This study evaluates the performance of different regression algorithms based on artificial intelligence in the quantification of ellagic and gallic acids from ultraviolet (UV) data. Samples of Libidibia ferrea, which is known for its medicinal properties and mostly consumed as tea, were used. The medicinal properties are related to gallic and ellagic acids present in the species. Thus, quantifying gallic and ellagic acids is important to differentiate the biological activities. This quantification is generally performed by high-performance liquid chromatography (HPLC). However, HPLC is expensive and uses polluting solvents. A potential alternative to HPLC is spectrophotometry, which is less expensive and easier to implement. However, the spectrophotometric signal has many overlapping components. This characteristic makes it difficult to quantify substances by spectrophotometry. Computational tools and chemometric methods are used to overcome this limitation. To perform the analysis, eight regression models were used: linear regression, support vector machine, random forest, random tree, and four configurations of extreme learning machines, with kernel: linear, third-degree polynomial, sigmoid, and sine. The Pearson correlation coefficient ( r) was the main metric used to evaluate the performances of the algorithms. The regression algorithms were shown to deliver effective concentration predictions even from raw spectrophotometric measurements, providing results closer to the values using HPLC, with a maximum r value of 0.93 for ellagic acid and 0.83 for gallic acid. Bioactive compound characterization is an essential step for utilizing pomegranate peel waste as food and nutraceuticals ingredients. In the present investigation, the effects of different drying methods (freeze, tray-oven, and sun) and extraction solvents such as methanol, ethanol, water, acetone, and hexane were investigated on the extraction and recovery of major bioactive compounds (ellagic acid, gallic acid, quercetin, and punicalagin) of pomegranate peel for two pomegranate varieties (i.e., Bhagwa and Ganesh) using high-performance liquid chromatography (HPLC). The results indicated that the freeze dried pomegranate peel powder of both pomegranate varities potential to extraction higher amount of bioactive compounds with methanol as extraction solvent as compared to other drying methods and solvents. Freeze-dried peel powder of Bhagwa pomegranate showed a higher amount of gallic acid (32.2 mg/g), ellagic acid (13.6 mg/g), punicalagin (15.2 mg/g), and quercetin (2.5 mg/g) with methanol solvent as compared to the other extract of Bhagwa and Ganesh varieties. The basis on the results of the current study, it can be concluded that the freeze-drying method of drying pomegranate peel powder and methanol as an extraction solvent are effective to recover higher amounts of bioactive compounds that can be utilized in food and pharmaceutical sectors at commercial scale. Pomegranate flowers (PFs) were reported to possess various biological activities such as antidiabetic, anti-inflammatory and hepatoprotective activities, and using to treat diabetes. Although chemical constituents and pharmacological activities of PFs have been studied, unfortunately, there was no report on the pharmacokinetic profile of PFs in vivo. In this study, a selective high-performance liquid chromatography triple quadrupole tandem mass spectrometry (HPLC–QQQ–MS/MS) method was developed and validated for simultaneous quantification of four compounds (corilagin, ellagic acid, gallic acid and brevifolincarboxylic acid) in rat plasma after oral administration of PFs. The good linearity concentration ranges for the four analytes were from 2.5 to 3000 ng/mL with coefficient value R 2 > 0.99 in calibration curves. The intra- and inter-day accuracy of the four analytes was in the range of 85.33–102.50%, with relative standard deviation (RSD) of <14.81%. The stability results showed that accuracy of the four analytes was in the range of 81.88–104.74%, with RSD of <14.86%. The validation method was successfully applied to pharmacokinetic profiles of the four analytes in rats after oral administration of PFs extract. Polyphenols, the secondary metabolites distributed in different parts of a plant, have major role in protecting the plants from deleterious effects of ultraviolet radiations and various diseases caused