evoked similar phantom sensations, albeit in the absence of motor evoked potentials in the stump. These data indicate that body parts that have never been physically developed can be represented in sensory and motor cortical areas. Both genetic and epigenetic factors, such as the habitual observation of other people moving their limbs, may contribute to the conscious experience of aplasic phantoms.

Background. Neurorehabilitation interventions to improve lower limb function and neuropathic pain have had limited success in people with chronic, incomplete spinal cord injury (iSCI). Objective. We hypothesized that intense virtual reality (VR)–augmented training of observed and executed leg movements would improve limb function and neuropathic pain. Methods. Patients used a VR system with a first-person view of virtual lower limbs, controlled via movement sensors fitted to the patient’s own shoes. Four tasks were used to deliver intensive training of individual muscles (tibialis anterior, quadriceps, leg ad-/abductors). The tasks engaged motivation through feedback of task success. Fourteen chronic iSCI patients were treated over 4 weeks in 16 to 20 sessions of 45 minutes. Outcome measures were 10 Meter Walking Test, Berg Balance Scale, Lower Extremity Motor Score, Spinal Cord Independence Measure, Locomotion and Neuropathic Pain Scale (NPS), obtained at the start and at 4 to 6 weeks before intervention. Results. In addition to positive changes reported by the patients (Patients’ Global Impression of Change), measures of walking capacity, balance, and strength revealed improvements in lower limb function. Intensity and unpleasantness of neuropathic pain in half of the affected participants were reduced on the NPS test. Overall findings remained stable 12 to 16 weeks after termination of the training. Conclusions. In a pretest/posttest, uncontrolled design, VR-augmented training was associated with improvements in motor function and neuropathic pain in persons with chronic iSCI, several of which reached the level of a minimal clinically important change. A controlled trial is needed to compare this intervention to active training alone or in combination.

Electromyographic (EMG) activity was analyzed for the occurrence of synergistic patterns during the steady hold periods of force in the precision grip. To establish the presence of muscle synergies in the amplitude (spatial) domain, the EMG activation levels of pairs of simultaneously active muscles were linearly correlated. Cross-correlations of EMG activity were computed to quantify muscle synergies in the spatiotemporal domain (synchronization). A muscle pair was defined to be synergistically coupled or synchronously activated when the correlation (amplitude domain) or cross-correlation (time domain) was significant for at least two of the three steady state force levels. Muscle synergies in the amplitude domain were found in one-third of the 213 muscle pairs tested, distributed among 47 of the 82 tested muscle combinations. Coactivation was the predominant synergistic pattern, whereas trade-off comprised not more than 23% of the synergies. Cross-correlation peak size varied between 5 and 39% of the autocorrelation size, with delays in the range of +/- 8 ms and base width between 12 and 20 ms. Synchronization was found in one-fourth of the 213 muscle pairs tested and among 35 of the 82 muscle combinations, i.e., less frequently than covariation of EMG activity levels. However, the interindividual prevalence was higher for synchronization than for synergies in the amplitude domain, since, for the synergistic muscle combinations, almost twice as many muscle pairs were found to be synchronized than coupled in the amplitude domain. Synergies in the two domains occurred independently in some pairs and concurrently in other cases, and were observed between muscles moving the thumb, the index finger, or both digits. Synchronization was more frequent in pairs of muscles supplied by branches of the same peripheral nerve (46%) than in those innervated by different nerves (18%). Synergies in the amplitude domain were distributed in similar proportions across intrinsic, extrinsic, and combinations of both types of muscles, whereas synchronization mainly occurred in pairs of intrinsic muscles. When the task was repeated with slightly lower target forces, there were fewer synergies in the amplitude domain (in 52 of the 213 pairs, distributed among 35 of 82 muscle combinations) and their distribution changed, indicating a flexible, force-dependent mechanism. In conclusion, no strictly coherent interindividual pattern of synergies in the spatial domain could be established.

