Histopathological studies of parasitic infections in fish from the natural environment of Brazilian Amazon, are quite scarce. In this study, we investigated the histopathological changes of the proximal intestine of specimens of the Amazonian fish Hoplias malabaricus infected by the hematophagous nematode Procamallanus (Spirocamallanus) paraensis. The prevalence of the infection was 60%, with an average abundance of 1.46 and an average intensity of 2.43 parasites/fish. No significant difference was observed in the prevalence of infection and fish size or sex, but larger fish showed greater infection intensity, which was also significantly higher in male hosts. Histological sections of the proximal intestine showed reduction and loss of the epithelial lining, exposure of the lamina propria where the nematode interacts with the intestine wall, through the insertion of the buccal capsule and fish cellular debris in the intestinal lumen. Tilapia is one of the most important farmed fish in the world and the most cultivated in Brazil. The increase of this farming favors the appearance of diseases, including bacterial diseases. Therefore, the aim of this study was to evaluate the bactericidal activity of copaiba oil, Copaifera duckei, against Streptococcus agalactiae and Flavobacterium columnare and the dietary effect of copaiba oil on zootechnical performance, hematological, biochemical, immunological, and histological analysis before and after an intraperitoneal infection (body cavity) with S. agalactiae in Nile tilapia. For this, fish were randomly distributed into 15 fiber tanks in five treatments (0, 0.25, 0.50, 0.75, and 1.0%) and fed with a commercial diet supplemented with copaiba oil for 30 days. After this period, the fish were randomly redistributed for the experimental challenge with S. agalactiae into six treatments (T0, T1, T2, T3, T4, and T5), the fish were anesthetized, and blood samples were collected to assess hematological, biochemical, immunological, and histological parameters. Copaiba oil showed bactericidal activity against Streptococcus spp. and Flavobacterium spp. in vitro. In addition, concentrations of 0.75 and 1.0% of copaiba oil have an anti-inflammatory effect and improve hematological and immunological parameters, increasing leukocyte numbers, albumin, and serum lytic activity. Furthermore, there is an increase in the intestinal villus length and tissue damage in groups at concentrations of 0.75 and 1.0% of copaiba oil. In conclusion, copaiba oil presented bactericidal activity against Streptococcus spp. and Flavobacterium spp. in vitro, and oral supplementation at concentrations of 0.75 and 1.0% compared to the control group enhanced non-specific immune parameters and digestibility in Nile Tilapia., (© 2024. The global ornamental fish trade carries important risk factors for spreading pathogens between different countries and regions, not only for ornamental fish but also for cultured fish and even other animal species. In the current study, we reported the capacity of Aeromonas veronii and A. hydrophila isolated from ornamental fish to experimentally infect the reared Amazonian fish Colossoma macropomum. For this, those bacteria were identified, and a primary characterization was performed. Fish were inoculated with 0.1 mL of increasing concentrations of A. hydrophila or A. veronii (C1 = 1 × 10 2 ; C2 = 1.8 × 10 4 ; C3 = 2.1 × 10 6 ; C4 = 2.4 × 10 8 bacterial cells per mL) in the coelomic cavity. In the control group, fish received the same volume of sterile saline solution (0.9 %). Fish presented petechiae, skin suffusions, and mortality rates up to 100 % according to the inoculum concentration. Histopathologically, fish presented necrosis with karyolysis, loss of the cytoplasmic delimitation of cells of the renal tubules and hepatocytes, hemorrhage, cellular edema, and the presence of bacterial cells. The LD 50-96h of A. veronii on C. macropomum was estimated