by pathogens. Considering the fact that these metabolites possess tremendous medicinal properties, extensive research has been carried out during the past few decades to explore their potential health benefits. Further, polyphenols are documented to possess different activities such as antioxidant, anticarcinogenic, anti-inflammatory, antimicrobial and antiulcer. The present study pertained to analyze different polyphenolic compounds in pollen grains of 9 medicinally important plant species viz., Bauhinia variegata, B.purpurea, Cassia biflora, C. fistula, C. glauca, C. saimea and Delonix regia belonging to Fabaceae family, Hibiscus rosa-sinensis belonging to Malvaceae family and Melia azadirach belonging toMeliaceaefamily using Ultra high-performance liquid chromatography (UHPLC). Various polyphenolic compounds likecaffeic acid, catechin, chlorogenic acid, coumaric acid, ellagic acid, epicatechin, gallic acid, kaempferol, quercetin, rutin, tert-butyl hydroquinone andumbelliferonewere found to be present in the pollen grains of these plant species. The order of plant species in terms of maximum number of polyphenolic compounds was observed as Cassia saimea (10) > B. purpurea (9) = C. fistula (9) = Hibiscus rosa-sinensis (9) > Delonix regia (8) > B. variegata (6) > C. glauca (4) = Melia Azadirach (4) > C. biflora (3). Electrochemistry of gallic acid (GA) and ellagic acid (EA) was investigated by cyclic voltammetry (CV) using a bare boron‐doped diamond (BDD) electrode. CVs indicate that the electro‐oxidation of both GA and EA are quasi‐reversible processes. High‐performance liquid chromatography (HPLC) coupled with a BDD electrode poised at+1.4 V offers the limit of detection (LOD, S/N=3) of 60 and 200 nM for GA and EA, respectively. The optimized method was then applied to the detection of both acids in Islay, Highland and Scotch whiskeys, with the highest concentrations found in a 14‐year‐old Highland whiskey. This work evaluated the antioxidant properties and in vivo antinociceptive and anti-inflammatory effects of extracts obtained from fruit peels of Myrciaria floribunda (H. West ex Willd.) O. Berg (Myrtaceae). This plant is popularly known in Brazil as Cambuí or camboim. Different extracts were submitted to comparative analysis to determine the content of selected phytochemical classes (levels of total phenols, flavonoids, and monomeric anthocyanins) and the in vitro antioxidant potentials. The extract with higher potential was selected for in vivo evaluation of its antinociceptive and anti-inflammatory action. Finally, the chemical characterization of this extract was performed by high-performance liquid chromatography (HPLC). MfAE (extract obtained using acetone as solvent) showed the higher levels of phenols (296 mg GAE/g) and anthocyanins contents (35.65 mg Cy-3-glcE/g) that were associated with higher antioxidant activity. MfAE also exhibited in vivo anti-inflammatory and analgesic propertiers. This fraction inhibited the inflammatory and neurogenic phases of pain, and this effect was reversed by naloxone (suggesting the involvement of opioidergic system). MfAE reduced the abdominal contortions induced by acetic acid. The HPLC analysis revealed the presence of gallic acid (and its derivatives) and ellagic acid. Poincianella pluviosa has already been described as capable of healing skin wounds. In an attempt to prolong contact of the drug with the wound, it was proposed in this study to evaluate wound healing using a crude extract (CE) of P. pluviosa incorporated in carboxymethylcellulose polymer films. The chromatographic profile of the semipurified fraction of P. pluviosa was evaluated by ultra-high performance liquid chromatography (UHPLC), confirming the compounds gallic acid, geraniin, and ellagic acid. The films were evaluated for their physical and mechanical properties, water vapor permeability, moisture absorption capacity, and FTIR spectroscopy. For in vivo experiments, wounds were made on the back of rats and treated daily for 4, 7, 10, or 14 days with film containing CE or control film. At the end of each period, skin permeation analysis and histological analysis were made using re-epithelialisation, cell proliferation, and collagen formation. Statistical significance was determined by GraphPad Prism using t test and Mann-Whitney test. Anti-staphylococcal activity was evaluated with standard strains of Staphylococcus aureus, methicillin-resistant, and coagulase negative. It was demonstrated that the presence of CE in the films increased the capacity to absorb water and decreased resistance and permeability. The CE of the film permeated the skin, reaching the dermis and was able to influence re-epithelisation, cell proliferation, and collagen formation. Satisfactory results were observed against S. aureus strains, particularly coagulase negative. Films with CE of P. pluviosa can be an alternative in the wound healing, protecting against opportunistic infections and giving comfort to the patient. [ABSTRACT FROM AUTHOR]