Electromyographic (EMG) activity was examined in six normal subjects, producing low isometric forces between thumb and index finger in a visually guided step-tracking task. Target forces ranged between 0.5 and 3.0 N. EMG activity of all 15 muscles acting on thumb or index finger was screened with simultaneous recordings of up to 8 muscles. Linear regression was applied to quantify the EMG activity as a function of force. The intrinsic muscles and the long flexors of the index finger had a tight relation to force, as indicated by the high correlation coefficient, as did the adductor and short flexor of the thumb. In contrast, the long extensors of the index finger did not show force-related activity. The other muscles, including the long flexor and extensor of the thumb, had varying, on average moderate, correlations to force. The slope of the regression lines, a measure for the amount of EMG modulation with increasing force, revealed the same trends. Thus the majority of the intrinsic muscles were as closely related to force as the long flexors, suggesting a more important role in production of low isometric forces in the grip than previously believed, perhaps even a primary role. Systematic interindividual differences were rarely observed. Analysis of the trial-by-trial variability of EMG activity revealed that for most muscles the observed scatter was produced by varying background activity and was not a random fluctuation of relative increases in activity from one force level to the next.

The introduction of functional neuroimaging has resulted in a profusion of knowledge on various topics, including how blood oxygenation level dependent (BOLD) signal in the brain is related to force. To date, studies that have explicitly examined this relationship have used block designs. To gain a better understanding of the networks involved in human motor control, analyses sensitive to temporal relationships, such as Granger Causality or Dynamic Causal Modeling, require event-related designs. Therefore the goal of this experiment was to examine whether similar or even better relationships between BOLD and force during precision grip could be determined with an event-related design. Five healthy subjects exerted forces at 10%, 20% and 30% of maximum voluntary force, along with an observation condition. We report that the BOLD signal was linearly correlated with precision grip force in primary sensorimotor cortex and cerebellum, showing slightly better correlations than previous work. The results provide a clearer picture regarding the sensitivity of BOLD signal to force and show that event-related designs can be more appropriate than block designs in motor tasks.

Several studies about noise-enhanced balance control in humans support the hypothesis that stochastic resonance can enhance the detection and transmission in sensorimotor system during a motor task. The purpose of the present study was to extend these findings in a simpler and controlled task. We explored whether a particular level of a mechanical Gaussian noise (0–15 Hz) applied on the index finger can improve the performance during compensation for a static force generated by a manipulandum. The finger position was displayed on a monitor as a small white point in the center of a gray circle. We considered a good performance when the subjects exhibited a low deviation from the center of this circle and when the performance had less variation over time. Several levels of mechanical noise were applied on the manipulandum. We compared the performance between zero noise (ZN), optimal noise (ON), and high noise (HN). In all subjects (8 of 8) the data disclosed an inverted U-like graph between the inverse of the mean variation in position and the input noise level. In other words, the mean variation was significantly smaller during ON than during ZN or HN. The findings suggest that the application of a tactile-proprioceptive noise can improve the stability in sensorimotor performance via stochastic resonance. Possible explanations for this improvement in motor precision are an increase of the peripheral receptors sensitivity and of the internal stochastic resonance, causing a better sensorimotor integration and an increase in corticomuscular synchronization.

Previous studies investigating the blood oxygen level-dependent (BOLD) signal in the human sensorimotor cortex during static force (maintained for a few seconds) and dynamic force (repetitive force pulses) resulted in contradictory findings. Therefore, we conducted a whole-brain functional magnetic resonance imaging study during a visuomotor task requiring the production of either dynamic or static power grip force. Thereby we aimed at clarifying whether the BOLD signal behaves differently with dynamic and static force in the primary motor cortex, and whether it behaves in the same way in all areas and regions involved in force production. In the static condition, participants applied visually guided, isometric grip force on a dynamometer of 20% maximal voluntary contraction (MVC) and held this force for 21 s. In the dynamic condition, self-paced force pulses of 20% MVC were produced at a rate of 0.5 Hz. Static and dynamic force production activated an overlapping network of sensorimotor cortical and subcortical regions. However, the production of a significantly higher mean static force compared with the dynamic force resulted in a significantly smaller BOLD signal in the contralateral motor cortex, confirming observations of an earlier investigation. In addition, we found that the ipsilateral anterior cerebellum behaved similar to the motor cortex, whereas in all other activated regions the activation during static and dynamic force did not significantly differ. These findings demonstrate that various regions of the sensorimotor network participate differentially in the production and control of low static and dynamic grip force, and raise important questions concerning the interpretation of the BOLD signal with respect to mechanisms of neurovascular coupling.

