Simple Summary Neuropeptides regulate most insect biological functions. One such group of peptides, the pyrokinins (PKs), are distinguished by a C-terminal FXPRLamide. While widely distributed in most insects, they are poorly characterized in plant bugs. To address this limitation, we identified the PK transcript in the western tarnished plant bug (Lygus hesperus) and examined its expression. The Lygus PK transcript is predicted to yield three PK-like peptides but only two (LyghePKa and LyghePKb) have the characteristic C-terminal amide. The transcript is expressed throughout development and is most abundant in heads. A custom FXPRLamide antibody revealed immunoreactive cells throughout the Lygus central nervous system consistent with typical neuropeptide expression. To assess potential functional roles of the peptides, a fluorescence-based Ca2+ influx assay using cultured insect cells stably expressing a moth PK receptor was performed. LyghePKa was unable to stimulate receptor activation, whereas LyghePKb triggered a robust response. The in vivo pheromonotropic activity of the two peptides was likewise assessed using three different moth species. Like the cell culture system, only the LyghePKb peptide was active. The study suggests evolutionary divergence of the PK gene in plant bugs and provides critical insights into likely biological functions in the western tarnished plant bug. Abstract The pyrokinin (PK) family of insect neuropeptides, characterized by C termini consisting of either WFGPRLamide (i.e., PK1) or FXPRLamide (i.e., PK2), are encoded on the capa and pk genes. Although implicated in diverse biological functions, characterization of PKs in hemipteran pests has been largely limited to genomic, transcriptomic, and/or peptidomic datasets. The Lygus hesperus (western tarnished plant bug) PK transcript encodes a prepropeptide predicted to yield three PK2 FXPRLamide-like peptides with C-terminal sequences characterized by FQPRSamide (LyghePKa), FAPRLamide (LyghePKb), and a non-amidated YSPRF. The transcript is expressed throughout L. hesperus development with greatest abundance in adult heads. PRXamide-like immunoreactivity, which recognizes both pk- and capa-derived peptides, is localized to cells in the cerebral ganglia, gnathal ganglia/suboesophageal ganglion, thoracic ganglia, and abdominal ganglia. Immunoreactivity in the abdominal ganglia is largely consistent with capa-derived peptide expression, whereas the atypical fourth pair of immunoreactive cells may reflect pk-based expression. In vitro activation of a PK receptor heterologously expressed in cultured insect cells was only observed in response to LyghePKb, while no effects were observed with LyghePKa. Similarly, in vivo pheromonotropic effects were only observed following LyghePKb injections. Comparison of PK2 prepropeptides from multiple hemipterans suggests mirid-specific diversification of the pk gene.

RNAi efficiency in insects is different from species to species; some species in Coleoptera are relatively more amenable to RNA interference (RNAi) than other species. One of the major factors is the presence of dsRNA-degrading enzymes, called dsRNases, in saliva, gut, or hemolymph in insects, which degrade the double-stranded RNA (dsRNA) introduced, resulting in the low efficacy of RNAi. In this study, we report a dsRNA-degrading activity in the gut homogenates from the spotted-wing drosophila, Drosophila suzukii, by ex vivo assay. Then, we identified two Drosophila suzukii dsRNase genes, named DrosudsRNase1 and DrosudsRNase2. In silico analysis shows that the gene structures are similar to dsRNases found in other insects. When dsRNases expressed in Sf9 cells were compared for their dsRNA degrading activities, dsRNase1 was more vital than dsRNase2. Both dsRNases were expressed highly and exclusively in the gut compared to the rest of body. Also, they were highly expressed during larval and adult stages but not in embryonic and pupal stages, suggesting the dsRNases protect foreign RNA molecules received during the feeding periods. DsRNase1 was expressed at a higher level in adults, whereas dsRNase2 showed more expression in early larvae. Our study on the tissue and development-specific patterns of dsRNases provides an improved understanding of the RNAi application for the management of D. suzukii.

The discovery of new insecticides improves integrated pest management (IPM), but is usually a long high-risk process with a low probability of success. For over two decades, insect neuropeptides (NPs) and their G-protein coupled receptors (GPCRs) have been considered as biological targets for insect pest control, because they are involved in almost all physiological processes associated with insect life stages. A key roadblock to success has been the question of how large volume chemical libraries can be efficiently screened for active compounds. New genomic and proteomic tools have advanced and facilitated the development of new approaches to insecticide discovery. In this study, we report a novel GPCR-based screening technology that uses millions of short peptides randomly generated by bacteriophages, and a method using an insect Sf9 cell expression system. The fire ant is a good model system, since bioactive peptides have been identified for a specific GPCR. The novel small peptides could interfere with the target GPCR-ligand functions. Therefore, we refer to this new mechanism as “receptor interference” (RECEPTORi). The GPCR-based bioactive peptide screening method offers multiple advantages. Libraries of phage-displayed peptides (~109 peptides) are inexpensive. An insect cell-based screening system rapidly leads to target specific GPCR agonists or antagonists in weeks. Delivery of bioactive peptides to target pests can be flexible, such as topical, ingestion, and plant-incorporated protectants. A variety of GPCR targets are available, thus minimizing the development of potential insecticide resistance. This report provides the first proof-of-concept for the development of novel arthropod pest management strategies using neuropeptides, and GPCRs.