Objective: Pomegranate is scientifically known as Punica granatum L. which is a nutrient dense fruit rich in phytochemical compounds. Phenolic content is the main compound attribute for the most of the functional properties in pomegranate. The aim of this study is to quantify and optimize the composition of phenolic acids extracted from pomegranate extract by using High-performance Liquid Chromatography (HPLC). Method: The pomegranate extracted with three different methods by using two different solvents which is 50% ethanol and water. The methods were blended (aril+ seed), Soaking (aril+ seed) and soaking + squeezed manually. HPLC-PDA was used as equipment to quantify and optimize the phenolic acids extracted from pomegranate. Result: Validation method of HPLC was analysed according to the percentage of recovery, LOD, LOQ and coefficient correlation. Result showed that GA was detected in all sample from different method of extraction applied while CA compound not detected in any of extraction method applied. FA compound was only detected in blended method by 50% ethanol and water as solvent while EA compound was detected only in water extraction of all three methods applied. Conclusion: As a conclusion, according to the standard calibration data curve showed that this method proved to detect and quantify the targeted compounds. By comparing the data obtained from this study, it showed that water blended extract method is significantly higher content of targeted compound except for the CA compound. Ingestion of antioxidants from the diet is associated with improved health. Total antioxidant capacity as gallic acid equivalents (GAE) of distilled spirits was determined using a modified and validated silver nanoparticle (SNaP) assay utilizing silver nanoparticles pre‐capped with starch and microwave technology. GAE concentrations determined by the SNaP assay for the distilled spirits were validated by high‐performance liquid chromatography (HPLC) measurements of key antioxidant components including gallic acid, ellagic acid, caffeic acid, syringic acid, syringaldehyde and 2‐furaldehyde. SNaP assay and HPLC measurements of GAE were compared via statistical and graphical methods to demonstrate comparability. Application of the assay to 33 different distilled spirits demonstrated good agreement with published literature values, with the highest total antioxidant capacity values observed for bourbon samples followed by whisky, tequila and other types of liqueur. Context: Natural products play a vital role in the discovery of leads for novel pharmacologically active drugs. Geraniin (GE) was identified as the major compound in the rind ofNephelium lappaceumL.(Sapindaceae), while ellagic and gallic acids have been shown to be its main metabolites. GE and its metabolites possess a range of bioactive properties including being an anti-infective, anticarcinogenic, antihyperglycemic, and antihypertensive. Objective: GE and its metabolites were investigated to establish its gastrointestinal absorption and physicochemical properties. Materials and methods: GE was purified fromN. lappaceumrind extract using reverse-phase C18 column chromatography. Lipophilicity (logP) was determined using the 1-octanol/water shake-flask method. Equilibrium solubility of GE and its metabolites (20 mg) was determined in water and four biorelevant media: simulated gastric, simulated intestinal, fasted state-simulated intestinal, and fed state-simulated intestinal after 72 h. Results and discussion: The purification yield was 10.8%; where a 97–99% pure GE was obtained. LogPvalues for GE, ellagic, and gallic acids were established as −0.73 ± 0.17, 0.11 ± 0.06, and 0.71 ± 0.21, respectively, establishing them as polar compounds. All three compounds were found to exhibit