Objective: Non-alcoholic fatty liver disease (NAFLD) represents the most common liver disease worldwide. The Latin American population has the highest obesity rates in the world. The aim was to research the factors associated with NAFLD in young adults. Methods: A cross-sectional study was performed on 171 participants with an average age of 21 years who underwent a physical and laboratory examination, anthropometric evaluation, and abdominal ultrasound. Results: Subjects with NAFLD were significantly overweight, with dyslipidemia, and with atherogenic risk. They had the presence of metabolic syndrome compared to those without NAFLD. Bivariate logistic regression showed that body mass index, atherogenic risk, Homeostatic Model Assessment (HOMA) value, and metabolic syndrome were associated with NAFLD development. Conclusions: The variables of HOMA, metabolic syndrome, and atherogenic risk were most associated as risk predictors of this pathology in young adults.

This work presents the effect of atmospheric-pressure plasma (APP) treatment on mesoporous TiO2 (mp-TiO2) layers for antimony sulfide (Sb2S3) solar cells. For this work, it was prepared heterojunctions of antimony sulfide (Sb2S3) thin films solid solution as the absorbing materials and cadmium sulfide (CdS) as a sensitized layer deposited by the successive ionic layer adsorption and reaction (SILAR) technique, the Sb2S3 was obtained by chemical bath deposition (CBD). The photovoltaic characteristics of the solar cells with APP treatment showed a power conversion efficiency (PCE) of 0.54% to 1.04% with APP 0- and 60-seconds treatment, respectively.

Inorganic arsenic (iAs) exposure is a serious health problem that affects more than 140 million individuals worldwide, mainly, through contaminated drinking water. Acute iAs poisoning produces several symptoms such as nausea, vomiting, abdominal pain, and severe diarrhea, whereas prolonged iAs exposure increased the risk of several malignant disorders such as lung, urinary tract, and skin tumors. Another sensitive endpoint less described of chronic iAs exposure are the non-malignant health effects in hepatic, endocrine, renal, neurological, hematological, immune, and cardiovascular systems. The present review outlines epidemiology evidence and possible molecular mechanisms associated with iAs-toxicity in several non-carcinogenic disorders.

Morphine promotes neuroinflammation after NOD-like receptor protein 3 (NLRP3) oligomerization in glial cells, but the capacity of other opioids to induce neuroinflammation and its relationship to the development of analgesic tolerance is unknown. We studied the effects of morphine and fentanyl on NLRP3 inflammasome activation in glial and neuronal cells in the dorsal raphe nucleus (DRN), a region involved in pain regulation. Male Wistar rats received i.p. injections of morphine (10 mg/kg) or fentanyl (0.1 mg/kg) 3 × daily for 7 days and were tested for nociception. Two hours after the last (19th) administration, we analyzed NLRP3 oligomerization, caspase-1 activation and gasdermin D-N (GSDMD-N) expression in microglia (CD11b positive cells), astrocytes (GFAP-positive cells) and neurons (NeuN-positive cells). Tolerance developed to both opioids, but only fentanyl produced hyperalgesia. Morphine and fentanyl activated NLRP3 inflammasome in astrocytes and serotonergic (TPH-2-positive) neurons, but fentanyl effects were more pronounced. Both opioids increased GFAP and CD11b immunoreactivity, caspase-1 and GSDMD activation, indicating pyroptotic cell death. The opioid receptor antagonist (−)-naloxone, but not the TLR4 receptor antagonist (+)-naloxone, prevented microglia activation and NLRP3 oligomerization. Only (+)-naloxone prevented astrocytes’ activation. The anti-inflammatory agent minocycline and the NLRP3 inhibitor MCC950 delayed tolerance to morphine and fentanyl antinociception and prevented fentanyl-induced hyperalgesia. MCC950 also prevented opioid-induced NLRP3 oligomerization. In conclusion, morphine and fentanyl differentially induce cell-specific activation of NLRP3 inflammasome and pyroptosis in the DRN through TLR4 receptors in astrocytes and through opioid receptors in neurons, indicating that neuroinflammation is involved in opioid-induced analgesia and fentanyl-induced hyperalgesia after repeated administrations.

