Decrypting the dynamics of receptor dimerization on cell membranes bears great importance in identifying the mechanisms regulating diverse cellular activities. In this regard, long-term monitoring of single-molecule behavior during receptor dimerization allows deepening insight into the dimerization process and tracking of the behavior of individual receptors, yet this remains to be realized. Herein, real-time observation of the receptor tyrosine kinases family (RTKs) at single-molecule level based on plasmon rulers was achieved for the first time, which enabled precise regulation and dynamic monitoring of the dimerization process by DNA programming with excellent photostability. Additionally, those nanoprobes demonstrated substantial application in the regulation of RTKs protein dimerization/phosphorylation and activation of downstream signaling pathways. The proposed nanoprobes hold considerable potential for elucidating the molecular mechanisms of single-receptor dimerization as well as the conformational transitions upon dimerization, providing a new paradigm for the precise manipulation and monitoring of specific single-receptor crosslink events in biological systems.

Energy carrier evolution is crucial for material performance. Ultrafast microscopy has been widely applied to visualize the spatiotemporal evolution of energy carriers. However, direct imaging of small amounts of energy carriers on nanoscale remains difficult due to extremely weak transient signals. Here we present a method for ultrasensitive and high-throughput imaging of energy carrier evolution in space and time. This method combines femtosecond pump-probe techniques with interferometric scattering microscopy (iSCAT), named Femto-iSCAT. The interferometric principle and unique spatially-modulated contrast enhancement increase the transient image contrast by >2 orders of magnitude and enable the exploration of new science. We address three important and challenging problems: transport of different energy carriers at various interfaces, heterogeneous hot electron distribution and relaxation in single plasmonic resonators, and distinct structure-dependent edge state dynamics of carriers and excitons in optoelectronic semiconductors. Femto-iSCAT holds great potential as a universal tool for ultrasensitive imaging of energy carrier evolution in space and time.

The development of uranyl ion detection technology has exhibited its significance in public security and environmental fields for the radioactivity and chemical toxicity of uranyl ion. The WHO standard of uranyl ion makes it necessary to develop highly sensitive uranyl rapid warning system in drinking water monitoring. Herein, a visualized rapid warning system for trace uranyl ion is carried out based on electrochemiluminescence (ECL) imaging technology to give an ultra-low limit of detection (LOD) and high selectivity. Amidoxime, a bi-functional group with both uranyl ion capturing and co-reactive functions, is modified on a conjugated polymer backbone with strong ECL signal to be prepared into three-in-one polymer nanoparticles (PNPs) with self-enhanced ECL property. The captured uranyl ion can enhance the ECL signal of PNPs via resonance energy transfer process to give the LOD as 0.5 ng/L, which is much lower than the known luminescent uranyl sensors. Furthermore, ECL imaging technology is introduced into realizing visualized uranyl rapid warning, and can be successfully applied on natural water samples. This study provides a novel strategy for uranyl rapid warning, and shows its potential meaning in public security and environmental fields.

Understanding the pH evolution during endocytosis is essential for our comprehension of the fundamental processes of biology as well as effective nanotherapeutic design. Herein, we constructed a plasmonic Au@PANI core-shell nanoprobe, which possessed significantly different scattering properties under acidic and basic conditions. Encouragingly, the scattering signal of Au@PANI nanoprobes displayed a positive linear correlation with the pH value not only in PBS but also in nigericin-treated cells. Ultimately, benefiting from the excellent response properties as well as the excellent biocompatibility and stability, the Au@PANI nanoprobes have successfully enabled a dynamic assessment of the evolving pH in the endosomal package as the endosome matured from early to late, and eventually to the lysosome, by reporting scattering signal changes.

By assembling nanotweezers with ATP-splitting aptamers on gold nanorods (AuT123L), we constructed a near-infrared-activated ATP sensing device that could time-controllably image ATP levels in living cells. By replacing the aptamers on the nanotweezers, the nanoplatform can be applied to other important biomolecules, opening up more possibilities for the study of time controllable nanodevices.

Single-cell protein therapeutics is expected to promote our in-depth understanding of how does a specific protein with therapeutic dosing treat the cell without population averaging. However, it has not yet been tackled by current single-cell nanotools. We address this challenge by utilization of a double-barrel nanopipette, in which one lumen was used for electroosmotic cytosolic protein delivery and the other was customized for ionic evaluation of the consequence. Upon injection of protein DJ-1 via delivery lumen, upregulation of the antioxidant protein could protect neural PC-12 cells against oxidative stress from phorbol myristate acetate exposure, which could be transduced by targeting the cytosolic hydrogen peroxide by the detecting lumen. This work has devised a nanotool for single-cell protein therapeutics and provided a perspective for futuristic single-cell therapeutics involving different therapeutic modalities e.g. peptides, enzymes and nucleic acids.