at 2.4 × 10 6  CFU mL -1 and of A. hydrophila at 1.408 × 10 5  CFU mL -1 . The results demonstrated that it is possible that Aeromonas species isolated from ornamental fish affect C. macropomum, causing similar clinical signs and lesions. Acanthocephalosis is caused by the endoparasite Neoechynorhyncus buttnerae and affects fish farmed in the Amazon. This study assessed the efficacy of therapeutic levamisole hydrochloride (LVC) baths against N. buttnerae and its effects on juvenile tambaqui blood parameters. In vitro and in vivo tests were carried out, the latter employing two experimental therapeutic LVC bath protocols. Concerning in vitro efficacy, the T75 (75 mg.L -1 LVC) and T100 (mg.L -1 LVC) treatments were 100% effective in 15 min, while the T50 (50 mg.L -1 LVC) and T25 (25 mg.L -1 LVC) treatments required parasite exposure for 45 and 60 min, respectively. During exposure, the parasites displayed reduced motility, proboscis retraction, coiling into a spiral shape, body rigidity and swelling. The LVC LC 50-72h for juvenile tambaqui was 115 mg.L -1 . Regarding in vivo efficacy for Protocol I (8-h bath), the T125 resulted in 82% effectiveness, while in Protocol II (two 8-h baths with a 24-h interval), the T115 treatment (115 mg.L -1 LVC) achieved 95.6% effectiveness without clinical intoxication signs, despite behavioural changes. No significant changes were observed in fish blood parameters. The present study describes two new Myxobolus species infecting the gills of Semaprochilodus insignis, the most consumed freshwater fish species in the Brazilian Amazon. The fish specimens were caught in the Tapajós River, in the state of Pará, and the morphological, ultrastructural, small subunit ribosomal DNA (ssrDNA), and phylogenetic data of the myxosporean species were obtained. Two species of Myxobolus were found in the gills of S. insignis. Myxobolus maiai n. sp. developed in the gill filaments, and mature myxospores were round-shaped from the frontal view, measuring 12.5-14.8 (13.9 ± 0.5) μm in length, 11.4-13.8 (12.3 ± 0.5) μm in width, and have a thickness of 6.4-7.7 (6.9 ± 0.6) μm in the lateral view, with symmetric values. Its polar capsules were 4.4-6.6 (5.5 ± 0.5) μm in length and 2.3-3.7 (3.0 ± 0.3) μm in width, and the polar tubules had 4 - 5 coils. Myxobolus iarakiensis n. sp. was found infecting the gill arch. Mature myxospores were oval-shaped from the frontal view, and measured 6.7-8.6 (8.0 ± 0.4) μm in length, 4.5-6.3 (5.6 ± 0.4) μm in width, and had a thickness of 2.7-4.7 (3.8 ± 0.5) μm in the lateral view, with symmetric values. Its polar capsules were 2.1-3.7 (2.9 ± 0.3) μm in length and 1.1-2.0 (1.5 ± 0.2) μm in width, and its polar tubules had 4 - 5 coils. Objective was the relationship of the parasite Rhadinorhynchus plagioscionis with its host Plagioscion squamosissimus, providing morphological and morphometric data to prove the specificity of this parasite with its host, with reports occurring only in rivers in the Brazilian Amazon. Rhadinorhynchus plagioscionis infected P. squamosissimus captured in the municipal region of Santarém, in the state of Pará, in the eastern Brazilian Amazon. The specimens were analyzed by light and scanning electron microscopy. Morphological data obtained confirmed conspecificty with R. plagioscionis and supplemental information was added to the morphological concept of this species. The body of R. plagioscionis is cylindrical and milky-coloured with dorsal curvature, a long proboscis displaying ventral curvature with 12 longitudinal rows containing 22 to 24 hooks, which are larger and more prominent towards the end of the ventral curvature, and decrease in size in the posterior extremity of the proboscis. The present study describes, in addition to the morphological and morphometric characteristics of R. plagioscionis, which have host specificity for P. squamosissimus, a relationship being reported exclusively in the Brazilian Amazon.

