Climate change and chemical contamination are global environmental threats of growing concern for the scientific community and regulatory authorities. Yet, the impacts and interactions of both stressors (particularly ocean warming and emerging chemical contaminants) on physiological responses of marine organisms remain unclear and still require further understanding. Within this context, the main goal of this study was to assess, for the first time, the effects of warming (+ 5 °C) and accumulation of a polybrominated diphenyl ether congener (BDE-209, brominated flame retardant) through dietary exposure on energy budget of the juvenile white seabream (Diplodus sargus). Specifically, growth (G), routine metabolism (R), excretion (faecal, F and nitrogenous losses, U) and food consumption (C) were calculated to obtain the energy budget. The results demonstrated that the energy proportion spent for G dominated the mode of the energy allocation of juvenile white seabream (56.0-67.8%), especially under the combined effect of warming plus BDE-209 exposure. Under all treatments, the energy channelled for R varied around 26% and a much smaller percentage was channelled for excretion (F: 4.3-16.0% and U: 2.3-3.3%). An opposite trend to G was observed to F, where the highest percentage (16.0 ± 0.9%) was found under control temperature and BDE-209 exposure via diet. In general, the parameters were significantly affected by increased temperature and flame retardant exposure, where higher levels occurred for: i) wet weight, relative growth rate, protein and ash contents under warming conditions, ii) only for O:N ratio under BDE-209 exposure via diet, and iii) for feed efficiency, ammonia excretion rate, routine metabolic rate and assimilation efficiency under the combination of both stressors. On the other hand, decreased viscerosomatic index was observed under warming and lower fat content was observed under the combined effect of both stressors. Overall, under future warming and chemical contamination conditions, fish energy budget was greatly affected, which may dictate negative cascading impacts at population and community levels. Further research combining other climate change stressors (e.g. acidification and hypoxia) and emerging chemical contaminants are needed to better understand and forecast such biological effects in a changing ocean., (Copyright © 2018 Elsevier Inc. The effects of two different slipping methods on the survival, physical and physiological response of sardines, Sardina pilchardus, captured in a purse-seine fishery were investigated in southern Portugal. Sardines were collected and transferred into holding tanks onboard a commercial fishing vessel after being captured, crowded and deliberately released using two slipping procedures: standard and modified. The standard slipping procedure aggregated fish at high densities and made them "roll over" the floatline, while the modified procedure aggregated the fish at moderate densities and enabled them to escape through an opening created by adding weights to the floatline. Both slipping methods were compared with minimally harmed non-slipped sardines (sardines collected from the loose pocket of the purse seine). Survival rates were monitored in captivity over 28 days using three replicates for each treatment. The estimated survival of sardines was 43.6% for the non-slipped fish, 44.7% for the modified slipping and 11.7% for the standard slipping treatments. Scale loss indicated the level of physical impact experienced, with dead fish from the non-slipped and modified slipping technique showing significantly lower scale loss than those fish from the standard slipping treatment within the same period. Of the physiological indicators of stress measured, cortisol, glucose, lactate and osmolality attained peak values during slipping and up to the first hours after introduction to captivity. Atlantic sardines (Sardina pilchardus) captured in the Portuguese mainland coast and maintained in captivity for one year feeding on artificial diet showed significant differences in muscle total lipids (TL) content in relation to wild fish (25% after one year compared to 14% in wild). Captive sardines had higher percentage of non-polar lipids (NPL) in their muscle, presented higher levels of 18:1n-9 and lower of 20:5n-3 and 22:6n-3 in NPL than those observed in wild fish. TL in the liver were not significantly different between captive and wild fish, but the NPL fraction of captive sardines presented higher levels of 22:6n-3 and 20:5n-3 than those registered in the wild. Polar lipids (PL) were less influenced by captivity since the fatty acids profile was more stable. Increases in carbon dioxide (CO 2 ) and other greenhouse gases emissions are changing ocean temperature and carbonate chemistry (warming and acidification, respectively). Moreover, the simultaneous occurrence of highly toxic and persistent contaminants, such as methylmercury, will play a key role in further shaping the ecophysiology of marine organisms. Despite recent studies reporting mostly additive interactions between contaminant and climate change effects, the consequences of multi-stressor exposure are still largely unknown. Here we disentangled how Argyrosomus regius physiology will be affected by future stressors, by analysing organ-dependent mercury (Hg) accumulation (gills, liver and muscle) within isolated/combined warming (ΔT=4°C) and acidification (ΔpCO 2 =1100μatm) scenarios, as well as direct deleterious effects and phenotypic stress response over multi-stressor contexts. After 30days of exposure, although no mortalities were observed in any treatments, Hg concentration was enhanced under warming conditions, especially in the liver. On the other hand, elevated CO 2 decreased Hg accumulation and consistently elicited a dampening effect on warming and contamination-elicited oxidative stress (catalase, superoxide dismutase and glutathione-S-transferase activities) and heat shock responses. Thus, potentially unpinned on CO 2 -promoted protein removal and ionic equilibrium between hydrogen and reactive oxygen species, we found that co-occurring acidification decreased heavy metal accumulation and contributed to physiological homeostasis. Although this indicates that fish can be physiologically capable of withstanding future ocean conditions, additional experiments are needed to fully understand the biochemical repercussions of interactive stressors (additive, synergistic or antagonistic)., (Copyright © 2017 Elsevier B.V. The behavioural effects of confinement of sardine Sardina pilchardus in a purse seine were evaluated through three laboratory experiments simulating the final stages of purse seining; the process of slipping (deliberately allowing fishes to escape) and subsequent exposure to potential predators. Effects of holding time (the time S. pilchardus were held or entangled in the simulation apparatus) and S. pilchardus density were investigated. Experiment 1 compared the effect of a mild fishing stressor (20 min in the net and low S. pilchardus density) with a control (fishing not simulated) while the second and third experiments compared the mild stressor with a severe stressor (40 min in the net and high S. pilchardus density). In all cases, sea bass Dicentrarchus labrax were used as potential predators. Results indicated a significant effect of crowding time and density on the survival and behaviour of slipped S. pilchardus. After simulated fishing, S. pilchardus showed significant behavioural changes including lower swimming speed, closer approaches to predators and higher nearest-neighbour distances (wider school area) than controls, regardless of stressor severity. These results suggest that, in addition to the delayed and unobserved mortality caused by factors related to fishing operations, slipped pelagic fishes can suffer behavioural impairments that may increase vulnerability to predation. Possible sub-lethal effects of behavioural impairment on fitness are discussed, with suggestions on how stock assessment might be modified to account for both unobserved mortality and sub-lethal effects, and possible approaches to provide better estimates of unobserved mortality in the field are provided., (© 2013 The Fisheries Society of the British Isles.)