Summary Background Spain is one of the European countries most affected by the COVID-19 pandemic. Serological surveys are a valuable tool to assess the extent of the epidemic, given the existence of asymptomatic cases and little access to diagnostic tests. This nationwide population-based study aims to estimate the seroprevalence of SARS-CoV-2 infection in Spain at national and regional level. Methods 35 883 households were selected from municipal rolls using two-stage random sampling stratified by province and municipality size, with all residents invited to participate. From April 27 to May 11, 2020, 61 075 participants (75·1% of all contacted individuals within selected households) answered a questionnaire on history of symptoms compatible with COVID-19 and risk factors, received a point-of-care antibody test, and, if agreed, donated a blood sample for additional testing with a chemiluminescent microparticle immunoassay. Prevalences of IgG antibodies were adjusted using sampling weights and post-stratification to allow for differences in non-response rates based on age group, sex, and census-tract income. Using results for both tests, we calculated a seroprevalence range maximising either specificity (positive for both tests) or sensitivity (positive for either test). Findings Seroprevalence was 5·0% (95% CI 4·7–5·4) by the point-of-care test and 4·6% (4·3–5·0) by immunoassay, with a specificity–sensitivity range of 3·7% (3·3–4·0; both tests positive) to 6·2% (5·8–6·6; either test positive), with no differences by sex and lower seroprevalence in children younger than 10 years (10%) and lower in coastal areas (

Chronic exposure to inorganic arsenic (iAs) through contaminated drinking water is an important health problem in certain countries. The use of phytochemicals such as curcumin has recently emerged as an alternative strategy for preventing cellular damage caused by iAs. The Epstein-Barr virus (EBV) affects ~90% of the population and experimental evidence suggested that curcumin mediates cytotoxicity against EBV-infected cells. Due to the potential for an interaction of these factors, the aim of the present study was to evaluate the effect of this phytochemical on iAs-related toxicity in EBV-infected cells. Two independent EBV-immortalized human lymphoblastoid cell lines (LCLs) were used as the model. The cell lines were first incubated with increasing concentrations of curcumin or iAs for 24 and 15 h, respectively, to determine the individual effects of each exposure on cell death. In the next experiment, cell cultures were pre-incubated with 5 µM curcumin for 9 h prior to treatment with 10 µM iAs for 15 h, followed by evaluation of cell death and the cell cycle profile via flow cytometry. The results indicated that individual treatment with either curcumin or iAs induced cell death in a concentration-dependent manner. Furthermore, curcumin pre-treatment enhanced iAs-induced cell death and promoted cell cycle arrest in G1 phase. Taken together, these results suggested that curcumin sensitizes EBV-positive LCLs to the cytotoxic effects of iAs.

In regards to breast cancer (BC), survival or disease-free periods are still compromised mainly in Triple Negative (TN) and HER2 tumors. The participation of estrogen receptor (ER) has been reported as crucial in the signaling pathways, including the NOTCH pathway. The study was aimed to evaluate the expression of NOTCH1 and NOTCH3 in BC and its relationship with the presence of ER, as well as with relapses.NOTCH1 and NOTCH3 expression was evaluated in BC using Oncomine database, Breast Cancer Gene Expression Miner database and Kaplan Meier Plotter. Subsequently, detection of NOTCH1 and NOTCH3 in 100 paraffin-embedded BC samples from Mexican patients was achieved by immunohistochemistry (IHC) and RT-qPCR, a group of benign breast tumors were included as controls. Relapses were evaluated by BC subtypes and their relationship with NOTCH1 and NOTCH3 expression, as well as with ER expression.The analyses from public databases of TN and HER2 groups, which are estrogen receptor-negative (ERN), revealed NOTCH1 and NOTCH3 expression variability. The overexpression was associated with lower relapse-free survival (P = 0.00019). These data were concordant with results from tumor samples of patients included in this study, which showed overexpression of NOTCH1 and NOTCH3 in ERN tumors, as well as lower relapse-free survival (P0.0001).NOTCH1 and NOTCH3 were found to be overexpressed mainly in ERN tumors. HER2 and TN groups, are related to higher relapse rates. Therefore, anti-NOTCH therapy could be justified and implemented in conventional treatments of high-risk BC groups.