Drosophila suzukii (Matsumura) (Diptera: Drosophilidae) also known as spotted-wing drosophila (SWD), is a pest native to Southeast Asia. In the last few decades, the pest has expanded its range to affect all major European and American fruit production regions. SWD is a highly adaptive insect that is able to disperse, survive, and flourish under a range of environmental conditions. Infestation by SWD generates both direct and indirect economic impacts through yield losses, shorter shelf life of infested fruit, and increased production costs. Fresh markets, frozen berries, and fruit export programs have been impacted by the pest due to zero tolerance for fruit infestation. As SWD control programs rely heavily on insecticides, exceedance of maximum residue levels (MRLs) has also resulted in crop rejections. The economic impact of SWD has been particularly severe for organic operations, mainly due to the limited availability of effective insecticides. Integrated pest management (IPM) of SWD could significantly reduce chemical inputs but would require substantial changes to horticultural management practices. This review evaluates the most promising methods studied as part of an IPM strategy against SWD across the world. For each of the considered techniques, the effectiveness, impact, sustainability, and stage of development are discussed.

Neuromedin U (NmU) is a neuropeptide regulating diverse physiological processes. The insect homologs of vertebrate NmU are categorized as PRXamide family peptides due to their conserved C-terminal end. However, NmU homologs have been elusive in Mollusca, the second largest phylum in the animal kingdom. Here we report the first molluscan NmU/PRXamide receptor from the slug, Deroceras reticulatum. Two splicing variants of the receptor gene were functionally expressed and tested for binding with ten endogenous peptides from the slug and some insect PRXamide and vertebrate NmU peptides. Three heptapeptides (QPPLPRYa, QPPVPRYa and AVPRPRIa) triggered significant activation of the receptors, suggesting that they are true ligands for the NmU/PRXamide receptor in the slug. Synthetic peptides with structural modifications at different amino acid positions provided important insights on the core moiety of the active peptides. One receptor variant always exhibited higher binding activity than the other variant. The NmU-encoding genes were highly expressed in the slug brain, while the receptor gene was expressed at lower levels in general with relatively higher expression levels in both the brain and foot. Injection of the bioactive peptides into slugs triggered defensive behavior such as copious mucus secretion and a range of other anomalous behaviors including immobilization, suggesting their role in important physiological functions.

Simple Summary Spotted-wing drosophila, Drosophila suzukii, is an invasive pest of soft-skinned fruits. Adult female flies oviposit, or lay eggs, into fruits where the larvae develop, making infested fruit unmarketable. The flies rely on alternative hosts, both cultivated and wild, to survive and maintain populations throughout the year. Better understanding of how the flies migrate between different hosts could be beneficial to improving management of the pest in crops. This study demonstrates potential to discriminate larval host of adult flies by analysis of fatty acids carried from the larvae to the adult stage in the body using a machine learning algorithm as an alternative to linear discriminant methods. Our study shows that fatty acids in adult flies can be used to determine larval host and that the machine learning algorithm can perform the discriminant analysis without making any assumptions about the data. Abstract Drosophila suzukii is a severe economic invasive pest of soft-skinned fruit crops. Management typically requires killing gravid adult female flies with insecticides to prevent damage resulting from oviposition and larval development. Fruits from cultivated and uncultivated host plants are used by the flies for reproduction at different times of the year, and knowledge of D. suzukii seasonal host plant use and movement patterns could be better exploited to protect vulnerable crops. Rearing and various marking methodologies for tracking movement patterns of D. suzukii across different landscapes have been used to better understand host use and movement of the pest. In this study, we report on potential to determine larval host for adult D. suzukii using their fatty acid profile or signature, and to use larval diet as an internal marker for adult flies in release-recapture experiments. Fatty acids can pass efficiently through trophic levels unmodified, and insects are constrained in the ability to synthesize fatty acids and may acquire them through diet. In many holometabolous insects, lipids acquired in the larval stage carry over to the adult stage. We tested the ability of a machine learning algorithm to discriminate adult D. suzukii reared from susceptible small fruit crops (blueberry, strawberry, blackberry and raspberry) and laboratory diet based on the fatty acid profile of adult flies. We found that fatty acid components in adult flies were significantly different when flies were reared on different hosts, and the machine learning algorithm was highly successful in correctly classifying flies according to their larval host based on fatty acid profile.

Drosophila suzukii differs from other members of the genus Drosophila in its host preference and oviposition behavior. The flies are attracted to ripening fruits, and females have a serrated ovipositor enabling eggs to be laid inside the fruit. In addition to its huge economic impact, its unique chemoecological, morphological, and physiological characteristics have garnered considerable research interests. In this study, we analyzed D. suzukii antennal transcriptomes to identify sex-biased genes by comparison of differential gene expressions between male antennae (MA) and female antennae (FA). Among 13,583 total genes of the fly genome, 11,787 genes were expressed in either MA or FA. There are only 132 genes (9 in MA, 7 in FA, and 116 in both, FPKM >1) were expressed in antennae exclusively, and 2,570 genes (9 in MA, 0 in FA, and 2,561 in both) were enriched in antennae containing 185 and 113 sex-biased genes in MA and FA, respectively. Interestingly, many immune-related genes were highly expressed in MA, whereas several chemosensory genes were at high rank in FA. We identified 27 sex-biased chemosensory genes including odorant and gustatory receptors, odorant-binding proteins, chemosensory proteins, ionotropic receptors, and cytochrome P450s, and validated the gene expressions using quantitative real-time PCR. The highly expressed sex-biased genes in antennae are likely involved in the fly specific mating, host-finding behaviors, or sex-specific functions. The molecular results demonstrated here will facilitate to find the unique chemoreception of D. suzukii, as well as on the development of new management strategies for this pest.

