Traffic accident injuries are relevant causes of mortality in industrialized countries. Among traffic accidents, the most destructive effect on the victim's body is produced by trains, which in a very short time provoke a complete distortion of the normal anatomical profiles and extensive amputations of the body. Deaths by severe body mutilation and/or decapitation require complex forensic investigation, as the victims' identification and the recognition of their body are not often feasible. In this study, a retrospective analysis of a series of cases of railway accidents is presented, with the aim of showing potential identification strategies, including a systematic physical re-assembling of the body. Among the railway fatalities assessed at the University Institute of Legal Medicine of Milan from 2016 until 2020, 61 cases were analyzed, focusing on the collection of postmortem data to improve accuracy for the identification of the victims. The results showed that the identification could be based on elements traditionally called "secondary identifiers" which, however, are becoming more and more important for positive identification, especially when dealing with migrants, vagrants, and homeless people. However, several issues need to be considered such as the limitations of the public prosecutors to perform expensive analyses (i.e., genetic testing), and the lack of antemortem data to be compared with postmortem findings, both odontological and fingerprint. In cases where it is not possible to proceed with identification or formal recognition of victims of railroad accidents with severe mutilations, a systematic physical re-assembling of the body is critical to reaching positive identification. Simple Summary: The delimitation of the postmortem interval (PMI) is a very important parameter in veterinary forensic medicine. Indeed, the PMI can provide law enforcement with useful information to correctly determine a temporal relationship between a suspect's actions and murder or to deny or confirm the testimony of a witness. Unfortunately, postmortem cadaveric changes, which are useful in investigating the PMI, have been poorly investigated in animals. In the present study, we evaluated the postmortem modifications of desmin and dystrophin over time and their correlation with PMI in dogs. To this aim, 10 dead adult dogs were evaluated for 4 days; for each animal, a cube of muscle tissue was collected from the vastus lateralis and triceps brachii. Muscle tissues were removed at 3 h postmortem and every 24 h until 96 h after death. Protein expression levels were analyzed by immunohistochemical examination and immunoblot analysis. The obtained results showed that dystrophin proteins had a higher degradation rate as compared to desmin. Overall, this study identified proteins with different postmortem degradation resistance, providing a reference basis for better investigating PMI in dogs. The obtained results showed rapid dystrophin degradation, with complete disappearance at 72 h after death. Image acquisition of dead bodies, particularly using postmortem computed tomography (PMCT), has become common in forensic investigations worldwide. Meanwhile, in countries such as Japan which have an extremely low rate of autopsy, PMCT is being increasingly used in the clinical field to certify the cause of death (COD) without performing an autopsy or toxicological tests, even in cases of unnatural death. Additionally, these PMCT images are predominantly interpreted by clinical personnel such as emergency physicians or clinicians who are not trained in PMCT interpretation and who work for the police, that is, the so‐called police doctors. Many potential pitfalls associated with the use of PMCT have been previously described in textbooks and published papers, including the pitfalls of not performing a complete forensic pathology investigation, and the use of physicians without appropriate PMCT training to interpret PMCT and direct death investigation and certification. We describe five examples in which apparent misdiagnosis of COD based on PMCT misinterpretation was revealed by autopsy. Here are the five examples of errors: (1) Postmortem changes were misinterpreted as COD, (2) resuscitation effects were misinterpreted as COD, (3) COD was determined after an incomplete examination, (4) fatal findings caused by external origin were wrongly interpreted as 'of internal origin' based on PMCT, and (5) non‐fatal findings on PMCT were wrongly interpreted as fatal. Interpretation of PMCT by appropriately trained physicians and an accompanying complete forensic investigation, including autopsy when indicated, is necessary to prevent significant errors in COD determination and related potential adverse medicolegal consequences. • It is a new idea to solve the species identification by screening differentially expressed proteins (DEPs) of fly pupae. • It is the first study to identify species of fly pupae using a label-free proteomics technique and to further validate their results with PRM. • Our study investigated DEPs via the label-free technique during the intrapuparial stage in the Ch. megacephala , and S. nudiseta and found 132 DEPs between Ch. megacephala , and S. nudiseta groups, with 68 and 64 proteins being up-regulated and down-regulated between the two