Crustaceans of the subclass Copepoda are an important component of the invertebrate aquatic fauna. They occur in all aquatic environments and include some representatives that are free-living organisms and others that have a parasitic lifestyle. The genus Therodamas comprises marine and freshwater copepods whose females are parasites of fish in their adult phase, with only seven species described so far. During a field survey of fish parasites in the Jari River, a large tributary of the Amazon River system, in Brazil, we found a new species of the genus Therodamas infecting Leporinus fasciatus. Therodamas longicollum n. sp. is the second strictly freshwater species known. Phylogenetic analysis showed that the new species is grouped in the family Ergasilidae, and divergence estimates showed that T. longicollum n. sp. diverged from its ancestor at around 66.34 Ma, in the late Upper Cretaceous. Therodamas longicollum n. sp. differs from its congeneric in that it does not have lobes and/or expansion of the anterior neck region. Besides describing a new Therodamas species, thereby increasing the diversity of the genus to eight species, this study points out the existence of a lineage of these copepods that has adapted to the freshwater environment of the Amazon. Streptococcosis causes great economic losses in intensive culture of tilapia. Vaccination is the most effective and safest way to tackle infectious diseases. Thus, this study sought the more effective and safer antigenic fraction after sonication of Streptococcus agalactiae to elaborate a vaccine against streptococcosis in Nile tilapia. For this, twenty-one days after vaccination with different fractions (soluble and insoluble) of S. agalactiae, the fish were challenged with the homologous strain (LD50). Then, samples were taken at zero, 14, 28, 60 and 90 days post-vaccination (DPV, n = 7). Blood and organs (cranial kidney, spleen and liver) were collected from vaccinated and unvaccinated fish. Finally, insoluble fraction vaccine presented the best effect, resulting in a 100% relative percent of survival (RPS) and without clinical manifestations. In view of the results, it was to evaluate the role of the insoluble fraction of the antigen in the protective immunity against streptococcosis. The results indicate that the spleen might be the main organ in the vaccine response in Nile tilapia due to the great morphological and immunological differences in vaccinated fish, evidenced by the greater of melanomacrophage centers (MMC) and IgM + lymphocytes in relation to the non-vaccinated fish. At 60 DPV, it was observed the peak of the protective immunity related to the maximum concentration of proteins, circulating leukocytes, antibody titers in the serum and tissue changes with greater expression of IgM + and MMC number in the spleen and kidney of Oreochromis niloticus. This study undertook the first investigation of malacosporean infections in Neotropical fish. We used polymerase chain reaction detection with a primer set generally targeting known malacosporeans to assay for infection in the kidney of 146 fish in 21 species belonging to 12 families collected from two areas in the Amazon Basin. Infections were found in 13 fish variously belonging to seven species in six families and included the first identification of a malacosporean infection in cartilaginous fish (a freshwater stingray). Based on ssrDNA, all infections represented a single Buddenbrockia species (Buddenbrockia sp. E) that demonstrates an exceptionally broad range of fish species infected, and countered our expectations of high Neotropical malacosporean diversity. Infections were characterized at varying and often high prevalences in fish species but sample sizes were small. Ascertaining whether highly divergent malacosporeans have not been detected by current primers, and more comprehensive sampling may reveal whether malacosporeans are truly as species poor in the Amazon Basin as present data suggest. Our results prompt speculations about evolutionary scenarios including introduction via marine incursions and patterns of host use over time.

This study aimed to investigate the presence of nematodes Spirocamallanus krameri and Contracaecum sp. in Hoplerythrinus unitaeniatus caught in a lake in the state of Pará, in northern Brazil, eastern Amazon region (Brazil). From 92 hosts, 83.7% were infected, and a total of 401 parasites were covered. There was a high prevalence of S. krameri, while the predominance was of Contracaecum sp. larvae. Both parasite species presented aggregated dispersion, and abundance of these parasites presented positive correlation with weight and length of the hosts. Morphological and morphometric data of third-stage larvae of Contracaecum sp. are described.

