In the modern world with climate changes and increasing pollution, different types of stress are becoming an increasing challenge. Hence, the identification of reliable biomarkers of stress and accessible sensors to measure such biomarkers are attracting increasing attention. In the current study, we demonstrate that the activity, but not the expression, of the ubiquitous enzyme topoisomerase 1 (TOP1), as measured in crude cell extracts by the REEAD sensor system, is markedly reduced in response to thermal stress in both fruit flies (Drosophila melanogaster) and cultivated human cells. This effect was observed in response to both mild-to-moderate long-term heat stress and more severe short-term heat stress in D. melanogaster. In cultivated HeLa cells a reduced TOP1 activity was observed in response to both cold and heat stress. The reduced TOP1 activity appeared dependent on one or more cellular pathways since the activity of purified TOP1 was unaffected by the utilized stress temperatures. We demonstrate successful quantitative measurement of TOP1 activity using an easily accessible chemiluminescence readout for REEAD pointing towards a sensor system suitable for point-of-care assessment of stress responses based on TOP1 as a biomarker. Aging is a multifactorial trait caused by early as well as late-life circumstances. A society trend that parents deliberately delay having children is of concern to health professionals, for example as advanced parental age at conception increases disease risk profiles in offspring. We here aim to study if advanced parental age at conception affects mitochondrial DNA content, a cross-species biomarker of general health, in adult human twin offspring and in a model organism. We find no deteriorated mitochondrial DNA content at advanced parental age at conception, but human mitochondrial DNA content was higher in females than males, and the difference was twofold higher at advanced maternal age at conception. Similar parental age effects and sex-specific differences in mitochondrial DNA content were found in Drosophila melanogaster. In addition, parental longevity in humans associates with both mitochondrial DNA content and parental age at conception; thus, we carefully propose that a poorer disease risk profile from advanced parental age at conception might be surpassed by superior effects of parental successful late-life reproduction that associate with parental longevity. The article discusses results of a study that determines how laboratory selection on diets with various protein content affect body composition, body size, body fat and viability in Drosophila melanogaster. The study material and methods are discussed, including fly populations and larvae diets and use of analysis of variances (ANOVA). The study showed both plastic and evolutionary consequences on life-history traits when Drosophila melanogaster was exposed to nutritional environments with different protein composition.

The increased homozygosity due to inbreeding leads to expression of deleterious recessive alleles, which may cause inbreeding depression in small populations. The severity of inbreeding depression has been suggested to depend on the rate of inbreeding, with slower inbreeding being more effective in purging deleterious alleles of smaller effect. The effectiveness of purging is however dependent on various factors such as the effect of the deleterious, recessive alleles, the genetic background of inbreeding depression and the environment in which purging occurs. Investigations have shown inconclusive results as to whether purging efficiently diminish inbreeding depression. Here we used an ecologically relevant inbreeding coefficient (f ≈ 0.25) and generated ten slow and ten fast inbred lines of Drosophila melanogaster by keeping the effective population size constant at respectively 32 and 2 for 19 or 2 generations. These inbred lines were contrasted to non-inbred control lines. We investigated the effect of inbreeding and inbreeding rate in traits associated with fitness including heat, cold and desiccation stress resistance, egg-to-adult viability, development time, productivity, metabolic rate and wet weight under laboratory conditions. The results showed highly trait specific consequences of inbreeding and generally no support for the hypothesis that slow inbreeding is less deleterious than fast inbreeding. Egg-to-adult viability and development time were investigated under both benign and heat stress conditions. Reduced viability and increased developmental time were observed at stressful temperatures and inbreeding depression was on average more severe at stressful compared to benign temperatures. The article investigates the association of Drosophila melanogaster's adult heat tolerance variation to heat-inducible heat shock proteins (Hsp70) expression. Study reveals no significant correlation between Hsp70 expression and the laboratory assays of adult heat tolerance used in Drosophila. It finds out that the variation in the expression explains a small proportion of variation in adult heat tolerance. This suggests that its influence on intra-specific heat tolerance variation may be specific to life, though it serves as a primary component of cellular heat stress response.