Drosophila suzukii, spotted-wing drosophila (SWD), is a fly pest of small fruits and cherries often managed by chemical insecticides, and finding a convenient, human-safe alternative is a challenge. Erythritol, a non-caloric sugar, is safe for humans and toxic to D. suzukii by causing an osmotic imbalance. Combining erythritol with sucrose as a phagostimulant can enhance fly mortality. However, sucrose is sticky when applied on plants and provides flies with nutritional carbohydrate if a non-lethal dose is ingested. Therefore, our objectives were to find a sucrose alternative, a non-caloric sugar that also has phagostimulative properties, where the formulation will enhance mortality, and reduce daily oviposition rates before females die. Through capillary no-choice and choice experiments, flies fed on 0.1 M sucralose (Sul) readily, and flies consumed more 1.5 M erythritol: 0.1 M sucralose (E:Sul) formulation than erythritol alone. In eleven different mortality assays, E:Sul induced mortality as did the 1.5 M erythritol: 0.5 M sucrose (E:S) formulation, while E:Sul was more successful than E:S when D. suzukii could also drink water to offset water loss. Sucralose (0.1 M) itself had modest insecticidal properties, but when combined with erythritol it increased the insecticidal effect. Both E:Sul and E:S formulations reduced oviposition when it was directly fed to D. suzukii in a cotton wick or applied on blueberry fruit. The latter suggests that the sugary coating on fruits has a deterrent effect. Given the potential of E:Sul, additional studies on the effect of sucralose on fly osmolar regulation, sugar metabolism, and field efficacy trials are underway to provide growers an efficacious tool.

In this study, we investigated the effects of non-nutritive sugars and sugar alcohols on the survivorship of spotted wing drosophila, Drosophila suzukii, and found erythritol and erythrose as potentially insecticidal to the fly. In a dose-dependent study, erythritol and erythrose significantly reduced fly longevity, with 100% mortality with 1, 0.5, 0.1 & 0.05M doses after feeding for 7days. When sucrose and erythritol solutions were provided separately to flies for 7days, there was no effect on survivorship regardless of erythritol concentrations. However, with a serial combination of sucrose and erythritol solutions, fly survivorship was significantly decreased for the same period. Also, the higher dose of erythritol regardless of the sucrose dose combined showed greater mortality. In a no-choice assay, D. suzukii ingested more erythritol than sucrose or water, indicating the fly continuously fed on erythritol for 72h. Also under no-choice conditions, erythritol and sucrose-fed flies gained more weight than water-fed flies. However, in two-choice assays, the amount of erythritol ingested was less than sucrose or water. Total sugar and glycogen levels among erythritol and erythrose-fed flies were significantly less than mannitol, sorbitol, xylitol, and sucrose-fed flies after 48h. This indicates that these two non-nutritive sugars can't be used a substrate for enzymes involved in sugar metabolism. Although the metabolism of erythritol and erythrose is unknown in insects, the mortality of D. suzukii flies ingesting these sugars might be caused by two potential physiological changes. The fly is starved by feeding of non-metabolizable erythritol and erythrose, or experiences abnormally high osmotic pressure in the hemolymph with erythritol molecules diffused from the midgut. Non-nutritive sugars might be used as an insecticide alone or combined with conventional or biological insecticides to enhance efficacy. If other sugar sources are present, a palatable sugar might be mixed with erythritol to elicit feeding.

The codling moth, Cydia pomonella, is a worldwide pest of pome fruits. Neuropeptides regulate most physiological functions in insects and represent new targets for the development of control agents. The only neuropeptides reported from the codling moth to date are the allatostatin A family peptides. To identify other neuropeptides and peptide hormones from codling moth, we analyzed head transcriptomes, identified 50 transcripts, and predicted 120 prepropeptides for the codling moth neuropeptides and peptide hormones. All transcripts were amplified, and these sequences were verified. One of the notable findings in this study is that diapause hormones (DHs) reported from Tortricid moths, including the codling moth, do not have the WFGPRL sequence in C-terminal ends in the pban genes. The C-terminal motif is critical to characterize insect DH peptides, and always conserved in pban/dh genes in Lepidoptera and many insect orders. Interestingly, the WFGPRL sequence was produced only from the capa gene in the codling moth. The allatostatin A-family encoding transcript predicted nine peptides, seven of which, as expected, are identical to those previously isolated from the moth. We also identified new codling moth orthologs of insect neuropeptides including CCHamides, allatostatin CC, RYamides, and natalisins. The information provided in this study will benefit future codling moth investigations using peptidoproteomics to determine peptide presence and functions.

Drosophila suzukii, the spotted wing drosophila (SWD), is currently a major pest that causes severe economic losses to thin-skinned, small fruit growers in North America and Europe. The monitoring and early detection of SWD in the field is of the utmost importance for its proper management. Although SWD traps using various attractants have been developed and used for SWD monitoring in the field, they are not specific to SWD; consequently, other insects, including Drosophilidae flies that share high morphological similarities with SWD, are also easily trapped. Thus, all captured flies need to be identified under microscopes by an expert in fly morphological characteristics. In this study, we developed a simple molecular detection tool that rapidly and accurately identifies SWD without sophisticated instruments or expertise. We first identified a gene, Ds10_00012111, that is present in the SWD genome but not in that of any other insect species. Then, we developed the loop-mediated isothermal amplification (LAMP) assay, which was designed based on genomic DNA of the Ds10_00012111 gene. This LAMP assay detected only genomic DNA from SWD—not from Drosophila melanogaster. The minimum amount of genomic DNA required for the LAMP assay was 1.0 picogram, and the duration of this assay at temperatures ranging from 58 °C to 62 °C was 30 min. In addition, this assay could detect genomic DNA in SWD geographical strains collected from 8 different locations in Asia, Europe, Hawaii, and the USA. Our LAMP assay could be a useful detection tool for identifying SWD rapidly in the field.