Longitudinal studies to evaluate the effect of rehabilitative therapies require an objective, reproducible and quantitative means for testing function in vivo. An fMRI assessment tool for upper extremity related brain activation using an MRI-compatible manipulandum was developed and tested for use in neurorehabilitation research.Fifteen healthy, right-handed subjects participated in two fMRI sessions, which were three to four weeks apart. A block design paradigm, composed of three conditions of subject-passive movement, subject-active movement and rest, was employed for the fMRI recordings. During the rest condition, subjects simply held the device handle without applying any force or movement. The same type of auditory and visual instructions were given in all the three conditions, guiding the subjects to perform the motor tasks interactively with the MRI-compatible arm manipulandum. The tasks were controlled across the fMRI sessions. The subjects' brain activation was recorded by fMRI, and their behavioral performance was recorded by the manipulandum. The brain network activated by the subjects' interaction with the manipulandum was identified, and the reproducibility and reliability of the obtained activation were determined.All subjects completed the trial protocol. Two subjects were excluded from analysis due to head motion artifacts. All passive movements were performed well. Four out of the total 780 active movements were missed by two subjects. Brain activation was found in the contralateral sensorimotor cortex, secondary somatosensory cortex and non-primary motor cortex as well as in subcortical areas in the thalamus, basal ganglia and the cerebellum. These activations were consistent across the two fMRI sessions.The MRI-compatible manipulandum elicited robust and reproducible brain activations in healthy subjects during the subject-active and subject-passive upper extremity motor tasks with a block design paradigm. This system is promising for many applications in neurorehabilitation research and may be useful for longitudinal studies.

Force scaling in the sensorimotor network during generation and control of static or dynamic grip force has been the subject of many investigations in monkeys and human subjects. In human, the relationship between BOLD signal in cortical and subcortical regions and force still remains controversial. With respect to grip force, the modulation of the BOLD signal has been mostly studied for forces often reaching high levels while little attention has been given to the low range for which electrophysiological neuronal correlates have been demonstrated. We thus conducted a whole‐brain fMRI study on the control of fine‐graded force in the low range, using a power grip and three force conditions in a block design. Participants generated on a dynamometer visually guided repetitive force pulses (ca. 0.5 Hz), reaching target forces of 10%, 20%, and 30% of maximum voluntary contraction. Regions of interest analysis disclosed activation in the entire cortical and subcortical sensorimotor network and significant force‐related modulation in several regions, including primary motor (M1) and somatosensory cortex, ventral premotor and inferior parietal areas, and cerebellum. The BOLD signal, however, increased monotonically with force only in contralateral M1 and ipsilateral anterior cerebellum. The remaining regions were activated with force in various nonlinear manners, suggesting that other factors such as visual input, attention, and muscle recruitment also modulate the BOLD signal in this visuomotor task. These findings demonstrate that various regions of the sensorimotor network participate differentially in the production and control of fine‐graded grip forces. 1. Single cell activity was investigated in the precentral motor (MI) and postcentral somatosensory (SI) cortex of the monkey to compare the neuronal activity related to the control of isometric force in the precision grip and to assess the participation of SI in motor control. 2. Three monkeys (Macaca fascicularis) were trained in a visual step-tracking paradigm to generate and precisely maintain force on a transducer held between thumb and index finger. Great care was taken to have the monkeys use only their fingers without moving the wrist or proximal joints. In two monkeys electromyographic (EMG) activity was checked in 23 muscles over several sessions. 3. Five similar classes of task-related firing patterns were found in both SI and MI cortical hand and finger representations, but their relative proportions differed. The majority of the SI neurons were phasically or phasic-tonically active (61%), whereas in MI the neurons that decreased their firing rate with force were most frequent (42%). 4. The timing of activity changes related to the onset of force increase from low to higher levels strongly differed in the two neuronal populations. In SI, only 14% of the task-related neurons increased or decreased their firing rate before the onset of force increase, in contrast to 56% in MI. Only 3% of the SI neurons showed changes before the earliest EMG activation. 5. In both SI and MI neurons with tonic and phasic-tonic, increasing or decreasing discharge patterns disclosed a relationship between neuronal activity and static force. Distinction was made between neurons modulating their activity in a monotonic way and those that were active only at one force level and had a kind of recruitment or deactivation threshold. The latter ones were more frequent in MI than in SI, and in the neuron population with decreasing firing patterns. For the neurons with increases in activity, statistically significant linear correlations between firing rate and force were found more frequently in MI than in SI, where the proportion of nonsignificant correlations was relatively high (35% vs. 15% in MI). In SI the indexes of force sensitivity, calculated from the slopes of the regression lines, covered a wider range than in MI; and their distribution was bimodal, with one mean of 30 Hz/N and the other of 155 Hz/N. The purpose of the present study was to examine the impact of the congenital absence of one hand on cortical organization of the sensorimotor cortex (S1/M1). We investigated the tongue representation in S1/M1 in nine participants with normally developed limbs, comprising the control group, and in eight persons with a congenitally completely missing hand (i.e. unilateral hand amelia). All participants were examined by fMRI while performing horizontal tongue movements. The significantly activated clusters covering S1/M1 in both hemispheres were analyzed with respect to the number and intensity of activated voxels, as well as the location of the activation. In the right-handed control group, the number of activated voxels was significantly higher in the left as compared to the right hemisphere demonstrating a clear left hemispheric motor dominance for horizontal tongue movements. In the amelic individuals, no such hemispheric lateralization effect was observed. The neural activation pattern underlying tongue movement, however, was enlarged and displaced in the hemisphere contralateral to the missing limb when compared to the respective motor non-dominant, right hemisphere of the control group participants. The present findings suggest that congenital absence of one hand leads to an appreciably altered topological organization of S1/M1 consisting of an enlargement of the tongue representation and a shift towards the "hand" area which, however, had never received any input from a hand.