Abstract: Formulation of diets according to fish amino acid (AA) qualitative requirements may improve fish growth. Two diets with different AA profiles were tested on 16‐day‐old meagre during 20 days. The first diet had an AA profile closer to meagre larval AA requirements (BAL), whereas the second had a different AA profile (UNBAL). Results showed that larvae fed the diet with higher similarities to the AA profile of meagre carcass had a higher final weight (19.8 ± 0.5 mg versus 13.4 ± 1.8 mg) and lower ammonia excretion after being fed (0.6 ± 0.1 ml/(g DW. larvae. h) and 0.8 ± 0.1 ml/(g DW larvae. h)). A higher mean fibre area was obtained in 36 DAH larvae fed the BAL diet (224.8 ± 33.3 μm2 versus 158.8 ± 34.5 μm2) as well as higher larval weight. When fibre area distribution was analysed, a higher frequency of larger fibres was observed in 36 DAH BAL larvae, suggesting that fibre hypertrophy had a higher importance in this treatment. In conclusion, this study shows that meagre larvae fed a diet with an AA profile closer to their requirements had lower nitrogen losses and higher final weight, as result of higher muscle hypertrophy. In this study, two growth trials were conducted to evaluate the effect of dietary protein and lipid levels on growth and feed utilization of white sea bream ( Diplodus sargus) juveniles. For the first trial, five diets were formulated to contain 120 g kg−1 lipid and increasing levels of protein, ranging from 400 to 600 g kg−1. Two additional diets were formulated with 400 and 600 g kg−1 protein and 180 g kg−1 lipids. The diets were fed to apparent visual satiety to duplicate groups of fish with a mean weight of 1.5 g for 10 weeks. For the second growth trial, four diets were formulated to contain 120 g kg−1 lipid and 380–520 g kg−1 protein. Two additional diets were formulated with 380 and 520 g kg−1 protein and 180 g kg−1 lipids. The diets were fed to apparent visual satiety to triplicate groups of fish with a mean weight of 41 g for 12 weeks. At the end of both trials, there were no growth differences among groups independent of the dietary protein content. In the first trial, growth was negatively correlated to dietary lipid levels. No significant differences of feed intake were detected among groups in both trials, but a direct correlation between feed efficiency and dietary protein level was observed. Protein efficiency ratio and nitrogen (N) retention (% N intake) significantly decreased with the increase of dietary protein levels. In both trials, energy retention (% energy intake) was highest in groups fed on diets with the highest protein-to-energy (P/E) ratio. At the end of both trials, no significant differences in whole-body composition were observed among groups. Specific activity of enzymes involved in amino acid catabolism [aspartate aminotransferase (AST), alanine aminotransferase (ALT) and glutamate dehydrogenase (GDH)] showed no significant differences with dietary protein level in both trials. Nevertheless, in the first trial, a significantly lower GDH activity was observed in fish fed with higher dietary lipid levels. No differences were found for specific activity of the lipogenic enzymes, fatty aid synthetase and glucose-6-phosphate dehydrogenase, in the second trial. Results of this study indicate that a diet with a protein level of 380–420 g kg−1 and a P/E ratio of 20 g protein MJ−1 satisfies the growth requirements of D. sargus juveniles. Abstract: The present work aims to study the effect of a soluble non-starch polysaccharide (NSP), guar gum on white sea bream intestinal health through oxidative status and distal intestine morphology evaluation. A control diet was formulated to contain 40% crude protein, 14% crude lipids and 35% pregelatinized maize starch. Three other diets were formulated to include the same levels of all ingredients as control diet except for guar gum, which was included at 4% (diet GG4), 8% (diet GG8) or 12% (diet GG12). Compared to the control, lower catalase (CAT), superoxide dismutase (SOD) and glutathione reductase (GR) activities and higher SOD, glutathione peroxidase (GPX) and glucose-6-phosphate dehydrogenase (G6PD) activities were observed in the whole intestine of fish fed with diets GG4 and GG12, respectively. Thiobarbituric acid-reacting substances (TBARS) levels were unaffected by diet composition. No morphological alterations in the distal intestine were found among groups. Overall, data on key enzymes of the antioxidant enzymatic defense mechanism suggested that diet-induced reactive oxygen species (ROS) generation is dependent on guar gum inclusion levels and low dietary inclusion of soluble NSP (up to 4%) may have a gut oxidation stress prevention effect.