The Noctuidae are one of the most speciose moth families and include the genera Helicoverpa and Heliothis. Females use (Z)-11-hexadecenal as the major component of their sex pheromones except for Helicoverpa assulta and Helicoverpa gelotopoeon, both of which utilize (Z)-9-hexadecenal. The minor compounds found in heliothine sex pheromone glands vary with species, but hexadecanal has been found in the pheromone gland of almost all heliothine females so far investigated. In this study, we found a large amount (0.5-1.5 μg) of hexadecanal and octadecanal on the legs of males of four heliothine species, Helicoverpa zea, Helicoverpa armigera, H. assulta, and Heliothis virescens. The hexadecanal was found on and released from the tarsi, and was in much lower levels or not detected on the remaining parts of the leg (tibia, femur, trochanter, and coxa). Lower amounts (0.05-0.5 μg) of hexadecanal were found on female tarsi. This is the first known sex pheromone compound to be identified from the legs of nocturnal moths. Large amounts of butyrate esters (about 16 μg) also were found on tarsi of males with lower amounts on female tarsi. Males deposited the butyrate esters while walking on a glass surface. Decapitation did not reduce the levels of hexadecanal on the tarsi of H. zea males, indicating that hexadecanal production is not under the same neuroendocrine regulation system as the production of female sex pheromone. Based on electroantennogram studies, female antennae had a relatively high response to hexadecanal compared to male antennae. We consider the possible role of aldehydes and butyrate esters as courtship signals in heliothine moths.

Previous studies have demonstrated various combinations of non-nutritive erythritol and sucrose having detrimental effects on Drosophila suzukii (Matsumura). Fly mortality is likely caused by 1) starvation from feeding on non-metabolizable erythritol; and 2) physiological imbalance with abnormally high osmotic pressure in the hemolymph. While erythritol kills D. suzukii in controlled environments, flies in the field can access naturally-occurring sugar sources. We evaluated fly mortality in the presence or absence of wounded fruits, and an erythritol mixture of 2.0 M erythritol:0.5 M sucrose (E+S), or erythritol- and sucrose-only controls. When provided E+S, survival was consistently lower than sucrose controls with/out wounded fruit, suggesting that this mixture still has a detrimental effect in the presence of competing sugar sources. Our second study examined the effects of diet on fecundity and egg load of female D. suzukii. Females laid fewer eggs on blueberries when fed E+S or erythritol-only than sucrose. Unexpectedly, females fed E+S had more ovarial eggs than sucrose-fed females, suggesting that erythritol might inhibit D. suzukii laying eggs. Lastly, we evaluated honey bee survivorship by enclosing bees with one of four diets in a cage. The erythritol mixture had no discernible impact on adult survivorship during 7 d.

Insect neuropeptides represent more than 90% of all insect hormones. The pheromone biosynthesis activating neuropeptide (PBAN)/pyrokinin family is a major group of insect neuropeptides. These neuropeptides regulate a variety of biological functions from embryo to adult in moths including, sex pheromone biosynthesis and diapause. Other functions are yet to be determined. The identification of suitable target genes is most important for the successful application of RNA interference (RNAi) for pest insect control. Insect neuropeptide genes including PBAN are known to have multiple functions and could be a good target for RNAi suppression. In this study, we selected the PBAN gene and its neuropeptide products as an RNAi target for two economically important moth species, the corn earworm, Helicoverpa zea (Boddie), and the tobacco budworm, Heliothis virescens (Fabricius). We investigated RNAi effects on immature moths that had ingested the specific double-stranded RNA (dsRNA) starting at the first instar larva through pupation. We report that RNAi treatments resulted in delay of larval growth, interference of pupal development, and mortality in the two pest moths. In addition, we selected small interfering RNAs (siRNAs) to determine if they have negative phenotypic effects similar to their full-length RNAi parents. This is one of the few examples of negative RNAi effects on lepidopteran pests via feeding and suggests possible RNAi-based control of pest moths.

Previously, we studied various combinations of non-nutritive sugars including erythritol and erythrose having a potentially insecticidal effect on Drosophila suzukii. The study suggested two potential physiological changes causing fly mortality: 1) starvation from the feeding of non-metabolizable erythritol and erythrose; 2) abnormal osmotic pressure increased in the hemolymph with erythritol transported from the midgut. In the present study, sucrose and erythritol were applied to blueberries and effects of these combinations on fly mortality and fecundity were monitored in the lab and greenhouse. In the lab, two sucrose/erythritol formulations (0.5M sucrose/2M erythritol, 1M sucrose/2M erythritol) resulted in the highest mortality and the lowest fecundity among D. suzukii adults. Two formulations, therefore, were selected for further evaluation with blueberry bushes and fruits in the greenhouse; fly survival with 0.5M sucrose/2M erythritol was significantly lower than 1M sucrose/2M erythritol for 7days. Unlike the smaller container, mortality occurred faster in the greenhouse probably because flies moved more in the bigger cage accelerating the exhaustion of energy reserves in the body. We examined presence of erythritol in the hemolymph and frass to determine the nutritional metabolism and absorption of erythritol in D. suzukii. Unlike sucrose, a large amount of erythritol was observed in the hemolymph of the fly that ingested 0.5M sucrose/0.5M erythritol. Erythritol was also found in the frass of the same fly. The results imply that erythritol might be directly transported from the midgut without being metabolized and stored, but is accumulated in the hemolymph which in turn elevates the osmotic pressure in the fly hemolymph. For practical application, the sucrose/erythritol combination would be more effective than erythritol alone because the combination tastes sweeter to elicit more feeding. This erythritol formulation can be a potential insecticide used alone or as a delivery agent combined with conventional or biological insecticides to enhance their efficacy.

