This study aimed to determine the stability of PCs in grape canes extracts stored at different temperatures and light conditions. The PCs composition was monitored every-two weeks during three months by liquid chromatography coupled to diode array and fluorescence detectors (LC-DAD-FLD). Initially, stilbenes represented 87 % of total PCs. Storage at -20 and 5 °C reduced PCs 8 and 6 %, respectively. When extracts were exposed to 25 and 40 °C, the degradation of (+)-catechin and (-)-epicatechin was faster than under lower temperatures, and light accelerated the degradation kinetics. trans-piceatannol showed particularly sensitive to temperature increase, being mostly degraded after two weeks stored at 40 °C. Conversely, degradation of trans-resveratrol and ε-viniferin was mostly catalyzed by light, since nearly 70 % of them were degraded at 40 °C under light, in comparison with a 23 % reduction of trans-resveratrol and no changes of ε-viniferin at 40 °C in darkness.

Irisin is a myokine regulated by peroxisome proliferator-activated receptor gamma co-activator-1α (PGC-1α) in the exercising skeletal muscle and released into the bloodstream after cleavage of FNDC5. Circulating irisin can up-regulate UCP-1 expression in white adipose tissue (WAT) promoting the formation of brown-like adipocytes. The aim of this study was to evaluate if supplementation with a grape pomace extract (GPE) could activate the FNDC5/irisin pathway via PGC-1α in rats fed a high fat diet (HFD). For this purpose we characterized the activation of: i. the FNDC5/irisin pathway and AMPK in skeletal muscle and ii. proteins involved in the formation of brown-like cells in epididymal WAT (eWAT). Consumption of the GPE activated the FNDC5/irisin pathway, increased AMPK phosphorylation in skeletal muscle and enhanced irisin plasma levels. In eWAT, the GPE increased the level of proteins involved in WAT browning, i.e. PGC-1α, PPARγ, PRDM16 and UCP-1. The GPE also prevented HFD-induced adipocyte hypertrophy and systemic insulin resistance. Consistently, in L6 myotubes, (?)-epicatechin (EC), a flavonoid abundant in the GPE, prevented palmitate-mediated downregulation of FNDC5/irisin protein expression and secretion, in part via PGC-1α activation. Consumption of the GPE, a winemaking residue rich in bioactive compounds, could be a beneficial strategy to counteract the adverse effects of Western style diets through the promotion of WAT browning. Grapevine woody by-products contain bioactive substances, mainly phenolic compounds (PCs), whose beneficial health effects initially depends on their levels of intake and bioavailability. Therefore, in-vitro simulated gastrointestinal digestion (GID; oral, gastric and intestinal phases) was performed to evaluate the bioaccessibility and antioxidant capacity (AC) of PCs extracts recovered from grapevine bunch stem and cane from Malbec grape cultivar. The total PCs in cane and bunch stem extracts were 74 and 20% bioaccessible, respectively. Syringic acid, cinnamic acid, ε-viniferin, naringenin and myricetin were highly bioaccessible, noticeably ε-viniferin in cane extract with 137%. The high bioaccessibility observed, particularly for compounds at high concentration such as ε-viniferin, will help to better understand the bioactive potential of these by-products. In this sense, bunch stems and canes can be considered as new and sustainable sources of bioactive substances for applications as functional ingredients or nutraceuticals in food and pharmaceutical industries.

Metabolic syndrome (MetS) is a risk factor for sudden cardiac death in humans, but animal models are needed for the study of this association. Grape pomace (GP), obtained from the winemaking process, contains phenolic compounds with potential cardioprotective effects. The aim of this study was to evaluate if a high-fat-fructose (HFF) diet facilitates the occurrence of arrhythmias during the reperfusion, and if a GP supplementation could counteract these effects. Wistar rats were fed with control (Ctrl), HFF diet and HFF plus GP (1 g kg-1 day-1) for six weeks. The HFF diet induces characteristic features of MetS (higher systolic blood pressure, dyslipidemia and insulin resistance) which was attenuated by GP supplementation. In addition, HFF induced increased reperfusion arrhythmias that were reduced upon GP supplementation. GP also reduced the non-phosphorylated form of connexin-43 (Cx43) while enhancing heart p-AKT and p-eNOS protein levels and reducing Nox4 levels enhanced by the HFF diet, indicating that GP may increase NO bioavailability in the heart. We found a murine model of MetS with increased arrhythmogenesis and translational value. Furthermore, GP prevents diet-induced heart dysfunction and metabolic alterations. These results highlight the potential utilization of winemaking by-products containing significant amounts of bioactive compounds to prevent/attenuate MetS-associated cardiovascular pathologies.

