The global concerns regarding the emergence of fungicide-resistant strains and the impact of the excessive use of fungicidal practises on our health, food, and environment have increased, leading to a demand for alternative clean green technologies as treatments. Photosensitization is a treatment that utilises a photosensitiser, light and oxygen to cause cell damage to microorganisms. The effect of photosensitization mediated by curcumin on Aspergillus niger, Aspergillus flavus, Penicillium griseofulvum, Penicillium chrysogenum, Fusarium oxysporum, Candida albicans and Zygosaccharomyces bailii was investigated using three methods. The viability of spores/cells suspended in aqueous buffer using different concentrations of curcumin solution (100-1000μM) and light dose (0, 24, 48, 72 and 96J/cm 2 ) were determined. Spraying curcumin solution on inoculated surfaces of agar plates followed by irradiation and soaking spores/cells in curcumin solution prior to irradiation was also investigated. In aqueous mixtures, photosensitised spores/cells of F. oxysporum and C. albicans were inhibited at all light doses and curcumin concentrations, while inactivation of A. niger, A. flavus P. griseofulvum, P. chrysogenum and Z. bailii were highly significant (P<0.001) reduced by 99%, 88.9%, 78%, 99.7% and 99.2% respectively. On the surface of agar plates, spores/cells exposed to a light dose of 360J/cm 2 sprayed with curcumin at 800μM showed complete inhibition for A. niger, F. oxysporum, C. albicans and Z. bailii, while A. flavus P. griseofulvum, and P. chrysogenum reduced by 75%, 80.4% and 88.5% respectively. Soaking spores/cells with curcumin solution prior to irradiation did not have a significant effect on the percentage reduction. These observations suggest that a novel photosensitization mediated curcumin treatment is effective against fungal spores/cells and the variation of percentage reduction was dependent on curcumin concentration, light dosage and fungal species., (Copyright © 2017 Elsevier B.V. Fungal pathogens represent a significant threat to immunocompromised patients or individuals with traumatic injury. Strategies to efficiently remove fungal spores from hospital surfaces and, ideally, patient skin thus offer the prospect of dramatically reducing infections in at-risk patients. Photodynamic inactivation of microbial cells using light holds considerable potential as a non-invasive, minimally destructive disinfection strategy. Recent data indicate that high-intensity blue light effectively removes bacteria from surfaces, but its efficacy against fungi has not been fully tested. Here we test a wide range of fungi that are pathogenic to humans and demonstrate that blue light is effective against some, but not all, fungal species. We additionally note that secondary heating effects are a previously unrecognized confounding factor in establishing the antimicrobial activity of blue light. Thus blue light holds promise for the sterilization of clinical surfaces, but requires further optimization prior to widespread use.