Rapid cold hardening is a naturally occurring phenomenon in insects that is thought to be responsible for increased cold tolerance during diurnal variations in temperature. The underlying physiological mechanisms are still not fully resolved but, in Drosophila melanogaster (Meigen 1830), rapid cold hardening is accompanied by specific changes in the membrane lipid composition. To further understand the link between rapid cold hardening and adjustments in the membrane lipid composition, the present study investigates how different rates of cooling affect thermotolerance and the composition of phospholipid fatty acids. Female Drosophila are cooled gradually from 25 to 0 °C at 0.01, 0.05, 0.1 or 0.5 °C min−1, respectively, and, subsequently, phospholipid fatty acid composition and survival after a 1-h cold shock at −5 °C is measured. The rapid cold hardening treatments all influence cold tolerance differently so that short and intermediate rapid cold hardening treatments (0.05, 0.1 or 0.5 °C min−1 cooling rates) increase cold shock survival, whereas the slow cooling treatment (0.01 °C min−1) decreases survival relative to an untreated control. The intermediate rapid cold hardening treatments (0.05 or 0.1 °C min−1) induce a similar type of response characterized by an increase in the molar percentage of linoleic acid, 18:2(n-6), at the expense of 16:0 and 18:1(n-9), which leads to an increase in the degree of unsaturation. The slowest cooling treatment (0.01 °C min−1) results in a large increase in cis-16:1(n-7) and significant reductions in the saturated phospholipid fatty acids 16:0, 18:0 and the unsaturated 16:1(n-9) and 18:2(n-6) fatty acids. These changes cause a slight decrease in the average length of the phospholipid fatty acids and an increase in the overall ratio of unsaturated vs. saturated fatty acids. These findings demonstrate that the rate of cooling is important for both the reorganization of membrane lipids, and for the degree of acquired cold tolerance during rapid cold hardening, and they suggest an important role for rapid cold hardening during diurnal rather than seasonal temperature changes. Age is known to have a large effect upon fertility and survival. Here we study the impact of maternal and grandmaternal age on wing size and developmental instability (DI) in offspring using a parthenogenetic strain ofDrosophila mercatorum. This enabled us to cancel out any genetic variance between individuals. The mean centroid size of the wings was estimated in offspring from four maternal/grandmaternal age groups. Further, DI was calculated by measuring phenotypic variability and correcting this index for the bias produced by the environmental variability. The offspring developed from eggs laid by mothers belonging to four different age groups with young (Y) or old (O) mothers and grandmothers. The age groups are: YY, YO, OY and OO, with the first letter referring to the age of the grandmother and the second letter to the age of the mother. We consider flies between three to six days of age as young and flies between 15 and 18 days of age as old. We found that environmental variability was present in this study and therefore the correction for its bias on phenotypic variability was shown to be necessary for a correct interpretation of the results. DI displayed, for most of the traits investigated, a significantly higher level in the OY and OO age groups, as compared to the YY and YO age groups. The mean centroid size of the wings were in almost all cases significantly different among the age groups with the highest mean value in the OY group for all traits. Simple Summary: Most laboratory experiments on insects to date have been conducted using constant temperature settings. Even when the purpose of the study was to investigate effects of temperature, insects have mostly been kept at different but constant temperatures ignoring natural variation in temperature. Here we investigated effects of simple daily temperature fluctuation (22.5/27.5 °C and 20/30 °C) on some development characteristics in five species of fruit flies (Drosophila) originating from areas with different temperature profiles. We demonstrated how species of the same genus can show substantial differences when developing at fluctuating temperatures not always predictable by development at comparable constant temperature (25 °C). Temperature has profound effects on biochemical processes as suggested by the extensive variation in performance of organisms across temperatures. Nonetheless, the use of fluctuating temperature (FT) regimes in laboratory experiments compared to constant temperature (CT) regimes is still mainly applied in studies of model organisms. We investigated how two amplitudes of developmental temperature fluctuation (22.5/27.5 °C and 20/30 °C, 12/12 h) affected