Neuropeptides control various physiological functions and constitute more than 90% of insect hormones. The pheromone biosynthesis activating neuropeptide (PBAN)/pyrokinin family is a major group of insect neuropeptides and is well conserved in Insecta. This family of peptides has at least two closely related G-protein-coupled receptors (GPCRs) activated by PBAN and a diapause hormone (DH). They have been shown to control several biological activities including pheromone production and diapause induction in moths. However, beyond some moth species, the biological function(s) of PBAN/pyrokinin peptides are largely unknown although these peptides are found in all insects. In this study we identified and characterized PBAN/pyrokinin peptides and corresponding GPCRs from the mosquito, Aedes aegypti. Ae. aegypti PBAN mRNA encodes four putative peptides including PBAN and DH, and is expressed in females and males during all life stages. The PBAN receptor (PBAN-R) and the DH receptor (DH-R) were functionally expressed and confirmed through binding assays with PBAN and DH peptides. These receptors are differentially expressed from eggs to adults with the relative gene expression of the PBAN-R significantly lower during the 4th instar larval (L4) and pupal (P1-P2) stages compared to the 2nd and 3rd instar larval stages (L2 and L3). However, DH-R expression level is consistently 4-10 times higher than the PBAN-R in the same period, suggesting that PBAN-R is downregulated in the late larval and pupal stages, whereas DH-R stays upregulated throughout all developmental stages. PBAN/pyrokinin mRNA expression remains high in all stages since it produces PBAN and DH peptides. This study provides the foundation for determining the function(s) of the PBAN/pyrokinin peptides in mosquitoes and establishes data critical to the development of methods for disruption of these hormone actions as a novel strategy for mosquito control.

RNA interference (RNAi) is a convenient tool to identify and characterize biological functions in organisms. Recently, it has become an alternative to chemical insecticides as a biologically based control agent. This promising technology has the potential to avoid many problems associated with conventional chemical insecticides. In order for RNAi application to be practical for field use, a major hurdle is the development of a cost-effective system of double-stranded RNA (dsRNA) production for a large quantity of dsRNA. A handful of research reports has demonstrated microbial-based dsRNA production using L4440 vector and HT115 (DE3) Escherichia coli for application to vertebrate and invertebrate systems. However, the dsRNA yield, production efficiency, and biological purity from this in vitro system is still unclear. Thus, our study detailed biochemical and molecular tools for large-scale dsRNA production using the microbial system and investigated the production efficiency and yield of crude and purified dsRNAs. An unrelated insect gene, green fluorescent protein (GFP), and an insect neuropeptide gene, pyrokinin (PK) identified from Drosophila suzukii, were used to construct the recombinant L4440 to be expressed in the HT115 (DE3) cell. A considerable amount of dsRNA, 19.5 µg/mL of liquid culture, was isolated using ultrasonic disruption followed by phenol extraction. The sonication method was further evaluated to extract crude dsRNA without the additional phenol extraction and nuclease treatments and also to reduce potential bacterial viability. The results suggest that the ultrasonic method saved time and costs to isolate crude dsRNA directly from large volumes of cell culture without E coli contamination. We investigated whether the injection of PK dsRNA into flies resulted in increased adult mortality, but it was not statistically significant at 95% confidence level. In this study, the microbial-based dsRNA production has potential for applied RNAi technology to complement current insect pest management practices.

The pyrokinin/pheromone-biosynthesis-activating neuropeptide (PBAN) family of peptides found in insects is characterized by a 5-amino-acid C-terminal sequence, FXPRLamide. The pentapeptide is the active core required for diverse physiological functions, including the stimulation of pheromone biosynthesis in female moths, muscle contraction, induction of embryonic diapause, melanization, acceleration of puparium formation, and termination of pupal diapause. We have used immunocytochemical techniques to demonstrate the presence of pyrokinin/PBAN-like peptides in the central nervous system of the fire ant, Solenopsis invicta. Polyclonal antisera against the C-terminal end of PBAN have revealed the location of the peptide-producing cell bodies and axons in the central nervous system. Immunoreactive material is detectable in at least three groups of neurons in the subesophageal ganglion and corpora cardiaca of all adult sexual forms. The ventral nerve cord of adults consists of two segmented thoracic ganglia and four segmented abdominal ganglia. Two immunoreactive pairs of neurons are present in the thoracic ganglia, and three neuron pairs in each of the first three abdominal ganglia. The terminal abdominal ganglion has no immunoreactive neurons. PBAN immunoreactive material found in abdominal neurons appears to be projected to perisympathetic organs connected to the abdominal ganglia. These results indicate that the fire ant nervous system contains pyrokinin/PBAN-like peptides, and that these peptides are released into the hemolymph. In support of our immunocytochemical results, significant pheromonotropic activity is found in fire ant brain-subesophageal ganglion extracts from all adult fire ant forms (queens, female and male alates, and workers) when extracts are injected into decapitated females of Helicoverpa zea. This is the first demonstration of the presence of pyrokinin/PBAN-like peptides and pheromonotropic activity in an ant species.

Spotted wing drosophila, Drosophila suzukii Matsumura, is an invasive and economically damaging pest in Europe and North America. The females have a serrated ovipositor that enables them to infest almost all ripening small fruits. To understand the physiological and metabolic basis of spotted wing drosophila food preferences for healthy ripening fruits, we investigated the biological and biochemical characteristics of spotted wing drosophila and compared them with those of Drosophila melanogaster Meigen. We found that the susceptibility to oxidative stressors was significantly increased in spotted wing drosophila compared with those of D. melanogaster. In addition, we found that spotted wing drosophila had significantly reduced glutathione-S transferase (GST) activity and gene numbers. Furthermore, fructose concentrations found in spotted wing drosophila were significantly lower than those of D. melanogaster. Our data strongly suggest that the altered food preferences of spotted wing drosophila may stem from evolutionary adaptations to fresh foods accompanied by alterations in carbohydrate metabolism and GST activities.