Electrogenerated chemiluminescence microscopy (ECLM) provides a real-time imaging approach to visualize the surface-dependent catalytic activity of nanocatalysts, which helps to rationalize the design of catalysts. In this study, we first propose super-resolution ECLM that could measure the facet- and site-specific activities of a single nanoparticle with nanometer resolution. The stochastic nature of the ECL emission makes the generation of photons obey Poisson statistics, which fits the requirement of super-resolution radial fluctuation (SRRF). By processing an SRRF algorithm, the spatial resolution of ECL images achieved ca. 100 nm, providing more abundant details on electrocatalytic reactivities at the subparticle level. Beyond conventional wide-field ECL imaging, super-resolution ECLM provided the spatial distribution of catalytic activities at a Au nanorod and nanoplate with scales of a few hundred nanometers. It helped uncover the facet- and defect-dependent surface activity, as well as the dynamic fluctuation of reactivity patterns on single nanoparticles. The super-resolution ECLM provides high spatiotemporal resolution, which shows great potential in the field of catalysis, biological imaging, and single-entity analysis.

Cuproptosis is a new form of programmed cell death and exhibits enormous potential in cancer treatment. However, reducing the undesirable Cu ion release in normal tissue and maximizing the Cu-induced therapeutic effect in cancer sites are two main challenges. Herein, we constructed a photothermally triggered nanoplatform (Au@MSN-Cu/PEG/DSF) to realize the on-demand delivery for synergistic therapy. The released DSF chelated with Cu2+ in situ to generate the high cytotoxic bis(diethyldithiocarbamate)-copper (CuET) causing cell apoptosis and formed Cu+ species promoting toxic mitochondrial protein aggregation, leading to cell cuproptosis. Synergistic with photothermal therapy, the Au@MSN-Cu/PEG/DSF could effectively kill the tumor cell and inhibit tumor growth (inhibition rate up to 80.1%). These results provide a promising perspective for potential cancer treatment based on cuproptosis, and also inspire the design of advanced nano-therapeutic platforms.

A novel mass spectrometric method for probing the flash chemistry of electrogenerated reactive intermediates was developed based on rapid collision mixing of electrosprayed microdroplets by using a theta-glass capillary. The two individual microchannels of the theta-glass capillary are asymmetrically or symmetrically fabricated with a carbon bipolar electrode to produce intermediates in situ . Microdroplets containing the newly formed intermediates collide with those of the invoked reactants at sub-10 microsecond level, making it a powerful tool for exploring their ultrafast initial transformations. New tools for single-cell interrogation enable deeper understanding of cellular heterogeneity and associated cellular behaviors and functions. Information of RNA expression in single cell could contribute to our knowledge of the genetic regulatory circuits and molecular mechanism of disease development. Although significant progresses have been made for intracellular RNA analysis, existing methods have a trade-off between operational complexity and practical feasibility. We address this challenge by combining the ionic current rectification property of nanopipette reactor with duplex-specific nuclease-assisted hybridization chain reaction for signal amplification to realize a simple and practical intracellular nanosensor with minimal invasiveness, which enables single-cell collection and electrochemical detection of intracellular RNA with cell-context preservation. Systematic studies on differentiation of oncogenic miR-10b expression levels in non-malignant breast cells, metastatic breast cancer cells as well as non-metastatic breast cancer cells were then realized by this nanotool accompanied by assessment of different drugs effects. This work has unveiled the ability of electrochemistry to probe intracellular RNA and opened new opportunities to study the gene expression and heterogeneous complexity under physiological conditions down to single-cell level.

The worldwide application of nuclear power has created the potential risk of a nuclear accident, which has been a challenge to public security. In nuclear leakages, I2 radioisotopes would cause rapid, global pollution. Therefore, highly sensitive and selective I2 sensors exhibit their significance in nuclear accident early warnings and treatments. Herein, a conjugated polymer was developed for I2 vapor monitoring with an ultra-low limit of detection (LOD). This polymer, modified with a tertiary amine as a co-reactive group, exhibits aggregation-induced electrochemiluminescence (AIECL) and self-enhanced ECL behaviors. It is noteworthy that the tertiary amine also acts as I2 vapor capturing and sensing groups to give a LOD of 0.13 ppt. Excellent selectivity was obtained in various interfering atmospheres. A new mechanism was discovered for designing vapor sensors, which is summarized as co-reactive group poisoning (CGP). To meet the high efficiency requirement of nuclear emergency monitoring, an I2 sensor modified screen printed carbon electrode was used due to its low cost, lack of need for pretreatment and suitability for mass production. A matching upwardly photosensitive ECL dark box was further designed. This study reports ECL vapor monitoring for the first time and provides a novel strategy for early warning of a nuclear emergency, suggesting its significance in environmental and public security fields.