groups. • The C 1 -tetrahydrofolate synthase, Malate dehydrogenase, Transferrin, Protein disulfide-isomerase, and Fructose-bisphosphate aldolase, could be used for species identification of Ch. megacephala , and S. nudiseta. Here, we used the label-free proteomics technique to discover the DEPs and further validate using the parallel reaction monitoring technique (PRM) in the common flies. In this study, we reared the Chrysomya megacephala and Synthesiomyia nudiseta at constant temperature, and then we sampled at least four pupae at 24 h intervals until the end of the intrapuparial stage. We found 132 DEPs between Ch. megacephala , and S. nudiseta groups, with 68 and 64 proteins being up-regulated and down-regulated between the two groups. Among the 132 DEPs, we selected five proteins having potential for further development and utilization, such as C 1 -tetrahydrofolate synthase, Malate dehydrogenase, Transferrin, Protein disulfide-isomerase, and Fructose-bisphosphate aldolase, for further validation using PRM-targeted proteomics, with the trends of PRM results being consistent with the label-free data for corresponding proteins. We present the results of the first study on the decomposition of hanging carcasses in a forest habitat of Poland. The decomposition of 12 hanging pigs and 12 ground pigs, which were used as control, in spring, summer, and autumn was examined during a two-year study. Mosaic decomposition was observed with bloating, active decay, and advanced decay irrespective of the carrion treatment (hanging vs ground). Bloating was less visible or did not occur at all in hanging pigs. After undergoing advanced decay, carcasses became mummified and remained in the hanging position. Complete skeletonization was observed only within the head in one case in summer. Irrespective of the carrion treatment, the active decay was driven by the same blow fly species with a similar seasonal pattern of larval dominance. The season was found to affect the decomposition of the hanging carcasses as revealed by a distinct delay in the onset of bloating in autumn and the late onset of active decay in spring. The duration of active decay was the shortest in summer and the longest in autumn. The effect of carrion treatment on the rate of decomposition was statistically insignificant. Nevertheless, some differences in the duration of decomposition between hanging and ground pigs were observed. The most distinct difference was that earlier decomposition proceeded faster in hanging pigs in spring and a delayed decomposition was observed in autumn. In summer, decomposition was similar in all the carcasses. The presented results are largely in contrast to the findings of previous studies on decomposition of hanging pig carrion. Human bone histological analysis is a useful tool to assess post mortem diagenesis and to predict successful nuclear DNA typing of forensic material. This study is part of a series of studies developed by the authors intended to improve the understanding of post mortem diagenesis and to develop applications for DNA analysis of skeletal species from tropical soils, in order to optimize genetic and anthropological protocols. The aim of this study was to analyze the impact of burial period on the integrity of exhumed compact bone microstructure from tropical climate. In fragments of exhumed human femora from 39 individuals from the same cemetery (exhumed group) and 5 fresh femora from routine autopsies (control group), sections stained by hematoxylin-eosin were analyzed in order to measure bone microstructural integrity. We found that bone integrity index in exhumed group was negatively influenced by the period of burial (r = -0.37, p < 0.05) and highly significantly decreased (p < 0.0001) in comparison to control group. The period of burial and nitric acid decalcification time was positively correlated (r = 0.51; p < 0.01), leading to imply a bone petrification process during inhumation. Exhumed group showed higher level of matrix bone loss (p < 0.001), as expected, and 87% of cases analyzed were "tunneled" as described by Hackett. Bone integrity index and bone matrix tend to decrease in bones buried in tropical soil between 8-14 years of inhumation. This period is short if we consider cases in which there are preserved bones interred for longer periods in other environments. These data must be considered in cases where genetic identification of exhumed skeletons from tropical environment is required. In forensic death investigations, estimating the postmortem interval (PMI) is critical. An accurate PMI estimate increases the speed and accuracy of identifying the remains by narrowing the time frame in which the death occurred, thus reducing the pool of possible decedents. Cape Town, South Africa has a high level of unnatural death, and due to a burdened death investigation system, many remain unidentified. There has been a tendency to broadly apply quantitative models of decomposition across biogeographically unique circumstances. A prime example is the widespread application of the total body score (TBS)/accumulated degree day (ADD) model developed by Megyesi et al. (2005), later refined by Moffatt et al. (2016). However, the appropriateness of applying a single model to a wide range of locations with unique geography and