The objective of this study was reports, through morphological and small subunit ribosomal DNA (SSU rDNA) sequencing, two novel myxobolid myxosporeans infecting Piaractus brachypomus, an economicaly important Amazonian fish popularly known as "pirapitinga". Of a total of 25 specimens of P. brachypomus examined 68% had the gill filament parasitized by Henneguya tapariensis n. sp. and 16% had infection of Myxobolus arapiuns n. sp. in the pyloric cecum. The morphological analysis revealed H. tapariensis n. sp. myxospores with an ellipsoid shape and caudal process larger than the length of the body. The polar capsules of same size were elongated and occupied less than half the body. Sequencing of the SSU rDNA generated a partial sequence of 1946 bp. In M. arapiuns n. sp. the myxospores had oval-shaped body and polar capsules of the same size, occupying less than half the body. Sequencing of the SSU rDNA generated a partial sequence of 1950 bp. Phylogenetic analysis revealed a cluster according to the order/family of the host, where H. tapariensis n. sp. was grouped in a subclade with Henneguya brachypomus and Henneguya piaractus and M. arapiuns grouped in a subclade with Myxobolus colossomatis, Myxobolus matosi and Myxobolus pirapitingae., Competing Interests: Declaration of Competing Interest The authors declare that there are no conflicts of interest., (Copyright © 2020 Elsevier B.V. Clinostomid trematodes are only widely studied due to the ability to infect their metacercariae, which can affect amphibians, fish, snakes and occasionally mammals, with occasional records in humans. The Loricariids constitute the most diverse family of neotropical fish, with more than 800 registered species. They present a large heterogeneity of colors and body forms that reflect its high degree of ecological specialization and importance on economic aspects such as ornamentation and food. In spite of these factors, studies involving the diversity of parasitic fauna that affect in these fish is still scarce. In this aspect, our study presents the first record of occurrence of the parasite Clinostomum marginatum in association with the Loricariid Pterygoplichthys pardalis in the Amazon region of Brazil. In September 2018, in the river mouth of Tapajós river, tributary of Amazon river, 32 specimens of P. pardalis and six specimens of Aphanotorulus emarginatus were captured. The parasitological studies revealed the occurrence of metacercariae of C. marginatum in association with P. pardalis, representing a prevalence of 12.5% with a mean abundance of 0.18 and a medium intensity of infection of 1.5. No C. marginatum parasitism was observed in A. emarginatus.

Hoplias aff. malabaricus is abundant in the Mogi-Guaçu River. The aim of this study was to perform an inventory of the species of metazoan that parasite this species of fish taken from oxbow lakes of the Mogi-Guaçu River. The Mann-Whitney test was used to statistically analyze the possible influence of the sex of the host on the group with the highest parasite richness and the greatest abundance of parasites. Simpson’s diversity index was used to determine parasite diversity among the zoological groups of parasites of H. aff. malabaricus with the highest index. A total of 78 specimens of H. aff. malabaricus were examined. Among the zoological groups of metazoans found, the phylum Nematoda had the greatest number of species. Among these, the larval stage of Contracaecum sp. was most abundant. The sex of the host had a significant effect, with parasites more abundant in female fish (Z(U)=0.043; p<0.05). The digenean Parspina argentinensis, the nematodes Procamallanus (S.) iheringi, Rhabdochona acuminata and Hysterothylacium sp. and copepods Vaigamus sp. and Lernaea cyprinacea have not previously been recorded as parasites of H. aff. malabaricus.

Aeromonas bacteria can cause an infection characterized by septicemia and is one of the most common pathogens in tropical fish. This disease is responsible for high morbidity and mortality rates, causing considerable losses in aquaculture. Thus, the understanding of its pathophysiology is crucial to develop control strategies of this bacterial infection in farmed fish. This study aimed to characterize early pathological aspects of acute sepsis in pacu (Piaractus mesopotamicus) experimentally infected with Aeromonas hydrophila. A total of 160 juvenile pacus were inoculated intraperitoneally with A. hydrophila (1.78 x 109 CFU/mL) and at 0 (control), 1, 3, 6, and 9 hours post-inoculation (hpi), animals were anesthetized and samples were collected for microbiological, light microscopy and transmission electron microscopy (TEM) analyzes. The results showed the occurrence of hemodynamic alterations, such as hemorrhage and congestion, which were observed mainly after 6 and 9 hpi. It was possible to re-isolate Aeromonas at all sampling times except in control group. However, just after 9 hpi it was possible to find the bacteria in all fish and tissues. Light microscopy analyses revealed a degenerative process, necrosis and vascular damage mainly at 6 and 9 hpi. According to the ultrastructural examination, areas of cellular death were identified in all examined tissues, especially at 6 and 9 hpi. However, the most severe, related to necrosis, were observed after 6 and 9 hpi. The findings suggested that this bacterium spreads in the first hpi through the fish organs, mainly affecting spleen, liver and kidney, causing irreversible lesions at the molecular level., Competing Interests: The authors have declared that no competing interests exist.