Las empresas importadoras y exportadoras, en comparación con las que no comercializan al exterior, tienen un riesgo adicional caracterizado por la volatilidad cambiaria. En esta investigación, se analizan las características que distinguen a la administración de las empresas con operaciones internacionales de la ciudad de México y Oaxaca, con altos rendimientos ante el riesgo cambiario 2014-2016 en ambientes contingentes, versus las empresas con bajos rendimientos, donde según la teoría de la contingencia las empresas usan diferentes mecanismos para adaptarse a los cambios del medio ambiente. Se usó una regresión logística (RL) para clasificar a las empresas con rendimientos superiores al 20 %, en función a sus características en la administración. Para caracterizar la administración se utilizaron 23 variables explicativas que abarcaron aspectos del administrador y los esquemas de cobertura empleados. Los resultados obtenidos encontraron que solo cuatro de las 23 variables fueron estadísticamente significativas para que las compañías lograran altos márgenes de rentabilidad, sobresaliendo la utilización de instrumentos financieros derivados como un factor determinante.

Arsenic, a metalloid and naturally occurring element, is one of the most abundant elements in the earth's crust. Water is contaminated by arsenic through natural sources (underground water, minerals and geothermal processes) and anthropogenic sources such as mining, industrial processes, and the production and use of pesticides. Humans are exposed to arsenic mainly by drinking contaminated water, and secondarily through inhalation and skin contact. Arsenic exposure is associated with the development of vascular disease, including stroke, ischemic heart disease and peripheral vascular disease. Also, arsenic increases the risk of tumors of bladder, lungs, kidneys and liver, according to the International Agency for Research on Cancer and the Food and Drug Administration. Once ingested, an estimated 70-90% of inorganic arsenic is absorbed by the gastrointestinal tract and widely distributed through the blood to different organs, primarily to the liver, kidneys, lungs and bladder and secondarily to muscle and nerve tissue. Arsenic accumulates in the organs, especially in the liver. Its excretion mostly takes place through urination. The toxicokinetics of arsenic depends on the duration of exposure, pathway of ingestion, physicochemical characteristics of the compound, and affected biological species. The present review outlines of arsenic toxic effects focusing on different cancer types whit highest prevalence's by exposure to this metalloid and signaling pathways of carcinogenesis.

Inorganic arsenic (iAs) is an important pollutant associated with various chronic-degenerative diseases. The cytoprotective protein nuclear factor erythroid 2-related factor (NRF2) has been proposed as an important responsive mechanism against iAs exposure. The aim of this study was to determine whether the risk of skin lesions in people exposed to iAs-contaminated water could be modified by the presence of single nucleotide polymorphisms in the NRF2 coding gene. We studied 117 individuals with long-term iAs exposure and 120 nonexposed individuals. Total As was determined in water, meanwhile iAs and its metabolites were measured in urine. The iAs-induced skin lesion status was evaluated by expert dermatologists. We sequenced the promoter region of NRF2 in a sample of 120 healthy donors. We found four polymorphisms previously reported and one novel polymorphism in the 5′ regulatory region of the NRF2. In this study, we did not find allelic and genotype association of NRF2 polymorphisms with iAs-related skin lesion. However, the analysis of haplotypes composed by –653GA, and –617CA NRF2 single nucleotide polymorphisms showed a significant association with protection against skin lesions in the low-As exposure group. This is the first report studying the association between NRF2 polymorphisms and susceptibility of As-related skin lesions. Increasing the sample size will allow us to confirm this data.