climates remains in question. The aim of the study was to evaluate and compare the accuracy of Megyesi and Moffatt models for estimating PMI in Cape Town, South Africa. Using pig carcasses, Finaughty established baseline data on the rates and patterns of terrestrial decomposition in summer and winter in two different locations in a forensically significant area of Cape Town. Among the baseline data, Finaughty derived TBS values using the Megyesi criteria. The present study used these values to estimate the ADD per the Megyesi and Moffatt models, which would correspond to an estimated PMI. These estimated values were compared to actual ADD values. Estimates of ADD were inaccurate for both models in winter, and only partially in summer. The Moffatt model was more accurate in earlier decomposition stages, with the Megyesi model more accurate in later decomposition stages. These results indicate the Cape Town environments may contain factors that the two models do not consider, producing inaccurate PMI estimations at various TBS' values. ADD does not depict the entire taphonomic story; the decomposition process appears to be too complex for universal modelling based on a single or narrow suite of variables. Seasonality was an important factor in determining the accuracy of the models, primarily resulting in underestimations of the true PMI values. These findings show the impracticality of applying models developed for- or in one region to any other and support the need to establish regionally-specific equations for estimating PMI in a forensic context. Post-mortem measurements were made of the body temperature of dogs. The aim of the study was to evaluate and verify a reliable mathematical model that can be used to establish the time elapsed since the death of a dog during the initial post-mortem period at room temperature, using the eye (vitreous body), internal organs (heart, liver, kidney and lung), and rectum as sites for temperature measurement. The measurements were performed at six points in the body using an electronic thermometer in conjunction with a temperature probe. The method of temperature measurement is simple and does not cause perceptible macroscopic changes or disfigure the carcass. Multiple regression analysis was shown to be suitable for estimating the time elapsed from death to the discovery of the body for a period up to 12h post-mortem. The proposed multiple regression equation using body weight and the temperature at a specific site reduces manipulation of the carcass to a minimum and thus reduces error in establishing the time of death. Over the past decade, Structure from Motion (SfM) has increasingly been used as a means of digital preservation and for documenting archaeological excavations, architecture, and cultural material. However, few studies have tapped the potential of using SfM to document and analyze taphonomic processes affecting burials for forensic sciences purposes. This project utilizes SfM models to elucidate specific post-depositional events that affected a series of three human cadavers deposited at the South East Texas Applied Forensic Science Facility (STAFS). The aim of this research was to test the ability for untrained researchers to employ spatial software and photogrammetry for data collection purposes. For a series of three months a single lens reflex (SLR) camera was used to capture a series of overlapping images at periodic stages in the decomposition process of each cadaver. These images are processed through photogrammetric software that creates a 3D model that can be measured, manipulated, and viewed. This project used photogrammetric and geospatial software to map changes in decomposition and movement of the body from original deposition points. Project results indicate SfM and GIS as a useful tool for documenting decomposition and taphonomic processes. Background: The correct estimation of the post mortem interval (PMI) can be crucial on the success of a forensic investigation. Diverse methods have been used to estimate PMI, considering physical changes that occur after death, such as mortis algor, livor mortis, among others. Degradation after death of dental pulp is a complex process that has not yet been studied thoroughly. It has been described that pulp RNA degradation could be an indicator of PMI, however this study is limited to 6 days. The tooth is the hardest organ of the human body, and within is confined dental pulp. The pulp morphology is defined as a lax conjunctive tissue with great sensory innervation, abundant microcirculation and great presence of groups of cell types.Aim: The aim of this study is to describe the potential use of pulp post mortem alterations to estimate PMI, using a new methodology that will allow obtainment of pulp tissue to be used for histomorphological analysis. The current study will identify potential histological indicators in dental pulp tissue to estimate PMI in time intervals of 24h, 1 month, 3 months and 6 months.Materials and Method: This study used 26 teeth from individuals with known PMI of 24h, 1 month, 3 months or 6 months. All samples were manipulated with the new methodology (Carrasco, P. and Inostroza C. inventors; Universidad de los Andes, assignee. The dental pulp tissues obtained were fixed in formalin for the subsequent generation