Moth sex-pheromone biosynthesis follows a circadian cycle, which is cued by the release of the neurohormone pheromone biosynthesis activating neuropeptide (PBAN) to the hemolymph. PBAN binds to a G protein-coupled receptor (GPCR), in pheromone glands, (PG) initially identified by us in Helicoverpa zea moths (HezPBAN-R). In this study, the sequences of the seven transmembrane helices of HezPBAN-R were identified, built, packed and oriented correctly after multiple sequence alignment of the HezPBAN-R and several other GPCRs using the X-ray structure of rhodopsin as a template. Molecular dynamics simulations were run on three different beta-turn types of the C-terminal hexapeptide of PBAN and the results clustered into 12 structurally distinct groups. The lowest energy conformation from each group was used for computer-simulated docking with the model of the HezPBAN-R. Highest scoring complexes were examined and putative binding sites were identified. Experimental studies, using in vitro PG, revealed lower levels of pheromonotropic activity when challenged with pyrokinin-like peptides than with HezPBAN as ligand. Thus, the Drosophila melanogaster pyrokinin-1 receptor (CG9918) was chosen to create chimera receptors by exchanging between the three extracellular loops of the HezPBAN-R and the CG9918 for in silico mutagenesis experiments. The predicted docking model was validated with experimental data obtained from expressed chimera receptors in Sf9 cells.

Pheromone biosynthesis-activating neuropeptide (PBAN) is a peptide used by a variety of moths to regulate pheromone production. Pyrokinins are peptides that activate muscle contraction in a variety of insects. These peptides have a common FXPRLamide C-terminal ending that is required for activity. Receptors have been identified from a moth and Drosophila as belonging to the rhodopsin family of G-protein coupled receptors (GPCRs) with sequence similarity to neuromedin U receptors from vertebrates. No insect GPCR has been characterized with regard to role of extracellular domains required for peptide binding and receptor activation. To begin characterizing these GPCRs we created chimera receptors using a PBAN-receptor from a moth and pyrokinin-receptors from Drosophila where extracellular domains were swapped. The N-terminal of the moth GPCR has two N-glycosylation sites that when replaced with glutamines, activity was reduced but not absent, indicating these sites contribute to receptor stability. Activity was greatly reduced by replacing the 2nd extracellular loop that has an N-glycosylation site and a cysteine that can form a disulfide bridge with a cysteine at the beginning of the 3rd transmembrane domain. Exchange of the 3rd extracellular loop between the moth and Drosophila receptor resulted in differential activation by PBAN or a diapause hormone peptide. This result indicates that the 3rd extracellular loop is directly involved in peptide ligand recognition. Results are discussed in context of the structural features of insect GPCRs that are required for receptor activation as compared to vertebrate receptors.

Fatty acid synthesis produces long-chain fatty acids that are principal forms of stored energy and essential constituents of cellular membrane lipids. In animals fatty acid synthesis is catalyzed by fatty acid synthase (FAS) from acetyl-coenyzyme A (CoA) and malonyl-CoA. Cerulenin and C75, potent FAS inhibitors, can inhibit feeding in mammals. Using these inhibitors we examined the effect of feeding inhibition during H. zea larval stage. Growth of larvae injected (30 μg/g body weight) with C75 or cerulenin was significantly delayed during the first 8 hrs after injection, but recovered to normal levels within 20 hrs. During the first 8 hr period, the amount of consumed diet in the inhibitor treated larvae was significantly less than the control group. The retardation of larval development could be caused from the reduction of food intake after injection of the inhibitor. The result indicates that C75 or cerulenin inhibits fatty acid synthesis, resulting in feeding suppression in the larval moth as demonstrated in vertebrates. Pheromone production was significantly decreased in the isolated pheromone gland of H. zea females treated with FAS inhibitors. Pheromone production was inhibited by blocking fatty acid synthesis, even though PBAN stimulated pheromone biosynthesis. After topical application of D3-16: Acid to pheromone glands the relative labeled pheromone amount was increased when the gland was incubated with C75. This result indicates that a part of the pheromone amount could be synthesized from 16: Acid directly when fatty acid synthesis was blocked. These results indicate that the inhibitors have a potential possibility to control insect feeding activity and inhibit pheromone biosynthesis in moths.

The pyrokinin/pheromone biosynthesis activating neuropeptide (PBAN) family of peptides found in insects is characterized by a 5-amino-acid C-terminal sequence, FXPRLamide. The pentapeptide is the active core required for diverse physiological functions, including stimulation of pheromone biosynthesis in female moths, stimulation of muscle contraction, induction of embryonic diapause in Bombyx mori, and stimulation of melanization in some larval moths. Recently, this family of peptides has been implicated in accelerating the formation of the puparium in a dipteran. Using bioassay and immunocytochemical techniques, we demonstrate the presence of pyrokinin/PBAN-like peptides in the central nervous system of Drosophila melanogaster. Pheromonotropic activity was shown in the moths Helicoverpa zea and Helicoverpa armigera by using dissected larval nervous systems and adult heads and bodies of D. melanogaster. Polyclonal antisera against the C-terminal ending of PBAN revealed the location of cell bodies and axons in the central nervous systems of larval and adult flies. Immunoreactive material was detected in at least three groups of neurons in the subesophageal ganglion of 3rd instar larvae, pupae, and adults. The ring gland of both larvae and adults contained immunoreactivity. Adult brain-subesophageal ganglion complex possessed additional neurons. The fused ventral ganglia of both larvae and adults contained three pairs of neurons that sent their axons to a neurohemal organ connected to the abdominal nervous system. These results indicate that the D. melanogaster nervous system contains pyrokinin/PBAN-like peptides and that these peptides could be released into the hemolymph.