Chlamydomonas reinhardtii is able to accumulate large amounts of triacylglycerides, the major feedstock for biodiesel production, when grown under stress conditions. In order to characterize gene transcripts induced under nitrogen, iron, and sulfur deprivation in C. reinhardtii; 583 expressed sequence tags (ESTs) were generated through a cDNA library. These sequences were subjected to contig assembly resulting in 30 contigs and 76 singletons. The comparison of the ESTs obtained with public databases allowed to assign putative functions to 66.7 % of the sequences. An important group of the identified genes are related to the lipid metabolic process. A phylogenetic analysis of these sequences identified five isoforms of diacylglycerol O-acyltransferase type 2 (DGAT-2). These genes were selected to measure their relative expression under these stress conditions by means of qRT-PCR. According to the results, the accumulation of DGTT1 mRNA increases considerably under nitrogen and iron inanition when compared to the other isoforms, which indicated that each isoform participates at different levels under each stress condition. These results can help to identify potential genes to be overexpressed by genetic engineering in C. reinhardtii.

Cellular response to arsenic is strongly dependent on p53 functional status. Primarily arresting the cell cycle in G1 or G2/M phases, arsenic treatment also induces an increase in the S-phase time in wild-type p53 cells. In contrast, cells with a non-functional p53 display only a subtle increase in the S phase, indicating arsenic differentially affects the cell cycle depending on p53 status. Importantly, it has been reported that arsenic induces reactive oxygen species (ROS), a process counteracted by p53. To evaluate the participation of p53 in the lengthening of the S phase and the connection between the transient cell cycle arrest and oxidative stress, we evaluated the cell response to arsenic in MCF-7 and H1299 cells, and analyzed p53’s role as a transcription factor in regulating genes involved in ROS reduction and S phase transition. Herein, we discovered that arsenic induced an increase in the population of S phase cells that was dependent on the presence and transcriptional activity of p53. Furthermore, for the first time, we demonstrate that arsenic activates p53-dependent transcription of ROS detoxification genes, such as SESN1, and by an indirect mechanism involving ATF3, genes that could be responsible for the S phase cell cycle arrest, such as CDC25A.

The role of progesterone in women's cancers as well as the knowledge of the progesterone receptor (PR) structure has prompted the design of different therapies. The aim of this review is to describe the basic structure of PR agonists and antagonists as well as the recent treatments for illness associated with the progesterone receptor. The rational design for potent and effective drugs for the treatment of female cancer must consider the structural changes of the androgen and progestogen skeleton which are an indicator of their activity as progestins or antiprogestins. The presence of a hydroxyl group at C-17 in the progesterone skeleton brings about a loss of progestational activity whereas acetylation induces a progestational effect. The incorporation of an ethynyl functional group to the testosterone framework results in a loss of androgenic activity with a concomitant enhancement of the progestational effect. On the other hand, an ester function at C-3 of dehydroepiandrosterone skeleton induces partial antagonism to the PR.

The high demand for less polluting, newer, and cheaper fuel resources has increased the search of the most innovative options for the production of the so-called biofuels. Chlamydomonas reinhardtii is a photosynthetic unicellular algae with multiple biotechnological advantages such as easy handling in the laboratory, a simple scale-up to industrial levels, as well as a feasible genetic modification at nuclear and chloroplast levels. Besides, its fatty acids can be used to produce biofuels. Previous studies in plants have found that the over expression of DOF-type transcription factor genes increases the synthesis and the accumulation of total lipids in seeds. In this context, the over-expression of a DOF-type transcription factor in C. reinhardtii was applied as approach to increase the amount of lipids. The results indicate higher amounts (around 2-fold) of total lipids, which are mainly fatty acids, in the genetically C. reinhardtii modified strains when compared with the non-genetically modified strain. In order to elucidate the possible function of the introduced Dof-type transcription factor, we performed a transcription profile of 8 genes involved in fatty acid biosynthesis and 6 genes involved in glycerolipid biosynthesis, by quantitative real time (qRT-PCR). Differential expression profile was observed, which can explain the increase in lipid accumulation. However, these strains did not show notable changes in the fatty acid profile. This work represents an early effort in generating a strategy to increase fatty acids production in C. reinhardtii and their use in biofuel synthesis.