of histological sections, stained with Hematoxylin Eosin and Masson's Trichrome. All sections were observed under an optical microscope using magnifications of 10× and 40×.Results: The microscopic analysis of the samples showed a progressive transformation of the cellular components and fibers of dental pulp along PMI. These results allowed creating a chart of qualitative and quantitative parameters to be used on the estimation on PMI based on microscopic degradation of dental pulp.Conclusions: The histological transformations of dental pulp as a function of time can be used as PMI indicators. This study aimed to develop an aquatic decomposition scoring ( ADS) method and investigated the predictive value of this method in estimating the postmortem submersion interval ( PMSI) of bodies recovered from the North Sea. This method, consisting of an ADS item list and a pictorial reference atlas, showed a high interobserver agreement (Krippendorff's alpha ≥ 0.93) and hence proved to be valid. This scoring method was applied to data, collected from closed cases-cases in which the postmortal submersion interval ( PMSI) was known-concerning bodies recovered from the North Sea from 1990 to 2013. Thirty-eight cases met the inclusion criteria and were scored by quantifying the observed total aquatic decomposition score ( TADS). Statistical analysis demonstrated that TADS accurately predicts the PMSI ( p < 0.001), confirming that the decomposition process in the North Sea is strongly correlated to time. The precision of death time estimation by vitreous potassium using 5 different formulas is compared on a random sample of 600 cases. Mean differences, standard deviations and 95% limits of confidence between real and extrapolated time since death were calculated. The best results were obtained using equations with "mean" slopes of about 0,17 or 0,19mmol/l per hour. Equations with a steeper or flatter slope reveal already great mean deviations between real and extrapolated time since death and do not allow a reliable death time estimation. Microbially mediated mechanisms of human decomposition begin immediately after death, and are a driving force for the conversion of a once living organism to a resource of energy and nutrients. Little is known about post-mortem microbiology in cadavers, particularly the community structure of microflora residing within the cadaver and the dynamics of these communities during decomposition. Recent work suggests these bacterial communities undergo taxa turnover and shifts in community composition throughout the post-mortem interval. In this paper we describe how the microbiome of a living host changes and transmigrates within the body after death thus linking the microbiome of a living individual to post-mortem microbiome changes. These differences in the human post-mortem from the ante-mortem microbiome have demonstrated promise as evidence in death investigations. We investigated the post-mortem structure and function dynamics of Staphylococcus aureus and Clostridium perfringens after intranasal inoculation in the animal model Mus musculus L. (mouse) to identify how transmigration of bacterial species can potentially aid in post-mortem interval estimations. S. aureus was tracked using in vivo and in vitro imaging to determine colonization routes associated with different physiological events of host decomposition, while C. perfringens was tracked using culture-based techniques. Samples were collected at discrete time intervals associated with various physiological events and host decomposition beginning at 1h and ending at 60 days post-mortem. Results suggest that S. aureus reaches its highest concentration at 5-7 days post-mortem then begins to rapidly decrease and is undetectable by culture on day 30. The ability to track these organisms as they move in to once considered sterile space may be useful for sampling during autopsy to aid in determining post-mortem interval range estimations, cause of death, and origins associated with the geographic location of human remains during death investigations. Introduction: The microscopic study and the interpretation of skin samples with advanced post-mortal phenomena or with particular destructive injuries is problematic for the forensic pathologist. In an attempt of restoring the histological architecture of cadaveric skin and overcoming these types of problem, the Authors performed a histological processing that was longer than the standard: it was extended until 62 days to evaluate the improvement of the microscopic morphological aspect.Materials and Methods: Cutaneous samples were taken from 25 cadavers (5 typologies of skin: charred, putrified, corifed, mummified and partially skeletonized), fixed with a 10%-buffered formalin and then processed in two different ways: one half of the samples was routinely addressed to the standard-time automatic technique, while the other half was manually processed with prolonged times. All the slides were then stained in Hematoxylin-Eosin.Results: The standard-processed slides demonstrated marked morphological alterations and artefacts at the microscopic observation; conversely, those processed with the prolonged manual technique showed an improvement in the morphological structure, sometimes permitting the identification of the