several fitness-related traits in five Drosophila species with markedly different thermal resistance. Egg-to-adult viability did not change much with temperature except in the cold-adapted D. immigrans. Developmental time increased with FT among all species compared to the same mean CT. The impact of FT on wing size was quite diverse among species. Whereas wing size decreased quasi-linearly with CT in all species, there were large qualitative differences with FT. Changes in wing aspect ratio due to FT were large compared to the other traits and presumably a consequence of thermal stress. These results demonstrate that species of the same genus but with different thermal resistance can show substantial differences in responses to fluctuating developmental temperatures not predictable by constant developmental temperatures. [Display omitted] • We present a system to study navigation of small motile animals in thermal landscapes. • In this system we exposed fruit flies to thermal gradients and extreme temperatures. • Some flies showed thermal boldness (TB, voluntary exposure to critical temperatures) • Mild fasting was not required but enhanced TB, and TB varied among strains. • TB did not correlate with physiological tolerances to temperature. A dominant perception is that small and motile ectothermic animals must use behavior to avoid exposure to critical or sub-critical temperatures impairing physiological performance. Concomitantly, volunteer exploration of extreme environments by some individuals may promote physiological adjustments and enhance ecological opportunity. Here we introduce to the literature a Thermal Decision System (TDS) which is fully modular, thermally stable, versatile, and adaptable to study navigation through thermal landscapes in insects and other small motile animals. We used a specific setting of the TDS to investigate volunteer navigation through critical cold and hot temperatures in Drosophila melanogaster. We demonstrate that a thermally bold behavior (volunteer crossings through a Critical Temperature Zone, CTZ) characterized a fraction of flies in a sample, and that such a fraction was higher in an outbred population relative to isofemale lines. As set, the TDS generated a thermal gradient within the cold and hot CTZs, and the exploration of this gradient by flies did not relate simply with a tendency to be thermally bold. Mild fasting affected thermal exploration and boldness in complex manners, but thermal boldness was evident in both fasted and fed flies. Also, thermal boldness was not associated with individual critical temperatures. Finally, some flies showed consistent thermal boldness, as flies that performed an extreme thermal cross were more likely to perform a second cross compared with untested flies. We hypothesize that a simple "avoidance principle" is not the only behavioral drive for D. melanogaster facing extreme temperatures over space, and that this pattern may characterize other small motile ectothermic animals with analogous natural history. The effects of cold- and heat hardening on resistance to both low and high temperature stress was examined in Drosophila melanogaster lines selected for resistance to either cold or heat. The hardening effect was positive when the hardening was of the same type as the stress in all selection regimes. The effect of cold hardening on survival after heat stress was further examined in the lines selected for cold resistance and corresponding controls. A cross-protection effect (increased heat resistance after cold hardening) was present and this effect was lower in the lines selected for resistance to cold than in the controls. The level of Hsp70 expression induced by a non-lethal cold hardening was examined, showing that cold hardening induced Hsp70 expression. The results suggest that the cross-protection effect is at least partly due to Hsp70 expression induced by cold exposure. [Copyright &y& Elsevier]

Abstract: A short exposure to a mild cold stress is sufficient to increase cold tolerance in many insects. This phenomenon, termed rapid cold hardening (RCH) expands the thermal interval that can be exploited by the insect. To investigate the possible role of altered metabolite levels during RCH, the present study used untargeted 1H NMR metabolomic profiling to examine the metabolomic response in Drosophila melanogaster during the 72h following RCH and cold shock treatment. These findings are discussed in relation to the costs and benefits of RCH that are measured in terms of survival and reproductive output. Cold shock caused a persistent disturbance of the metabolite profile that correlated well with a delayed onset of cold shock mortality. The disruption of metabolite homeostasis was smaller following RCH, where control levels were fully recovered after 72h. RCH