Electrogenerated chemiluminescence, also known as electrochemiluminescence (ECL), is an electrochemically induced production of light by excited luminophores generated during redox reactions. It can be used to sense the charge transfer and related processes at electrodes via a simple visual readout; hence, ECL is an outstanding tool in analytical sensing. The traditional ECL approach measures averaged electrochemical quantities of a large ensemble of individual entities, including molecules, microstructures and ions. However, as a real system is usually heterogeneous, the study of single entities holds great potential in elucidating new truths of nature which are averaged out in ensemble assays or hidden in complex systems. We would like to review the development of ECL intensity and imaging based single entity detection and place emphasis on the assays of small entities including single molecules, micro/nanoparticles and cells. The current challenges for and perspectives on ECL detection of single entities are also discussed., We summarize the history and recent development that has been made in the ECL detection of single entities.

Three new ruthenium(ii) complexes containing an N-heterocyclic carbene (NHC) ligand (RuNHC) have been successfully synthesized and proved to be efficient near-infrared (NIR) ECL (electrogenerated chemiluminescence) luminophores. In addition to the advantages of the lower-charge main motif (+1), the much lower oxidation potentials, and the longer metal to ligand charge transfer (MLCT) absorption bands, most importantly, these RuNHC complexes show higher, or at least comparable, ECL efficiency compared with Ru(bpy)32+ under the same experimental conditions; this demonstrates their great potential for applications in the NIR ECL imaging field in the future.

We report a new mass spectrometric method for detecting electrogenerated intermediates. This approach is based on simultaneous activation of electrospray ionization and redox reaction on a wireless bipolar ultramicroelectrode, which is fabricated in the tip of a quartz nanopipette. The hollow structure of the ultramicroelectrode permits rapid transferring the transient species from electrode-electrolyte interfaces into the gas phase for mass spectrometric identification on the time scale of microseconds. The long-sought fleeting intermediates including TPrA.+ , whose lifetime in solution is only 200 μs, and catecholamine o-semiquinone radicals, the second-order rate constant of which is typically 109 m-1 s-1 , were successfully identified, helping clarify the previously hidden reaction pathways. Accordingly, our method may have wide applicability in exploring the dynamics of many electrochemical reactions, especially their ultrafast initial steps.

Combining nanomaterials and nucleic acid technology, fluorescent nucleic acid-functionalized gold nanoprobes possess many advantages, such as improved stability, good biocompatibility, unique optical properties and precise programmability, which open a new era for live-cell imaging. In particular, amplified nucleic acid-functionalized gold nanoprobes are ideal for in situ detecting powerful but low-abundance biomarkers. In this review, from the perspective of live-cell fluorescence imaging and analysis, the major properties, design principles and applications of nucleic acid-functionalized gold nanoprobes are introduced.

Single-nanoparticle electrochemistry offers electrochemical behaviors of individual entities beyond the ensemble system. An electric double layer (EDL) exists on any charged particle-liquid interface because of counter-ion accumulation, while direct measuring of the interfacial ion migration remains a challenge. Herein, a plasmonic-based transient microscopic method, with a temporal resolution of 1-2 μs, was demonstrated to directly track the ion migration dynamics on single charged nanoparticles. We found that the dynamics of EDL formation might deviate significantly from the prediction made by using the classical resistance-capacitance (RC) model under nanoscale and transient conditions. Under ultrafast charging, due to the limit migration rate of ions in the solution, the actual time scale of the EDL formation could be up to 5 times slower than the predicted value from the RC model. We then proposed a new theoretical model to describe the transient dynamics of EDL formation. These results may expand our current knowledge about nano-electrochemistry and transient electrochemistry.

A label-free and fast approach for positive electrochemiluminescence (ECL) imaging of single cells by bipolar nanoelectrode array is proposed. The reduction of oxygen at a platinized gold nanoelectrode array in a closed bipolar electrochemical system is coupled with an oxidative ECL process at the anodic side. For elevating the ECL imaging contrast of single cells, a driving voltage of -2.0 V is applied to in situ generate oxygen confined beneath cells that is subsequently used for ECL imaging at 1.1 V. High oxygen concentration in the confined space resulting from steric hindrance generates prominent oxygen reduction current at the cathodic side and higher ECL intensity at the anodic side, allowing positive ECL imaging of the cells adhesion region with excellent contrast. Cell morphology and adhesion strength can be successfully imaged with high image acquisition rate. This approach opens a new avenue for label-free imaging of single cells.