Malbec wines from 27 parcels from the three most important winemaking regions of Mendoza, Argentina, were produced under standardized winemaking conditions, and analyzed for phenolic composition and by means of sensory descriptive analysis. Different methods of characterization and cluster analysis for each data matrix showed that some locations of Mendoza could be significantly separated from each other. The results of unsupervised statistical methods were compared using a test for similarities and divergences, also showing that different locations may be associated. The current report is the first one characterizing Malbec wines from the three major producing regions of Argentina using two different ways for locations classification. The effects of climate and geographical origin of Malbec grapes on the quality parameters of resulting wines were also evidenced. These results have enological and viticulture interest for the winemaking industry as the vineyard site selection for Malbec can considerably affect quality attributes. Fil: Urvieta, Roy Alexander. The suitability of dispersive liquid–liquid microextraction (DLLME) and gas chromatography accurate mass spectrometry (GC–MS), based on a time-of-flight (TOF) MS analyzer and using electron ionization (EI), for the characterization of volatile and semi-volatile profiles of grape marc distillates (grappa) are evaluated. DLLME conditions are optimized with a selection of compounds, from different chemical families, present in the distillate spirit. Under final working conditions, 2.5 mL of sample and 0.5 mL of organic solvents are consumed in the sample preparation process. The absolute extraction efficiencies ranged from 30 to 100%, depending on the compound. For the same sample volume, DLLME provided higher responses than solid-phase microextraction (SPME) for most of the model compounds. The GC-EI-TOF-MS records of grappa samples were processed using a data mining non-targeted search algorithm. In this way, chromatographic peaks and accurate EI-MS spectra of sample components were linked. The identities of more than 140 of these components are proposed from comparison of their accurate spectra with those in a low resolution EI-MS database, accurate masses of most intense fragment ions of known structure, and available chromatographic retention index. The use of chromatographic and spectral data, associated to the set of components mined from different grappa samples, for multivariate analysis purposes is also illustrated in the study. A new reliable method for the determination 3-alkyl-2-methoxypyrazines (MPs) in wine samples based on the sequential combination of solid-phase extraction (SPE), dispersive liquid–liquid microextraction (DLLME) and gas chromatography (GC) quadrupole time-of-flight accurate tandem mass spectrometry (QTOF-MS/MS) is presented. Primary extraction of target analytes was carried out by using a reversed-phase Oasis HLB (200 mg) SPE cartridge combined with acetonitrile as elution solvent. Afterwards, the SPE extract was submitted to DLLME concentration using 0.06 mL carbon tetrachloride (CCl4) as extractant. Under final working conditions, sample concentration factors above 379 times and limits of quantification (LOQs) between 0.3 and 2.1 ng L−1 were achieved. Moreover, the overall extraction efficiency of the method was unaffected by the particular characteristics of each wine; thus, accurate results (relative recoveries from 84 to 108% for samples spiked at concentrations from 5 to 25 ng L−1) were obtained using matrix-matched standards, without using standard additions over every sample. Highly selective chromatographic records were achieved considering a mass window of 5 mDa, centered in the quantification product ion corresponding to each compound. Twelve commercial wines, elaborated with grapes from different varieties and geographical origins, were processed with the optimized method. The 2-isobutyl-3-methoxypyrazine (IBMP) was determined at levels above the LOQs of the method in half of the samples. Fil: Fontana, Ariel Ramón. Plants are able to synthesize a large number of organic compounds. Among them, primary metabolites are known to participate in plant growth and development, whereas secondary metabolites are mostly involved in defense and other facultative processes. In grapevine, one of the major fruit crops in the world, secondary metabolites, mainly polyphenols, are of great