In 'Hass' avocado (Persea americana), fruit presence reduces next season flowering. Recent fruit tree studies proposed that heavy fruit load (HFL) generates an auxin (indole-3-acetic acid, IAA) signal in the buds that represses flowering. However, the nature of this signal remains unknown. Here, we investigated the effect of avocado HFL on bud IAA accumulation and flowering transition. We found that IAA–aspartate and IAA–glutamate conjugate levels were significantly higher in buds from fully loaded ('on') than low-loaded ('off') trees, hinting that free IAA levels were higher in the former. Expression analysis showed that coinciding with flowering reduction, HFL induced the floral repressor PaTFL1 , and suggested that accumulation of IAA in buds as imposed by HFL was associated with its conjugation to aspartate and glutamate and resulted both from de novo IAA synthesis and from reduced IAA export. Accordingly, experiments involving radiolabelled [14C]IAA demonstrated that HFL reduced shoot basipetal IAA transport. Finally, we confirmed the negative effects of IAA on flowering, showing that IAA and polar auxin transport blocker (2,3,5-triiodobenzoic acid) treatments delayed 'off' trees' inflorescence development, reducing their inflorescence axis and inducing PaTFL1 expression. In many fruit trees, heavy fruit load in one year reduces flowering in the following year, creating a biennial fluctuation in yield termed alternate bearing (AB). In subtropical trees, where flowering induction is mostly governed by the accumulation of chilling hours, fruit load is thought to generate a signal (AB signal) that blocks the perception of cold induction. Fruit removal during a heavy-fruit-load year is effective at inducing flowering only if performed one to a few months before the onset of the flowering induction period. We previously showed that following fruit removal, the content of the auxin indoleacetic acid (IAA) in citrus buds is reduced, suggesting that the hormone plays a role in the AB signal. Here, we demonstrate that fruit presence generates relatively strong polar auxin transport in citrus and olive stems. Upon fruit removal, polar auxin transport is reduced and allows auxin release from the bud. Furthermore, using immunolocalization, hormone, and gene expression analyses, we show that in citrus, IAA level in the bud and specifically in the apical meristem is reduced upon fruit removal. Alternate bearing (AB) is the process in fruit trees by which cycles of heavy yield (ON crop) one year are followed by a light yield (OFF crop) the next. Heavy yield usually reduces flowering intensity the following year. Despite its agricultural importance, how the developing crop influences the following year's return bloom and yield is not fully understood. It might be assumed that an 'AB signal' is generated in the fruit, or in another organ that senses fruit presence, and moves into the bud to determine its fate--flowering or vegetative growth. The bud then responds to fruit presence by altering regulatory and metabolic pathways. Determining these pathways, and when they are altered, might indicate the nature of this putative AB signal. We studied bud morphology, the expression of flowering control genes, and global gene expression in ON- and OFF-crop buds. In May, shortly after flowering and fruit set, OFF-crop buds were already significantly longer than ON-crop buds. The number of differentially expressed genes was higher in May than at the other tested time points. Processes differentially expressed between ON- and OFF-crop trees included key metabolic and regulatory pathways, such as photosynthesis and secondary metabolism. The expression of genes of trehalose metabolism and flavonoid metabolism was validated by nCounter technology, and the latter was confirmed by metabolomic analysis. Among genes induced in OFF-crop trees was one homologous to SQUAMOSA PROMOTER BINDING-LIKE (SPL), which controls juvenile-to-adult and annual phase transitions, regulated by miR156. The expression pattern of SPL-like, miR156 and other flowering control genes suggested that fruit load affects bud fate, and therefore development and metabolism, a relatively long time before the flowering induction period. Fruit tree production is challenged by climate change, which is characterized by heat waves, warmer winters, increased storms, and recurrent droughts. The technology of top netting may provide a partial solution, as it alleviates climatic effects by microclimate manipulation. The tree physiological performance is improved under the nets, with an increased productivity and quality. The application of photoselective nets, which also alter the light spectrum, may result in additional horticultural improvements. We present the results of a 5-year experimental study on Valencia oranges, examining three nets: red, pearl, and transparent. Each net was tested at three fertigation conditions: a field standard (100%, I100) and two reduced fertigation regimes, which were 80% (I80) and 60% (I60) of the standard. The average multi-annual yield under the red and pearl nets with I100 and I80 and transparent net with I100 was significantly higher than that of the control trees. While the multi-annual yield increase under the red net I80 was due to the increase in the fruit number, in other treatments, the effect was mostly due to induction in the individual fruit weight. The data presented here show that an increased productivity of orange trees grown under photoselective nets, particularly the red net, with its specific spectral properties, was achieved with a considerable water-saving effect. Top-netting technology improves crop microclimate, and its utilization in fruit tree plantations may mitigate deleterious weather effects. The technology is associated with improved tree water relations, and may result in significant water savings. Photoselective nets may provide additional, crop- and target-dependent, beneficial effects. A previous paper reported results of a five-year trial in Valencia orange, in which three nets were used, pearl, transparent and low shading red, in combination with three irrigation regimes, 100% (I100), standard commercial fertigation, and two reduced rates, 80% (I80) and 60% (I60). Among the different netting types, the combination of red nets and I80 resulted in the highest yield on a multi-annual basis. The current study provides data regarding water relations during stage II, i.e., cell expansion, of fruit development. The red net had higher radiation transmittance and canopy resistance than the pearl net. The higher resistance is an expression of the lower transpiration of the red net, monitored by sap flow, as compared to the other treatments, especially under deficient irrigation. The beneficial effect of the red net was also evidenced from multi-annual stem water potential. These data supplement the enhanced yield effect of the red net, suggesting they are associated with its spectral properties. • Water relations were tested in orange trees under three photo-selective top-netting. • Red net resulted in lower transpiration as compared to pearl and transparent nets. • Trees under the red net also displayed higher stem water potential values. • The above findings supplemented the enhanced yield effect of the red net.
