Phenotypic expression of the murine intraspecies and interspecies antigenic determinants of the major type C viral structural 30,000-dalton polypeptide, p30, was measured by radioimmunoassay inhibition in cell lines from different species. Uninfected normal rat kidney (NRK) cells did not contain detectable levels of murine intraspecies and interspecies p30 antigen, whereas rat cells transformed by and producing murine sarcoma virus (MSV)-Moloney leukemia virus (M-MSV-MuLV) contained high levels of both murine intraspecies and interspecies p30 antigen. Significant amounts of murine intraspecies and interspecies p30 antigen were detected in wild-type MSV-transformed nonproducer NRK cells. The control of p30 antigen expression was examined in temperature-sensitive MSV-transformed nonproducer cells [NRK(MSV-1b)] which are cold sensitive for maintenance of the transformed phenotype. Both murine intraspecies and interspecies p30 antigens were detected in NRK(MSV-1b) cells when grown at the permissive (39 C) or nonpermissive (33 C) temperature, suggesting that p30 antigen expression is not correlated with maintenance of the transformed phenotype. The results demonstrate that previously undetectable p30 antigens are expressed in MSV-transformed nonproducer NRK cells, and suggest that the expression of p30 antigen may be a useful marker for viral gene expression in mammalian cells.

Two murine sarcoma viruses, the Kirsten and the Harvey, were isolated by passage of mouse type C leukemia viruses through rats. These sarcoma viruses have genomes containing portions of their parental type C mouse leukemia virus genomes, in stable association with specific rat cellular sequences that we find to be quite likely not those of a rat type C leukemia virus. To determine if these murine sarcoma viruses provide a model relevant to the events occurring in spontaneous tumors, we have hybridized DNA and RNA prepared from rat tumors and normal rat tissues to [3H]DNA prepared from the Kirsten murine sarcoma virus. We have also hybridized these rat tissue nucleic acids to [3H]DNA prepared from a respresentative endogenous rat type C leukemia virus, the WFU (Wistar-Furth). Sarcoma-viral rat cellular sequences and endogenous rat leukemia viral sequences were detected in the DNA of both tumor and normal tissues, with no evidence of either gene amplification or additional sequences being present in tumor DNA. Sarcoma-viral rat cellular sequences and endogenous rat leukemia viral sequences were detected at elevated concentrations in the RNA of many rat tumors and in specific groups of normal tissues.

Cell lines of four mammalian species were each examined for the number of Moloney murine sarcoma virus (M-MSV) DNA copies in total cellular DNA after M-MSV transformation. Sarcoma-positive, leukemia-negative (S+L-) M-MSV-transformed cells were compared to M-MSV-transformed cells infected with a replicating leukemia virus. Both unfractionated M-MSV complementary DNA and complementary DNA representing the MSV-specific and the MSV-murine leukemia virus-common regions of the M-MSV genome were hybridized to total cellular DNA of various species. DNAs of mouse, cat, dog, and human S+L-cells contained from less than one to a few proviral M-MSV DNA copies per haploid genome. In contrast, helper virus-coinfected, M-MSV-producing cells of each species showed a 3- to 10-fold increase in M-MSV proviral DNA over that found in corresponding S+L- cells. MSV-specific and MSV-murine leukemia virus-common nucleotide sequences were each increased to a similar degree. A corresponding examination of cellular DNA of leukemia virus-infected normal or S+L- mammalian cells was performed to establish the resulting number of leukemia proviral DNA copies. The infection of normal or S+L- mammalian cells with several leukemia-type viruses that did not have nucleotide sequences closely related to the cell before infection resulted in the appearance of one to three corresponding leukemia proviral DNA copies.

Mouse cells transformed by murine sarcoma virus were made resistant to 8-azaguanine. Resistant cells and cell clones isolated from them were deficient in hypoxanthine-guanine phosphoribosyl transferase (HGPRT) activity. They did not grow in HATG medium, did not incorporate labeled hypoxanthine, and had negligible HGPRT activity. The resistance was genetically stable. The resistant cells were hyperdiploid and contained telocentric chromosomes only. The resistant cells as well as the progenitor cells were slightly tumorigenic in mice, the plating efficiency in soft agar was very low. The parental cells and aza-G resistant cells produced C-type viral particles having RNA-dependent DNA polymerase activity. The resistance to aza-G did not influenced the expression of murine sarcoma virus genome in cells. The resistant cells are suitable for preparation of cell hybrids.

Polyions were tested for effects on some membrane-related functions. Both polycations investigated reduced the negative surface charge of assay cells and enhanced in vitro infectivity of murine C-type viruses, but had no influence on leukemia-virus-induced XC cell syncytia formation. Three polyanions increased the net outer cell charge, while only one of four inhibited infectivity and two of three impeded syncytia formation. Polyions had a slight, probably toxic, effect on the transmembrane potential, independent of their charge. Cells treated with fluorescent DEAE-dextran showed diffuse staining, which 4 h later had been modified into a granular fluorescence with unstained areas now present. This change correlated with a loss of enhancement of viral infectivity. The only polyanion which inhibited viral infectivity had a strong antihyaluronidase activity, and hyaluronidase and Ca++ both increased viral infectivity. It is suggested, therefore, that polyions may in part work on virus-cell membrane interactions by influencing membrane enzymes and not necessarily by simply changing the net outer cell surface charge.