interest for the wine industry. Even though there is an extensive literature on the content and profile of those compounds in berries, scarce or no information is available regarding polyphenols in other organs. In addition, little is known about the effect of plant growth regulators (PGRs), ABA and GA3 (extensively used in table grapes) on the synthesis of primary and secondary metabolites in wine grapes. In table grapes, cultural practices include the use of GA3 sprays shortly before veraison, to increase berry and bunch size, and sugar content in fruits. Meanwhile, ABA applications to the berries on pre-veraison improve the skin coloring and sugar accumulation, anticipating the onset of veraison. Accordingly, the aim of this study was to assess and characterize primary and secondary metabolites in leaves, berries and roots of grapevine plants cv. Malbec at veraison, and changes in compositions after ABA and GA3 aerial sprayings. Metabolic profiling was conducted using GC-MS, GC-FID and HPLC-MWD. A large set of metabolites was identified: sugars, alditols, organic acids, amino acids, polyphenols (flavonoids and non-flavonoids) and terpenes (mono-, sesqui-, di- and triterpenes). The obtained results showed that ABA applications elicited synthesis of mono- and sesquiterpenes in all assessed tissues, as well as L-proline, acidic amino acids and anthocyanins in leaves. Additionally, applications with GA3 elicited synthesis of L-proline in berries, and mono- and sesquiterpenes in all the tissues. However, treatment with GA3 seemed to block polyphenol synthesis, mainly in berries. In conclusion, ABA and GA3 applications to grapevine plants cv. Malbec influenced the synthesis of primary and secondary metabolites known to be essential for coping with biotic and abiotic stresses.

In-vial filtration with dispersive solid-phase extraction (d-SPE) clean-up of QuEChERS (quick, easy, cheap, effective, rugged and safe) extracts is proposed for the determination of ethylphenols (EPs) in red wines. Analytes were extracted from 5mL wine sample (previously alkalinized with 0.5% sodium hydroxide) using 5mL acetonitrile. For phase separation, 1.5g NaCl and 4g anhydrous MgSO4 were added. Then, a 0.5mL aliquot of the partitioned supernatant was cleaned-up using d-SPE and in-vial filtration with a combination of anhydrous CaCl2 (100mg) and primary-secondary amine (PSA, 25mg) as sorbents. The proposed method provided limits of quantification (LOQs) ranging from 0.024 to 0.04mgL-1. Considering matrix-matched calibration as quantification technique, the recoveries (accuracy) ranged between 73% and 116%. The method was applied for the determination of EPs in 15 commercial wines of Argentina, where 4-EP was quantified at concentrations ranging from 0.25 to 3.01mgL-1.

In this study the effect of diet supplementation with grape pomace (GP) and grape pomace extract (GPE) on insulin sensitive tissues (adipose, liver and muscle) was evaluated in an experimental model of metabolic syndrome (MetS). MetS was developed by giving a high-fat-fructose (HFF) diet to Wistar rats. Six weeks of HFF diet induced weight gain, which was partially attenuated by GP (1 g per kg per day) and GPE (300 mg per kg per day) supplementation. HFF diet increased systolic blood pressure, triglycerides, insulin resistance (HOMA:IR) and inflammation (c-reactive protein (CRP)). Supplementation with GP prevented SBP, triglycerides and CRP increased and partially attenuated insulin resistance. On the other hand, GPE partially reduced SBP and triglycerides and significantly prevented insulin resistance and inflammation. Also, HFF diet induced higher triglycerides content and enhanced NADPH oxidase activity in the liver. Also, HFF diet increased the epididymal adipose tissue weight, enlarged adipocyte size, and c-jun N-terminal kinase (JNK) activation, probably contributing to a pro-inflammatory cytokine pattern (higher resistin) and lower adiponectin protein expression. These alterations may result in an impairment of insulin signaling cascade observed in adipose, liver and muscle tissue (IRS1, Akt, and extracellular signal-regulated kinases (ERK1/2)) from HFF rats. Supplementation with GP and to a greater extent GPE attenuated liver triglyceride content and adiposity and restored adipose, liver and muscle response to insulin. These findings show that supplementation with GP and GPE to a greater extent can counteract