5% Degussa P25 TiO2 was spray-coated onto black-light and white-light fluorescent lamps, using five different binders, namely DURAMAX B-1000, DURAMAX D-3005, silane-69, and two polyethylene glycols with molecular weight 1000 (PEG-1000) and 700 (PEG-700). The coated lamps were tested with Staphylococcus epidermidis, Escherichia coli, spores of Bacillus subtilis and spores of Aspergillus niger. It was found that 0.5% B-1000 and 1% PEG-1000 gave the highest inactivation rates: 93-96% from coated black-light lamps and 85-88% from coated white-light lamps for bacteria. In the case of spores, 70-72% and 55-57% inactivation rates were recorded from coated black-light and coated white-light lamps, respectively. The effects of UVA irradiance and face velocity were also examined. Significant improvement was observed from coated white-light lamps when the UVA irradiance increased. The objective of this study was to determine the effects of light intensity and duration (photoperiod) on the sporulation (discharge of primary conidia) and conidia germination (from non-infective primary conidia to infective capilliconidia) of Neozygites floridana isolates from Tetranychus urticae originating from Norway and Brazil. Two light intensities (40 and 208 μmolm(-2)s(-1)), three photoperiods (24 h of continuous light (24 h D), 12 h of darkness followed by 12 h of light (12 h D: 12 h L) and 24 h of continuous darkness (24 h D)) and two temperatures (18°C and 23°C) were tested. The fungus produced similar amounts of primary conidia and capilliconidia at 12 h D:12 h and 24 h D, indicating that the fungus discharges almost all of its conidia during the first 12 h of darkness. Light had less of an effect on the production of primary conidia than on capilliconidia formation. At 24 h L, capilliconidia formation was significantly lower for all tested light intensities, temperatures and isolates compared to 12 h D:12 h L and 24 h D. At both light intensities, 24 h L resulted in a significantly lower capilliconidia formation for the Norwegian isolate compared to the Brazilian isolate. Our data suggest that, even though 24 h L reduced sporulation, some capilliconidia formation may occur at the low light intensities found on the underside of strawberry leaves during parts of the day as well as the top of a non-shaded strawberry leaf during the dim evening and morning hours in the tropics and during the dim, long summer days in temperate regions., (Copyright © 2013 Elsevier Inc. Unlabelled: Blue light (470 nm) LED antimicrobial properties were studied alone against bacteria and with or without the food grade photosensitizer, erythrosine (ERY) against filamentous fungi. Leuconostoc mesenteroides (LM), Bacillus atrophaeus (BA) or Pseudomonas aeruginosa (PA) aliquots were exposed on nutrient agar plates to Array 1 (AR1, 0·2 mW cm(-2)) or Array 2 (AR2, 80 mW cm(-2)), which emitted impure or pure blue light (0-300 J cm(-2)), respectively. Inoculated control (room light only) plates were incubated (48 h) and colonies enumerated. The antifungal properties of blue light combined with ERY (11·4 and 22·8 μmol l(-1)) on Penicillium digitatum (PD) and Fusarium graminearum (FG) conidia were determined. Conidial controls consisted of: no light, room light-treated conidia and ERY plus room light. Light-treated (ERY + blue light) conidial samples were exposed only to AR2 (0-100 J cm(-2)), aliquots spread on potato dextrose agar plates, incubated (48 h, 30°C) and colonies counted. Blue light alone significantly reduced bacterial and FG viability. Combined with ERY, it significantly reduced PD viability. Blue light is lethal to bacteria and filamentous fungi although effectiveness is dependent on light purity, energy levels and microbial genus., Significance and Impact of the Study: Light from two arrays of different blue LEDs significantly reduced bacterial (Leuconostoc mesenteroides, Bacillus atrophaeus and Pseudomonas aeruginosa) viabilities. Significant in vitro viability loss was observed for the filamentous fungi, Penicillium digitatum and Fusarium graminearum when exposed to pure blue light only plus a photosensitizer. F. graminearum viability was significantly reduced by blue light alone. Results suggest that (i) the amount of significant loss in bacterial viability observed for blue light that is pure or with traces of other wavelengths is genus dependent and (ii) depending on fungal genera, pure blue light is fungicidal with or without a photosensitizer., (© 2012 The Society for Applied Microbiology.)

The accident at the Chernobyl Atomic Energy Station resulted in radiation contamination of large tracts of land and particularly the reactor building itself. Sustained exposure of microfungi to radiation appears to have resulted in formerly unknown adaptive features, such as directed growth of fungi to sources of ionizing radiation. We evaluate here spore germination and subsequent emergent hyphal growth of microfungi in the presence of pure gamma or mixed beta and gamma radiation of fungi isolated from a range of long term background radiation levels. Conidiospore suspensions were exposed to collimated beams of radiation and percent spore germination and length of emergent hyphae were measured. All fungal species isolated from background radiation showed inhibition or no response in germination when irradiated. Isolates from sites with elevated radiation showed a stimulation in spore germination (69% mixed radiation and 46% for gamma irradiation). Most isolates from low background radiation sites showed a significant reduced or no response to exposure to either source of radiation, whereas the stimulatory effect of experimental exposure to radiation appeared to increase in magnitude as prior exposure to radiation increased. We propose that the enhanced spore germination and hyphal growth seen in the exposure trials is induced by prior long term exposure to radiation and these factors could be important in controlling the decomposition of radionuclide-bearing resources in the environment.