With reduced background and high sensitivity, photoelectrochemistry (PEC) may be applied as an intracellular nanotool and open a new technological direction of single-cell study. Nevertheless, the present palette of single-cell tools lacks such a PEC-oriented solution. Here a dual-functional photocathodic single-cell nanotool capable of direct electroosmotic intracellular drug delivery and evaluation of oxidative stress is devised by engineering a target-specific organic molecule/NiO/Ni film at the tip of a nanopipette. Specifically, the organic molecule probe serves simultaneously as the biorecognition element and sensitizer to synergize with p-type NiO. Upon intracellular delivery at picoliter level, the oxidative stress effect will cause structural change of the organic probe, switching its optical absorption and altering the cathodic response. This work has revealed the potential of PEC single-cell nanotool and extended the boundary of current single-cell electroanalysis.

Trace Ir(III) complex enhanced aggregation-induced electrochemiluminescence (AIECL) of poly-tetraphenylethene (pTPE) in aqueous media was investigated for the first time. The poly-TPE end-capped by Ir(III) complex (Ir@pTPE) and its corresponding model polymer poly-TPE (Ph@pTPE) could be synthesized by Suzuki coupling polymerization reaction of 1,2-bis(4-bromophenyl)-1,2-diphenylethene ( M-1 ) with 1,2-diphenyl-1,2-bis(4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)phenyl)ethene( M-2 ) and the continuous Suzuki coupling end-capped reaction of poly-TPE-pinacol boronate with (pq)2Ir(pico)Br and bromobenzene, respectively. Subsequently, the corresponding Ir@pTPE Pdots and Ph@pTPE Pdots encapsulated with poly(styrene- co -maleicanhydride) (PSMA) could be obtained by nanoprecipitation method. Compared with Ph@pTPE Pdots, the Ir@pTPE Pdots with a trace amount of Ir(III) complex (1.34‰ of Ir(III) content, wt) could exhibit 9.9-fold enhancement of the electrochemiluminescence (ECL) signal for visual emission. This work provided a novel strategy on designing highly efficient ECL materials based on trace Ir(III)-end capping AIE-active polymer dots.

A rational combination of different therapeutic modalities within one single nanostructure is promising to enhance the therapeutic response, especially to achieve a synergistic therapeutic efficacy for tumor treatment. Herein, a near-infrared (NIR) photothermally activated nanoagent, which could achieve a spatially controllable codelivery of different curative molecules and a temporally controlled responsive release, was designed to perform effective gene-chemo-photothermal therapy of malignant tumors. The nanoagent consisted of a gold nanorod (AuNR) functionalized with mPEG, DNA, and small interfering RNA (siRNA). With the aid of aptamer AS1411-mediated recognition and endocytosis, the nanoagents were selectively delivered into cancer cells; subsequently, the photothermal conversion of AuNRs happened while under NIR irradiation, which successfully achieved an effective photothermal therapy, induced dehybridization of DNA duplexes, and simultaneously released doxorubicin (DOX) and siRNA. The aggregation-induced electrochemiluminescence (AIECL) of carboranyl carbazoles in aqueous media was investigated for the first time. Quantum yields, morphologies, and particle sizes were observed to determine the electrochemiluminescence (ECL) performance of these aggregated organic dots (ODs). All compounds exhibit much higher ECL stability and intensity than the carborane-free compound, demonstrating the essential role of the carboranyl motif. Moreover, the results of cyclic voltammetry (CV) suggest that oxidation/reduction reactions take place at the carboranyl motif. The excited states of ODs were proposed to be generated by the mechanism of surface state transitions. More importantly, these compounds show a reductive-oxidative mechanism in contrast to other organic materials that show oxidative-reductive mechanisms. Our experiments and data have established the relation between AIE organic structures and ECL properties that has a strong potential for biological and diagnostic applications.