adiposity, inflammation, liver damage and impaired insulin signaling associated to MetS, supporting the utilization of winemaking residues in food industry/human health due to their high amount of bioactive compounds. Dispersive solid-phase extraction (DSPE) is proposed for the first time as a simplified, fast and low cost clean-up technique of biological sample extracts for polybrominated diphenyl ethers (PBDEs) determination. The combination of a traditional extraction technique, such as ultrasound-assisted leaching (USAL) with DSPE was successfully applied for sample preparation prior to gas chromatography–tandem mass spectrometry (GC–MS/MS) analysis. The analytes were first extracted from 1 g homogenized sample in n-hexane:dichloromethane (8:2) by applying USAL technique and further cleaned-up using DSPE with 0.20 g C 18 -silica as sorbent material. Different solvent mixtures, sorbent type and amount, and lipid digestion procedures were evaluated in terms of clean-up and extraction efficiency. Under optimum conditions, the method detection limits (MDLs) for PBDEs, calculated as three times the signal-to-noise ratio (S/N) were within the range 9–44 pg g −1 wet weight. The calibration graphs were linear within the concentration range of 53–500,000 pg g −1 , 66–500,000 pg g −1 , 89–500,000 pg g −1 and 151–500,000 pg g −1 for BDE-47, BDE-100, BDE-99 and BDE-153, respectively; and the coefficient of determination ( r 2 ) exceeded 0.9992 for all analytes. The proposed methodology was compared with a reference solid-phase extraction technique. The applicability of the methodology for the screening of PBDEs has been demonstrated by analyzing spiked and real samples of biological nature (fish, egg and chicken) with different lipid content as well as reference material (WELL-WMF-01). Recovery values ranged between 75% and 114% and the measured concentrations in certified material showed a reasonable agreement with the certified ones. BDE-47, BDE-100 and BDE-99 were quantified in three of the seven analyzed samples and the concentrations ranged between 91 and 140 pg g −1 . In addition, this work is the first description of PBDEs detected in fish of Argentinean environment.

In this work, a reliable and selective procedure for the determination of thirteen fungicides in red and white wine samples is proposed. Solid-phase extraction (SPE) and liquid chromatography (LC) tandem mass spectrometry (MS/MS), based on a hybrid quadrupole time-of-flight (QTOF) system, were used as sample preparation and determination techniques, respectively. Extraction and purification of target analytes was carried out simultaneously by using a reversed-phase Oasis HLB (200 mg) SPE cartridge combined with acetonitrile as elution solvent. Fungicides were determined operating the electrospray source in the positive ionization mode, with MS/MS conditions adjusted to obtain at least two intense product ions per compound, or registering two transitions per species when a single product was noticed. High selective MS/MS chromatograms were extracted using a mass window of 20 ppms for each product ion. Considering external calibration as quantification technique, the overall recoveries (accuracy) of the procedure ranged between 81% and 114% for red and white wine samples (10-20 mL), spiked at different concentrations between 5 and 100 ng mL-1. Relative standard deviations of the above data stayed below 12% and the limits of quantification (LOQs) of the method, calculated for 10 mL of wine, varied between 0.1 ng mL-1 for cyprodinil (CYP) and 0.7 ng mL-1< for myclobutanil (MYC). The optimized method was applied to seventeen commercial wines produced in Spain and obtained from local supermarkets. Nine fungicides were determined, at levels above the LOQs of the method, in the above samples. The maximum concentrations and the highest occurrence frequencies corresponded to metalaxyl (MET) and iprovalicarb (IPR). Fil: Fontana, Ariel Ramón. A novel and efficient analytical methodology is proposed for extracting and preconcentrating polybrominated diphenyl ethers (PBDEs) from samples of environmental interest prior gas chromatography-mass spectrometry (GC-MS) analysis. It is based on the induction of micellar organized medium by using a non-ionic surfactant (Triton X-114) to extract the target PBDEs. To enable coupling the efficient extracting technique with GC analysis, ultrasound-assisted back-extraction (UABE) into an organic