The pathogenesis of sepsis involves complex systems and multiple interrelationships between the host and pathogen producing high mortality rates in various animal species. In this study, hematological disturbances, innate immunity and survival during the septic process in Piaractus mesopotamicus inoculated with Aeromonas hydrophila were studied. For this aim, fish blood samples were taken from control and infected groups 1, 3, 6, and 9 h post-inoculation (HPI). Leukogram showed reduction in the number of leukocytes and thrombocytes, followed by cessation of leukocyte chemotaxis 6 HPI and severe morphological changes in leukocytes and erythrocytes. At 3 HPI production of reactive oxygen species increased and at 6 HPI decreased. There was no change in serum lysozyme concentration and lytic activity of the complement system, despite the progressive increase in serum lytic activity and bacterial agglutination. Finally, the changes in clinical signs due to aeromonosis and increasing septicemia resulted in a reduction in survival to 57.14% after 36 HPI. It was possible concluded that these hematological and immune are crucial event in the worsening of sepsis in P. mesopotamicus, and these findings are utility for diagnosing and understanding the pathophysiology sepsis in pacu induced by A. hydrophila., (Copyright © 2019 Elsevier Ltd. The study investigated the monogenoids infracommunity from the gills of Cichla monoculus and C. pinima living sympatrically in the lower Tapajós River, State of Pará (Brazil). A total of 561 monogenoids were collected of C. monoculus, which was host to seven species of these parasites, and 672 monogenoids were collected of C. pinima, which was host to eight species of these parasites. The monogenoids infracommunities of C. monoculus and C. pinima had a high qualitative similarity (88.0%), and quantitative (87%), and were composed by Gussevia arilla, G. longihaptor, G. tucunarense, G. undulata, Sciadicleithrum ergensi, S. umbilicum and S. uncinatum. There was a predominance of G. arilla and a low level of infection by Tucunarella cichlae. The prevalence, intensity and abundance of monogenoids were similar for C. monoculus and C. pinima, except for the intensity of G. undulata that was higher in C. monoculus. Parasite species displayed an aggregated dispersion, but G. longihaptor and S. ergensi exhibited random dispersion. This was the first study of monogenoids for C. pinima, and the first record of G. arilla, G. tucunarense, S. ergensi, S. umbilicum and S. uncinatum for C. monoculus. This study therefore extended the geographic distribution of these parasites to the lower Tapajós River in eastern Amazon.

The ciliate Ichthyophthirius multifiliis is an important pathogen of freshwater fish that occurs in both temperate and tropical regions around the world. The purpose of present study was to report an outbreak of I. multifiliis in Steindachneridium parahybae from the Paraiba do Sul River in state of São Paulo (Brazil). The fins, tegument and gills of S. parahybae were examined for the presence of I. multifiliis, and blood parameters (Red Blood Cell, thrombocytes and leukocytes) were evaluated. All the examined fish (100%) were parasitized with I. multifiliis and the mean infection intensity was 15.5 parasites per fish. All the fish died 72 hours as a consequence of ichthyophthiriasis with alterations to the tegument coloration and open opercula. Red Blood Cell counts and hemoglobin concentration were low, indicating an anemic process.

The present study describes, for the first time, histopathological alterations in the gills of Macrobrachium amazonicum caused by infestation of Probopyrus bithynis (Isopoda: Bopyridae). In every case (100%), the infestation by P. bithynis was by a single pair of parasites (male and female) and occurred in the right or left side of the branchial chamber; the gill structures were visibly compressed due to the presence of parasites. The gills of M. amazonicum parasitized by P. bithynis exhibited a chronic inflammatory response, with the presence of edema, greater quantities of hemocytes, necrosis, epithelial cell hyperplasia, rupture of the pillar cells at the ends of the gill lamellae, desquamation of the cuticle, lamellar fusion and rupture of the lamellar epithelium. Tissue lesions were found in the histological sections of the gills of the parasitized M. amazonicum. Structural alterations in the branchial chamber of the hosts caused by the presence of P. bithynis can lead to physiological changes that can impair host respiratory performance. Finally, histopathological alterations in the branchial chamber of hosts suggest that P. bithynis feed directly on the gill tissues of this shrimp.