Mouse neuroblastoma cells containing intracisternal type A particles were treated with iododeoxyuridine and dexamethasone to induce the release of type C oncornavirus particles. For 5 days after treatment, antigenic markers and DNA polymerase activities specific to particles of each of the two types were assayed in the cells and in pellets obtained by high-speed centrifugation of the culture fluid. There was a marked release of C-particle antigen (p30) and DNA polymerase activity in extracellular particulate form, reaching a maximum on day 3 after treatment and falling thereafter. In contrast, no extracellular A-particle antigen was detected, and A-particle-specific DNA polymerase activity in the medium pellets did not increase from the original very low level. Electron microscopy confirmed the presence of free type C virus particles, but not intracisternal type A particles, in the culture fluid. Although intracellular levels of C-particle antigen rose 20- to 30-fold per milligram of cell protein, intracellular A-particle antigen and DNA polymerase activity did not vary more than two-fold. The relative rate of A-particle synthesis in the treated cells, as judged by incorporation of radioactive amino acids into the major structural protein (P73), was also unchanged over the period of observation. Thus, the induction of type C virus particle formation in cultured neuroblastoma cells had no detectable effect on the quantity, synthesis rate, or location of intracisternal type A particles.

Guinea pig embryo (GEP) cells were transformed in vitro by the Kirsten strain of mouse sarcoma virus (Ki-MSV). The transformed cells were found to release infectious virus continuously and produced high titers of group-specific (gs) complement-fixing (CF) antigen characteristics of the murine sarcoma-leukemia virus complex. Foci of transformed cells were similar in appearance to those obtained with Ki-MSV in mouse and rat cells. The transformed cells produced RNA dependent DNA polymerase and type C virus particles with a density of approximately 1.15 g/ml in sucrose gradients by 3H-uridine labeling. The transformed cells produced tumors when transplanted into newborn guinea pigs. A number of focus-derived clonal lines from Ki-MSV transformed cells were isolated and characterized. All the focus-derived lines were found to be either producers or nonproducers (NP). The NP guinea pig cells produced neither infectious virus nor viral antigens of the murine sarcoma-leukemia virus complex although they were morphologically indistinguishable from virus-releasing MSV transformed GPE lines and produced tumors when transplanted into newborn guinea pigs. However, the sarcoma virus genome could be rescued in these NP cells by cocultivation with "helper" murine leukemia virus (MuLV) releasing GPE cells. Particles resembling guinea pig leukemia virus were activated from guinea pig NP cells or cultured normal guinea pig cells following chemical treatment. These particles were approximately 100 nm in the mature form and had a density of 1.16-1.17 g/ml. They contained RNA dependent DNA polymerase activity.

In short-term cultures of BALB/c spleen cells, treatment with a combination of 5-bromo-2'-deoxyuridine (BrdU) and either lipopolysaccharide W. Escherichia coli or concanavalin A resulted in release of C-type virus into the medium. Only lipopolysaccharide induced virus release when given alone. This could be potentiated by a combined treatment with BrdU. In contrast, phytohemagglutinin at mitogenic concentration had no effect with or without BrdU, suggesting that inducibility may vary between various mitogen-responsive spleen cell populations. In AKR mice, spontaneous virus release was detectable in nonstimulated spleen cell cultures. This could be potentiated by lipopolysaccharide, whereas no further increase occurred upon additional BrdU treatment. The induced viruses had C-type characteristics in that they contained reverse transcriptase that could be distinguished from cellular enzymes by template-primer preference experiments. Furthermore, the enzyme activities were particle-associated, banding in isopycnic sucrose gradients at 1.15-1.17 g/cm-3. The presence of C-type viruses was confirmed by electron microscopy.

We have previously shown that dexamethasone stimulates production of type-C virus from seemingly normal murine fibroblasts (BALB/3T3) and from transformed (Kirsten sarcoma-leukemia virus) nonproducing cells (BALB/K3T3) induced by 5-iododeoxyuridine. In this report, we further examine the mechanism of this effect by using BALB/K3T3 cells. Several observations suggest that this effect is post-transcriptional. The optimal stimulation by dexamethasone is obtained when dexamethasone is given 24 to 48 h after 5-iododeoxyuridine induction. Although this effect is late, time course experiments suggest that dexamethasone does not act to promote release of preformed virions. The stimulation by dexamethasone is blocked when cells are treated with cordycepin (3'-deoxyadenosine) during the first 24 h of induction, but not when cordycepin is added later. Conversely, interferon, which inhibits virus production, interferes with dexamethasone when it is added late or after removal of the steroid. The results of molecular hybridization experiments show that there is no detectable increase in Kirsten sarcoma-leukemia virus-specific RNA in dexamethasone-treated cells (with or without 5-iododeoxyuridine). The results of the time course studies, and the cordycepin, interferon, and hybridization experiments, suggest that the effect of dexamethasone on type-C virus production in this system is post-transcriptional.