Execution and imagination of movement activate distinct neural circuits, partially overlapping in premotor and parietal areas, basal ganglia and cerebellum. Can long-term deafferented/deefferented patients still differentiate attempted from imagined movements? The attempted execution and motor imagery network of foot movements have been investigated in nine chronic complete spinal cord-injured (SCI) patients using fMRI. Thorough behavioral assessment showed that these patients were able to differentiate between attempted execution and motor imagery. Supporting the outcome of the behavioral assessment, fMRI disclosed specific patterns of activation for movement attempt and for motor imagery. Compared with motor execution data of healthy controls, movement attempt in SCI patients revealed reduced primary motor cortex activation at the group level, although activation was found in all single subjects with a high variability. Further comparisons with healthy subjects revealed that during attempt and motor imagery, SCI patients show enhanced activation and recruitment of additional regions in the parietal lobe and cerebellum that are important in sensorimotor integration. These findings reflect central plastic changes due to altered input and output and suggest that SCI patients may require additional cognitive resources to perform these tasks that may be one and the same phenomenon, or two versions of the same phenomenon, with quantitative differences between the two. Nevertheless, the retained integrity of movement attempt and motor imagery networks in SCI patients demonstrates that chronic paraplegics can still dispose of the full motor programs for foot movements and that therefore, attempted and imagined movements should be integrated in rehabilitative strategies.

Although corticomuscular synchronization in the beta range (15-30 Hz) was shown to occur during weak steady-state contractions, an examination of low-level forces around 10% of the maximum voluntary contraction (MVC) is still missing. We addressed this question by investigating coherence between electroencephalogram (EEG) and electromyogram (EMG) as well as cortical spectral power during a visuomotor task. Eight healthy right-handed subjects compensated isometrically static forces at a level of 4% and 16% of MVC with their right index finger. While 4% MVC was accompanied by low coherence values in the middle to high beta frequency range (25-30 Hz), a significant increase of coherence mainly confined to low beta frequencies (19-20 Hz) was observed with force of 16% MVC. Furthermore, this increase was associated with better performance, as reflected in decreased relative error in force during 16% MVC. We additionally show that periods of good motor performance within each condition were associated with higher values of EEG-EMG coherence and spectral power. In conclusion, our results suggest a role for beta-range corticomuscular coherence in effective sensorimotor integration, thus stabilizing corticospinal communication.