solvent was required. Several factors, including surfactant type and concentration, equilibration temperature and time, ionic strength, pH and buffers nature and concentration were studied and optimized over the extraction efficiency of the proposed technique. Under optimal experimental conditions, the target analytes were quantitatively extracted achieving an enrichment factor of 250 when 10 mL aliquot of ultrapure water spiked with PBDE-standard mixture (10 pg mL-1 each PBDE) was extracted. Method detection limits (MDLs) calculated with aqueous PBDEs solutions as three times the signal-to-noise ratio (S/N), ranged from 1 to 2 pg mL-1 with RSDs values ≤8.5% (n = 5). The coefficients of estimation of the calibration curves obtained following the proposed methodology were ≥0.9987 and linear range of all PBDEs was 4-150 pg mL-1. The proposed methodology was validated by carrying out a recovery study by spiking the samples at two different concentration levels of PBDEs (10 and 50 pg mL-1 for waters samples). Recoveries values in the range of 96-106% for water samples were obtained showing satisfactory robustness of the method for analyzing PBDEs in water samples. A simple, efficient, innovative and environmentally friendly analytical technique was successfully applied for the first time for the extraction and preconcentration of polybrominated diphenyl ethers (PBDEs) from water samples. The PBDEs selected for this work were those most commonly found in the literature in natural water samples: 2,2′,4,4′-tetraBDE (BDE-47), 2,2′,4,4,5-pentaBDE (BDE-99), 2,2′,4,4,6-pentaBDE (BDE-100) and 2,2,4,4′,5,5′-hexaBDE (BDE-153). The extracted PBDEs were separated and determined by gas chromatography-mass spectrometry (GC-MS). The extraction/preconcentration technique is based on ultrasound-assisted emulsification-microextraction (USAEME) of a water-immiscible solvent in an aqueous medium. Several variables including, solvent type, extraction time, extraction temperature and matrix modifiers were studied and optimized over the relative response the target analytes. Chloroform was used as extraction solvent in the USAEME technique. Under optimum conditions, the target analytes were quantitatively extracted achieving enrichment factors (EF) higher than 319. The detection limits (LODs) of the analytes for the preconcentration of 10 mL sample volume were within the range 1-2 pg mL-1. A rapid and economically affordable reverse-phase chromatographic approach based on a core-shell column with high-performance liquid chromatography multi-wavelength detector (HPLC-MWD) is proposed for the quantification and quality control of multiclass polyphenols (PPs). The separation of 20 relevant polyphenols from grape pomace extracts (GPEs) was achieved in less than 12 min by using a Kinetex C18 column (3.0 mm x 100 mm, 2.6 µm) with a gradient system of ultrapure water (0.1 % formic acid) and acetonitrile, a temperature of 35 °C and a flow rate of 0.8 mL min-1. The maximum backpressure reached was 327 bar, meaning the developed method is adequate for standard HPLC instruments. The applicability of the method was demonstrated by the determination of PPs in GPEs of different red grape varieties. Cabernet Sauvignon GPE showed the highest content of studied PPs (9804.2 µg g-1 GPE) followed by Bonarda GPE (7302.0 µg g-1 GPE). Besides the methodological development for a high throughput routine quality control of GPEs, this is the first report of PPs content for Bonarda and Aspirant Bouchet GPE, so the results add knowledge for these grape varieties cultivated in Argentina. Low molecular weight polyphenols (LMW-PPs) and anthocyanins, along with the antioxidant capacity, were assessed in grape pomace extract (GPE) of red grape (Vitis vinifera L.) cv. Malbec. Twenty-six phenolics (13 LMW-PPs and 13 anthocyanins) were characterized and quantified by HPLC-MWD and UPLC?ESI-MS. The maximum concentrations of LMW-PPs corresponded to the flavanols (+)-catechin and ()-epicatechin, whereas malvidin-3-glucoside was the most abundant anthocyanin. Piceatannol, a stilbene analogue to resveratrol with higher antioxidant activity, was firstly identified and quantified in GPE of the cv. Malbec. The antioxidant activity for Malbec GPE determined by oxygen radical absorbance capacity (ORAC) assay was 2756 lmol TE g1 GPE. Therefore, the data reported sustain the use of winemaking by-products as a cheap source of phenolic compounds