Using an MLg cell line originating from a single newborn ddY mouse, N-tropic C-type virus able to infect the cells or origin was induced by 5-iodo-2'-deoxyuridine (IUdR). All the clones isolated from the MLg cell line released the virus after IUdR-treatment. There was more than a 100-fold difference in the virus inducibility between the highly and lowly inducible clones. The [125I]IUdR incorporation was, however, the same for the both types of clones. When the induced virus was titrat-d in the S+L-C182 cells, the XC plaque titre was about tenfold higher than murine sarcoma virus rescue focus titre.

A tumor line (58-2T) was established from a slowly growing tumor in a BALB/c mouse inoculated with M58-2 cells. The latter clonal cell line was isolated after bromodeoxyuridine treatment as a flat variant from nonproducer BALB/3T3 cells transformed by the Kirsten sarcoma virus. The 58-2T cells produced type C virus with two discrete virus-specific RNA species. One of the species, which was probably an endogenous virus RNA subunit, had a sedimentation coefficient of 35S as the largest major subunit, and had sequences similar to Rauscher leukemia virus RNA based on nucleic acid hybridization. The other RNA species had 30S as the largest major subunit and corresponded to Kirsten sarcoma virus-specific RNA. These two RNA species formed heterogeneous, 60 to 70S, high-molecular-weight RNA in virions.DNA transcripts (58-2T DNA) from the activated virus contained base sequences complementary to Rauscher leukemia virus and Kirsten sarcoma virus. The Kirsten sarcoma virus-specific DNA sequences (58-2TS) were purified from 58-2T DNA by eliminating RLV-specific sequences.

A rat tropic rat hepatoma virus pseudotype Kirsten sarcoma virus has been rescued from a co-culture of rat hepatoma tissue culture cells, HTC-HI, with Kirsten murine sarcoma transformed non-producer rat kidney cells (K-NRK). The virus complex, RHHV-KiMSV, released from the co-culture exhibited cell transformation ability on normal rat kidney (NRK) cells and showed a restricted host range limited to rat embryo fibroblasts and other normal rat cell lines. Evidence derived from indirect immunofluorescence assays demonstrated the association of rat helper virus specific gs-I and gs-3 antigens with RHHV-KiMSV transformed cells. RHHVKiMSV is currently being cultured at a titre as high as 10(4) f.f.u./ml in tissue culture, and thus offering opportunity for biochemical studies of rat viruses.

The host range of the C particle produced by FLOPC-1 myeloma cells, FLOPC-1 murine myeloma-associated virus (FL-MuMAV), was assessed in terms of its ability to productively infect and/or induce new viral antigens in a variety of different cell lines. Production of C particle-like structures by cells exposed to FL-MuMAV) was determined by incorporation of [3H]uridine into particles with a density of 1.16 g/ml and/or measurement of RNA-dependent DNA polymerase activity in concentrated culture medium. to FL-MuMAV was capable of infecting NIH/3T3, normal rat kidney (NRK) cell, BALB/c 3T3, and the A31 clone of BALB/3T3 cells but not rabbit cell line, SIRC. Thus, it is an N, B-tropic murine virus as replication in NRK cells has been shown not to delineate a group of murine viruses with a separate host range (M. M. Lieber, C. J. Sherr, and G. J. Todero, 1974). Further neoantigens, reactive with anti-FL-MuMAV serum, were detected on infected cells. Production of the MuMAV-like particle and MuMAV-associated cell antigens in infected NIH/3T3 and NRK cells persisted for three subcultures. The limited production could not be explained by the lack of an RNA-dependent DNA polymerase or high-molecular-weight RNA as the particles possessed both of these properties. The particles produced by infected NIH/3T3 or NRK cells were antigenically and physicochemically similar to FL-MuMAV and not K-MuLV. The MuMAV-like particles produced by infected NIH/3T3 were capable of limited replication in NIH/3T3 and and BALB/3T3 cells, whereas NRK-MuMAV replicated for a limited period in NIH/3T3, NRK, and SIRC cells; i.e., they had a different host range than FL-MuMAV. The particles produced by infected BALB/3T3 and A31 cells had the same host range as FL-MuMAV. In certain situations, isotopically labeled particles with a density of 1.16 g/ml were produced which appeared to lack RNA-dependent DNA polymerase.

A sensitive biological assay has been developed for studying the frequency of virus induction from mouse cells containing poorly infectious endogenous C-type viruses. A low spontaneous frequency of virus activation of one but not a second endogenous virus of BALB/c cells has been demonstrated. The resistance of BALB/c cells to superinfection by the more inducible virus appears to be absolute, whereas resistance to the other virus is only relative. Thus, the evidence indicates that the BALB/c cell exerts differential effects at two levels of regulation on the expression of its biologically distinguishable endogenous C-type viruses.

Murine ribonucleic acid tumor viruses and C-type virus particles are produced in relatively large quantities in roller bottle cultures. The viruses present in large volumes of culture fluids can be purified by a simple two-step procedure involving polyethylene glycol precipitation and equilibrium centrifugation in sucrose density gradients.