Background Over the last few years much research has been devoted to investigating the synchronization between cortical motor and muscular activity as measured by EEG/MEG-EMG coherence. The main focus so far has been on corticomuscular coherence (CMC) during static force condition, for which coherence in beta-range has been described. In contrast, we showed in a recent study [1] that dynamic force condition is accompanied by gamma-range CMC. The modulation of the CMC by various dynamic force amplitudes, however, remained uninvestigated. The present study addresses this question. We examined eight healthy human subjects. EEG and surface EMG were recorded simultaneously. The visuomotor task consisted in isometric compensation for 3 forces (static, small and large dynamic) generated by a manipulandum. The CMC, the cortical EEG spectral power (SP), the EMG SP and the errors in motor performance (as the difference between target and exerted force) were analyzed. Results For the static force condition we found the well-documented, significant beta-range CMC (15–30 Hz) over the contralateral sensorimotor cortex. Gamma-band CMC (30–45 Hz) occurred in both small and large dynamic force conditions without any significant difference between both conditions. Although in some subjects beta-range CMC was observed during both dynamic force conditions no significant difference between conditions could be detected. With respect to the motor performance, the lowest errors were obtained in the static force condition and the highest ones in the dynamic condition with large amplitude. However, when we normalized the magnitude of the errors to the amplitude of the applied force (relative errors) no significant difference between both dynamic conditions was observed. Conclusion These findings confirm that during dynamic force output the corticomuscular network oscillates at gamma frequencies. Moreover, we show that amplitude modulation of dynamic force has no effect on the gamma CMC in the low force range investigated. We suggest that gamma CMC is rather associated with the internal state of the sensorimotor system as supported by the unchanged relative error between both dynamic conditions., BMC Neuroscience, 8, ISSN:1471-2202

The beta-range synchronization between cortical motor and muscular activity as revealed by EEG/MEG-EMG coherence has been extensively investigated for steady-state motor output. However, there is a lack of information on the modulation of the corticomuscular coherence in conjunction with dynamic force output. We addressed this question comparing the EEG-EMG coherence and the cortical motor spectral power in eight healthy subjects in a visuomotor task, in which the subjects exerted a steady-state or periodically modulated dynamic isometric force output with their right-index finger to keep a visual cursor within a target zone. In the static condition, significant coherence was confined to the beta-range. In the dynamic condition, the most distinct coherence occurred in the gamma-range and the significant beta-range coherence was strikingly reduced. The cortical motor power in the beta-range during dynamic force output was decreased, whereas the power in the gamma-range remained without significant change. We conclude that during dynamic force the corticospinal oscillation mode of the sensorimotor system shifts towards higher (principally gamma) frequencies for the rapid integration of the visual and somatosensory information required to produce the appropriate motor command.

Little is known about the influence of the afferent peripheral feedback on the sensorimotor cortex activation. To answer this open question we investigated the alpha and beta band task-related spectral power decreases (TRPow) in the deafferented patient G.L. and compared the results to those of six healthy subjects. The patient has been deafferented up to the nose for 24 years but her motor fibers are unaffected and she can perform complex motor tasks under visual control. We recorded EEG (58 scalp positions) as well as the exerted force during a visuomotor task. The subjects had to maintain in precision grip an isometric force at 15% of the maximal voluntary contraction. In the patient we found a significantly higher alpha band spectral power during rest and larger alpha TRPow decreases during the motor task when compared to the healthy subjects' data. In contrast, we did not observe any significant differences between patient and controls for the beta band TRPow. The results indicate an altered functional alpha band network state in the patient, probably due to the chronic deafferentation leading to a deep 'idling' state of the contralateral sensorimotor area.

Conflicting reports exist about the occurrence, reliability and localization of activation in the ipsilateral primary motor cortex (M1). We re-examined this issue with functional magnetic resonance imaging in 12 volunteers performing right hand, finger, wrist, elbow, foot and tongue movements in two separate sessions. Ipsilateral M1 activation was inconsistently and non-reliably present during all movements: in 54% of all hand, 50% elbow, 46% finger, 33% wrist, and in 17% of all foot experiments. When compared to contralateral M1, the volumes and maximum t-values were always smaller. The ipsilateral M1 body representation was somatotopically organized with coordinates similar to the contralateral M1. Finally, the presence of ipsilateral M1 activation depended on the global activation level in other motor-related areas, which was significantly increased, when ipsilateral M1 activation was detected.