improved both survival and reproductive output after a subsequent cold shock but the RCH treatment alone was associated with costs in terms of reduced survival and reproductive output. The most pronounced changes following the RCH treatment were elevated levels of glucose and trehalose. Although, it is difficult to discern if a change in a specific metabolite is linked to physiological processes of adaptive, neutral or detrimental nature we observed that the onset and magnitude of the increased sugar levels correlated tightly with the improved chill tolerance following RCH. These findings suggest a putative role of cryoprotectants during RCH which are discussed in the light of the existing literature on the mechanistic background of RCH. [Copyright &y& Elsevier]

Abstract: The dependency of knock-down resistance on age, from 0 to 12 days post eclosion, was studied in two lines of Drosophila melanogaster, one selected for knock-down resistance and one unselected control. Additionally, the inducible Hsp70 expression level was assessed at maintenance temperature and after a heat hardening treatment (1 h at 35 °C) at the same ages. Knock-down resistance decreased roughly linearly with age in both control and knock-down selected lines and in both sexes regardless of maintenance temperature. Hsp70 expression, however, fell only from day 0 to day 3 and was thereafter changed little. Acclimation did not result in detectable difference between lines. The effect of hardening was only significant for control flies. This suggests that the increased knock-down resistance of the selected line was reached at the expense of hardening ability. Abstract: The effects a stressful developmental temperature and maternal and grandmaternal age was investigated on plasticity of wing shape, using the Procrustes method, in two parthenogenetic strains of Drosophila mercatorum. Shape was significantly modified by both factors. Maternal and grandmaternal age were more important than temperature when compared in terms of larger Mahalanobis distances. Our results suggest that, in populations harbouring low genetic variability, the direct shape changes caused by different developmental temperatures may be smaller than indirect changes mediated through changes in age composition of the maternal and grandmaternal generation. Genomewide gene expression patterns were investigated in inbred and noninbred Drosophila melanogaster lines under benign and stressful (high temperature) environmental conditions in a highly replicated experiment using Affymetrix gene chips. We found that both heat-shock protein and metabolism genes are strongly affected by temperature stress and that genes involved in metabolism are differentially expressed in inbred compared with noninbred lines, and that this effect is accentuated after heat stress exposure. Furthermore we show that inbreeding and temperature stress cause increased between-line variance in gene expression patterns. We conclude that inbreeding and environmental stress both independently and synergistically affect gene expression patterns. Interactions between inbreeding and the environment are often observed at the phenotypic level and our results reveal some of the genes that are involved at the individual gene level. Our observation of several metabolism genes being differentially expressed in inbred lines and more so after exposure to temperature stress, together with lower fitness in the investigated inbred lines, supports the hypothesis that superiority of heterozygous individuals partly derives from increased metabolic efficiency. Abstract: Maternal age is known to be of importance for the fitness of the offspring. Few studies have, however, been able to analyse this phenomenon as an isolated effect without confounding effects through genetic variation. This difficulty can be circumvented by working with parthenogenetic organisms. We investigated the effect of maternal and grandmaternal age on wing traits, pupal survival and developmental instability (DI) in both a benign and a high temperature environment using two different parthenogenetic strains of Drosophila mercatorum. Both the maternal and grandmaternal age was found to influence all the traits. Two opposing factors seem to shape the effects of maternal age. Senescence in older mothers leads to a reduction in offspring fitness, whereas, plastic responses lead to more competitive and stress resistant offspring from older mothers. The relative importance of these factors is trait specific and is influenced by environmental factors. DI is mostly influenced by senescence whereas wing sizes are influenced mostly by plastic responses towards higher competition. This means that any analysis of fitness should take age composition of at least two generations into account. [Copyright &y& Elsevier]