poor solubility in gastric (0.61–8.10 mg/mL) but good solubility in intestinal fluids (3.59–14.32 mg/mL). Conclusion: The above results indicate that the compounds have limited gastrointestinal absorption due to its polarities. Compared with abundant catechins, strictinin is a minor constituent in teas and has been demonstrated to possess inhibitory potency on influenza virus. In this study, strictinin was found as the major phenolic compound in Pu'er teas produced from leaves and buds of wild trees. Due to its thermal instability, strictinin, in tea infusion or in an isolated form, was completely decomposed to ellagic acid and gallic acid after being autoclaved for 7 minutes. A plaque reduction assay was employed to compare the relative inhibitory potency between strictinin and its thermally degraded products against human influenza virus A/ Puerto Rico/8/34. The results showed that the antiviral activity of ellagic acid regardless of the presence or absence of gallic acid was significantly higher than that of strictinin. Thermal degradation of strictinin to ellagic acid and gallic acid seems to be beneficial for the preparation of Pu'er teas in terms of enhancing antiviral activity. [ABSTRACT FROM AUTHOR]

A novel method of extraction and separation of gallic acid and ellagic acid from Euonymus alatus was developed by microwave-assisted extraction (MAE) coupled with aqueous surfactant two-phase extraction (ASTPE). Using PEG-400/(NH 4) 2 SO 4 system as the extractant, the effects of key factors on the extraction yields of the phenolic acids were investigated by response surface methodology. The optimized extraction conditions were as follows: (NH 4) 2 SO 4 concentration 17.74% (w/w), PEG-400 concentration 25.94% (w/w), extraction temperature 95.40 °C and liquid-to-material ratio of 59.99:1. The results of validation experiments were close to the predictedvalues of the model. The extraction yields of the phenolic acids for MAE-ASTPE were superior to that of conventional methods. The proposed approach coupled to high-performance liquid chromatography was successfully applied for extraction and determination of the phenolic acids in three batches of Euonymus alatus. The extraction yields of gallic acid and ellagic acid were 0.4532 ± 0.0031, 0.5864 ± 0.0061, 0.4966 ± 0.0056 mg/g and 0.1713 ± 0.0043, 0.3072 ± 0.0030, 0.1374 ± 0.0015 mg/g, respectively. The extract obtained by MAE-ASTPE had stronger antioxidant activity, and EC 50 values of scavenging DPPH (2,2-diphenyl-1-picrylhydrazyl), hydroxyl, superoxide and ABTS (2,2′-azinobis-3-ethylbenzothiazoline-6-sulfonic acid) radicals were respectively 198.5 ± 1.2, 358.0 ± 2.8, 776.3 ± 3.3 and 333.6 ± 1.8 μg/mL. Gallic acid and ellagic acid have been identified in the seed ofCornus officinalisby the use of an ultrahigh performance liquid chromatography (UHPLC) method coupled with a diode array detector (DAD). The water extract ofC. officinalisseed contained the highest gallic acid content (3.03 ± 0.10 mg/g seed), which was followed by aqueous methanol extract (2.43 ± 0.10 mg/g seed) and aqueous ethanol extract (1.53 ± 0.25 mg/g seed). But a higher concentration (12.32 ± 0.45 mg/g seed) of ellagic acid was obtained from extraction with aqueous methanol than with aqueous ethanol (11.03 ± 0.42 mg/g seed) and water (7.28 ± 0.16 mg/g seed). After heat treatment and acid hydrolysis,C. officinalisseed had higher concentrations of gallic acid and ellagic acid, contributing to more potent antioxidant activity. A simple, rapid, and economic simultaneous HPLC method was developed and validated for the quantification of Gallic acid (GA), Ellagic acid (EA), and Ascorbic acid (AA), in Emblica officinalis Linn. (aamla) and in two poly herbal Unani formulations, containing aamla as an ingredient. Separation was achieved on a reverse phase C18 (250 × 4.6 mm, 5 µm) column with a mobile phase of 0.1% orthophosphoric acid and acetonitrile in gradient elution. The flow rate was kept as 1 mL min−1 and detection was carried out at 254 nm. The proposed method was validated as per the ICH guidelines for accuracy, precision, robustness, LOD, and LOQ. The proposed method was linear over a wide range of concentration with a good regression coefficient of more than 0.99. The LOD of method was 0.035 µg mL−1, 0.31 µg mL−1, and 0.38 µg mL−1, whereas LOQ was 0.1 µg mL−1, 1.0 µg mL−1 and 1.3 µg mL−1 for ascorbic acid, ellagic