anatomical components.Conclusion: Though it is characterized by the inconvenience of protracted times, the application of a long-term manual histological processing to cadaveric skin samples with advanced post-mortal alteration permits to better observe the anatomical architecture of skin and it could be useful and helpful in the evaluation of such cases. Penetrating trauma has been cited as a significant factor in the rate of decomposition. Therefore, penetrating trauma may have an effect on estimations of time-since-death in medicolegal investigations and on research examining decomposition rates and processes when autopsied human bodies are used. The goal of this study was to determine if there are differences in the rate of decomposition between autopsied and non-autopsied human remains in the same environment. The purpose is to shed light on how large incisions, such as those from a thorocoabdominal autopsy, effect time-since-death estimations and research on the rate of decomposition that use both autopsied and non-autopsied human remains. In this study, 59 non-autopsied and 24 autopsied bodies were studied. The number of accumulated degree days required to reach each decomposition stage was then compared between autopsied and non-autopsied remains. Additionally, both types of bodies were examined for seasonal differences in decomposition rates. As temperature affects the rate of decomposition, this study also compared the internal body temperatures of autopsied and non-autopsied remains to see if differences between the two may be leading to differential decomposition. For this portion of this study, eight non-autopsied and five autopsied bodies were investigated. Internal temperature was collected once a day for two weeks. The results showed that differences in the decomposition rate between autopsied and non-autopsied remains was not statistically significant, though the average ADD needed to reach each stage of decomposition was slightly lower for autopsied bodies than non-autopsied bodies. There was also no significant difference between autopsied and non-autopsied bodies in the rate of decomposition by season or in internal temperature. Therefore, this study suggests that it is unnecessary to separate autopsied and non-autopsied remains when studying gross stages of human decomposition in Central Texas and that penetrating trauma may not be a significant factor in the overall rate of decomposition. Abstract: The most common task of a forensic entomologist is to determine an accurate minimum post-mortem interval (PMI) using necrophagous fly larvae found on carrion. More often, blowflies (Diptera: Calliphoridae) are the first insects to detect the cadaver and, if the circumstances are favourable, to leave eggs on the body. However, several studies reveal that products such as gas or paint found on the cadaver induce a delay in the colonisation of the body, leading to an under-estimate of the PMI. Six common household products (gas, mosquito citronella repellent, perfume, bleach, hydrochloric acid and soda) were added to dead rats (Rattus norvegicus) in a field (Lille Forensic Institute, France). The presence of necrophagous flies was checked at regular intervals during 1 month. This experiment was repeated at the same period for four consecutive years. Results clearly showed the repellent effect of three of the six tested substances: gas (petroleum spirit), perfume and mosquito citronella repellent, which resulted in a mean delay of several days in the appearance of the first Dipteran species. Experiments were then carried out in controlled conditions in order to confirm previous observations. An olfactometer was specially designed to observe the behaviour of female Calliphora vicina (Diptera: Calliphoridae) in response to mice (Mus musculus) cadaver odour stimuli combined with household products. Dead mouse odour was a strong attractive stimulus for most of the tested individuals. Furhermore, it was noticed that the presence of mosquito citronella repellent, perfume, hydrochloric acid and paradichlorobenzene produced a significant repellent effect on female flies. All these results together confirm the repellent effect of some household products on flies and the necessity for forensic entomologists to consider this hypothesis when estimating the PMI. Purpose: The decomposition process of human bodies in marine environment is not well understood, and it is influenced by external variables related to the geographical area where the body is submerged. We report the application of two decomposition scores, the Heaton's score and the van Daalen's score, on a casuistry of human bodies recovered from the Northern Adriatic Sea. The aims of this study are to verify whether the marine environment of a Mediterranean climate area may affect the applicability of both scores and to develop a prediction model that can be applied on bodies recovered in salt water.Methods: A retrospective study was performed on 61 human bodies recovered between 2005 and 2019 from coastal water of the Northern Adriatic Sea nearby the Italian regions Emilia-Romagna and Marche. For each of the 61 cases included, the Total Aquatic Decomposition Score (TADS) was calculated with the Heaton's score and the Van Daalen's score. The prediction model was assessed through