Arsenic can modify the urinary excretion of porphyrins in animals and humans. Arsenic also interferes with the activities of several enzymes of the heme biosynthesis pathway, such as aminolevulinate synthase (ALA-S), porphobilinogen deaminase (PBGD), uroporphyrinogen III synthase (Uro III S), uroporphyrinogen decarboxylase (URO-D), coproporphyrinogen oxidase (COPRO-O), ferrochelatase and heme oxygenase (H-O). This review deals with HPLC-based techniques for the analysis of porphyrins in human and rodent urine and several heme enzymes with discussion on their usefulness as early biomarkers of arsenic toxicity.

Background and Aims The diagnosis of Clostridium difficile -associated disease (CDAD) is based on the detection of toxins from stool samples. There are several immunoassays for this purpose. The aim of this study was to determine the concordance between the two immunoassays and their performance in comparison to the toxigenic culture as part of the initial evaluation of a suspected case of CDAD. Methods All fecal samples submitted for detection of C. difficile toxins during a 5-month period to our laboratory were analyzed by two immunoassays, VIDAS Toxin CDA/B assay (BioMerieux) and ImmunoCard Toxins A/B (Meridian Bioscience). We cultured on cycloserine-cefoxitin-fructose agar and PCR was used for detection of toxigenic genes. Real-time PCR was performed directly from samples to detect the tcd C gene. Results At the end of the study we processed 230 samples, 13 were positive using VIDAS CDA/B (5.6%), and 14 using ImmunoCard A/B (6.0%); κ coefficient was 0.857. With ImmunoCard A/B we obtained a sensitivity of 80%, a specificity of 99%, positive predicitive value (PPV) 86% and negative predictive value (NPV) 98%, as compared to toxigenic culture. For VIDAS CDA/B we obtained a sensitivity of 90%, a specificity of 98%, PPV 69% and NPV 99%, compared to the same standard. There were seven undetermined results (3.0%) by VIDAS CDA/B. Five of these had a positive culture and all the patients had symptoms of CDAD. Considering these undetermined results as positive, we calculated a sensitivity of 93%, specificity of 97%, PPV 71% and NPV of 99% for this test, and a κ of 0.856. Both immunoassays showed similar results and are suitable for the initial evaluation of patients with suspected CDAD. Conclusions Our data suggest that an undetermined result of VIDAS CDA/B should be considered as positive if CDAD is suspected. Additionally, both immunoassays showed similar results and are suitable for the initial evaluation of patients with suspected CDAD.

We have studied hepatic function in individuals chronically exposed to arsenic (As) via drinking water in Region Lagunera, Mexico. We studied 51 individuals living in three villages exposed to As in water. Nazareno (0.014 mgAs/l), Santa Ana (0.1 mgAs/l) and Benito Juarez (0.3 mgAs/l). We determined the serum activity of aspartate aminotransferase (SAT) and alanine aminotransferase (ALT) as indicators of hepatocellular injury and that of gamma-glutamyl-transpeptidase (GGT) and alkaline phosphatase (ALP) as indicators of cholestasic injury. Serum bilirubin was used as an indicator of organic conjugated anion transport. Total proteins, albumin and globulin fraction in serum were used as indicators of biosynthetic liver capacity. The main findings of this study were the predominantly conjugated hyperbilirubinemia and increased serum ALP activity which were related to the concentration of total arsenic (TAs) in urine, suggesting the presence of cholestasis in As-exposed individuals. No significant changes were observed in the other parameters studied.