Development of simple and effective synergistic therapy by combination of different therapeutic modalities within one single nanostructure is of great importance for cancer treatment. In this study, by integrating the anticancer drug DOX and plasmonic bimetal heterostructures into zeolitic imidazolate framework-8 (ZIF-8), a stimuli-responsive multifunctional nanoplatform, DOX-Pt-tipped Au@ZIF-8, has been successfully fabricated. Pt nanocrystals with catalase-like activity were selectively grown on the ends of the Au nanorods to form Pt-tipped Au NR heterostructures. Under single 1064 nm laser irradiation, compared with Au NRs and Pt-covered Au NRs, the Pt-tipped Au nanorods exhibit outstanding photothermal and photodynamic properties owing to more efficient plasmon-induced electron–hole separation. The heat generated by laser irradiation can enhance the catalytic activity of Pt and improve the O2 level to relieve tumor hypoxia. Meanwhile, the strong absorption in the NIR-II region and high-Z elements (Au, Pt) of the DOX-Pt-tipped Au@ZIF-8 provide the possibility for photothermal (PT) and computed tomography (CT) imaging. Both in vitro and in vivo experimental results illustrated that the DOX-Pt-tipped Au@ZIF-8 exhibits remarkably synergistic plasmon-enhanced chemo-phototherapy (PTT/PDT) and successfully inhibited tumor growth. Plasmonic photocatalysts have opened up a new direction in utilization of visible light and promoting photocatalytic efficiency. An electrochemical deposition method is reported to synthesise metal@semiconductor (M@SC) core-shell nanocrystals. Due to the strong affinity of Au atoms to S2- and Se2- reduced at negative potential, CdS, CdSe and ZnS were selectively deposited on the surface of the Au core to form a uniform shell with a clear metal/semiconductor interface, which conquered the barrier caused by the large lattice mismatch between the two components. Plasmonic effects increased the photocatalytic performance, as well as provided a chance to in situ monitor the surface nucleation and growth. The structure formation process could be observed under dark-field microscopy (DFM) in real-time and precisely controlled via the scattering color, intensity and wavelength. The proof-of-concept strategy combines the electrochemical deposition and plasmonic imaging, which provides a universal approach in controllable synthesis of core-shell heterostructures, and leads to the improvement of plasmonic photocatalysts.

Uncovering the relationship between the structure, surface properties and electrochemical activity of nanoparticles is of great importance for constructing novel nanocatalysts and highly efficient electrocatalytic devices. Here we report a study of the heterogeneously distributed electrocatalytic activity on individual 2D gold nanoplates. On the basis of electrogenerated chemiluminescence (ECL) microscopy, the size, shape, and site-specific catalytic activity of 2D nanocrystals could be directly imaged at the single particle level with submicron resolution. Since the microelectrode effect with higher fluxes at the perimeter was offset by diffusion of excited species of Ru(bpy)32+, calculated by finite element simulation, the ECL distribution was supposed to be uniform on the micro-sized plates. Therefore, it is highly possible that the observed nonuniform ECL distribution at single nanoplates reflected distinct surface electrocatalytic activities at different sites. Furthermore, ECL microscopy allows continuous in situ imaging, which elucidates the time-varying changes in the spatial distribution of electrocatalytic activity on individual nanoplates, indicating that the corners and edges with more defect sites exhibit higher reactivity, but lower stability than the flat facet. We believe that real-time and high-throughput ECL microscopy may lead to more comprehensive understanding of reactivity patterns of single nanocatalysts.

Herein, we proposed a 30 nm nanopipette functionalized with G-quadruplex DNAzyme as an efficient biomimetic recognizer for ROS generation at subcellular level via the changes of current-voltage relationship. Taking advantages of the ultra-small tip, the nanopipette could penetrate into a single living cell repeatedly or keep measuring for a long time without compromising the cellular functions. Coupled with precision nanopositioning system, generation of ROS in mitochondria in response to cell inflammation was determined with high spatial resolution. Meanwhile, the changes of aerobic metabolism in different cell lines under drug-induced oxidative stress were monitored continuously. We believe that the ICR-nanopipette could be developed as a powerful approach for the study of cellular activities via electrochemical imaging in living cells.

Noble metal nanoparticles are promising catalysts in electrochemical reactions, while understanding the relationship between the structure and reactivity of the particles is important to achieve higher efficiency of electrocatalysis, and promote the development of single-molecule electrochemistry. Electrogenerated chemiluminescence (ECL) was employed to image the catalytic oxidation of luminophore at single Au, Pt, and Au-Pt Janus nanoparticles. Compared to the monometal nanoparticles, the Janus particle structure exhibited enhanced ECL intensity and stability, indicating better catalytic efficiency. On the basis of the experimental results and digital simulation, it was concluded that a concentration difference arose at the asymmetric bimetallic interface according to different heterogeneous electron-transfer rate constants at Au and Pt. The fluid slip around the Janus particle enhanced local redox reactions and protected the particle surface from passivation.

Hierarchical CoS2 microspheres were prepared and modified with a cationic ionic liquid (IL). The composite shows enhanced oxygen evolution reaction (OER) performances attributed to the fact that the OER equilibrium is driven forward by the IL, according to Le Chatelier's principle, and by the electrostatic affinity generated at the CoS2/electrolyte interface.