Peripheral and central nervous system lesions can induce reorganization within central somatosensory and motor body representations. We report changes in brain activation patterns during movements of non-affected body parts in paraplegic patients with spinal cord injury (SCI). Nine SCI patients and 12 healthy controls underwent blood oxygen level dependent signal functional MRI during sequential finger-to-thumb opposition, flexion and extension of wrist and of elbow, and horizontal movements of the tongue. Single subject and group analyses were performed, and the activation volumes, maximum t values and centres of gravity were calculated. The somatotopical upper limb and tongue representations in the contralateral primary motor cortex (M1) in the SCI patients were preserved without any shift of activation towards the deefferented and deafferented M1 foot area. During finger movements, however, the SCI patients showed an increased volume in M1 activation. Increased activation was also found in non-primary motor and parietal areas, as well as in the cerebellum during movements of the fingers, wrist and elbow, whereas no changes were present during tongue movements. These results document that, in paraplegic patients, the representation of the non-impaired upper limb muscles is modified, though without any topographical reorganization in M1. The extensive changes in primary and non-primary motor areas, and in subcortical regions demonstrate that even distant neuronal damage has impact upon the activation of the whole sensorimotor system.

The human cortex reportedly contains at least five nonprimary motor areas: in the frontolateral convexity, the dorsal and ventral premotor cortex (PMd and PMv), and in the frontomesial wall, the presupplementary and supplementary motor areas (pre-SMA and SMA), and the rostral, dorsal and ventral cingulate areas (CMAr, CMAd, and CMAv). Activation of these regions in neuroimaging studies has been generally associated either with the performance of complex motor tasks or with reorganization occurring with motor recovery in the presence of pathology. Recent evidence from neuroimaging studies suggests that the same areas are activated with well controlled simple movements in healthy subjects providing support to the observation that their contribution may be more quantitative rather than exclusively specific to a certain aspect of motor behaviour. An important consequence of this observation is that activation of multiple nonprimary motor areas during simple motor tasks should not be considered unique to patients with upper or lower motoneuron lesions but rather as a normal physiological process.

The relationship between single cell activity in the “motor” thalamus and the generation of isometric force between the fingers has been investigated in 2 monkeys. Neurons related to the task were found in the thalamic motor regions VLo, VPLo, and VA where microstimulation occasionally elicited motor reactions in hand and fingers. 58% of these 55 neurons, designated “typical”, showed modulation of their discharge patterns with force similar to neurons in precentral cortex and could be assigned to one of 5 discharge patterns described for the motor cortex. Only a small percentage of the thalamic neurons were found to have phasic activity. The other “atypical” neurons (42%) had discharge patterns with complex sequences of phasic and tonic activation with respect to force. For 18 typical and atypical neurons with tonic and phasic-tonic modulation of their firing rate with force significant regression coefficients between firing rate and static force were observed. The mean index of force sensitivity (rateforce slope) was 54.5 Hz/N for the neurons increasing their discharge rate with force, i.e. approximately that of precentral cells. Neurons tested for their sensory properties had receptive fields located on hand and/or fingers and were activated mainly by stimulation of muscle and joint receptors. The characteristics of these thalamic neurons are compared to those of precentral cells recorded under identical experimental conditions and are discussed in relation to the known input-output relationships of the motor thalamic nuclei. The data strongly support the hypothesis that parameters of movement, in particular force, are represented by the activity of neurons in the “motor” thalamus.

Background During isometric compensation of modulated low-level forces corticomuscular coherence (CMC) has been shown to occur in high-beta or gamma-range. The influence of the frequency of force modulation on CMC has up to now remained unexplored. We addressed this question by investigating CMC, motor performance, and cortical spectral power during a visuomotor task in which subjects had to compensate a modulated force of 8% of the maximum voluntary contraction exerted on their right index finger. The effect of three frequencies of force modulation (0.6, 1.0 and 1.6 Hz) was tested. EEG, EMG from first dorsal interosseus, hand flexor and extensor muscles, and finger position were recorded in eight right-handed women. Results Five subjects showed CMC in gamma- (28-45 Hz) and three in beta-range (15-30 Hz). Beta- and gamma-range CMC and cortical motor spectral power were not modulated by the various frequencies. However, a sharp bilateral CMC peak at 1.6 Hz was observed, but only in the five gamma-range CMC subjects. The performance error increased linearly with the frequency. Conclusions Our findings suggest that the frequency of force modulation has no effect on the beta- and gamma-range CMC during isometric compensation for modulated forces at 8% MVC.