suitable for biotechnological applications, as a strategy for sustainable oenology. In this work, a reliable, simple, fast, inexpensive and robust sample preparation approach for the determination of multiclass polyphenols in wine samples is proposed. The polyphenols selected for this work were gallic acid, (+)-catechin, (-)-epicatechin, caffeic acid, syringic acid, coumaric acid, ferulic acid, trans-resveratrol, quercetin and cinnamic acid. The method is based on QuEChERS (quick, easy, cheap, effective, rugged and safe) extraction technique coupled with dispersive solid-phase extraction (d-SPE) clean-up. Under optimized conditions, the analytes were extracted from 5 mL wine samples (previously acidified with 1 % formic acid) using 2.5 mL acetonitrile. For phase separation, 1.5 g NaCl and 4 g anhydrous MgSO4 were added. Then, a 1 mL aliquot of the partitioned supernatant was cleaned-up using d-SPE with a combination of 150 mg CaCl2, 50 mg primary-secondary amine (PSA) and 50 mg C18 as sorbents. A 250 µL aliquot of the obtained cleaned extract was concentrated to dryness and taken up with the initial mobile phase previous to liquid chromatography-multi-wavelength detection (LC-MWD). The proposed method provided limits of detection (LODs) ranging from 0.004-0.079 µg mL-1 and an inter-day variability below 12 % RSD for all analytes in red and white wine samples. Considering external calibration (red wines) and matrix-matched calibration (white wines) as quantification techniques, the overall recoveries (accuracy) of the method ranged between 75.0 % and 119.6 % for red and white wine samples, respectively. The developed method was applied for the determination of polyphenols in 10 wines produced in Argentina. Nine phenolic compounds were determined, at concentrations above detectable levels in the method. The maximum concentrations corresponded to (-)-epicatechin in white wines, while gallic acid and (+)-catechin were the most abundant in red wines. Fil: Fontana, Ariel Ramón. A novel method for the residue analysis of wine spoilage compound 2,4,6-trichloroanisole is reported. Wine (60 ml) was extracted with 2 ml toluene in presence of 24 g MgSO4 and 6 g NaCl. Cleanup of the toluene phase by dispersive solid phase extraction with mixture of 100 mg CaCl2, 25 mg primary secondary amine and 50 mg MgSO4 was effective in minimising co-extractives and matrix effects. Time-of-flight and tandem mass spectrometric parameters were optimised to achieve linearity over 0.25–500 ng ml1 and method detection limit 0.0083 ng ml1 which is well below the odour threshold of 0.04 ng ml1 . Coacervative microextraction ultrasound-assisted back-extraction technique (CME-UABE)is proposed for the first time for extracting and preconcentrating organophosphates pesticides (OPPs) from honey samples prior to gas chromatography-mass spectrometry (GC-MS) analysis.The extraction/preconcentration technique is supported on the micellar organized medium based on non-ionic surfactant. To enable coupling the proposed technique with GC, it was required to back extract the analytes into hexane. Several variables including, surfactant type and concentration, equilibration temperature and time, matrix modifiers, pH and buffers nature were studied and optimized over the relative response of the analytes. The best working conditions were as follows: an aliquot of 10 mL 50 g L-1 honey blend solution was conditioned by adding 100 µL 0.1 mol L-1 hydrochloric acid (pH 2) and finally extracted with 100 µL Triton X-114 100 g L-1 at 85 °C for 5 min by using CME technique. Under optimal experimental conditions, the enrichment factor (EF) was 167 and limits of detection (LODs), calculated as three times the signal-to-noise ratio (S/N=3), ranged between 0.03 ng g-1 and 0.47 ng g-1. The method precision was evaluated over five replicates at 1 ng g-1 with RSDs lower than 9.5%.The calibration graphs were linear within the concentration range of 0.3-1000 ng g-1 for Chlorpirifos; and 1-1000 ng g-1 for Fenitrothion, Parathion and Methidathion; respectively. The coefficients of correlation were higher than 0.9992.Validation of the methodology was performed by standard addition method at two concentration levels (2 and 20 ng g-1). The recoveries were higher than 90%, indicating satisfactory robustness of the methodology, which could be successfully applied for determination of OPPs in honey samples of different Argentinean regions.