acid, and gallic acid, respectively. The results demonstrated that the proposed method is a simple, reproducible, accurate, economic, and suitable method for the quality control of aamla, its formulations, as well as a large population of plants with anti-oxidant property. Abstract: The anti-oxidant content and potential health benefits associated with consuming pomegranate and pomegranate-containing products has lead to increased consumer demand for this crop resulting in it becoming a high value crop. The potential health benefits and high anti-oxidant content of this fruit is attributed to the polyphenolic compounds it contains, including the ubiquitous phenolic acids, gallic acid and ellagic acid, and punicalagin A and punicalagin B, two polyphenolics unique to this fruit. A rapid HPLC–UV method targeting these four metabolites requiring minimal sample cleanup and offering runtimes half as long as existing methods was established. Within day and inter-day run-to-run variability for the four metabolites ranged from 1.9% to 6.6% and 5.3% to 11.4%, respectively. Spike recovery percentages for gallic acid, punicalagin A, punicalagin B and ellagic acid were found to be 98.5%, 92.4%, 95.5%, and 96.5%, respectively. This method was applied to the evaluation of various pomegranate products, including commercial drinks, handmade juice, and marc extracts. Plinia cauliflora (Mart.) Kausel, widespread in South America, has edible fruits, and its bark is commonly used against diarrhea and other disorders, on account of its astringency. Because diarrhea is still one of the most important causes of illness and death among children in developing countries, where the population turns to traditional medicine for its treatment, the present study determined the composition of fruit and leaf extracts of P. cauliflora, analyzed the activity against diarrhea by antimicrobial and gastrointestinal motility, and evaluated the cytotoxicity of the extracts. Chemical composition was determined by high-performance liquid chromatograpy-ultraviolet/photodiode array detection. Antimicrobial activity was analyzed by agar diffusion and the microdilution method against etiological agents of diarrhea. The effect on gastrointestinal motility was analyzed using an experimental model in mice. Cytotoxicity was evaluated in vitro with the fibroblast cell line SIRC CCL 60, and leaf extract showed a 50% inhibitory concentration of 0.48 μg/mL. Gallic acid, ellagic acid, and flavonoid derivatives were detected in the extracts. It was observed that fruit and leaf extracts showed some activity against Enterococcus faecalis, Escherichia coli, Salmonella sp., and Shigella sp. Abstract: Gallic acid (GA) and ellagic acid (EA) have been identified in longan seed and mango kernel by the use of reversed-phase high-performance liquid chromatography (RP-HPLC) coupled with photodiode array detection (DAD). The ethanolic extract of longan seed contained 23.3 and 156mg/100 seeds of GA and EA, respectively. The ethanolic extracts of mango kernel contained approximately 87% more GA than the longan seed ethanolic extracts but about 32% less EA. After heat treatment and acid hydrolysis, mango kernel had higher concentrations of GA and EA, contributing to more potent antioxidant activity. • Compounds of high biological value were extracted of pequi peel flours. • Non-extractable phenolics were determined for the first time in this food material. • Higher yields of phenolics were detected in the non-extractable fractions. • Lutein and β-carotene were the carotenoids identified with the highest amounts. • The presence of saponins and hydrolyzable tannins were observed for pequi peels. The bioactive compounds of pequi peel flours were characterized. Flavonoid contents ranged from 19.67 to 87.61 mg/100 g, high in comparison to many vegetables described in the literature. Gallic acid (11.52–418.67 mg/100 g), gallate ethyl (2026.75 – 5205.90 mg/100 g), ellagic acid (509.47 – 1630.66 mg/100 g), lutein (0.17–1.36 mg/100 g), β-carotene (0.82 – 1.49 mg/100 g), and β-cryptoxanthin (0.07 – 0.11 mg/100 g) were identified by HPLC, contributing to a greater valorization of the pequi peel flours. Phytochemical tests indicated the presence of hydrolyzable tannins and saponins. The detection of these compounds makes the product commercially attractive, in addition to generating value for an agro-industrial residue.