In vivo and in vitro antiparasitic activity of the essential oil of Lippia sidoides and blood and histological alterations were assessed in Colossoma macropomum (tambaqui). Essential oil concentrations of 10, 20, 40, 80, 160 and 320mg/L were assayed in vitro against monogenoideans Anacanthorus spathulatus, Notozothecium janauachensis and Mymarothecium boegeri from fish gills. Lippia sidoides essential oil concentrations of 320 and 160mg/L were 100% effective against monogenoideans in 10min and 1h of exposure, respectively. However, the effectiveness of 100% concentrations of 80mg/L and 40mg/L occurred in 3 and 6h, respectively. In the in vivo tests, juvenile fish were submitted to 60min of baths with 10mg/L and 15min of baths with 20mg/L of the essential oil of L. sidoides. These therapeutic baths were not efficient against Ichthyophthirius multifiliis, and monogenoideans present in the gills of C. macropomum. In addition, 10 and 20mg/L of the essential oil of L. sidoides caused an anesthetic effect on the fish and did not influence total glucose and protein plasma levels; however, it decreased the number of total erythrocytes in fish exposed to the higher concentration of this essential oil. Severe alterations and irreversible damage were observed in the fish gills just after L. sidoides essential oil baths and after 24h of recovery. The most recurrent lesions found were hyperplasia and fusion of the lamellar epithelium, vasodilation, detachment of the gill epithelium and lamellar aneurism, epithelial breakdown with hemorrhage, congestion, edema and necrosis, proliferation of the mucous cells and chloride cells and lamellar hypertrophy. Therefore, since the essential oil of L. sidoides has in vitro antiparasitic activity and low concentrations of it have shown toxic effects, the bioactive potential of its main chemical components should be investigated, as well as more efficient forms of its administration in therapeutic baths in order to eliminate fish parasites., (Copyright © 2016 Elsevier B.V. This study describes the hematological alterations in Astyanax altiparanae associated with infestation with Lernaea cyprinacea. The study was carried out in a lagoon of the Mogi-Guaçu River, in the municipality of Pirassununga, in the state of São Paulo, Brazil. Of 46 fish examined, 45.6% had their integument infested by L. cyprinacea, with a mean intensity of 4.9 parasites per fish and a mean abundance of 2.2, giving a total of 139 recovered crustaceans. The abundance of L. cyprinacea correlated positively with the length and weight of the hosts, and the intensity of infestation was higher in female hosts. Macroscopic observation of lesions associated with the parasite showed a severe inflammatory reaction around the site of attachment of L. cyprinacea, associated with a lower relative condition factor and blood parameters. The hematocrit, number of red blood cells and thrombocytes were higher in non-parasitized than in parasitized fish. However, the hemoglobin concentration, hematimetric indices and the number of white blood cells were not influenced by infestation.

The aim of this study was to investigate the component community of parasite metazoans of Piaractus brachypomus in the lower Amazon River, northern Brazil. From 34 necropsied fish, 27,384 metazoan parasites were collected, such as Anacanthorus spathulatus, Mymarothecium viatorum and Notozothecium janauachensis (Monogenoidea); Spectatus spectatus and Contracaecum sp (Nematoda); Clinostomum marginatum and Dadaytrema oxycephala (Digenea); and Argulus carteri and Ergasilus sp. (Crustacea). The dominant species was S. spectatus followed by monogenoidean species, and there was aggregated dispersion of parasites, except for D. oxycephala and Contracaecum sp., which presented random dispersion. Positive correlation among the abundance of the three monogenoideans species were found, thus indicating that there was no competition between the species of these parasites on the gills of hosts. The abundances of some parasite species showed positive correlations with the size of the hosts, but the condition factor of the fish was not affected by the parasitism levels. It showed that this host had a metazoan community characterized by high species richness of metazoans, low evenness and high diversity of parasites, with prevalence of endoparasites, including larval stages. This was the first record of C. marginatum, A. carteri, Ergasilus sp. and Contracaecum sp. for P. brachypomus.