multiple regression analyses, and the determination coefficients (r2) between TADS and PMSI (expressed in days) and between TADS and Accumulate Degrees Days (ADD) were studied. The prediction model was applied to the entire case sample, to bodies recovered during the warm season and to bodies recovered during the cold season.Results: All bodies were recovered floating, and a very poor scavenging activity was observed. The regression analyses showed a strong correlation between the TADS and the total case sample using both scores and both independent variables (PMSI and ADD). The determination coefficients were greater than 0.95 also when considering the total case sample.Discussion: The proposed prediction models are not significantly influenced by seasonality, contrarily to what observed on bodies recovered in fresh water in the same climate area. However, the ADD model, which also consider the water temperature, should be preferred for higher decomposition stages. While cerebral edema is a live-threatening condition in living persons, also an edema-like fluid redistribution can occur post mortem. In deceased, usually macroscopic signs are evaluated during autopsy in order to determine the presence or absence of cerebral edema. As a quantitative and objective classification is beneficial, an already existing method (Radojevic et al., 2017), which is based on a mathematical formula using the intracranial dimensions and the cerebral weight, was compared to the evaluation of macroscopic signs in 31 cases. The results showed an excellent agreement for the comparison between the raters as well as the measurement methods (at opened skull or in CT images). However, both measurement methods only poorly agree with the macroscopic edema evaluation. In order to find a more concordant method, the normalized cerebral weight, which puts the cerebral weight in relation to the intracranial volume, was calculated for 115 cases. This method resulted in an excellent agreement with the macroscopic rating and showed a clear numerical difference between the edematous and nonedematous group. While the influence of the post mortem time and the cooling time was found to be negligible, the age at death might confound the edema classification due to pre-existing cerebral atrophy leading to lower cerebral weights. In summary, the present study compared different assessment methods to classify cerebral edema and developed a rater independent, objective and quantitative classification method, which was as reliable as the rating of the forensic pathologists. Gold standard for the estimation of the time since death in the early postmortem period is the temperature based nomogram method together with time of death dependent criteria of postmortem lividity, rigor mortis and supravital reactions. There is also a huge literature on chemical methods proposed for estimating the time since death which however play obviously no role in forensic practice. Especially the rise of vitreous potassium has been studied intensively. Also immunohistochemical methods have been proposed for estimating the time since death but obviously not yet applied in casework. We present a case where a woman was found murdered 8 days after having been seen last alive. Due to lack of putrefactive changes postmortem interval was thought to be not more than 2 days. However, immunohistochemical stainings and vitreous potassium concentration revealed that time since death was more than 6 days and the woman was obviously murdered immediately after she was seen lastly alive. Twenty mummies discovered in a forensic context between 2002 and 2016 were compiled in this work. 15 cases were excluded and 15 cases of forensic mummies were found in the literature. In the current work the percentage of mummification was calculated by "the rule of nines" used for describing burned injuries in livings. Dry and hot environments, emaciation, little access to flies are favorable conditions for mummification. Nevertheless mummification was also observed in other cold and humid environments. Extensive mummification (defined in this work as "at least 50% of mummification of the body skin) has occurred in as little as three weeks in the current series. The Virginia opossum (Didelphis virginiana) is a highly active scavenger whose behavior has significant impacts on rates of decomposition and skeletonization, which have previously not been addressed. In this study, scavenging by the opossum led to the skeletonization of carcasses in half of the accumulated degree days (ADD) of a comparable non-scavenged control carcass. Opossums used body orifices, as well as natural tears caused by the decomposition process, to access internal tissues and consume them. This activity resulted in little movement of the carcass and the retained appearance of natural undisturbed decomposition. This concealed activity has the potential to cause drastically incorrect estimates of time since deposition and post-mortem interval. Scavenging by opossums was also found to leave distinct tooth mark and other defects on bone, which have not been previously distinguished in the literature. This research suggests, beyond effects on PMI, that scavenging by opossums has been historically overlooked and misattributed to canid scavengers. [ABSTRACT FROM AUTHOR]