Sex pheromone biosynthesis is induced in many moths by a neuropeptide, PBAN, consisting of 33-amino acid amidated at the C-terminus. The present study is concerned with cloning and characterizing the partial sequence of Hea-PBAN cDNA which is isolated from the brain and suboesophageal ganglion complex (Br-Sg) of Helicoverpa armigera adults. From the cDNA sequence, it can be predicted that the cDNA has a PBAN domain with 33 amino acids, LSDDMPATPADQEMYRQDPEQIDSRTKYFSPRL, with FSPRL amidated at the C-terminus. The amino acid sequence of predicted peptides including the PBAN, is identical to that of H. zea and H. assulta.

Insect neuropeptide hormones represent more than 90% of all insect hormones. The PBAN/pyrokinin family is a major group of insect neuropeptides, and they are expected to be found from all insect groups. These species-specific neuropeptides have been shown to have a variety of functions from embryo to adult. PBAN is well understood in moth species relative to sex pheromone biosynthesis, but other potential functions are yet to be determined. Recently, we focused on defining the PBAN gene and peptides in fire ants in preparation for an investigation of their function(s). RNA interference (RNAi) technology is a convenient tool to investigate unknown physiological functions in insects, and it is now an emerging method for development of novel biologically-based control agents as alternatives to insecticides. This could be a paradigm shift that will avoid many problems associated with conventional chemical insecticides. In this study, we selected the PBAN gene and its neuropeptide products as an RNAi target from two insect groups; a social insect, the fire ant (Solenopsis invicta) and a non-social insect, the corn earworm (Helicoverpa zea). Both insects are economically important pests. We report negative impacts after PBAN dsRNA treatment to suppress PBAN gene transcription during developmental and adult stages of both species, e.g. increased adult and larval mortality, delayed pupal development and decreased sex pheromone production in the moth. This is an important first step in determining the multiple functions of the PBAN gene in these two insects. This work illustrates the variety of phenotypic effects observed after RNAi silencing of the PBAN gene and suggests the possibility of novel biologically-based insect pest control methods.

Neuropeptides are the largest group of insect hormones. They are produced in the central and peripheral nervous systems and affect insect development, reproduction, feeding and behavior. A variety of neuropeptide families have been identified in insects. One of these families is the PBAN/pyrokinin family defined by a common FXPRLamide or similar amino acid fragment at the C-terminal end. These peptides, found in all insects studied thus far, have been conserved throughout evolution. The most well studied physiological function is regulation of moth sex pheromone biosynthesis through the Pheromone Biosynthesis Activating Neurohormone (PBAN), although several developmental functions have also been reported. Over the past years we have extended knowledge of the PBAN/pyrokinin family of peptides to ants, focusing mainly on the fire ant, Solenopsis invicta. The fire ant is one of the most studied social insects and over the last 60 years a great deal has been learned about many aspects of this ant, including the behaviors and chemistry of pheromone communication. However, virtually nothing is known about the regulation of these pheromone systems. Recently, we demonstrated the presence of PBAN/pyrokinin immunoreactive neurons in the fire ant, and identified and characterized PBAN and additional neuropeptides. We have mapped the fire ant PBAN gene structure and determined the tissue expression level in the central nervous system of the ant. We review here our research to date on the molecular structure and diversity of ant PBAN/pyrokinin peptides in preparation for determining the function of the neuropeptides in ants and other social insects.

Neuropeptide hormones produced by neurosecretory cells in the central or peripheral nervous systems regulate various physiological and behavioral events during insect development and reproduction. PBAN/Pyrokinin is a major neuropeptide family, characterized by a 5-amino-acid C-terminal sequence, FXPRLamide. This family of peptides has been implicated in regulating various physiological functions including, pheromone biosynthesis, muscle contraction, diapause induction or termination, melanization, and puparium formation in different insect species. In the present study, we report a new member of the PBAN family from the red imported fire ant, Solenopsis invicta, Soi-PBAN, composed of 26-AA (GSGEDLSYGDAYEVDEDDHPLFVPRL). Three additional peptides were deduced from Soi-PBAN cDNA: 15-AA (TSQDIASGMWFGPRL), 8-AA (QPQFTPRL) and 9-AA (LPWIPSPRL), that correspond to diapause hormone (DH), beta-neuropeptide (NP), and gamma-NP, which are found in many lepidopteran moths. Five peptides, DH, alpha, beta, gamma NPs, and PBAN are encoded from PBAN genes of lepidopteran moths, but in the fire ant the alpha-NP is missing. Each of the four synthetic peptides from the fire ant Soi-PBAN cDNA showed significant pheromonotropic activity in a moth model, indicating that these peptides are cross-reactive. Soi-beta-NP induced the highest amount of pheromone production of the four peptides evaluated. The Soi-DH homologue had the lowest pheromonotropic activity, but was still significantly greater than control values. When the deduced amino acid sequences (entire ORF domains) from Soi-PBAN cDNA were compared with other known sequences, the fire ant was most similar to the honey bee, but phylogenetically distant from moth and beetle species. Soi-PBAN (26-AA) unlike the other three peptides shows a low degree of sequence identity with honeybee PBAN (33-AA). Based on the amino acid sequences encoded from insect PBAN genes identified to date, neuropeptide diversity is correlated with the taxonomic or phylogenetic classification of Insecta. From the present study we report the first neuropeptide identified and characterized from the central nervous system of Formicidae.