This study examined the efficacy of the following treatments to reduce selected fungal spore and mycotoxin levels on materials commonly found in home contents: (1) gamma irradiation at a 10-13 kiloGray exposure, (2) a detergent/bleach wash, and (3) a steam cleaning technique. A minimum of six replicates were performed per treatment. Paper, cloth, wood, and carpet were inoculated with either fungal spores (Stachybotrys chartarum, Aspergillus niger, Penicillium chrysogenum, or Chaetomium globosum) at 240,000 spores/2.54 cm2 of material or with the mycotoxins roridin A, T-2, and verrucarin A at 10 microg per 2.54 cm2 of material. Treatments were evaluated with an agar plating technique for fungal spores and a yeast toxicity culture assay for mycotoxins. Results showed that gamma irradiation inactivated fungal spores, but the treatment was not successful in inactivating mycotoxins. The washing technique completely inactivated or removed spores on all materials except for C. globosum, which was reduced on all items except paper (p < 0.05). Washing inactivated all mycotoxins on paper and cloth but not on carpet or untreated wood (p < 0.001). The steam cleaning treatment did not completely eliminate any fungal spores; however, it reduced P. chrysogenum numbers on all materials, C. globosum was reduced on wood and carpet, and S. chartarum was reduced on wood (p < 0.05). Steam cleaning was unsuccessful in inactivating any of the tested mycotoxins. These results show that the bleach/detergent washing technique was more effective overall in reducing spore and mycotoxin levels than gamma irradiation or steam cleaning. Growth and sporulation of phytopathogenic microscopic fungi were studied under a static magnetic field. The applied flux densities were 0.1, 0.5, and 1 mT. The magnetic field decreased the growth of colonies by 10% using this flux density region. At 0.1 mT flux density, the deviations are significant, P =.001, while in other cases the deviations generally are not significant. At the same time, the number of the developed conidia of Alternaria alternata and Curvularia inaequalis increased by 68-133%, but the number of Fusarium oxysporum conidia decreased by 79-83%. Levels of fungi growing on insulation within air-handling units (AHUs) in an office building and levels of airborne fungi within AHUs were measured before the use of germicidal UV light and again after 4 months of operation. The fungal levels following UV operation were significantly lower than the levels in control AHUs.

Of the six species of microorganisms isolated from the mesosphere, five contained pigments and were more resistant to UV radiation compared with their pigment-free mutants. The black pigment isolated from the conidia of Aspergillus niger considerably increased the UV resistance of the unpigmented mutant conidia of Penicillium notatum, the spore Circinella muscae and the vegetative cells of Micrococcus albus. From the data it is possible to conclude that in the upper layers of the Earth's atmosphere the predominant proportion of pigmented microorganisms is the consequence of natural selection by UV radiation.

The present review describes the effects of light on reproductive processes in fungi, mainly when action spectra are available. The study of these has resulted in three kinds of photoresponses observable in fungi: - responses only to UV light (230-380 nm) - responses to NUV and blue light (300-520nm) - responses to great wavelengths of the visible spectrum. The photomorphogenetic processes on the control of these same photoreceptor pigments are reviewed.

Dormant sporangiospores of Phycomyces blakesleeanus were activated by Cs-137 gamma rays. After a dose of about 300 krad, between 80 and 90% of the spores germinated normally. However, further development of the mycelium was inhibited. Even with dry spores a partial activation was obtained. Activation by gamma rays was not accompanied by an increase in trehalase activity.

Lower doses of gamma and X-rays (5 and 1 Krad, respectively) stimulated the spore germination and growth of 7 tested fungi. Higher doses (500 and 100 Krad, respectively) revealed that Epicoccum nigrum was the most radioresistant, while Alternaria humicola, Stemphylium verruculosum, Rhizopus nigricans, and Cladosporium herbarum were of moderate resistance. Aspergillus niger and Penicillium oxalicum were the most radiosensitive. Protein and polysaccharide synthesis were inhibited in the mycelium developed from irradiated spores; the inhibition was a function of dose. Study of the different factors (morphological, biological, and biochemical) indicates that variability in fungal resistance to ionizing radiation is, most probably, an inherent character, connected with mycelial water content and the natural production of a chemical substance(s) that acts as a radioprotector(s).