Pheromone-biosynthesis-activating neuropeptide (PBAN) regulates sex pheromone production in many female moths. PBAN-like peptides, with common FXPRLamide C-terminals are found in other insect groups where they have other functions. The ubiquity and multifunctional nature of the pyrokinin/PBAN family of peptides suggests that the PBAN receptor proteins could also be present in a variety of insect tissues with alternative functions from that of sex pheromone biosynthesis. Previously we showed the presence of the PBAN-R in Helicoverpa armigera at the protein level. In the present study we confirm the similarities between the two Helicoverpa species: armigera and zea by (1) demonstrating the presence of the receptor protein in Sf9 cells, cloned to express the HezPBAN receptor, as compared with the endogenous receptor protein, previously shown in H. armigera pheromone glands, and (2) by identifying the nucleotide sequence of the PBAN-R from mRNA of H. armigera pheromone glands. Sequences of the two Helicoverpa spp. are 98% identical with most changes taking place in the 3'-end. We demonstrate the spatial distribution of the PBAN receptor protein in membranes of H. armigera brain (Br), thoracic ganglion (TG) and ventral nerve cord (VNC). We also demonstrate the presence and differential expression of the PBAN receptor gene (using reverse transcription-polymerase chain reaction and reverse transcription-quantitative real-time polymerase chain reaction, respectively) in the neural tissues (Br, TG and VNC) of adult H. armigera female moths as compared with its presence in pheromone glands. Surprisingly, the gene for the PBAN receptor is also detected in the male tissue homologous to the female pheromone gland, the aedeagus, although the protein is undetectable and PBAN does not induce physiological (pheromone production) or cellular (cyclic-adenosine monophosphate production) responses in this tissue. Our findings indicate that PBAN or PBAN-like receptors are present in the neural tissues and may represent a neurotransmitter-like function for PBAN-like peptides. In addition, the surprising discovery of the presence of the gene encoding the PBAN receptor in the male homologous tissue, but its absence at the protein level, launches opportunities for studying molecular regulation pathways and the evolution of these G protein coupled receptors (GPCRs).

Pheromone biosynthesis-activating neuropeptide (PBAN), a peptide produced by the subesophageal ganglion, is used by a variety of moths to regulate pheromone production. PBAN acts directly on pheromone gland cells by using calcium and cAMP as second messengers. We have identified a gene encoding a G protein-coupled receptor (GPCR) from pheromone glands of the female moth Helicoverpa zea . The gene was identified based on sequence identity to a group of GPCRs from Drosophila that are homologous to neuromedin U receptors in vertebrates. The full-length PBAN receptor was subsequently cloned, expressed in Sf9 insect cells, and shown to mobilize calcium in response to PBAN. This response was dose-dependent (EC 50 = 25 nM) with a maximum response at 300 nM and a minimal observable response at 10 nM. Four additional peptides produced by the PBAN-encoding gene were also tested for activity, and it was determined that three had similar activity to PBAN and the other was slightly less active. Peptides belonging to the same family as PBAN, namely pyrokinins, as well as the vertebrate neuromedin U peptide also induced a calcium response. We have identified a GPCR for the PBAN/pyrokinin family of peptides with a known function of stimulating pheromone biosynthesis in female moths. It is related to several receptors from insects ( Drosophila and Anopheles ) and to neuromedin U and ghrelin receptors from vertebrates.

Our understanding of insect chemical communication including pheromone identification, synthesis, and their role in behavior has advanced tremendously over the last half-century. However, endocrine regulation of pheromone biosynthesis has progressed slowly due to the complexity of direct and/or indirect hormonal activation of the biosynthetic cascades resulting in insect pheromones. Over 20 years ago, a neurohormone, pheromone biosynthesis activating neuropeptide (PBAN) was identified that stimulated sex pheromone biosynthesis in a lepidopteran moth. Since then, the physiological role, target site, and signal transduction of PBAN has become well understood for sex pheromone biosynthesis in moths. Despite that PBAN-like peptides (∼200) have been identified from various insect Orders, their role in pheromone regulation had not expanded to the other insect groups except for Lepidoptera. Here, we report that trail pheromone biosynthesis in the Dufour's gland (DG) of the fire ant, Solenopsis invicta, is regulated by PBAN. RNAi knock down of PBAN gene (in subesophageal ganglia) or PBAN receptor gene (in DG) expression inhibited trail pheromone biosynthesis. Reduced trail pheromone was documented analytically and through a behavioral bioassay. Extension of PBAN's role in pheromone biosynthesis to a new target insect, mode of action, and behavioral function will renew research efforts on the involvement of PBAN in pheromone biosynthesis in Insecta.

While carabid beetles have been shown to feed on a variety of crop pests, little is known about their species assemblages in US annual ryegrass crops, where invertebrate pests, particularly slugs, lepidopteran larvae and craneflies, incur major financial costs. This study assesses the biological control potential of carabid beetles for autumn- and winter-active pests in annual ryegrass grown for seed by: (a) investigating the spatial and temporal overlap of carabids with key pests, and (b) molecular gut content analysis using qPCR. Introduced Nebria brevicollis was the only common carabid that was active during pest emergence in autumn, with 18.6% and 8.3% of N. brevicollis collected between September and October testing positive for lepidopteran and cranefly DNA, respectively, but only 1.7% testing positive for slug DNA. While pest DNA was also detected in the guts of the other common carabid species&mdash, Agonum muelleri, Calosoma cancellatum and Poecilus laetulus&mdash, these were active only during spring and summer, when crop damage by pests is less critical. None of the four carabid species was affected by disk tilling and only N. brevicollis was significantly associated with a vegetated field margin. However, as its impact on native ecosystems is unknown, we do not recommend managing for this species.
